Description
本發明係有關一製備式(II)鹵化吡啶衍生物之方法,
其始自式(I)化合物,
經由式(IIIa)或(IIIb)中間物
其中該顯示於式(I)、(II)、(IIIa)與(IIIb)之結構元件具下列定義。本發明更有關此類鹵化吡啶衍生物與中間物。
式(II)鹵化吡啶衍生物在製藥及農化工業上具有重大的產業意義,此外,其亦為諸如製備可作為殺蟲劑之有效化合物的重要反應夥伴。
文獻上已描述吡啶於鋰鹼或鎂鹼(LDA、TMPLi、TMPMgCl.LiCl)存在下之金屬化反應,且必須在極低溫下進行(鋰為-78℃至-100℃,鎂為-78℃ 至-30℃),請見如Tetrahedron 2001(57),p.4059ff,Journal of the American Chemical Society 2010(132),p.2410ff and Organic Letters 2011(13),p.2306ff。以錯合物鎂鹼TMP2Mn.2MgCl2.4LiCl進行2,5-雙(經取代)吡啶之去質子化亦描述於Synlett 2015(26),p.1515ff,但僅進行氧化同元耦合。在鋅鹼存在下,一些經取代吡啶的位置4金屬化反應為習知,且描述於Angewandte Chemie 2007(46),p.7685ff or Organic Letters 2009(11),p.1837ff,但無2,5-雙(經取代)吡啶的位置6金屬化反應。
先前技術已描述的鹵化吡啶衍生物化學合成法常使用從產業觀點來看相當不經濟,及/或具有其他缺點的方法。
特別是在鋰鹼與鎂鹼的情況下,缺點為化學產率低、須在非常低的溫度下進行,以及由於彼等試劑的高反應性,去質子化反應中的空間和化學選擇性困難。有時,為進行進一步的選擇性反應,如所述Negishi交聯耦合反應,必須使用鋅鹽如氯化鋅進行金屬轉化反應,請見如Organic Letters 2008(10),p.2497ff。該製備法因而非常昂貴(形成許多鹽類),且不適於工業規格商用製程。
關於上述缺點,迫切需要一個簡化、工業上和經濟上可執行的製程,以製備鹵化吡啶衍生物,尤其是式(II)鹵化吡啶衍生物。此製程所得鹵化吡啶衍生物應較佳具高產率、高純度,且以較經濟方式進行。
發現到,令人驚訝地,在使用有機鋅鹼的製程中,可優勢製備式(II)鹵化吡啶衍生物。
據此,本發明提供用於製備式(II)化合物之方法
其中(構形1)Q與W各獨立地為鹵素,以及 Y為鹵素、CO2R1或NO2,其中R1為(C1-C6)-烷基或(C1-C6)-鹵烷基,其特徵為,在第一方法步驟a)中,式(I)化合物
其中W與Y各具有上述定義,係與具結構(NR3R4)-Zn-R2或(NR3R4)2-Zn之有機鋅鹼反應,其中R2為鹵素或-O-三甲基乙醯基,以及R3與R4共同形成-(CH2)4-、-(CH2)5-或-(CH2)2O(CH2)2-基團,其中彼等基團各可任意地經1、2、3或4個R5基取代,以及R5選自於由甲基、乙基、正丙基與異丙基組成之群組,以取得式(IIIa)或式(IIIb)化合物
其中W、Y與R2各具有上述定義,以及此式(IIIa)或(IIIb)化合物係於第二方法步驟b)中,與結構為Q-X之化合物反應,其中X為鹵素,且Q具有上述定義,以取得式(II)化合物。
化合物Q-X,如Q與X定義中所述,為鹵素間化合物,較佳為鹵素元素。
