Definitions
- the present invention relates to a microscope technique capable of observing a sample to be observed in atmospheric pressure or a predetermined gas atmosphere.
- a scanning electron microscope (SEM), a transmission electron microscope (TEM), or the like is used to observe a minute region of the object.
- SEM scanning electron microscope
- TEM transmission electron microscope
- a housing for placing a sample is evacuated, and a sample atmosphere is vacuumed to image the sample.
- biochemical samples and soft material materials are damaged or changed in state by vacuum.

- These devices in principle, provide a diaphragm or minute hole through which an electron beam can pass between the electron optical system and the sample to separate the vacuum state from the atmospheric state, both of which are between the sample and the electron optical system. It is common in the point that a diaphragm is provided.
- Patent Document 1 describes a sample storage container in which a diaphragm through which an electron beam passes is provided on the upper surface side. SEM observation is performed by detecting reflected electrons or secondary electrons from the sample in contact with the diaphragm.
- the sample is held in a space constituted by an annular member placed around the diaphragm, and the space is further filled with a liquid such as water.
- Patent Document 2 the electron source side of the electron optical lens barrel is oriented downward, and the objective lens side is oriented upward, and the electron beam through the O ring on the electron beam emission hole at the end of the electron optical lens barrel.
- an SEM provided with a diaphragm through which In the invention described in the document, a sample to be observed is directly mounted on a diaphragm, and a primary electron beam is irradiated from the lower surface of the sample to detect reflected electrons or secondary electrons and perform SEM observation.
- the sample is held in a space constituted by an annular member placed around the diaphragm, and the space is further filled with a liquid such as water.
- the sample storage container for performing sample observation under these conventional atmospheric conditions is mainly made for observing substances in liquid, and basically, the sample and the diaphragm should be observed in complete contact with each other Is assumed. Also, even if the sample and the diaphragm are not placed in complete contact with each other, the sample is moved independently of the diaphragm in the local environment formed in the vacuum chamber of the charged particle beam device to make the sample and the diaphragm It was not possible to adjust the relative positional relationship of Therefore, there is a problem that a sample not in the field of view of the diaphragm can not be observed. Also, for the same reason, it is difficult to observe a solid sample.
- the present invention has been made in view of such problems, and it is an object of the present invention to provide a sample storage container, a charged particle beam apparatus, and an image acquisition method capable of observing most of the sample regardless of the sample form.

- the present invention is also applicable to a scanning electron microscope, a scanning ion microscope, a scanning transmission electron microscope, a combined apparatus of these and a sample processing apparatus, or an analysis / inspection apparatus to which these are applied.
- atmospheric pressure means an atmospheric atmosphere or a predetermined gas atmosphere in a slightly negative pressure or pressurized state, and means an atmospheric pressure or a pressure environment similar to this. Specifically, the pressure is about 10 5 Pa (atmospheric pressure) to about 10 3 Pa or so.
- FIG. 1 shows the entire configuration of the sample storage container of the present embodiment.
- the sample storage container shown in FIG. 1 mainly includes the storage container 100, the lid 101, the sample table 102, the sample stage 103 having a drive mechanism for changing the position of the sample table 102, and the sample stage 103 from the outside of the sample storage container
- a plurality of operation units 104 for movement, a mechanical element 128 between the operation unit 104 and the sample stage 103, a diaphragm 10 for passing or transmitting charged particle beams, and a diaphragm holding member 106 for holding the diaphragm 10 are provided.
- the sample 6 is placed on the sample table, and is stored together with the sample table in the storage container 100 which is a closed space.
- a vacuum sealing member 107 such as an O-ring or packing is provided between the lid 101 and the storage container 100 in order to keep the gas species and the pressure state inside the sample storage container separated from each other.
- the lower surface of the sample stage 103 and the bottom surface of the storage container 100 are fixed by screws or the like (not shown).
- the lid 101 can be detached from the storage container 100.
- the inside of the storage container 100 in the state of being disposed inside the charged particle beam device is the atmospheric pressure or a desired gas pressure space, and the outside of the storage container 100 is in a vacuum state. Therefore, a force acts in the direction in which the lid 101 is pushed from inside the storage container 100. Therefore, the combination of the protrusion 109 connected to the lid 101 and the protrusion 110 connected to the storage container 100 prevents the lid 101 from being dismounted even if a force acts in a direction to be pushed from inside the storage container 100. May be In this case, the lid 101 can be attached to and detached from the storage container 100 by sliding in the direction perpendicular to the sheet of the drawing.
- the lid 101 and the storage container 100 may be fixed by a screw or the like (not shown).
- the storage container 100 and the lid 101 may be screwed and tapped on their own, and may be coupled by combining and rotating.
- the fixing means of the lid 101 is not limited to the above example, as long as the lid 101 and the storage container 100 are fixed by a force that can withstand the pressure difference between the inside and the outside of the sample storage container.
- the sample stage 103 is capable of driving the position of the sample 6 in a direction toward or away from the diaphragm, and an XY axis capable of driving in the lateral direction or the vertical direction in the drawing.

