Abstract
本発明が解決しようとする課題は、副作用を心配することなく、日常的に摂取可能であって、肌を内部から美しいものとすると同時に全身のスキンケアを可能とする医薬品及び飲食品を提供することであり、その解決手段は、乳清から培養されてなる乳清ケフィアを有効成分として含んでなる内服用美肌剤である。
Description
本発明は、乳清から培養されてなる乳清ケフィアを有効成分として含んでなる、内服用美肌剤に関する。
美しい肌に対する関心は、近年ますます高いものになっており、美しく白い肌(美白の肌)や、気になる赤みのない肌や、美しくうるおいのある肌や、美しくきめのこまかな肌が、望まれるようになってきた。このような美しい肌となるために、美肌用の製品が市販されているが、その大半はいずれも肌(皮膚)に直接適用する外用化粧料であった。このような外用化粧料は、直接に塗布した部位でしかその効果を発揮せず、さらに発汗や摩擦などではがれやすく、短時間毎に塗り直すなどが必要であった。そのために、肌を内部から美しいものとすると同時に全身のスキンケアが可能となる内服用(経口用)の美肌製品が、求められるようになってきた。
ケフィアは、ロシアのコーカサス地方原産の発酵乳であり、コーカサス地方に由来するケフィアグレイン(ケフィア粒、ケフィア菌)を種菌(スターター)として牛乳等の獣乳を発酵させて製造される。ケフィアグレインは、それ自体があたかも一つの生体として振る舞うものであるが、学術的には各種微生物が天然の共生体となったものであり、Lactobacillus kefiri、並びにLeuconostoc属, Lactococcus属 及び Acetobacter属の各種微生物の1種以上、さらに、乳糖発酵性酵母(例えばKluyveromyces marxianus)及び非乳糖発酵性酵母(例えばSaccharomyces unisporus, Saccharomyces cerevisiae, Saccharomyces exiguus)を含んでいる。ケフィアは、発酵乳として美味である点に加えて、各種の健康維持増進機能が指摘されており、健康によい食品として普及しつつある。
このケフィアの優れた特性を生かすために、ケフィアに種々の食品成分を添加して、さらに健康維持増進機能に優れた食品を創り出す試みが行われてきた。例えば、特許文献1(WO2002/076240)は、ケフィアにナットウキナーゼを添加した食品を開示している。
乳清(ホエー)は、牛乳からチーズ製造を行う際の副産物として、大量に生産される。乳清は、低カロリーでミネラルやビタミンが豊富であることから、健康的な食品素材として、近年注目されている。しかし、乳清は、チーズ生産量と比較して需要が極めて少ないために、その多くが廃棄物として処理される。
上述のように、肌を内部から美しいものとすると同時に全身のスキンケアが可能となる、新規な内服用美肌剤が求められていた。
また、上述のように、チーズ生産量と比較して需要が極めて少ないために、その多くが廃棄物として処理される乳清(ホエー)を、医薬品及び飲食品の原材料として、有効に利用することが、求められていた。
従って、本発明の目的は、新規な内服用美肌剤を提供することにある。
さらに、本発明の目的は、乳清(ホエー)を原材料とした新規な医薬品及び飲食品を提供することにもある。
本発明者等は、ケフィアについて鋭意研究開発を行ってきた。その結果、乳清(ホエー)を原材料として製造した乳清ケフィアによって、上記目的が達成されることを見出して、本発明を完成した。乳清ケフィアは、乳清を原材料としているために、通常のケフィアとは異なる飲食品であり、ケフィア様飲食品というべきものである。
従って、本発明は、次の[1]~[5]にある。

