Abstract

본 발명은 탈분화 식물 프로토플라스트 내에 활성 물질이 인입된 구조체, 이의 제조방법 및 이를 포함하는 화장료 조성물에 관한 것으로, 더욱 상세하게는 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체, 이의 제조방법 및 이를 유효 성분으로 포함하는 것을 특징으로 하는 화장료 조성물에 관한 것이다. 상기 구조체는 기존에 식물 세포가 갖고 있는 활성 성분의 안정성이 그대로 유지되고 추가적으로 세포 내 인입된 활성 물질의 안정성이 높으며, 이를 포함하는 화장료 조성물은 상기 활성 물질에 의한 효능 또한 향상되어 다양한 효능을 확보할 수 있다.

Description

탈분화 식물 프로토플라스트 내에 활성 물질이 인입된 구조체, 이의 제조방법 및 이를 포함하는 화장료 조성물
본 발명은 활성 물질의 안정성 및 효능을 높이기 위해 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체, 이의 제조방법 및 이를 유효 성분으로 포함하는 화장료 조성물에 관한 것이다.
천연에 존재하는 식물에는 각기 다른 수많은 활성 물질(phyto-chemical)이 내재되어 있으며, 그 종류 또한 테르페노이드, 플라보노이드, 플라보놀, 폴리페놀, 아미노산, 리그난, 알칼로이드, 비타민, 카테킨 화합물 등 무수한 종류로 이루어져 있다.
이에 식물에서 특정 성분만을 추출하거나 식물 세포를 이용한 화장료 조성물이 활발히 제시되고 있다.
일례로, 국제특허 WO2013-180526호에서는 에델바이스 추출물이 피부 재생을 촉진시켜 화장료로서 이용 가능하고, 국제특허 WO2012-102456호에서는 비단풀 추출물이 주름 개선에 효과적이어서 다양한 화장료로 사용될 수 있음을 언급하고 있다.
또한, 국제특허 WO2009-139581호는 주목의 형성층 또는 전형성층에서 유도되고, 다음의 특성을 갖는 세포주 등이 항산화 및 항염증 효과가 있어 노화를 방지하고 멜라닌 생성을 억제함에 따라 화장료 조성물로 사용 가능함을 개시하고 있고, 일본 공개특허 제2012-102136호는 피부 탈색 및 라이트닝을 위해 크리스테마린(Criste Marine)과 같은 호염성 식물의 탈분화 식물 세포의 동결 건조물을 포함하는 화장품 조성을 제시하고 있다.
한편, 레티놀 등과 같은 유효 활성 물질은 인체 내 생리학적 특성에 있어서 유용한 기능을 가짐에도 안정성 면에서 자유롭지 못해, 이의 안정성을 높이기 위해 다양한 방법이 제시되고 있다.
일례로, 활성 물질에 인지질 혹은 계면활성제를 이용한 리포좀, 세라마이드를 이용한 안정화된 세라좀, 액정구조로 안정화시킨 액정좀, 모노글리세라이드를 이용한 입방상 큐보좀 적용 등 많은 시도가 이루어지고 있다.
이와 더불어 식물 세포를 화장료 조성물에 적용하고자 하는 연구 또한 진행되고 있다.
국제특허 WO2012-173458호는 내부에 갈릭산(Gallic acid), 아미노산 등의 활성물질이 안정화된 식물 세포를 함유하는 화장료 조성물을 제시하고 있으며, 이때 상기 식물 세포는 세포벽으로서, 세포벽 내에 활성 물질을 인입시키는 경우 활성 물질의 안정성이 높아질 수 있음을 제시하였다.
식물 세포의 세포벽은 과량의 셀룰로오스를 포함하는데, 이로 인해 다른 물질의 침투가 용이하지 않아 활성 물질의 인입을 위해선 제거가 필요하고, 세포벽과 세포막을 모두 제거할 경우 인입된 활성 물질의 안정성이 오히려 저하될 수 있다.

이에 본 발명자들은 식물 세포 내에 활성 물질을 높은 인입률로 안정하게 포접할 수 있도록 다각적으로 연구를 수행한 결과, 식물 세포의 세포벽을 제거하고 세포막 및 세포소로 이루어진 프로토플라스트(protoplast)를 사용할 경우 생체에 대한 침투성 뿐만 아니라 활성 물질의 인입률을 높여 화장료 조성물로서 사용 가능함을 확인하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 활성 물질이 높은 인입률로 인입되어, 상기 활성 물질의 효능을 극대화할 수 있는 구조체 및 이의 제조방법을 제공하는 것이다.
또한, 본 발명의 다른 목적은 상기 구조체를 유효 성분으로 포함하는 화장료 조성물을 제공하는 것이다.
상기 목적을 달성하기 위해, 본 발명은 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체를 제공한다.
또한, 본 발명은
식물의 잎, 줄기, 뿌리, 꽃, 열매 및 씨앗에서 식물의 세포를 수득하는 단계;
상기 수득된 식물 세포를 옥신을 이용하여 탈분화하여 탈분화된 식물 세포를 얻는 단계;
상기 얻어진 탈분화 식물 세포를 대량으로 배양하는 단계;
상기 대량 배양된 탈분화 식물 세포의 세포벽을 효소 반응을 이용하여 제거하여 프로토플라스트를 얻는 단계;
상기 얻어진 탈분화 식물 프로토플라스트 내에 가압 삼투 공정으로 활성 물질을 인입하는 단계; 및
후처리하는 단계를 포함하는
탈분화 식물 프로토플라스트 내에 활성 물질이 인입된 구조체의 제조방법을 제공한다.
