ここで、アルキル基、アリール基、アリールカルボニル基、アルキルカルボニル基、アリールスルホニル基、アルキルスルホニル基は、置換基を有していてもよい炭素原子における各定義と同様である。

R3’及びR4’は、式(1’)で表される光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン骨格中の窒素原子をオニウムとすることができる置換基であれば良く、R3’とR4’は、同一又は相異なる1価の有機基であるか、又は、該アゼピン骨格中の窒素原子とR3’及びR4’が一緒になって環状構造を為す有機基である。
アゼピン骨格中の窒素原子とR3’とR4’が一緒になって環状構造を為す有機基(以下、「環状構造を為す有機基」と呼称する場合がある。)は、アゼピン骨格の窒素原子以外に、置換基を有してもよい炭素原子、及び、必要に応じて、置換基を有してもよい窒素原子、酸素原子、硫黄原子、及び-SO2-基からなる群より選ばれる原子又は基によって構成され、アゼピン骨格中の窒素原子と、それらの原子或いはそれらの原子と基の合計数は5~63である。

[式中、Arは前記と同じ意味を、rcは、水素原子又はC1~C8アルキルを示し、或いはrc同士が一緒になって、ピナコール基等の環状ジアルコキシ基を示す。]で表されるホウ素化合物を、パラジウム触媒と塩基の存在下、適宜な溶媒中において作用させて、化合物(3”)を得る工程である。
式(8)で表されるホウ素化合物は、化合物(3’)に対して、2.0倍モル~10.0倍モル、好ましくは2.1倍モル~5.0倍モル、より好ましくは2.5倍モル~3.5倍モルである。
パラジウム触媒は、ビス[1,2-(ジフェニルホスフィノ)エタン]パラジウム、ビス(ジベンジリデンアセトン)パラジウム、トリス(ジベンジリデンアセトン)ジパラジウム、Pd(PPh3)4等の0価のパラジウム錯体が好適に使用でき、化合物(3’)に対して、0.005当量~0.2当量、好ましくは0.001当量~0.1当量、より好ましくは0.01当量~0.05当量である。
また、パラジウム触媒は、2価パラジウム化合物にホスフィンを作用させることによって、反応系中で調製し使用することもできる。2価パラジウムは、塩化パラジウム、臭化パラジウム等のハロゲン化パラジウム、酢酸パラジウム、アセト酢酸パラジウム等が挙げられ、好ましくは、酢酸パラジウムである。ホスフィンには特に制限は無いが、入手容易なトリアリールホスフィン類が好ましく使用でき、トリフェニルホスフィンがより好ましい。2価パラジウム化合物とホスフィンのモル比率は、1:1~1:10、好ましくは1:2~1:5である。
溶媒は、ベンゼン、トルエン、キシレン等の芳香族系溶媒、エーテル、イソプロピルエーテル、THF、tert-ブチルメチルエーテル、CPME等のエーテル系溶媒、DMF、N-メチルピロリジン-2-オン(NMP)、N,N’-ジメチルイミダゾリジン-2-オン(DMI)等のアミド系溶媒が好ましく使用できる。溶媒量(L)は化合物(3’)(kg)に対して、容積(L)/重量(kg)で好ましくは1.0倍~100倍、より好ましくは3倍~30倍である。
反応は、溶媒の融点から沸点までの適宜な温度で、好ましくは0℃~120℃で、より好ましくは室温~100℃である。
反応は攪拌することにより行われ、反応時間は反応速度に応じて適宜に設定できるが、好ましくは3~50時間、より好ましくは6~24時間である。
ハロゲン化剤は、塩素、N-クロロコハク酸イミド、臭素、N-ブロムコハク酸イミド(NBS)等、ヨウ素等が使用でき、NBSが好適であり、化合物(3”)に対して、好ましくは2.0倍モル~5.0倍モル、より好ましくは2.1倍モル~3.0倍モルである。
反応開始剤として、AIBN、BPO(過酸化ベンゾイル)等のラジカル反応開始剤を使用することが好ましく、化合物(3”)に対して、0.05倍モル~1.0倍モル、より好ましくは0.1倍モル~0.5倍モルである。

