Description
Foreliggende oppfinnelse vedrører nye glycerolderivater, en fremgangsmåte til fremstilling derav og legemidler på basis av disse glycerolderivater.
Glycerolderivatene som fremstilles ifølge oppfinnelsen er forbindelser med formelen
hvori en av restene R<1>, R<2>og R<3>er en gruppe U med formel OY<1>eller -X<1>-CO-(A<1>)n-Z<1>,
en annen enn gruppe V med formelen
OY<2>eller -X2-CO-(A<2>) P-Z<2>,
og den gjenværende en gruppe W med formel
-X3 T-(C2 - 6-alkylen)-N+R A"
hvori en av X<1>, X<2>og X<3>er oksygen eller en gruppe NQ<1>og de to andre er oksygen,
Y<1>er Cio-26-alkyl eller Cio- 2e-alkenyl,
Y<2>er Ci-e-alkyl, C2-e-alkenyl, C3- 6-cykloalkyl,
C5- 6-cykloalkenyl, fenyl, benzyl eller 2-tetrahydropyranyl,
A<1>og A<2>oksygen eller en gruppe NQ<2>,
n og p tallet 1 eller 0,
Z<1>Cg-25-alkyl eller C9-2s-alkenyl,
Z<2>Ci-5-alkyl, C2-g<->alkenyl, fenyl eller hvis A<2>ikke er

eller 6-leddet aromatisk gruppe bundet til et kvaternært nitro-gen, eventuelt med et ytterligere heterogenatom 0, S eller N, eventuelt med tilkondensert benzen og eventuelt monosubstituert med alkyl eller alkoksy med opptil 4 C-atomer, hydroksy, nitro, karbamoyl eller ureido, Y<3>og Y<4>har Ci-6-alkyl eller tilsammen C3-e-alkylen og Y' Ci - 6 ,
hvorunder, hvis R<1>er en gruppe OY<1>eller OY<2>og samtidig R<3>en gruppe W hvori X<3>betyr oksygen og T metylen eller karbonyl, inneholder R<2>gruppen 0C00 eller et substituert N-atom,
og deres hydrater.
De her anvendte uttrykk "alkyl" og "alkenyl" referer til rettkjedede eller forgrenede, mettede henholdsvis monoumettede rester såsom metyl, etyl, propyl, ispropyl, 2-propenyl, butyl, isobutyl, heksadecyl, heptadecyl, octadecyl og octadecenyl, spesielt metyl og octadecyl. Eksempler på C3- 6-cykloalkylrester er cyklopropyl og cykloheksyl, blant Cs - 6-cykloalkenylrestene 2-cyklopentenyl og spesielt 2-cykloheksenyl. C2-e-alkylengrupper kan være rettkjedede eller forgrenede, eksempler på dette er n-butylen, 2-metylpropylen og spesielt etylen og propylen. Eksempler på rester -N+ (Y3 , Y4 , Y<5>) er 1-metylpiperidinium og trimetylammonium. Eksempler på heterocykliske grupper N<+>R er oxazolium, isoxazolium, pyridinium, pyridazinium, kinolinium, isokinolinium og spesielt N-metylimidazolium og tiazolium.
Eksempler på anioner av sterke organiske eller uorganiske syrer er Ci - 4-alkylsulfonyloksy, fenylsulfonyloksy, tosyloksy, kamfer-10-sulfonyloksy, henholdsvis Cl-, Br~, J~, CICm, SCm~, PCm - - og NO3 .
Forbindelsen med formel I kan være hydratiserte. Hydra-tiseringen kan finne sted under fremstillingsprosessen eller etter hvert som følge av de hygroskopiske egenskapene til den første vannfrie forbindelse med formel I.
Forbindelsene med formel I inneholder minst et asymmetrisk C-atom og kan dermed foreligge som optisk aktive enantiomere, som diastereomere eller som blandinger derav, f.eks. som racemater.
Foretrukne forbindelser med formel I er de hvori R<1>er en gruppe U, R<2>er en gruppe V og R<3>er en gruppe W.
Spesielt foretrukne forbindelser med formel I er de hvori R<1>betyr oktadecanoyloksy, oktadecylkarbamoyloksy, oktadecyloksy, oktadecyloksyformamido, N-isopropyloktadecyloksyformamido eller l-isopropyl-3-oktadecylureido, og/eller R<2>acetamido, metoksyformamido, metoksy, metylkarbamoyloksy eller metoksykarbonyloksy og/eller R<3>[4-(trimetylammonio)-n-butyryloksy]klorid,[2-(1-metylpiperidino)etoksykarbonyloksy]klorid, [4-(3-tiazolio)-n- butoksy]bromid, [4-(trimetylammonio)-n-butoksy]bromid eller [4-(3-metylimidazolio)-n-butoksy]bromid.
Forbindelsene med formel I og deres hydrater kan fremstilles ved at man
a) omsetter en forbindelse med formel
hvori restene R4 , R<3>og R<6>har den samme betydning som
restene R<1>, R<2>henholdsvis R3 , hvorunder det imidlertid foreligger en avspaltbar gruppe A- i gruppen W istedet for resten
N+ R A" ,
med et amin med formel NR, eller
b) omsetter en forbindelse med formel
hvori restene R7 , R<8>og R<9>har den samme betydning som
restene R1 , R<2>henholdsvis R3 , hvorunder imidlertid en hydroksy-eller en aminogruppe foreligger i stedet for gruppene U og V,
med en syre med formelen
eller et reaktivt derivat derav, eller med et isocyanat med formelen eller et imidazolid med formelen eller c) omsetter en forbindelse med formelen
hvori R01 ,R02og R°<3>har den samme betydning som resten R1 ,
R<2>henholdsvis R<3>, hvorunder imidlertid en gruppe -N(Y<3>, Y<4>) i gruppen W istedet for gruppen -N* R A",
med et Ci -6-alkylhalogenid, hvorunder R<1>,R<2>,R<3>. -N+ R,A", U, V, W, Z1 , Z2, A<1>, A2 , Y3 , Y4 , n og p har den ovenfor angitte betydning.
