背景技術
[0002] 遺伝子組換え技術の発達によって、種々のタンパク質製剤が安定した供給量で提 供されるようになった。これらの製剤は安定性を確保するため、凍結乾燥したタンパク 質成分粉末とこれを溶解するための水溶性希釈液とを別途包装し、使用時に溶解す る形態で提供されるか、あるいは安定性を向上させるための添加剤を加えたタンパク 質溶液製剤の形で提供されて 、る。
[0003] 近年、種々の抗体製剤が開発され実用に供されているが、多くの抗体製剤は静脈 注射用製剤として用いられている。一方、医療現場のニーズにより、抗体含有製剤を 自己注射可能な皮下注射用製剤として開発する要望が高くなつている。
[0004] 皮下注射用の抗体含有製剤を設計するにあたっては、 1回あたりの抗体投与量が 大量となる一方で (100〜200mg-程度)、皮下注射では一般的に注射液量の制限があ ることから、投与液中の抗体の高濃度化が必要となる。そこで、凍結乾燥濃縮を利用 した高濃度の抗体含有製剤の開発が求められている。

用により粘度の高い溶液を形成する傾向にある。さらに、凍結乾燥製剤の製造にお いては、そのケーキ形成や安定性を維持するために、通常、糖などの凍結乾燥保護 剤(リオプロテクタント)を大量に添加した状態で凍結乾燥される。しかし、糖は分子間 相互作用を増強し、粘度を増大させることになり、高い粘度の製剤は、調剤、シリンジ への吸引及び皮下注射を行うのが困難となる。したがって、糖類の添加を伴わずに、 かつ安定な凍結乾燥製剤を製造する新規な方法が求められている。

[0008] 本発明の目的は、糖類の添加を伴わず、かつ凍結乾燥濃縮を利用した高濃度の 抗体含有製剤を製造する工程及び得られた高濃度の凍結乾燥製剤の保存時、及び 再溶解時にぉ 、て、抗体の二量体や低分子量分解物などの生成が少な!、安定な抗 体製剤を提供することである。
課題を解決するための手段
[0009] 上記目的を達成するために鋭意研究した結果、本発明者らは、アルギニン、ヒスチ ジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオニンからなる群力も選ば れる 1種または 2種以上のアミノ酸又はその塩を添加することにより、賦形剤として糖 類を添加しなくても高濃度の抗体含有凍結乾燥製剤となしうることを見いだし本発明 を完成した。
[0010] すなわち、本発明は以下のものを提供する。
(1)賦形剤として還元糖、非還元糖、糖アルコールまたは多糖類を含まず、アルギ- ン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオニンからなる群 力 選ばれる 1種または 2種以上のアミノ酸又はその塩を含む抗体含有凍結乾燥製 剤。
(2)凍結乾燥製剤の再溶解後の抗体濃度が lOmgZmL以上である(1)に記載の製 剤。
(3)凍結乾燥製剤の再溶解後の抗体濃度が 50mgZmL以上である(2)に記載の製 剤。
(4)凍結乾燥製剤の再溶解後の抗体濃度が lOOmgZmL以上である(3)に記載の
製剤。
(5)アミノ酸又はその塩の含有量力 抗体 1モルに対して 270モル以上である(1)一 ( 4)の 、ずれかに記載の製剤。
(6)アミノ酸又はその塩の含有量力 抗体 1モルに対して 380モル以上である(5)に 記載の製剤。
(7)アミノ酸又はその塩の含有量力 抗体 1モルに対して 540モル以上である(6)に 記載の製剤。
(8)再溶解後における溶液の pHが 4〜8である(1)一(7)のいずれかに記載の製剤
(9)再溶解後における溶液の pHが 5. 0〜7. 5である(8)に記載の製剤。
(10)抗体がキメラ抗体、ヒト化抗体又はヒト抗体である(1)一(9)のいずれかに記載 の製剤。
(11)抗体が抗 IL 6レセプター抗体である(10)に記載の製剤。
(12)アミノ酸又はその塩力 アルギニン、ヒスチジン及びリジン力もなる群力も選ばれ る 1種または 2種以上のアミノ酸又はその塩である(1)一(11)のいずれかに記載の製 剤。
(13)アミノ酸又はその塩が、アルギニン又はその塩である(12)に記載の製剤。
(14)再溶解後における粘度が 20mPa' s以下である(1) - (13)のいずれかに記載 の製剤。
(15)再溶解後における粘度が 15mPa · s以下である(14)に記載の製剤。
(16)再溶解後における粘度が 12mPa · s以下である( 15)に記載の製剤。
(17)アルギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ ニン力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテク タント或いは賦形剤として含む抗体含有凍結乾燥製剤であって、製剤中の抗体濃度 が 20mg/ml以上のときはアミノ酸又はその塩を 25mg/ml以上、抗体濃度が 30mg/ml以 上のときはアミノ酸又はその塩を 12.5mg/ml以上、抗体濃度が 40mg/ml以上のときは アミノ酸又はその塩を 6.25mg/ml以上含んでいることを特徴とする前記抗体含有凍結 乾燥製剤。
(18)製剤中の抗体濃度が 30mg/ml以上のときはアミノ酸又はその塩を 25mg/ml以上 、抗体濃度が 40mg/ml以上のときはアミノ酸又はその塩を 12.5mg/ml以上含んで 、る ことを特徴とする(17)記載の製剤。
(19)アルギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ ニン力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテク タント或いは賦形剤として含む抗体含有凍結乾燥製剤の製造方法であって、還元糖 、非還元糖、糖アルコールおよび多糖類の総濃度が 20mgZmL未満である凍結乾 燥前溶液を凍結乾燥する工程を含む、前記抗体含有凍結乾燥製剤の製造方法。
(20)アルギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ ニン力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテク タント或いは賦形剤として含む抗体含有凍結乾燥製剤であって、バイアル当たりの抗 体重量と還元糖、非還元糖、糖アルコールおよび多糖類の総重量との比が 1 :0. 5 未満である前記抗体含有凍結乾燥製剤。

