Es wird ein neues Verfahren zur Herstellung von 4,4-Diphenylcyclohexanol beschrieben.
Description
Verfahren zur Herstellung von 4,4-Diphenylcyclohexanol
Die Erfindung betrifft den in den Patentansprüchen gekennzeichneten Gegenstand, das heißt ein neues Verfahren zur Herstellung von 4,4- Diphenylcyclohexanol.
Die Verwendung von Kontrastmitteln zur Darstellung des Intravasalraumes (Blood pool imaging) ist einer der bedeutenden Anwendungen in der MRI- Angiographie. Hierbei hat sich besonders eine Verbindung bewährt, das so genannte MS-325 (US-6,676,929 und WO 96/23526) das von der EMEA unter dem Namen Vasovist® zugelassen wurde. Die Synthese von MS-325 ist in Synthetic Communications, 26(13), 2511-2522 (1996) und Synthetic Communications, 29(14), 2377-2391 (1999) beschrieben. Im Rahmen der Entwicklung dieser Verbindung entstand der Wunsch größere Substanzmengen zu liefern. Da die Substanz am Menschen appliziert wird, müssen strikte Anforderungen an die Reinheit des Endproduktes, sowie an die Zwischenprodukte gestellt werden. Aufgrund der zu erwartenden großen Anwendungspalette, sollte ein derartig hochwertiges Produkt auch in zu vertretendem preislichen und zeitlichen Aufwand herstellbar sein. Es besteht daher der Wunsch eine möglichst ökonomisch günstige Synthese in Händen zu haben.
Ein sehr wichtiges Zwischenprodukt der Synthese von MS-325 ist das 4,4- Diphenylcyclohexanol (I)
Die bisher im Stand der Technik offenbarten Verfahren zur Herstellung von 4,4- Diphenylcyclohexanol (I) aus 4,4-Diphenylcyclohex-2-enon (II)
waren stets zweistufig, wobei zunächst die Doppelbindung heterogen- katalytisch, vorzugsweise unter Verwendung eines Palladium-Katalysators hydriert wird (Amedio et. al., Synt. Comm. 28(20) 1998, 3895-3906, Freeman et. al., J. Org. Chem. 54(4) 1989, 782-789). Nach Isolierung des gebildeten 4,4Z Diphenylcyclohexanons erfolgt dann in einem zweiten Schritt die Reduktion der Ketofunktion mit komplexen Metallhydriden, beispielsweise Natriumborhydrid oder Lithiumaluminiumhydrid.
Im Hinblick auf Rohstoff- und Entsorgungskosten sowie Durchsatzzeit ist dagegen jedoch eine zweistufige Reaktionsführung nachteilig. Weiterhin werden nicht die kostengünstigsten Reduktionsmittel (möglichst Wasserstoff) eingesetzt was zu einer weiteren Erhöhung der Produktionskosten führt.
Eine direkte, einstufige Hydrierung wurde in der verfügbaren Literatur lediglich im Patent DE1793611, Beispiel Nr. 8 genannt. Dort diente die beschriebene Umsetzung unter Verwendung von Platinoxid zur Herstellung von 4,4- Diphenylcyclohexanol. Eine genauere Angabe der Reaktionsbedingungen fehlt. Das gebildete 4,4zDiphenylcyclohexanol wurde ohne Charakterisierung weiter umgesetzt, die Gesamtausbeute über 2 Stufen betrug nur 41% vom Einsatz 4,4- Diphenylcyclohex-2-enon.
Es besteht daher der Bedarf an einem neuen Verfahren zur Synthese von 4,4- Diphenylcyclohexanol aus 4,4-Diphenylcyclohex-2-enon, das sowohl kostengünstig ist als auch möglichst wenig Zeit benötigt.
Das erfindungsgemäße Verfahren erfüllt die gestellten Anforderungen in hohem Maße.
Die Erfindung betrifft ein Verfahren zur Herstellung der Verbindung 4,4- Diphenylcyclohexanol der Formel (I)
dadurch gekennzeichnet, dass die Verbindung 4,4-Diphenylcyclohex-2-enon der
Formel (II)
\n Gegenwart eines Nickelkatalysators mit Wasserstoff hydriert wird wobei die Reaktionszeit mindestens 1 min beträgt, und die Reaktionstemperatur 00C bis 100° beträgt und das erhaltene 4,4-Diphenylcyclohexanol gegebenenfalls aufgereinigt wird.
Das Lösungsmittel ist bevorzugt ausgewählt aus Isopropanol, Methanol, Ethanol, n-Propanol, n-Butanol, THF, Aceton, Ethylacetat, Dichlormethan, Toluol oder Mischungen davon.