Eksempler på avspaltbare grupper i forbindelsene med formel II er klor, brom, jod, mesyloksy, fenylsulfonyloksy og tosyloksy. Reaksjonen av en forbindelse II med et amin NR kan man utføre på i og for seg kjent måte, f.eks. ved en temperatur opp til reaksjonsblandingens tilbakeløpstemperatur, hensiktsmessig ved ca. 80°C, eventuelt i et løsningsmiddel såsom acetonitril, nitrometan eller et aromatisk hydrokarbon, f.eks. toluen eller benzen, eller i tetrahydrofuran.
Eksempler på reaktive derivater av syre med formel IV er syrebromider eller syreklorider og anhydrider. Omsetningen av en slik syre eller en av dens reaktive derivater med en forbindelse med formel III kan utføres på i og for seg kjent måte. Et syreklorid eller syrebromid kan omsettes i et løsningsmiddel i nærvær av en base med en temperatur fra ca. 0 til 80°C med forbindelsen III. Et anhydrid kan omsettes i nærvær av en katalysator såsom dimetylaminopyridin, hensiktsmessig i et løsningsmiddel, med forbindelsene III. Som løsningsmiddel kan man anvende halogenerte hydrokarboner såsom kloroform eller dikloretan, og som baser organiske baser såsom trietylamin, kinolin eller pyridin, eller uorganiske baser, såsom alkali-eller jordalkalimetallhydroksyder, -karbonater eller -bikarbo-nater, f.eks. Na2CO3 , KHCO3eller Ca2CO3 .
Omsetningen av en forbindelse med formel III med et isocyanat med formel V eller med tilsvarende imidazolid med formel VI kan utføres i et løsningsmiddel såsom kloroform, aceton eller dimetylformamid ved en temperatur mellom ca. 0 og 100°C, fortrinnsvis ved ca. 40-60°C, hensiktsmessig i nærvær av en katalysator såsom en Lewis-base, f.eks. trietylamin eller diisopropyletylamin. Reaksjonen kan eventuelt også utføres uten nærvær av et løsningsmiddel.
Omsetningen av et amin med formel VII med et Ci-6-alkylhalogenid kan man også utføre på i og for seg kjent måte, f.eks. i et løsningsmiddel såsom diklormetan eller kloroform ved en temperatur mellom romtemperatur og 50°C, fortrinnsvis ved romtemperatur. Som halogenid anvender man fortrinnsvis jodidet.
Forbindelsen med formel II kan fremstilles fra sådanne med formel IX eller X, og forbindelsene med formel III ut fra sådanne med formel VIII etter det følgende reaksjonsskjema:
I forbindelsen med formel VIII har restene R10 , R<20>ogR30 den samme betydning som restene R<1>, R<2>og R<3>, hvorunder imidlertid det i stedet for en av gruppene U eller V foreligger en eventuelt beskyttet hydroksy- eller aminogruppe eller en azidogruppe.
I forbindelsene med formel IX har restene R1 1 , R1 2 ogR13 den samme betydning som restene R<1>, R<2>og R3 , hvorunder det imidlertid i stedet for gruppen W foreligger en eventuelt beskyttet hydroksy- eller aminogruppe eller en azidogruppe.
I forbindelsen med formel X er en av restene R14 , R15 ogR16 en eventuelt beskyttet hydroksy- eller aminogruppe eller en azidogruppe, en annen er en gruppe U eller V, og den gjenværende er en gruppe W hvori det i stedet for -N<+>R A- foreligger en avspaltbar gruppe.
I forbindelsen med formel XI er en av restene R17 , R<1>8 og R<19>en eventuelt beskyttet hydroksy- eller aminogruppe eller en azidogruppe, en annen er en eventuelt beskyttet hydroksygruppe og den gjenværende er en gruppe U eller V.
Eksempler på beskyttede hydroksy- og aminogrupper er etergrupper såsom benzyloksy, trityloksy eller 2-tetrahydro-pyranyloksy, henholdsvis succinimid, ftalimid, benzyloksy-karbonylamino eller t-butoksykarbonylamino.
For å fremstille en forbindelse med formel II kan man omsette en forbindelse med formel IX hvori f.eks. R<13>er en hydroksy, med et halogenid med formel Hal-T-(C2-e-alkylen)-G, hvori G er en avspaltbar gruppe, Hal er et halogenatom og T har ovenfornevnte betydning i nærvær av en base såsom pyridin, eller med et isocyanat med formelen 0=C=N-(C2-e-alkylen)-G.
Alternativt kan man overføre en forbindelse med formel X, hvori f.eks. R<14>er hydroksy, i den tilsvarende forbindelse med formel II, hvori R<4>er en gruppe U eller V, hvilket kan foretas på samme måte som den ovenfor beskrevne omvandling av en forbindelse III i en forbindelse I. Analogt kan man overføre en forbindelse med formel VII, hvori f.eks. R<17>er hydroksy og R<19>en beskyttet hydroksy- eller aminogruppe, i den tilsvarende forbindelsen med formel IX hvori R<11>er en gruppe U eller V.
En azidogruppe eller beskyttet hydroksy- eller aminogruppe i en forbindelse med formel IX, f.eks. R13 , kan på i og for seg kjent måte overføres i den fri hydroksy- eller aminogruppe. En benzylgruppe kan avspaltes ved hydrogenolyse, f.eks. over palladium, en tritylgruppe ved hjelp av trifluoreddiksyre eller fortynnet saltsyre, og en tetrahydropyronylgruppe ved hjelp av fortynnet saltsyre. En azidogruppe kan overføres i aminogruppen med et komplekst hydrid såsom litiumaluminiumhydrid eller ved hjelp av H2/Pd-C. Analogt kan en azidogruppe i en forbindelse med formel VIII, X eller XI eller beskyttet hydroksy- eller aminogruppe overføres i den fri hydroksy- eller aminogruppe. På denne måte overføres en forbindelse med formel VIII hvori f.eks. R<10>er en beskyttet hydroksy- eller aminogruppe i den tilsvarende forbindelse med formel III hvori R<7>er hydroksy.