発明を実施するための最良の形態
[0011] 本発明の製剤および方法に使用される抗体は、所望の抗原と結合する限り特に制 限はなぐポリクローナル抗体であってもモノクローナル抗体であってもよいが、均質 な抗体を安定に生産できる点でモノクローナル抗体が好ましい。
[0012] 本発明で使用されるモノクローナル抗体としては、ヒト、マウス、ラット、ノ、ムスター、 ゥサギ、ヒッジ、ラタダ、サル等の動物由来のモノクローナル抗体だけでなぐキメラ抗 体、ヒト化抗体、 bispedfic抗体など人為的に改変した遺伝子組み換え型抗体も含ま れる。また、抗体の免疫グロブリンクラスは特に限定されるものではなぐ IgGl、 IgG2 、 IgG3、 IgG4などの IgG、 IgA、 IgD、 IgE、 IgMなどいずれのクラスでもよいが、 IgG 及び IgMが好ましい。

[0015] モノクローナル抗体を産生するハイプリドーマは、基本的には公知技術を使用し、 以下のようにして作製できる。すなわち、所望の抗原や所望の抗原を発現する細胞 を感作抗原として使用して、これを通常の免疫方法にしたがって免疫し、得られる免 疫細胞を通常の細胞融合法によって公知の親細胞と融合させ、通常のスクリーニン グ法により、モノクローナルな抗体産生細胞 (ノヽイブリドーマ)をスクリーニングすること によって作製できる。ノ、イブリドーマの作製は、たとえば、ミルスティンらの方法 (Kohle r. G. and Milstein, C, Methods Enzymol. (1981) 73: 3-46 )等に準じて行うことがで きる。抗原の免疫原性が低い場合には、アルブミン等の免疫原性を有する巨大分子 と結合させ、免疫を行えばよい。
[0016] また、抗体遺伝子をハイプリドーマ力 クローユングし、適当なベクターに組み込ん で、これを宿主に導入し、遺伝子組換え技術を用いて産生させた遺伝子組換え型抗 体を用いることができる(例えば、 Carl, A. K. Borrebaeck, James, W. Larrick, THER APEUTIC MONOCLONAL ANTIBODIES, Published in the United Kingdom by MA
CMILLAN PUBLISHERS LTD, 1990参照)。具体的には、ハイプリドーマの mRNAか ら逆転写酵素を用いて抗体の可変領域 (V領域)の cDNAを合成する。目的とする抗 体の V領域をコードする DNAが得られれば、これを所望の抗体定常領域 (C領域)を コードする DNAと連結し、これを発現ベクターへ組み込む。または、抗体の V領域を コードする DNAを、抗体 C領域の DNAを含む発現ベクターへ組み込んでもよい。発 現制御領域、例えば、ェンハンサー、プロモーターの制御のもとで発現するよう発現 ベクターに組み込む。次に、この発現ベクターにより宿主細胞を形質転換し、抗体を 発現させることができる。
[0017] 本発明では、ヒトに対する異種抗原性を低下させること等を目的として人為的に改 変した遺伝子組換え型抗体、例えば、キメラ (Chimeric)抗体、ヒトイ匕 (Humanized )抗 体などを使用できる。これらの改変抗体は、既知の方法を用いて製造することができ る。キメラ抗体は、ヒト以外の哺乳動物、例えば、マウス抗体の重鎖、軽鎖の可変領域 とヒト抗体の重鎖、軽鎖の定常領域からなる抗体であり、マウス抗体の可変領域をコ ードする DNAをヒト抗体の定常領域をコードする DNAと連結し、これを発現ベクター に組み込んで宿主に導入し産生させることにより得ることができる。
