明 細 書
安定なイ ンタ一フ エ ロ ン 3組成物
技 術 分 野
本発明は、 イ ンターフ ヱロン (以下、 I F N— と 略す) の局所投与、 あるいは全身投与を目的と した安定 な I F N— 3組成物に関し、 特に軟膏剤に適した安定化 組成物に関する。
背 景 技 術
ゥィ ルス増殖抑制作用をもつ I F N— 3は、 細胞培養 法や遺伝子組換え法によつて大量に製造する方法が開発 され、 すでに医薬品と して市販されている。 医薬品と し ての要請から I F N— /3は高度に精製されるが、 精製が 進むに従って安定性が低下する問題が存在した。 I F N — /3の安定性の向上に関しては、 ポ リオールの使用 (特 開昭 58— 9 2 6 1 9号公報など) 、 ヒ ト血清アルブミ ンの使用 (国際公開 WO 83 Z 0 1 1 98) 、 有機酸緩 衝剤の使用 (特開昭 58 - 9 26 2 1号公報) 、 力ルポ キシメ チルセルロースの使用 (特開昭 62 - 1 5 3 22 6号公報) などの方法がすでに提案され、 一定の安定化 効果が達成されている。
また、 I F N— 3の製剤と しては、 ポ リエチレングリ コール、 プロ ピレ ングリ コール、 ヒ ト血清アルブ ミ ン、 ポリ ビニルピロ リ ドン、 メチルまたはプロ ピル、 p— ヒ ドロキンベンゾェ一 トを基剤とする軟膏が Serono社 (ィ タ リア) および Inter-Yeda社 (イ スラエル) で市販され
ており、 4〜 8 Cの低温保存で一年間安定であることが 能書に記載されている。
上記従来技術により I F N— /3の安定化を図ることは できるが、 I F N— ^を特に軟膏剤とするためには、 増 粘剤および保存剤など種々の添加物を加える必要があり、 増粘剤としてカルボキンメチルセルロース ( "ダイセル 220 0 " ) を加えた場合、 たとえ特開昭 58— 926 1 9号公報に示された 3 0 %以上 (実験では 50 %) の グリセリ ン、 ヒ ト血清アルブミ ン (国際公開 W 083 Z 0 1 1 98) 、 有機酸緩衝剤 (特開昭 58— 926 2 1 号公報) 、 および 2 %カルボキシメチルセルロース (
"ダイセル 1 240 " ) (特開昭 52— 1 53226号 公報) の 4種類の I F N— 安定化剤が添加されても、 2 ( w Z w ) %カルボキシメチルセルロース ( "ダイセ ル 1 240 " ) - 50 ( /w) %グリセリ ン一 48
(w/w) % 0. 1 Mクェン酸緩衝液 (p H 5) 中の I F N— ^に比べ安定性が著しく劣る。 また、 保存剤とし て知られている P—ヒ ドロキシベンゾエー ト類を添加し た場合も、 特開昭 59 - 1 762 1 6号公報にも記載さ れているように、 I F N - /3は失活する。
また、 前記のイタリアおよびイスラエルで市販されて いる I F N— ^の保存期間は、 4〜 8。(:保存で一年間で ある。 しかし、 市中の薬局において I F N— の組成物 を市販する場合および輸送における簡便性を考えるなら ば、 室温保存が可能な I F N— S組成物を開発する必要
がある。 したがって本発明の目的は、 室温でも安定な I F N— ^組成物の処方を見い出すことにある。
発 明 の 開 示
本発明は、 ポ リオールを 6 5〜 9 0重量%と、 p — ヒ ドロキシベンゾェ一 ト類とを含有してなる安定なィ ンタ 一フ ロ ン ^組成物である。
本発明により、 室温で長期間保存可能で、 さ らに種々 の添加物を含んでいても I F N - Sが失活せず、 I F N
- の安定化効果が長期間持続する組成物となる。
図面の簡単な説明
第 1図は、 実施例 1 〜 4および比較例 1 〜 4で測定し た I F N— 3の力価残存率を示すものである。
