Abstract
Description
〔グルタチオン産生促進剤,グルタチオンの欠乏に起因する疾患の予防・治療剤〕
本実施形態のグルタチオン産生促進剤及びグルタチオンの欠乏に起因する疾患の予防・治療剤は、リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物(以下、単に「甘草抽出組成物」という場合がある。)を有効成分として含有する。
本実施形態の飲食品又は飼料は、リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を配合したものである。
甘草(Glycyrrhiza glabra)の根茎を粗砕したチップ状物10kgに10Lの水を加え、ゆるく攪拌しながら抽出し、デカンテーションで固液分離して抽出液を得た。得られた抽出液をペースト状になるまで減圧濃縮した後、70容量%エタノール水溶液を得られた濃縮物の10倍量加え、攪拌機を用いて分散させた。分散液を5℃にて一夜保管し、生じた沈殿を濾紙で吸引濾過して除去し、濾液のエタノール水溶液を減圧下で留去した。

甘草(Glycyrrhiza uralensis)の根茎を粗砕したチップ状物10kgに10Lの3%アンモニア水を加える以外は実施例1と同様にして処理し、黄褐色粉末410gを得た(試料2)。得られた粉末を実施例1と同様にして高速液体クロマトグラフ法で分析した結果、グリチルリチン酸含量0.9質量%、リクイリチン含量5.5質量%、イソリクイリチン含量4.6質量%、リクイリチゲニン含量3.9質量%、イソリクイリチゲニン含量3.5質量%であった。
甘草(Glycyrrhiza inflata)の根茎を粗砕したチップ状物10kgに10Lの水を加え、ゆるく攪拌しながら抽出し、デカンテーションで固液分離して抽出液を得た。得られた抽出液を攪拌しながら、50%硫酸を加えて抽出液のpHを2に調整し、酸性析出処理を行い、ろ過により沈殿物を集めた。得られた沈殿物に水を加え、攪拌して分散させ、静置後上澄みをデカンテーションで除去した。この操作を繰り返して硫酸を除去した後、濾過を行って沈殿物を得た。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして表皮角化細胞におけるグルタチオン産生促進作用を試験した。
式中、Aは「試料無添加時の細胞中における総タンパク量当たりのグルタチオン量(対照)」を表し、Bは「試料添加時の細胞中における総タンパク量当たりのグルタチオン量」を表す。
結果を表1に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして皮膚線維芽細胞におけるグルタチオン産生促進作用を試験した。
式中、Aは「試料無添加時の細胞中における総タンパク量当たりのグルタチオン量(対照)」を表し、Bは「試料添加時の細胞中における総タンパク量当たりのグルタチオン量」を表す。
結果を表2に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにしてB16メラノーマ細胞におけるグルタチオン産生促進作用を試験した。
式中、Aは「試料無添加時の細胞中における総タンパク量当たりのグルタチオン量(対照)」を表し、Bは「試料添加時の細胞中における総タンパク量当たりのグルタチオン量」を表す。
結果を表3に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして肝細胞におけるグルタチオン産生促進作用を試験した。
式中、Aは「試料無添加時の細胞中における総タンパク量当たりのグルタチオン量(対照)」を表し、Bは「試料添加時の細胞中における総タンパク量当たりのグルタチオン量」を表す。
結果を表4に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして表皮角化細胞におけるγ-グルタミルシステインシンテターゼmRNA発現促進作用を試験した。
式中、Aは「試料無添加時のmRNA発現量(対照)」を表し、Bは「試料添加時のmRNA発現量」を表す。
結果を表5に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして皮膚線維芽細胞におけるγ-グルタミルシステインシンテターゼmRNA発現促進作用を試験した。

