Abstract
高コレステロール症、高トリグリセリド症、耐糖能異常、高血圧、肥満はそれぞれの症状が軽度であっても、これらが複合的に発生するメタボリックシンドロームが、大きな死亡要因の1つである循環器疾患の原因となっている。アスタキサンチン含有抽出を有効成分として含有するメタボリックシンドロームの改善・予防効果を有する組成物ならびにアスタキサンチン含有抽出を有効成分として含有するメタボリックシンドロームの飲食物や医薬品を提供する。また、アスタキサンチン含有抽出はメタボリックシンドロームの症状である高血圧、高脂血、糖尿病、肥満の改善・予防にも効果がある。

[0001] 本発明は、ァスタキサンチンを有効成分として含有するメタボリックシンドロームの改 善及び Z又は予防効果を有する組成物に関する。さらに詳しくは、密閉型培養装置 で培養されるへマトコッカス藻を乾燥粉砕し、溶媒で抽出したのち、溶媒を除去して 得られるァスタキサンチンを有効成分として含有するメタボリックシンドロームの改善 及び Z又は予防効果を有する組成物に関する。また、ァスタキサンチン及び 1種又 は 2種以上の活性剤を有効成分として含有するメタボリックシンドロームの予防'改善 効果を有する組成物に関する。
背景技術
[0002] 日本国において脳血管障害や心血管障害などの循環器疾患が全死亡率の約 30 %を占め、癌とともに最も大きな死亡原因となっている。また死亡とならないまでも、発 病によって大きな後遺症を残すことも大きな問題となっている。循環器疾患の原因と して、これまで高脂血症、高血圧、高血糖などの症状がそれぞれ進行したことが主な 原因と考えられてきた。しかし、近年、これらも単独で原因とはなっているが、それより も高コレステロール症、高トリグリセリド症、耐糖能異常、高血圧、肥満のそれぞれが 特に症状が進行していなくても 3つ以上が合併した場合の発病率が数十倍も高いこ とが分力つてきた。 1個人にこれらの症状で軽度なものが集合しているマルチプルリス クファクター症候群をメタボリックシンドロームとよばれて!/、る。
[0003] メタボリックシンドロームは、内臓脂肪の蓄積による肥満細胞の増大が最大の原因と して知られている。肥満細胞の増大は、肥満細胞が分泌している種々の生理活性物 質の異常や血中への遊離脂肪酸の放出増加をもたらす。肥満細胞が分泌している 生理活性物質としては、インスリン抵抗性に関する TNFひやレジスチン、血栓形成に 関連する PAI— 1、高血圧に関連するアンジォテンシンシノーゲンなどがある。
[0004] メタボリックシンドロームを形成する個々の症状である高脂血症、高血圧、高血糖な どについては、それぞれの症状の治療薬は数多く開発されている。しかし、医薬品の
場合、薬の副作用や併用 (飲み合わせ)による弊害があり、長期間摂取したり、また、 予防のために摂取したりするためには好ましくないものが多ぐ副作用や毒性がなく 長期間に渡って摂取できる飲食物の形態で摂取できるメタボリックシンドロームの改 善'予防用の飲食物や薬剤が望まれている。
[0005] ァスタキサンチンは、 j8—力口テンと同じカロテノイドの一種で、ェビ、力-等の甲殻 類、サケ、タイ等の魚類、緑藻へマトコッカス等の藻類、赤色酵母ファフィァ等の酵母 類等、天然、特に海洋に広く分布しており、赤色色素として広く用いられている。ァス タキサンチンはビタミン Ε ( α—トコフエロール)の約 1000倍、 13—カロテンの約 40倍 もの強力な抗酸ィ匕作用を有することが見いだされ、現在は最も有効性のある健康食 品として用いられている。
[0006] ァスタキサンチンの有する主な機能特性として、動脈硬化の防止効果を有すること ( 特許文献 1)、高血圧の防止効果を有すること (特許文献 2)、インスリン依存真性糖 尿病の治療剤 (特許文献 3)が報告されている。また、ァスタキサンチンの有するその 他の機能特性として、抗炎症作用、光傷害力もの網膜保護作用、 日周リズム調節作 用、免疫賦活作用、抗ストレス作用、精子の質向上作用や膀胱がん誘発抑制、筋肉 持続力向上作用等数多くの報告がなされて!/ヽる。