式(9)で表されるアミン化合物は、化合物(3''')に対して、0.5倍モル~5.0倍モル、好ましくは0.75倍モル~2.0倍モルである。
塩基は、水酸化リチウム、水酸化ナトリウム、炭酸水素ナトリウム、ナトリウムメトキシド、ナトリウムエトキシド、水酸化カリウム、炭酸カリウム、カリウムメトキシド、カリウムエトキシド、カリウムtert-ブトキシド、水酸化セシウム、炭酸セシウム等の無機塩基が、好ましく使用でき、化合物(3’)に対して、2倍モル~30倍モル、好ましくは3倍モル~20倍モルである。

本反応を相間移動反応条件下に行うことも可能であり、この場合にはテトラブチルアンモニウムブロミド、ベンジルトリメチルアンモニウムブロミド、ベンジルトリエチルアンモニウムブロミド、18-クラウン-6-エーテル、15-クラウン-4-エーテル等の一般的な相間移動触媒を化合物(3''')に対して0.005倍モル~0.2倍モル、好ましくは0.01~0.05倍モル共存させることによる。この場合には前記の塩基を、水溶液或いは固体のままできる。
反応は攪拌することにより行われ、溶媒の融点から沸点までの適宜な温度で、好ましくは室温~100℃である。反応時間は反応速度に応じて適宜に設定できるが、好ましくは3~50時間、より好ましくは6~24時間である。

式(11)で表される2官能性化合物は、式(10)で表される光学活性ビフェニル誘導体に対して、1.0倍モル~5.0倍モル、好ましくは1.05倍モル~2.0倍モルである。
塩基は、水酸化リチウム、水酸化ナトリウム、炭酸水素ナトリウム、ナトリウムメトキシド、ナトリウムエトキシド、水酸化カリウム、炭酸カリウム、カリウムメトキシド、カリウムエトキシド、カリウムtert-ブトキシド、水酸化セシウム、炭酸セシウム等の無機塩基が、好ましく使用でき、化合物(10)に対して、2倍モル~30倍モル、好ましくは3倍モル~20倍モルである。
溶媒は、反応を阻害しなければ特に制限はないが、ヘキサン、シクロへキサン、ベンゼン、トルエン等の炭化水素系、塩化メチレン、クロロホルム、クロロベンゼン等塩素系溶媒、アセトニトリル、ベンゾニトリル等のニトリル系溶媒、アセトン、エチルメチルケトン、tert-ブチルメチルケトン等のケトン系溶媒、DMF、N-メチルピロリジン-2-オン(NMP)、N,N’-ジメチルイミダゾリジン-2-オン(DMI)等のアミド系溶媒、DMSO等が使用できる。溶媒量(L)は式(10)で表される光学活性ビフェニル誘導体(kg)に対して、容積(L)/重量(kg)で好ましくは1.0倍~100倍、より好ましくは5倍~30倍である。
本反応を相間移動反応条件下に行うことも可能であり、この場合にはテトラブチルアンモニウムブロミド、ベンジルトリメチルアンモニウムブロミド、ベンジルトリエチルアンモニウムブロミド、18-クラウン-6-エーテル、15-クラウン-4-エーテル等の一般的な相間移動触媒を式(10)で表される光学活性ビフェニル誘導体に対して0.005倍モル~0.2倍モル、好ましくは0.01~0.05倍モル共存させることによる。この場合には前記の塩基を、水溶液或いは固体のままできる。
反応は攪拌することにより行われ、溶媒の融点から沸点までの適宜な温度で、好ましくは0℃~80℃である。反応時間は反応速度に応じて適宜に設定できるが、好ましくは3~50時間、より好ましくは6~24時間である。
本発明の光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン誘導体の存在下に、以下の反応を行うことができる。

また、「置換基を有していてもよいC6~C14アリール基」の「置換基」としては、ベンゼン環上の置換基であり、その数は単数でも複数でもよく、その種類も単一でも複数でもよく、その置換位置にも特に制限は無いが、原料の入手し易さから、

