また、式(I)の化合物には、不斉炭素原子や軸不斉を有する場合があり、これに基づく光学異性体が存在しうる。本発明は、式(I)の化合物の光学異性体の分離されたもの、あるいはそれらの混合物も包含する。
式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P. G. M. Wuts)及びグリーン(T. W. Greene)著、「Greene's Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行なったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。
なお、R1'中に置換基として-CO2-tert-ブチル基が存在する場合、当該tert-ブチル基は、本工程で同時に脱保護される。
上記製造法における原料化合物は、例えば下記の方法、後述の製造例に記載の方法、公知の方法、あるいはそれらの変法を用いて製造することができる。
本工程は、化合物2aと化合物2bとのアミド化により化合物2cを得る工程である。
本工程は、化合物2aと化合物2bとを当量若しくは一方を過剰量用い、これらの混合物を縮合剤の存在下、反応に不活性な溶媒中、冷却下から加熱還流下、好ましくは-20℃~60℃において、通常0.1時間~5日間撹拌することによって行われる。ここで用いられる溶媒の例としては、特に限定はされないが、ベンゼン、トルエン、キシレン等の芳香族炭化水素類、ジクロロメタン、1,2-ジクロロエタン、クロロホルム等のハロゲン化炭化水素類、ジエチルエーテル、テトラヒドロフラン、1,4-ジオキサン、ジメトキシエタン等のエーテル類、N,N-ジメチルホルムアミド(DMF)、ジメチルスルホキシド(DMSO)、酢酸エチル、アセトニトリル、水、あるいはこれらの混合物が挙げられる。縮合剤の例としては、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド塩酸塩、ジシクロヘキシルカルボジイミド、1,1'-カルボニルビス-1H-イミダゾール、ジフェニルリン酸アジド、オキシ塩化リン、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(HATU)、(1-シアノ-2-エトキシ-2-オキソエチリデンアミノオキシ)ジメチルアミノ-モルホリノ-カルベニウム ヘキサフルオロホスファート(COMU)等が挙げられるが、これらに限定されるものではない。1H-ベンゾトリアゾール-1-オール等の添加剤を用いることが反応の進行に好ましい場合もある。また、トリエチルアミン、N,N-ジイソプロピルエチルアミン、N-メチルモルホリン、ジメチルアミノピリジン、ピリジン等の有機塩基、又は炭酸カリウム、炭酸ナトリウム、水酸化カリウム等の無機塩基を用いることが反応の進行に好ましい場合もある。
本工程は、化合物2cと化合物2dとのエステル化により化合物2eを得る工程である。
本工程は、原料合成1の第一工程に準じた方法を用いることができる。
本工程は、化合物2eのR4がベンジル基である場合に、ベンジル基を脱保護した化合物2fを得る工程である。
本工程は、水素雰囲気下、反応に不活性な溶媒中、化合物2eを金属触媒存在下で、通常1時間~5日間撹拌する。この反応は、通常、冷却下から加熱下、好ましくは室温で行われる。ここで用いられる溶媒の例としては、特に限定されないが、メタノール、エタノール、2-プロパノール等のアルコール類、ジエチルエーテル、テトラヒドロフラン、1,4-ジオキサン、ジメトキシエタン等のエーテル類、水、酢酸エチル、N,N-ジメチルホルムアミド、ジメチルスルホキシド及びこれらの混合物が挙げられる。金属触媒としては、パラジウム炭素、パラジウム黒、水酸化パラジウム等のパラジウム触媒、酸化白金等の白金触媒、還元ニッケル、ラネーニッケル等のニッケル触媒、トリス(トリフェニルホスフィン)クロロロジウム等のロジウム触媒が好適に用いられる。水素ガスの代わりに、化合物2eに対し等量から過剰量のギ酸またはギ酸アンモニウムを水素源として用いることもできる。

単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行なわれる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
1.ヒト及びマウストリプシン阻害活性の確認
実験にはヒトリコンビナントトリプシン(rh-trypsin ; 和光純薬工業、カタログ番号206-17171)及びマウストリプシン(m-trypsin ; マウス小腸内容物より自社にて精製)を使用した。マウス小腸内容物からのm-trypsinの精製法を以下に示す。
マウス10匹分の小腸内容物及び消化管をリン酸緩衝生理食塩水(PBS)中でポリトロンを用いてホモジナイズし、15,000×g、10分間の遠心処理を数回行った。その上清を、Benzamidine Sepharose 4 Fast Flow樹脂(GE Healthcare:#17-5123-10)に4℃で16時間混和した。PBSで樹脂を洗浄した後、グリシン緩衝液(pH3.0)でm-trypsinを溶出し精製を行った。得られた精製画分は抗マウスTrypsin抗体(Santa Cruz社:sc-67388)で認識されることをウェスタンブロット解析にて確認した。トリプシン阻害活性測定法は以下に示す。
化合物をジメチルスルホキシド(DMSO)で溶解し、任意の濃度に希釈した(A)。AをBuffer(0.1M Tris(pH8.0)、0.15M NaCl、10mM CaCl2、0.05% Brij35)で100倍希釈した(B)。rh-trypsinは0.088μg/mLとなるよう、m-trypsinは上記精製画分液を上記Bufferで50倍希釈した(C)。m-trypsinの希釈倍率はカイネティクス解析により0.088μg/mL rh-trypsinと同じ活性を示すように設定した。トリプシン酵素の基質であるBZiPAR溶液(Rhodamine標識基質)は5μmol/Lになるよう上記Bufferで希釈した(D)。B;5μL、C;5μL、D;10μLを384穴プレートに添加し、室温にて30分インキュベーションした。蛍光シグナルはTecan Safire Fluorometerを使用して最大励起(Ex)/蛍光波長(Em)=497nm/520nmにて検出した。化合物は2500, 750, 250, 75, 25, 7.5, 2.5, 0.75, 0.25, 0.075, 0.025, 0.0075nMで検討し、酵素存在下で化合物未添加時(DMSOのみ)を0%阻害、酵素非存在下で化合物未添加時を100%阻害とし、各化合物の阻害率を算出した。得られた阻害率を基に非線形回帰からトリプシン阻害活性(IC50値,nM)を算出した。
