斯かる本発明の化合物は、単独で用いてもよく、2種以上を混合して用いてもよい。
本発明の化合物のうち、昆虫忌避効果及び使用感の点から好適なものとして、例えば、オクタヒドロ-7-メチル-1,4-メタノナフタレン-6(2H)-オン:フィルメニッヒ(プリカトン)、3-メチル-5-プロピル-2-シクロヘキセン-1-オン(グラベノン)、6-メチルキノリンが挙げられる。
ここで、二酸化炭素応答抑制とは、昆虫が有する二酸化炭素受容応答を無力化又は低減化して、二酸化炭素に対する認知感覚を制御し、二酸化炭素源への誘引行動を抑制することを意味する。昆虫の二酸化炭素応答は、公知の単一感覚毛記録法(Nature 461, 277-281 (10 September 2009))を用いて、電極を感覚毛に刺入後、二酸化炭素刺激に対する神経発火パターンを確認することにより測定することができる。
二酸化炭素応答抑制作用を有する香料化合物としては、例えば、後述の参考例に示す化合物、すなわち、2-メトキシ-4-プロピルフェノール、2-イソプロピル-4-メチルチアゾール、イソシクロシトラール、9-デセン-1-オール、1-オクテン-3-オール、1-デカノール、2-エチルヘキサン酸エチル、酢酸 トランス-2-ヘキセニル、2-シクロヘキシルプロピオン酸エチル、フェニル酢酸エチル、2,4,6-トリメチル-3-シクロヘキセン-1-メタノール、1-オクタノール、2,4-ジメチルシクロヘキセ-3-エン-1-メタノール、o-tert-ブチルシクロヘキシル アセテート、2-メチル酪酸、プロピオン酸ベンジル、3-メチル-2-ペンチルシクロペント-2-エン-1-オン、cis-3-ヘキセニル アセテート等が挙げられる。このうち、2,4-ジメチルシクロヘキセ-3-エン-1-メタノール(フロラロール)、1-オクタノール、cis-3-ヘキセニル アセテート等が好ましく、2,4-ジメチルシクロヘキセ-3-エン-1-メタノールがより好ましい。
ここで、「昆虫忌避」とは、昆虫を対象物へ寄せ付けないこと、或いは昆虫が対象物へ寄り付かないことを意味し、「昆虫の熱受容抑制」とは、昆虫が有する熱受容応答を無力化又は低減化して、熱源に対する認知感覚を抑制し、熱源への誘引行動を抑制することを意味し、昆虫に対して殺虫力を有し、昆虫を駆除するものとは異なる。
尚、熱受容はプレートヒーターのような熱源を配置したケージの中に昆虫を放ち、熱源へ誘引された昆虫の数を計測することにより測定することができる。
また、添加物としては、種々の化成品、化粧品等に一般に用いられる添加物が挙げられ、具体的には、界面活性剤、有機溶媒、油性成分、保湿剤、粉体、可溶化剤、増粘剤、樹脂、洗浄剤、防腐剤、紫外線吸収剤、無機物、香料、色素、消臭剤、精油、薬剤、植物抽出物、その他の忌避剤等が挙げられる。

<24><13>、<16>、<22>又は<23>において、香料化合物は、2,4-ジメチルシクロヘキセ-3-エン-1-メタノール、1-オクタノール及びcis-3-ヘキセニル アセテートから選ばれる1種以上である。
<25><13>、<16>、<22>、<23>又は<24>において、<12>の化合物、<15>の化合物、<20>の化合物又は<21>の化合物は、オクタヒドロ-7-メチル-1,4-メタノナフタレン-6(2H)-オン又は3-メチル-5-プロピル-2-シクロヘキセン-1-オンである。
(1)ヒトスジシマカの準備
ヒトスジシマカは、住友テクノサービス株式会社より購入した卵を成長させたものを使用した。透明なプラスチックパンに水を1cm程度張り、卵が産み付けられているろ紙を入れた。毎日、幼虫用餌として、熱帯魚用エサ(テトラミン)を与えた。約1週間後、蛹をスポイトで回収し、20mL用プラスチックカップに移し、網を張ったケージに移した。成虫用の餌として、10質量%スクロースを25mLプラスチックチューブに入れたものを与えた。羽化後、5日間オスとメスを同じケージで飼育することで交尾を行わせた。飼育5日後、成虫を吸虫管を用いて集め、氷上で5分間麻酔後に、目視下でオスとメスを分け、メスのみを回収した。
約150-200匹のメスヒトスジシマカをプラスチックケージ(30×30×30cm)に移した。メス蚊の行動をケージの上部からモニターするため、プラスチックケージの上面を透明なアクリル板に取り換えた。また、熱源となるプレートヒーター(小動物用マルチパネルヒーター16W ビバリア製)はケージの一隅に5×3cm露出するように、メッシュの外側から配置した。上面から熱源が撮影できるように上部にビデオカメラ(ソニー製)を配置し、実験を行った。熱源に両面テープを用いて、試料塗布用のシートを固定した。
表1に示す香料を、エタノールを用いて濃度0.1%に調整したものを試料とし、当該試料あるいは溶媒エタノール50μL投与して実験を行った。メス蚊の活性化のため、二酸化炭素1%を上部から1秒間投与し、その後、熱源をケージに接触させた。30秒間接触後の熱源を写真に撮り、誘引された蚊の数を数えた。アッセイが一度終了するたびに熱源に使用したシートは廃棄し、新たなシートを使用した。また、蚊への試料の影響を考え、一つのケージで行う評価は最大で(7試料+1コントロール)とした。抑制効果は、同じアッセイ(7試料+1コントロール)内でエタノール溶媒のみを塗布した場合の降着数と比較して、下記式により抑制効果率を計算した。