さらに、本発明は、乳清から培養されてなる乳清ケフィアの、美肌のためのスキンケアの使用、肌を保湿するための使用、肌を美白するための使用、肌の肌理(肌のきめ)を改善するための使用、肌の赤みを低減するための使用にもある。
さらに、本発明は、内服用美肌剤、内服用肌保湿剤、内服用肌美白剤、内服用肌理改善剤、または内服用肌赤み低減剤を製造するための、乳清から培養されてなる乳清ケフィアの使用にもある。
本発明によれば、肌の保湿、肌の美白、肌の肌理(肌のきめ)の改善、肌の赤みの低減が可能であり、このように美肌のためのスキンケアが可能である。本発明による内服用美肌剤、内服用肌保湿剤、内服用肌美白剤、内服用肌理改善剤、及び内服用赤み低減剤は、乳清ケフィアを有効成分としているので、副作用を心配することなく、安心して、日常的に摂取可能である。
また、本発明によれば、このように優れた医薬品及び機能性食品を、その多くが廃棄物として処理される乳清(ホエー)を原材料として、製造することができるので、環境負荷の低減と優れた経済性を両立するものとなっている。
以下に発明の実施の形態を示して本発明を詳細に説明する。本発明は、以下に例示する実施の形態に限定されるものではない。WO2002/076240号公報の内容は、本明細書の一部として取り込む。特に記載のない限り、%は質量パーセントを表す。
本発明に係る内服用美肌剤、内服用肌保湿剤、内服用肌美白剤、内服用肌理改善剤、または内服用肌赤み低減剤は、乳清ケフィアを有効成分として含んでなるものである。
いわゆる通常のケフィアは、牛乳等の獣乳を原材料として、ケフィアスターターを接種して発酵させて得られたものである。一方、本発明の乳清ケフィアは、乳清を原材料として、ケフィアスターターを接種して発酵させて得られるものである。
乳清(ホエー)は、牛乳からチーズ製造を行う際の副産物として大量に生産され、チーズ生産量と比較して需要が極めて少ないために、その多くが廃棄物として処理されるが、食品素材としては伝統的なものであって、人類史的な食経験の蓄積がある食品素材であるために、長期にわたって日常的に摂取した場合にも、その安全性には何ら心配がない。また、ケフィアグレインによる発酵もまた伝統的に行われているものであって、こちらも人類史的な食経験の蓄積がある食品であるために、発酵生成物の安全性には、長期にわたって日常的に摂取した場合にも、何ら心配がない。
本発明に係る有効成分である乳清ケフィアは、乳清を含む原料溶液から、次の工程:

好適な実施の態様において、乳清を含む原料溶液は、乳清及び乳糖を含んでなるものとすることができる。乳清は、市販の乳清、例えば、液体の乳清を使用することができ、乾燥した乳清粉末(ホエーパウダー)を使用することもでき、液体の乳清とホエーパウダーを混合して使用することもできる。乳糖は、市販の乳糖を使用することができる。
好適な実施の態様において、原料溶液は、ケフィアスターターの接種の前に、滅菌処理が行われる。滅菌処理としては、例えば加熱処理を挙げることができる。加熱処理は、例えば、80~95℃での10~120分、好ましくは30分~90分の加熱保持によって行うことができる。あるいはUHT法による殺菌又はHTST法による殺菌を行うこともできる。
好適な実施の態様において、ケフィアスターターとして、ケフィアグレインを投入して培養した培養液を使用することができ、例えば、ケフィアグレインを牛乳中へ投入して継代培養した培養液を使用することができるが、ケフィアスターターとしてケフィアグレインを使用することもできる。好適な実施の態様において、ケフィアスターターは、原料溶液100質量部に対して、例えば、1~20質量部、好ましくは2~14質量部、さらに好ましくは2~10質量部、さらに好ましくは3~8質量部の範囲で接種することができる。
原料溶液に接種されたケフィアスターターは、20~33℃、好ましくは24~33℃の温度で、10~25時間、好ましくは15~20時間の間、培養することによって、発酵させる。