実験には6週齢の雄性ICRマウスを用い、1群5例にて実施した。マウスを15時間絶食後、対照群は0.5%メチルセルロース溶液(MC)を、被験薬群は化合物(5mg/kg)を0.5%MCに溶解又は懸濁し強制経口投与した。その直後から絶食を解除し自由摂餌(CE-2)を開始させ、3時間後から9時間後までの糞を回収し重量を測定した。得られた糞は6mLの蒸留水で懸濁し、1,940×gで10分間遠心分離した。得られた上清中の蛋白質濃度をブラッドフォード法にて測定し、糞中蛋白質量を糞重量で除することで糞1g中に含まれる糞中蛋白質濃度として算出し、対照群に対する比から薬効を検討した。複数回評価した化合物については、平均値を算出した。
10週齢の雄性Wistarラットに左腎摘出術を行い、その1週後にDXR(5mg/kg)を尾静脈内投与することにより腎症モデルを作製した。1群10例にて試験を実施し、化合物投与期間中は昼間のみ給餌を行い、夜間は絶食とした。被験化合物(10mg/kg)はモデル作製翌日から0.5%メチルセルロース溶液(MC)に懸濁し1日2回(朝及び夜)強制経口投与した。sham(正常)群、control群には0.5% MCを強制経口投与した。被験化合物投与開始から1、2、3週間後にそれぞれ24時間採尿を行い、尿中蛋白排泄量を測定した。3週目の採尿終了後に採血を行い、血漿中クレアチニン濃度を測定した。
本試験の結果より、当該化合物は尿中蛋白排泄及び血漿中クレアチニン濃度の低下作用を示し、腎症の進行が抑制されることを確認した。
投与は錠剤、丸剤、カプセル剤、顆粒剤、散剤、液剤等による経口投与、又は、関節内、静脈内、筋肉内等の注射剤、坐剤、点眼剤、眼軟膏、経皮用液剤、軟膏剤、経皮用貼付剤、経粘膜液剤、経粘膜貼付剤、吸入剤等による非経口投与のいずれの形態であってもよい。
経口投与のための液体組成物は、薬剤的に許容される乳濁剤、溶液剤、懸濁剤、シロップ剤又はエリキシル剤等を含み、一般的に用いられる不活性な希釈剤、例えば精製水又はエタノールを含む。当該液体組成物は不活性な希釈剤以外に可溶化剤、湿潤剤、懸濁剤のような補助剤、甘味剤、風味剤、芳香剤、防腐剤を含有していてもよい。
tert-ブチル 4-メチルチオフェン-2-カルボキシラート(12.0g)、N-ブロモスクシンイミド(10.8g)、2,2'-アゾビス(イソブチロニトリル)(496mg)、及び四塩化炭素(119mL)の混合物を90℃で1時間撹拌した。さらにN-ブロモスクシンイミド(1.08g)を加え、90℃で1時間撹拌した。反応懸濁液を室温まで冷却したのち、不溶物を濾別し、濾液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 4-(ブロモメチル)チオフェン-2-カルボキシラート(16.3g)を得た。
tert-ブチル 4-(ブロモメチル)チオフェン-2-カルボキシラート(9.90g)のN,N-ジメチルホルムアミド(100mL)溶液にtert-ブチル グリシナート 塩酸塩(18.0g)、及びトリエチルアミン(19.9mL)を加え、60℃で15時間撹拌した。反応懸濁液を室温まで冷却した後、トリアセトキシ水素化ホウ素ナトリウム(22.7g)を加え、室温で5時間撹拌した。反応懸濁液に水、炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出した。有機層を5%塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(5.67g)を得た。
6-ヒドロキシ-2-ナフトエ酸(220mg)のN,N-ジメチルホルムアミド(3.30mL)溶液にtert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(383mg)、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(489mg)、及びN,N-ジイソプロピルエチルアミン(500μL)を加え、室温で20時間撹拌した。さらにO-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(222mg)、及びN,N-ジイソプロピルエチルアミン(200μL)を加え、室温で6時間撹拌した。反応溶液に水を加え、酢酸エチルにて抽出した。有機層を水、飽和塩化ナトリウム水溶液の順に洗浄した後、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)(6-ヒドロキシ-2-ナフトイル)アミノ]メチル}チオフェン-2-カルボキシラート(277mg)を得た。
4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(194mg)のジクロロメタン(7.29mL)溶液にN-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(118mg)、tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)(6-ヒドロキシ-2-ナフトイル)アミノ]メチル}チオフェン-2-カルボキシラート(255mg)、及び4-ジメチルアミノピリジン(18.8mg)を加え、室温で24時間撹拌した。反応液に水を加え、クロロホルムで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-({[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-ナフトイル](2-tert-ブトキシ-2-オキソエチル)アミノ}メチル)チオフェン-2-カルボキシラート(137mg)を得た。
6-メトキシ-1-ベンゾチオフェン-2-カルボン酸(960mg)のジクロロメタン(5.76mL)溶液に氷冷下1M三臭化ホウ素のジクロロメタン溶液(37.5mL)を10分間かけて滴下した後、室温で3時間撹拌した。反応液を氷に滴下した後、撹拌した。析出物をろ取し、減圧下乾燥して6-ヒドロキシ-1-ベンゾチオフェン-2-カルボン酸(920mg)を得た。
2-tert-ブチル 3,7-ジメチル (3R)-3,4-ジヒドロイソキノリン-2,3,7(1H)-トリカルボキシラート(345mg)のメタノール(7.00mL)溶液に1M水酸化ナトリウム水溶液(3.50mL)を加え、室温で3時間撹拌した。1M塩酸(3.50mL)を加え中和した後、水を加え酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮して、(3R)-2-(tert-ブトキシカルボニル)-1,2,3,4-テトラヒドロイソキノリン-3,7-ジカルボン酸(328mg)を得た。