또한, 본 발명은 상기 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체를 유효 성분으로 포함하는 화장료 조성물을 제공한다.
본 발명에 따른 구조체는 기존에 식물 세포가 갖고 있는 활성 성분의 안정성이 그대로 유지되고 추가적으로 세포 내 인입된 활성 물질의 안정성이 높아, 상기 활성 물질에 의한 효능 또한 향상되어 화장료의 유효 성분 조성으로 바람직하게 사용 가능하다.

이하 본 발명을 더욱 상세히 설명한다.
본 발명에서는 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체를 제시한다.
프로토플라스트는 세포의 세포벽은 제거되고 세포막이 존재하는 원형질체로, 상기 세포막이 내부에 화장료 조성으로 사용 가능한 다양한 활성 물질이 인입이 가능하다. 상기 세포벽은 과량의 셀룰로오스를 포함하는데, 이로 인해 다른 물질의 침투가 용이하지 않아 활성 물질의 인입을 위해선 제거가 필요하고, 세포벽과 세포막을 모두 제거할 경우 인입된 활성 물질의 안정성이 오히려 저하될 수 있으므로, 세포막이 존재하는 프로토플라스트가 바람직하다.
활성 물질은 친수성 물질 또는 소수성 물질일 수 있으며, 본 발명에서 특별히 한정하지 않는다. 대표적으로, 활성 물질로는 유기산, 비타민, 알부틴, 아데노신, 나이아신아마이드, 폴리페놀, 플라보노이드, 레티놀, 사이클로헥산다이올 비스에틸헥사노에이트, 비스레틴아미도 메틸펜탄, 베타라파촌, 성장인자, 성장인자 복합 펩타이드, 및 이들의 조합으로 이루어진 군에서 선택된 1종이 가능하다.
이러한 활성 물질은 그 자체로서 화장료 조성물의 유효 성분으로 사용되나, 안정성이 낮아 그 활성을 충분히 발현할 수 없었으나 프로토플라스트 내에 인입되어 활성 안정성이 높아지고, 상기 프로토플라스트로 인해 생체 적합성 또한 크게 향상되는 이점이 있다.

식물 세포 수득은 본 발명에서 특별히 한정하지 않으며, 공지의 방법이 사용될 수 있다. 일례로, 본 발명의 실시예에서는 멸균 후 계면활성제가 첨가된 염소산나트륨 수용액에 담근 후 세척하여 식물 세포를 얻을 수 있었다.
사용 가능한 식물로는 본 발명에서 특별히 한정하지 않으며, 모든 식물이 가능하다. 대표적으로, 커리플랜트(curry plant, Helichrysum italicum), 구갈(Commiphora wightii, Commiphora myrrha), 부채 선인장(Opuntia Ficus indica), 작약(Paeonia lactiflora), 석화(Adenium obesum), 님파이아 오도라타(Nymphaea coerulea), 유칼리투스(Eucalyptus punctata), 은행나무(Ginkgo biloba), 나리꽃(Lilium candidum), 감람나무(Olea europaea), 파피루스(Cyperus papyrus), 연꽃(Nelumbo nucifera), 레드우드(Sequoia sempervirens), 가리카 로즈(Rosa gallica officinalis), 커피나무(Coffea arabica), 플루메리아(Plumeria obtusa), 치자나무(Gardenia jasminoides), 부겐베리아(Bougainvillea spectabilis), 자스민(Jasminum sambac), 로사 센티폴리아(Rosa centifolia), 페퍼민트(Mentha piperita), 로사 다마스케나(Rosa damascena), 붓꽃(Iris pallida), 포도나무(Vitis vinifera), 백장미(Rosa alba), 일랑일랑(Cananga odorata), 아몬드 나무(Prunus amygdalus dulcis), 사과나무(Malus domestica), 살구나무(Prunus armeniaca), 인삼(Panax ginseng), 블랙베리(Rubus fruticosus), 금영화(Eschscholtzia californica), 병풀(Centella asiatica), 신양벚나무(Prunus cerasus), 하와이 무궁화(Hibiscus rosa sinensis), 주니퍼베리(Juniperus communis), 목화(Gossypium arboreum), 대추야자(Phoenix dactylifera), 생강(Zingiber officinale), 무궁화(Hibiscus syriacus), 푸에라리아투베로사(Pueraria tuberosa), 석류나무(Punica granatum), 붐박스 코스타튬(Bombax costatum), 사프란(Crocus sativus), 살비아(Salvia officinalis), 수련(Nymphaea alba) 및 이들의 조합으로 이루어진 군에서 선택된 1종이 가능하며, 바람직하기로 커리플랜트(curry plant, Helichrysum italicum), 구갈(Commiphora wightii, Commiphora myrrha), 부채 선인장(Opuntia Ficus indica) 등을 사용한다.
단계 (b): 탈분화 단계
단계 (b)에서는 상기 수득된 식물 세포를 옥신을 이용하여 탈분화하여 탈분화된 식물 세포를 얻는다.
