Description
本發明係關於一種藥物載體,特別係關於一種具有螯合型複合微胞之藥物載體及其配方與形成方法。
癌症病患接受放射線治療與化學治療時所引發之副作用眾人皆知,抗癌治療藥物的毒性也一向為病患所詬病,因為嚴重副作用會造成許多癌症病患之二度傷害與更大之夢靨;如果病患因為放射線治療與化學治療時所引發之嚴重副作用而不能完成預定之療程,不僅未能達到療效也造成醫療資源之浪費。放射線治療與化學治療不論對正常細胞或癌細胞都無選擇性,因此在癌細胞被消滅的同時,正常細胞亦遭受池魚之殃,病人常見治療後有口腔潰瘍、食慾不振、腹瀉、落髮及白血球、血小板下降等副作用,甚至引發其他可致命的併發症。副作用往往也限制了治療劑量的給予,影響了療效;部份副作用可以靠改變給藥方法予以減輕,如分散或長時間給藥,或以局部給藥方式,像腹膜內或動脈化療,讓高濃度的藥物只局限於病灶所在,不致引發全身性的副作用;或待藥物作
用時間過後,馬上給予某種拮抗藥物(解毒劑),減除抗癌藥部份細胞殺傷力。然而上述方法並無選擇性,只能稍減對正常細胞的毒性,相對於癌細胞的療效也被打折扣,他法即是迅速治療發生的副作用,儘量降低傷害的程度,如白血球生成素及強效止吐劑的注射,但此法亦僅部份減輕已形成的副作用,並無預防功效,因此療效與零副作用猶同魚與熊掌,不可兼得。又腫瘤形成後,癌細胞不斷滋生,經由淋巴管及血管蔓延各處,族群越來越複雜,最後生出有抗藥性的細胞群,此時腫瘤即到達無法根治的地步,任何治療皆告無效;故抗癌治療藥物的毒性與放射線治療無法有效控制腫瘤滋生蔓延,是目前治癌無法奏全功的絆腳石。據此,為令癌症病患免除或減少副作用,並能完成放射線治療與化學治療之療程以增加療效,重點即在於如何能在攻擊癌細胞時同時保護正常細胞。部分的化學治療與放射線治療作用機轉來自於自由基傷害,如果能夠選擇性的利用具抗氧化與自由基物質,可以降低這些治療對正常細胞的傷害。
氨磷汀Amifostine(Ethyol;WR-2721)係一種含磷酸硫(thiophosphate)的抗輻射細胞保護劑,其為phosphorylated aminothiol的前驅物,此藥物須經由存在生物體內的細胞、體液和血液的鹼
性磷酸酶(alkaline phosphatase)的去磷酸化作用(dephosphorylated),此為一種鹼性磷酸酵素作用,之後才能轉換成有保護細胞作用的活性代謝物free thiol,其通稱為WR-1065,可使細胞不受輻射及化學藥物的傷害,進而使癌症病患免於放射線治療與化學治療時所引發之嚴重副作用。由於癌細胞之鹼性磷酸酵素含量較正常細胞低得多,癌細胞內的血流狀況及較酸環境亦不利鹼性磷酸酵素作用,Amifostine的組織保護作用機轉可與alkylating agents、platinum analogs的活性物質結合成thioether conjugates,防止其與正常組織的DNA或RNA結合;另外WR-1065可部分逆轉已形成DNA-platinum的內轉產物;協助移除DNA-platinum,讓正常的DNA繼續進行作用;亦可提供H基給DNA的游離子修補DNA的損傷。WR-1065同時也是一個強力的ROS(Reactive oxygen species)淨化劑,可清除放射線治療及特定化學治療藥物所產生的ROS,預防細胞損傷。在體外亦可有效清除-OH基相關的spin-trap訊號、過氧化陰離子及Doxorubincin衍生的過氧化陰離子,另外WR-1065對AOS的清除作用,可預防Bleomycin所誘發的肺臟發炎及纖維化。
氨磷汀Amifostine可減少癌症化學治療引起的毒性作用、保護正常的組織、提高化療及電療的有效治療劑量、增加反應率,此外,氨磷汀Amifostine更可配合其他生長因子,細胞素共同使用,具有加成保護造血幹細胞效果。氨磷汀Amifostine注射5-10分鐘後即開始產生細胞保護作用,具短暫的血中半衰期(βt1/2=8.8min),在血漿中快速的清除,約90%會在6分鐘內清除。迅速分佈於各組織,在正常組織中經由酵素相關的去磷酸化作用,快速利用,滯留時間長,達到穩定狀態約10分鐘。相反的,此作用在腫瘤組織中則利用緩慢或分佈極少。氨磷汀Amifostine注射後,於正常細胞內的濃度較癌細胞高十倍,包括腎臟、肺臟、肝臟、皮膚、骨髓、腸道及脾臟等,較少存於腦組織、橫紋肌及腫瘤細胞,因此氨磷汀Amifostine對正常細胞有選擇性的保護作用,但須於化療或放射治療前15~30分鐘給予。然而,一般的治療期間需有幾個小時的療程,因此,如何能延長氨磷汀Amifostine於人體內的半衰期即為當前產業亟需發展之重要標的。