(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)酢酸(1.03g)のN,N-ジメチルホルムアミド(20.5mL)溶液にtert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート 塩酸塩(1.96g)、トリエチルアミン(762μL)、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(1.00g)、及び1H-ベンゾトリアゾール-1-オール(705mg)を添加し、室温で13時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を水、飽和塩化ナトリウム水溶液で順次洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製し、tert-ブチル 3-({(2-tert-ブトキシ-2-オキソエチル)[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセチル]アミノ}メチル)ベンゾアート(2.41g)を得た。
窒素雰囲気下、tert-ブチル 3-(クロロメチル)ベンゾアート(29.1g)のN,N-ジメチルホルムアミド(300mL)溶液にtert-ブチル グリシナート 塩酸塩(43.0g)、及びトリエチルアミン(71.6mL)を加え、60-63℃で3時間撹拌した。反応混合物を氷冷した後、水を加え酢酸エチルで抽出した。有機層を10%塩化アンモニウム水溶液、20%塩化ナトリウム水溶液で順次洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣を酢酸エチル(400mL)に溶解した後、窒素雰囲気下氷冷中4M塩化水素の酢酸エチル溶液(32.1mL)を滴下し1時間撹拌した。析出物をろ取し、酢酸エチルで洗浄した後、50℃で減圧下乾燥し、tert-ブチル 3-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート 塩酸塩(28.5g)を得た。
エチル (6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセタート(2.08g)及び48%臭化水素酸(40.0mL)の混合物を、120℃で17時間撹拌した。反応混合物を放冷した後、減圧下濃縮した。残渣にテトラヒドロフラン(100mL)を加え、室温で1時間撹拌した後、析出物をろ取した。ろ液を減圧下濃縮した後、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製した。精製物にジイソプロピルエーテル(15.0mL)を加えた後、室温で1時間撹拌した。析出物をろ取し、ジイソプロピルエーテルにて洗浄した後、室温で減圧下乾燥し、(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)酢酸(1.06g)を得た。
4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(1.97g)のジクロロメタン(48.0mL)溶液にtert-ブチル 3-({(2-tert-ブトキシ-2-オキソエチル)[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセチル]アミノ}メチル)ベンゾアート(2.40g)、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(1.17g)、及び4-ジメチルアミノピリジン(173mg)を添加し、室温で2時間撹拌した。さらに4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(179mg)、及びN-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(100mg)を加え、室温で1時間撹拌した。反応混合物を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 3-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(3.71g)を得た。

3-[(ベンジル{[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]ペンタン二酸(886mg)、N,N-ジメチルホルムアミド ジ-tert-ブチル アセタール(1.60mL)、及びトルエン(4.43mL)の混合物を80℃で4時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、ジ-tert-ブチル 3-[(ベンジル{[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]ペンタンジオアート(232mg)を得た。
3,7-ジベンジル 2-tert-ブチル (3R)-3,4-ジヒドロイソキノリン-2,3,7(1H)-トリカルボキシラート(413mg)に4M塩化水素の1,4-ジオキサン溶液(4.00mL)を加え、室温で終夜撹拌した。反応懸濁液を減圧下濃縮し、残渣を減圧下乾燥し、ジベンジル (3R)-1,2,3,4-テトラヒドロイソキノリン-3,7-ジカルボキシラート 塩酸塩(358mg)を得た。
6-ヒドロキシ-1-ベンゾチオフェン-3-カルボン酸(100mg)、tert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(186mg)、及びN,N-ジイソプロピルエチルアミン(88.2μL)のN,N-ジメチルホルムアミド(3.00mL)溶液に(1-シアノ-2-エトキシ-2-オキソエチリデンアミノオキシ)ジメチルアミノ-モルホリノ-カルベニウム ヘキサフルオロホスファート(243mg)を加え、室温で16時間撹拌した。反応混合物を酢酸エチルにて希釈し、有機層を水、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-({(2-tert-ブトキシ-2-オキソエチル)[(6-ヒドロキシ-1-ベンゾチオフェン-3-イル)カルボニル]アミノ}メチル)チオフェン-2-カルボキシラート(230mg)を得た。
tert-ブチル [4-(アミノメチル)フェニル]アセタート(1.00g)のアセトニトリル(15.0mL)溶液にトリエチルアミン(693μL)、及びtert-ブチル ブロモアセタート(668μL)を加え、室温で4時間撹拌した。反応液を減圧下濃縮した後、酢酸エチルを加えた。有機層を0.1M塩酸、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-[4-(2-tert-ブトキシ-2-オキソエチル)ベンジル]グリシナート(747mg)を得た。
