Abstract

しかし、ウイルス特有の2本鎖RNAとは異なり、一本鎖RNAやDNAは宿主由来の核酸と大きな違いはなく、TLRによるリガンド認識機構が厳密に制御されなければ、自己に対する反応を惹起し、自己免疫疾患に陥ってしまう。
これに対し、抗微生物ペプチドや自己抗体との相互作用によって自己の核酸が分解に耐性を有するようになり、エンドリソソームに到達できるようになると、TLR7依存性自己免疫反応が引き起こされる。実際、TLR7については、乾癬や全身性エリテマトーデス(systemic lupus erythematosus;SLE)との関連が示唆されている(非特許文献2~4)。
安全性及び特異性の面では抗体医薬が望ましいが、上述のとおり、TLR7は、自己免疫反応を制限するためにエンドリソソームに局在し、細胞表面からは隔絶されていると考えられてきたため、細胞表面にのみ作用する抗体は使用できないと考えられており、抗体医薬を使用する試みはこれまでなされなかった。
したがって、TLR9も、乾癬やSLEを含む自己免疫疾患の治療標的と考えられてきたが、同様に細胞表面には発現していないと考えられており、抗体医薬を使用する試みはこれまでなされなかった。
そして、抗TLR7抗体が、脾臓由来B細胞、骨髄由来マクロファージ(BM-MCs)、骨髄由来古典的樹状細胞(BM-cDCs)、及び骨髄由来形質細胞様樹状細胞(BM-pDCs)において、TLR7応答を阻害することを確認した。さらに、マウスの炎症性疾患モデルに対して抗TLR7抗体を投与すると、病態が有意に改善されること、抗TLR7抗体がin vivoでTLR7リガンドの投与によって産生される炎症性サイトカインを抑制すること、マウス乾癬モデルにおいても抗TLR7抗体が炎症を抑制することを実証し、これまでのその存在を認識されていなかった細胞表面TLR7及びTLR9が炎症性疾患の標的として有用であることを確認して、本発明を完成するに至った。
即ち、本発明は、
〔1〕抗TLR7抗体又は抗TLR9抗体を含む、炎症性疾患の治療又は予防剤;
〔2〕前記抗TLR7抗体が、以下の少なくとも1つを含む、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:3で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:4で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:5で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:6で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:7で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:8で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:3~8のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:3~8のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
〔3〕前記TLR7抗体が、以下のいずれかである、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:9で表されるアミノ酸配列を含む重鎖と、配列番号:10で表されるアミノ酸配列を含む軽鎖とを含む抗体;
(2) 配列番号:9及び/又は10で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:9及び/又は10で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
〔4〕前記抗TLR7抗体が、配列番号:1で示されるTLR7のアミノ酸配列のうち、27位~457位の領域内に特異的に結合する、上記〔1〕に記載の炎症性疾患の治療剤又は予防剤;
〔5〕前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の228位~364位の領域内に特異的に結合する、上記〔1〕に記載の炎症性疾患の治療剤又は予防剤;
〔6〕前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の275位~313位の領域を含む領域に特異的に結合する、上記〔1〕に記載の炎症性疾患の治療剤又は予防剤;
〔7〕前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の313位~364位の領域を含む領域に特異的に結合する、上記〔1〕に記載の炎症性疾患の治療剤又は予防剤。
〔8〕前記抗TLR9抗体が、以下の少なくとも1つを含む、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:11で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:12で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:13で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:14で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:15で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:16で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:11~16のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び

〔9〕前記抗TLR9抗体が、以下の少なくとも1つを含む、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:17で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:18で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:19で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:20で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:21で表されるアミノ酸配列からなる軽鎖CDR2;

(2) 配列番号:29及び/又は30で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:29及び/又は30で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
〔12〕前記TLR9抗体が、以下のいずれかである、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:31で表されるアミノ酸配列を含む重鎖と、配列番号:32で表されるアミノ酸配列を含む軽鎖とを含む抗体;
(2) 配列番号:31及び/又は32で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:31及び/又は32で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
〔13〕前記TLR9抗体が、以下のいずれかである、上記〔1〕に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:33で表されるアミノ酸配列を含む重鎖と、配列番号:34で表されるアミノ酸配列を含む軽鎖とを含む抗体、
(2) 配列番号:33及び/又は34で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体、及び
(3) 配列番号:33及び/又は34で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
〔14〕前記炎症性疾患が、自己免疫疾患である、上記〔1〕から〔13〕のいずれか1項に記載の炎症性疾患の治療又は予防剤;
〔15〕前記自己免疫疾患が、全身性エリテマトーデス、又は乾癬である、上記〔14〕に記載の炎症性疾患の治療又は予防剤;
〔16〕以下のいずれかの抗TLR7抗体:
(i) 以下の少なくとも1つを含む抗体:(a) 配列番号:3で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:4で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:5で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:6で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:7で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:8で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:3~8のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:3~8のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