発明を実施するための最良の形態 本発明のイ ンターフエ口 ン ; 3組成物は、 保存剤、 増粘 剤などが添加された場合に安定性の高いものを得るため に、 6 5〜 9 0重量%のポ リオールを含有する必要があ り、 7 0〜 8 5重量%含有することがより好ま しい。 本発明のポ リオ一ルとは 2価以上のァルコールであり 具体的にはプロ ピレングリ コール、 エチレングリ コール グリセ リ ンまたはポ リ グリセリ ンなどが挙げられ、 これ らのポ リオールは 1種でも 2種以上を併用してもよい。
さ らに、 本発明のポリオールは 3価のアルコールが好 ま しく 、 特にグリセリ ンが好ま しい。
本発明で用いる P—ヒ ドロキンベンゾェ一 ト類と して は、 例えばメ チル、 ェチル、 プロ ピル、 ブチルエステル
等力 挙げられる。 これらの 一 ヒ ドロキシベンゾエー ト 類は 1種でも 2種以上併用して使用してもよい。 上記 p ーヒ ドロキシベンゾェ一 ト類の中では、 ェチル p —ヒ ド ロキシベンゾ、ェ一 ト、 プロピノレ p —ヒ ドロキシベンゾ、ェ — トが特に好ま しい。 p —ヒ ドロキシベンゾエー ト類の 含有量は、 通常 0 . 0 1 〜 0 . 2重量%の範囲で用いら れる。
また、 特に軟膏を調整する際には、 ポリオ一ル、 p— ヒ ドロキシべンゾエー ト類以外に力ルボキシメチルセル ロースおよびその塩、 メチルセルロース、 ヒ ドロキシェ チルセルロース、 デンプン、 微小織維状セルロース等の 增粘剤、 ヒ ト血清アルブミ ン、 ヒ ト血清グロブリ ン等の 安定化剤を添加することが好ま しい。 増粘剤と しては、 カルボキシメチルセルロースあるいはその塩が好ま しく . 特に 1 %水溶液と して 2 5 。C、 6 0 Γ ριπ の条件下に B型 粘度計により測定した時の粘度が 3 0 〜 2 0 0 eps であ るものが好ま しい。 具体的には、 ダイセル社製の "ダイ セル 1 2 4 0 " 、 "ダイセル 1 2 6 0 " 、 "ダイセル 1 3 4 0 " 、 "ダイセル 2 2 0 0 " 等のカルボキシセル口 —スのナ ト リ ウム塩が挙げられる。 増粘剤の含有量は、 通常 0 . 1 〜 2 . 5重量%の範囲で用いられる。 安定化 剤と しては、 例えばヒ ト血清アルブミ ン、 ヒ ト血清グロ ブリ ン等が挙げられるが、 ヒ ト血清アルブミ ンが好ま し く用いられる。 安定化剤の含有量は、 通常 0 . 1 〜 1重 量%の範囲で用いられる。
さ らに、 クェン酸緩衝剤等の配合剤を適宜配合しても よい。
本発明の組成物に配合する I F N— /3は、 ヒ ト由来の ものであれば細胞培養法で製造したものでも、 遺伝子組 換え法によって製造されたものでもよい。 I F N— /3の 配合量は特に限定されないが、 1 X 1 0 4 国際単位 ( I U) 以上の力価を有する I F N— Sを配合すること が好ま しい。
次に実施例により本発明をさ らに説明する。 なお、 実 施例中のイ ンターフヱロン力価の測定はシン ドビスウイ ルスあるいは V S Vウィ ルスおよびヒ ト羊膜由来の株化 細胞 (F L細胞) を用い、 細胞変性効果 ( C P E ) 法に より行い国際単位 ( I U) に換算した。
実 施 例 1
細胞増療法によつて製造されたヒ ト 2倍体線維芽細胞 由来の I F N— /3 〔 "フュロ ン" (東レ株式会社製) 〕 を使用し、 次に示す組成で I F N— S組成物を調製した c なお、 組成比は重量パーセン 卜で示した。

I F N - ^ ix 105 IU/g 以上のような組成で調製した I F N - /3組成物のサン プルを 3 0でで放置し、 4 , 1 2, 24, 36および 5 2週間後にサンプリ ングを行い、 力価を測定し、 初期力 価 ( 1 X 1 05 IU/g) を 1 0 0 %として I F N— ^の力 価残存率を算出した。 結果を第 1図に示す。
