膜安定化剤は、脂質膜を物理的又は化学的に安定させたり、脂質膜の流動性を調 節したりするために添加できる脂質膜の任意成分であり、その含有量は脂質膜構成 成分の総量の通常 0〜70% (モル比)、好ましくは 0〜30% (モル比)、さらに好ましく は 0〜10% (モル比)である。特に糖修飾脂質膜においては、膜安定化剤の含有量 が脂質膜構成成分の総量の 0〜60% (モル比)であることが好ましぐ 0〜30% (モル 比)であることがさらに好ましぐ 0〜 10% (モル比)であることが最も好ましい。糖修飾 脂質膜における膜安定化剤の含有量が上記範囲にあると、核膜孔複合体に結合し た糖の核内への引き抜きによる糖修飾脂質膜の崩壊性を向上させることができるの で、リボソームの内部に封入された目的物質の核膜孔を通じた核内送達効率を向上 させることがでさる。

[0033] 膜融合性機能を付与することができる脂質 (膜融合性脂質)としては、例えば、ジォ レオイルホスファチジルエタノールァミン等の中性脂質;コレステロールコハク酸、力 ルジオリピン等のァ-オン性脂質 (酸性脂質)が挙げられる。リボソームが多重膜リポ ソームであり、当該リボソームが細胞膜、エンドソーム膜等と融合させることを目的とし た脂質膜を糖修飾脂質膜の他に備える場合、当該脂質膜は膜融合性脂質を含有す ることができる。脂質膜に含有される膜融合性脂質は、細胞膜、エンドソーム膜と融 合可能である限り特に限定されるものではない。脂質膜に含有される膜融合性脂質 量は特に限定されるものではないが、脂質膜に含有される総脂質量の通常 40% (モ ル比)以上、好ましくは 70% (モル比)以上、さらに好ましくは 90% (モル比)以上であ る。脂質膜に含有される膜融合性脂質量を上記範囲とすることにより、細胞膜、エンド ソーム膜等に対する十分な融合能を脂質膜に付与することができる。なお、脂質膜 に含有される膜融合性脂質量の上限値は、脂質膜に含有される総脂質量の 100% である。

1.リボソームの作製
(a)ポリカチオンによる DNAの凝集化
DNAとしては、ルシフェラーゼ遺伝子及びその上流に CMVプロモーターを有する 全長 7037bpのプラスミド DNA (CMVプロモーターを有する pcDNA3. 1プラスミド にルシフェラーゼ遺伝子を組み込んだもの)を使用し、ポリカチオンとしては、硫酸プ ロタミンを使用した。 DNA及びポリカチオンをそれぞれ 10mM HEPES緩衝液 (pH 7. 4)に溶解し、 DNA溶液(0. lmgZmL)及びポリカチオン溶液(0. 07mg/mL) をボルテックス下、室温にて混合し、 DNAを凝集化させた。 NZP (nitrogen/phospha te)比 = 1で調製した DNAZポリカチオン複合体(DPC)懸濁液の DNA量は 0. 05 mgZ mLであつ 7こ。

DNAの凝集化の後、 DPC懸濁液 0. lmLを脂質膜にカ卩え、 15分間インキュベー シヨンして水和した。脂質膜は、ジォレオイルホスファチジルエタノールァミン(dioleoy 1 phosphatidyl ethanolamine: DOPE)と、 N—ァセチルダルコサミン導入ポリエチレン グリコールに結合したコレステロール(GlcNAc— PEG— Choi)とを 2 : 1又は 9 : 1 (モ ル比)でクロ口ホルムに溶解して混合した後(総脂質量 55nmol)、脂質膜の表面をォ クタアルギニン (R8)で修飾するためにステアリルォクタアルギニン(STR— R8)溶液 (脂質の 5モル%)を混合液に加え、ガラス試験管内で溶媒を除去することにより形成 させた。水和後の脂質の最終濃度は 0. 55mMであった。