(ii) 以下のいずれかの抗体:(1) 配列番号:9で表されるアミノ酸配列を含む重鎖と、配列番号:10で表されるアミノ酸配列を含む軽鎖とを含む抗体;(2) 配列番号:9及び/又は10で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び(3) 配列番号:9及び/又は10で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
(iii)配列番号:1で示されるTLR7のアミノ酸配列のうち、27位~457位の領域内に特異的に結合するモノクローナル抗体;
(iv)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の228位~364位の領域内に特異的に結合する抗体;
(v)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の275位~313位の領域を含む領域に特異的に結合する抗体;又は
(vi)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の313位~364位の領域を含む領域に特異的に結合する抗体;及び
〔17〕以下のいずれかのTLR9抗体:
(i) 以下の少なくとも1つを含むモノクローナル抗体:(a) 配列番号:11で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:12で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:13で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:14で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:15で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:16で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:11~16のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:11~16のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;

本発明に係る炎症性疾患の治療又は予防剤は、抗TLR7抗体又は抗TLR9抗体を含む。
TLRはI型の膜タンパク質で細胞外にLeucine rich repeat(LRR)を有する。病原体成分を認識すると、細胞内のToll/IL-1R homology(TIR)ドメインによりシグナル伝達を行う。リガンドを認識したTLRはTIRドメインを介したシグナルを細胞内へ伝えることにより、最終的にNF-κBやInterferon-Regulatory Factor(IRF)ファミリーなどの転写因子を活性化させ、炎症性サイトカイン(IL-6やIL-12、TNFαなど)や炎症性ケモカイン(RANTESなど)、I型インターフェロン(IFNαやIFNβ)の産生を誘導し、局所において適切な自然免疫応答を引き起こす。これらのTLRを介した免疫応答は生体防御において必要不可欠であり、TLR応答に関連する分子が欠損すると、様々な病原体に感染し易くなることが報告されている。しかし、何らかの原因で、自己に由来する物質がTLRの内因性リガンドとなると、慢性的な炎症を引き起こす可能性が指摘されている。
抗TLR7抗体は、後述する実施例に示すとおり、細胞表面TLR7に結合し、当該細胞のTLR7応答を阻害することにより、免疫の異常な活性化を防いで炎症性疾患の治療又は予防に寄与する TLR7は、配列番号:1に示すアミノ酸配列を有するI型の膜タンパク質である。
TLR9は、配列番号:2に示すアミノ酸配列を有するI型の膜タンパク質である。
(a) 配列番号:3で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:4で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:5で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:6で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:7で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:8で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:3~8のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:3~8のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR。
配列番号:3~8で表されるアミノ酸配列からなるCDRを有する抗体は、実施例に示されるとおり、免疫細胞におけるTLR7応答を阻害し、マウスの炎症性疾患モデルの病態を改善させる。
本明細書において用語「アミノ酸」は、その最も広い意味で用いられ、天然のアミノ酸のみならずアミノ酸変異体及び誘導体といったような非天然アミノ酸を含む。アミノ酸の例としては、天然タンパク原性L-アミノ酸;D-アミノ酸;アミノ酸変異体及び誘導体などの化学修飾されたアミノ酸;ノルロイシン、β-アラニン、オルニチンなどの天然非タンパク原性アミノ酸;及びアミノ酸の特徴である当業界で公知の特性を有する化学的に合成された化合物などが挙げられるがこれらに限定されない。非天然アミノ酸の例としては、α-メチルアミノ酸(α-メチルアラニンなど)、D-アミノ酸、ヒスチジン様アミノ酸(2-アミノ-ヒスチジン、β-ヒドロキシ-ヒスチジン、ホモヒスチジン、α-フルオロメチル-ヒスチジン及びα-メチル-ヒスチジンなど)、側鎖に余分のメチレンを有するアミノ酸(「ホモ」アミノ酸)及び側鎖中のカルボン酸官能基アミノ酸がスルホン酸基で置換されるアミノ酸(システイン酸など)が挙げられるがこれらに限定されない。
本明細書において、「配列番号:Xに示すアミノ酸配列に対してY%以上の同一性を有する」とは、2つのポリペプチドのアミノ酸配列の一致が最大になるように整列(アライメント)させたときに、共通するアミノ酸残基数の、配列番号:Xに示す全アミノ酸数に対する割合が、Y%以上であることを意味する。
本発明に用いられる抗TLR7抗体においては、結果として得られるCDRのセットが抗TLR7抗体のCDRとしての機能を保持する限り、配列番号:3~8に示されるアミノ酸配列と90%以上、95%以上、98%以上の同一性を有するアミノ酸配列からなるものとすることができる。
(1) 配列番号:9で表されるアミノ酸配列を含む重鎖と、配列番号:10で表されるアミノ酸配列を含む軽鎖とを含む抗体、
(2) 配列番号:9及び/又は10で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体、
(3) 配列番号:9及び/又は10で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。及び
(4) 上記(1)から(3)のいずれかの抗体と同じエピトープを認識する抗体
したがって、上記(1)~(4)で示される抗体は、A94B10抗体と同様に、免疫細胞におけるTLR7応答を阻害すると考えられる。
配列番号:1で示されるマウスTLR7のアミノ酸配列のうち275位~313位の領域を含む領域としては、例えば、275位~313位の領域を含む120アミノ酸以下の領域、110アミノ酸以下の領域、100アミノ酸以下の領域、90アミノ酸以下の領域とすることができる。
配列番号:1で示されるマウスTLR7のアミノ酸配列のうち313位~364位の領域を含む領域としては、例えば、313位~364位の領域を含む120アミノ酸以下の領域、110アミノ酸以下の領域、100アミノ酸以下の領域、90アミノ酸以下の領域、80アミノ酸以下の領域、70アミノ酸以下の領域、60アミノ酸以下の領域、とすることができる。

配列番号:33で表されるアミノ酸配列を含む重鎖と、配列番号:34で表されるアミノ酸配列を含む軽鎖を含む抗体は、C34A1抗体である。
本発明は、上述した炎症性疾患の治療剤又は予防剤に含まれる抗TLR7抗体又は抗TLR9抗体自体も提供するものである。
本発明で用いられる抗TLR7抗体又は抗TLR9抗体の作製方法は限定されないが、例えば、抗TLR7モノクローナル抗体は、TLR7又はその断片で免疫した非ヒト哺乳動物から抗体産生細胞を単離し、これを骨髄腫細胞等と融合させてハイブリドーマを作製し、このハイブリドーマが産生した抗体を精製することによって得ることができる。また、抗TLR7ポリクローナル抗体は、TLR7はその断片で免疫した動物の血清から得ることができる。免疫に用いるTLR7の断片は、得られる抗体が細胞表面TLR7に結合してその機能を阻害する限り特に限定されないが、例えば、配列番号:1の27位~457位アミノ酸のTLR7断片が挙げられる。
