單株抗體(mAb)的生產及其純化由於該過程的高成本持續造成問題。分離過程中生物分子必須經歷的數個純化步驟導致了高成本。舉例來說,純化步驟中的一個步驟是蛋白質A親和力層析,其中葡萄球菌的蛋白質A以高選擇性以及與Fab區域(藥物分子的活性區域)極小的交互作用與第1、2及4亞型的IgG分子結合。隨著生物科技市場的快速成長,這些純化步驟的改良對於將生物製劑及時地並以降低的成本引進市場變得更令人嚮往也更有價值。

所述共聚物的分子量範圍可以是任何合適的範圍,例如,從50到4000 kD、100到3500 kD、或200到1500 kD、50到3126 kD。最佳的分子量範圍是從207 kD到1270 kD,且在具體實例中,從50到3126 kD,且較佳地從207 kD到1270 kD,其中所述分子量是以三合一偵測GPC分析來測定。

在具體實例中,對於膜的應用,影響層析應用中的鹽耐受度同時允許有效率地回收蛋白質之單體組合的範圍是從莫耳比例1:0.25:2到莫耳比例2:0.5:1。此範圍特別可在使用包含以下者的三單體來形成在膜上交聯的聚合物的情況下應用:磺酸2-丙烯醯胺基-2-甲基丙酯、N-(3-胺基丙基)甲基丙烯醯胺與任何烷基丙烯醯胺(諸如N,N-二甲基丙烯醯胺、N-三級丁基丙烯醯胺、N-苯基丙烯醯胺、N,N-二乙基丙烯醯胺、N-異丙基丙烯醯胺、N-乙基丙烯醯胺、N,N-二乙基甲基丙烯醯胺、N-(異丁氧基甲基)甲基丙烯醯胺、丙烯醯胺及N-(三苯甲基)甲基丙烯醯胺)。
在具體實例中,當使用磺酸2-丙烯醯胺基-2-甲基丙酯、N-(3-胺基丙基)甲基丙烯醯胺及N,N-二甲基丙烯醯胺之三單體來形成在用於純化蛋白質的膜上交聯的聚合物時,提供高鹽耐受度並同時提供高達200 mg/mL IgG之高動態結合能力的聚合物濃度範圍為以重量計從1% - 10%。
本發明進一步提供產生多孔膜的方法,其包含:
(i) 使單體(A)、(B)及(C)之混合物聚合,以獲得三元共聚物,以及
(ii) 將(i)中所獲得的三元共聚物塗覆在多孔撐體上並原位交聯,以獲得配置於所述多孔撐體上的共聚物塗層。
單體的聚合可使用合適的起始劑進行–自由基、陽離子或陰離子起始劑,且特別是自由基起始劑,例如:熱活化自由基起始劑,諸如偶氮基化合物、過硫酸鹽、過氧化物、過酸、過乙酸鹽、及有機金屬化合物。自由基起始劑的例子包括AIBN、4,4-偶氮基雙(4-氰戊酸)、1,1'-偶氮基雙(環己烷碳腈)(1,1'-azobis(cyclohexanecarbonitrile))、過氧化苯甲醯、2,2-雙(三級丁基過氧)丁烷、1,1-雙(三級丁基過氧)環己烷、二異丙苯基過氧化物、過氧苯甲酸三級丁酯、過氧苯甲酸三級戊酯及過硫酸鉀。
根據所述方法,所述交聯包含二或多個基團,例如2、3、4或更多個交聯官能基,例如:選自二環氧化物、二醛、二鹵化物及二酯之交聯劑。例如,所述交聯劑是選自:乙二醇二環氧丙基醚、丙二醇二環氧丙基醚、甘油基三環氧丙基醚、聚乙二醇二環氧丙基醚、二甲磺酸二醇酯與二甲苯磺酸酯及其任何組合。
本發明進一步提供一種處理含有痕量金屬之流體的方法,所述方法包含使所述流體與如上所述之介質或多孔膜接觸,並回收含有降低濃度的痕量金屬之流體。
根據本發明的一個具體實例,多孔膜可以是奈米多孔膜,例如:孔直徑為1 nm至100 nm之間的膜,或是微米多孔膜,例如:孔直徑為0.005 µm至10 µm之間的膜。