탈분화(dedifferentiation)에 사용하는 옥신(auxin)은 식물의 생장 조절 물질의 하나로, 저농도에서는 세포 신장을 촉진하나 고농도에서는 생장을 억제한다. 이에 옥신으로는 알파-나프탈렌 아세트산(α-Naphtalene acetic acid), 2,4-디클로로페녹시 아세트산, 인돌-3-아세트산(Indole-3-acetic acid), 및 이들의 조합으로 이루어진 군에서 선택된 1종이 가능하며, 이들을 1∼5㎎/ℓ의 농도로 포함하는 배지에 식물 세포를 배양하여 상기 식물 세포의 탈분화를 유도할 수 있다.
상기 배지는 본 발명에서 특별히 한정하지 않으며, 식물 세포 배양에 사용되는 모든 배지가 사용될 수 있다.
단계 (c): 탈분화 식물 세포의 배양 단계
단계 (c)에서는 상기 단계 (b)에서 얻어진 탈분화 식물 세포를 대량으로 배양한다.
탈분화 식물 세포의 대량 배양은 다양한 배지 내에서 이루어지며, 이때 배지로는 MS 배지, B5 배지, WHITE 배지, N6 배지, SH 배지, Anderson 배지 등 공지된 바의 배지가 사용될 수 있으며, 바람직하기로 MS 배지를 사용한다.
또한, 배양 조건 및 기간은 본 발명에서 특별히 언급하지 않으며, 공지된 바의 조건 하에서 수행할 수 있다.
단계 (d): 효소 반응을 통한 세포벽 제거 단계
단계 (d)에서는 대량으로 배양된 탈분화 식물 세포의 세포벽을 효소 반응을 이용하여 제거하여 세포막과 세포소 기관만 남은 프로토플라스트를 얻는다.
상기 효소는 셀룰라아제(EC 3.2.1.4), 펙티나아제(EC 3.2.1.15), 크실라나아제(EC 3.2.1.8), 키티나아제(EC 3.2.1.14), 헤미셀룰라아제 및 이들의 조합으로 이루어진 군에서 선택된 1종이 가능하다.
이들 효소는 0.01∼10 중량%의 다양한 농도로 사용될 수 있으며, 바람직하기로 셀룰라아제 0.1∼5 중량%, 펙티나아제 0.01∼2.5 중량%, 헤미셀룰라아제 0.1∼5 중량%로 사용하고, 더욱 바람직하기로 셀룰라아제 1 중량%, 헤미셀룰라아제 2 중량% 및 펙티나아제 0.5 중량%의 다성분 효소 혼합물을 사용한다.
상기 효소 반응은 4℃∼40℃의 온도 범위에서 (더 적절하게는 10℃∼25℃) 15시간∼24시간 동안 10∼50rpm의 속도로 반응시켜 이루어지며, 이러한 효소 반응에 의해 탈분화 식물 세포의 세포벽만이 제거되고 세포막은 안정하게 보존된 프로토플라스트를 얻을 수 있다.
단계 (e): 가압 삼투 공정에 의한 활성 물질 인입 단계
단계 (e)에서는 상기 얻어진 탈분화 식물 프로토플라스트 내에 가압 삼투 공정으로 활성 물질을 인입한다.
구체적으로, 탈분화 식물 프로토플라스트를 저급 알코올(C1~C4 알코올)에 용해시키고, 이를 활성 물질 및 물과 혼합한 후 염화 나트륨을 첨가하게 되면, 삼투화에 의해 상기 활성 물질이 프로토플라스트 내부로 인입된다.
이러한 공정은 가압 삼투 공정으로 진행되며, 구체적으로는 0.01∼ 10MPa, 바람직하게는 0.05∼1MPa, 가장 바람직하게는 0.05∼0.5MPa에서 수행하며, 이때 염화 나트륨의 첨가에 의해 농도를 1% 이상, 바람직하기로 1∼50%가 되도록 한다. 만약, 상기 압력 및 농도가 상기 범위 미만이면 활성 물질의 인입이 효과적으로 일어나지 않으며, 반대로 상기 범위를 초과하면 프로토플라스트가 파괴될 우려가 있으므로, 상기 범위 내에서 적절히 사용한다.
상기 인입은 활성 물질의 효능이 충분히 발휘될 수 있는 농도가 되도록 수행하며, 이는 활성 물질에 따라 달라질 수 있으나, 0.001∼20%의 인입률(중량% 기준)을 갖는다. 일례로, 실시예 1의 활성 물질인 사이클로헥산다이올 비스에틸헥사노에이트의 경우 0.001∼15 중량%, 바람직하기로 0.01∼10 중량%, 더욱 바람직하기로 0.1∼10 중량% 농도일 경우 우수한 활성을 갖는다.
또한, 실시예 2의 활성 물질인 비스레틴 아미도메틸펜탄의 경우 0.005∼5.0 중량%, 바람직하기로 0.01∼2 중량% 농도일 경우 우수한 활성을 가지며, 실시예 3의 활성 물질인 베타라파촌의 0.005∼10.0 중량%, 바람직하기로 0.01∼5.0 중량% 농도일 경우 우수한 활성을 갖는다. 그리고 실시예 4의 성장인자 복합 펩타이드의 경우 10∼5000ppm, 바람직하기로 50∼5000ppm, 더욱 바람직하기로 100∼500ppm 농도일 수 있다.