除上述氨磷汀Amifostine外,一般藥物劑型在體內代謝半衰期都很短,經服用或以針劑方式進入人體後,會廣泛的散佈至全身各組織,為了維持長時間
的藥效和有效的藥物分佈,須用比實際藥物濃度需求量更多的劑量或以多次投藥方式,方能達到預期療效。但其也因此毒害正常組織,且引起不必要的副作用。為了改善傳統藥劑重複多次投藥的缺點,避免藥物過量和浪費,提出藥物傳釋系統(drug delivery system;DDS)的概念,以增加藥物的療效並減少用藥次數。其中一種商業用的載體如脂質體(或稱微脂囊;Liposome),其係為一種球形載體,主由單層或多層的卵磷脂(Phosphatidylcholin;PC)所構成,由於結構限制只能攜帶親水性的藥物(包覆于內層),尤其容易累積在肝臟內,其對於溫度相當敏感,因而保存困難,也不易以乾粉的型態運輸,參考C.Chen,D.Han,et al.(2010)."An overview of liposome lyophilization and its future potential." 高分子奈米微胞是極具潛力的藥物載體之一,其優點包括提高藥物效率、降低細胞毒性、保護與安定藥物、提供標的效果等,而奈米尺寸的微胞具有延長體內循環時間、躲避巨噬細胞(mononuclear phagocyte system,MPS)與內質網(reticular epithelial system,RES)的功能,因此是目前最受
矚目的藥物控制釋放系統。一般的奈米微胞由兩性團聯共聚物(amphiphilic copolymer)構成。在水相環境下,高分子鏈段因親疏水性的不同會排列形成核殼結構(core-shell structure)微胞,因此提供了疏水性藥物(如:indomethacin、adriamycin、amphotericin B)絕佳的保護環境,以高分子微胞攜帶抗癌藥物亦可有效提高藥物的安定性與效率。高分子微胞可藉由物理性包覆、化學鍵結或靜電作用力來包覆藥物。物理性包覆的驅動力主要是疏水性鏈段與藥物間的疏水性作用力。化學鍵結方式即利用共價鍵鍵結藥物,如醯胺鍵(amide bond),此類鍵結非常安定,不易被酵素分解或水解,因此須在藥物與高分子之間導入一特定環境下會斷裂之隔離基(spacer)以利藥物釋放。以物理作用力在包覆疏水性藥物時往往會造成較多的損耗;而以化學作用力包覆藥物則因合成步驟較繁複,載藥量會較低。
除上述兩性質包覆藥物的方法外,目前也有用靜電作用力的高分子微胞。通常這種高分子微胞會設計成一端的不帶電性的高分子形成殼(shell),而另一端則具有電荷鏈段,其在經過相反電荷之基因或是藥物中和電性後會變成較疏水性的核(core),藉此可形成具有殼核(core-shell)結構的聚離子錯合型微胞
(Polyion Complex micelles;PIC micelles)。然而,利用靜電吸附藥物亦有其限制,分子量太小或水溶性藥物很容易被取代置換,如同離子交換樹脂般,無法形成微胞的核。另一種方式則在於直接鍵結藥物,尤其是含金屬之藥物,如Cisplatin(順鉑)、Carboplatin(卡鉑)、Oxaliplatin(奧沙利鉑)等等,然而,由於這些含金屬之藥物於此係用以當成路易士酸,配位子與鉑Pt鍵結後藥物的官能基已被取代,不是原本藥物結構,因此必須重新驗證藥物的安全性與有效性等,恐將導致成本大幅增加。據此,更是需要發展一種新的藥物釋放之載體即為當前產業亟欲發展之重要標的。
鑒於上述之發明背景中,為了符合產業上特別之需求,本發明提供一種具有配位鍵之螯合型藥物載體可用以解決上述傳統技藝未能達成之標的。