好適な実施の態様において、培養工程の後には、培養液の滅菌処理を行う滅菌工程、及び、滅菌された培養液のろ過処理を行う清澄化工程、が行われる。滅菌工程は、上述の滅菌工程と同様の熱処理によって行うことができる。ろ過処理を、滅菌と清澄化を兼ねた処理として行うこともできる。ろ過処理は、透過孔の小さなフィルターでのろ過に先立って、プレフィルターによるろ過処理、あるいはフィルタープレスによる加圧ろ過処理を行ってもよい。透過孔の小さなフィルターでのろ過としては、0.20~20μmの範囲の透過孔のフィルターを使用することができる。このうち、0.20~0.45μmの範囲の透過孔のフィルターは、滅菌(除菌)を兼ねたフィルターとして使用することができる。ろ過処理の結果、培養液は清澄なものとなり、清澄化された乳清ケフィアを得ることができる。
本発明に係る乳清ケフィアは、これを有効成分として、内服用美肌剤、内服用肌保湿剤、内服用肌美白剤、内服用肌理改善剤、または内服用肌赤み低減剤として使用することができる。これらは、好ましくは経口的に摂取されるが、経腸的に摂取することもできる。これらは、医薬品として使用することもできるが、日常的に安心して摂取し続けることができる点から、飲食品として好適であり、特にいわゆる機能性食品、健康食品として好適に摂取することができる。すなわち、本発明は、乳清ケフィアを有効成分として含んでなる機能性食品、乳清ケフィアを有効成分として含んでなる特定保健用食品、乳清ケフィアを有効成分として含んでなる食品添加剤、にもある。さらに、本発明は、美肌用、肌保湿用、肌美白用、肌理改善用、及び赤み低減用の各用途の、医薬品、飲食品、機能性食品、特定保健用食品、食品添加剤、医薬組成物、内服用組成物、及び飲食用組成物にもある。
本発明に係る医薬品及び飲食品の摂取量は、有効成分である乳清ケフィア(清澄化された、発酵後の培養液)の質量に換算して、例えば成人1日あたり1~300g、好ましくは5~200g、さらに好ましくは10~100gを摂取することができるが、これらの量に限定されるものではない。本発明の有効成分である乳清ケフィアは、副作用の心配なく飲食品として日常的に摂取することができるものであるので、これよりも多量の摂取をすることには何ら問題がない。摂取は、例えば毎日であってもよく数日毎であってもよいが、本発明による健康の維持と増進の効果を有効に発揮するためには、継続的に毎日摂取することが好ましい。好適な摂取の期間は、例えば1週間以上、好ましくは2週間以上、さらに好ましくは3週間以上、さらに好ましくは4週間以上を挙げることができるが、本発明による健康の維持と増進の効果を有効に発揮するためには、さらに長期にわたって継続的に摂取することが好ましい。本発明の有効成分である乳清ケフィアは、副作用の心配なく飲食品として日常的に摂取することができるものであるので、これよりも長期間の摂取をすることには何ら問題がない。本発明の有効成分である乳清ケフィアは、液体の形態のままで摂取することもでき、食品用として公知の添加物を添加して摂取することもでき、他の飲食品に添加して摂取することもでき、医薬品に使用される公知の添加物とともに製剤して摂取することもでき、あるいは公知の方法で乾燥して例えば乾燥粉末の形態として摂取することもでき、この乾燥品を他の飲食品に添加して摂取することもでき、この乾燥品を他の飲食品に添加して摂取することもでき、この乾燥品を医薬品に使用される公知の添加物とともに製剤して摂取することもできる。
以下に実施例を示して本発明を詳細に説明する。本発明は、以下に示す実施例に限定されるものではない。なお、以下で%(パーセント)の表記が使用される場合は、特に示さない限りはいずれも重量パーセントである。
[製造例]
乳清ケフィアを次のように製造した。