6-ヒドロキシ-1-ナフトエ酸(190mg)のN,N-ジメチルホルムアミド(2.85mL)溶液にtert-ブチル 4-({[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ}メチル)チオフェン-2-カルボキシラート(345mg)、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(422mg)、及びN,N-ジイソプロピルエチルアミン(190μL)を加え、50℃で19時間撹拌した。反応溶液に水を加え、酢酸エチルにて抽出した。有機層を水、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-({[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル](6-ヒドロキシ-1-ナフトイル)アミノ}メチル)チオフェン-2-カルボキシラート(101mg)を得た。
N-[(ベンジルオキシ)カルボニル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニン(500mg)のテトラヒドロフラン(4.00mL)、及びtert-ブチルアルコール(4.00mL)の溶液に、ジ-tert-ブチルジ炭酸エステル(656mg)、及び4-ジメチルアミノピリジン(30.6mg)を加え、室温で終夜撹拌した。反応溶液に水を加え、酢酸エチルで抽出した。有機層を水、飽和炭酸水素ナトリウム水溶液、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮して、tert-ブチル N-[(ベンジルオキシ)カルボニル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(641mg)を得た。
(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)酢酸(150mg)、tert-ブチル N-(2-tert-ブトキシ-2-オキソエチル)-L-フェニルアラニナート(293mg)、及びN,N-ジイソプロピルエチルアミン(147μL)のN,N-ジメチルホルムアミド(4.50mL)溶液に(1-シアノ-2-エトキシ-2-オキソエチリデンアミノオキシ)ジメチルアミノ-モルホリノ-カルベニウム ヘキサフルオロホスファート(368mg)を加え、60℃で8時間撹拌した。反応混合物を酢酸エチルにて希釈し、有機層を水、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-(2-tert-ブトキシ-2-オキソエチル)-N-[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセチル]-L-フェニルアラニナート(347mg)を得た。
5-メチル-1-ベンゾチオフェン-2-カルボン酸(1.50g)のN,N-ジメチルホルムアミド(10.5mL)溶液に1,1'-カルボニルビス-1H-イミダゾール(1.27g)を加え、室温で2時間30分間撹拌した。反応混合物にtert-ブチルアルコール(1.44mL)、及び1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン(1.17mL)を加え、50℃で24時間撹拌した。反応混合物を酢酸エチルにて希釈した後、有機層を0.1M塩酸、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 5-メチル-1-ベンゾチオフェン-2-カルボキシラート(1.78g)を得た。
tert-ブチル 5-メチル-1-ベンゾチオフェン-2-カルボキシラート(1.77g)、四塩化炭素(17.7mL)、及びN-ブロモスクシンイミド(2.16g)の混合物に2,2'-アゾビス(イソブチロニトリル)(58.5mg)を加え90℃で終夜撹拌した。反応懸濁液をろ過し、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン‐酢酸エチル)にて精製し、tert-ブチル 5-(ブロモメチル)-1-ベンゾチオフェン-2-カルボキシラート(1.51g)を得た。
tert-ブチル 5-(ブロモメチル)-1-ベンゾチオフェン-2-カルボキシラート(1.00g)のN,N-ジメチルホルムアミド(10.0mL)溶液にtert-ブチル グリシナート 塩酸塩(1.02g)、及びトリエチルアミン(1.70mL)を加え、85-95℃で終夜撹拌した。反応混合物を室温まで冷却した後、酢酸エチルで抽出した。有機層を25%塩化アンモニウム水溶液で洗浄後、無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 5-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}-1-ベンゾチオフェン-2-カルボキシラート(359mg)を得た。
2-tert-ブチル 3-メチル (3R)-7-ヒドロキシ-3,4-ジヒドロイソキノリン-2,3(1H)-ジカルボキシラート(1.00g)のジクロロメタン(20.0mL)溶液に氷冷下トリフルオロメタンスルホン酸無水物(770μL)、及び2,6-ジメチルピリジン(800μL)を加え、氷冷下2時間撹拌した。反応混合物に水を加え、クロロホルムで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣にN,N-ジメチルホルムアミド(15.0mL)、メタノール(3.00mL)、酢酸パラジウム(II)(37.0mg)、1,1'-ビス(ジフェニルホスフィノ)フェロセン(90.0mg)、及びトリエチルアミン(1.10mL)を加え、一酸化炭素雰囲気下80℃で終夜撹拌した。反応混合物を室温まで冷却後、減圧下濃縮した。残渣に水を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄し、無水硫酸ナトリウムにて乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、2-tert-ブチル 3,7-ジメチル (3R)-3,4-ジヒドロイソキノリン-2,3,7(1H)-トリカルボキシラート(350mg)を得た。
(3R)-2-(tert-ブトキシカルボニル)-1,2,3,4-テトラヒドロイソキノリン-3,7-ジカルボン酸(320mg)のN,N-ジメチルホルムアミド(6.00mL)溶液に炭酸カリウム(315mg)、及び臭化ベンジル(275μL)を加え、室温で終夜撹拌した。反応懸濁液に水を加え、酢酸エチルにて抽出した。有機層を水、飽和塩化ナトリウム水溶液で順次洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、3,7-ジベンジル 2-tert-ブチル (3R)-3,4-ジヒドロイソキノリン-2,3,7(1H)-トリカルボキシラート(415mg)を得た。