本發明之一目的係提供一種具有金屬離子核心之載體結構,本發明係以至少一金屬離子(包含過渡金屬)為中心,並與具有螯合端之高分子(包含崁段共聚物;block copolymer)形成具有配位鍵之藥物載體,以使得可提供電子對之藥物(包含具有官能基
如羧酸carboxylic acids,醇alcohols,酮ketones,呋喃furans,胺amines,苯胺anilines,吡咯pyrroles,硫醇thiols,酯esters,醯胺amides,亞胺imines,吡啶pyridines,嘧啶pyrimidins,咪唑imidazoles,比唑pyrazols,磺胺sulfonamides,膦酸phosphonic acids等)與金屬離子藉由配位鍵鍵結,藉此可與具有螯合端之高分子同時形成螯合型複合微胞(Chelating Complex Micelles;CCM)。眾所周知,配位鍵(又稱配位共價鍵,或簡稱配鍵;Coordinate bond)是一種特殊的共價鍵,當共價鍵中共用的電子對由其中一原子獨自供應時,即稱之為配位鍵。配位鍵的形成需要兩個條件:一是中心原子或離子,它必須有能接受電子對的空軌道;二是配位體,組成配位體的原子必須能提供配對的孤對電子。當一路易士鹼供應電子對給路易士酸而形成化合物時,配位鍵就形成了。在配位化合物中,由電負性小的元素原子向電負性大的元素原子提供孤對電子形成配位鍵時,每個有一對孤對電子的前者(電負性小的原子)顯示正價,後者顯示負價。反之,由電負性大的元素原子提供孤對電子與電負性小的元素原子之間形成配位鍵時,兩種元素都無價態變化。
本發明之螯合型複合微胞(CCM)在製備上比一般物理/化學作用力包覆藥物簡單。使用物理或是化學性包覆藥物往往受到鏈段設計上的限制,通常僅能包覆疏水性藥物,而這些藥物與高分子進行包覆時由於溶解度不佳,造成在水溶液中會有很多損耗。因此,本案架構僅需藥物具有可提供電子對官能基,且能與金屬形成配位鍵者,即可混成具有多牙配位子(polydentate ligands)之螯合型複合微胞。不僅可以包覆疏水性藥物也能夠包覆親水性藥物,所以能更加廣泛在其他類型的藥物傳輸系統上;而且配位鍵與藥物間的吸引力較親疏水作用力強進而能提高包覆效率以及增加藥物溶解度。
再者,本發明之金屬離子核心除可提供作為高分子與藥物連結中心外,亦可提供如使用釓Gd可作為MRI磁振造影、鎝99mTc(SPECT單光子電腦斷層)、鎵68Ga(PET/CT正子電腦斷層)、錸188Re(體內放射療法)以及銦111In(白血球造影)等顯影功能,本發明藉此應用而提供一即時監控造影與治療(Real-time monitoring and therapy)的技術手段,若再加上可提供標靶效果的標的分子(targeting moiety),如葉酸(folic acid),等,能累積至腫瘤組織以達到攻擊癌細胞的目的。
本發明之另一目的係提供一種細胞保護劑載體,其係以一具有配位基之金屬(包含過渡金屬)離子為核心(Core),其分別與一細胞保護劑(包含氨磷汀Amifostine、WR-1065)以及至少一嵌段共聚物(包含聚乙二醇-聚麩胺酸崁段共聚物;PEG-b-PGA)以配位方式鍵結形成細胞保護劑載體,此外,本發明之崁段共聚物中不具螯合能力之高分子鏈段會延伸在螯合型複合微胞外側,以穩定微胞結構與強化分散性。
據此,本發明之螯合型複合微胞同時具有多牙配位子以及金屬離子,其具備可包覆親、疏水性藥物、生物可分解、流體分散性好、可顯影、延展藥物之等特質,故其可廣泛地應用於醫藥之商業與工業中以達成產業亟需發展之重要標的。
根據本發明上述之目的,本發明提供一種螯合型複合微胞,該螯合型複合微胞包含一金屬離子核心,該金屬離子核心為路易士酸;與至少一配位子,該配位子係與該金屬離子核心藉由配位鍵彼此鍵結。上述之金屬離子核心係選自下列族群中之一者或其任意組合或其衍生物:鐵Fe,銅Cu,鎳Ni,銦In,鈣Ca,鈷Co,鉻Cr,釓Gd,鋁Al,錫Sn,鋅Zn,鎢W,鈧Sc,鈦Ti,錳Mn,釩V,鎂Mg,鈹Be,鑭