包含於本發明方法之式(I)、(II)、(IIIa)與(IIIb)中的Q、W、R1、R2、X與Y基之較佳與特佳定義,係於下文中闡明,下文更具體描述有機鋅鹼,因此特別指出該鹼的較佳構型。
(構形2)
Q與X具有相同定義,且各為碘或溴,W較佳為氟或氯,R2較佳為鹵素,尤其是氯、溴或碘,以及Y較佳為氯、溴、CO2R1或NO2,其中R1為(C1-C4)-烷基。
(構形3)
Q與X具有相同定義,且各特佳為碘,W特佳為氟,R2特佳為為氯,以及Y較佳為氯、溴、CO2R1或NO2,其中R1為甲基。
以上提供的基團定義和闡述皆可應用於最終產物與中間物,以及原料。彼等基團定義可根據需要彼此組合,即包括各較佳範圍間之組合。

優勢地,式(II)鹵化吡啶衍生物可利用本發明方法製備,其具高產率和高純度。由於鋅試劑具有極佳官能基耐受性,鋅鹼便極具吸引力。在化學計量之選擇性鹼存在下,吡啶的空間和化學選擇性金屬化作用是可能的,即使是在高溫下,而未發生芳炔消去反應或敏感官能基受攻擊。所形成之鋅化合物中間物隨後可以各種親電性試劑清除,如Organic Letters 2009(11),p.1837ff之實施例所述。彼等新穎之經取代吡啶衍生物之後可作為有價值的合成子進一步反應。
特別的是,金屬化反應之極佳與令人意外之高空間選擇性為其優勢,其發生於吡啶骨架之位置6,特別是在相對高溫下(諸如加熱至40至80℃,如60℃)。據此,若希望進一步改進空間選擇性,加熱特別具有優勢。舉例而言,以位置6與4之碘清除吡啶混合物可於室溫下形成,而在相對高溫下吡啶則於位置6被碘清除。

在此流程圖中,Q、W、R1、R2、X與Y具上述意義。括號中顯示的化合物為中間物(式IIIa或式IIIb),其可進一步反應得式(II)化合物。因此,本發明方法可分為步驟a)與b)二步,步驟a)為式(I)化合物轉換為相對應之中間物,且步驟b)進一步將中間物轉換為式(II)化合物。
在本發明內文中,術語鹵素(Hal),除非另有指明,包含選自於由氟、氯、溴與碘所組成群組之彼等元素。
術語「鹵化物」與本發明結合,係描述週期表上的鹵素與其他族元素之間的化合物,其可得鹵化物鹽類(離子化合物(鹽類),其由陰離子與陽離子組成,由於二元素間電負度差異大,且以靜電作用力維持),或共價鹵化物(其中電負度差異並不像前述離子化合物那麼大,但鍵結仍具電荷極性),取決於化學鍵性質。本發明特佳為鹵化物鹽類。
術語「三甲基乙醯基」在本發明中係描述具有經驗式(CH3)3CCO2H之新戊酸(X)去質子化基。
「O-三甲基乙醯基」相應地代表三甲基乙醯基基團鍵結係經由酸性基團上氧原子去質子化而得。
任意地經取代基團可為單或多取代,其中在多取代基情況下,該取代基可為相同或不同。
經一或多個鹵素原子(-Hal)取代之烷基為,例如,選自於三氟甲基(CF3)、二氟甲基(CHF2)、CF3CH2、ClCH2或CF3CCl2。
本文中之烷基,除非另有指明,為直鏈、支鏈或環狀飽和烴基。
C1-C12-烷基之定義包含本文所定義烷基之最廣範圍。特別是,此定義包含如甲基、乙基、正-、異丙基、正-、異-、第二-與第三丁基、正戊基、正己基、1,3-二甲基丁基、3,3二甲基丁基、正庚基、正壬基、正癸基、正十一烷基、正十二烷基。
