- the CD34 positive rate was defined as the purity of the CD34 cells, and the number of cells obtained by multiplying the CD34 positive rate and the number of recovered cells was defined as the number of recovered CD34 positive cells. As a result, a purity of 45.2% and a recovery rate of 62.6% were obtained. From this, it was confirmed that a device for separating and separating hematopoietic undifferentiated cells consisting of a column device packed with gel was effective.
- the remaining cells were reacted with anti-HPCA-2 antibody (Becton) on ice for 30 minutes, washed with cold saline containing 2% fetal calf serum, and collected by centrifugation. The cells were measured for CD34 positivity using a Coulter flow cytometer.
- the CD34 positive rate is defined as the purity of the CD34 cells.
- the number of multiplied cells was defined as the number of collected CD34-positive cells. As a result, the purity was 36.1% and the recovery rate was 35.3%. This indicated that the single-chain antibody produced in the production of Example 21 separated CD34-positive cells.
- An antibody column to which the single-chain antibody produced in Example 21 was bound was prepared using CNBr-activated Sepha rose4B (Pharmacia). The method of binding the antibody to Sepharose4B beads was in accordance with the attached instruction manual. The single-chain antibody solution was reacted with 4.0 ml of the gel to prepare an affinity gel with a coupling efficiency of 99.0%.
- the CD34 positive rate was defined as the purity of CD34 cells, and the number of cells obtained by multiplying the CD34 positive rate by the number of recovered cells was defined as the number of recovered CD34 cells. As a result, a result with a purity of 52.8% and a recovery rate of 41.0% was obtained. From this, it was confirmed that a device for separating and separating hematopoietic undifferentiated cells consisting of a column device packed with gel was effective.
- the pCANMYlO vector prepared in Example 21 was modified to prepare an antibody capable of improving the binding to a water-insoluble carrier. Following the E-tag sequence at the carboxyl terminus of the single-chain antibody, three amino acid lysine residues were introduced. Using a restriction enzyme site of the pCANMY10 vector, a sequence containing a lysine residue was introduced by the PCR method. The sequence is shown in Sequence Listing 4.
- This vector was introduced into E. coli HB2151 cells, and a single-chain antibody was prepared in the same manner as in Example 21.
- the obtained antibody was prepared using a CNB r-activated Separose 4B (Pharmacia) with an antibody column to which a single-chain antibody was bound.