La,金Au,銀Ag,鎘Cd,汞Hg,鈀Pd,錸Re,鎝Tc,銫Cs,鐳Ra,銥Ir,鎵Ga,及其組合。其中該配位子係選自下列族群中之一者或其任意組合或其衍生物:羧酸carboxylic acids,醇alcohols,酮ketones,呋喃furans,胺amines,苯胺anilines,吡咯pyrroles,硫醇thiols,酯esters,醯胺amides,亞胺imines,吡啶pyridines,嘧啶pyrimidins,咪唑imidazoles,比唑pyrazols,磺胺sulfonamides,膦酸phosphonic acids。
上述之螯合型複合微胞更包含一藥物,該藥物係為路易士鹼,且該藥物係與該金屬離子核心藉由配位鍵彼此鍵結。上述之藥物係包含一或多個官能基,該官能基其係選自下列族群中之一者或其任意組合或其衍生物:羧酸carboxylic acids,醇alcohols,酮ketones,呋喃furans,胺amines,苯胺anilines,吡咯pyrroles,硫醇thiols,酯esters,醯胺amides,亞胺imines,吡啶pyridines,嘧啶pyrimidins,咪唑imidazoles,比唑pyrazols,磺胺sulfonamides,膦酸phosphonic acids,其中上述之藥物係選自下列族群中之一者或其任意組合或其衍生物:Amifostine(氨磷汀),WR-1065,Doxorubicin(阿黴素),Pemetrexed(培美曲塞),

根據本發明上述之目的,本發明提供一種具有螯合型複合微胞之藥物載體,該具有螯合型複合微胞之藥物載體包含至少一高分子聚合物,該高分子聚合物係為路易士鹼;與一金屬離子核心,該金屬離子核心係為路易士酸,且該金屬離子核心係與該高分子聚合物形成配位鍵,且該具有螯合型複合微胞之藥物載體用以搭載一為路易士鹼之藥物,,其中上述之金屬離子核心、藥物的選擇係相同於本發明之螯合型複合微胞的內容。此外,上述之高分子聚合物為下列之一:unidentate ligands(單牙配位子),bidentate ligands(雙牙配位子),tridentate ligands(三牙配位子),hexadentate ligands(六牙配位子),polydentate ligands(多牙配位子),且該高分子聚合物的分子量(Molecular weight)範圍約為1,000-100,000道爾頓(Dalton),其中上述之高分子聚合物係選自下列族群中之一者或其任意組合或其衍生物:聚乙二醇Poly(ethylene glycol),聚天

根據本發明上述之目的,本發明提供一種具有螯合型複合微胞之醫藥組合物,該具有螯合型複合微胞之醫藥組合物包含一嵌段共聚物,該嵌段共聚物係為路易士鹼,其中,該嵌段共聚物更包含一螯合端以進行配位鍵結,與一分散端以使該醫藥組合物能穩定而良好地分散於生物流體中;一藥物,該藥物係包含路易士鹼之官能基;與一金屬離子核心,該金屬離子核心係為路易士酸,且該金屬離子核心係分別與該嵌段共聚物以及該藥物形成配位鍵。上述之金屬離子核心
係為一二價鐵離子、三價鐵離子或三價釓離子,且上述之藥物係為一氨磷汀(Amifostine)或WR-1065,而上述之嵌段共聚物係為一聚乙二醇-聚麩胺酸嵌段共聚物(Poly(ethylene glycol)-b-Poly(glutamic acid);PEG-b-PGA),其中,聚麩胺酸為該螯合端,且聚乙二醇為該分散端。
根據本發明上述之目的,本發明提供一種具有螯合型複合微胞之醫藥組合物的製造方法,該具有螯合型複合微胞之醫藥組合物的製造方法包含提供一原料,該原料包含一氨磷汀(Amifostine)、一聚乙二醇-聚麩胺酸嵌段共聚物(PEG-b-PGA)與一氯化亞鐵(FeCl2);與將該原料置入緩衝液中進行一勻相擾動程序,藉此,該氨磷汀(Amifostine)經由該亞鐵離子(Fe2+)與該聚乙二醇-聚麩胺酸嵌段共聚物藉由配位鍵直接進行自組裝(self-assembly)反應,上述之氨磷汀(Amifostine)之重量為0.1-10mg、該聚乙二醇-聚麩胺酸嵌段共聚物(PEG-b-PGA)之重量為0.1-100mg,及該氯化亞鐵(FeCl2)之重量為0.01-50mg。上述之氨磷汀(Amifostine)濃度約為0.01-10mg/mL,且上述之緩衝液可為HEPES[4-(2-hydroxyethyl)-1-piperazinee-thanesulfonic acid)],其中,上述之緩衝液的pH