추가로, 상기 인입은 인입률을 높이기 위해 탈수 반응을 더욱 수행할 수 있다.
단계 (f): 후처리 단계
단계 (f)에서는 후처리를 통해 프로토플라스트 내에 활성 물질이 인입된 구조체를 후처리를 통해 얻는다.
이러한 후처리는 통상의 후처리 공정으로, 원심 분리 및 세척을 통해 미반응 물질(인입이 되지 않은 활성 물질) 및 염을 제거한다.
상기 단계를 거쳐 얻어진 프로토플라스트 내에 활성 물질이 인입된 구조체는 기존에 식물 세포가 갖고 있는 활성 성분의 안정성이 그대로 유지되고 추가적으로 세포 내 인입된 활성 물질의 안정성이 높으며, 이에 따라 상기 활성 물질에 의한 효능 또한 향상되어 화장료의 유효 성분 조성으로 바람직하게 사용 가능하다.
이러한 구조체는 전체 화장료 조성물 총 중량에 대하여 0.001 내지 99.0 중량%, 바람직하게는 0.01 내지 10.0 중량%, 더욱 바람직하게는 0.1 내지 3 중량%로 포함한다.
또한, 화장료 조성물은 공지된 바의 어떠한 제형으로 제조될 수 있으며, 화장수, 영양로션, 영양크림, 마사지크림, 에센스, 팩, 페이스트, 겔, 크림, 로션, 파우더, 비누, 오일, 파운데이션, 왁스 또는 스프레이형태로 제형화할 수 있다.
또한, 각 제형의 화장료 조성물에 있어서, 상기의 세포 이외에 다른 성분들을 기타 화장료의 제형 또는 사용 목적 등에 따라 임의로 선정하여 배합할 수 있다. 또한, 각 제형의 조성들은 그 제형의 제제화에 필요하고 적절한 각종의 기제와 첨가물을 함유할 수 있으며, 그 효과를 떨어트리지 않는 범위 내에서 비이온 계면활성제, 실리콘 폴리머, 체질안료, 향료, 방부제, 살균제, 산화 안정화제, 유기 용매, 이온성 또는 비이온성 증점제, 유연화제, 산화방지제, 자유 라디칼 파괴제, 불투명화제, 안정화제, 에몰리언트(emollient), 실리콘, α-히드록시산, 소포제, 보습제, 비타민, 곤충 기피제, 향료, 보존제, 계면활성제, 소염제, 물질 P 길항제, 충전제, 중합체, 추진제, 염기성화 또는 산성화제, 또는 착색제 등 공지의 화합물을 포함하여 제조된다.
본 발명의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로하이드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌 글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타하이드록사이드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 더욱 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
제조예 1: 탈분화 식물 원형질체 세포 제조
(1) 탈분화 식물 세포의 수득
커리플랜트(curry plant, Helichrysum italicum), 구갈(Commiphora wightii, Commiphora myrrha), 부채 선인장(Opuntia Ficus indica)의 줄기, 잎 등에서 조직 절편 (최소 3㎝)을 절단하였다. 본 조작 단계에서, 모든 작업은 무균 조건하에 무균 작업대에서 수행하였다.
식물 재료를 멸균하기 위하여, 조직은 70% 에탄올(Ethanol, Sigma, USA)에 60초, 30% 과산화수소(Hydrogen peroxide, LG Chemical, Korea)에 15분 담그고 용제를 제거하며, 이후 이들 조직은 무균 H2O로 3∼5회 씻어내고 몇 방울의 Tween 20이 첨가된 염소산나트륨(Sodium hydrochlorite, Sigma, USA)에 15분간 담그고 무균 H2O로 3∼5회 씻어냈다.
조직 배양을 위하여, 이들 조직 단편은 무균 페트리 접시(125㎜)에 집어넣고 절단하며 (2∼3㎜) 표백된 부분을 조심스럽게 제거하고, 이렇게 수득된 시료는 가늘게 절개하여 고형 배양 배지(하기 표 1)에 도말하고 반쯤 묻었다.

(2) 탈분화 식물세포의 재식(Replanting)
압설자(Spatula)로 2∼3개의 세포 클러스터(1∼2cm)를 채취한 후, 새로운 배지에 도말하고 분산시켰다. 이 모든 과정은 무균 조건하에 무균 작업대에서 수행하였다.
(3) 액상 배양 배지에서 탈분화 식물 세포의 증식
상기의 탈분화된 식물 세포를 하기 표 2의 액상 배양 배지로 이전한 다음, 25℃, 암조건 하에서 50∼150rpm의 회전 교반기에서 배양하였으며, 이때 이들의 계대 배양 주기는 매10일로 고정하였다.

(4) 탈분화 식물세포의 세포벽을 제거한 프로토플라스트 수득
상기 얻어진 탈분화 식물세포가 증식된 액상 배양 배지에 다성분 효소 혼합물(셀룰라아제 1%, 헤미셀룰라아제 2% 및 펙티나아제 0.5%)을 첨가하고 25℃의 온도 범위에서 20시간 동안 50rpm의 속도로 반응시켜 세포벽을 제거하였다.
이어, 배양액 중의 원형질체를 200xg 하에서 15분간 원심분리하여 수득한 후, 다시 5,000xg 하에서 15분간 원심분리하여 추가로 정제하였다.