將塗層提供於其上的多孔撐體可以是任何合適的材料,例如:聚合物、金屬或陶瓷,且特別是聚合物,例如:聚碸(PSU)、聚醚碸(PES)、聚苯醚(PPE)、聚伸苯醚碸(PPES)、聚伸苯醚(PPO)、聚碳酸酯(PC)、聚(呔酮醚碸酮)(poly(phthalazinone ether sulfone ketone),PPESK)、聚醚醚酮(PEEK)、聚醚酮酮(PEKK)、聚醚醯亞胺(polyetherimide,PEI)及其摻合物。
在一個具體實例中,所述共聚物被塗覆且經交聯於以下者上以產生多孔介質或膜:多孔高密度聚乙烯膜、聚醚碸(polyether sulfone)膜、聚偏二氟乙烯膜、聚四氟乙烯膜、聚苯基碸(polyphenylsulfone)膜、聚伸苯硫(polyphenylene sulfide)膜、聚乙烯和聚丙烯膜、聚酯膜、熔噴(melt blown)聚酯膜、熔噴聚丙烯膜、纖維素膜、尼龍膜和聚氯乙烯/丙烯腈。
根據本發明之具體實例的多孔介質可以配置於容納裝置(housing)中,其包含至少一個入口及至少一個出口,並限定出至少一個入口及出口之間的流體流動路徑,其中至少一個本發明之膜或包含至少一個本發明之膜的濾器橫越所述流體流動路徑,以提供過濾裝置或過濾組件。在一個具體實例中,提供包含容納裝置的過濾裝置,所述容納裝置包含入口以及第一出口,並限定出所述入口及第一出口之間的第一流體流動路徑;以及至少一個本發明的膜或包含至少一個本發明之膜的濾器,所述本發明的膜或包含至少一個本發明之膜的濾器被配置在容納裝置中而橫越所述第一流體流動路徑。
對於交錯流的應用,在一個具體實例中,至少一個本發明的多孔膜或包含至少一個本發明之膜的濾器被配置在容納裝置中,所述容納裝置包含至少一個入口以及至少兩個出口,並至少限定出所述入口及第一出口之間的第一流體流動路徑,以及所述入口及第二出口之間的第二流體流動路徑,其中本發明的膜或包含至少一個本發明之膜的濾器橫越所述第一流體流動路徑,以提供過濾裝置或過濾組件。在一個例示性的具體實例中,所述過濾裝置包含交錯流過濾組件、容納裝置,其包含入口、包含濃縮出口的第一出口以及包含可滲透出口的第二出口,並限定出所述入口及第一出口之間的第一流體流動路徑,以及所述入口及第二出口之間的第二流體流動路徑,其中至少一個本發明的膜或包含至少一個本發明之膜的濾器被設置於橫越所述第一流體流動路徑。
所述過濾裝置或組件可為可滅菌的。可採用任何具有合適形狀且提供入口及一或多個出口的容納裝置。
所述容納裝置可由任何合適的堅固的無法滲透的材料組裝,包含任何無法滲透的熱塑性材料,其與將處理的流體相容。舉例來說,所述容納裝置可以由金屬組裝,諸如不銹鋼,或由聚合物組裝,例如:透明或半透明的聚合物,諸如丙烯酸、聚丙烯、聚苯乙烯或聚碳酸酯樹脂。

實施例1
此實施例例示根據本發明的一個具體實例之三元共聚物溶液之製備方法。
製備聚(磺酸丙烯醯胺基-2-甲基丙酯-共-(胺基丙基)甲基丙烯醯胺-共-N,N-二甲基丙烯醯胺)的10%三元共聚物溶液:將磺酸2-丙烯醯胺基-2-甲基丙酯(15.6 g,75 mM)、N-(3-胺基丙基)甲基丙烯醯胺鹽酸鹽(4.5 g,25 mM)及N,N-二甲基丙烯醯胺(10 g,100 mM)與DI水(260 g)以及8 M NaOH(17.1 g)混合,使混合物脫氣5分鐘,並添加過硫酸銨(0.690 g,3 mM)並混合。將反應混合物於60°C在氮氣下攪拌2小時,並使其冷卻至室溫以獲得三元共聚物溶液。
實施例2