值約為6.5~7.5之間,此外,上述之勻相擾動程序係於4-40℃下進行。

根據本發明之一第三實施例,本發明提供一種具有螯合型複合微胞之之醫藥組合物,如:細胞保護劑,具有螯合型複合微胞之醫藥組合物更包含一金屬離子核心、至少一嵌段共聚物(block copolymer)與藥物,其中,金屬離子核心用以當成路易士酸
(Lewis acid),且至少一嵌段共聚物(block copolymer)與藥物皆當成路易士鹼(Lewis base),並藉由路易士鹼(Lewis base)提供電子對而使得金屬離子核心能與嵌段共聚物(block copolymer)與藥物形成配位鍵。
上述之金屬離子核心更包含一Fe鐵離子或Gd釓離子,且上述之藥物更包含氨磷汀Amifostine、WR-1065,而上述之嵌段共聚物更包含一端為螯合端可與鐵離子進行鍵結,另一端為分散端以便使具有螯合型複合微胞之醫藥組合物能穩定而良好地分散於生物流體中,例如,聚乙二醇-聚麩胺酸崁段共聚物(Poly(ethylene glycol)-b-Poly(glutamic acid);PEG-b-PGA)。上述之聚乙二醇-聚麩胺酸崁段共聚物係以具有高生物適應性之聚麩胺酸提供螯合端(-COO-)以便與鐵離子產生配位鍵,同時藉由具水溶性之聚乙二醇的高分散性使得整體藥劑能穩定而良好地分散於生物流體中,其中值得注意的是,聚乙二醇本身並無可提供任何具配位的官能基,聚乙二醇必須經由適當地改質始得與金屬形成配位鍵,本實施例所使用之聚麩胺酸的目的之一亦在於連結分散度高的聚乙二醇與鐵離子,且聚麩胺酸係為一種生物可分解性高分子天然物質。
上述之藥物更包含氨磷汀Amifostine、WR-1065,Amifostine具有五個提供電子對之處,其分別位於兩個-OH、-P=O、NH與-NH2;WR-1065則具有三個提供電子對之處,其分別位於-SH、-NH與-NH2。
根據本實施例,參考第一圖所示,具有螯合型複合微胞之醫藥組合物100之製作方法如下,取0.1-10mg細胞保護劑氨磷汀(Amifostine)110,0.1-100mg聚乙二醇-聚麩胺酸崁段共聚物(PEG-b-PGA)120與0.01-50mg氯化亞鐵(FeCl2)130。然後,將上述混合原料置入1-10mL緩衝液中於4-40℃下進行0至48小時之勻相擾動程序,藉由前述的鍵結原理,氨磷汀Amifostine 110可經由Fe離子130與PEG-b-PGA 120直接進行自組裝(self-assembly)反應而形成粒徑約為10~300nm之間的具有螯合型複合微胞之細胞保護劑300,其中上述之緩衝液可為0.05M之HEPES(4-(2-hydroxyethyl)-1-piperazineethane-sulfonic acid),其pH值約為6.5~7.5之間。
根據上述實施例,上述之氨磷汀(Amifostine)及其衍生物的結構如第二A圖與第二B圖所示,而聚乙二醇-聚麩胺酸崁段共聚物(PEG-b-PGA)之結
構式如第三圖所示,且具有螯合型複合微胞之細胞保護劑的結構式如第四圖所示。根據本實施例,第五圖所示係為以聚乙二醇PEG為巨起始劑(macroinitiator)合成聚乙二醇-聚麩胺酸崁段共聚物PEG-b-PGA,其重量平均分子量Mw 5700,數目平均分子量Mn 4900,分布係數PdI 1.16,其中,第六圖所示則為PEG-b-PGA之氫核磁共振光譜(Proton Nuclear Magnetic Resonance,1H-NMR)與相對應官能基(functional groups)之化學位移(chemical shift)。第七圖所示則為以粒徑分析儀分析螯合型複合微胞(Chelating Complex Micelles;CCM),平均粒徑為31.61nm。