[0018] ヒト化抗体は、再構成 (reshaped)ヒト抗体とも称され、ヒト以外の哺乳動物、たとえば マウス抗体の相補性決定領域(CDR; complementarity determining region)をヒト抗 体の相補性決定領域へ移植したものであり、その一般的な遺伝子組換え手法も知ら れている。具体的には、マウス抗体の CDRとヒト抗体のフレームワーク領域(framewor k region ;FR)を連結するように設計した DNA配列を、末端部にオーバーラップする 部分を有するように作製した数個のオリゴヌクレオチドから PCR法により合成する。得 られた DNAをヒト抗体定常領域をコードする DNAと連結し、次いで発現ベクターに 組み込んで、これを宿主に導入し産生させることにより得られる(欧州特許出願公開 番号 EP 239400、国際特許出願公開番号 WO 96/02576参照)。 CDRを介して連結 されるヒト抗体の FRは、相補性決定領域が良好な抗原結合部位を形成するものが選 択される。必要に応じ、再構成ヒト抗体の相補性決定領域が適切な抗原結合部位を 形成するように抗体の可変領域のフレームワーク領域のアミノ酸を置換してもよ ヽ(Sa to, K.et al" Cancer Res. (1993) 53, 851—856)。
[0019] また、ヒト抗体の取得方法も知られて 、る。例えば、ヒトリンパ球を in vitroで所望の 抗原または所望の抗原を発現する細胞で感作し、感作リンパ球をヒトミエローマ細胞 、例えば U266と融合させ、抗原への結合活性を有する所望のヒト抗体を得ることもで きる(特公平 1-59878参照)。また、ヒト抗体遺伝子の全てのレパートリーを有するトラ ンスジエニック動物を抗原で免疫することで所望のヒト抗体を取得することができる( 国際特許出願公開番号 WO 93/12227, WO 92/03918, WO 94/02602, WO 94/255 85, WO 96/34096, WO 96/33735参照)。さらに、ヒト抗体ライブラリーを用いて、パン ニングによりヒト抗体を取得する技術も知られている。例えば、ヒト抗体の可変領域を 一本鎖抗体 (scFv)としてファージディスプレイ法によりファージの表面に発現させ、 抗原に結合するファージを選択することができる。選択されたファージの遺伝子を解 析すれば、抗原に結合するヒト抗体の可変領域をコードする DNA配列を決定すること ができる。抗原に結合する scFvの DNA配列が明らかになれば、当該配列を含む適当 な発現ベクターを作製し、ヒト抗体を取得することができる。これらの方法は既に衆知 であり、 WO 92/01047, WO 92/20791, WO 93/06213, WO 93/11236, WO 93/1917 2, WO 95/01438, WO 95/15388を参考にすることができる。
[0020] 抗体遺伝子を一旦単離し、適当な宿主に導入して抗体を作製する場合には、適当 な宿主と発現ベクターの組み合わせを使用することができる。真核細胞を宿主として 使用する場合、動物細胞、植物細胞、真菌細胞を用いることができる。動物細胞とし ては、(1)哺乳類細胞、例えば、 CHO, COS,ミエローマ、 BHK (baby hamster kidney ) , HeLa, Vero, (2)両生類細胞、例えば、アフリカッメガエル卵母細胞、あるいは (3) 昆虫細胞、例えば、 sl9, sf21, Tn5などが知られている。植物細胞としては、ニコティア ナ(Nicotiana)属、例えば-コティアナ ·タパカム(Nicotiana tabacum)由来の細胞が知 られており、これをカルス培養すればよい。真菌細胞としては、酵母、例えば、サッカ 口ミセス (Saccharomyces )属、例えばサッカロミセス 'セレビシェ (Saccharomyces sere visiae)、糸状菌、例えば、ァスペルギルス(Aspergillus )属、例えばアスペスギルス. 二ガー (Aspergillus niger )などが知られている。原核細胞を使用する場合、細菌細 胞を用いる産生系がある。細菌細胞としては、大腸菌 . coli )、枯草菌が知られて いる。これらの細胞に、目的とする抗体遺伝子を形質転換により導入し、形質転換さ
れた細胞を in vitroで培養することにより抗体が得られる。