ヒトスジシマカは、住友テクノサービス株式会社より成虫として購入した生後5-10日のメス220匹、オス80匹を30cm×30cm×30cmのプラスチックケージ(bug dorm)に入れた。
評価試料として、2,4-ジメチルシクロヘキセ-3-エン-1-メタノール(「フロラロール」、IFF社)、オクタヒドロ-7-メチル-1,4-メタノナフタレン-6(2H)-オン(「プリカトン」、FIRMENICH社)をエタノールでそれぞれ0.4%に濃度を調整した。また、エタノールのみの試料を対照とした。
綿100%手袋を二重に片手にはめ、そこに上記評価試料及び対照試料をスプレーにて表裏に計14プッシュ(0.6g)吹き付けた。
試料付与後、1分間放置しエタノールを揮発させたのち、試料付与した手袋をはめた手をプラスチックケージに入れ、1分ごとに手袋に降着した蚊の数をカウントした。
1分,2分,3分のそれぞれの降着数、及びそれらの総計をカウントし、対照試料付与を100%としたときの降着率を計算した。同じ実験を3回繰り返した。結果を表3に示す。本発明の化合物は、フロラロール(2,4-ジメチルシクロヘキセ-3-エン-1-メタノール)との併用により、蚊のヒト腕への誘引行動がより効果的に抑制されることが確認された。

1Mol/L水酸化カリウム水溶液を50mLシリンジに注入し、シリンジをクランプでマグネットスタンドに水平に固定した。電極ホルダーに固定したタングステンワイヤーを水平方向からシリンジの中へと挿入し、顕微鏡下で観察しながら電気分解により、研磨を行なった。交流電源からワニ口クリップで、電極ホルダー、シリンジへと電流を供給した。
スライドグラス上に準備したヒトスジシマカを顕微鏡下に置き、×10対物レンズで触角が視野の中心部に来るように位置を調節した。基準電極を昆虫の目に刺入した。匂い刺激用のピペットを小顎髭の近くに設置し、対物レンズを×100へと変換した。記録電極を感覚毛に近づけ、微動マニュピレーターを用いて慎重に感覚毛に刺入した。ヒトスジシマカの二酸化炭素感受性の感覚毛は、ペグ状の形状を持つことが顕微鏡下で確認できる。電極を感覚毛に刺入後、自発的な神経発火パターンを確認した。0.1%濃度に調整した二酸化炭素をシリコンチューブで匂い刺激装置につなぎ、小顎髭を1秒間刺激し、二酸化炭素応答を確認した。
試料は、表4に示す香料を、パラフィンオイルを用いて濃度1%に調整したものを使用した。試料をフィルターペーパー(3mm×50mm)上に15μL滴下し、パスツールピペット内に挿入した。ビニールチューブを用いて、パスツールピペットと刺激装置を接続し、空気中の二酸化炭素への応答をモニターしながら、1秒間の匂い刺激を行い、二酸化炭素への応答抑制効果を評価した。応答抑制効果は、溶媒であるパラフィンオイルで刺激後1秒間の神経の発火数、それぞれの試料で刺激後1秒間の神経の発火数を計測し、その比率による評価を行った(N=1)。