In einer insbesondere bevorzugten Ausführungsform ist enthält das Lösungsmittel THF, insbesondere ist das Lösungsmittel eine Mischung von THF/Methanol im Verhältnis 1:10 bis 10:1.
Ganz besonders bevorzugt ist die Verwendung von reinem THF als Lösungsmittel (d.h. es sind keine weiteren Lösungsmittel im Reaktionsansatz vorhanden).
Unter reinem THF wird THF mit einer Reinheit von mindestens 99 Gew. % verstanden.
Unter einem Lösungsmittel wird eine Flüssigkeit verstanden in der 4,4- Diphenylcyclohexanol vollständig löslich ist bei mindestens einer Temperatur zwischen 00C und 1000C in einer Konzentration von mindestens 1 Gew %, vorzugsweise mindestens 5 Gew. %, insbesondere mindestens 10 Gew %.
In einer bevorzugten Ausführungsform wird 4,4-Diphenylcyclohex-2-enon mit dem organischen Lösungsmittel im Verhältnis 1 :100 bis 10:1 , insbesondere im Verhältnis 1:50 bis 1:1 , ganz besonders bevorzugt, im Verhältnis 1:20 bis 1:3 gemischt.
In einer ganz besonders bevorzugten Ausführungsform wird 4,4- Diphenylcyclohex-2-enon mit THF im Verhältnis 1 :3 bis 1 :20 gemischt. Die Hydrierung erfolgt in Gegenwart des Nickelkatalysators, bevorzugt Raney- Nickel.
Die Aufreinigung erfolgt nach den dem Fachmann bekannten Methoden, bevorzugt wird das Reaktionsgemisch filtriert, eingeengt und ein Lösungsmittel, bevorzugt Methanol im Verhältnis 1 :10 bis 10:1 zugegeben und das Lösungsmittelgemisch abdestilliert und das 4,4-Diphenylcyclohexanol durch Filtration isoliert.
Bevorzugt beträgt die Reaktionszeit mindestens 5 min, insbesondere bevorzugt mindestens 10 min, am meisten bevorzugt mindestens 20 min. Die Reaktion kann beliebig lange durchgeführt werden, aus ökonomischen Gründen wird jedoch die Reaktionszeit üblicherweise 48 h nicht überschreiten.
Bevorzugt beträgt die Reaktionstemperatur 100C bis 800C, insbesondere bevorzugt zwischen 15°C und 5O0C, ganz besonders bevorzugt zwischen 2O0C und 40°C.
Das so erhaltene 4,4-Diphenylcyclohexanol kann dann zur Synthese von Vasovist in seiner formulierten Form, zur Synthese von Gadofosveset oder eines seiner Salze, insbesondere des Natriumsalzes, oder zur Synthese von Fosveset eingesetzt werden, wie beispielsweise in US 6,676,929 oder US 5,919,967 beschrieben.
Der Ausgangsstoff 4,4-Diphenylcyclohex-2-enon ist dem Fachmann allgemein bekannt und kann z.B. wie in Amedio et. al., Synt. Comm. 28(20) 1998, 3895- 3906, Zimmermann et al. J. Am. Chem. Soc. 84, 1962, 4527 oder Bordwell et. al. J. Org. Chem. 28, 1963, 2544 beschrieben synthetisiert werden.
Die Erfindung betrifft weiterhin ein Verfahren zur Herstellung von MS-325, gekennzeichnet durch die Schritte a) Umsetzung der Verbindung 4,4-Diphenylcyclohex-2-enon der Formel (II)
in einem organischen Lösungsmittel gelöst wird und anschließend In Gegenwart eines Nickelkatalysators mit Wasserstoff hydriert wird wobei die Reaktionszeit mindestens 1 min beträgt, und die Reaktionstemperatur O0C bis 100° beträgt
zu 4,4-Diphenylcyclohexanol b) gegebenenfalls Aufreinigen von 4,4-Diphenylcyclohexanol
c) Umsetzung von 4,4-Diphenylcyclohexanol mit PCI3 und anschließend Imidazol in THF zum bis(amino)phosphino Reaktionsprodukt, und d) Umsetzung des in c) erhaltenen Reaktionsproduktes zu Fosveset e) Umsetzung von Fosveset und Gd2O3 zu Gadofosveset (MS-325)
Die Schritte b) bis e) sind in US 5,919967 beschrieben, und auf die Offenbarung der Patentschrift wird hiermit ausdrücklich Bezug genommen und der Inhalt der Patentschrift wird in dem Umfang in dem sie sich auf die Herstellung von MS- 325 ausgehend von 4,4-Diphenylcyclohexanol bezieht in diese Anmeldung aufgenommen.