- These driving mechanisms are installed inside the sample storage container, and the sample stage 103 is operated by the operation unit 104 provided outside the sample storage container via the mechanical element 128.
- the mechanical element 128 is, for example, a rotating shaft or a rod.
- the operation unit 104 can be operated by rotating or pushing or pulling.
- a vacuum seal member 107 such as an O-ring or packing is provided between the containment vessel 100 and the mechanical element 128 so that the type of gas and the pressure inside the sample storage container do not change.
- the pressure difference between the inside and the outside of the sample storage container is maintained, and the sample 6 can be driven independently of the diaphragm 10 while maintaining the atmosphere (pressure, gas type) inside the sample storage container. That is, according to the position adjustment mechanism described above, the position of the sample 6 with respect to the diaphragm 10 can be adjusted from the outside of the sample storage container.
- the sample storage container is disposed on a flat portion such as a stage or a stand inside the charged particle beam apparatus, and the operation unit 104 is operated while observing the sample with an optical microscope or the like.
- the operation unit 104 be disposed on the side of the sample storage container as shown in the figure. Since the sample 6 and the diaphragm 10 are not in contact and it is possible to move the sample independently of the diaphragm 10 in a direction parallel to the diaphragm 10, a very wide range (at least a range larger than the area of the diaphragm) It becomes possible to observe. In addition, since the sample and the diaphragm are not in contact with each other, it is not necessary to exchange the diaphragm every time the sample is exchanged.

- the sample 6 is mounted on a sample stage 102 provided on the sample stage 103.
- only the sample 6 may be removed, or the entire sample table 102 may be removed.
- the lid 101 is provided with a diaphragm holding member 106 provided with the diaphragm 10. Airtightness is maintained between the lid 101 and the diaphragm holding member 106 by the presence of an adhesive, double-sided tape, vacuum grease, O-ring, packing or the like.
- the charged particle beam is emitted from the upper portion of the lid 101 in the drawing to irradiate the diaphragm 10 and the sample 6 with the charged particle beam, so the lid 101 is provided with the opening 112.
- a detector for detecting secondary charged particles emitted from the sample is disposed at the top of the lid 101. Therefore, in order to detect secondary charged particles efficiently, it is desirable that the opening 112 be shaped such that the opening area is larger at the upper surface than at the lower surface of the lid 101.
- the figure illustrates the configuration in which the opening 112 has a tapered shape.

- the side shown in FIG. 2, that is, the side on which the tapered portion 113 is present is disposed on the vacuum side (upper side in the drawing). This is to efficiently detect the secondary charged particles emitted from the sample by the detector 3.
- the sample storage container is provided with a gas inlet 114 and a gas outlet 115. These are provided with a valve mechanism that can separate or communicate the atmosphere state of the outer space of the sample storage container with the gas atmosphere state of the inner space 12. While the storage container 100 is closed by the lid 101, a desired gas is introduced from the gas introduction outlet 114. However, if the pressure inside the sample storage container rises too much, there is a risk that the diaphragm 10 may rupture. Therefore, when gas is introduced from the gas inlet / outlet 114 with the gas outlet port 115 open, the internal space 12 is filled with a gas atmosphere of a desired pressure without applying pressure to the diaphragm 10.
- the gas outlet port 115 may be a safety valve or the like that automatically opens when the pressure in the internal space 12 becomes higher than the external space of the sample storage container.
- a vacuum pump can be attached to the gas outlet port 115, in which case the sample storage container can be brought into a low pressure state of a desired gas type. Not only gas but also liquid may be introduced and discharged from the gas inlet / outlet and piping described above.