For fremstilling av en forbindelse med formel X, hvori f. eks. R<16>er gruppe W hvori det foreligger en avspaltbar gruppe i stedet for -NR<+>A-, kan man behandle den tilsvarende forbindelse med formel XI, hvori R<19>er hydroksy, på samme måte som lenger ovenfor beskrevet for overføringen av en forbindelse IX i en med formel II.
Overføringen av en forbindelse med formel X i en med formel VIII kan utføres på samme måte som overføringen av en forbindelse II i en forbindelse I.
Man kan fremstille en forbindelse med formel VII ut fra den tilsvarende forbindelse med IX, hvori restene R<11>, R<12>og R13 har den samme betydning som restene R<1>, R<2>og R3 , hvorunder det imidlertid istedet for gruppen W foreligger en hydroksy- eller aminogruppe. Således kan man omsette en forbindelse med formel IX, hvori f.eks. R<13>er hydroksy, i et løsningsmiddel såsom diklormetan med fosgen i toluen og deretter med en alkohol med formelen HO-(C2-e-alkylen)-N(Y3 , Y<4>), hvori Y<3>og Y<4>har den ovennevnte betydning, f.eks. med 1-(2-hydroksyetyl)-piperidin, til den tilsvarende forbindelse med formel VII, hvori resten X<3>T i gruppen W er en OCOO-gruppering.
Forbindelsen med formel II, hvori en av restene R4 , R<5>ogR<6>inneholder en aminogruppe X<1>, X<2>eller X<3>, kan man fremstille ut fra en forbindelse med formelen
hvori en av restene R41,R51 og R<61>er en gruppe N(H,Q), en annen en hydroksygruppe og den gjenværende f.eks. en gruppe 0-(C2-6-alkylen)-G' , hvori Q har den samme betydning som Q1 , Q<2>eller Q3 , og G' er en beskyttet hydroksygruppe såsom trityloksy eller benzyloksy. Fremstillingen av forbindelsene med XII og deres overføring i II kan utføres på i og for seg kjent måte, f.eks. som beskrevet i eksemplene 5A. og 7A.
Forbindelsene med formelene II, III og VIII er nye. Som sådanne er forbindelsene med formel III og VIII bestanddel av oppfinnelsen.
Forbindelsen med formel I og deres hydrater hemmer blod-plateaktiveringsfaktoren (PAF) og kan derfor anvendes ved bekjempelse henholdsvis forebygning av sykdommer såsom trombose, apopleksi, hjerteinfarkt, angina pectoris, høyt blodtrykk og bronkeal astma forårsaket av allergi og videre som betennelses- hemmer og antirevmatika. Videre har forbindelsen med formel I og deres hydrater en hemmende virkning på veksten av tumorceller og kan derfor anvendes som antitumormidler.
Hemningsvirkningen på PAF kan påvises som følger:
Blodplaterikt plasma (PRP) fremstilles ved sentrifugering av 1/10 volumer 90 mM trinatriumcitratholdig kaninblod. Aggregeringen av blodplatene måles ved 37°C under røring ved hjelp av et aggregometer. To minutter etter tilsetning av forsøkssubstansen til PRP utløses blodplateaggregeringen ved en submaksimal dose av PAF (4 nM). ICso-verdien (mM) som er angitt i den etterfølgende tabell tilsvarer den konsentrasjon av forsøkssubstansen som nedsetter aggregeringen av blodplaten som er utløst med PAF til halvparten.
Som nevnt innledningsvis er legemidler som inneholder en forbindelse med formel I eller et hydrat derav likeledes gjen-stand for foreliggende oppfinnelse, videre også en fremgangsmåte ved fremstilling av slike legemidler som erkarakterisert vedat man bringer en eller flere forbindelser med formel I eller et hydrat derav og om ønsket et eller flere andre terapeutiske verdifulle stoffer i en galenisk administreringsform.
Legemidlene kan gis enteralt, f.eks. oralt i form av
tabletter, lakktabletter, drageer, hård- og mykgelatinkapsler, løsninger, emulsjoner eller suspensjoner eller rektalt, f.eks. i form av suppositorier, eller som spray. Administreringen kan imidlertid også skje parenteralt, f.eks. i form av injeksjons-løsninger.
For fremstilling av tabletter, lakktabletter, drageer og hårdgelatinkapsler kan virkestoffet blandes med farmasøytiske inerter, uorganiske eller organiske fyllstoffer. Som sådanne fyllstoffer kan man for tabletter, drageer og hårdgelatinkapsler f.eks. anvende laktose, maisstivelse eller derivater derav, talkum, stearinsyre eller dens salter. Egnede fyllstoffer for mykgelatinkapsler er f.eks. vegetabilske oljer, vokser, fett, halvfaste og flytende polyoler; avhengig av virkestoffets beskaffenhet er fyllstoffene imidlertid overhodet ikke nødvendig ved mykgelatinkapsler. For fremstilling av løsninger og sirups er egnede fyllstoffer f.eks. vann, polyoler, sakkarose, invert sukker og glukose, for injeksjonsløsninger egner f.eks. vann, alkoholer, polyoler, glyceroler og vegetabilske oljer seg, og for suppositorier egner f.eks. naturlige eller herdede oljer, vokser, fett og halvtflytende eller flytende polyoler seg.