第八圖所示,取5mg氨磷汀(Amifostine)、20mg PEG-b-PGA(分子量5700Dalton)與1.25-10mg FeCl2,重量比為(1:4:0.25至1:4:2)。然後,將上述原料置入5mL緩衝液中於25℃下進行24小時之均勻擾動程序,並置入分子量3500之透析膜模擬藥物釋放。結果顯示FeCl2 10mg時,多數藥物已與鐵離子與PEG-b-PGA自組裝(self-assembly)反應而形成螯合型複合微胞,因此停留在透析膜內未釋放。其中上述之緩衝液為0.05M之
HEPES(4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid),其pH值約為7.0。
第九圖所示,取5mg氨磷汀(Amifostine)、20mg PEG-b-PGA(分子量5700Dalton)與10mg FeCl2。然後,將上述原料置入5mL緩衝液中於25℃下進行不同時間之均勻擾動程序,並置入分子量3500之透析膜模擬藥物釋放。結果顯示均勻擾動24小時藥物氨磷汀(Amifostine)釋放到透析膜外之比例較低,多數已與鐵離子與PEG-b-PGA自組裝(self-assembly)反應而形成螯合型複合微胞。其中上述之緩衝液為0.05M之HEPES(4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid),其pH值約為7.0。
一般的藥劑上通常無法直接與高分子連接改質,因此,藉由本發明的配位共價的概念,利用金屬離子當作路易式酸來自行同時結合藥劑與高分子,所形成的具有螯合型複合微胞不但具有如同螯合劑(EDTA)的結構,同時適度地延長了藥物於人體內半衰期(可於人體內循環更久而不衰退),而且不會造成人體毒性(EDTA會攫取人體內的金屬元素)。此外,配位共價鍵亦可彌補以電荷相吸的鍵結強度不足之問題。再者,金屬離子本身多易沈澱,而PEG
本身雖然具有很好的流體分散度,但並無官能基可接其他物質,因此,必須透過PGA的改質以便與金屬離子鍵結,以便於整體載體能均勻分散於流體中而不致因金屬離子造成沈澱阻塞的現象。值得注意的是,本發明之載體能適用各種親水(hydrophilic)或親油(hydrophobic)的藥物,而提升了相當高的應用價值。此外,對於已經符合FDA或相關醫藥檢驗規範的藥物而言,若是破壞原藥物的結構,即使僅佔極小比例,都很可能被視為新藥而需重新進行安全性與有效性之驗證,耗費成本極高,相對而言,本發明完全不會破壞藥物本身結構,不會造成重新驗證的問題,對於製程相容性而言,具有極高的成本價值。
顯然地,依照上面實施例中的描述,本發明可能有許多的修正與差異。因此需在其附加的權利請求項之範圍內加以理解,除上述詳細描述外,本發明還可以廣泛地在其他的實施例中施行。上述僅為本發明之較佳實施例而已,並非用以限定本發明之申請專利範圍;凡其它未脫離本發明所揭示之精神下所完成的等效改變或修飾,均應包含在下述申請專利範圍內。

- 如申請專利範圍第1項所述之螯合型複合微胞,其中上述之配位子係選自下列族群中之一者或其任意組合:羧酸(carboxylic acids),醇(alcohols),酮(ketones),呋喃(furans),胺(amines),苯胺(anilines),吡咯(pyrroles),硫醇(thiols),酯(esters),醯胺(amides),亞胺(imines),吡啶 (pyridines),嘧啶(pyrimidins),咪唑(imidazoles),比唑(pyrazols),磺胺(sulfonamides),膦酸(phosphonic acids)。
- 如申請專利範圍第1項所述之螯合型複合微胞,該螯合型複合微胞更包含一藥物,該藥物係為路易士鹼,且該藥物係與該無配位基金屬離子核心藉由配位鍵彼此鍵結。

- 一種具有螯合型複合微胞之藥物載體,該具有螯合型複合微胞之藥物載體包含:至少一高分子聚合物,該高分子聚合物係為路易士鹼;與一無配位基(ligand free)金屬離子核心,該無配位基金屬離子核心係為路易士酸,且該無配位基金屬離子核心係與該高分子聚合物形成配位鍵。