抗TLR9モノクローナル抗体及びポリクローナル抗体も同様に作製することができる。
例えば、多数のランダムな配列のペプチドを固相担体に固定してアレイ化し、抗体Xと反応させ、酵素標識2次抗体で結合を検出して、抗体Xが特異的に結合するペプチドのアミノ酸配列を調べ、このアミノ酸配列と抗原タンパク質のアミノ酸配列の相同性を検索することによって、抗原タンパク質上のエピトープを決定することが可能である。固相担体に固定するペプチドを、予め、抗原タンパク質の部分ペプチド群としてもよい。
また、抗原タンパク質の種々の部分ペプチドの存在下で、抗体Xと抗原タンパク質との結合をELISA法で検出し、競合活性の有無を調べることによっても、抗原タンパク質上のエピトープを決定することが可能である。
エピトープの配列を決定することができれば、これに特異的に結合する抗体Yは、公知の方法にしたがって当業者が作製することができる。例えば、エピトープ配列を含むペプチドを固相担体に固定し、当該ペプチドと種々の抗体の結合を検出することにより、同エピトープに特異的に結合する抗体を得ることができる。
ここで、「種々の抗体」としては、動物を抗原タンパク質又はその部分ペプチドで免疫することによって得たものを用いてもよいし、ファージディスプレイ法によって作製した抗体ライブラリ又は抗体フラグメントライブラリを用いてもよい。ファージディスプレイ法によるライブラリを用いる場合、エピトープ配列を含むペプチドを固相担体に固定しパニングを繰り返すことによって、同エピトープに特異的に結合する抗体Yを得ることもできる。
例えば、ヒト型キメラ抗体の場合、マウス抗体を産生するハイブリドーマのmRNAから逆転写酵素によりcDNAを合成し、重鎖可変領域(VH)及び軽鎖可変領域(LH)をPCRでクローニングして配列を解析する。次に、一致率の高い抗体塩基配列から、リーダー配列を含む5’プライマーを作製し、5’プライマーと可変部3’プライマーによって上記cDNAから、シグナル配列から可変領域の3’末端までをPCRでクローニングする。一方で、ヒトIgG1の重鎖及び軽鎖の定常領域をクローニングし、重鎖と軽鎖それぞれについて、マウス抗体由来可変領域と、ヒト抗体由来定常領域とをPCRによるOverlapping Hanging法で連結し、増幅する。得られたDNAを適当なベクターに挿入し、これを形質転換して、ヒト型キメラ抗体を得ることができる。
「配列番号:Xに示されるアミノ酸配列において1又は2個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR」や、「配列番号:Xに示されるアミノ酸配列に対してY%以上の同一性を有するアミノ酸配列からなるCDR」は、部位特異的変異導入法、ランダム変異導入法、チェーンシャフリング法、CDRウォーキング法などの公知の方法を用いて作製され得る。
これらの方法により、ファージディスプレイ法によってCDRに種々の変異を有する抗体又は抗体断片をファージ表面に提示させ、抗原を使用してスクリーニングすることにより、より親和性が成熟したCDRを得られることが当業者によく知られている(例えば、Wu et al., PNAS, 95:6037-6042(1998); Schier, R. et al., J. Mol. Bio. 263:551-567(1996); Schier, R. et al., J. Mol. Biol. 255:28-43(1996); Yang, W.P. et al., J. Mol. Biol., 254:392-403(1995)。)。本発明は、このような方法で成熟させたCDRを含む抗体も包含する。
ある抗TLR7抗体又は抗TLR9抗体を、本発明に係る炎症性疾患の予防又は治療剤に用いることができるか否かは、当業者が適宜決定することができる。
例えば、実施例に示される方法にしたがって、(i) 得られた抗体が細胞表面のTLR7又はTLR9に結合するか否か;(ii) 免疫細胞をTLR7又はTLR9のリガンドによって刺激しつつ、得られた抗体と接触させたときに、免疫細胞から分泌される炎症性サイトカイン量が抑制されるか否か;(iii) B細胞をTLR7又はTLR9のリガンドによって刺激しつつ、得られた抗体と接触させたときに、B細胞の増殖が抑制されるか否か;及び、(iv) 炎症性疾患モデル動物に得られた抗体を投与することによって、病態が改善されるか否か、の少なくとも1つを確認することにより、得られた抗体から、自己免疫疾患の予防剤又は治療剤として用いられるものを選択することが可能である。
中でも、その発症の機序にTLR7及び/又はTLR9が関与することが報告されている、SLEや乾癬に有用と考えられる。
本発明は、対象に本発明に係る抗体を投与する工程を含む自己免疫疾患の治療又は予防方法も包含する。
〔材料と方法〕
マウスと細胞
野生型C57BL/6マウスおよび野生型Balb/cマウスは日本SLCより購入した。Unc93B1 D34A変異マウス(Unc93b1D34A/D34A)は福井竜太郎助教より提供頂いた。TLR7欠損マウス(TLR7-/-)は審良静男博士(大阪大学)より提供頂いた。マウスはSPF環境にて飼育し、東京大学の倫理規定に基づいて実験を行った。プロB細胞由来の細胞株であるBa/F3細胞はRPMI培養液(10%FCS、50μM 2ME、ペニシリン/ストレプトマイシン/1-グルタミン混合溶液、IL-3を含む)を用いて培養した。C末端側にFlag-His6を付加させたマウスおよびヒトTLR3、TLR7、TLR8、TLR9は福井竜太郎助教から分与して頂いた(Fukui, R. et al., J Exp Med 2009)。PRAT4AがノックダウンされたM12細胞は柴田琢磨特任助教から分与して頂いた(Takahashi, K. et al. J Exp Med 2007)。
CpGB(5' -TCCATGACGTTCCTGATGCT- 3')DNA(配列番号:35)及びCpGA(5' - GGGGTCAACGTTGAGGGGGG-3')DNA(配列番号:36)、PolyU (5' -UUUUUUUUUUUUUUUUUUU-3`)(配列番号:37)、RNA9.2s-DR (5' -UGUCCUUCAAUGUCCUUCAA-3`)(配列番号:38)は北海道システムサイエンスに合成を委託した。
LipidAは深瀬浩一博士(大阪大学)に合成を依頼し、提供を受けた。
Pam3CSK4、Poly(I:C)、Loxoribine(ロキソリビン)、およびImiquimod(イミキモド)はinvivogenより購入した。
リコンビナントマウスgranulocyte macrophage colony-stimulating factor(GM-CSF)およびリコンビナントマウスmacrophage colony-stimulating factor(M-CSF)、マウスFlt3リガンドはpeprotechより購入した。
ピューロマイシン、ネオマイシンはSIGMAより購入した。RPMI培養液、DMEM培養液はGIBCOより購入した。リコンビナントマウスIL-3はCHO細胞を使用して当研究室により作製したものを使用した。
TLR7は切断されることが報告されている(Ewald et al. Nature 2008)。そこで、TLR7の切断アミノ酸部位を決定するためにEdman分解法によりN末端アミノ酸配列分析を行った。サンプルの調整方法を以下のように示す。
N末端アミノ酸配列分析に使用した細胞はC末端側にGreen Fluorescent Protein(GFP)を付加させたTLR7 (TLR7-GFP)を強制発現させたRAW264.7細胞を用いて、1×1010まで増やし、回収した。
回収した細胞を氷冷lysis buffer(組成は表1)により30分かけて溶解し、遠心後のlysateを回収した。回収したlysateを抗GFP抗体(FM264)ビーズに加え、4℃で12時間撹拌させ免疫沈降した。免疫沈降したビーズを回収し、Washing buffer(0.1% Triton X-100, 30 mM Tris/HCl pH 7.4, 150mM NaCl)により4回洗浄し、Elution buffer(0.1% Triton X-100, 30 mM Glycine/HCl pH 2.5, 150mM NaCl)でTLR7-GFPを溶出した。溶出したタンパク質はポリアクリルアミド電気泳動をして、クマシーブリリアントブルー染色によりTLR7-GFPを確認した。溶出したタンパク質を確認した後、TLR7-GFPが含まれた溶液をdialysis buffer(0.1% Triton X-100, 10mM NaCl)により透析した。透析した溶液は回収した後、4.5時間かけて凍結乾燥させた。凍結乾燥させたタンパク質を50μl Sample buffer(62.5 mM Tris/HCl pH 6.8, 10 % Glycerol, 2 % sodium dodecyl sulfate (SDS), 0.025 % bromophenol blue, 5 % 2-mercaptoethanol)で溶解させ、ポリアクリルアミド電気泳動して、CAPS transfer buffer(1 x CAPS pH 11.0, 10 % methanol)の中でPVDF膜に転写した。転写したTLR7はクマシーブリリアントブルー染色により可視化し、切断されたTLR7 C末端タンパク質のバンドを切り取った。切り取ったPVDF膜はCAPS bufferにより2回洗浄した後、そのサンプルはEdman分解法によるN末端アミノ酸配列決定を行うために、APRO社に解析を依頼した。
