Definitions
- the present invention relates to a method for obtaining optically pure amino acids using extraction methods including optical splitting and optical conversion.
- Optically pure amino acids are industrially important compounds because they are widely used as ligands for asymmetric catalysts or as starting materials or intermediates for the synthesis of various pharmaceuticals and bioactive substances (Helmchen, G .; Pfaltz, A. Acc. Chem. Res . 2000 , 33 , 336-345).
- amino acids obtainable through fermentation are limited to L-amino acids among natural amino acids.
- Optically pure D-amino acids and non-natural amino acids are produced through enzymatic and optical splitting methods, but due to high manufacturing costs, their unit cost is 5-10 times higher than that of natural L-amino acids produced by fermentation.
- Are experiencing difficulties Maruoka, K .; Ooi, T. Chem. Rev. 2003 , 103 , 3013.).
- the present inventors have developed a method of converting L-amino acids to D-amino acids by recognizing chiral amino alcohols and chirality of amino acids through imine bonds using a binaphthol derivative of the following formula having an aldehyde group ((a) Park , H .; Kim, K. M .; Lee, A .; Ham, S .; Nam, W .; Chin, J. J. Am. Chem. Soc. 2007 , 129, 1518-1519; (b) Kim, K. M .; Park, H .; Kim, H .; Chin, J .; Nam, W. Org. Lett. 2005 , 7 , 3525-3527.).
- the binaphthol derivatives form chiral selective imines with amino acids and can achieve L-D optical conversion of amino acids in organic solvents such as DMSO.
- organic solvents such as DMSO.
- Existing patents and articles (Park, H .; Kim, K. M .; Lee, A .; Ham, S .; Nam, W .; Chin, J. J. Am. Chem. Soc. 2007 , 129, 1518-1519) is an optical conversion in DMSO to extract the entire solution using water and an organic solvent.
- the amino acid is separated into the water layer
- the binaphthol derivative is separated into the organic layer
- the binaphthol derivative of the organic layer is recovered through concentration and reused.
- the L-D optical conversion takes about 24 to 48 hours for optical conversion, and the DMSO solvent cannot be recovered because the DMSO solvent is mixed in both the water and organic layers during extraction. Moreover, in order to recover the binaphthol derivatives, DMSO must be completely removed from the organic layer. In order to do so, a large amount of water must be used, and the working volume becomes large, thereby decreasing productivity. In addition, using another solvent that is not mixed with water instead of DMSO has a problem that the L-D optical conversion time is longer.
- the present invention is to solve the above problems of the prior art, the optical conversion time is significantly shortened, it is possible to repeatedly use the organic solution containing the chiral selective receptor without the concentration process for the organic solvent used It is an object of the present invention to provide a method for obtaining optically pure amino acids which are very efficient and economical.

- It provides a method for obtaining an optically pure amino acid comprising a third step of obtaining a D-type or L-type amino acid from the acidic aqueous solution separated in step 2.
- the process of the first step and the second step may be further repeated one or more times.
- the amino acid included in the basic aqueous solution may be in the form of Li +, Na + or K + salt.
- the basic aqueous solution may further include a racemization catalyst for racemization of amino acids.

- the organic solvent included in the organic solution may be a mixed solvent of an organic solvent that is not mixed with water at all and an organic solvent containing a highly polar functional group.
- the organic solution may further include a Phase Transfer Catalyst (PTC).
- PTC Phase Transfer Catalyst
- the amino acid may be alpha-amino acid or beta-amino acid.
- a racemization catalyst in a basic aqueous solution containing amino acids, and the aqueous solution is heated to 50 to 100 ° C.
- the method of the present invention significantly shortens the optical conversion time, and it is possible to repeatedly use an organic solution containing a chiral selective receptor without the concentration process for the organic solvent used.
- optically pure amino acids can be obtained very efficiently by a simple method and can be mass produced very economically.
- Figure 2 is a 1 H NMR graph carried out to confirm the chiral selectivity of the imine formed by the binding of the amino acid and chiral selective receptor in Example 5 of the present invention.

At this time, the amount of NaOH or KOH may be put in the number of moles such as amino acids or slightly more or less than this. This amount can be adjusted according to the stability of the amino acid, the degree of racemization, chiral selectivity, and the like.
- the basic aqueous solution may further include a racemization catalyst for racemization of amino acids.

- all derivatives comprising salicylic aldehyde groups capable of forming amino acids and imines can be used in the process of the invention as long as they have chiral selectivity.
- Even compounds without salicylic aldehyde groups can be used in the present invention as long as they have chiral selectivity to amino acids and satisfy the aforementioned conditions.

- MC methylenechloride
- EA ethylacetate
- toluene 2-pentanone
- butyronitrile tolulenonitrile
- methyl Isobutyl ketone a mixed solvent thereof.
- organic solvents with high polar nitrile, carbonyl and sulfoxide groups alone increases the chiral selectivity, but these solvents may be mixed in water. This is because it is possible to prevent mixing with water by increasing the selectivity while using a solvent which is not mixed with water at all.
- the mixing ratio of the organic solvent which does not mix with the water at all and the organic solvent containing a highly polar functional group may be 1: 9 to 9: 1 by volume ratio.
- the mixed solvent examples include a mixed solvent of methylene chloride and butyronitrile, a mixed solvent of chloroform and tolulenonitrile, and the like.