De farmasøytiske preparater kan dertil også inneholde konserveringsmidler, løsningsformidlere, stabiliseringsmidler, fuktemidler, emulgeringsmidler, søtningsmidler, farvemidler, aromatiseringsmidler, salter for endring av det osmotiske trykk, buffere, overtrekksmidler eller antioksydanter.
Doseringen av virkestoffer kan variere innenfor vide grenser og må naturligvis i hvert enkelt tilfelle tilpasses de indivi-duelle forhold. Generelt skulle en dose på ca. 0,1 til 20 mg/kg, fortrinnsvis på ca. 0,5 til 4 mg/kg pr. dag være passende ved oral administrering til voksne, hvorunder imidlertid også den nettopp angitte øvre grense kan overskrides når dette skulle vise seg nødvendig.
Eksempel 1
A. Fremstilling av utgangsmaterialet
a) 15,35 g (RS)-l-O-benzyl-3-O-tritylglycerol (36,15 mM) (Heiv. Chim. Acta 65, 1982, 1059) ble oppløst i 75 ml kloroform. 10 Ml
pyridin ble tilsatt etterfulgt av 10,5 g tosylklorid. Etter 24 timer ved romtemperatur ble kloroformet destillert av. Resten ble opptatt i 50 ml pyridin. 10 Ml H20 og deretter 10 g KHC03ble tilsatt. Etter avdestillering av løsningsmidlet ble resten tatt opp i toluen, den faste del ble skilt fra, så ble den organiske fase rystet med H20, tørket og inndampet. Produktet krystalliserer fra smeiten ved avkjøling, utbytte 95%, smeltepunkt 98-100°C.
b) Det erholdte (RS)-1-0-benzyl-2-0-tosyl-3-0-tritylglycerol ble under oppvarming overført i (RS)-l-0-benzyl-2-deoksy-2-azido-3-O-tritylglycerol ved hjelp av natriumazid i tørt DMF. c) En løsning av det erholdte azid i tørr dietyleter ble ved hjelp av en suspensjon av LiAlH4i tørr dietyleter redusert til (RS)-l-0-benzyl-2-deoksy-2-amino-3-0-tritylglycerol, smeltepunkt 67-69° C. d) En løsning av det erholdte amin i dioksan ble overført med fortynnet saltsyre i (RS)-l-0-benzyl-2-deoksy-2-aminoglycerol-hydroklorid, smeltepunkt 148-149°C. e) En vandig løsning av det erholdte hydroklorid gjøres alkalisk med NaOH-løsning, det frigjorte amin tas opp i metylen-klorid og løsningsmidlet fjernes. f) En løsning av 1,6 g av det erholdte (RS)-l-0-benzyl-2-deoksy-2-aminoglycerol (8,8 mM) i 10 ml diklormetan ble tilsatt 2 g K2CO3i 3 ml H20. Så ble 2 ml eddiksyreanhydrid dryppet til under røring. Etter 1 time ble reaksjonsblandingen opparbeidet. For rensing ble forbindelsen kromatografert over kiselgel med eter/metanol (9:1). Man fikk 1,4 g (RS)-l-0-benzyl-2-deoksy-2-acetamidoglycerol i væskeform (81% av teoretisk).
g) En løsning av 1,4 g (RS)-l-0-benzyl-2-deoksy-2-acetamido-glycerol i 10 ml kloroform og 1,5 ml pyridin ble tildryppet 2,1 g
stearoylklorid i 5 ml kloroform. Etter 1 time ved romtemperatur ble blandingen opparbeidet. For rensing ble forbindelsen kromatografert over kiselgel med diklormetan/eter (1:1). Etter
krystallisering fra n-heksan erholdtes 2,4 g hvite krystaller (78% av teoretisk), smeltepunkt 65-66°C.
h) En løsning av 1,5 g av det erholdte (RS)-1-0-stearoyl-2-deoksy-2-acetamido-3-0-benzylglycerol i 20 ml THF ble hydrogenert
i nærvær av 0,5 g 10%ig Pd-karbon under lett H2-overtrykk. (RS)-1- 0-stearoyl-2-deoksy-2-acetamidoglycerol erholdtes i kvantitativt utbytte, smeltepunkt 89-90°C (fra n-heksan).
i) Til en løsning av 1,1 g (RS)-l-0-stearoyl-2-deoksy-2-acetamidoglycerol (2,75 mM) i 20 ml kloroform og 0,5 ml trietylamin dryppet man i isbad 0,4 g 4-klorsmørsyreklorid (2,8 mM) i 5 ml kloroform under utelukkelse av fuktighet. Reaksjonsblandingen ble rørt 2 timer. Etter opparbeiding ble (RS)-1-O-stearoyl-2- deoksy-2-acetamido-3-0 -(4-klorbutyryl)glycerol krystallisert fra n-heksan. Man fikk 1,39 g hvite krystaller (utbytte: kvantitativt). Smeltepunkt 51-52°C.
B. Fremstilling av produktet
0,45 g (RS)-l-0-stearoyl-2-deoksy-2-acetamido-3-0 -(4-klorbutyryl)glycerol ble blandet med 2 ml av en 20%ig løsning av trimetylamin i acetonitril og brakt til omsetning i bomberør ved 80°C i 24 timer. Som rensing ble forbindelsen kromatografert på kiselgel med kloroform/metanol (7:3). Man fikk 0,22 g [3-[[(RS)~2-acetamido-3-[oktadecanoyloksy] propoksy]karbonyl]propyl]-trimetylammoniumklorid (utbytte: 44% av teoretisk). Smeltepunkt 205° C.