抗マウスTLR7抗体A94B10は以下のように作製した。
免疫動物にはC57BL/6バックグラウンドのTLR7欠損マウスをBalb/cマウスへ6回戻し交配を行い、Balb/cバックグラウンドのTLR7欠損マウスを用いた。抗原には、Flag-His6エピトープを付加したマウスTLR7(mTLR7-fH)を遺伝子導入したBa/F3細胞を使用した。
免疫は、免疫原をマウスの腹腔内に投与することで行った。免疫初日にCFAと混合した抗原を、免疫8日目にIFAと混合した抗原を、更に1週間毎に1 x PBSで希釈した抗原を3回免疫した。最終免疫日から5日目に脾臓を摘出し、得られた脾臓細胞をマウスミエローマ由来のSp2/o細胞と細胞融合させた。このハイブリドーマから得られるモノクローナル抗体を、A94B10抗体と呼ぶ場合もある。
マウスTLR7を特異的に認識する抗体を培養上清中に産生するハイブリドーマのクローンを選択するために、抗原であるmTLR7-fHを強制発現させたBa/F3細胞に対し、0.1%サポニン含有FACS溶液(1 x PBS、2.5%FBS、0.1%NaN3)を用いて細胞内染色を行い、フローサイトメトリーにより選別した。得られたハイブリドーマ細胞の中から抗マウスTLR7抗体を産生するモノクローナル抗体産生ハイブリドーマ株A94B10を樹立した。
得られたクローンA94B10のアイソタイプはIgG1であった。また、この抗マウスTLR7抗体はBM-MCsおよびBM-cDCs、BM-pDCs、脾臓細胞由来のB細胞に発現している内在性TLR7をフローサイトメトリー、免疫沈降および共焦点顕微鏡で検出することができた。これらの事実は、樹立したハイブリドーマ株がマウスTLR7特異的な抗体を産生していることを示す。
マウスの大腿骨および脛骨より採取した骨髄細胞を溶血処理した後、BM-MCsおよびBM-cDCs、BM-pDCsを誘導した。BM-MCsは骨髄細胞を5 x 106/径10cmバクテリアシャーレ(greiner Bio-one)に調整して播き、100ng/mlのリコンビナントマウスM-CSF(peprotech)を含むDMEM培養液で7日間培養することにより誘導した。BM-cDCsは骨髄細胞を1 x 107/径10cm細胞培養用シャーレ(greiner Bio-one)に調整して播き、10ng/mlのマウスGM-CSF(peprotech)を含むRPMI培養液で7日間培養することにより誘導した(Kaisho, T. et al., J Immunol 2001)。BM-cDCsの誘導では、2日毎に上記培養液を半量ずつ新たなGM-CSF含有RPMI培養液に交換し、細胞増殖とともに培養液を増やした。
BM-pDCsは骨髄細胞を1 x 107/径10cm細胞培養用シャーレに調整して播き、30ng/mlのマウスFlt3リガンド(peprotech)を含むRPMI培養液で7日間培養することにより誘導した。BM-cDCsの誘導では、誘導期間中毎日、誘導している培養液を半量ずつ新たなFlt3リガンド含有RPMI培養液に交換し、細胞増殖とともに培養液を増やした。BM-pDCsは細胞表面マーカーとしてCD11c陽性B220陽性細胞集団であることが知られている。そこで、誘導したBM-pDCをanti-mouse CD11c-PEおよびanit-mouse B220-APCを用いて染色し、FACS Aria(BD)によりCD11c陽性B220陽性細胞を単離した。
CD43は休止期や通常の末梢B細胞には発現しない。これを利用し、CD43陰性の脾臓細胞分画を回収することで脾臓B細胞を精製した(Nagai, Y. et al., J Immunol 2005)。野生型マウスより脾臓を摘出し、スライドガラスを用いて脾臓組織より細胞を単離した。溶血処理後、脾臓細胞にCD43 MicroBeads(Miltenyi Biotec株式会社)を加え、4℃で30分間混和した。Running buffer(1 x PBS、2% FBS、2mM EDTA)により2回洗浄した後、AutoMACSのdeplete programでCD43陰性細胞群を分離した。B細胞の分離効率はFACS Callibur(BD)を用いたフローサイトメトリーにより確認した。分離したB細胞の培養は10%FBS添加RMPI培養液で行った。
細胞増殖能は細胞への[3H]thymidine取り込み量を測定することで定量した。脾臓細胞より精製したB細胞を1 x 105/wellで調整し、96穴平底プレートに藩種し、複数種のTLRリガンドで刺激した。72時間刺激後に[3H]thymidine 1μCi/ml存在下で更に5時間培養し、セルハーベスターシステム(Inotech)を用いて細胞内に取り込まれた[3H]thymidine量を測定した(Nagai et al.J Immunol 2005)。
96穴平底プレートに1 x 105/wellで調整した細胞に、複数種のTLRリガンドを添加した。24時間後の培養上清をサンドイッチELISAに供し、TNFα、IL-6、IL-12p40、RANTESの濃度をELISA Kit(R&D Systems)およびIFN-αの濃度をELISA Kit(PBL Interferon Source)により測定した。
フローサイトメトリーはFACS Callibur(BD)で解析した。解析する細胞をFACS溶液で洗浄した後、染色に使用した。抗体は以下を用いた。
Fluorescein (FITC)-conjugated anti-mouse CD4(L3T4), Phycoerythrin(PE)-conjugated anti-mouse CD62L(MEL-14), Allophycocyanin(APC)-conjugated anti-mouse CD8α(53-6.7), APC-conjugated anti-mouse Ly6G(1A8), PEcy7-conjugated anti-mouse CD19(1D3), Streptavidin-APCはBD pharmingenより購入した。Biotin conjugated anti-mouse ICOS(7E.17G9), Biotin mouse IgG1,κ Isotype ctrl antibody(MOPC-21), FITC-conjugated anti-mouse B220(RA3-6B2), FITC- conjugated anti-mouse CD11b(M1/70), FITC-conjugated anti-mouse CD71(R17217), PE-conjugated anti-mouse Ter119(TER119), PerCP/Cy5.5-conjugated anti-mouse CD11c(N418), APC-conjugated anti-mouse/human CD44(lM7), APC-conjugated anti-mouse B220(RA3-6B2), Alexa488-conjugated anti-mouse GL-7(GL-7)はeBioscienceより購入した。PE-conjugated anti-mouse CD11c(N418), PerCP/Cy5.5-conjugated anti-mouse Ly6C(HK1.4), PerCP-conjugated anti-mouse CD4(GK1.5), APC-conjugated anti-mouse IgD(11-26c,2a), Pacific Blue-conjugated anti-mouse PDCA-1(927), Streptavidin-PEはBiolegendより購入した。R-PE conjugated goat anti-mouse IgG(H+L) (Absorbed against human immunoglobulins), APC-conjugated anti-mouse Lamp1(1D4B)はSouthern Biotechより購入した。Biotin-conjugated anti-mouse Flag(M2)はSigma Aldrichより購入した。Biotin-conjugated anti-mouse TLR4/MD2(MTS510)は当研究室により作製したものを使用した(Akashi, S. et al., J Immunol 2000)。
C末端側にFlag-His6を付加させたヒトTLR7は薬剤耐性遺伝子のないレトロウイルスベクターpMXsに組み込んだ。
抗TLR7モノクローナル抗体のエピトープ決定を行うために、TLR7の切断アミノ酸部位を境にN末端領域TLR7(27番目のフェニルアラニンから457番目のプロリン)およびC末端領域TLR7(461番目のグルタミン酸から837番目のアスパラギン酸)を作製し、InFusion酵素(TaKaRa)によりpDisplayベクター(Invitorgen)に組み込み、さらにピューロマイシン耐性遺伝子済みのレトロウイルスベクターpMXpに組み込んだ。このベクターは組み込んだ遺伝子により発現した目的タンパク質を細胞表面に表示させ、そのN末端にHA抗原を付加することが可能である。
レトロウイルスベクターのレトロウイルス粒子への取り込みにはHEK293由来パッケージング細胞株であるPlat-E細胞にFuGene6(Roche)を用いて遺伝子導入した。導入後24時間後に培養上清を回収し、ウイルス懸濁液として確保した。このウイルス懸濁液にDOTAP(Roche)を混和させ、遺伝子を導入する対象の細胞に加えて2000rpmで1時間の遠心を行った。