2-ブロモ-1,3,5-トリメチルベンゼン(925μL)のテトラヒドロフラン(20.0mL)溶液に、-78℃下1.59M n-ブチルリチウムのヘキサン溶液(3.86mL)を滴下し、-78℃で30分間撹拌した。反応混合物にtert-ブチル (4-ブロモ-2-チエニル)アセタート(1.55g)のテトラヒドロフラン(15.0mL)溶液を滴下し、-78℃で30分間撹拌した。続いて反応混合物に1.59M n-ブチルリチウムのヘキサン溶液(3.51mL)を滴下し、-78℃で30分間撹拌した。反応混合物にさらにN,N-ジメチルホルムアミド(451μL)を滴下し、-78℃で1時間撹拌した。反応液に塩化アンモニウム水溶液、酢酸エチルを加え、有機層を抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル (4-ホルミル-2-チエニル)アセタート(355mg)を得た。
tert-ブチル (4-ホルミル-2-チエニル)アセタート(350mg)、tert-ブチル グリシナート(243mg)、及び酢酸(265μL)のジクロロメタン(4.05mL)溶液に、氷冷下トリアセトキシ水素化ホウ素ナトリウム(656mg)を加え、室温で3時間撹拌した。反応混合物に飽和炭酸水素ナトリウム水溶液を加えて中和した後、クロロホルムで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-{[5-(2-tert-ブトキシ-2-オキソエチル)-3-チエニル]メチル}グリシナート(237mg)を得た。
tert-ブチル [3-(アミノメチル)フェニル]アセタート(1.00g)、tert-ブチル ブロモアセタート(700μL)、炭酸カリウム(650mg)、及びアセトニトリル(20.0mL)の混合物を室温で終夜撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-[3-(2-tert-ブトキシ-2-オキソエチル)ベンジル]グリシナート(1.14g)を得た。
N-ベンジル-N-(シクロペンタ-3-エン-1-イルメチル)-2-(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセトアミド(430mg)、炭酸カリウム(174mg)、及びN,N-ジメチルホルムアミド(4.30mL)の混合物に臭化ベンジル(177μL)を加え、50℃で6時間撹拌した。反応混合物を冷却した後、水を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、N-ベンジル-2-[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]-N-(シクロペンタ-3-エン-1-イルメチル)アセトアミド(416mg)を得た。
N-ベンジル-2-[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]-N-(シクロペンタ-3-エン-1-イルメチル)アセトアミド(100mg)、tert-ブチルアルコール(2.40mL)、及び水(600μL)の混合物に2.5%四酸化オスミウムのtert-ブチルアルコール溶液(269μL)、及び4-メチルモルホリン 4-オキシド(75.5mg)を加え、室温で2時間撹拌した。反応混合物にチオ硫酸ナトリウム水溶液を加え、酢酸エチルで抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮して、N-ベンジル-2-[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]-N-[(3,4-ジヒドロキシシクロペンチル)メチル]アセトアミド(107mg)を得た。
N-ベンジル-2-[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]-N-[(3,4-ジヒドロキシシクロペンチル)メチル]アセトアミド(106mg)、ヨードベンゼン ジアセタート(322mg)、ジクロロメタン(3.00mL)、及び水(1.00mL)の混合物に1-メチル-2-アザアダマンタン-N-オキシル(3.33mg)を加え、室温で3時間撹拌した。反応混合物に20%チオ硫酸ナトリウム水溶液を加え、室温で5分間撹拌した。続いて1M塩酸を加え、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムにて乾燥し、減圧下濃縮して、3-[(ベンジル{[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]ペンタン二酸(110mg)を得た。
[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体の(1S)-1-フェニルエタンアミン塩(370mg)、及び酢酸エチル(4.50mL)の混合物に3M塩酸(4.50mL)を加え、室温で2時間撹拌した。反応混合物を酢酸エチルにて抽出した。有機層を無水硫酸ナトリウムにて乾燥後、減圧下濃縮し、[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(243mg)を得た。
[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体の(1R)-1-フェニルエタンアミン塩(450mg)、及び酢酸エチル(5.51mL)の混合物に4M塩化水素の酢酸エチル溶液(4.13mL)を加え、室温で2時間撹拌した。反応混合物に水を加え、酢酸エチルにて抽出した。有機層を飽和塩化ナトリウム水溶液にて洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮して[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(290mg)を得た。
製造例29で得られた[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(238mg)のジクロロメタン(8.00mL)溶液に、氷冷下1M三臭化ホウ素のジクロロメタン溶液(2.20mL)を加え、室温で2時間撹拌した。反応混合物に氷水を加え、酢酸エチルにて抽出した。有機層を水で洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮して、[(1R)-6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(147mg)を得た。
製造例30で得られた[(1R)-6-メトキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(288mg)のジクロロメタン(9.77mL)溶液に氷冷下1M三臭化ホウ素のジクロロメタン溶液(2.66mL)を滴下した後、室温で2時間撹拌した。反応混合物に氷水を加え、酢酸エチルにて抽出した。有機層を飽和塩化ナトリウム水溶液で洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、[(1R)-6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(230mg)を得た。