Eksempel 2
A. Fremstilling av utgangsmaterialet
a) En løsning av 450 mg (RS)-l-0-benzyl-2-deoksy-2-amino-glycerol-hydroklorid i metanol ble blandet med en løsning av KOH
i vann. Metanolen ble avdestillert, resten ble tilsatt vann og diklormetan og blandingen ble blandet med metylklorformiat. Man fikk 470 mg (95%) (RS)-l-0-benzyl-2-deoksy-2-(1-metoksyformamido) glycerol som olje.
b) 0,45 g (RS)-l-0-benzyl-2-deoksy-2-(1-metoksyformamido)-glycerol (1,88 mM) ble blandet med 0,6 g octadecylisocyanat og løsningen oppvarmet 1 time ved 90°C. Den ble kromatografert på kiselgel med en blanding av toluen og eddikester (4:1). Etter krystallisering fra n-heksan fikk man 0,65 g (RS)-l-O-benzyl-2-deoksy-2-(1-metoksyformamido) -3-0-(oktadecylkarbamoyl)glycerol. Smeltepunkt 65-67°C.
c) 4,9 g (RS)-l-0-benzyl-2-deoksy-2-(1-metoksyformamido)-3-0-(oktadecylkarbamoyl)glycerol oppløst i 75 ml THF ble hydrogenert
i nærvær av 1 g 10%ig Pd-karbon ved svakt H2-overtrykk. Man fikk 4,05 g (RS)-2-deoksy-2-(1-metoksyformamido) -1-O-oktadecyl-karbamoylglycerol, smeltepunkt 86°C (fra n-heksan).
d) Til en løsning av 2 g (RS)-2-deoksy-2-(1-metoksyformamido)
-1-O-oktadecylkarbamoylglycerol (4,5 mM) i 20 ml kloroform og
0,7 ml trietylamin i isbad dryppet man 0,75 ml 4-klorsmørsyre-klorid (6,68 mM) i 5 ml kloroform under fuktighetsbeskyttelse. Reaksjonsblandingen ble rørt 2 timer ved romtemperatur. Etter opparbeiding ble reaksjonsproduktet filtrert på kiselgel med diklormetan/eter (1:1). Man fikk 2,3 g (RS)-1-0-(4-klorbutyryl)-2-deoksy-2-(1-metoksyformamido) -3-0-(oktadecylkarbamoyl)glycerol etter krystallisering fra n-heksan, smeltepunkt 68-70°C.
B. Fremstilling av produktet
Analogt eksempel IB fremstiltes ut fra 0,4 g av det under A.d) erholdte klorid 0,31 g (70%) [3-[[ (RS)-2-(1-metoksyformamido) -3-(oktadecylkarbamoyloksy)propoksy]karbonyl]propyl]-trimetylammoniumklorid, smeltepunkt 200°C (spaltn.).

Eksempel 3
A. Fremstilling av utgangsmaterialet
En løsning av 500 mg (1,4 mMol) (RS)-2-0-metyl-l-0-okta-decylglycerol i 5 ml diklormetan ble ved 0° C blandet med 2 ml
(4 mMol) av en 20%ig løsning av fosgen i toluen. Så rørte man 3 timer uten kjøling. Overskuddet av fosgen og diklormetan ble avdampet. Så omsattes en løsning av 500 mg (3,9 mMol) l-(2-hydroksyetyl)-piperidin i 10 ml diklormetan. Etter 2 timers røring ble løsningsmidlet fordampet. Resten ble kromatografert på kiselgel. Ved eluering med toluen-etylacetat (1:1) og
deretter etylacetat fikk man (RS)-2-0-metyl-l-O-oktadecyl -3-0-[[2-(1-piperidino)etoksy]karbonyl]glycerol, smeltepunkt 34°C.
B. Fremstilling av produktet
En løsning av 100 mg (0,195 mMol) av det under 3A. erholdte produkt i 5 ml diklormetan ble blandet med 100 mg (0,7 mMol) metyljodid og fikk stå 3 dager ved romtemperatur. Løsningsmidlet ble avdampet og resten perkolert med metanol-diklormetan (4:1) over kloridholdig anionbytterharpiks. Løsningsmidlet ble avdampet og resten ble kromatografert på kiselgel. Eluering med kloroform-metanol-vann (60:35:5) ga 1-[2-[[[(RS)-2-metoksy -3-(oktadecyloksy)propoksy]karbonyl]oksy]etyl] -1-metylpiperidinium-klorid, smeltepunkt 59°C.
Eksempel 4
A. Fremstilling av utgangsforbindelsen
a) En løsning av 0,9 g (RS)-l-deoksy-l-amino-3-O-benzylglycerol (5 mM) i 20 ml diklormetan ble tilsatt en vandig KOH-løsning.
Blandingen ble tildryppet en løsning av 1,7 g oktadecylklorformiat i 10 ml diklormetan under røring. Etter 1 times røring ved romtemperatur opparbeidet man, og den erholdte forbindelse ble kromatografert på kiselgel med n-heksan/eter (1:1). Etter krystallisering fra n-heksan fikk man 1,6 g krystaller, smeltepunkt 58-69° C.
b) 2 g (RS)-l-0-benzyl-3-deoksy -3-[1-(oktadecyloksy)formamido] glycerol ble oppvarmet med 2 ml metylisocyanat og 0,5 ml
trietylamin i 2 timer ved 40°C. Etter fjerning av reagensoverskuddet ble produktet omkrystallisert fra n-heksan, og derved fikk man 2 g (89,3%) (RS)-l-O-benzyl-3-deoksy -3-[1-(oktadecyloksy ) f ormamido] -2-0-metylkarbamoylglycerol, smeltepunkt 95-96 , 5° C. c) Analogt eksempel lA.h) og i) ble (RS)-1-0-(4-klorbutyryl) - 3-deoksy-3-[1-(oktadecyloksy)formamido] -2-O-metylkarbamoyl-glycerol fremstilt gjennom (RS)-3-deoksy-3-[1-(oktadecyloksy)-formamido] -2-0-metylkarbamoylglycerol.
B. Fremstilling av produktet
Analogt eksempel IB. ble [3-[[(RS)-2-metylkarbamoyloksy-3-[1-(oktadecyloksy)formamido]propoksy]karbonyl]propyl]trimetyl-ammoniumklorid fremstilt, smeltepunkt > 200°C.
