たとえば、AC220(Ambit社)は、III型チロシンキナーゼ(FLT3、c-KIT、FMS、PDGFR)を選択的に阻害する化合物であり、AMLを対象にした開発が行われている(特許文献1)。
一方、生体たんぱく質に共有結合することにより、活性や持続性に優れた薬剤が開発・上市されている。たとえば、分子内にアクリル基を有するEGFR阻害剤としてアファチニブ(BIBW2992)が報告されており(特許文献2)、米国で上市されている。

化合物Aを溶媒1に懸濁し、酸を添加して加熱溶解させた後、冷却し、次いで、溶媒2を加えることによって、化合物Aの塩を製造することができる。

他の好ましい例は、粉末X線回折において、回折角度(2θ)12.8、16.1、21.4および28.0°に回折ピークを有する、化合物Aのコハク酸塩の結晶(以下、β型結晶ともいう。)である。
他の好ましい例は、粉末X線回折において、回折角度(2θ)8.6、13.7、17.8および23.0°に回折ピークを有する、化合物Aのフマル酸塩の結晶である。
本発明の化合物Aの塩の結晶の好ましい例は、赤外吸収スペクトル(ATR法)において、波数2937、2218、1441、1304および1242cm-1に吸収ピークを有する、化合物Aのコハク酸塩のα型結晶である。
他の好ましい例は、赤外吸収スペクトル(ATR法)において、波数2219、1660、1512、1239および1121cm-1に吸収ピークを有する、化合物Aのコハク酸塩のβ型結晶である。
他の好ましい例は、赤外吸収スペクトル(ATR法)において、波数2220、1594、1517、1428および1080cm-1に吸収ピークを有する、化合物Aのフマル酸塩の結晶である。
本発明の化合物Aの塩またはその結晶を含有する医薬組成物は、本発明の種々の化合物Aの塩またはそれらの結晶のうち、一種のみを含有してもよく、または二種以上を含有してもよい。

化合物A3.50gのアセトン70mL懸濁液に、室温でコハク酸779mgを加え、加熱還流下、目視にて溶解を確認した。反応混合物を室温まで徐々に冷却し、1日間静置した。固形物を濾取し、アセトンで洗浄後、減圧下で乾燥させ、白色固体4.08gを得た。

化合物A5.50gのアセトン110mL懸濁液に、室温でコハク酸1.22gを加え、加熱還流下、目視にて溶解を確認した。反応混合物を室温まで徐々に冷却し、1日間静置した。固形物を濾取し、アセトンで洗浄後、減圧下で乾燥させ、淡黄色固体6.22gを得た。
得られた淡黄色固体150mgの1,4-ジオキサン3.0mL懸濁液を加熱還流し、目視にて溶解を確認した。この溶解液を室温まで徐々に冷却し、12日間静置した。固形物を濾取し、1,4-ジオキサンで洗浄後、減圧下で乾燥させ、化合物Aのコハク酸塩のβ型結晶141mgを得た。

化合物A1.50gのエタノール30mL懸濁液に、室温でフマル酸328mgを加え、70℃で加熱攪拌し、目視にて溶解を確認した。反応混合物を室温まで徐々に冷却し、3日間静置した。固形物を濾取し、エタノールで洗浄後、減圧下で乾燥させ、白色固体1.67gを得た。
得られた白色固体1.67gのエタノール30mL懸濁液に、化合物A0.53gを加え、80℃で加熱攪拌し、目視にて溶解を確認した。この溶解液を室温まで徐々に冷却し、6時間静置した。固形物を濾取し、エタノールで洗浄後、減圧下で乾燥させ、白色固体の化合物Aのフマル酸塩1.96gを得た。
水分含量:1.0%(重量比)