製備聚(苯乙烯磺酸-共-(胺基丙基)甲基丙烯醯胺-共-N,N-二甲基丙烯醯胺)的10%溶液:將4-乙烯基苯磺酸鈉(15.46 g,75 mM)、N-(3-胺基丙基)甲基丙烯醯胺鹽酸鹽(4.5 g,25 mM)及N,N-二甲基丙烯醯胺(10 g,100 mM)與DI水(260 g)以及8 M NaOH(17.1 g)混合,使混合物脫氣5分鐘,且添加過硫酸銨(0.690 g,3 mM)。將反應混合物於60°C在氮氣下攪拌2小時,並使其冷卻至室溫以獲得三元共聚物溶液。
實施例3
此實施例例示根據本發明之另一具體實例的四元共聚物溶液之製備方法。
製備聚(磺酸丙烯醯胺基-2-甲基丙酯-共-(胺基丙基)甲基丙烯醯胺-共-N,N-二甲基丙烯醯胺-共-三級丁基丙烯醯胺),9.5%溶液:將磺酸2-丙烯醯胺基-2-甲基丙酯(10.45 g,50 mM)、N-(3-胺基丙基)甲基丙烯醯胺鹽酸鹽(2.24 g,12.5 mM)、N,N-二甲基丙烯醯胺(5 g,50 mM)及三級丁基丙烯醯胺[1.6 g,在IPA(9 g)中12.6 mM]與DI水(164.7 g)及8 M NaOH(10 g)混合,使混合物脫氣5分鐘,並添加過硫酸銨(0.431 g,1.9 mM)。將反應混合物於60°C在氮氣下攪拌2小時,並使其冷卻至室溫以獲得上述四元共聚物溶液。
實施例4

製備聚(磺酸丙烯醯胺基-2-甲基丙酯-共-(胺基丙基)甲基丙烯醯胺-共-N,N-二甲基丙烯醯胺-共-丙烯酸),10%溶液:將磺酸2-丙烯醯胺基-2-甲基丙酯(7.3 g,35.3 mM)、N-(3-胺基丙基)甲基丙烯醯胺鹽酸鹽(2.52 g,14.1 mM)、N,N-二甲基丙烯醯胺(7 g,70.6 mM)及丙烯酸[2.54 g,在IPA(9 g)中35.3 mM]在DI水(155 g)及8 M NaOH(10 g)中混合,使混合物脫氣5分鐘,並添加過硫酸銨(0.532 g,2.3 mM)。將反應混合物於60°C在氮氣下攪拌2小時,並使其冷卻至室溫以獲得上述四元共聚物溶液。
實施例5