遺伝子導入後、pMXpを遺伝子導入した細胞にはピューロマイシン2μg/ml、pSSCNを遺伝子導入した細胞にはネオマイシン 4μg/mlを添加することで遺伝子導入細胞の選択を行った。
レトロウイルス作製に必要なpMXとその派生レトロウイルスベクター(pMX, pMXp)およびパッケージング細胞株であるPlat-Eは東京大学医科学研究所 北村俊雄教授より分与して頂いた。
TLR7のタンパク質発現をウエスタンブロットにより解析した。細胞は1×PBSにより2回洗浄し、回収した。タンパク質発現の検出は回収した細胞を氷冷溶解用緩衝液(1% Digitonin, 20mM Tris-HCl (pH 7.4), 150mM NaCl, 1mM CaCl2, 1mM MgCl2, 10% Glycerol, 1mM DTT 及びComplete protease Inhibitor Cocktail (Roche))により30分かけて溶解し、遠心後のlysateを回収した。回収したlysateを抗TLR7モノクローナル抗体(A94B10)と結合させたN-hydroxysuccinimide-activated Sepharose 4FFビーズに加え、2時間、4℃で撹拌した。この行程により、細胞内に発現しているTLR7を免疫沈降した。撹拌後、ビーズを氷冷した洗浄用緩衝液(0.1% Digitonin、20mM Tris/HCl(pH 7.4)、150mM NaCl、1mM CaCl2、1mM MgCl2、0.1% Glycerol、1mM DTT)で3回洗浄した。洗浄したビーズにSDSサンプル緩衝液(125 mM Tris/HCl [pH 6.8], 20% glycerol, 4% SDS, 10% 2-ME, 0.005% bromophenol blue)を加え、96℃で5分間加熱してタンパク質を変性処理した。これら調整したサンプルはポリアクリルアミド電気泳動して、タンパク質をPVDF膜に転写し、ウエスタンブロットした。
ウエスタンブロットに使用した抗体は以下の通りである。Rabbit anti-mouse Grb2(C-23)はSanta Cruz Biotechnologyから購入した。抗マウスTLR7ポリクローナル抗体(TLR7N)はeBioscienceから購入した。
細胞表面をビオチン化した免疫沈降およびウエスタンブロットは以下のような方法で行った。
EZ-Link Sulfo-NHS-LC-Biotin(Thermo scientific)をBiotin labeling buffer (1×PBS, 15mM HEPES, 150mM NaCl)で10mg/mlに調整したビオチン化試薬を用いて、HBSS(-)で2回洗浄した細胞の表面を5 x 107細胞あたり0.1mgでビオチン化した。ビオチン化した細胞をHBSS(-)で3回洗浄し、回収した。回収した細胞を氷冷溶解用緩衝液により30分かけて溶解し、遠心後のlysateを回収した。回収したlysateを抗TLR7モノクローナル抗体(A94B10)と結合させたN-hydroxysuccinimide-activated Sepharose 4FFビーズに加え、12時間、4℃で撹拌した。この行程により、内在性TLR7を免疫沈降した。撹拌後、ビーズを氷冷した洗浄用緩衝液で3回洗浄した。洗浄したビーズにSDSサンプル緩衝液を加え、調整した。これら調整したサンプルはポリアクリルアミド電気泳動して、タンパク質をPVDF膜に転写し、ウエスタンブロットした。
細胞表面ビオチン化によるウエスタンブロットに使用した抗体はストレプトアビジン標識西洋ワサビペルオキシダーゼ(StAv-HRP)により検出した。抗マウスTLR7ポリクローナル抗体(TLR7N)はeBioscienceから購入した。
抗体によるマウス投与実験において、抗TLR7モノクローナル抗体投与群とコントロール抗体投与群の間におけるデータの有為差検定はStudent t検定により算定した。T検定における危険率が0.01未満の場合、比較群間の差が優位であると判定した。
(試験管内試験)
細胞はBM-MCs、BM-cDCs、BM-pDCsおよび脾臓細胞から単離したB細胞を用いた。96well平底プレートにBM-MCs、BM-cDCs、BM-pDCsを1×105播種し、抗TLR7抗体を各々の濃度で添加した。抗体を添加してから4時間後、TLRリガンドを藩種した培養細胞に加えた。TLRリガンド添加した24時間後、培養上清液を回収した。回収した培養上清液はリガンド刺激によって、産生されたサイトカインを測定するためにELISA法により検出した。
(生体試験)
また、Unc93B1 D34A変異マウスはTLR7過剰応答により、血小板減少症および脾腫を呈することが報告されている(Fukui et al. immunity 2011)。
そこで、Unc93B1 D34A変異マウスが血小板の減少を認めた12週齢から16週齢(野生型マウスが85.3[104/μl]に対して、Unc93B1 D34A変異マウスが32.4[104/μl])において、抗体投与を開始した。投与抗体量は静脈注射により200μg/マウスで行った。用いた抗体はA94B10抗体およびコントロール抗体であり、その投与頻度は毎週行った。抗体投与による血小板減少の影響を精査するために、抗体を投与してから2週間毎に尾採血を行い、血球計数器(MEK-6450セルタックα;NIHON KOHDEN)により血小板値を測定した。抗体投与から10週後、D34A変異マウスから脾臓を摘出し、重量を測定した。さらに、同マウスから摘出した脾臓、リンパ節(上腕部、液窩、鼠径リンパ節)はフローサイトメトリーにより細胞集団を解析した。
QIAGENから購入したRNeasy Mini Kitを用いてハイブリドーマ株A94B10、B33A4、C34A1よりtotal RNAを単離、精製した。Clontechから購入したSMARTer RACE cDNA Amplification Kitを用いてtotal RNAより重鎖アイソタイプ及び軽鎖特異的な1stストランドcDNA合成を行った後5'-RACE PCRを行い、可変領域を含む全長重鎖及び軽鎖cDNAを合成した。これらcDNAをTaKaRaより購入したMighty TA-cloning Kitを用いてpMD20-T vectorへ挿入し、FASMACへ塩基配列解析を依頼し、塩基配列よりアミノ酸配列を決定した。CDR配列は、チューリッヒ大学より提供されているデータベースAHo's Amazing Atlas of Antibody Anatomyを用いて、アミノ酸配列を比較する事により決定した。
〔結果〕
抗TLR7モノクローナル抗体の樹立と細胞表面TLR7の検出
一次免疫細胞(primary immune cell)の内因性TLR7の細胞内局在を調べるため、マウスTLR7に対するモノクローナル抗体A94B10抗体を樹立した。TLR7は細胞内小胞に局在すると信じられていたが、抗TLR7モノクローナル抗体は、骨髄由来マクロファージの細胞表面にTLR7が存在することを明らかにした(図1A)。
染色の特異性は、TLR7ノックアウトマウスの骨髄由来マクロファージが染色されないことによって確認した。さらに、TLR7の細胞表面での発現を確認するため、細胞表面をビオチン化し、TLR7を骨髄由来マクロファージから免疫沈降させた。沈降したTLR7はストレプトアビジン又は抗TLR7抗体で検出された。TLR7はエンドリソソームでタンパク質分解的に切断された。全長TLR7と切断されたTLR7はいずれも、野生型細胞でストレプトアビジンによって検出されたが、TLR7ノックアウト細胞では検出されなかった(図1B)。このことは、プロセシングを受けたTLR7も受けていないTLR7も細胞表面に発現していることを示す。全長及び切断されたTLR7は、それぞれゴルジ装置又はエンドリソソームに由来するものと考えられる。
細胞表面TLR7は、骨髄由来の古典的又は形質細胞様樹状細胞ではほとんど検出されなかったが(図1A)、脾臓由来のレジデントpDC又はcDCでは検出された。in vitroの分化したDCとin vivoのレジデントDCでの細胞表面TLR7の違いの理由は不明である。脾臓におけるCD11b+マクロファージ/単核球及び好中球も細胞表面TLR7を発現していたが、脾臓由来DCよりはかなり少なかった(データ示さず)。B細胞もTLR7リガンドに反応したが、細胞表面TLR7は検出されなかった(データ示さず)。
TLR7及び9の、小胞体(ER)からエンドリソソームへの輸送には、2つのERレジデントタンパク質PRAT4A及びUnc93B1を必要とする。細胞表面TLR7は、Unc93B1機能欠損マウスであるUnc93B13d/3dマウス(図1C)又はPRAT4A-/-マウス(データ示さず)由来の脾臓DCでは検出されなかった。このことは、PRAT4A及びUnc93B1がいずれもTLR7の細胞表面への輸送に必要とされることを示す。細胞表面TLR7は、脾臓MyD88-/- cDCでは検出された(データ示さず)。このことは、TLRシグナルは、TLR7の細胞表面での発現に必要とされないことを示す。B細胞におけるTLR7発現は、I型インターフェロンにより持続することが知られる。しかしながら、IFNAR-/- cDCは、TLR7の細胞表面での発現に変化は見られなかった。I型インターフェロンは、TLR7の細胞表面発現には必要とされていない。