In einer weiteren Ausführungsform betrifft die vorliegende Erfindung ein Verfahren zur Herstellung eines MS-325 enthaltenden Präparats geeignet für die Diagnose mittels MRI, dadurch gekennzeichnet, dass zunächst MS-325 nach dem oben aufgeführten Verfahren hergestellt wird und anschließend mit in der Galenik üblichen Hilfs- und Zusatzstoffen in eine für die diagnostische Anwendung am Menschen annehmbare Form gebracht wird. In einer besonders bevorzugten Ausführungsform ist das Präparat für die i.v. Anwendung geeignet und ist insbesondere ein Präparat das unter dem Markennamen Vasovist® erhältlich ist.
Die Vorteile des neuen Verfahrens sind :
1. Deutlich höhere Gesamtausbeute gegenüber dem Stand der Technik (siehe unten)
2. Einfache Verfahrensweise (einstufige Reduktion) 3. Preiswerte Hydrierungsmittel (Wasserstoff)
4. Hohe Reinheit des erhaltenen 4,4-Diphenylcyclohexanol
Beispiele
Beispiel 1
100 g 4,4-Diphenylcyclohex-2-enon werden in 750 ml Tetrahydrofuran gelöst. Es werden 10 ml einer zuvor salzfrei gewaschenen, wässrigen Suspension von Raney-Nickel zugegeben. Die Suspension wird unter Rühren bei einem
Wasserstoffdruck von 5 bar und einer Temperatur von 25 bis 3O0C 60 min hydriert. Es wird vom Nickelkatalysator abfiltriert und das Filtrat unter Normaldruck bis zu einer Temperatur von 80 bis 85°C eingeengt. Anschließend werden 500 ml Methanol zugegeben, auf ein Restvolumen von 200 ml eingeengt und auf 0°C abgekühlt. Das Kristallisat wird isoliert, mit Methanol gewaschen und getrocknet.
Ausbeute: 95g (entspricht 93% d. Th.) Gehalt: 99,7 Flächen-% (HPLC).
Beispiel 2
80,0 g 4,4-Diphenylcyclohex-2-enon werden in 750 ml Tetrahydrofuran / Methanol-Gemisch 7:3 (v/v) gelöst. Es werden 10 ml einer zuvor salzfrei gewaschenen, wässrigen Suspension von Raney-Nickel zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 5 bar und einer Temperatur von 25 bis 300C 60 min hydriert. Es wird vom Nickelkatalysator abfiltriert und das Filtrat unter Vakuum bis zu einer Temperatur von 40 bis 45°C eingeengt. Anschließend werden 400 ml Methanol zugegeben und unter Vakuum 250 ml Lösungsmittel abdestilliert. Es wird auf 00C abgekühlt, das Kristallisat isoliert, mit Methanol gewaschen und getrocknet.
Ausbeute: 68,5g (entspricht 84% d. Th.). Gehalt: 99,7 Flächen-% (HPLC).
Beispiel 3
80,0 g 4,4-Diphenylcyclohex-2-enon werden in 750 ml Isopropanol suspendiert. Es werden 10 ml einer zuvor salzfrei gewaschenen, wässrigen Suspension von Raney-Nickel zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 5 bar bis auf eine Temperatur von 700C erwärmt und 120 min hydriert. Es wird heiß vom Nickelkatalysator abfiltriert, das Filtrat unter Vakuum bis auf ein Volumen von 300 ml eingeengt und auf O0C abgekühlt. Das Kristallisat wird isoliert, mit Isopropanol gewaschen und getrocknet.
Ausbeute: 66,0 g (entspricht 81% d. Th.). Gehalt: 99,1 Flächen-% (HPLC).
Beispiel 4
80,0 g 4,4-Diphenylcyclohex-2-enon werden in 750 ml Methanol suspendiert. Es werden 10 ml einer zuvor salzfrei gewaschenen, wässrigen Suspension von Raney-Nickel zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 5 bar bis auf eine Temperatur von 7O0C erwärmt und 90 min hydriert. Es wird heiß vom Nickelkatalysator abfiltriert und das Filtrat unter Vakuum bis auf ein Volumen von ca. 150 ml eingeengt. Anschließend wird auf O0C abgekühlt, das Kristallisat isoliert, mit Methanol gewaschen und getrocknet.
Ausbeute: 72g (entspricht 89% d. Th.). Gehalt: 73,8 Flächen-% 4,4-Diphenylcyclohexanol (HPLC) 25,4 Flächen-% 4,4-Diphenylcyclohex-2-enon (HPLC).
Beispiel 5 (Vergleichsversuch)
57,0 g 4,4-Diphenylcyclohex-2-enon werden in 1000 ml Ethanol gelöst.
Es wird 1 ,0g Palladium auf Aktivkohle-Träger (Palladiumgehalt 5 bis 10 Gew.-
%) zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 5 bar bis auf eine Temperatur von 800C erwärmt und 75 min hydriert.