製造例31で得られた[(1R)-6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]酢酸又はその鏡像異性体(145mg)のN,N-ジメチルホルムアミド(2.90mL)溶液にtert-ブチル 4-({[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ}メチル)チオフェン-2-カルボキシラート(255mg)、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(295mg)、及びN,N-ジイソプロピルエチルアミン(135μL)を加え、室温で終夜撹拌した。反応溶液に水を加え、酢酸エチルで抽出した。有機層を水で洗浄した後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-[([(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]{[(1R)-6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]チオフェン-2-カルボキシラート又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(237mg)を得た。

製造例35で得られたtert-ブチル 4-[([(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]{[(1R)-6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]チオフェン-2-カルボキシラート又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(328mg)、4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(282mg)、4-ジメチルアミノピリジン(22.7mg)、及びジクロロメタン(4.92mL)の混合物に、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(154mg)を加え、室温で4時間撹拌した。反応混合物を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-[({[(1R)-6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ)メチル]チオフェン-2-カルボキシラート又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(460mg)を得た。
tert-ブチル 3-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(395mg)をUVトリガーによる超臨界クロマトグラフィー法(二酸化炭素-メタノール)にて分取精製後、減圧下濃縮し、tert-ブチル 3-{[{[(1R)-6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート及びその鏡像異性体としてPEx.37-1(188mg、RT:4.02min)及びPEx.37-2(187mg、RT:4.87min)を得た。なお、RTを求める為に行った超臨界クロマトグラフィー法の分析条件を以下に示す。

6-ヒドロキシ-1-ナフトエ酸(150mg)、tert-ブチル 4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(200mg)、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(160mg)、1H-ベンゾトリアゾール-1-オール(110mg)、及びN,N-ジメチルホルムアミド(4.00mL)の混合物を室温で終夜撹拌した。反応混合物に水を加え、1時間撹拌した。析出物をろ取し、水で洗浄後、減圧下乾燥し、tert-ブチル 4-(tert-ブトキシカルボニル)-N-(6-ヒドロキシ-1-ナフトイル)-L-フェニルアラニナート(183mg)を得た。
tert-ブチル 4-(tert-ブトキシカルボニル)-N-(6-ヒドロキシ-1-ナフトイル)-L-フェニルアラニナート(180mg)、4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(180mg)、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(90.0mg)、4-ジメチルアミノピリジン(15.0mg)、及びジクロロメタン(2.00mL)の混合物を室温で2時間撹拌した。反応混合物をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル N-[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1-ナフトイル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(303mg)を得た。
6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-2-カルボン酸(200mg)のN,N-ジメチルホルムアミド(3.00mL)溶液にtert-ブチル 4-{[(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(341mg)、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N',N'-テトラメチルウロニウム ヘキサフルオロホスファート(435mg)、及びN,N-ジイソプロピルエチルアミン(196μL)を加え、室温で2時間撹拌した。反応混合物に水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-({(2-tert-ブトキシ-2-オキソエチル)[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-2-イル)カルボニル]アミノ}メチル)チオフェン-2-カルボキシラート(522mg)を得た。