[0008] 本発明者らは、上記課題を解決するべぐメタボリックシンドロームの改善及び Ζ又 は予防効果を有する物質を探索した結果、ァスタキサンチンカ^タボリックシンドロー ムを改善及び Ζ又は予防する効果があることを見出した。本発明は、力かる知見に 基づき完成されたものであり、ァスタキサンチンを有効成分とするメタボリックシンドロ ームの改善及び Ζ又は予防効果を有する組成物、並びにァスタキサンチンを有効成
分とするメタボリックシンドロームの改善及び z又は予防効果を有する医薬品、飲食 物を提供するものである。
[0009] 本発明は、ァスタキサンチンを有効成分とするメタボリックシンドロームの改善及び
Z又は予防効果を有する組成物、並びにァスタキサンチンを有効成分とするメタボリ ックシンドロームの改善及び Z又は予防効果を有する医薬品及び飲食物を提供する ことを目的とする。本発明の組成物、医薬品および飲食物は、メタボリックシンドロー ムを改善及び Z又は予防することにより、メタボリックシンドロームによって生じる疾病 の改善及び Z又は予防にも有用である。
課題を解決するための手段
[0010] 本発明者らは上記課題を解決するために鋭意研究した結果、ァスタキサンチンが 特に優れたメタボリックシンドロームの改善及び Z又は予防効果を有することを見出 した。本発明は係る知見に基づくものである。
発明の効果
[0011] 本発明のァスタキサンチンを有効成分として含有するメタボリックシンドロームの改 善及び Z又は予防効果を有する組成物を、飲食物または医薬品の形態で投与する ことによって、メタボリックシンドロームの改善及び Z又は予防することができる。 図面の簡単な説明

発明を実施するための最良の形態
[0013] 本発明においてァスタキサンチンとは、天然物由来のものまたは合成により得られ るものを意味する。天然物由来のものとしては、例えば、ェビ、ォキアミ、力-などの 甲殻類の甲殻、卵および臓器、種々の魚介類の皮および卵、緑藻へマトコッカスなど の藻類、赤色酵母ファフィァなどの酵母類、海洋性細菌、福寿草および金鳳花など の種子植物力 得られるものをあげることができる。天然からの抽出物および化学合 成品は市販されており、入手は容易である。
[0014] ァスタキサンチンは、例えば、赤色酵母ファフィァ、緑藻へマトコッカス、海洋性細菌
などを、公知の方法に準拠して、適宜な培地で培養することにより得られる。培養や 抽出のしゃすさ、ァスタキサンチンを最も高濃度で含有することや生産性の高さから 緑藻へマトコッカスが最も好適である。
[0015] ァスタキサンチンの使用形態としては、前記方法で得たァスタキサンチンの抽出物 およびそれらを含有した粉末や水溶液、または赤色酵母ファフィァ、緑藻へマトコッカ ス、海洋性細菌などの乾燥品およびそれらの破砕品を用いることができる。
[0016] ァスタキサンチンを含むへマトコッカス藻の培養とへマトコッカス藻からァスタキサン チンの抽出方法について以下に具体的に説明する。
へマトコッカス緑藻類の培養方法としては、異種微生物の混入'繁殖がなぐその他 の夾雑物の混入が少ない密閉型の培養方法が好ましぐ例えば、一部解放型のドー ム形状、円錐形状又は円筒形状の培養装置と装置内で移動自在のガス吐出装置を 有する培養基を用いて培養する方法 (国際公開第 99Z50384号公報)や、密閉型 の培養装置に光源を入れ内部から光を照射して培養する方法、平板状の培養槽ゃ チューブ型の培養層を用いる方法、ならびにピーク波長が約 540nm以下の波長の 光を照射することによって培養する方法 (特開 2004— 147641号公報)が適してい る。
[0017] 本発明のへマトコッカス藻力 抽出物を得る方法としては、常法に従って、へマトコ ッカス藻を破砕し溶媒で抽出することができ、例えば、(1)へマトコッカスを破砕 ·乾燥 したあと、溶媒で抽出を行い、溶媒を除去して抽出物を得る方法、(2)へマトコッカス (湿末)を有機溶媒に懸濁した後、粉砕機に通して細胞を粉砕して抽出し、有機溶媒 を除去して抽出物を得る方法があげられる。夾雑物が最も少ないこと工程が少なく余 分な熱を加えな 、ことから、好ま 、のは(2)の粉砕抽出法である。