式(A)で表されるα-置換グリシン誘導体又は式(A’)で表されるグリシン誘導体と式(1’)で表されるアゼピン誘導体のモル比は、20,000/1から10/1、好ましくは10,000/1から100/1、より好ましくは5,000/1から500/1である。
式(A)で表されるα-置換グリシン誘導体又は式(A’)で表されるグリシン誘導体と式(C)の化合物のモル比は、1/0.75から1/10、好ましくは1/1から1/5、より好ましくは1/1.1から1/2である。
式(A)で表されるα-置換グリシン誘導体又は式(A’)で表されるグリシン誘導体と塩基のモル比は、1/1から1/100、好ましくは1/1から1/10、より好ましくは1/1.1から1/3であり、塩基は、水酸化リチウム、水酸化ナトリウム、炭酸水素ナトリウム、ナトリウムメトキシド、ナトリウムエトキシド、水酸化カリウム、炭酸カリウム、カリウムメトキシド、カリウムエトキシド、カリウムtert-ブトキシド、水酸化セシウム、炭酸セシウム等の無機塩基を、単独或いは混合して使用できる。塩基を水溶液として使用する場合には、その濃度(w/w)は、5%から過飽和まで適宜に行われるが、好ましくは、10%から飽和濃度である。
反応は攪拌することにより行われ、溶媒の融点から沸点までの適宜な温度で、好ましくは-10℃~60℃である。反応時間は反応速度に応じて適宜に設定できるが、好ましくは3~60時間、より好ましくは6~24時間である。
キラルビスフェノール(0.5mmol)と炭酸カリウム塩基(1.5~2mmol)をアセトニトリル(50mL)溶媒に溶かした。還流させながら、1,4-ジブロモブタンをゆっくり滴下した。3時間後、アセトニトリルを減圧留去した後、抽出・乾燥をおこなった。減圧濃縮後、シリカゲルショートカラムにて簡単な精製を行い、次工程に用いた(収率87%)。

アリールブロマイド体(S)-1をアルゴン雰囲気にて、酢酸パラジウム(5mol%)p-トリルトリフェニルホスフィン(20mol%)触媒存在下、リン酸カリウム(5e.q.)を塩基として用い、DMF(5mL)溶媒中、アリールボロン酸(1.5mmol)と鈴木カップリングさせた。反応混合物を90℃にて12時間攪拌した。反応終了後、セライトにて触媒をろ過した後、抽出し乾燥を行なった。減圧濃縮の後、カラムクロマトグラフィー(ヘキサン:酢酸エチル=10:1)にて精製し次工程に用いた(収率94%)。

得られた(S)-2に、NBS(2.2e.q.)、AIBN(10mol%)を加え、ベンゼン溶媒中(20mL)、還流下3時間攪拌を行なった。反応終了後、抽出・乾燥・減圧濃縮後、カラムクロマトグラフィー(ヘキサン:酢酸エチル=15:1)にて精製を行った(収率83%)。

触媒前駆体であるビスブロマイド(S)-3に対してn-ジブチルアミンを過剰量(2~3倍モル量)加え、炭酸カリウム(2~3倍モル量)存在下、アセトニトリル溶媒中、40℃にて3時間攪拌を行なった。TLCにて反応終了を確認した後、抽出・乾燥・濃縮を行なった。カラムクロマトグラフィー(塩化メチレン:メタノール=10:1)にて精製した(収率63%)。

実施例1-1と同様にして実施例1-2の化合物を合成した。
実施例1-1と同様にして実施例1-3の化合物を合成した。

反応混合物を抽出・濃縮・乾燥後、カラムクロマトグラフィーにて精製を行った。このときイミン部位が脱保護されたアミン体として目的物を得た。このアミン体を塩化メチレン溶液に溶かし、塩化ベンゾイル(0.25mmol)を滴下した。1時間後、アンモニア水によって反応を終了させた後、抽出・カラム精製(ヘキサン:酢酸エチル=7:1)を行い、ベンゾイル化体を得た。光学純度は、このベンゾイル化体をHPLC分析(キラルセルAD-H、ヘキサン:イソプロパノール:エタノール=90:5:5)することにより決定した(95%ee)。