5 [0081] まず、コレステロール 10gをピリジン lOOmLに溶解し、 p-TsCl 10gを加え、室温 で 16時間撹拌した。反応後、得られた液体をシリカゲルクロマトグラフィーで精製する ことによりィ匕合物 1 (収量 13g)を得た。続いて、化合物 l (lg)を真空下にてよく脱気し た後、ジォキサン 18mLに懸濁した。これに巿販のテトラエチレングリコール 2gをカロ え、不活性条件下、オイルバスで 4時間還流した。得られた液体を濃縮後、シリカゲ ルクロマトグラフィーで精製することにより薄黄色透明の液体として化合物 2 (収量 0. 144g)を得た。化合物 2 (120mg)を真空下でよく脱気した後、 CH CI lOmLに溶

解し、活性ィ匕したモレキュラーシーブス (4 A) (2g程度)をカ卩えて撹拌し、これに 3,4,5 —tri—0—Acetyト 1—thiophenyト 2— N—Troc— glucosamine 240mgをカ卩え、続 ヽて N—ヒド 口キシコハク酸イミド 145mg及びトリフルォロメタンスルホン酸(純度 98%,和光純薬 製) 10 μ Lを加えて反応を開始した。不活性及び禁水条件下で 3時間程度反応させ 、得られた反応溶液をクロ口ホルムで希釈後、チォ硫酸ナトリウム飽和水溶液、続い て水で洗浄した。得られた有機層を硫酸ナトリウムで乾燥後、濃縮し、シリカゲルクロ マトグラフィ一にて精製し、化合物 3 (収量 130mg)を得た。化合物 3 (130mg)に無 水酢酸(和光純薬製) 3mL、酢酸(和光純薬製) 3mL、 THF 5mLを加えて室温で 撹拌した。 lOOmMの硫酸銅五水和物水溶液に少量の Znパウダーをくわえ、 Znを 活性化した後、これを濾別したもの (触媒量)を反応溶液に加えて反応を開始し、 5時 間程度反応させた。得られた化合物 4 (収量 180mg)は精製せずに次の反応へ用い た。化合物 4の粗生成物 180mgをメタノール 5mLに溶かし、撹拌した後、 28質量 %ナトリウムメトキシド一メタノール溶液 19 /z Lをカ卩えて反応を開始した。 1時間ほど 室温で撹拌した後、 DOWEX(50WX8- 200 ion-exchange resin) (l〜2g程度)をカロ えて溶液を中和し、続いて綿栓濾過によってこれを取り除いた。得られた溶液を濃縮 後、シリカゲルクロマトグラフィーによって精製し、 目的物である化合物 5 (収量 30mg )を得た。各化合物の同定は NMR及び質量分
祈によって行った。
[0082] (c)超音波処理による凝集化 DNAのパッケージング
DPCを脂質でコーティングするために、ガラス試験管を超音波槽(125 W, Branson Ultrasonics)で約 1分間超音波処理した。こうして、脂質膜の表面に N—ァセチルダ ルコサミン (N ァセチルダルコサミンはポリエチレングリコールを介して脂質膜に結 合する)及びォクタアルギニンを有するリボソームを作製した (粒径: 200nm、ゼータ 電位: + 15mV)。リボソームの流体力学的直径は準弾性光散乱方法によって測定し 、ゼータ電位は、電気泳動的光散乱分光測光器(ELS-8000, Otsuka electronics)に よって測定した。