[0018] 上記(1)の方法は、へマトコッカスを破砕 ·乾燥したあと、溶媒で抽出を行い、溶媒 を除去して抽出物を得る方法である。粉砕方法は、湿式、乾式のどちらでもよぐビー ズミル、ロールミル、ハンマーミル、ジェットミル、ピンミルなど常法に従って行うことが できる。このとき、ァスタキサンチンの酸化を抑制'防止するため、トコフエロールやトコ トリェノールなど既存の酸ィ匕防止剤を添加することができる。乾燥方法は、棚式乾燥
、流動層乾燥、フラッシュ乾燥、噴霧乾燥など常法にしたがって行うことができる。粉
砕-乾燥の具体的な方法としては、本出願人が先に出願した WO2002Z077105の 方法によって行うことができる。続いて、この乾燥破砕藻を溶媒で抽出する。抽出〖こ 用いる溶媒は、例えば、アセトン、アルコール、酢酸ェチル、ベンゼン、クロ口ホルム、 二酸化炭素、アンモニアなどが適している。抽出時の温度は溶媒が、沸騰しない温 度でよぐ 0〜60°Cが適している。続いてこの抽出液を減圧蒸留などを行い、溶媒を 除去して抽出物を得る。
[0019] 抽出溶媒として、二酸ィ匕炭素やアンモニアを用いる場合は、超臨界抽出を行う。超 臨界抽出は、常法に従って、例えば、特開 2004— 41147号公報に記載の方法で 行うことができる。粉砕処理したへマトコッカス藻をペレット状に成形し、超臨界状態 の二酸ィ匕炭素をまたは超臨界点近傍の状態の二酸ィ匕炭素をそのペレットを充填した 層を通過させて抽出する。減圧を行って二酸ィ匕炭素を除去して抽出物を得る。ペレ ットからの抽出性を高めるために高級不飽和脂肪酸や、グリセリン、アルコール、水な どを加えて良い。
[0020] 上記(2)の方法は、本出願人が先に出願した特願 2004— 253525の方法により 行うことができる。へマトコッカス藻を湿末のまま、上述の有機溶媒に懸濁させて、粉 砕器で粉砕しながら同時に抽出を行う。有機溶媒の除去の方法は上述同様に常法 に従って行うことができる。
[0021] 上述の 2種の製法で得られた抽出物は、所望により分離カラムやリパーゼ分解によ りさらに精製することができる。さらに溶媒を除去したい場合は、分子蒸留装置などを 用!/、ることができる。
[0022] 密閉型の培養装置を用 、て 、るため異種微生物由来の供雑物がなく、また穏和な 条件でァスタキサンチン含有抽出物を得ていることから抽出物の酸ィ匕による破壊が 少なぐァスタキサンチンの効果をより効果的に発揮することができる。
[0023] ァスタキサンチンは、 3, 3' ジヒドロキシー 13 , β一力口テン 4, 4'ージオンであり 、立体異性体を有する。具体的には、(3R, 3'R)—ァスタキサンチン、 (3R, 3'S) - ァスタキサンチンおよび(3S, 3'S)ーァスタキサンチンの 3種の立体異性体が知られ ている。
[0024] 本発明の記載で、特に記載がない限り、ァスタキサンチンはァスタキサンチン及び
z又はそのエステルを含む。さらに、ァスタキサンチンのエステルにはモノエステル体 及び Z又はジエステル体を含む。
[0025] ァスタキサンチンは突然変異原性が観察されず、安全性が高い化合物であることが 知られており、食品添加物として広く用いられて 、る(高橋ニ郎ほか:へマトコッカス 藻ァスタキサンチンの毒性試験 Ames試験、ラット単回投与毒性試験、ラット 90日 反復経口投与性毒性試験 臨床医薬、 20 : 867— 881 2004)。
[0026] 本発明のァスタキサンチン含有抽出物には、ァスタキサンチンとして、ァスタキサン チンの遊離体、モノエステル体、ジエステル体の少なくとも一種以上が含まれている。 ジエステル体は 2つの水酸基がエステル結合により保護されているため物理的に遊 離体やモノエステル体よりも安定性が高く飲食物中や医薬品中で酸化分解されにく い。しかし、生体中に取り込まれると生体内酵素により速やかに遊離体のァスタキサ ンチンに加水分解され、効果を示すものと考えられている。
