Claims (12)
- 피루베이트 디카르복실라아제 (Pyruvate decarboxylase, PDC)의 활성이 내재적 활성에 비해 불활성화되고, ATP-시트르산 분해효소 (ATP-citrate lyase, ACL) 활성이 도입되고, 피루베이트 생합성 경로가 내재적 생합성 경로에 비하여 강화되도록 변이된, 젖산 생산능을 갖는 사카로마이세스 속 (Saccharomyces sp.) 미생물.
- 제1항에 있어서, 상기 피루베이트 생합성 경로의 강화는 포스포에놀 피루베이트 카르복시키나아제 1 (Phosphoenolpyruvate carboxykinase 1, PCK1) 또는 피루베이트 키나아제 2 (Pyruvate kinase 2, PYK2), 또는 모두의 활성이 내재적 활성에 비하여 강화되도록 변이시킨, 미생물.
- 제1항에 있어서, 상기 피루베이트 생합성 경로의 강화는 말레이트 디히드로게네이즈 2 (Malate dehydrogenase 2, MDH2) 또는 세포 기질 말릭 효소 1 (cytosolic Malic enzyme 1, cytosolic MAE1), 또는 모두의 활성이 내재적 활성에 비하여 강화되도록 변이시킨, 미생물.
- 제1항에 있어서, 상기 피루베이트 디카르복실라아제는 서열번호 39, 41, 및 43의 아미노산 서열로 이루어진 군에서 선택된 1개 이상의 아미노산 서열로 기재되는 효소인, 미생물.
- 제1항에 있어서, 상기 ATP-시트르산 분해효소는 서열번호 29의 아미노산 서열로 기재되는 효소인, 미생물.
- 제2항에 있어서, 상기 포스포에놀 피루베이트 카르복시키나아제 1은 서열번호 31의 아미노산의 서열로 기재되는 효소이며, 피루베이트 키나아제 2는 서열번호 33의 서열로 기재되는 효소인, 미생물.
- 제3항에 있어서, 상기 말레이트 디히드로게네이즈 2는 서열번호 35의 아미노산 서열로 기재되는 효소이며, 세포 기질 말릭 효소 1은 서열번호 37의 아미노산 서열 또는 서열번호 52의 아미노산 서열로 기재되는 효소인, 미생물.
- 제1항에 있어서, 상기 미생물은 추가적으로 락테이트 디히드로게네이즈 (Lactate dehydrogenase, LDH) 활성이 도입된, 미생물.
- 제8항에 있어서, 상기 락테이트 디히드로게네이즈는 서열번호 49의 아미노산 서열로 기재되는 효소인, 미생물.
- 제1항에 있어서, 상기 미생물은 추가적으로(i) 알코올 디히드로게네이즈 1 (Alcohol dehydrogenase 1, ADH1)의 활성이 내재적 활성에 비해 불활성화되도록 변이됨;(ii) 피루베이트 디카르복실라아제 1 (Pyruvate decarboxylase 1, PDC1)의 활성이 내재적 활성에 비해 불활성화되도록 변이됨; 및(iii) D-락테이트 디히드로게네이즈 1 (D-lactate dehydrogenase 1, DLD1)의 활성이 내재적 활성에 비해 불활성화되도록 변이된 것인, 미생물.
- 제1항에 있어서, 상기 사카로마이세스 속 미생물은 사카로마이세스 세레비지애 (Saccharomyces cerevisiase)인 미생물.
- (i) 제1항 내지 제11항 중 어느 한 항의 사카로마이세스 속 (Saccharomyces sp.) 미생물을 배지에서 배양하는 단계; 및(ii) 상기 배양에 따른 배지 또는 상기 미생물로부터 젖산을 회수하는 단계를 포함하는, 젖산 제조 방법.