- 如申請專利範圍第7項所述之具有螯合型複合微胞 之藥物載體,該具有螯合型複合微胞之藥物載體用以搭載一為路易士鹼之藥物。
- 如申請專利範圍第9項所述之具有螯合型複合微胞之藥物載體,其中上述之藥物係選自下列族群中之一者或其任意組合:Amifostine(氨磷汀),WR-1065,Doxorubicin(阿黴素),Docetaxel(多西紫杉醇),Epirubicin(表柔比星),Amphotericin(兩性黴素),Ciprofloxacin(環丙氟哌酸)。
- 如申請專利範圍第7項所述之具有螯合型複合微胞之藥物載體,其中上述之高分子聚合物為下列之一:unidentate ligands(單牙配位子),bidentate ligand s(雙牙配位子),tridentate ligands(三牙配位子),hexadentate ligands(六牙配位子),polydentate ligands(多牙配位子)。
- 如申請專利範圍第7項所述之具有螯合型複合微胞之藥物載體,其中該高分子聚合物的分子量(Molecular weight)範圍約為1,000-100,000道爾頓(Dalton)。
- 如申請專利範圍第7項所述之具有螯合型複合微胞之藥物載體,其中上述之高分子聚合物係選自下列族群中之一者或其任意組合:聚乙二醇[Poly(ethylene glycol)],聚天門冬氨酸[Poly(aspartic acid)],聚麩胺酸[Poly(glutamic acid)],聚賴氨酸(Polylysine),聚丙烯酸[Poly(acrylic acid)],幾丁聚醣(chitosan),聚乙烯亞胺(Polyethyleneimine),聚甲基丙烯酸[Poly(methacrylic acid)],透明質酸(hyaluronic acid),膠原蛋白(collagen),聚(氮-異丙基丙烯醯胺)[Poly(N-isopropyl acrylamide)],直鏈澱粉(amylose),纖維素(cellulose),聚羥基丁酸酯[poly(hydroxybutyrate)],聚乳酸[poly(lactic acid)],聚琥珀酸丁酯[poly(butylenesuccinate)],聚己內酯[poly(caprolactone)],羧甲基纖維素(carboxymethylcellulose),糊精(dextran),環糊精(cyclodextrin)。

- 如申請專利範圍第14項所述之具有螯合型複合微胞之醫藥組合物,其中上述之嵌段共聚物係為一聚乙二醇-聚麩胺酸嵌段共聚物[Poly(ethylene glycol)-b-Poly(glutamic acid);PEG-b-PGA],其中,聚麩胺酸為該螯合端,且聚乙二醇為該分散端。
- 一種具有螯合型複合微胞之醫藥組合物的製造方法,該具有螯合型複合微胞之醫藥組合物的製造方法包含:提供一原料,該原料包含氨磷汀(Amifostine)、聚乙二醇-聚麩胺酸嵌段共聚物(PEG-b-PGA)與氯化亞鐵(FeCl2);與將該原料置入緩衝液中進行一勻相擾動程序,藉此,該氨磷汀(Amifostine)經由該亞鐵離子(Fe2+)與該聚乙二醇-聚麩胺酸嵌段共聚物藉由配位鍵直接進行自組裝(self-assembly)反應。

- 如申請專利範圍第18項所述之具有螯合型複合微胞之醫藥組合物的製造方法,其中上述之氨磷汀(Amifostine)濃度約為0.01-10mg/mL。
- 如申請專利範圍第18項所述之具有螯合型複合微胞之醫藥組合物的製造方法,其中上述之緩衝液可為HEPES[4-(2-hydroxyethyl)-1-piperazine-ethanesulfonic acid)]。
- 如申請專利範圍第18項所述之具有螯合型複合微胞之醫藥組合物的製造方法,其中上述之緩衝液的pH值約為6.5~7.5之間。
- 如申請專利範圍第18項所述之具有螯合型複合微胞之醫藥組合物的製造方法,其中上述之勻相擾動程序係於4~40℃下進行。