- the current introduction terminal 116 can be used for application of an electric field, temperature heater, temperature measurement and the like. Further, by arranging the detection element inside the sample storage container and connecting the signal line from the detection element to the current introduction terminal 116, it becomes possible to acquire the signal generated inside the sample storage container.
- the current introduction terminal 116 can also be used as an electrical signal lead-out terminal.
- the sample stage 102 below the sample 6 is a detection element such as a scintillator or a semiconductor detector that converts charged particle beams into light or electric signals
- the transmitted charged particle beams transmitted from the sample 6 are As it can be acquired, it becomes possible to acquire sample internal information. Since the inside of the sample container is in the atmospheric state or gas state, it is desirable to make the distance between the sample and the detection element shorter than the distance at which most of the transmitted charged particle beam scatters when detecting information inside the sample. That is, it is necessary to shorten the mean free path of the charged particle beam to be transmitted.
- the distance between the diaphragm and the sample and the allowable distance between the sample and the detector also change depending on the irradiation conditions such as the accelerating voltage of the charged particle beam, in reality, for example, it is necessary to be 1 mm or less Become. Therefore, it is desirable to place the sample 6 directly on the detection element.

If an optical microscope is provided in the apparatus, it is possible to adjust the sample position with high throughput because it can be adjusted by the operation knob of the sample storage container while it is on the stage 5 when it is desired to adjust the sample position. Become. Further, since the operation knob is on the side of the container, the above-described operation can be easily implemented. Furthermore, if the mounting position of the optical microscope on the lid member 50 is the same as the installation position of the charged particle optical lens barrel on the lid member when the lid member 50 is closed, the lid member 50 and the support are adjusted after adjustment to the sample position to be observed. When the sample storage container is placed in the housing 7 together with the plate 108, since the diaphragm center position 54 and the sample site to be observed are already present on the optical axis of the charged particle optical lens barrel, observation is performed. Can be done more easily.
- the optical microscope does not necessarily have to be included in the apparatus, and the sample storage container may be disposed on the base of the optical microscope outside the charged particle beam apparatus to perform observation and sample position adjustment.
- FIG. 5 shows an example in which a U-shaped lid 70 is placed on the sample storage container below the optical microscope 60 so as to cover the diaphragm 10.
- the lid 70 is shaped so as to form a closed space between it and the diaphragm in a state of covering the diaphragm 10, and immediately above the diaphragm so that the diaphragm can be observed with an optical microscope in the state of covering the diaphragm.
- a window 72 for observation is provided.
- FIG. 5 shows a space 71 which can be vacuumed, a window 72 which can be confirmed by the optical microscope 60, and a vacuum pump 73 and a vacuum meter 74 which are connected to the lid 70 through a pipe 75.
- a vacuum sealing member 76 (not shown) is provided between the window 72 and the lid 70.
- the diaphragm is broken, it is possible to grasp the situation such as the time to reach the desired degree of vacuum is delayed by the vacuum meter 74.
- the valve 114 or the like of the sample storage container is opened, the space 71 does not become vacuum if the diaphragm is broken, so it is possible to check the breakage of the diaphragm. is there.
- the sample position is adjusted in the third step, the presence or absence of breakage of the diaphragm 10 may be confirmed by the optical microscope 60 in some cases.
- the diaphragm 10 it may be possible to confirm the presence or absence of breakage of the diaphragm 10 by checking the wrinkles of the diaphragm that would normally be present, dust on the diaphragm, foreign matter, or marks on the diaphragm intentionally created. In that case, this fourth step is not necessarily required.
- the inside 11 of the case 7 is evacuated by a vacuum pump. At this time, the inside of the sample storage container is maintained at atmospheric pressure or a desired gas type and a desired pressure.
- the position of the sample storage container is confirmed from the image obtained by irradiating the primary charged particle beam.
- the stage 5 is adjusted so that the diaphragm center position 54 and the optical axis 55 of the charged particle optical lens barrel coincide with each other.