Eksempel 5
A. Fremstilling av utgangsforbindelsen
a) Til 30 ml 1,4-butandiol og 0,2 ml BF3 . Et2O dryppet man under isbadkjøling 5 ml epiklorhydrin. Man lot reagere natten
over ved romtemperatur. Overskuddet av butandiol ble først avdestillert, deretter 10 g (85%) RS-l-klor-l-deoksy-3-O- (4-hydroksybutyl)glycerol som farveløs væske, kokepunkt 110°C/0,1 mm Hg; IR (cm"<1>): 3360 (OH); 1124 (eter); 1058 (alkohol-II bånd).
b) Diolet fra a) ble omsatt med tritylklorid i pyridin. Man fikk RS-l-klor-l-deoksy-3-O-[4-(trityloksy)butyl]glycerol som
viskøs væske, IR (cm"<1>): 3554 (OH); 1596 og 1490 (aromat); 1120 (eter); 1072 og 1032 (alkohol); 764, 746, 706 (monosubst. benzen).
c) For fremstilling av 1,2-epoxy-3-[4-(trityloksy)butoksy]-propan ble 4,85 g av produktet fra b) oppløst i 20 ml THF og så
omsatt med 2 g kalium-t-butylat under tilbakeløp. Man fikk produktet i kvantitativt utbytte, IR (cm"<1>): 1596 og 1490 (aromat); 1090 og 1073 (eter); 764, 747og 706 (monosubst. benzen).
d) Omsetningen av epoksyder fra c) med isopropylamin i trykk-kolbe ved 70° C i 8 timer ga (RS) -3-deoksy-3- (isopropylamino-) -1-0-[4-(trityloksy)butyl]glycerol i kvantitativt utbytte, IR (cm-<1>): 3380 og 3301 (OH og NH); 1597 og 1490 (aromat); 1123 og 1074 (eter); 764, 746 og 706 (monosubst. benzen). e) 2,4 g av aminet fra d) og 2 g oktadecylklorformiat i 20 ml CH2 CI2 ble rørt ved romtemperatur i 1 time i nærvær av 2 ml 30%ig kalilut. Produktet ble kromatografert på kiselgel med eter-kloroform-pyridin (49,5:49,5:1). Man fikk 3,1 g (78%) farveløs væske, IR (cm-<1>): 3421 (OH); 1696, 1669 og 1489 (aromat); 1206 (ester); 1124 og 1074 (eter); 774, 764, 745 og 705 (monosubst. benzen). f) Omsetningen av produktet fra e) med metylisocyanat ga (RS)-1-deoksy-l-[N-isopropyl-1-(oktadecyloksy)formamido]-2-0-(metyl-karbamoyl)-3-0-[4-(trityloksy)butyl]glycerol som farveløs væske. g) Produktet fra f) ble omsatt med vandig HC1 i dioksan ved 95°C. Kromatografi på kiselgel med EtOAc ga (RS)-1-deoksy-l-[N-isopropyl-1-(oktadecyloksy)formamido]-3-0-(4-hydroksybutyl)-2-0(metylkarbamoyl)glycerol som farveløs væske, IR (cm-<1>): 3353 og 3058 (NH og OH); 1702 (karbamat); 1539 (amid); 1255 (ester), 1120, 1095 og 1071 (alkohol og eter). h) Produktet fra g) ble bromert med trifenylfosfindibromid i nærvær av Et3N (støkiometrisk mengde). Fra 1,2 g utgangs-forbindelse fikk man etter kromatografi på kiselgel med toluen-EtOac (1:1) 0,9 g (67%) bromid som farveløs væske, IR (cm"<1>): 3356 (NH); 1702 (karbamat); 1530 (amid); 1251 (NH-CO); 1132 (eter).
B. Fremstill ing av produktet
0,3 g (RS)-1-0-(4-brombutyl)-3-deoksy-3-[N-isopropyl-3-(oktadecyloksy)formamido]-2-0-(metylkarbamoyl)glycerol ble blandet med 0,3 ml tiazol og reaksjonsblandingen holdt 5 timer ved 80°C. Etter avdestillering av reagensoverskuddet ble forbindelsen renset på kiselgel med CHCI3-MeOH (7:3). Man fikk 0,17 g (50%) 3-[4-[ (RS)-3-[N-isopropyl-1-(oktadecyloksy)formamido] -2-[(metylkarbamoyl)oksy]propoksy]butyl]tiazoliumbromid, MS: m/e = 626 (M<+>av kationet).
Eksempel 6
0,4 g av det samme utgangsmaterialet som i eksempel 5 ble oppløst i 3 ml THF og blandet med 2 ml av en 20%ig (CH3)3N-løsning i THF. Man lot reagere 2 timer i trykkolbe ved 80°C. For å rense filtrerte man over kiselgel med CHCI3-MeOH (1:1). Man fikk 0,42 g (96%) [4-[(RS)-3-[N-isopropyl(1-oktadecyloksy)-formamido]-2-[metylkarbamoyl)oksy]propoksy]butyl]trimety1-ammoniumbromid, MS: m/e = 600 (M<+>av kationet).