実施 例 2

I F N — . IX 105 IU/g 以上の様な組成で調製した I F N— 組成物のサンプ ルを 3 0 で放置し、 1 2、 24、 36、 5 2および 6

0週間後にサンプリ ングを行い、 力価を測定した。 初期 力価 ( I x l O 5 I V / g ) を 1 0 0 %と して I F N— ^の力価残存率を算出した。
結果を表 1 に示す。

本発明の組成物は、 室温で長期間安定な I F N — ^組 成物であり、 さらに、 本発明組成物は種々の添加剤を含 んでいても I F N— /3が失活せず、 I F N — ^の安定化 効果が長期間持続する優れた組成物である。
よって本発明の組成物は、 溶剤、 ゲル、 スプレー、 軟 骨等の種々の剤形に製剤化が可能であるが、 特に軟骨剤 に適している。
Claims
1 . ポ リ オ一ルを 6 5 〜 9 0重量%と、 p — ヒ ドロキシ ベンゾエー ト類とを含有してなる安定なイ ンタ一フエ ロ ン ^組成物。
2 . p — ヒ ドロキシベンゾエー ト類の含有量が、 0 , 0 1 〜 0 . 2重量%である請求の範囲第 1項記載の安定 なイ ン タ ー フ ェ ロ ン /3組成物。
3 . P — ヒ ドロキシベンゾエー ト類がェチノレ p — ヒ ドロ キシベンゾエー ト も し く は、 プロ ピノレ p — ヒ ドロキシ ベンゾエー トである請求の範囲第 1項記載の安定なィ ンターフヱロ ン /3組成物。
4 . ポ リオールの含有量が、 7 0 〜 8 5重量%である請 求の範囲第 1項記載の安定なイ ンタ一フ ュ ロ ン 組成 物。
5 . ポ リオールが 2価以上のアルコールである請求の範 囲第 1项記載の安定なイ ンターフェ ロ ン /5組成物。
6 . ポ リオールが 3価のアルコールである請求の範囲第 5項記載の安定なィ ンタ一フ ロ ン ^組成物。
7 . ポ リオールがグリ セ リ ンである請求の範囲第 6項記 載の安定なイ ンタ一フ ュロ ン /3組成物。
8 . 增粘剤および安定化剤をさ らに含有する請求の範囲 第 1項記載の安定なイ ンタ一フュ ロ ン /3組成物。
9 . 増粘剤の含有量が 0 . 1 〜 2 . 5重量%である請求 の範囲第 8項記載の安定なィ ンターフ ロ ン 5組成物。
10. 增粘剤がカルボキシメ チルセルロースおよびその塩、
メ チルセルロース、 ヒ ドロキシェチルセルロース、 デ ンプンおよび微小繊維状セルロースから選ばれたもの である請求の範囲第 8項記載の安定なィ ンターフェ口 ン 3組成物。
11. 増粘剤がカルボキシメチルセルロースも しく はその 塩である請求の範囲第 1 0項記載の安定なィ ンターフ ェロン ^組成物。
12. 力ルボキシルメチルセルロースが 1 %水溶液と して 2 5。C、 6 O rpin の条件下に、 B型粘度計により測定 したときの粘度が 3 0〜 2 0 ◦ cps である請求の範囲 第 1 1項記載の安定なィ ンターフェ口 ン ^組成物。
13. 安定化剤の含有量が 0 . 1〜 1重量%である請求の 範囲第 8項記載の安定なィ ンタ一フユ口 ン S組成物。
14. 安定化剤がヒ ト血清アルブミ ンまたはヒ ト血清グロ プリ ンである請求の範囲第 8項記載の安定なィ ンター フエロ ン ^組成物。
15. 安定化剤がヒ 卜血清アルブミ ンである請求の範囲第 1 4項記載の安定なィンターフェロン /3組成物。
16. イ ンターフェロ ン ^力 ヒ ト由来のものである請求の 範囲第 1項記載の安定なィ ンターフニ口 ン ^組成物。
17. イ ンタ一フエロ ン Sが 1 X 1 0 4 国際単位 ( I U ) Z g以上の力価を有する請求の範囲第 1項記載の安定 なイ ンタ 一 フ エ ロ ン β組成物。