- the charged particle beam that has passed through or transmitted through the diaphragm 10 is irradiated to the sample 6.
- the sample position confirmed in the third step on a charged particle microscope image. If it is desired to adjust the position of the sample 6 observed through the diaphragm 10, the inside 11 of the housing 7 is opened to the atmosphere, and the process returns to the third step to perform sample position adjustment under an optical microscope.
- the area of the diaphragm is very small due to the requirement that the diaphragm must be maintained at a differential pressure between atmospheric pressure and vacuum. Therefore, there is a problem that the place which can be observed in the method of the conventional known literature is very narrowly limited by the area of the diaphragm.
- the sample can be freely moved independently of the diaphragm while confirming the sample position with the optical microscope, various parts of the sample can be observed. In particular, since the observation position adjustment work can be performed while maintaining the local atmosphere, the convenience of the user is greatly improved.
- ⁇ Contact prevention member> As described above, since the diaphragm 10 and the sample 6 need to be about 1000 ⁇ m or less, if the operation knob of the sample storage container is operated, there is a possibility that the sample and the diaphragm may be accidentally collided to damage the diaphragm. So, in a present Example, the contact prevention member which prevents that the diaphragm 10 and the sample 6 contact is arrange
- the contact preventing member 400 is provided so as to protrude from the sample table 401, and as shown in FIG. 6A, the tip of the contact preventing member 400 is always disposed closer to the diaphragm than the sample 6.
- the sample table 401 is disposed on the sample stage 103. Then, as shown in FIG. 6B, when the position of the sample table 401 is made close to the direction of the diaphragm 10, the diaphragm 10 and the sample 6 are firstly brought into contact with the lid 101 by the contact preventing member 400. It is possible to prevent contact.
- the height B of the sample 6 may change depending on the sample. Therefore, it is necessary to have an adjustment mechanism capable of adjusting the height A of the contact prevention member 400 according to the height of the sample B.
- the contact prevention member 400 is a male screw, and by changing the sample stand 401 to an internal thread 402, the height A of the contact prevention member 400 can be changed by turning the screw portion of the contact prevention member 400.
- the adjustment mechanism may be any mechanism that can move the position of the contact prevention member 400 at which the sample contacts the diaphragm in the optical axis direction of the charged particle optical lens barrel.
- FIG. 6 (a) and 6 (b) show that a vacuum sealing member 407 is provided for vacuum sealing between the lid 101 and the diaphragm 10.
- the vacuum sealing member 407 is, for example, an adhesive or a double-sided tape. If a vacuum seal member is present between the lid 101 and the diaphragm 10, the distance C is the sum of the thicknesses of the vacuum seal member of the lid 101 and the diaphragm 10 and the diaphragm 10.
- the distances A, B, and C are not known, use a device that can measure the height using a laser or light with the sample table 401 on which the sample 6 is mounted and the lid 101 holding the diaphragm 10 outside the device. The distances of A, B and C can be observed. If the sample 6 and the diaphragm 10 always have the same height, once the distance A from the sample table 401 to the contact preventing member 400 is determined, there is no need to adjust the contact preventing member 400 again. As described above, the contact preventing member also has an effect that the distance from the diaphragm to the surface of the sample can be kept constant by bringing the contact preventing member into contact with the diaphragm holding member.
- the sample 6 is mounted on the sample table 401.
- the contact preventing member 400 is inserted into the female screw 402.
- the distance between the surface of the sample 6 and the upper portion of the contact prevention member 400 is a term of (AB) shown by [Expression 1] or [Expression 2].
- A-B distance between the surface of the sample 6 and the upper portion of the contact prevention member 400 is a term of (AB) shown by [Expression 1] or [Expression 2].
- measurement or recording is performed using a device capable of measuring the height using a laser or light.
- the sample holder with the contact prevention member 400 and the sample 6 provided is placed on the sample stage 5 in the sample storage container.
- the lid 101 of the sample storage container is put on.
- the contact preventing member 400 and the lid 101 are brought into contact with each other.

Abstract