Mittels DC-Kontrolle der Hydriersuspension kann keine Bildung von 4,4-
Diphenylcyclohexanol festgestellt werden.
Es wird erneut 1 ,0g Palladium auf Aktivkohle-Träger (Palladiumgehalt 5 bis 10
Gew.-%) zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 6,8 bar bis auf eine Temperatur von 1000C erwärmt und
60 min hydriert. Mittels DC-Kontrolle der Hydriersuspension kann die Bildung von ca. 10 bis 20% 4,4-Diphenylcyclohexanol festgestellt werden. Es wird 1 ,0g
Platin auf Aktivkohle-Träger (Platingehalt ca. 10 Gew.-%) zugegeben. Die
Suspension wird unter Rühren bei einem Wasserstoffdruck von 7,0 bar bis auf eine Temperatur von 100°C erwärmt und 60 min hydriert. Mittels DC-Kontrolle der Hydriersuspension kann die Bildung von 50% 4,4-Diphenylcyclohexanol festgestellt werden.
Es wird vom Katalysator abfiltriert und das Filtrat bis auf ein Volumen von ca.
100 ml eingeengt. Anschließend werden bei einer Temperatur von 55 bis 6O0C 250 ml Wasser zugegeben und auf 100C abgekühlt. Das Kristallisat wird isoliert, mit Wasser gewaschen und getrocknet.
Ausbeute: 56,0g (entspricht 96 % d. Th.). Gehalt: 50% 4,4-Diphenylcyclohexanol sowie 50% Summe aus 4,4-Diphenylcyclohex-2-enon und
4,4-Diphenylcyclohexanon (bestimmt durch DC).
Beispiel 6 (Vergleichsversuch)
55,0 g Reaktionsprodukt aus Beispiel 5 werden in 1000 ml Tetrahydrofuran gelöst.
Es werden 3,0g Platin auf Aktivkohle-Träger (Platingehalt ca. 10 Gew.-%) zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von
5 bar bei einer Temperatur von 20 bis 25 0C 60 min hydriert. Mittels DC-
Kontrolle der Hydriersuspension kann keine Veränderung gegenüber dem
Einsatzmaterial festgestellt werden.
Es werden erneut 2,0g Platin auf Aktivkohle-Träger (Platingehalt ca. 10 Gew.- %) zugegeben. Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 5 bar bei einer Temperatur von 25 bis 30 0C 60 min hydriert. Mittels DC-
Kontrolle der Hydriersuspension kann keine Veränderung gegenüber dem
Einsatzmaterial festgestellt werden.
Die Suspension wird unter Rühren bei einem Wasserstoffdruck von 6,6 bar bei einer Temperatur von 80 0C 30 min hydriert. Mittels DC-Kontrolle der
Hydriersuspension kann die Bildung von unpolaren Nebenverbindungen in der
Größenordnung von 30 bis 50% festgestellt werden.
Claims
1. Verfahren zur Herstellung von 4,4-Diphenylcyclohexanol (I),
dadurch gekennzeichnet, dass die Verbindung der Formel (II)
in einem organischen Lösungsmittel gelöst wird und anschließend In Gegenwart eines Nickelkatalysators mit Wasserstoff hydriert wird wobei die Reaktionszeit mindestens 1 min beträgt, und die Reaktionstemperatur 00C bis 100° beträgt und das erhaltene 4,4-Diphenylcyclohexanol gegebenenfalls aufgereinigt wird.
2. Verfahren zur Herstellung von MS-325, gekennzeichnet durch folgende Schritte: a) Durchführung des Verfahrens nach Anspruch 1 , b) gegebenenfalls Aufreinigen von 4,4-Diphenylcyclohexanol c) Umsetzung von 4,4-Diphenylcyclohexanol mit PCb und anschließend Imidazol in THF zum bis(amino)phosphino Reaktionsprodukt, und d) Umsetzung des in c) erhaltenen Reaktionsproduktes zu Fosveset e) Umsetzung von Fosveset und Gd2θ3 zu Gadofosveset (MS-325)
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass das organische Lösungsmittel Isopropanol, Methanol, Ethanol, n-Propanol, n- Butanol, THF, Aceton, Ethylacetat, Dichlormethan, Toluol oder Mischungen davon ist.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass das organische Lösungsmittel THF enthält.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass das organische Lösungsmittel THF ist und keine weiteren Lösungsmittel im Reaktionsansatz vorhanden sind.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet dass der Katalysator Raney-Nickel ist.
7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet dass die Reaktionstemperatur zwischen 100C und 800C beträgt.
8. Verfahren nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet dass die Reaktionszeit mindestens 10 min beträgt.