실시예 1: 탈분화 식물 프로토플라스트 내에 사이클로헥산다이올 비스에틸헥사노에이트가 인입된 구조체의 제조
사이클로헥산다이올 비스에틸헥사노에이트(Cyclohexandiol Bis-Ethylhexanoate, RNS, Korea)를 에탄올에 30분간 교반하여 10 중량% 농도의 용액을 제조하였다.
얻어진 사이클로헥산다이올 비스에틸헥사노에이트 용액 20 중량%에, 상기 제조예 1에서 제조한 프로토플라스트 20 중량%, 및 증류수 60 중량%에 넣고, 패들믹서(PL-S10, Poonglim, Korea)를 이용하여 500rpm, 25℃ 조건으로 1시간 동안 교반하였다.
이어, 활성물질 용액, 프로토플라스트 및 증류수의 혼합액에 염화나트륨(NaCl, Sigma, USA)을 2 중량% 넣고 고압반응기(Miniclave, Buchi AG, Switzerland)를 이용하여 0.5MPa, 25℃ 1시간 조건으로 역삼투, 탈수 및 사이클로헥산다이올 비스에틸헥사노에이트의 인입 반응을 유도했다.
그 후, 원심분리기(Supra 22K, Hanil, Korea)를 이용하여 5,000xg 하에서 20분간 원심분리하여 프로토플라스트 내에 사이클로헥산다이올 비스에틸헥사노에이트가 인입된 구조체를 수득하였다.
이렇게 얻어진 구조체는 보존을 위해 구조체 20 중량%에 글리세린 80 중량%를 가하여 세포 조성물을 얻었다.
실시예 2: 탈분화 식물 프로토플라스트 내에 비스레틴아미도 메틸펜탄이 인입된 구조체의 제조
상기 실시예 1과 동일하게 수행하되, 활성 물질로 사이클로헥산다이올 비스에틸헥사노에이트 대신 비스레틴아미도 메틸펜탄(Bis-retinamido methylpentane, DermaLab, Korea)를 사용하여, 프로토플라스트 내에 비스레틴아미도 메틸펜탄이 인입된 구조체를 얻었다(구조체/글리세린, 20/80 중량%).
실시예 3: 탈분화 식물 프로토플라스트 내에 베타라파촌이 인입된 구조체의 제조
상기 실시예 1과 동일하게 수행하되, 활성 물질로 사이클로헥산다이올 비스에틸헥사노에이트 대신 베타라파촌(β-Lapachone, Sigma, USA)을 사용하여, 프로토플라스트 내에 베타라파촌이 인입된 구조체를 얻었다(구조체/글리세린, 20/80 중량%).
실시예 4: 탈분화 식물 프로토플라스트 내에 성장인자 복합 펩타이드(growth factor mimic peptides)가 인입된 구조체의 제조
상기 실시예 1과 동일하게 수행하되, 활성 물질로 사이클로헥산다이올 비스에틸헥사노에이트 대신 하기 조성의 성장인자 복합 펩타이드 조성을 사용하여, 프로토플라스트 내에 성장인자 복합 펩타이드가 인입된 구조체를 얻었다(구조체/글리세린, 20/80 중량%).
성장인자 복합 펩타이드 조성은 올리고펩타이드-34(Oligopeptide-34, Caregen, Korea), 올리고펩타이드-24(Oligopeptide-24, Caregen, Korea), 데카펩타이드-4(Decapeptide-4, Caregen, Korea), 아세틸 데카펩타이드-3(Acetyl Decapeptide-3, Caregen, Korea) 및 rh-폴리펩타이드-4(rh-Polypeptide-4, Bio-FD&C, Korea)를 3차 증류수에 각각 4,000 ppm(총 20,000ppm)으로 가하여 30분간 교반한 것을 사용하였다.
실험예 1: 활성 물질이 인입된 구조체의 성분 분석
본 실험에서는 상기 실시예 1 내지 4에서 제조한 구조체에서 프로토플라스트 내에 활성 물질이 잘 인입되었는지를 GC(gas chromatography, 가스크로마토그래피)를 통해 확인하였으며, 이때 비교를 위해 각 활성 물질의 표준 시약을 사용하였다.
도 2의 (a)는 표준시료인 사이클로헥산다이올 비스에틸헥사노에이트, (b)는 실시예 1의 프로토플라스트 내에 사이클로헥산다이올 비스에틸헥사노에이트가 인입된 구조체의 GC 스펙트럼으로, 세포 내에 활성 물질인 사이클로헥산다이올 비스에틸헥사노에이트가 인입되어 있음을 알 수 있다.
이러한 결과는 실시예 2 및 3에서도 동일하게 나타남을 확인하였다.
도 3의 (a)는 표준시료인 비스레틴아미도 메틸펜탄, (b)는 실시예 2의 프로포플라스트 내에 비스레틴아미도 메틸펜탄이 인입된 구조체의 GC 스펙트럼으로, 세포 내에 활성 물질인 비스레틴아미도 메틸펜탄이 인입되어 있음을 알 수 있다.
또한, 도 4의 (a)는 표준시료인 베타라파촌, (b)는 실시예 3의 프로포플라스트 내에 베타라파촌이 인입된 구조체의 GC 스펙트럼으로, 세포 내에 활성 물질인 베타라파촌이 인입되어 있음을 알 수 있다.