しかし、本発明を、凍乾前より少ない容量の水を用いて凍結乾燥製剤を再溶解する ことにより高濃度溶液製剤を調製する、いわゆる凍乾濃縮技術を適用する場合には 、凍乾前の抗体濃度は lmgZmL以上であるものが好ましぐさらには lOmgZmL 以上であるものが好ましぐ特に凍乾前の抗体濃度が 20mgZmL以上であるものが 好ましい。また、凍乾濃縮技術における抗体の濃縮倍率は 2〜50倍であることが好ま しぐさらには 2〜20倍であることが好ましぐ特に抗体の濃縮倍率が 2〜6倍であるこ とが好ましい。

[0033] 本発明の製剤では、賦形剤として還元糖、非還元糖、糖アルコールまたは多糖類 を添加しないことを特徴とする。通常、凍結乾燥製剤の賦形剤として還元糖、非還元 糖、糖アルコールまたは多糖類をタンパク質含有製剤に添加する場合、凍乾前溶液 として 20mgZmL以上必要である。従って、本発明における「賦形剤として還元糖、 非還元糖、糖アルコールまたは多糖類を含まない」とは、タンパク質含有凍結乾燥製 剤の製造時に凍乾前溶液に含まれる還元糖、非還元糖、糖アルコールまたは多糖 類の合計の濃度が 20mgZmL未満、好ましくは lOmgZmL以下、さらに好ましくは 5mgZmL以下、さらに好ましくは lmgZmL以下、特に好ましくは実質的に含まな いものを意味する。
[0034] 本発明はまた、アルギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン 及びスレオニン力 なる群力 選ばれる 1種または 2種以上のアミノ酸又はその塩をリ ォプロテクタント或いは賦形剤として含む抗体含有凍結乾燥製剤であって、ノ ィアル 当たりの抗体重量と還元糖、非還元糖、糖アルコールおよび多糖類の総重量との比 が 1 : 0. 5未満である前記抗体含有凍結乾燥製剤を提供する。バイアル当たりの抗 体重量と還元糖、非還元糖、糖アルコールおよび多糖類の総重量との比が好ましく は 1 : 0. 5未満であり、より好ましくは 1 : 0. 25以下であり、さらに好ましくは 1 : 0. 125 以下であり、さらに好ましくは 1 : 0. 025以下であり、最も好ましくは還元糖、非還元糖 、糖アルコールおよび多糖類を含まない。
[0035] ここで言う、還元糖としては、グルコース、フルクトース、マルトース、ラタトース;非還 元糖としては、スクロース、トレハロース、ラフイノース、ネオトレハロース;糖アルコー ルとしては、マンニトール、ソルビトール、マルチトール、エリスリトール;多糖類として