実施例2-1と同じアラニンシッフ塩基 (0.25mmol)、ベンジルブロマイド(0.3mmol)、実施例2-1と同じ相間移動触媒PTC(0.02mol%)をトルエン溶媒(2mL)に溶かした。混合溶液を激しく攪拌しながら、飽和KOH水溶液(0.5mL)を滴下した。アルゴン雰囲気下、25℃(恒温槽)にて反応が終了するまで激しく攪拌を行なった。反応終了後、有機相に1Nの塩酸を加え、室温にて3時間攪拌を行なった。反応終了後、水相を回収し、炭酸ナトリウムを加えて溶液をアルカリ性にした後、酢酸エチルで抽出し、脱保護されたアミン体を得た。このアミン体を塩化メチレン溶液に溶かし、塩化ベンゾイル(0.25mmol)を滴下した。1時間後、アンモニア水によって反応を終了させた後、抽出・カラム精製(ヘキサン:酢酸エチル=7:1)を行い、ベンゾイル化体を得た。光学純度は、このベンゾイル化体をHPLC分析(キラルセルAD-H、ヘキサン:イソプロパノール:エタノール=90:5:5)することにより決定した(97%ee)。

或いは、R1とR2は一緒になって、Rと同じ定義の2価の基を示す。
R3及びR4は、同一又は相異なって、置換基を有していてもよい炭素原子、及び、必要に応じて、置換基を有していてもよい窒素原子、酸素原子、硫黄原子及び-SO2-基からなる群から選ばれる原子又は基によって構成され、それらの原子或いはそれらの原子と基の合計数が1から30の1価の基を示し、
或いは、R3とR4は一緒になって、-CH2-N+-CH2-と共に合計員数が5から63の環を形成する。
Arは、置換基を有していてもよいC6~C14アリール基、又は、置換基を有していてもよいC3~C8ヘテロアリ-ル基を示す。

(式中R’は、置換基を有していてもよい炭素原子、及び、必要に応じて酸素原子又は-SO2-基からなり、それらの原子或いはそれらの原子と基の合計数が1から8の鎖状の2価の基を示す。)で表される基であることを特徴とする、請求項1又は2記載の光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン誘導体。 - 前記式(1’)又は(1)中の、R1及びR2が、同一又は相異なって、水素原子、ハロゲン原子、置換基を有していてもよい直鎖、又は分岐もしくは環状のC1~C8アルキル基、置換基を有していてもよいC6~C14アリール基であることを特徴とする、請求項1~3のいずれかに記載の光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン誘導体。
- 前記式(1)中の、R3及びR4は、同一又は相異なって、置換基を有していてもよい直鎖、分岐もしくは環状のC1~C30アルキル基又は置換基を有していてもよいC6~C14アリール基、或いは、R3とR4は一緒になって、置換基を有してもよい炭素数2から60の2価の基であることを特徴とする、請求項2~4のいずれかに記載の光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン誘導体。
- 前記式(1’)中の、Arは、置換基を有していてもよいC6~C14アリール基、又は置換基を有していてもよいC3~C8ヘテロアリ-ル基である、請求項1に記載の光学活性6,7-ジヒドロ-5H-ジベンゾ[c,e]アゼピン誘導体。
- 下記式(3)
R1及びR2は、同一又は相異なって、水素原子、ハロゲン原子、置換基を有していてもよい直鎖、分岐もしくは環状のC1~C8アルキル基、置換基を有していてもよいC2~C8アルケニル基、置換基を有していてもよいC2~C8アルキニル基、置換基を有していてもよい直鎖、分岐もしくは環状のC1~C8アルコキシ基、置換基を有していてもよいC6~C14アリール基、置換基を有していてもよいC3~C8ヘテロアリ-ル基、置換基を有していてもよいC1~C8アルキルカルボニル基、置換基を有していてもよいC1~C8アルキルスルホニル基、置換基を有していてもよいC6~C14アリールカルボニル基、又は置換基を有していてもよいC6~C14アリールスルホニル基を示し、或いは、R1とR2は一緒になって、2価の有機基を示す。
