Claims (11)

1. Способ получения 4D термочувствительного гидрогеля на основе хитозана, включающий следующие стадии:
(1) при комнатной температуре взвешивание хитозана, растворение хитозана в растворе уксусной кислоты, перемешивание до полного растворения хитозана с получением раствора хитозана; использование 4D-биопринтера для печати раствора хитозана в термочувствительный гидрогель на основе хитозана с размером пор 50-100 мкм в соответствии с заданными параметрами печати, и при желании формование термочувствительного гидрогеля на основе хитозана; лиофилизация после экстракции растворителем с получением лиофилизированного хитозана;
(2) загрузка сверхчистой воды и β-глицерофосфата натрия в чайник с водяной баней при температуре 60-70°С; после растворения β-глицерофосфата натрия естественное охлаждение до комнатной температуры с получением водного раствора β- глицерофосфата натрия;
(3) приготовление водного раствора карбоксиметилхитозана со сверхчистой водой при комнатной температуре, добавление по каплям водного раствора β- глицерофосфата натрия, полученного на стадии (2), в водный раствор карбоксиметилхитозана и тщательное перемешивание с получением смеси; и
(4) сшивание лиофилизированного хитозана, полученного на стадии (1), со смесью, полученной на стадии (3), в течение 1-2 мин с получением 4D термочувствительного гидрогеля на основе хитозана с однородным размером пор.
2. Способ получения 4D термочувствительного гидрогеля на основе хитозана по п.1, в котором молярная концентрация раствора уксусной кислоты, полученного на стадии (1), составляет 0,2 моль/л, и концентрация раствора хитозана составляет от 2,2 мас.% до 6,7 мас.%.
3. Способ получения 4D термочувствительного гидрогеля на основе хитозана по п.1, в котором концентрация водного раствора β-глицерофосфата натрия составляет от 6 мас.% до 8 мас.%.
4. Способ получения 4D термочувствительного гидрогеля на основе хитозана по п.1, в котором концентрация водного раствора карбоксиметилхитозана, полученного на стадии (3), составляет от 2,2 мас.% до 6,7 мас.%.
5. Способ получения 4D термочувствительного гидрогеля на основе хитозана по п.1, в котором концентрации хитозана, карбоксиметилхитозана и β-глицерофосфата натрия в термочувствительном гидрогеле на основе хитозана 4D, полученном на стадии (4), составляют 10-30 мас.%, 10-30 мас.% и 60-80 мас.%, соответственно.
6. Способ получения 4D термочувствительного гидрогеля на основе хитозана по любому из пп.1-5, в котором термочувствительный гидрогель на основе хитозана 4D можно хранить в стерильной среде при 4-15°С в течение 6-12 месяцев.
7. Способ получения 4D термочувствительного гидрогеля на основе хитозана по любому из пп.1-5, в котором после захвата лимбальных стволовых клеток 4D термочувствительный гидрогель на основе хитозана наносится на поверхность обожженной щелочью роговицы и высвобождает лимбальные стволовые клетки для восстановления и лечения раны.