tert-ブチル 4-({(2-tert-ブトキシ-2-オキソエチル)[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-2-イル)カルボニル]アミノ}メチル)チオフェン-2-カルボキシラート(520mg)のジクロロメタン(10.4mL)溶液に氷冷下4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]安息香酸(433mg)、N-[3-(ジメチルアミノ)プロピル]-N'-エチルカルボジイミド 塩酸塩(298mg)、及び4-ジメチルアミノピリジン(38.0mg)を添加し、室温で2時間撹拌した。反応溶液に水、1M塩酸を加えた後、クロロホルムで抽出した。有機層を無水硫酸マグネシウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、tert-ブチル 4-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-2-イル]カルボニル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(723mg)を得た。
(3R)-2-(tert-ブトキシカルボニル)-7-ヒドロキシ-1,2,3,4-テトラヒドロイソキノリン-3-カルボン酸(1.50g)のトルエン(60.0mL)、及びメタノール(9.00mL)の溶液に0.60M トリメチルシリルジアゾメタンのヘキサン溶液(10.0mL)を滴下した後、室温で30分間撹拌した。反応混合物に酢酸(300μL)を加えた後、炭酸水素ナトリウム水溶液を加え、酢酸エチルにて抽出した。有機層を水、飽和塩化ナトリウム水溶液にて順次洗浄後、無水硫酸ナトリウムにて乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル)にて精製して、2-tert-ブチル 3-メチル (3R)-7-ヒドロキシ-3,4-ジヒドロイソキノリン-2,3(1H)-ジカルボキシラート(1.47g)を得た。
ジベンジル (3R)-2-{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}-1,2,3,4-テトラヒドロイソキノリン-3,7-ジカルボキシラート(257mg)のエタノール(5.00mL)溶液に10% パラジウム-炭素(52.0mg、50%含水品)を加え、常圧、水素雰囲気下、室温で2時間撹拌した。反応混合物をセライト(登録商標)層を通してろ過した後、ろ液を減圧下濃縮して、(3R)-2-{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}-1,2,3,4-テトラヒドロイソキノリン-3,7-ジカルボン酸(213mg)を得た。
ジ-tert-ブチル 3-[(ベンジル{[6-(ベンジルオキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}アミノ)メチル]ペンタンジオアート(230mg)、10% パラジウム-炭素(38.1mg、50%含水品)、及びメタノール(4.60mL)の混合物を常圧、水素雰囲気下、室温で16時間撹拌した。反応混合物をセライト(登録商標)層を通してろ過した後、ろ液を減圧下濃縮して、ジ-tert-ブチル 3-({ベンジル[(6-ヒドロキシ-1,2,3,4-テトラヒドロナフタレン-1-イル)アセチル]アミノ}メチル)ペンタンジオアート(186mg)を得た。
tert-ブチル 4-({[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-2-ナフトイル](2-tert-ブトキシ-2-オキソエチル)アミノ}メチル)チオフェン-2-カルボキシラート(132mg)に4M塩化水素の1,4-ジオキサン溶液(2.02mL)を加え、室温で24時間撹拌した。反応懸濁液を減圧下濃縮した後、残渣をオクタデシルシリル(以下、ODSとする。)カラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、4-{[{6-[(4-カルバムイミドアミドベンゾイル)オキシ]-2-ナフトイル}(カルボキシメチル)アミノ]メチル}チオフェン-2-カルボン酸 一塩酸塩(62.7mg)を得た。
tert-ブチル 3-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート(528mg)のジクロロメタン(5.00mL)溶液にトリフルオロ酢酸(2.00mL)を加え、室温で2時間撹拌した。反応混合物を減圧下濃縮した後、残渣に1M塩酸(606μL)、及びアセトニトリル(10.0mL)を加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、減圧下乾燥して、3-{[({6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸 一塩酸塩(227mg)を得た。
tert-ブチル N-[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1-ナフトイル]-4-(tert-ブトキシカルボニル)-L-フェニルアラニナート(300mg)のジクロロメタン(1.50mL)溶液にトリフルオロ酢酸(1.50mL)を加え、室温で2時間撹拌した。反応混合物を減圧下濃縮した後、残渣に1M塩酸(1.00mL)、及びアセトニトリル(1.00mL)を加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、N-{6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1-ナフトイル}-4-カルボキシ-L-フェニルアラニン 一塩酸塩(158mg)を得た。
tert-ブチル 4-{[{[6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-2-イル]カルボニル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}チオフェン-2-カルボキシラート(720mg)のジクロロメタン(5.33mL)溶液にトリフルオロ酢酸(5.36mL)を加え、室温で5時間撹拌した。反応混合物にアセトニトリルを加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製して、4-{[({6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-2-イル}カルボニル)(カルボキシメチル)アミノ]メチル}チオフェン-2-カルボン酸 一塩酸塩(440mg)を得た。
