IgG1 항체 제제 (70mg/ml)(항체 B) 및 IgG2 항체 제제 (70mg/ml)(항체 C)가 10,000 MWCO 투석 튜우빙을 사용하여 각각 20mM 아세트산나트륨 pH 4.7 및 4.9에 대해 투석되었다. 항체 샘플은 30,000 MWCO 원심 필터 및 Allegra X-12R 원심분리기를 사용하여 농축되었다. 항체 샘플 농도는 Agilent UV/Vis 분광측정기를 사용하여 측정되었다. 부형제-함유 샘플은 예비-칭량된 고체 부형제로 벌크 농축된 용액을 첨가함에 의해 제조되었다. 점성 샘플은 양성 변위 피펫을 사용하여 이동되었다. 점도는 25℃에서 브룩필드 RV-DV III+ 프로그래밍가능한 유량계를 사용하여 측정되어 아래 표 2.1 및 2.2에 기록되었다.

Claims (28)
- 적어도 70 mg/ml 농도의 항체를 포함하는 액체 약제학적 제제의 점도를 감소시키는 방법이며,
상기 항체를, 점도-감소(viscosity-reducing) 농도의 n-아세틸 아르기닌, n-아세틸 라이신, n-아세틸 히스티딘, n-아세틸 프롤린 및 이들의 혼합물로 구성된 군으로부터 선택된 부형제와 조합하는 단계를 포함하고,

- 제21항 내지 제23항 중 어느 한 항에 있어서, 상기 항체가 인간, 인간화된 또는 키메라성 단클론성 항체인 동결건조된 분말.
- 멸균된 수성 희석제를 첨가하는 단계를 포함하는, 제21항 내지 제23항 중 어느 한 항의 동결건조된 분말을 재구성하는 방법.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 상기 항체가 인간, 인간화된 또는 키메라성 단클론성 항체인 방법.