ヒドロキシ尿素存在条件下(2. 5mM)、細胞にリボソーム(DNAlOpgZcell)を添カロ し、 37°Cで 3時間インキュベーションすることにより、細胞にリボソームをトランスフエク シヨンした。トランスフエクシヨンの 3時間後及び 6時間後、細胞を洗浄し、 reporter lysi s buffer (Promega社) 75 μ Lで溶解し、細胞溶解液 20 μ Lにルシフェラーゼアツセィ 試薬(Promega社) 50 μ Lをカ卩えて、ルミノメーター(Luminescencer- PSN)によりルシ フェラーゼ活性を測定した。また、 BCAプロテインアツセィキット(PIERCE社)により細 胞溶解液中のタンパク質量を測定した。トランスフエクシヨン 3時間後の遺伝子発現量 を図 2 (a)に、トランスフエクシヨン 6時間後の遺伝子発現量を図 2 (b)に示す。 [0088] 図 2 (a)に示すように、トランスフエクシヨン 3時間後、脂質膜の表面に N—ァセチル ダルコサミンもォクタアルギニンも有しな 、リボソーム、脂質膜の表面にォクタアルギ ニンを有するが N ァセチルダルコサミンは有しないリボソーム、及び脂質膜の表面 に N ァセチルダルコサミンを有するがォクタアルギニンは有しないリボソームは、脂 質組成に関わらず、遺伝子発現は全く認められな力つた。一方、脂質膜の表面に N —ァセチルダルコサミン及びォクタアルギニンを有するリボソームは、 DOPE: GlcN Ac— PEG— Chol= 2 : 1の場合には遺伝子発現は全く認められなかった力 DOPE : GlcNAc PEG Chol= 9: 1の場合には有意な遺伝子発現が認められた。
[0089] また、図 2 (b)に示すように、トランスフエクシヨン 6時間後、脂質膜の表面に N ァセ チルダルコサミンもォクタアルギニンも有しな ヽリボソーム、及び脂質膜の表面に N— ァセチルダルコサミンを有するがォクタアルギニンは有しな 、リボソームは、脂質組成 に関わらず、遺伝子発現が極めて低かった。脂質膜の表面にォクタアルギニンを有 するが N—ァセチルダルコサミンは有しないリボソームは、トランスフエクシヨン 3時間 後よりも若干高い遺伝子発現が認められた。一方、脂質膜の表面に N—ァセチルダ ルコサミン及びォクタアルギニンを有するリボソームは、 DOPE : GlcNAc— PEG— C hol= 2 : 1の場合、脂質膜の表面にォクタアルギニンを有するが N ァセチルダルコ サミンは有しな 、リボソーム(DOPE: Chol= 2: 1)の遺伝子発現よりも約 11倍高!ヽ 遺伝子発現が認められ、 DOPE: GlcNAc— PEG— Chol= 9 : 1の場合、脂質膜の 表面に N ァセチルダルコサミン及びォクタアルギニンを有するリボソーム(DOPE : GlcNAc - PEG - Chol= 2 : 1)の遺伝子発現よりも約 13倍高 、(すなわち、脂質膜 の表面にォクタアルギニンを有するが N ァセチルダルコサミンは有しないリボソーム (DOPE: Choi = 2: 1)の遺伝子発現よりも約 141倍高 ヽ)遺伝子発現が認められた
[0090] 脂質膜の表面にォクタアルギニンを有するが N ァセチルダルコサミンは有しな!/ヽ リボソームの遺伝子発現はほとんど認められな力つたことから、 N ァセチルダルコサ ミンは、細胞表面レセプター認識によりリボソームの細胞内取り込みを促進する素子 として機能するわけではなぐォクタアルギニンの機能によりリボソームが細胞内に取 り込まれた後に、リボソームの核移行を促進する素子として機能すると考えられる。脂 質膜の表面に存在する糖が核膜孔複合体へ結合することにより、リボソームが核移 行し、核膜孔複合体に結合した糖が核内へ引き抜かれることにより、脂質膜が崩壊し てリボソームの内部に封入された目的物質は核膜孔を通じて核内に放出されると考 えられる。リボソームの核移行には、主として、糖の核膜孔複合体への結合が関与し ていると考えられ、リボソーム封入物質の核内送達には、主として、核膜孔複合体に 結合した糖の核内への引き抜きによる脂質膜の崩壊、及び脂質膜に含有される膜安 定化剤 (例えばコレステロール等)の量の減少による脂質膜の崩壊性の向上が関与 しているものと考えられる。