Eksempel 7
A. Fremstilling av utganqsforbindelsen
a) Analogt eksempel 5A. a) fikk man ut fra epiklorhydrin og 4-benzyloksy-l-butanol (RS)-1-0-[4-(benzyloksy)butyl]-3-klor-3-deoksyglycerin som farveløs væske. b) 15 g av produktet fra a) ble tatt opp i 20 ml isopropylamin og behandlet 6 timer i trykkolbe ved 80°C. Så ble isopropylamin-overskuddet avdestillert. Det dannede hydroklorid ble felt med eter og kastet. Man fikk (RS)-1-0-[4-(benzyloksy)butyl]-3-deoksy-3-(isopropylamino)glycerol, IR (cm-<1>): 3302 (OH og NH) ; 1496 (aromat); 1103 (eter og alkohol-II bånd); 737 og 698 (monosubst. benzen). c) En organisk fase bestående av 20 ml CH2CI2 , 5,9 g av produktet fra b) og 6,65 g oktadecylkarbamidsyreklorid ble rørt 2 timer ved 5 ml 30%ig kalilut. Etter kromatografi på kiselgel med CHCI3-EtOAc-Toluen (1:1:1) fikk man 7,9 g (RS)-3-0-[4-(benzyl oksy)butyl]-l-deoksy-l-(1-isopropy1-3-oktadecylureido)-glycerol, MS (m/e): 72 (100%; = NHCH(CH3)2; 296 (2%; 0=CNH-Ci8 H3 7) . d) Til en løsning av 3 g av produktet fra c) i 5 ml CH2Cl2og 0,5 ml Et3N satte man 3 ml metylisocyanat. Man lot reagere i 5 timer ved 40°C. Produktet ble renset på kiselgel med toluen-EtOAc (1:1). Man fikk 2,5 g (76%) av en voksaktig forbindelse, IR (cm"<1>): 3340 (NH); 1707 (karbamat); 1634 (urinstoff); 1541 (amid-II bånd); 1101 (eter); 735, 697 (monosubst. benzen). e) Katalytisk hydrogenering over 10%ig Pd-C i THF av produktet fra d) ga (RS)-1-deoksy-3-0-(4-hydroksybutyl)-1-(l-isopropyl-3-oktadecylureido)-2-0-(metylkarbamoyl)glycerol i kvantitivt utbytte MS (m/e): 483 (100%; M-CHsNHCOO); 558 (25%; M+H<+>). f) Bromeringen av produktet fra e) ble utført analogt eksempel 5A. h). IR (cm-<1>): 3335 (NH); 2916 og 2849 (alif. CH); 1649
(karbamat); 1623 (urinstoff); 1556 og 1525 (amid-II bånd og CO-NH åpen).
B. Fremstilling av produktet
Analogt eksempel 5B. fikk man ut fra 0,3 g (RS)-3-0-(4-brombutyl)-1-deoksy-l-(l-isopropyl-3-oktadecylureido)-2-0-(metylkarbamoyl)glycerol og tiazol 0,17 g (50%) 3-[4-[(RS)-3-(1-isopropyl-3-oktadecylureido)-2-[(metylkarbamoyl)oksy]propoksy]-butyl]tiazoliumbromid, MS (m/e): 625 (M<+>av kationet).
Eksempel 8
0,2 g av det samme utgangsmaterialet som i eksempel 7 ble tatt opp i 0,5 ml N-metylimidazol. Man lot reagere 30 minutter ved 80°C. Etter avdestillering av reagensoverskuddet ble forbindelsen krystallisert fra metanol-eter. Man fikk 0,2 g (88%) l-[4-[(RS)-3-(l-isopropyl-3-oktadecylureido)-2-[(metyl-karbamoyl )oksy]propoksy]butyl]-3-metylimidazoliumbromid, MS (m/e): 622 (M<+>av kationet).
Eksempel 9
A. Fremstilling av utgangsforbindelsen
a) En blanding av 6 g (20 mM) (RS)-1-0-(4-benzyloksybutyl)-3-deoksy-3-isopropylaminoglycerol og 6,66 g oktadecylklorformiat i
50 ml CH2Cl2ble rørt 2 timer med 20 ml 30%ig kalilut. Etter isolering av produktet fra den organiske fase og kromatografi på kiselgel med toluen-eddikester (4:1) fikk man 11 g (91,5%) (RS)-1-0-(4-benzyloksybutyl)-3-deoksy-3-(N-isopropyl-l-oktadecyloksyformamido)glycerol, IR (cm-<1>): 3431 (OH), 1696 og 1670 (karbamat), 1119 (eter og alkohol-II bånd); 734 og 697 (monosubst. benzen). b) 4,5 g av produktet fra a) ble forestret med metylklorformiat i nærvær av pyridin til 3,8 g (80%) (RS)-1-0-(4-benzyloksybutyl)-3-deoksy-2-0-metoksykarbonyl-3-(N-isopropyl-1-oktadecyloksyformamido)glycerol, IR (cm"<1>): 1751 (karbonat C=0), 1701 (karbamat C=0), 1266 (ester), 1101 (eter); 734 og 697 (monosubst.
benzen). c) 3,65 g av produktet fra b) ble hydrogenert over 10%ig Pd-C i THF til 3,1 g (94%) (RS)-l-deoksy-3-0-(4-hydroksybutyl)-1-(N- isopropy1-1-oktadecyloksyformamido)-2-O-metoksykarbonylglycerol, smeltepunkt 28°C. d) Analogt eksempel 5A.h ble 2,9 g av produktet fra c) bromert til 2,5 g (77,5%) (RS)-1-0-(4-brombutyl)-3-deoksy-3-(N-isopropy1-1-oktadecyloksyformamido)-2-0-metoksykarbonylglycerol, IR (cm-<1>): 1751 (karbonat CO), 1700 (karbamat CO), 1266 (ester), OH-bånd forsvunnet.
B. Fremstilling av produktet
Analogt eksempel 5B ble 1,5 g av produktet fra A.d omsatt med 1 ml tiazol til 1,45 g (85%) 3-[4-[ (RS)-3-(N-isopropyl-1-oktadecyloksyformamido)-2-(metoksykarbonyloksy)propoksy]butyl]-tiazoliumbromid, smeltepunkt 124-125°C (fra en blanding av 2 ml aceton og 30 ml eter) .