リオプロテクタント或いは賦形剤として含む抗体含有凍結乾燥製剤であって、製剤中 の抗体濃度が 20mg/ml以上のときはアミノ酸又はその塩を 25mg/ml以上、抗体濃度 が 30mg/ml以上のときはアミノ酸又はその塩を 12.5mg/ml以上、抗体濃度が 40mg/ml 以上のときはアミノ酸又はその塩を 6.25mg/ml以上含んでいることを特徴とする前記 抗体含有凍結乾燥製剤を提供する。製剤中の抗体濃度が 30mg/ml以上のときはアミ ノ酸又はその塩を 25mg/ml以上、抗体濃度が 40mg/ml以上のときはアミノ酸又はその 塩を 12.5mg/ml以上含んで!/、ることが好まし!/、。

実施例 1:凍結乾燥製剤に対して安定化効果を有する賦形剤種の選択
抗 IL-6レセプターヒト化抗体を含む凍結乾燥製剤について、凍結乾燥工程前後及 び製剤自体の安定化を目的として、安定化効果を有する賦形剤種の検討を行った。

7)、セリン (試料 No.9)及びプロリン (試料 No.10)を添カ卩した試料について、凍結乾燥 前後における二量体の増加量が明らかに抑制されており、二量体生成抑制効果を 確認することができた。これらの効果は一般的な凍結乾燥保護剤として知られて 、る スクロース或いはトレハロースよりも高かった。なお、何れの試料についても、低分子 量分解物はほとんど認められなカゝつた。表 1において、 Dimerは二量体を、 LMWは低 分子量分解物を、 Preは主成分よりも短 、保持時間で溶出されるピークの総和をそれ ぞれ表す。

実施例 7:凍結乾燥製剤再溶解液の粘度に賦形剤種類が及ぼす影響
抗 IL- 6レセプターヒト化抗体を含む凍結乾燥製剤にっ ヽて、賦形剤種類が再溶解 液の粘度に及ぼす影響を評価した。

[0132] 本検討では、実施例 2で評価した試料のうち試料 No. 12について、以下に示したよ うに添加する注射用水の量を変動させることにより、試料 No. 49 53からなる抗体濃 度の異なる再溶解液試料を調製した。また、抗 IL-6レセプターヒト化抗体及びスクロ ースを含む凍結乾燥製剤である実施例 2で評価した試料 No. 16について、試料 No.5 4として抗体濃度が約 200 mg/mLとなるように調製した再溶解液試料の粘度を参考と して測定した。粘度測定については、実施例 7と同一の手法を用いた。

粘度測定結果を表 13に示した。抗体濃度が増大すると共に粘度は増大したが、抗 体濃度 200 mg/mLにおける粘度は 16.9 mPa'sとなり、スクロースを含む抗体濃度 195 mg/mLの再溶解液と比較して低値を示した。このように、アルギニン処方とスクロース 処方を比較すると、アルギニンの方が高モル濃度であるにもかかわらず高抗体濃度
における粘度は低ぐアルギニンは抗体溶液の粘度を低減させる可能性が示唆され た。