DOPEと、コレステロール(Choi)と、糖導入ポリエチレングリコールが結合したコレ ステロール(Sugar— PEG— Choi)とを 5 :5:0、 5:4:1、 5:3:2、 5:2:3、 5:1:4又 は 5: 0: 5 (モル比)でクロ口ホルムに溶解して混合して (総脂質量 55nmol)脂質膜を 形成させた点を除き、実施例 1と同様の操作を行い、脂質膜の表面に糖 (糖はポリエ チレングリコールを介して脂質膜に結合する)及びォクタアルギニンを有するリポソ一 ムを作製した(粒径: 220nm、ゼータ電位: + 10mV)。

[0099] 図 3に示すように、糖が N—ァセチルダルコサミンであるとき、脂質膜の表面に存在 する糖の量が脂質膜に含有される総脂質量の 10% (モル比)である場合に最も高い 遺伝子発現が認められた。なお、図 3中、「★」は対照であり、「X 127」等は対照に対 する遺伝子発現量を示す。また、図 4に示すように、糖がマンノースであるとき、脂質 膜の表面に存在する糖の量が脂質膜に含有される総脂質量の 40% (モル比)である 場合に最も高い遺伝子発現が認められた。なお、図 4中、「★」は対照であり、「X 12 10」等は対照に対する遺伝子発現量を示す。また、図 5に示すように、糖がガラクトー スであるとき、脂質膜の表面に存在する糖の量が脂質膜に含有される総脂質量の 10 % (モル比)である場合に最も高い遺伝子発現が認められた。なお、図 5中、「★」は 対照であり、「X 116」等は対照に対する遺伝子発現量を示す。

コレステロール(Choi)及び糖導入ポリエチレングリコールが結合したコレステロ一 ル(GlcNAc— PEG— Choi, Man -PEG -Choi, Gal— PEG— Choi)力脂質膜 に含有される (残りの成分は DOPE)リボソームを、 Choi含有量を変化させながら実 施例 1及び 2と同様にして調製し、リボソームを細胞にトランスフエクシヨンした後(6時 間後)の遺伝子発現量を実施例 1及び 2と同様にして測定した。なお、 GlcNAc— P EG— Choiの量は、脂質膜に含有される総脂質量の 10% (モル比)、 Man— PEG— Choiの量は、脂質膜に含有される総脂質量の 30% (モル比)、 Gal— PEG— Choi の量は、脂質膜に含有される総脂質量の 10% (モル比)とした。
[0101] その結果、図 6に示すように、糖の種類に関わらず、 Choi含有量が小さくなると、遺 伝子発現量が増加した。脂質膜の安定性が低減することにより、核膜上における脂 質膜の崩壊性が向上したため、リボソームの内部に封入された遺伝子の核内送達効 率を向上したものと考えられる。

DOPE :糖導入ポリエチレングリコールが結合したコレステロール(GlcNAc— PEG — Chol、 Man— PEG— Chol、 Gal -PEG -Choi) = 9 : 1の脂質組成からなるクロ 口ホルム溶液に、脂質膜の表面をォクタアルギニン (R8)で修飾するためにステアリ ルォクタアルギニン(STR—R8)溶液 (脂質の 5モル%)を加え、ガラス試験管内で溶 媒を除去することにより脂質膜を形成させた。この脂質膜に対して、実施例 1で調製 した DPC懸濁液をカ卩えて 15分間インキュベーションした後、超音波槽(125 W, Bran son Ultrasonics)で約 1分間超音波処理して、上記糖及びォクタアルギニンを有する リボソームを調製した。また、糖導入ポリエチレングリコールが結合したコレステロール に代えてコレステロールを使用したコントロールリボソームを調製した。
[0103] トランスフエクシヨンの 48時間前に 24穴プレートに 2 X 104 cells/500 μ L DMEM ( 10%血清を含む) Zdishとなるように HeLa細胞を播種し、 37°C、 5% CO環境下で