パモ酸73mgの水懸濁液に、室温で3.0mol/L水酸化ナトリウム水溶液126μLを加えた(溶解液1)。化合物A100mgのアセトン10mL懸濁液を、60℃で加熱攪拌し、目視にて溶解を確認した(溶解液2)。溶解液1に溶解液2を室温で加えた後、酢酸22μL、アセトンおよび水を加え、30分間攪拌した。固形物を濾取し、水で洗浄後、減圧下で乾燥させ、淡黄色固体の化合物Aのパモ酸塩132mgを得た。

化合物A150mgのアセトン4.5mL懸濁液を、加熱還流下、目視にて溶解を確認した。この溶解液を40℃まで徐々に冷却した後、4.0mol/L塩酸/1,4-ジオキサン溶液141μLを加え、室温で5日間静置した。固形物を濾取し、アセトンで洗浄後、減圧下で乾燥させ、白色固体の化合物Aの塩酸塩112mgを得た。

化合物A1.00gのエタノール20mL懸濁液を、70℃で加熱攪拌し、目視にて溶解を確認した。この溶解液にリン酸238μLを加え、室温まで徐々に冷却し、3時間30分静置した。固形物を濾取し、エタノールで2回洗浄後、減圧下で乾燥させ、淡黄色固体の化合物Aのリン酸塩0.75gを得た。

化合物A1.00gのエタノール20mL懸濁液を、70℃で加熱攪拌し、目視にて溶解を確認した。この溶解液に硫酸211μLを加え、室温まで徐々に冷却し、3時間静置した。固形物を濾取し、エタノールで2回洗浄後、減圧下で乾燥させ、白色固体の化合物Aの硫酸塩1.10gを得た。