第一方法步驟(步驟a)中,式(I)化合物轉換為式(IIIa)或(IIIb)化合物,係在結構(NR3R4)-Zn-R2或(NR3R4)2-Zn之有機鋅鹼存在下進行,其中(構形B-1)R2如上述定義(構形1)(因此為鹵素或-O-三甲基乙醯基),R3與R4共同形成-(CH2)4-、-(CH2)5-或-(CH2)2O(CH2)2-基團,其中彼等基團各任意地經1、2、3或4個R5基取代,以及R5選自於由甲基、乙基、正丙基與異丙基組成之群組。

本發明極佳之鹼類構形中,結構元件(NR3R4)為式(IV)四甲基哌啶(TMP)。
本發明之最佳有機鋅鹼據此特徵為鋅與TMP結合,尤其是以鹵化鋅形式,最佳為以氯化鋅形式。此類鹼具下列式(V)結構(構形B-4)(V) (TMP)xZnCl2-x,其中x為數字1或2。在彼等化合物中,較佳為具x=1(構形B-5)之式(VI)鹼:
在本發明方法之另一較佳實施例中,該有機金屬鹼係與鹼金屬鹵化物或鹼土金屬鹵化物結合存在。尤其是式(V)與(VI)之鹼。此類鹼金屬或鹼土金屬鹵化物特佳為氯化鋰與氯化鎂,特佳為氯化鋰。因此,本發明特佳之有機金屬鹼為TMP ZnCl.LiCl或(TMP)2 Zn.2LiCl(構形B-6)。最佳為TMP ZnCl.LiCl(VII:構形B-7)。
本發明式(I)、(II)和(IIIa)或(IIIb)化合物與鹼之組合列舉於下表1中,作為實例,彼等皆可用於本發明方法。由於在一些構形中,結構元件R2存在於本發明鹼與式(IIIa)化合物二者中,最嚴格的定義適用於每一情況下的R2。
較佳地,本發明方法使用之有機金屬鹼總量為0.5至5當量,較佳為0.8至2當量,更佳為1至1.5當量,特佳為1.0至1.2當量,以式(I)化合物為基準。在此方面,本發明方法之一優點為該有機金屬鹼可以實際上化學計量之量使用。
取決於結構元件(NR3R4)於所使用有機鋅鹼中存在一次或二次,式(IIIa)或式(IIIb)中間物係於步驟a)中形成。
式(IIIa)或(IIIb)化合物於第二步驟(步驟b)中轉換為式(II)化合物,其係於Q-X化合物存在下進行,其中Q與X各具有上述定義。由於Q與X二者皆為鹵素,該化合物為鹵素間化合物(interhalogen compound)。Q與X不一定為相同鹵素。舉例而言,Q可為碘或溴,且X可為氯、溴或碘。然而,較佳地,該化合物Q-X為鹵素元素,尤其是F2、Cl2、Br2或I2。特佳為I2或Br2,特佳為I2。
較佳地,本發明方法中化合物Q-X之總使用量為0.5至10.0當量,較佳為0.8至5當量,更佳為1至2.5當量,特佳為1.0至1.5當量,以式(I)化合物為基準。
在各情況下,本發明式(I)化合物轉換為式(IIIa)或(IIIb)化合物,以及進一步轉換為式(II)化合物,較佳為於有機溶劑存在下進行。原則上,可用之溶劑包括所有有機溶劑,其在所使用之反應條件下為惰性,其中待轉換之化合物具有適當溶解度。適用之溶劑尤其包括:四氫呋喃(THF)、1,4-二噁烷、乙醚、二甘醇二甲醚(diglyme)、甲基第三丁醚(MTBE)、第三-戊基甲醚(TAME)、2-甲基-THF、甲苯、二甲苯、均三甲苯、乙烯碳酸酯、丙烯碳酸酯、N,N-二甲基乙醯胺、N,N-二甲基甲醯胺(DMF)、N-甲基吡咯烷酮(NMP)、N-乙基-2-吡咯烷酮(NEP)、N-丁基-2-吡咯烷酮(NBP);N,N'-二甲基丙基烯脲(DMPU)、鹵化烴、芳烴,尤其是氯化烴如四氯乙烯、四氯乙烷、二氯丙烷、氯化甲烯、二氯丁烷、氯仿、四氯化碳、三氯乙烷、三氯乙烯、五氯乙烷、 二氟化苯、1,2-二氯乙烷、氯化苯、溴化苯、二氯化苯,尤其是1,2-二氯化苯、氯化甲苯、三氯化苯;4-甲氧基苯、氟化脂族與氟化芳族,如三氯三氟乙烷、三氟甲苯和4-氯三氟甲苯。亦可使用溶劑混合物,較佳為上述溶劑之混合物如四氫呋喃(THF)、1,4-二噁烷、乙醚、二甘醇二甲醚(diglyme)、甲基第三丁醚(MTBE)、第三戊基甲醚(TAME)、2-甲基四氫呋喃、甲苯、二甲苯均三甲苯、二甲基甲醯胺(DMF)。