結果を表6に示す。

結果を表7に示す。
上記のようにして得られた甘草抽出組成物(試料1~3)について、以下のようにして肝細胞におけるγ-グルタミルシステインシンテターゼmRNA発現促進作用を試験した。
式中、Aは「試料無添加時のmRNA発現量(対照)」を表し、Bは「試料添加時のmRNA発現量」を表す。
結果を表8に示す。
製造例1で得られた甘草抽出組成物(試料1)を養殖用飼料として用いて、下記条件で養殖した養殖魚の筋組織中に含まれるグルタチオン量を測定した。
(2)試験時期:9~10月
(3)試験尾数:10/6000(放尾数)
(4)投与量:20mg-試料1/kg-魚体重
(5)投与方法:4日給餌,3日休餌(1ヶ月)
(6)測定日:試験開始1ヶ月後
試料1投与群の養殖マダイの筋肉(背側)を5%5-スルホサリチル酸中でホモジナイズし、4℃にて遠心分離後、上清を試料溶液とした。96wellプレートに試料溶液25μL及び0.1Mのリン酸緩衝液125μLを加えた後、10mMの5,5'-dithiobis(2-nitrobenzoic acid)25μLを加え、5分後の波長412nmにおける吸光度を測定し、ΔOD/minを求めた。還元型グルタチオン濃度は、還元型グルタチオン標準品を用いて作成した検量線をもとに算出した。また、コントロール群として、試料1を与えない以外は同様の条件で養殖した養殖マダイの筋肉(背側)中の還元型グルタチオン濃度を同様にして算出した。
結果を表9に示す。
HR-1/Hos系の5週齢の雌性ヘアレスマウス(星野試験動物飼育所より購入)を1週間馴化した後、製造例1で得られた甘草抽出組成物(試料1)を粉末試料(ラボMRストック,清水実験材料社製)に混餌し、甘草抽出組成物(試料1)の1日あたりの摂取量が50(mg/kg-マウス体重)となるように、3週間毎日給餌した。給餌終了後、皮膚を採取し、テフロンホモジェナイザーを用いて、採取した皮膚1mgあたり100μLの1%Tritonx-100含有PBS中で皮膚を可溶化し、4℃にて遠心分離(12000rpm,15分間)した後、上清を測定試料とした。
結果を表10に示す。
常法により、以下の組成を有する配合飼料を製造した。
甘草抽出組成物(製造例1) 1質量%
魚粉 75質量%
小麦粉 10質量%
でん粉 7質量%
魚油 3質量%
ミネラル類 2質量%
ビタミン類 2質量%
常法により、以下の組成を有する錠剤を製造した。
甘草抽出組成物(製造例1) 30.0mg
乳清ミネラル(カルシウム25~30%含有) 100.0mg
ビタミンK2(1%含有粉末) 1.5mg
マルチトール 156.0mg
グリセリン脂肪酸エステル 12.5mg
常法により、以下の組成を有する錠剤を製造した。
甘草抽出組成物(製造例2) 15.0mg
ドロマイト(カルシウム20%、マグネシウム10%含有) 83.4mg
カゼインホスホペプチド 16.7mg
ビタミンC 33.4mg
マルチトール 126.8mg
コラーゲン 12.7mg
ショ糖脂肪酸エステル 12.0mg
常法により、以下の組成を有するカプセル剤を製造した。なお、カプセルとしては、1号ハードゼラチンカプセルを使用した。
<1カプセル(1錠200mg)中の組成>
甘草抽出組成物(製造例2) 20.0mg
コーンスターチ 60.0mg
乳糖 100.0mg
乳酸カルシウム 10.0mg
ヒドロキシプロピルセルロース(HPC-L) 10.0mg
常法により、以下の組成を有する経口液状製剤を製造した。
<1アンプル(1本100mL)中の組成>
甘草抽出組成物(製造例3) 0.4質量%
ソルビット 12.0質量%
安息香酸ナトリウム 0.1質量%
香料 1.0質量%
硫酸カルシウム 0.5質量%
精製水 残部
Claims (6)
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を有効成分として含有することを特徴とするグルタチオン産生促進剤。
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を有効成分として含有することを特徴とするグルタチオンの欠乏に起因する疾患の予防・治療剤。
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を配合したことを特徴とする飲食品。
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を配合したことを特徴とする飼料。
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を含有するグルタチオン産生促進剤を、ヒト又は動物の所定部位に投与することを特徴とするヒト又は動物の生体内細胞におけるグルタチオンの産生を促進する方法。
- リクイリチン、リクイリチゲニン、イソリクイリチン及びイソリクイリチゲニンを含有し、グリチルリチン酸を実質的に含有しない甘草抽出組成物を製造する方法であって、
甘草エキスからグリチルリチン酸を実質的に除去することを特徴とする製造方法。