また、TLR7の切断部位を調べたところ、460番目のロイシンと461番目のグルタミン酸および461番目のグルタミン酸と462番目のアラニンの間で切断されていることが明らかになった(図9A)。A94B10抗体は、N末端領域TLR7(TLR7N;27番目のフェニルアラニンから457番目のプロリン)に結合し、C末端領域TLR7(TLR7C;461番目のグルタミン酸から837番目のアスパラギン酸)には結合しないことがわかった(図9B)。
TLR7は、エンドリソソームでRNAを認識すると考えられていた(Ewald, S. E. et al. J Exp Med 2011)。RNA認識における細胞表面TLR7の役割を調べるために、抗TLR7モノクローナル抗体を、様々な免疫細胞に加えた。
予想に反して、抗TLR7モノクローナル抗体は、低分子のTLR7リガンドであるloxoribine、又は合成RNAで刺激した骨髄由来マクロファージによるIL-6、TNF-α、IL-12、及びRANTES (CCL5)の産生を濃度依存的に阻害した(図2A)。TLR9リガンドであるCpG-BによるTLR9依存性サイトカイン産生は変化しなかった。予想に反して、TLR7特異的阻害は、細胞表面TLR7がほとんど検出されなかったBM-cDCs及びBM-pDCs(それぞれ図2及び図3)でも見られた。ポリウリジル酸(PolyU)に対するBM?pDCs反応は、中程度ではあるが有意に阻害された(図4)。さらに、低分子のTLR7リガンドであるimiquimod応答性のB細胞増殖も、大きく阻害された(図5)。これらの結果は、抗TLR7モノクローナル抗体が、BM-cDCs BM-pDCs、BM-MCs及び脾臓由来B細胞のTLR7応答を阻害できることを示した。
次に、抗TLR7モノクローナル抗体の阻害効果を、TLR7依存性炎症性疾患を有するマウスで調べた。Unc93b1遺伝子の34位のアスパラギン酸がアラニンに置換されたUnc93b1D34A/D34Aマウスは、TLR7の過反応による全身性の致死性の炎症を生じる(Fukui et al. immunity 2011)。
全身性の炎症は、末梢血で簡単に監視できる血小板減少症を引き起こす。抗TLR7抗体の治療効果を調べるため、すでに血小板減少症を発症した3~4月齢のUnc93b1D34A/D34Aマウスにモノクローナル抗体を注射投与した。マウス当たり200μgのモノクローナル抗体を週1回注射し、末梢血の血小板を1週間おきに測定した。10回の治療後、抗TLR7モノクローナル抗体投与群のマウスの血小板は増加したが、コントロール抗体を投与した群では次第に減少した(図6A)。
抗TLR7モノクローナル抗体で10週間治療した後、マウスを犠死させ、病態を分析した。Unc93b1D34A/D34Aマウスでは、脾臓の骨髄細胞の赤芽球が増加した。脾臓における赤芽球の割合は、コントロール抗体投与群よりも、抗TLR7抗体投与群において著しく低かった(図6B)。T細胞は活性化され、細胞表面の共刺激分子ICOSの発現の亢進に伴って、メモリーT細胞へと分化した。Unc93b1D34A/D34AマウスにおけるCD4+細胞上のメモリーT細胞とICOSの発現の割合は、コントロール抗体で処理した群に比較して著しく低かった(図6C及び6D)。これらの結果は、抗TLR7抗体が、Unc93b1D34A/D34AマウスのTLR7依存性全身性炎症を改善することを示す。
次に、抗TLR7モノクローナル抗体が、in vivoでTLR7リガンドによって誘導されるサイトカイン産生を抑制するかどうか調べた。図10上段に示すスケジュールでA94B10抗体と、TLR7リガンドであるR848を投与し、血液中のIL-6とIFNαの濃度を測定した。
結果を図10下段に示す。A94B10抗体は、R848投与によって誘導されるサイトカイン産生を、濃度依存的に有意に抑制することが確認された。
次に、抗TLR7モノクローナル抗体が、in vivoでTLR7リガンドによって誘導される皮膚の炎症を抑制するかどうか調べた。図11上段に示すスケジュールでA94B10抗体を投与するとともに、TLR7リガンドであるImiquimodをマウスの耳に皮内投与(Intradermal;i.d.)し、耳介厚を測定することによって炎症の程度を評価した。
結果を図11に示す。A94B10抗体は、Imiquimodによって誘導される皮膚の炎症を有意に抑制することが確認された。
次に、抗TLR7モノクローナル抗体が、乾癬モデルマウスにおいて炎症を抑制するかどうか調べた。図12上段に示すスケジュールで、マウスにIL-23を耳に投与して乾癬モデルを作製し、A94B10抗体を投与した。IL-23をマウスに投与するとTLR7発現が亢進し(図12左下)、ヒトの乾癬に非常に近いモデルとなることが知られている。耳介厚を測定することによって炎症の程度を評価した。
結果を図12右下に示す。A94B10抗体は、IL-23で誘導した乾癬モデルマウスの炎症を有意に抑制することが確認された。
ハイブリドーマ株A94B10から得られるモノクローナル抗体の重鎖アミノ酸配列を配列番号:9に、軽鎖アミノ酸配列を配列番号:10に示す。また、同抗体の重鎖CDR1~3のアミノ酸配列を配列番号:3~5に、軽鎖CDR1~3のアミノ酸配列を配列番号:6~8に示す。
A94B10抗体のエピトープを決定するために、図13上段に示すように、配列番号:1に示すTLR7のアミノ酸27位から457位、27位から364位、27位から312位、及び27位から226位までの断片を細胞株に発現させ、各断片とA94B10抗体との結合をフローサイトメトリーで測定した。
結果を図13下段に示す。TLR7N27-312にはA94B10抗体が結合しなかったが、TLR7N27-364には結合したことから、A94B10抗体は、TLR7のアミノ酸配列の313位から364位の領域付近にエピトープを有すると考えられた。
mTLR71-227、mTLR71-275、及びmTLR71-313では、それぞれN末端から1位~227位、1位~275位、及び1位~313位をマウスTLR7の配列とし、C末端側をヒトTLR7の配列とした。また、mTLR7228-313、mTLR7228-275、mTLR7275-313では、N末端とC末端をヒトTLR7の配列とし、中央をマウスTLR7の配列とした。
結果を図14下段に示す。マウスTLR71-274を含むキメラTLR7にはA94B10抗体が結合しなかったが、マウスTLR71-313およびマウスTLR7275-313を含むキメラTLR7には結合したことから、A94B10抗体は、TLR7のアミノ酸配列の275位から313位の領域付近にエピトープを有すると考えられた。
〔材料と方法〕
抗マウスTLR9モノクローナル抗体の樹立
マウスTLR9に対するモノクローナル抗体(mAb)を樹立するために、BALB/c TLR9-/-マウスを、マウスTLR9を発現するBa/F3細胞(Ba/F3_mTLR9)で免疫した。最後の免疫の4日後、脾臓細胞をSP2/O骨髄腫細胞と融合させた。抗マウスTLR9 mAbを産生するハイブリドーマを、フローサイトメトリーにより、免疫に用いたBa/F3細胞の染色によって選択した。3つのモノクローナル抗体J15A7(IgG1/κ)、C34A1(IgG2a/κ)、及びB33A4(IgG2a/κ)が樹立された。
BM細胞及び脾臓細胞の染色のため、細胞は抗CD16/CD32抗体(clone:93)とプレインキュベートし、蛍光物質で標識した以下のマーカーに特異的な抗体を用いて染色し、フローサイトメトリーを行った:CD4(GK1.5)、CD8α(53-6.7)、CD11c(HL3)、B220(RA3-6B2)、PDCA-1(927, 129c1)。
脾臓細胞の単一細胞懸濁液をShibataらの方法に従って調製した(Shibata, T., et al. Int Immunol 2012)。細胞をビオチン化mAbと4℃でインキュベートし、染色用緩衝液(1 × PBS、2.5% FBS及び0.1% NaN3)で希釈した後、ストレプトアビジン結合抗体とインキュベートした。細胞内の染色のため、細胞は、染色用緩衝液中の0.05%又は0.1%のサポニン(Sigma-Aidrich (S5421)又はWAKO (193-00162))で透過処理し、続く工程もサポニン緩衝液中で行った。染色された細胞は、FACSCallibur又はFACSAriフローサイトメーター(BD Biosciences)で解析した。
細胞を溶解用緩衝液(1% Digitonin, 20mM Tris-HCl (pH 7.4), 150mM NaCl, 1mM CaCl2, 1mM MgCl2, 10% Glycerol, 1mM DTT 及びComplete protease Inhibitor Cocktail (Roche))で洗浄及び溶解した。氷上でインキュベートした後、溶解物を遠心処理し、不純物を除去した。免疫沈降のために、抗HA matrix(Roche clone: 3F10)又はN-ヒドロキシスクシンイミドで活性化した抗マウスTLR9mAb結合セファロース4FFビーズを、細胞溶解物に加え、4℃でインキュベートした。0.1%ジギトニンでビーズを3回洗浄し、結合したタンパク質を沸騰したSDSサンプル緩衝液(125 mM Tris/HCl [pH 6.8], 20% glycerol, 4% SDS, 10% 2-ME, 0.005% bromophenol blue)中で溶出させた。電気泳動後、サンプルをPVDF膜に移行し、イムノブロッティングに供した。