製備聚(磺酸丙烯醯胺基-2-甲基丙酯-共-(胺基丙基)甲基丙烯醯胺-共-N,N-二甲基丙烯醯胺-共-苯乙烯磺酸),8%溶液:將4-乙烯基苯磺酸鈉(4.16 g,20 mM)、N-(3-胺基丙基)甲基丙烯醯胺鹽酸鹽(1.8 g,10 mM)、磺酸2-丙烯醯胺基-2-甲基丙酯(4.18 g,20 mM)及N,N-二甲基丙烯醯胺(4 g,40 mM)在含有8 M NaOH(17.1 g)的DI水(156.34 g)中混合,脫氣5分鐘,接著添加過硫酸銨(0.690 g,3 mM)。將反應混合物於60°C在氮氣下攪拌2小時,並使其冷卻至室溫以獲得上述共聚物溶液。
實施例6
此實施例例示根據本發明的一個具體實例之多孔膜的製備方法。
5%磺化聚合物塗層(100 g混合):將10%來自實施例1的磺化聚合物(50 g)與水(49 g)、聚乙二醇二環氧丙基醚(150 mg)及Triton X-100(850 mg)混合,並攪拌至溶解。使用NaOH溶液將溶液的pH調整至10,以獲得塗層溶液。藉由浸泡塗覆以塗層溶液塗覆孔直徑3 µm的HDPE膜。將經塗覆的膜夾在兩個移液管中間並從上往下刮過以提供HDPE膜上的薄塗層。使所述膜上的塗層在65°C下交聯一小時或在室溫下交聯隔夜。將得到的膜在冷水中淋溶(leach)24小時並使其乾燥。
實施例7
此實施例例示根據本發明的另一個具體實例之多孔膜的製備方法。
使磺酸2-丙烯醯胺基-2-甲基丙酯(104.5 g,504 mmol)與N,N-二甲基丙烯醯胺(50 g,504 mmol)、N-(3-胺基丙基)甲基丙烯醯胺HCl(22.4 g,126 mmol)與含有170 g異丙醇的DI水(1.524 kg)混合。以8 M NaOH(100 g)將混合物的pH值調整至9.6。使反應混合物以氮氣脫氣15分鐘,並添加過硫酸銨(3.88 g 1.5 mol%)。將混合物在60°C下攪拌2小時,並冷卻至室溫以獲得10%的聚合物溶液。
從上述10%聚合物溶液與0.85% Triton X100界面活性劑及0.45%二環氧化物交聯劑(聚乙二醇二環氧丙基醚)製備4%聚合物塗層溶液。調整所述溶液的pH值至10,並以上述溶液浸泡塗覆孔直徑3 µm的HDPE膜。將經塗覆的膜在65°C下熱交聯一小時,在DI水中淋溶隔夜,並在65°C下乾燥隔夜。
實施例8
此實施例例示根據本發明的一個具體實例的膜移除痕量金屬的能力。
基線:0.2 µm膜樣本以100 mL的IPA(10 mL/min)、200 mL的DIW、100 mL的5% HCl、200 mL DI水以及200 mL的IPA沖洗。收集100 mL的樣本,並以ICP-MS進行金屬分析。
金屬試驗(challenge):將得自實施例7的0.2 µm膜樣本以加料的IPA(1 ppb的金屬)在7 mL/min的流出速度下進行試驗。將一開始的10 mL流出物棄置,並將接下來的10 mL流出物收集至PFA瓶中。使用ICP-MS進行金屬分析。以n=2重複此實驗。

- 如請求項1-9中任一項之多孔介質,其中所述共聚物包含一種經聚合的單體(A)、一種經聚合的單體(B)及兩種不同的經聚合的單體(C)。
- 如請求項11之多孔介質,其中在一種經聚合的單體(C)中的X是SO3 H且在另一種經聚合的單體(C)中的X是COOH。
- 如請求項1-12中任一項之多孔介質,其為多孔膜。
- 如請求項1-12中任一項之多孔介質,其為珠粒或纖維。
- 一種用於製造如請求項1-14中任一項之多孔介質的方法,其包含: (i) 使單體(A)、(B)及(C)之混合物聚合,以獲得三元共聚物,以及 (ii) 將(i)中所獲得的三元共聚物與交聯劑塗覆在多孔撐體上,並原位交聯以獲得配置於所述多孔撐體上經交聯的三元共聚物塗層。
- 如請求項15的方法,其中所述交聯劑包含二或多個選自二環氧化物、二醛、二鹵化物、二酯、二磺醯氯(disulfonyl chloride)及二醯氯(diacyl chloride)的交聯官能基。
- 如請求項15或16之方法,其中該交聯劑是選自乙二醇二環氧丙基醚、丙二醇二環氧丙基醚、甘油基三環氧丙基醚、聚乙二醇二環氧丙基醚、二甲磺酸二醇酯、及二醇二甲磺醯氯(glycol dimesoyl chloride)、二甲苯磺酸酯及其任何組合。