- the acidic aqueous solution may be prepared by adding various acids, for example, may be made acidic by adding HCl.
- the appropriate concentration is determined by considering the amino acid type and the time it takes for the imine bond to break.
- the two layers are stirred together in a general manner, and thus the binding with the chiral selective receptor (S-type or R-type) contained in the organic solution of the amino acid of the basic aqueous solution is performed.
- S-type or R-type the chiral selective receptor contained in the organic solution of the amino acid of the basic aqueous solution.
- D- or L-forms selectively form imines with chiral selective receptors.

- the time required for the selective transfer of amino acids from the basic aqueous solution layer to the organic solution layer is 2 to 3 hours, and the amino acid racemization in the basic aqueous solution can be completed in a short time using a suitable catalyst.
- LD optical conversion of amino acids can be completed in a short time.
- optical conversion is performed in DMSO according to the disclosed patent method, it takes at least several days, including reaction and recovery, and thus the method of the present invention can be said to be very innovative.
- the present invention is very economical because the organic solution layer containing the chiral selective receptor can be reused as it is without any recovery process.
- PLP pyridoxal phosphate
- basic aqueous solution PLP forms imine with amino acid and racemates amino acid.
- the amount of PLP is most suitable for 5% of the number of moles of amino acid, but can vary depending on the type of amino acid.
- the racemization of amino acids in PLP 5% proceeds slowly at room temperature and proceeds about 30% per week. This can be confirmed, for example, by adding PLP 5% in D 2 O to the Na + salt of phenylalanine and observing the degree of deuteration of alpha hydrogen. This is because the degree of deuteration of alpha hydrogen means the racemization of amino acids.
- Increasing the amount of PLP by 20% does not speed up the racemization of amino acids.
- the temperature of the aqueous solution is raised to 50 ⁇ 100 °C it was found that the racemization of amino acids can be made in a short time within 1 hour. Therefore, it is preferable to raise the temperature of basic aqueous solution to the said range in order to advance racemization in basic aqueous solution which supplies an amino acid.
- the temperature of the aqueous solution is heated to less than 50 °C in the above, the racemization promoting effect is insignificant, and is not heated above 100 °C because of the aqueous solution.
- the amino acid of the basic aqueous solution layer is moved to the organic solution layer in the above, the amino acid may be continuously added to continue the optical conversion.
- Chiral selectivity in the present invention is due to the structural stability of the imine, a detailed principle of which is given in several published papers.
- the published paper JACS 2007; Che. Eur. J 2008; Tetrahedron 2008) describes the use of binaphtholaldehyde or other derivatives to achieve chiral conversion of amino acids in a DMSO solvent.
- chiral conversion of amino acids generally does not occur easily.
- the method of the present invention provides a method that enables chiral conversion of all DL-amino acids. This method is possible because of the racemization of amino acids in the basic aqueous solution.
- chiral conversion can in principle be applied to all alpha-amino acids capable of racemization in aqueous solution.
- the method of the present invention can be effectively applied to chiral separation of general beta-amino acids.
- the method of the present invention can be applied to compounds of all amines capable of forming imines with chiral selective receptors.
- the racemization catalyst is put into a basic aqueous solution containing an amino acid, and the said aqueous solution is heated to 50-100 degreeC, It is related with the racemation promotion method.
- the racemization rate of amino acids does not increase.
- the racemization catalyst is added in the present invention and the temperature of the aqueous solution is heated to 50 to 100 ° C.
- the racemization of the amino acid may be performed in a short time within 1 hour. Therefore, in order to advance the racemization in the aqueous solution supplying the amino acid, it is preferable to heat the temperature of the aqueous solution to the above range.
- salicylic aldehyde derivatives may be used as the racemization catalyst.
- the salicylic aldehyde derivatives include PLP (pyridoxal phosphate), pyridoxal (pyridoxal) and the like, these may be used alone or in combination of two or more.
- PLP pyridoxal-5'-phosphate
- the pH of the basic aqueous solution may be more than 7 ⁇ 14, preferably pH 10 ⁇ 12.
- Phenylalanine (racemic, 30 g, 180 mmol) was added to water (100 ml), 1.0 equivalent of NaOH was added to dissolve all, and PLP 1.5 g was added to prepare a basic aqueous solution.
- the small peak indicated by L in FIG. 2 is for NH of imine made with L-phenylalanine and the NH labeled with imine made with D-phenylalanine.
- This chiral selectivity was identical to that of chiral selectivity when optical conversion of L-phenylalanine to D-phenylalanine in DMSO (Park, H .; Kim, KM; Lee, A .; Ham, S .; Nam, W .; Chin , JJ Am. Chem. Soc. 2007, 129, 1518-1519).
- the organic solution layer was separated from the aqueous layer using a separatory funnel and then stirred together with the acidic aqueous solution for 1 hour. After confirming by 1 H NMR, all the amino acids were separated from the compound 3 and the organic solution layer was confirmed that the compound 3 and Aliquat 336 exist in the same state as before the optical conversion. In addition, it was confirmed that D-phenylalanine separated from the organic solution layer was present in the acidic aqueous solution layer.
- Beta-amino acids such as DL-serine, DL-alanine, DL-valine, DL-leucine, DL-tryptophan and DL-3-aminobutyric acid, beta-phenylalanine, beta-leucine, beta-homophenylalanine, beta- Optical selective transfer experiments on beta-amino acids such as homoleucine and 3-aminoisobutyric acid were also performed in the same manner as described in Example 5 above. The chiral selectivity of amino acids moving from the aqueous solution layer to the organic solution layer was confirmed by area calculation of the 1 H NMR peak, and the results are shown in Table 1 below.
- the method of the present invention showed relatively high chiral selectivity for beta-phenylalanine, beta-leucine, beta-homophenylalanine, and beta-homoleucine even in beta amino acids.