TLR9N又はTLR9C特異的な抗TLR9モノクローナル抗体の樹立
内因性TLR9の研究のため、TLR9に対する3つのモノクローナル抗体を樹立した。モノクローナル抗体の特異性は、TLR3、7、8及び9を発現するBa/F3細胞の膜透過性染色によって確認した。抗マウスTLR9抗体は、マウスTLR9と特異的に反応し、ヒトTLR9に対する交差反応性はなかった(図7A)。
DNAセンシングにおけるTLR9のエクトドメインのタンパク質切断を考慮すると、これらのモノクローナル抗体のエピトープがTLR9NとTLR9Cのいずれであるかを決定することが重要であることから、まず、TLR9の切断部位を決定した。M12B細胞リンパ腫でTLR9-GFPを発現させ、PRAT41の発現を抑制したM12細胞で発現させた場合と比較した。PRAT4Aは、TLR特異的シャペロンであり、PRAT4A非存在下では、TLR9は、小胞体から出ることができないため、プロセシングを受けない。したがって、PRAT4A発現抑制M12細胞では見られない断片が、TLR9Cである可能性が高いと考えた(図7B)。
検出されたTLR9C断片を精製し、N末端アミノ酸配列を決定した。TLR9Cは、461T又は467Fから始まることがわかった(図7C)。決定した切断部位は、ロイシンリッチリピート14(LRR14)及びLRR15の間の領域であり(図7D)、過去の報告に基づく予想と一致した。N末端が2種類存在することは、TLR9がアスパラギンエンドペプチダーゼとカテプシンによって順次プロセシングを受けることにより説明される。このN末端アミノ酸配列にしたがって、N末端HAエピトープ、TLR9エクトドメイン、及び膜透過ドメインからなるキメラタンパク質をBa/F3細胞で発現させた。樹立した細胞を抗TLR抗体で染色した。2つのモノクローナル抗体J15A7及びB33A4はTLR9Nと反応したが、C34A1はTLR9Cに結合した(図7E)。J15A7のTLR9Nへの結合は、B33A4の結合よりもかなり弱かった。
TLR9は、細胞表面から分泌されると考えられていたが、一次免疫細胞表面で内因性TLR9が発現していることは、これまで報告されていなかったが、抗TLR9抗体を用いて、細胞表面のTLR9発現を調べた。
まず、Ba/F3細胞でTLR9を過剰発現させたところ、予想通り、TLR9発現Ba/F3細胞表面にTLR9は検出されず、膜透過性染色によってエンドソームのTLR9が検出されただけであった(図8A、上段)。Unc93B1は、TLR9を小胞体からエンドリソソームに輸送する。Unc93B1がTLR9を細胞表面にも輸送している可能性を考えて、TLR9を発現するBa/F3細胞でUnc93B1も過剰発現させた。J15A7とC34A1は細胞表面TLR9を検出したが、抗TLR9NのB33A4は検出しなかった(図8A、下段)。J15A7は、脾臓由来pDC、CD8α+ cDC、CD4+ cDC、及びCD4-CD8-cDCにおいても、細胞表面TLR9を検出した。J15A7染色の特異性は、Tlr9-/-DCで確認した。Unc93B1遺伝子に機能喪失型の変異を有するUnc93b13d/3dマウスの脾臓由来DCでは、細胞表面TLR9は検出されなかった。Unc93B1の過剰発現が、Ba/F3細胞における細胞表面TLR9の発現を誘導することを示すこれらの結果から、Unc93B1はTLR9の細胞表面への輸送において重要な役割を果たすことがわかった。

Claims (17)
- 抗TLR7抗体又は抗TLR9抗体を含む、炎症性疾患の治療又は予防剤。
- 前記抗TLR7抗体が、以下の少なくとも1つを含む、請求項1に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:3で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:4で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:5で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:6で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:7で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:8で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:3~8のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:3~8のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR。 - 前記TLR7抗体が、以下のいずれかである、請求項1に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:9で表されるアミノ酸配列を含む重鎖と、配列番号:10で表されるアミノ酸配列を含む軽鎖とを含む抗体;
(2) 配列番号:9及び/又は10で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:9及び/又は10で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。 - 前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列のうち、27位~457位の領域内に特異的に結合する、請求項1に記載の炎症性疾患の治療剤又は予防剤。
- 前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の228位~364位の領域内に特異的に結合する、請求項1に記載の炎症性疾患の治療剤又は予防剤。
- 前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の275位~313位の領域を含む領域に特異的に結合する、請求項1に記載の炎症性疾患の治療剤又は予防剤。
- 前記抗TLR7抗体が、配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の313位~364位の領域を含む領域に特異的に結合する、請求項1に記載の炎症性疾患の治療剤又は予防剤。
- 前記抗TLR9抗体が、以下の少なくとも1つを含む、請求項1に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:11で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:12で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:13で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:14で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:15で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:16で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:11~16のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:11~16のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR。 - 前記抗TLR9抗体が、以下の少なくとも1つを含む、請求項1に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:17で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:18で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:19で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:20で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:21で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:22で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:17~22のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:17~22のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR。 - 前記抗TLR9抗体が、以下の少なくとも1つを含む、請求項1に記載の炎症性疾患の治療又は予防剤:
(a) 配列番号:23で表されるアミノ酸配列からなる重鎖CDR1;
(b) 配列番号:24で表されるアミノ酸配列からなる重鎖CDR2;
(c) 配列番号:25で表されるアミノ酸配列からなる重鎖CDR3;
(d) 配列番号:26で表されるアミノ酸配列からなる軽鎖CDR1;
(e) 配列番号:27で表されるアミノ酸配列からなる軽鎖CDR2;