[0027] ァスタキサンチンのモノエステルとしては、低級または高級飽和脂肪酸、あるいは低 級または高級不飽和脂肪酸によりエステルイ匕されたエステル類をあげることができる 。前記低級または高級飽和脂肪酸、あるいは低級または高級不飽和脂肪酸の具体 例としては、酢酸、ラウリン酸、ミリスチン酸、ペンタデカン酸、パルミチン酸、ノ レミト ォレイン酸、へブタデカン酸、エライジン酸、リシノール酸、ベトロセリン酸、バタセン酸 、エレォステアリン酸、プ-シン酸、リカン酸、パリナリン酸、ガドール酸、 5—エイコセ ン酸、 5 ドコセン酸、セトール酸、エルシン酸、 5, 13 ドコサジェン酸、セラコール 酸、デセン酸、ステリング酸、ドデセン酸、ォレイン酸、ステアリン酸、エイコサオペンタ ェン酸、ドコサへキサェン酸、リノール酸、リノレン酸、ァラキドン酸などをあげることが できる。また、ァスタキサンチンのジエステルとしては前記脂肪酸力もなる群力も選択 される同一または異種の脂肪酸によりエステルイ匕されたジエステル類である。
[0028] さらに、ァスタキサンチンのモノエステルとしては、グリシン、ァラニンなどのアミノ酸; 酢酸、クェン酸などの一価または多価カルボン酸;リン酸、硫酸などの無機酸;ダルコ シドなどの糖;グリセ口糖脂肪酸、スフインゴ糖脂肪酸などの糖脂肪酸;グリセ口脂肪 酸などの脂肪酸;グリセ口リン酸などによりエステルイ匕されたモノエステル類である。前 記モノエステル類の塩も含まれて 、る。

Claims
[1] ァスタキサンチンを有効成分として含有するメタボリックシンドロームの改善'予防効 果を有する組成物。
[2] ァスタキサンチン及び 1種又は 2種以上の活性剤を有効成分として含有するメタボリッ クシンドロームの予防 ·改善効果を有する組成物。
[3] ァスタキサンチンが、密閉型培養装置で培養されるへマトコッカス藻を粉砕し、溶媒 で抽出したのち、溶媒を除去して得られるァスタキサンチンである請求項 1〜2の組 成物。
[4] 活性剤が、以下の群のいずれかの 1種または 2種以上である請求項 2〜3の組成物: ビタミン Α類、カロテノイド類、ビタミン B類、ビタミン C類、ビタミン D類、ビタミン E類、ト コトリエノール、ダルタチオン及びこれらの誘導体並びにこれらの塩; aーリポ酸、デ ォキシリボ核酸、リボ核酸、アデノシン三リン酸、アデノシン一リン酸、グリチルリチン 酸、グリチルレチン酸、グァニン、キサンチン、 ex または γ リノレイン酸、エイコサ ペンタエン酸、ドコサへキサェン酸、コハク酸、エストラジオール及びその誘導体並び にそれらの塩;ァスパラダン酸、グリコール酸、乳酸、リンゴ酸、クェン酸、サリチル酸 などの OCーヒドロキシ酸及びそれらの誘導体並びにそれらの塩;血清除蛋白抽出物 、脾臓抽出物、胎盤抽出物、鶏冠抽出物、ローヤルゼリー;酵母抽出物、乳酸菌抽 出物、ビフィズス菌抽出物、霊芝抽出物;ニンジン抽出物、センプリ抽出物、ローズマ リー抽出物、ォゥバタ抽出物、ニン-ク抽出物、ヒノキチオール、セファランチン、ァロ ェ抽出物、サルビア抽出物、アル二力抽出物、力ミツレ抽出物、シラカバ抽出物、オト ギリソゥ抽出物、ユーカリ抽出物、ムクロジ抽出物、センプクカ抽出物、ケィケットウ抽 出物、サンペンズ抽出物、ソゥハクヒ抽出物、トウキ抽出物、イブキトラノォ抽出物、ク ララ抽出物、サンザシ抽出物、シラユリ抽出物、ホップ抽出物、ノイバラ抽出物、ョクイ ニン抽出物; D フラクション、グリコーゲン、ォクタコサノール、ァリシン、コェンザィム Q10、カテキン、ポリフエノール、フラボノイド、カルノシン、カルニチン;システィン及 びその誘導体並びにその塩、ペプチド;リジン、硫化ァリル、ピオチン、パントテン酸、 コラーゲン、エラスチン、ケラチン及びこれらの誘導体並びにその塩類;ヒアルロン酸 、コンドロイチン硫酸、デルマタン硫酸、へパラン硫酸、へパリン、ケラタン硫酸;乳酸
菌;鉄、モリブデン、カルシウム、亜鉛、セレン、マンガン、銅、ヨウ素などのミネラル類
[5] 請求項 1〜4の組成物が医薬品である請求項 1〜4の組成物。
[6] 請求項 1〜4の組成物が飲食物である請求項 1〜4の組成物。
[7] 請求項 1〜4の糸且成物を投与することによって、メタボリックシンドロームを治療'予防 する方法。