請求の範囲
[I] 賦形剤として還元糖、非還元糖、糖アルコールまたは多糖類を含まず、アルギニン
、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオニンからなる群か ら選ばれる 1種または 2種以上のアミノ酸又はその塩を含む抗体含有凍結乾燥製剤
[2] 凍結乾燥製剤の再溶解後の抗体濃度が lOmgZmL以上である請求項 1に記載の 製剤。
[3] 凍結乾燥製剤の再溶解後の抗体濃度が 50mgZmL以上である請求項 2に記載の 製剤。
[4] 凍結乾燥製剤の再溶解後の抗体濃度が lOOmgZmL以上である請求項 3に記載 の製剤。
[5] アミノ酸又はその塩の含有量力 抗体 1モルに対して 270モル以上である請求項 1
4のいずれかに記載の製剤。
[6] アミノ酸又はその塩の含有量力 抗体 1モルに対して 380モル以上である請求項 5 に記載の製剤。
[7] アミノ酸又はその塩の含有量力 抗体 1モルに対して 540モル以上である請求項 6 に記載の製剤。
[8] 再溶解後における溶液の pH力 〜8である請求項 1—7のいずれかに記載の製剤
[9] 再溶解後における溶液の pHが 5. 0〜7. 5である請求項 8に記載の製剤。
[10] 抗体がキメラ抗体、ヒト化抗体又はヒト抗体である請求項 1 9の!、ずれかに記載の 製剤。
[II] 抗体が抗 IL 6レセプター抗体である請求項 10に記載の製剤。
[12] アミノ酸又はその塩が、アルギニン、ヒスチジン及びリジン力もなる群力も選ばれる 1 種または 2種以上のアミノ酸又はその塩である請求項 1 11のいずれかに記載の製 剤。
[13] アミノ酸又はその塩が、アルギニン又はその塩である請求項 12に記載の製剤。
[14] 再溶解後における粘度が 20mPa's以下である請求項 1— 13のいずれかに記載の
製剤。
[15] 再溶解後における粘度が 15mPa' s以下である請求項 14に記載の製剤。
[16] 再溶解後における粘度が 12mPa' s以下である請求項 15に記載の製剤。
[17] ァノレギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ-ン 力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテクタント 或いは賦形剤として含む抗体含有凍結乾燥製剤であって、製剤中の抗体濃度が 20 mg/ml以上のときはアミノ酸又はその塩を 25mg/ml以上、抗体濃度が 30mg/ml以上の ときはアミノ酸又はその塩を 12.5mg/ml以上、抗体濃度が 40mg/ml以上のときはァミノ 酸又はその塩を 6.25mg/ml以上含んでいることを特徴とする前記抗体含有凍結乾燥 製剤。
[18] 製剤中の抗体濃度が 30mg/ml以上のときはアミノ酸又はその塩を 25mg/ml以上、抗 体濃度が 40mg/ml以上のときはアミノ酸又はその塩を 12.5mg/ml以上含んでいること を特徴とする請求項 17記載の製剤。
[19] ァノレギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ-ン 力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテクタント 或いは賦形剤として含む抗体含有凍結乾燥製剤の製造方法であって、還元糖、非 還元糖、糖アルコールおよび多糖類の総濃度が 20mgZmL未満である凍結乾燥前 溶液を凍結乾燥する工程を含む、前記抗体含有凍結乾燥製剤の製造方法。
[20] ァノレギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ-ン 力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテクタント 或いは賦形剤として含む抗体含有凍結乾燥製剤であって、バイアル当たりの抗体重 量と還元糖、非還元糖、糖アルコールおよび多糖類の総重量との比が 1 : 0. 5未満 である前記抗体含有凍結乾燥製剤。
[21] 賦形剤として還元糖、非還元糖、糖アルコールまたは多糖類を含まず、アルギニン 、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオニンからなる群か ら選ばれる 1種または 2種以上のアミノ酸又はその塩を含むタンパク質含有凍結乾燥 製剤。
[22] ァノレギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオ-ン
力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテクタント 或いは賦形剤として含むタンパク質含有凍結乾燥製剤の製造方法であって、還元糖 、非還元糖、糖アルコールおよび多糖類の総濃度が 20mgZmL未満である凍結乾 燥前溶液を凍結乾燥する工程を含む、前記タンパク質含有凍結乾燥製剤の製造方 法。
ァノレギニン、ヒスチジン、リジン、セリン、プロリン、グリシン、ァラニン及びスレオニン 力もなる群力も選ばれる 1種または 2種以上のアミノ酸又はその塩をリオプロテクタント 或いは賦形剤として含むタンパク質含有凍結乾燥製剤であって、ノ ィアル当たりのタ ンパク質重量と還元糖、非還元糖、糖アルコールおよび多糖類の総重量との比が 1 : 0. 5未満である前記タンパク質含有凍結乾燥製剤。