較佳之溶劑為四氫呋喃、N,N-二甲基甲醯胺(DMF)、1,4-二噁烷、二甘醇二甲醚(diglyme)、甲基第三丁醚(MTBE)、第三-戊基甲醚(TAME)、2-甲基-THF、甲苯與4-甲氧基苯。
特佳之溶劑為THF與N,N-二甲基甲醯胺(DMF),特佳為THF。
溶劑亦可經除氣(無氧)。
步驟a)與b)兩步驟中較佳為使用相同溶劑。亦可能使用本發明之其他構形,其中步驟a)與b)使用不同溶劑,然而,在此情況下,溶劑同樣較佳為選自於上述溶劑,且各溶劑指定為較佳、更佳與最佳者,皆可應用於步驟a)或b)各流程。
方法步驟a)之轉換通常於溫度0℃至80℃間進行,較佳為介於10℃至75℃之間、介於20℃至70℃之間、介於30℃至70℃之間、介於40℃至68℃之間,最佳為介於50℃至65℃之間,如60℃。
方法步驟b)之轉換通常於溫度介於0℃至80℃之間進行,較佳為介於10℃至70℃之間、介於15℃至60℃之間、介於20℃至50℃之間、介於20℃至40℃之間,最佳為介於20℃至35℃之間,如室溫或25℃。
反應一般於標準壓力下進行,但亦可於升壓或減壓下進行。
可分離所需之式(II)化合物,例如,藉由在飽和氯化銨或硫代硫酸鈉溶液存在下,進行水處理,及/或之後進行層析法。彼等方法為本領域技術人員習知,亦包括由有機溶劑或溶劑混合物中結晶出。

本發明將以下列實施例更詳細地闡述,儘管彼等實施例不應理解為侷限本發明。
將溶解於四氫呋喃(2毫升)中之2-氯-5-氟吡啶(132毫克,1.0毫莫耳),於60℃氮氣下加入TMPZnCl.LiCl(1.31M之四氫呋喃溶液,0.84毫升,1.1毫莫耳),此反應溶液攪拌30分鐘。之後,於25℃加入碘(355毫克溶於4毫升四氫呋喃溶液),溶液繼續攪拌30分鐘。加入飽和氯化銨和硫代硫酸鈉溶液進行常規處理後,反應混合物係以乙酸乙酯萃取,合併之有機相以無水Na2SO4除水,並於膜幫浦真空中濃縮。經管柱層析純化後(乙酸乙酯/環己烷),得6-氯-3-氟-2-碘化吡啶(158毫克,62%),為黃色固體。HPLC-MS:log P=2.53;質譜(m/z):無離子化;1H-NMR(D6-DMSO):δ 7.80(m,1H),7.61(m,1H)。
將溶解於四氫呋喃(2毫升)中之甲基5-氟吡啶-2-羧酸酯(155毫克,1.0毫莫耳),於60℃氮氣下加入TMPZnCl.LiCl(1.31M之THF溶液,0.84毫升,1.1毫莫耳);此反應溶液攪拌30分鐘。之後,於25℃加入碘(355毫克溶於4毫升四氫呋喃溶液),溶液繼續攪拌30分鐘。進行常規處理後,以管柱層析法純化(乙酸乙酯/環己烷),得甲基5-氟-6-碘化吡啶-2-羧酸酯(197毫克,70%),為黃色固體。HPLC-MS:log P=1.85;質譜(m/z):281.9;1H-NMR(D6-DMSO):δ 8.10(m,1H),7.86(m,1H),3.89(s,3H)。