Eksempel A

Claims (6)
1. Fremgangsmåte ved fremstilling av glycerolderivater med formel
hvori en av restene R<1>, R<2>og R<3>er en gruppe U med formel OY<1>eller -X1 -CO- (A1 ) „ -Z1 ,
en annen en gruppe V med formel OY<2>eller -X2-CO-(A2)P-Z2 ,
og den gjenværende en gruppe W med formel -X3 T- (Cz - e -alkylen) -N+ R A" ,
hvorunder en av X<1>, X<2>og X<3>er oksygen eller en gruppe NQ<1>og de to andre oksygen,
Y<1>er Cio-26-alkyl eller Cio-2s-alkenyl,
Y<2>er Ci - 6-alkyl, C2- 6-alkenyl, C3-e-cykloalkyl,
Cs -e-cykloalkenyl, fenyl, benzyl eller 2-tetrahydro pyranyl,
A<1>og A<2>betyr oksygen eller en gruppe NQ2 ,
n og p tallet 1 eller 0,
Z<1>C9-2s-alkyl eller Cg - 2 3 -alkenyl,
Z<2>Ci-3-alkyl, C2-s-alkenyl, fenyl eller, hvis A<2>ikke
er oksygen, også hydrogen,
T karbonyl, COO eller CONQ<3>, eller, hvis X<3>er oksygen,
også metylen,
Q1 , Q<2>og Q<3>hydrogen, Ci-4-alkyl, C3- 6-cykloalkyl eller fenyl,
A- anionet av en sterk syre, -N+ R en gruppe -N4 (Y3 , Y4 , Ys ) eller, hvis minst av X1 , X2 , X3 , A<1>, A<2>og T inneholder et substituert N-atom, også en 5- eller 6-leddet aromatisk gruppe bundet til det kvaternære hydrogen, eventuelt med et ytterligere heterogenatom 0, S eller N, eventuelt med tilkondensert benzen og eventuelt monosubstituert med alkyl eller alkoksy med opp til 4 C-atomer, hydroksy, nitro, karbamoyl eller ureido, Y<3>og Y<4>Ci-6-alkyl eller tilsammen er C3- 6-alkylen, og Y<5>Ci-e-alkyl,
hvorunder, hvis samtidig R<1>er en gruppe OY<1>eller OY<2>og R<3>en gruppe W, hvori X<3>betyr oksygen og T metylen eller karbonyl, inneholder R<2>gruppen 0C00 eller et substituert N-atom,
og deres hydrater,karakterisert vedat man a) omsetter en forbindelse med formel
hvori restene R4 , R<5>og R<6>har den samme betydning som restene R<1>, R<2>henholdsvis R3 , hvorunder imidlertid det i gruppen W foreligger en avspaltbar gruppe i stedet for resten -N<+>R A-, med et amin med formelen NR, eller b) omsetter en forbindelse med formelen
hvori restene R7 , R<8>og R<9>har den samme betydning som restene R<1>, R<2>henholdsvis R3 , hvorunder det imidlertid istedet for en av gruppene U og V foreligger en hydroksy- eller en aminogruppe,
med en syre med formelen
eller et reaktivt derivat derav, eller med et isocyanat med formelen
eller et imidazolid med formelen
eller c) omsetteren forbindelse med formelen hvori R°1 ,R02og R<03>har den samme betydning som restene R1 , R<2>henholdsvis R<3>, hvorunder imidlertid det i gruppen W foreligger en gruppe -N(Y3 , Y<4>) istedet for gruppen -N<+>R A-,
med et Ci-e-alkylhalogenid, hvorunder R<1>, R<2>, R<3>, -N+ R, A-, U, V, W, Z1 , Z2 , A1 , A2 , Y3 , Y4 , n og p har den ovenfor angitte betydning.
2. Fremgangsmåte ifølge krav 1, hvori R<1>er gruppe U, R<2>en gruppe V og R<3>en gruppe W,
karakterisert vedat man anvender tilsvarende substituerte utgangsmaterialer.
3. Fremgangsmåte ifølge krav 1 eller 2, hvori R<1>er oktadecanoyloksy, oktadecylkarbamoyloksy, oktadecyloksy, oktadecyloksyformamido, N-isopropyloktadecyloksyformamido eller l-isopropyl-3-oktadecylureido,
karakterisert vedat man anvender tilsvarende substituerte utgangsmaterialer.
4. Fremgangsmåte ifølge krav 1, 2 eller 3, hvori R<2>er acetamido, metoksyformamido, metoksy, metylkarbamoyloksy eller metoksykarbonyloksy,
karakterisert vedat man anvender tilsvarende substituerte utgangsmaterialer.
5. Fremgangsmåte ifølge et av kravene 1-4, hvori R<3>er [4-(trimetylammonio)-n-butyryloksy]klorid, [2-(1-metylpiperidino)-etoksykarbonyloksy]klorid, [4-(3-tiazolio)-n-butoksy]bromid, [4-(trimetylammonio)-n-butoksy]bromid eller [4-(3-metylimidazolio)-n-butoksy]bromid,
karakterisert vedat man anvender tilsvarende substituerte utgangsmaterialer.
6. Fremgangsmåte ifølge et av kravene 1-5, hvori R<1>er oktadecanoyloksy, oktadecylkarbamoyloksy, oktadecyloksy, oktadecyloksyformamido, N-isopropyloktadecyloksyformamido eller 1-isopropyl-3-oktadecylureido, R<2>acetamido, metoksyformamido, metoksy, metylkarbamoyloksy eller metoksykarbonyloksy, R<3>[4-(trimetylammonio)-n-butyryloksy]klorid, [2-(1-metylpiperidino)- etoksykarbonyloksy]klorid, [4-(3-tiazolio)-n-butoksy]bromid, [4-(trimetylammonio)-n-butoksy]bromid eller [4-(3-metylimidazolio)-n-butoksy]bromid,
karakterisert vedat man anvender tilsvarende substituerte utgangsmaterialer.