化合物A200mgの水5mL懸濁液に、室温でベンゼンスルホン酸1水和物132mgを加え、50℃で加熱攪拌し、目視にて溶解を確認した。反応混合物を室温まで冷却した後、減圧下で溶媒を留去し、オイル状の化合物Aのベンゼンスルホン酸塩を得た。
化合物A150mgのアセトン4.5mL懸濁液を、加熱還流し、目視にて溶解を確認した。この溶解液に臭化水素酸64μLを加え、室温まで徐々に冷却した。固形物を濾取し、アセトンで洗浄後、減圧下で乾燥させ、淡黄色固体の化合物Aの臭化水素酸塩98mgを得た。
試験例1 FLT3酵素阻害試験
FLT3酵素阻害試験には、バキュロウィルス発現システムを用いて産生されたグルタチオンS-トランスフェラーゼ(GST)融合ヒトFLT3蛋白質(細胞内領域564‐993aa)(Carna Biosciences社)を用いた。
FLT3蛋白質と所定の濃度の試験化合物を含む9μL反応液(1.2μg FLT3、100mM HEPES、10mM MgCl2、25mM NaCl、0.01% BSA、1mM DTT、pH7.5)を15分間25℃で静置した。その後、基質ペプチドBiotin‐AAA‐AEEEEYFELVAKKK(東レ社)3μL(終濃度0.25μM)、ATP(Sigma‐Aldrich社)3μL(終濃度50μM)をそれぞれ添加し、2分間振盪後、さらに30分間25℃で静置して酵素反応を行った。
その後、Streptavidin‐Xlent(Cisbio社)とMab PT66‐K(Cisbio社)を含む酵素反応停止液(5μg/mL Streptavidin、0.19μg/mL PT66‐K、30mM HEPES(pH7.0)、150mM KF、75mM EDTA、0.15% BSA、0.075% Tween20)30μLを添加し、酵素反応を停止させると同時に、室温で1時間静置することにより抗原抗体反応を行った。その後、Envision(PerkinElmer社)を用いて615nm、665nmの時間分解蛍光を測定し、基質ペプチドのリン酸化を測定した。
結果を表7に示す。
白血病細胞株MV4-11(ATCC Number:CRL-9591)およびMOLM-13(DSMZ Number:ACC554)を用いて、白血病細胞増殖阻害試験を行った。
白血病細胞増殖阻害試験は、以下に記載の方法に従って行った。
化合物による増殖阻害を測定する目的で、ルシフェリン-ルシフェラーゼ反応を利用してATP濃度が定量できるCellTitet-Glo(Promega社)試薬を用い、全細胞ATP濃度を基にして全細胞数を定量化した。MOLM-13またはMV4-11細胞をペニシリン(100units/mL)/ストレプトマイシン(100μg/mL)、FBSを10%入れておいたRPMI培地に入れ、2×105個/mLとなるように調整し、96ウエルプレート(Corning社)にウエル1個当たり50μLずつ(10,000個)細胞を播種した。
前記細胞に化合物の段階希釈液または0.1%DMSO(溶媒対照)50μLを加えた後、前記細胞を標準的細胞増殖条件(37℃,5% CO2)下で72時間培養増殖させた。全細胞増殖を測定する目的で、CellTitet-Gloの使用説明書に従い、各ウエルに等しい体積のCellTitet-Glo反応液を加えた後、発光カウント数(相対的光単位,RLU)を定量した。
増殖阻害に関するGI50値は、培養72時間後のDMSO溶媒対照が示したRLUシグナルを0%阻害として定義し、そのDMSO溶媒対照における全細胞増殖の50%阻害をもたらす化合物液濃度に相当する。各データ点は、二重複サンプルにより得られた。GI50値は、XLfitソフトウエアを用い、シグモイド用量反応式による非線形回帰適合(Fit Model(205))によって算出された。
結果を表7に示す。
試験化合物として、実施例1および3の化合物を選択した。
比較化合物として、化合物Aを選択した。
水に試験化合物または比較化合物を添加した後、室温にて24時間撹拌した。メンブランフィルター(0.2μm)を用いて不溶物を濾去した。濾液をHPLCで分析し、溶解度を求めた。
結果を表8に示す。
試験物質として、実施例1および3の結晶を選択した。
試験物質200mgを開放状態のガラス瓶に入れ、保存条件1(25℃、相対湿度75%)または保存条件2(40℃、相対湿度75%)の条件で2週間保存した。試験開始前および試験終了後に試験物質の純度および水分含量を測定した。
試験開始前および試験終了後の試験物質の純度および水分含量を表9に示す。
試験物質として、実施例1の結晶を選択した。
試験物質200mgを2重のポリ袋に入れて開口部を縛り、保存条件1(25℃、相対湿度75%)または保存条件2(40℃、相対湿度75%)の条件で4週間保存した。試験開始前および試験終了後に試験物質の純度および水分含量を測定した。
試験開始前および試験終了後の試験物質の純度および水分含量を表10に示す。

- (S,E)-N-(1-((5-(2-((4-シアノフェニル)アミノ)-4-(プロピルアミノ)ピリミジン-5-イル)ペント-4-イン-1-イル)アミノ)-1-オキソプロパン-2-イル)-4-(ジメチルアミノ)-N-メチルブト-2-エンアミドのカルボン酸塩、鉱酸塩またはスルホン酸塩。
- カルボン酸塩である、請求項1に記載の塩。
- 鉱酸塩である、請求項1に記載の塩。
- カルボン酸塩が、ギ酸塩、酢酸塩、乳酸塩、安息香酸塩、クエン酸塩、シュウ酸塩、フマル酸塩、マレイン酸塩、コハク酸塩、リンゴ酸塩、酒石酸塩、アスパラギン酸塩、トリクロロ酢酸塩、トリフルオロ酢酸塩またはパモ酸塩である、請求項2に記載の塩。
- カルボン酸塩が、フマル酸塩、コハク酸塩またはパモ酸塩である、請求項2に記載の塩。
- 鉱酸塩が、塩酸塩、臭化水素酸塩、ヨウ化水素酸塩、硝酸塩、リン酸塩または硫酸塩である、請求項3に記載の塩。