將溶於四氫呋喃(5毫升)之2-溴-5-氟化吡啶(880毫克,5.0毫莫耳),於60℃氮氣下加入TMPZnCl.LiCl(1.31M之四氫呋喃溶液,4.2毫升,5.5毫 莫耳);此反應溶液攪拌30分鐘。之後,於25℃加入碘(1.78克溶於4毫升四氫呋喃溶液),溶液繼續攪拌30分鐘。加入飽和氯化銨和硫代硫酸鈉溶液進行常規處理後,反應混合物係以乙酸乙酯萃取,合併之有機相以無水Na2SO4除水,並於膜幫浦真空中濃縮。經管柱層析純化後(乙酸乙酯/環己烷),得6-溴-3-氟-2-碘化吡啶(1.31克,87%),為黃色固體。HPLC-MS:log P=2.67;質譜(m/z):無離子化;1H-NMR(D6-DMSO):δ 7.72(m,2H)。
Claims (17)
- 一種製備式(II)化合物之方法
- 如申請專利範圍第1項之方法,其特徵為Q與X具有相同定義,且各為碘或溴, W為氟或氯,R 2為鹵素,尤其是氯、溴或碘,以及Y為氯、溴、CO 2R 1或NO 2,其中R 1為(C 1-C 4)-烷基。
- 如申請專利範圍第1與2項中任一項之方法,其特徵為R 3與R 4共同形成-(CH 2) 5-基團,其中彼等基團各可任意地經1、2、3或4個R 5基取代,以及R 5選自於由甲基與乙基組成之群組。
- 如申請專利範圍第1至3項中任一項之方法,其特徵為Q與X具有相同定義,且各為碘,W為氟,R 2為氯,以及Y為氯、溴、CO 2R 1或NO 2,其中R 1為甲基。
- 如申請專利範圍第1至4項中任一項之方法,其特徵為R 3與R 4共同形成-(CH 2) 5-基,其經4個甲基取代。
- 如申請專利範圍第1至5項中任一項之方法,其特徵為該有機鋅鹼為式(V)化合物(V) (TMP) xZnCl 2-x,其中x為數字1或2。
- 如申請專利範圍第1至6項中任一項之方法,其特徵為該有機鋅鹼與鹼金屬鹵化物或鹼土金屬鹵化物結合存在,較佳為氯化鋰或氯化鎂。
- 如申請專利範圍第1至7項中任一項之方法,其特徵為該有機鋅鹼之總使用量為0.5至5當量,其係以式(I)化合物為基準。
- 如申請專利範圍第1至8項中任一項之方法,其特徵為該化合物Q-X為鹵素元素,尤其是F 2、Cl 2、Br 2或I 2。
- 如申請專利範圍第1至9項中任一項之方法,其特徵為該化合物Q-X之總使用量為0.5至10.0當量,其係以式(I)化合物為基準。
- 如申請專利範圍第1至10項中任一項之方法,其特徵為於溶劑存在下進行,該溶劑選自於由四氫呋喃(THF)、1,4-二噁烷、乙醚、二甘醇二甲 醚(digly me)、甲基第三丁醚(MTBE)、第三戊基甲醚(TAME)、2-甲基四氫呋喃、甲苯、二甲苯、均三甲苯、乙烯碳酸酯、丙烯碳酸酯、N,N-二甲基乙醯胺、N,N-二甲基甲醯胺(DMF)、N-甲基吡咯烷酮(NMP)、N-乙基-2-吡咯烷酮(NEP)、N-丁基-2-吡咯烷酮(NBP);N,N'-二甲基丙烯脲(DMPU)、鹵化烴、芳烴、氯化烴、四氯乙烯、四氯乙烷、二氯丙烷、氯化甲烯、二氯丁烷、氯仿、四氯化碳、三氯乙烷、三氯乙烯、五氯乙烷、二氟化苯、1,2-二氯乙烷、氯化苯、溴化苯、二氯化苯、1,2-二氯化苯、氯化甲苯、三氯化苯;4-甲氧基苯、氟化脂族、氟化芳族、三氯三氟乙烷、三氟甲苯,以及4-氯三氟甲苯,或彼等溶劑之至少二者與另一溶劑之混合物組成之群組。
- 如申請專利範圍第11項之方法,其特徵為該溶劑為四氫呋喃或N,N-二甲基甲醯胺(DMF)。