攪拌容器に、水(80質量部)、粉末乳清(ホエーパウダー)(10質量部)、及び乳糖(10質量部)を加えて、溶解するまで十分に攪拌した。ケフィアスターターとして、ケフィアグレイン(日本ケフィア社製)を牛乳培地に10%比で培養した培養液を別途用意した。上記攪拌した混合溶液を90℃で1時間殺菌した後に発酵原料とした。発酵原料の溶液を発酵タンクに投入して約30℃に保温し、ケフィアスターター(5.00質量部)を発酵原料に添加し、約30℃で約20時間保温して発酵させて発酵物を得た。得られた発酵物に対して、130℃20秒間のUHT殺菌、及び95℃で1時間の加熱殺菌、フィルタープレスによる加圧ろ過、フィルター(透過孔10μm)によるろ過を行って、清澄化された無菌状態の乳清ケフィアを得た。その後の飲用に供しやすいように、乳清ケフィア20mlを、水、香料、液体甘味料を添加して全量50mlへと希釈し、50mlビンに小分けして無菌充填してケフィアドリンクとした。
乳清ケフィアを次のように製造した。

試験は、4週間にわたって行った。試験食(乳清ケフィア)の摂取開始から終了時まで、被験者が新たなサプリメントの併用摂取を行わないこととした。ただし、被験者が、従来からの生活習慣として、試験開始前から継続して飲食していたサプリメントについては、その用法及び用量を変更しないまま継続するという条件で、併用可とした。被験者の生活状況は、摂食チェック表で管理した。
[測定評価方法]
被験者の顔面の肌の状態を、室温(25℃)にて、湿度45~50%の範囲の環境下で、洗顔の10分後に、ロボスキンアナライザーRSA-100システムセット(株式会社インフォワード製)を使用して、皮膚の状態について、以下のように各項目を測定して評価した。

水分量及び油分値は、油分水分センサーを用いて測定した。センサーを左右の頬に当て、以下の測定方法及び評価基準より測定結果を評価した。油分値(肌表面に分泌された皮脂量)は、センサー部分を押し当てることで圧延された油分を屈折率により計測し、面積で評価した。全面を油分が覆った状態を「油分が飽和した状態(100)」とし、全く油分が存在していない状態を「油分がゼロの状態(0)」として評価した。皮脂は、肌表面から水分が失われることを防ぎ、肌に柔軟性を与え、肌のバリア機能を維持するとされており、肌の油分量は加齢に伴い減少することが知られているので、数値が大きいほど、油分が多く、若々しい肌であることを意味する。水分値(生体表層の水分量)は、センサー部分を押し当てることで肌の静電容量を計測し、水分を表す値として計測した。生理食塩水そのものを計測した状態を「飽和した状態(100)」とし、全く水分が存在しない状態を「ゼロの状態(0)」として評価した。数値が大きいほど、水分が多く、ふっくらとした若々しい肌であることを意味する。
肌のきめ(皮溝密度)は、左右の頬を、マイクロスコープを用いて撮影し、撮影された画像をロボスキンアナライザーにて解析して、評価した。解析方法は、モノクロ画像に変換した撮影画像中における暗部を皮溝、明部を皮丘とし、明暗部を各々処理して二値化した結果と、一辺0.4mmの正三角形のきめモデル(アゴの下の皮膚の状態を参考にしたもの)とを比較し、モデルを100として撮影画像を評価した。数値が大きいほど、きめ(肌理)が整った、若々しい肌であることを意味する。
赤味は、全顔撮影ボックスにて撮影した肌画像の解析により評価した。撮影したカラー肌画像は、赤味を含む肌色部位群とそれらを含まない地肌部位群との2群に分け、この群間を繋ぐスペクトルを基準として画像の色情報を一次元化(モノクロ化)した。そして、測定領域に存在する「地肌に比べて赤いもの」を「赤味」と定義した。ニキビや紅班は「赤味」に含まれる。さらに、赤味は、その濃淡に応じてレベル(LV)を1~3に分類した。LV1とは薄いものから濃いものまでを包含し、LV2は検出感度換算でLV1の約54%、LV3はLV1の約35%(濃いものだけ)とした。したがって、LV1が検出感度を1番高く設定したもので、LV2はその約54%、LV3はその約35%ということになる。赤味は、ロボスキンアナライザーで、その領域の直径が0.6以上で1.2mm以下のものを「小」、1.2mmより大きいものを「大」として検出され、個数及び面積をカウントした。