製造例37で得られたtert-ブチル 3-{[{[(1R)-6-({4-[ N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート又はその鏡像異性体(PEx.37-1, 181mg)のジクロロメタン(3.00mL)溶液にトリフルオロ酢酸(686μL)を加え、室温で4時間撹拌した。反応溶液を減圧下濃縮した後、残渣に1M塩酸(208μL)、及びアセトニトリル(10.0mL)を加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、減圧下乾燥して、3-{[({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸又はその鏡像異性体の一塩酸塩(91mg, RT 10.6min)を得た。なお、RTを求める為に行ったキラルカラムクロマトグラフィー法の分析条件を以下に示す。

製造例37で得られたtert-ブチル 3-{[{[(1R)-6-({4-[ N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}(2-tert-ブトキシ-2-オキソエチル)アミノ]メチル}ベンゾアート又はその鏡像異性体(PEx.37-2, 183mg)のジクロロメタン(3.03mL)溶液にトリフルオロ酢酸(693μL)を加え、室温で4時間撹拌した。反応溶液を減圧下濃縮した後、残渣に1M塩酸(210μL)、及びアセトニトリル(10.0mL)を加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、減圧下乾燥して、3-{[({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸又はその鏡像異性体の一塩酸塩(98mg, RT 14.1min)を得た。なお、RTを求める為に行ったキラルカラムクロマトグラフィー法の分析条件を以下に示す。

製造例34で得られたtert-ブチル 4-[({[(1R)-6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ)メチル]チオフェン-2-カルボキシラート又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(324mg)のジクロロメタン(3.00mL)溶液にトリフルオロ酢酸(1.50mL)を加え、室温で終夜撹拌した。反応混合物を減圧下濃縮した後、残渣に1M塩酸(2.00mL)、及びアセトニトリル(1.50mL)を加え、減圧下濃縮した。残渣をODSカラムクロマトグラフィー(0.01M塩酸-アセトニトリル)にて精製し、4-({({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)[(1R)-1-カルボキシエチル]アミノ}メチル)チオフェン-2-カルボン酸又はその1,2,3,4-テトラヒドロナフタレン1位エピマーの塩酸塩(46mg)を得た。
製造例36で得られたtert-ブチル 4-[({[(1R)-6-({4-[N',N''-ビス(tert-ブトキシカルボニル)カルバムイミドアミド]ベンゾイル}オキシ)-1,2,3,4-テトラヒドロナフタレン-1-イル]アセチル}[(2R)-1-tert-ブトキシ-1-オキソプロパン-2-イル]アミノ)メチル]チオフェン-2-カルボキシラート又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(460mg)のジクロロメタン(3.07mL)溶液にトリフルオロ酢酸(2.06mL)を加え、室温で2時間撹拌した。反応混合物を減圧下濃縮した後、残渣に1M塩酸(588μL)、及びアセトニトリル(3.07mL)を加え、減圧下濃縮した。残渣に酢酸エチルを加え、減圧下濃縮した。得られた固体をアセトニトリルで洗浄し、4-({({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)[(1R)-1-カルボキシエチル]アミノ}メチル)チオフェン-2-カルボン酸又はその1,2,3,4-テトラヒドロナフタレン1位エピマー(44mg)を得た。
実施例6で得られた3-{[({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸又はその鏡像異性体の一塩酸塩(3.83g)の50%アセトニトリル水溶液(80.0mL)に対して、氷冷下1M水酸化ナトリウム水溶液(6.44mL)を滴下したのち、室温で3時間撹拌した。析出物を濾取したのち、50%アセトニトリル水溶液にて洗浄し、1時間通風乾燥した。乾燥物を50%アセトニトリル水溶液(400mL)に縣濁させたのち、120℃で30分間撹拌した。反応混合物を室温で12時間撹拌した。析出物を濾取したのち、50%アセトニトリル水溶液にて洗浄し、減圧下室温にて乾燥して3-{[({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸又はその鏡像異性体(2.89g)を得た。

- 式(I)の化合物又はその塩。
L1は結合又は-低級アルキレン-であり、
L2は置換されていてもよい低級アルキレンであり、
R1は置換されていてもよいアリール、置換されていてもよい芳香族へテロ環基及び-CO2Hからなる群より選択される置換基で置換されていてもよい低級アルキル又はHであり、あるいは、R1は結合する窒素原子及び窒素原子上のHO2C-L2基と一体となって-CO2Hで置換されていてもよい環状アミノを形成していてもよく、及び

- L2がメチレン又はメチルメチレンであり、R1が(-CO2Hで置換されたフェニル)-CH2-、(-CH2-CO2Hで置換されたフェニル)-CH2-又は(-CO2Hで置換されたチエニル)-CH2-である請求項8記載の化合物又はその塩。
- L1が結合又はメチレンであり、環Bがナフタレン-1,6-ジイル、ナフタレン-2,6-ジイル、1,2,3,4-テトラヒドロナフタレン-1,6-ジイル、1,2,3,4-テトラヒドロナフタレン-2,6-ジイル、2,3-ジヒドロインデン-1,5-ジイル、ベンゾチオフェン-3,6-ジイル、ベンゾフラン-3,6-ジイル又は2,3-ジヒドロベンゾフラン-3,6-ジイルであり、及び
a)L2がC1-3アルキレンであり、かつ、R1がi)-CO2H及び-CO2Hで置換された低級アルキルからなる群より選択される1以上の置換基で置換されていてもよいフェニル、及びii)-CO2H及び-CO2Hで置換された低級アルキルからなる群より選択される1以上の置換基で置換された、チエニル及びベンゾチエニルから選択される芳香族へテロ環基、からなる群より選択される1以上の置換基で置換され、更に1以上の-CO2Hで置換されていてもよい低級アルキルであるか、
b)L2が(-CO2Hで置換されたフェニル)で置換されたC1-3アルキレンであり、かつ、R1はHであるか、又は、
c)R1が結合する窒素原子及び窒素原子上のHO2C-L2基と一体となって、2個の-CO2Hで置換された1,2,3,4-テトラヒドロイソキノリン-2-イルを形成する、

3-{[({(1R)-6-[(4-カルバムイミドアミドベンゾイル)オキシ]-1,2,3,4-テトラヒドロナフタレン-1-イル}アセチル)(カルボキシメチル)アミノ]メチル}安息香酸、