실시예 4의 성장인자 복합 펩타이드가 인입된 세포는 MALDI-TOF 질량분석기(Matrix assisted laser desorption/ionization time-of-flight Mass Spectrometry)를 통해 이루어졌다.
도 5의 (a)는 표준시료인 성장인자 복합 펩타이드, (b)는 실시예 4의 프로포플라스트 내에 성장인자 복합 펩타이드가 인입된 구조체의 MALDI-TOF 질량분석 그래프로, 세포 내에 활성 물질인 성장인자 복합 펩타이드가 인입되어 있음을 알 수 있다.
효능 분석
상기 실시예 1 내지 4에서 얻어진 활성 물질이 포접된 세포의 화장료로서의 효과 및 효능을 확인하기 위해 하기와 같이 수행하였다.
실험예 2: 실시예 1의 구조체의 미백 효과 분석
사이클로헥산다이올 비스에틸헥사노에이트는 미백 효과가 있으며, 실시예 1에서 제조한 구조체와의 미백 효과를 비교하기 위해 멜라닌 생성 세포를 이용하여 실험하였다.
본 실험은 멜라닌생성세포(melanoma) 세포주와 멜라닌 자극호르몬을 이용한 in vitro assay계를 확립하여 사이클로헥산다이올 비스에틸헥사노에이트에 의한 멜라닌 생성 억제 효과, 티로시나제 활성 억제 효과를 측정함으로써 피부 미백 효과를 확인하였다.
(1) 세포주 및 세포배양
설치류 멜라노마 세포주인 B16 세포주(ATCC CRL-6323)를 37℃, 5% CO2의 조건에서 10% FBS (Gibco Co), 100 ㎍/㎖ 스트렙토마이신(streptomycin), 100 U/㎖ 페니실린(penicillin)이 첨가된 DMEM(Dulbecco's modified Eagle medium)을 사용하여 배양하였다(Incubator; Thermo-scientific, USA).
B16 세포는 100cm2 플라스크(Corning, USA)에서 충분히 증식시킨 후 배양 3일 간격으로 배양세포 표면을 PBS로 씻어준 후 0.25% 트립신-EDTA 용액을 넣고 배양기에서 3분간 처리한 다음 트립신-EDTA 용액을 버리고 37℃에서 5분간 보관하여 세포를 탈착시켰다. 탈착된 세포는 10% FBS가 포함된 DMEM 10㎖을 새로운 배양용기에 옮겨 1:5의 분할율 (split ratio)로 37℃, 5% CO2의 조건에서 계대 배양하였다.
(2) 멜라닌 생성억제 효과 측정
96-웰 플레이트에, 웰 당 1×105 개를 배지 DMEM을 이용하여 접종하고, 실시예 1에서 제조한 구조체와 미백효과가 알려진 사이클로헥산다이올비스에틸헥사노에이트, 알파-비사보롤을 각 농도별로 만들어 시료액 2ml을 처리하였다. 이후 α-Melanocyte stimulating hormone (α-MSH) 1ppm을 처리하고 72시간 동안 배양하였다. 다음으로, 인산완충식염수(phosphate buffer saline, PBS)로 배지를 씻어준 후 멜라닌 추출용액(1N NaOH+50 % DMSO)을 웰당 100㎕씩 첨가하여 80℃에서 세포를 용해시킨 후 ELISA 리더를 이용하여 492㎚에서 멜라닌의 흡광도를 측정하였다.

(3) 결과 분석
도 6은 실시예 1의 구조체, 사이클로헥산다이올 비스에틸헥사노에이트 및 알파-비사보롤의 각 농도에 따른 멜라닌의 생성 억제 효과를 보여주는 그래프이다.
도 6을 참조하면, 본 발명에 따른 실시예 1에서 제조한 프로토플라스트 내에 사이클로헥산다이올 비스에틸헥사노에이트이 인입된 구조체의 경우 멜라민 억제 효과가 사이클로헥산다이올 비스에틸헥사노에이트 및 알파-비사보롤 보다 우수하였으며, 이는 그 농도가 높을수록 멜라닌 생성 억제 효과 또한 크게 증가하였다.
실험예 3: 실시예 2의 구조체의 콜라겐 생합성 효과 분석
비스레틴아미도 메틸펜탄은 피부 재생 효과가 있으며, 실시예 2에서 제조한 구조체와의 콜라겐 생합성 효과를 비교하였으며, 이때 비교예로서 레티놀 및 아데노신을 추가로 사용하였다.
(1) 세포주 및 세포배양
인간 피부 섬유 아세포 (Human dermal fibroblast, Gibco, USA) 를 96-웰 마이크로 플레이트(96-wellmicroplate)의 각 웰(well)에 1×104세포가 되도록 접종하여 100 U/㎖ 페니실린, 100 ㎍/㎖ 스트렙토마이신이 보충된 10% DMEM(Dulbecco's Modified Eagle's Medium) 배지에서 배양하였다. 상기 배지는 부착된 세포들이 80% 합류 (confluence) 때까지 유지하고 80% 합류시기에 계대 배양을 수행하였다. 합류 때까지 배지는 2일 마다 교체하였다. 상기 계대 배양은 배양액을 제거한 플라스크를 인산완충식염수(phosphate buffer saline, PBS)로 세척하고 0.25% 트립신-EDTA (Gibco BRL, Grand Island, NY, USA)로 세포를 떨어뜨린 후 세포 부유액을 원심 분리한 후, 세포 수를 측정하고, 배양액에서 다시 3 배로 계대 배양하였다.