[統計学的検定法]
数値は平均±標準誤差で示した。試験食摂取前後の数値に対する統計解析は、two-tailed paired Student’s t-testを使用して行った。数値は平均±標準誤差で示した。有効性は、危険率5%未満を「有意差あり」と判定し、危険率5%以上で数値に増減がある場合、生理的に「改善傾向あり」と判定した。

[結果]
試験食投与開始例10例中,摂取期間中の脱落者はなかった。最大の有効性解析対象集団(Full Analysis Set,以下FAS)と安全性解析対象集団としてともに被験10例全例を採用した。被験者はいずれも女性であり、その背景を次の表2に示す。

BMI(ボディ・マス・インデックス)指数は、肥満度の判定に使用される評価値であり、次の式:

によって算出される。日本肥満学会では、BMIが22の場合が標準体重であるとし、BMIが25以上の場合を肥満としている。

摂取期間の前及び後に被験者に対して、顔面肌状態を示す各項目を測定して、得られた結果を次の表3に示す。
上記表3に示されるように、肌の赤みを示す評価は、本試験食の摂取期間の前後で比較して、いずれも有意に減少(改善)していた。このことから、本試験食(乳清ケフィア)が、肌の赤みを低減して、肌の美白に寄与することがわかった。
また、上記表3に示されるように、肌の水分値、肌の油分値、肌のきめ(肌理)を示す評価は、本試験食の摂取期間の前後で比較して、いずれも有意に増加(改善)していた。このことから、本試験食(乳清ケフィア)が、肌の水分を維持しあるいは増大させて、肌のうるおいと保湿に寄与すること、肌の油分を維持しあるいは増大させて、肌のうるおいと保湿に寄与すること、肌のきめ(肌理)のこまかさを維持しあるいは向上させて、肌理の改善に寄与することが、明らかになった。
本試験食の摂取期間及びウオッシュアウト期間を通じて、臨床検査、問診及び検診の結果から、試験食摂取に関連する、あるいは関連が否定できない有害事象はともに認められなかった。
本発明の内服用美肌剤、内服用肌保湿剤、内服用肌美白剤、内服用肌理改善剤、または内服用肌赤み低減剤は、肌の保湿、肌の美白、肌の肌理の改善、肌の赤みの低減、美肌のためのスキンケアを可能とすると同時に、乳清ケフィアを有効成分としているので、副作用を心配することなく、安心して、日常的に摂取可能であるために、本発明は産業上有用である。さらに、本発明によれば、このように優れた医薬品及び機能性食品を、その多くが廃棄物として処理される乳清(ホエー)を原材料として、製造することができるので、環境負荷の低減と優れた経済性を両立するものとなっており、本発明は産業上有用である。
Claims (7)
- 乳清から培養されてなる乳清ケフィアを有効成分として含んでなる、内服用肌保湿剤。
- 乳清から培養されてなる乳清ケフィアを有効成分として含んでなる、内服用肌理改善剤。
- 乳清から培養されてなる乳清ケフィアを有効成分として含んでなる、内服用肌赤み低減剤。
- 乳清から培養されてなる乳清ケフィアを有効成分として含んでなる、内服用美肌剤。
- 乳清を含む原料溶液にケフィアスターターを接種する接種工程、
ケフィアスターターが接種された原料溶液を培養する培養工程、
を含んでなる、乳清を含む原料溶液から乳清ケフィアを製造する方法。 - 原料溶液が、乳清、及び乳糖を含んでなる、請求項5に記載の方法。
- 培養工程の後に、