(f) 配列番号:28で表されるアミノ酸配列からなる軽鎖CDR3;
(g) 配列番号:23~28のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び
(h) 配列番号:23~28のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR。 - 前記TLR9抗体が、以下のいずれかである、請求項1に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:29で表されるアミノ酸配列を含む重鎖と、配列番号:30で表されるアミノ酸配列を含む軽鎖とを含む抗体;
(2) 配列番号:29及び/又は30で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:29及び/又は30で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。 - 前記TLR9抗体が、以下のいずれかである、請求項1に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:31で表されるアミノ酸配列を含む重鎖と、配列番号:32で表されるアミノ酸配列を含む軽鎖とを含む抗体;
(2) 配列番号:31及び/又は32で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び
(3) 配列番号:31及び/又は32で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。 - 前記TLR9抗体が、以下のいずれかである、請求項1に記載の炎症性疾患の治療又は予防剤:
(1) 配列番号:33で表されるアミノ酸配列を含む重鎖と、配列番号:34で表されるアミノ酸配列を含む軽鎖とを含む抗体、
(2) 配列番号:33及び/又は34で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体、及び
(3) 配列番号:33及び/又は34で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。 - 前記炎症性疾患が、自己免疫疾患である、請求項1から13のいずれか1項に記載の炎症性疾患の治療又は予防剤。
- 前記自己免疫疾患が、全身性エリテマトーデス、又は乾癬である、請求項14に記載の炎症性疾患の治療又は予防剤。
- 以下のいずれかの抗TLR7抗体:
(i) 以下の少なくとも1つを含む抗体:(a) 配列番号:3で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:4で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:5で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:6で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:7で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:8で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:3~8のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:3~8のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
(ii) 以下のいずれかの抗体:(1) 配列番号:9で表されるアミノ酸配列を含む重鎖と、配列番号:10で表されるアミノ酸配列を含む軽鎖とを含む抗体;(2) 配列番号:9及び/又は10で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び(3) 配列番号:9及び/又は10で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
(iii)配列番号:1で示されるTLR7のアミノ酸配列のうち、27位~457位の領域内に特異的に結合する抗体;
(iv)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の228位~364位の領域内に特異的に結合する抗体;
(v)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の275位~313位の領域を含む領域に特異的に結合する抗体;又は
(vi)配列番号:1で示されるマウスTLR7のアミノ酸配列、又は配列番号:39で示されるヒトTLR7のアミノ酸配列の313位~364位の領域を含む領域に特異的に結合する抗体。 - 以下のいずれかのTLR9抗体:
(i) 以下の少なくとも1つを含む抗体:(a) 配列番号:11で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:12で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:13で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:14で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:15で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:16で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:11~16のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:11~16のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
(ii) 以下の少なくとも1つを含む抗体:(a) 配列番号:17で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:18で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:19で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:20で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:21で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:22で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:17~22のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:17~22のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
(iii) 以下の少なくとも1つを含む抗体:(a) 配列番号:23で表されるアミノ酸配列からなる重鎖CDR1;(b) 配列番号:24で表されるアミノ酸配列からなる重鎖CDR2;(c) 配列番号:25で表されるアミノ酸配列からなる重鎖CDR3;(d) 配列番号:26で表されるアミノ酸配列からなる軽鎖CDR1;(e) 配列番号:27で表されるアミノ酸配列からなる軽鎖CDR2;(f) 配列番号:28で表されるアミノ酸配列からなる軽鎖CDR3;(g) 配列番号:23~28のいずれかのアミノ酸配列において、1又は2のアミノ酸の欠失、置換又は付加を有するアミノ酸配列からなるCDR;及び(h) 配列番号:23~28のいずれかのアミノ酸配列と90%以上の同一性を有するアミノ酸配列からなるCDR;
(iv) 以下のいずれかの抗体:(1) 配列番号:29で表されるアミノ酸配列を含む重鎖と、配列番号:30で表されるアミノ酸配列を含む軽鎖とを含む抗体;(2) 配列番号:29及び/又は30で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び(3) 配列番号:29及び/又は30で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;
(v) 以下のいずれかの抗体:(1) 配列番号:31で表されるアミノ酸配列を含む重鎖と、配列番号:32で表されるアミノ酸配列を含む軽鎖とを含む抗体;(2) 配列番号:31及び/又は32で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体;及び(3) 配列番号:31及び/又は32で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体;又は
(vi) 以下のいずれかの抗体: (1) 配列番号:33で表されるアミノ酸配列を含む重鎖と、配列番号:34で表されるアミノ酸配列を含む軽鎖とを含む抗体、(2) 配列番号:33及び/又は34で表されるアミノ酸配列において、1又は数個のアミノ酸の欠失、置換又は付加を有するアミノ酸配列を含む重鎖及び/又は軽鎖を含む抗体、及び(3) 配列番号:33及び/又は34で表されるアミノ酸配列と70%以上の同一性を有するアミノ酸配列を重鎖及び/又は軽鎖を含む抗体。
