(タンパク質の精製)
(1)ADF3Kai-small-Mのタンパク質を発現している大腸菌の湿菌体約15gに、1MのLiClを含むDMSOを45ml添加し、60℃で30分間溶解させた。その後、遠心分離機(トミー精工社製の「MX-305」)にて、20℃、11000×g、10分間遠心分離し、上清(溶解液)を回収した。
(2)回収した溶解液を透析することにより、DMSOと水の溶媒置換を行った。透析液(水)は6時間置きに交換し、トータル48時間透析を行った。透析後の溶解液を遠心分離機(トミー精工社製の「MX-305」)にて、20℃、11000×g、10分間遠心分離し、上清(溶媒置換後の溶解液)を回収した。
(3)溶媒置換後の溶解液40mlに50体積%のNiレジン(Hisに結合する)のスラリーを2ml加え、室温で1時間振とうした。次いで、Niレジンを回収し、溶出緩衝液(50mM Tris、50mM NaCl)10mlにて5回洗浄した後、300mMのイミダゾールを用いて目的タンパク質(ADF3Kai-small-M)を溶出させ、溶出液を回収した。
ADF3Kai-small-Mのタンパク質を発現している大腸菌の湿菌体約0.2gに純水500μlを入れ、超音波破砕し、超音波破砕液を得た。
ADF3Kai-small-Mのタンパク質を発現している大腸菌の湿菌体約15gに、1MのLiClを含むDMSOを45ml添加し、60℃で30分間溶解させた。その後、遠心分離機(トミー精工社製の「MX-305」)にて、20℃、11000×g、10分間遠心分離し、上清(溶解液)を回収した。

- 前記溶解用溶媒は、前記宿主の質量に対して、体積(mL)/質量(g)比として、0.5倍以上添加する請求項1~5のいずれか1項に記載の親水性組換えタンパク質の粗精製方法。
- 前記溶解液を、水系溶媒で溶媒置換する請求項1~6のいずれか1項に記載の親水性組換えタンパク質の粗精製方法。
- 前記溶媒置換された溶解液を、さらにカラム、濾過、遠心分離からなる群から選ばれる一種以上の手段で処理する請求項7に記載の親水性組換えタンパク質の粗精製方法。
- 前記親水性組換えタンパク質は、組換えクモ糸タンパク質、組換えレシリン、及び組換えコラーゲンからなる群から選ばれる1つのタンパク質である請求項1~8のいずれか1項に記載の親水性組換えタンパク質の粗精製方法。