(2) 콜라겐 생합성 효과
사람의 정상 섬유아세포(human normal fibroblast)를 10% FBS가 함유된 DMEM 배지가 들어 있는 48-웰 마이크로 플레이트에 1x105 cells/well의 농도로 접종시키고, 5% 농도(CO2)의 CO2 배양기에서 37℃, 24시간 배양하였다.
배양 후 실시예 2의 구조체, 비스레틴아미도 메틸펜탄, 레티놀 및 아데노신을 각 농도별로 조정한 혈청이 없는 DMEM배지로 교체한 후 48시간 더 배양 하였다. 배양 마지막 24시간 전에 아스코르빅산 50㎍/㎖을 첨가하여 콜라겐 합성을 촉진시킨다. 배양 후 각 웰을 세척하고 혈청이 함유되지 않은 DMEM 배지로 교체한 후 24시간 더 배양하였다.
배양 후 각 웰의 상층액을 모아 프로콜라겐 타입 아이씨-펩타이드(PICP, Type-I C-peptide)양을 키트(Kit, Takara Shuzo Co., Ltd, Japan)를 이용하여 새로 합성된 콜라겐 양을 PICP 양으로 측정하였으며, PICP 양은 ng/2×104 세포로 환산하여 콜라겐의 합성효과를 측정하였다
(3) 결과 분석
도 7은 실시예 2의 구조체, 비스레틴아미도 메틸펜탄, 레티놀 및 아데노신의 콜라겐 합성능을 보여주는 그래프로, 본 발명에 따른 실시예 2의 활성 물질 인입 세포의 콜라겐 합성능이 매우 우수하며, 농도가 높아질수록 상기 합성능이 더욱 향상됨을 보여준다.
실험예 4: 실시예 3의 구조체의 콜라겐 생합성 효과 분석
상기 실험예 3과 동일하게 수행하되, 배양 후 실시예 3에서 제조한 구조체, 베타라파촌, 레티놀 및 아데노신을 각 농도별로 조정한 혈청이 없는 디엠이엠 배지로 교체한 후 배양하여 콜라겐 합성 효과를 분석하였다.
도 8은 실시예 3의 구조체, 베타라파촌, 레티놀 및 아데노신의 콜라겐 합성능을 보여주는 그래프로, 본 발명에 따른 실시예 3의 활성 물질 인입 세포의 콜라겐 합성능이 매우 우수하며, 농도가 높아질수록 상기 합성능이 더욱 향상됨을 보여준다.
실험예 5: 실시예 4의 구조체의 효과 분석
(1) in-vitro 실험
상기 실험예 4과 동일하게 수행하되, 배양 후 실시예 4에서 제조한 구조체, 성장인자 복합 펩타이드, 레티놀 및 아데노신을 각 농도별로 조정한 혈청이 없는 디엠이엠 배지로 교체한 후 배양하여 콜라겐 합성 효과를 분석하였다.
도 9는 실시예 4의 구조체, 성장인자 복합 펩타이드, 레티놀 및 아데노신의 콜라겐 합성능을 보여주는 그래프로, 본 발명에 따른 실시예 3의 활성 물질 인입 세포의 콜라겐 합성능이 매우 우수하며, 농도가 높아질수록 상기 합성능이 더욱 향상됨을 보여준다.
(2) ex-vivo 실험
실시예 4에서 제조한 구조체의 사람 피부 조직에서의 표피 재생 효과와 글리코사미노글리칸의 증가 효과 및 콜라겐 증가 효과를 확인하기 위해, 하기와 같이 수행하였으며, 그 결과를 하기에 나타내었다.
1. 40세 전 후의 여성의 피부로부터 15개의 조직을 적출. 살아있는 상태로 배양한 피부조직 준비.
2. 이 피부 조직들은 다음과 같이 3 set 1조의 조건으로 실험.

3. D0 에서 피부조직에 샘플을 0.1g 도포 (시료처리는 D2, D5, D6 그리고 D8에도 동일하게 수행됨) - 시료의 농도: 0.2%
4. D0, 3개의 control batch 피부조직을 샘플링하고, D6과 D9, 각각 3개의 피부조직을 샘플링.
5. 파라핀에 고정. 5μm 두께로 섹션
6. 표피의 재생 정도를 확인: 파라핀 고정 조직을 마손 삼색 염색법(Masson s trichrome - Goldner variant, Masson-Goldner trichrome staining kit, MERCK, USA)으로 염색 후 CellD Olympus software(Olympus, Japan)를 이용하여 영상 분석.
7. 글리코사미노글리칸 증가 확인: 파라핀 고정 조직을 알시안 블루-PAS염색법(Alcian blue P.A.S., Alcian Blue/PAS Kit, Polyscience, Inc.,USA)으로 염색 후 글리코사미노글리칸의 증가량을 LEICA Q Win을 이용하여 확인하고 Kodak in vivo image analyzer (Eastern Kodak, Rochester, USA)를 이용하여 분석.