ュペートした。 PBS 0. 25mLで 3回細胞を洗浄したのち、 Reporter lysis buffer 75 μ Lを添加し、— 80°Cで約 30分凍結した。 30分後凍結したプレートを室温で融解させ 、氷上にてセルスクレーパーを用いて細胞を剥がしとり、細胞溶解液を回収し、上清 20 μ 1当たりのルシフェラーゼ活性 (RLU)を測定した。さらに上清を DDWで 5倍希 釈し、 BCA法によりタンパク定量を行い、得られる RLUをタンパク量で除することによ つて単位細胞当たりのルシフェラーゼ活性(RLU/mg protein)を算出した。
[0105] この結果、 GlcNAc、 Mannose、 Galactose, V、ずれの糖修飾リボソームにお!/、て も、過剰量の糖の存在下での遺伝子発現の変動は認められず、リボソームに含まれ る糖が細胞への取り込み過程にぉ 、て機能をして 、な 、ことが示唆された(図 7)。こ の実験結果は、本発明のリボソームが糖レセプターを介して細胞に取り込まれるので はなぐォクタアルギニンの機能により細胞内に導入され、細胞内に取り込まれた後 に糖が機能することを示すものである。

Label IT CX— Rhodamine Labeling Kit (Penverna Corp.)を用いて、添付のプロトコ一 ルに従い、実施例 1と同じプラスミド DNAをローダミン (Rhodamine)標識し、さらに 実施例 1と同様の操作を行って、ローダミン修飾 DPCを調製した。このローダミン修 飾 DPCを用 、、クロ口ホルム溶液に総脂質量の 1 %モルとなるように NBD— DOPE ( AVANTI POLAR LIPIDS) を添カ卩した他は全て実施例 4に示した方法によって、 NB Dで脂質がラベルされ、ローダミン修飾 DPCを封入した糖修飾リボソームを調製した

で、曝露させた。細胞を 20UnitsZmLへパリン(PBS溶液) lmLで 3回洗浄した後、 1ェ mgZmL Hoechst 33342(水溶液)を終濃度 5 gZmLとなるように添カ卩し、室温 、遮光条件下で 10分間インキュベートすることで核を染色した。 PBSで細胞を洗浄し たのち、培地を DMEMZF— 12に交換し、共焦点レーザースキャン顕微鏡にて解 祈した。
[0109] その結果、糖修飾を受けていないリボソームでは遺伝子の核への局在は全く認めら れなかったのに対して、糖修飾を受けたリボソームでは、遺伝子の核への局在が認 められた(図 8)。

[2] 前記糖が親水性ポリマーを介して前記脂質膜の表面に結合している請求の範囲第 1 項記載のベクター。
[3] 前記糖が 1〜20個の糖残基力もなる請求の範囲第 1項又は第 2項記載のベクター。
[4] 前記糖が単糖である請求の範囲第 1項又は第 2項記載のベクター。
[5] 前記糖がアミノ糖である請求の範囲第 1項又は第 2項記載のベクター。
[6] 前記糖の量が、前記脂質膜に含有される総脂質量の 0. 5〜65% (モル比)である請 求の範囲第 1項〜第 5項のいずれかに記載のベクター。
[7] 前記脂質膜に含有される膜安定化剤の量が、前記脂質膜構成成分の総量の 0〜60
% (モル比)である請求の範囲第 1項〜第 6項のいずれかに記載のベクター。
[8] 核内へ送達しょうとする目的物質が前記リボソームの内部に封入されている請求の 範囲第 1項〜第 7項のいずれかに記載のベクター。
[9] 請求の範囲第 8項記載のベクターを含有する目的物質の核内送達用組成物。