8. 콜라겐의 증가 확인: 냉동시킨 파라핀 고정 조직을 FITC가 라벨링 되어 있는 제1형 콜라겐의 안티바디인 래빗 폴리클로날 안티바디(Poab Rabbit anti Collagen I affinity purified Cat#PS047, Monosan, Netherlands)로 1시간 동안 상온에서 염색하고, 프로피디움 아이오다이드(Propidium Iodid, Sigma-Aldrich, USA)로 핵을 염색 후 콜라겐의 증가량을 LEICA Q Win을 이용하여 확인하고 Kodak in vivo image analyzer (Eastern Kodak, Rochester, USA)를 이용하여 분석.
도 10은 사람 피부조직에 대해 실시예 4의 구조체의 처치 전 및 후의 표피 재생을 보여주는 이미지로, 재생 효과가 매우 우수함을 알 수 있다.
이러한 결과는 글리코사미노글리칸의 증가 효과 및 콜라겐 증가 실험에서도 동일하게 나타났다.
도 11은 사람 피부조직에 대해 실시예 4의 구조체의 처치 전 및 후의 글리코사미노글리칸 증가 정도를 보여주는 이미지이고, 도 12는 타입 I 콜라겐의 증가 정도를 보여주는 이미지로서, 이 둘을 보면 본 발명에 따라 프로토플라스트 내에 성장인자 복합 펩타이드가 인입된 경우 상기 효과가 우수함을 알 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 제형예를 제시한다. 그러나 하기의 제형예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 제형예에 의해 본 발명의 내용이 한정되는 것은 아니다.
제형예 1: 영양 크림의 제조
하기의 표 4와 같은 조성을 갖도록 공지 방법을 이용하여 영양 크림을 제조하였다.

Claims (13)

  1. 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체.

  3. 제1항에 있어서, 상기 활성 물질은 친수성 물질 및 소수성 물질로 이루어진 군에서 선택된 1종인 것을 특징으로 하는 구조체.
  4. 제1항에 있어서, 상기 활성 물질은 유기산, 비타민, 알부틴, 아데노신, 나이아신아마이드, 폴리페놀, 플라보노이드, 레티놀, 사이클로헥산다이올 비스에틸헥사노에이트, 비스레틴아미도 메틸펜탄, 베타라파촌, 성장인자, 성장인자 복합 펩타이드, 및 이들의 조합으로 이루어진 군에서 선택된 1종인 것을 특징으로 하는 구조체.
  5. 제1항에 있어서, 상기 활성 물질의 인입률은 0.001∼20 중량%인 것을 특징으로 하는 구조체.
  6. 식물의 잎, 줄기, 뿌리, 꽃, 열매 및 씨앗에서 식물의 세포를 수득하는 단계;
    상기 수득된 식물 세포를 옥신을 이용하여 탈분화하여 탈분화된 식물 세포를 얻는 단계;
    상기 얻어진 탈분화 식물 세포를 대량으로 배양하는 단계;
    상기 대량 배양된 탈분화 식물 세포의 세포벽을 효소 반응을 이용하여 제거하여 프로토플라스트를 얻는 단계;
    상기 얻어진 탈분화 식물 프로토플라스트 내에 가압 삼투 공정으로 활성 물질을 인입하는 단계; 및
    후처리하는 단계를 포함하는
    탈분화 식물 프로토플라스트 내에 활성 물질이 인입된 구조체의 제조방법.
  7. 제6항에 있어서, 상기 옥신은 알파-나프탈렌 아세트산(α-Naphtalene acetic acid), 2,4-디클로로페녹시 아세트산, 인돌-3-아세트산(Indole-3-acetic acid), 및 이들의 조합으로 이루어진 군에서 선택된 1종으로 1∼5㎎/ℓ의 농도로 사용하는 것을 특징으로 하는 구조체의 제조방법.
  8. 제6항에 있어서, 상기 효소 반응은 셀룰라아제(EC 3.2.1.4), 펙티나아제(EC 3.2.1.15), 크실라나아제(EC 3.2.1.8), 키티나아제(EC 3.2.1.14), 헤미셀룰라아제 및 이들의 조합으로 이루어진 군에서 선택된 1종의 효소와 혼합하여 4℃∼40℃의 온도에서 15시간∼24시간 동안 반응시키는 것을 특징으로 하는 구조체의 제조방법.
  9. 제6항에 있어서, 상기 가압 삼투 공정은 0.01∼10MPa의 압력 하에 염화 나트륨을 첨가하여 1∼50% 농도가 되는 조건인 것을 특징으로 하는 구조체의 제조방법.
  10. 제6항에 있어서, 상기 활성 물질 인입 후 후처리 단계 이전에 탈수 반응을 더욱 수행하는 것을 특징으로 하는 구조체의 제조방법.
  11. 제6항에 있어서, 상기 후처리는 원심 분리 및 세척인 것을 특징으로 하는 구조체의 제조방법.
  12. 제1항의 탈분화 식물 프로토플라스트(protoplast) 내에 활성 물질이 인입된 구조체를 유효 성분으로 포함하는 것을 특징으로 하는 화장료 조성물.
  13. 제12항에 있어서, 상기 화장료 조성물은 화장수, 영양로션, 영양크림, 마사지크림, 에센스, 팩, 페이스트, 겔, 크림, 로션, 파우더, 비누, 오일, 파운데이션, 왁스 또는 스프레이의 제형인 것을 특징으로 하는 화장료 조성물.
