Description
Oppfinnelsen vedrører en fremgangsmåte for overføring av varig antimikrobiell aktivitet til en produsert gjenstand ved å danne gjenstanden av akrylpolymerfibrer eller -stoffer.
Det er kjent at antimikrobielle kjemiske midler kan spinnes inn i eller påføres lokalt på syntetiske fibrer. Topiske behandlinger utsettes imidlertid lett for overflate-slitasje og fjernes eventuelt fra fiberen. Additiver er ubestandige og etter et begrenset antall omganger med vask og bruk, kan det kjemiske middel bli oppbrukt og den antimikrobielle beskyttelse gå tapt. En viktig usikkerhet i forbindelse med disse fremgangsmåter er additivnivået. Mengden av additiv som trengs for å gi ønskede egenskaper ved slutt-bruken, vil avhenge av brukstiden til sluttproduktet, graden av blanding med andre fibrer, hyppigheten av vaske- og bruks-omganger og graden av slitasje under normal bruk. Kombina-sjonen av disse faktorer vil kunne fordre urimelige høye additivnivåer som blir uoverkommelige både økonomisk og teknisk.
Hovedproblemet med spinningsfremgangsmåten er toksisi-teten til additivet. Ved visse sluttanvendelser, som for eksempel barneklær, medisinske produkter og intimutstyr, vil bestandige additiver kunne ha potensielle giftige virkninger. Videre krever mange fremgangsmåter for fremstilling av syntetisk fiber forhøyede temperaturer i langvarige tidsperi-oder, eller det gjøres bruk av sjeldne oppløsningsmidler under varme og trykk som vil kunne føre til uønskede bireak-sjoner med additivene som gir giftige biprodukter (f.eks. dioksiner), korrosive forbindelser (f.eks. klorider) og/eller fjerner additivets antimikrobielle aktivitet. Dette vil kunne føre til produksjons-, håndterings- og kaseringspro-blemer, og vil kunne kreve ytterligere teknikker for å produsere fibren på en sikker måte.
Overflatebundne alkoksysilan-holdige kvaternære ammoniumklorider gir antimikrobiell fiberbeskyttelse. Det biologisk aktive kvaternære ammoniumalkoksysilan påføres fiberen eller sluttproduktet fra en behandlingsoppløsning i metanol, og bindes til overflaten ved hydrolyse og kondensa-sjon av alkoksysilanet til hydroksylgrupper i fiberen. Men i tilfellet med akrylfibrer, polyesterfibrer, nylonfibrer og andre fibrer som er laget av polymerer som ikke inneholder betydelige mengder hydroksylgrupper, kan alkoksysilanene ikke bli bundet til fiberoverflaten ved hjelp av hydrolyse- og etterfølgende kondensasjonsreaksjoner. Siloksanet vil følge-lig bare kunne utgjøre en topisk behandling av disse fibrene. Man har støtt på flere problemer ved denne fremgangsmåte.
Den overflatepåførte sluttbehandling har et bestandig klorid-motion som lett kan byttes ut med organiske fosfater og sulfonater; når produktet således vaskes, kan den antimikrobielle effektivitet av den overflatepåførte sluttbehandling bli mye redusert. Et annet betydelig problem er metanolav-givelse under alkoksysilanbindingsprosessen, som kan gi an-ledning til bekymringer vedrørende toksisitet og mulig brenn-barhet. Endelig er metoksysilylforbindelser uforenlige med de fleste vandige emulsjonsbaserte etterbehandlingsmidler for tekstiler, noe som fører til ugunstig påføring av etterbehandlingsmiddel og holdbarhetsproblemer.
Det er nå oppdaget at visse typer akrylpolymerer, -fibrer og -stoffer har bestandige antimikrobielle egenskaper. Ved foreliggende oppfinnelse ble det oppdaget at bestandig antimikrobiell aktivitet overføres til akrylpolymerer, fibrer eller stoffer laget derav, ved kopolymerisering av en akrylprotonisert amin-komonomer og/eller ved bruk av protoniserte aminendegrupper.
De protoniserte aminseter er en del av akrylpolymer-kjeden og følgelig er deres holdbarhet i sluttproduktet uavhengig av sluttanvendelsesegenskaper, som for eksempel holdbarhetstid for sluttproduktet, hyppighet av vask-og-bruk-omganger og grad av slitasje under normal bruk. Videre er diffunderbare giftige additiver som kan by på alvorlige betenkeligheter når det gjelder visse sluttanvendelser, ikke til stede. Etter som det antimikrobielle middel er en del av selve polymeren, oppstår det ikke giftige biprodukter fra forholdene under spinning og fiberbearbeiding.
Dertil er de protoniserte aminer fordelt utover i polymeren og er således til stede i alle deler av fiberen og dens overflate. Likevektsfordelingen av protoniserte aminer er en naturlig medfølgende del av fiberen. Oppdagelsen av antimikrobiell aktivitet hos selve fiberen er overraskende i lys av det faktum omtalt i US patentskrift nr. 3.197.430, at fibrer fremstilt fra akrylnitrilpolymerer ble antatt ikke å ha den iboende evne til å inhibere veksten av bakterier. Fiberen ifølge foreliggende oppfinnelse er svært stabil, særlig når det gjelder de overflatebundne alkoksysilanholdige kvaternære aminer som lett deaktiveres av såpeoppløsninger.
En vesentlig forutsetning for antimikrobiell virkning er effektiv kontakt mellom det kjemiske middel og mikroorga-nismen. Etter effektiv kontakt reagerer kjemiske middel med en eller annen kritisk bestanddel i oppbygningen av mikroben og/eller mikrobemetabolismen. Bestemte virkningsmåter for antimikrobielle forbindelser er vanskelig å bestemme og kompliseres av konsentrasjons-, pH-, temperatur-,oppløsnings-middel-,elektrolytt- og faseseparasjonsvirkninger.
Uttrykket antimikrobiell er brukt for å beskrive reduksjon i antallet levedyktige patanogene mikroorganismer eller mikroorganismer som forårsaker ubehageligheter (f.eks. lukt, biologisk eller fysisk nedbrytning). Uttrykket bestandig er brukt for å beskrive egenskapen til den antimikrobielle akrylbestanddel som, med mindre det skjer en eksponering av selve akrylpolymeren mot sterke betingelser med hensyn til hydrolyse eller dekomponering, ikke kan fjernes fra fiberen.
Disse bestandige antimikrobielle egenskaper ifølge oppfinnelsen er ideelle for reduksjon av sykdomsforårsakende mikroorganismer, beskyttelse mot biologisk nedbrytning og er nyttige for luktreduksjon og melduggresistens, etc.
Akrylnitrilpolymerer, -fibrer og -stoffer med antimikrobielle egenskaper ville være nyttige som antimikrobielle luft- og vannfiltere for isolasjonsformål, oppløsningsstabili-satorer og membraner for rense- og konsentrasjonsformål, medisinske anordninger for beskyttelse mot sykdom i bandasjer, sårkompresser, tørkekluter, tamponger, svamper, kapper, dreneringsrør, katetere, suturer, etc, hygieneprodukter for luktreduksjon i bleier, sanitetsbind, sokker, etc, industri-elle produkter for melduggresistente malinger, antimikro bielle innpakningsmaterialer for økt holdbarhetstid for frukt og grønnsaker, og for mange andre anvendelser hvor man støter på mikroorganismer.
Additiver kan også brukes sammen med denne oppfinnelse for å gi ytterligere antimikrobiell eller annen hygienisk, biologisk eller medisinsk beskyttelse til akrylpolymerer, -fibrer eller -stoffer i sluttanvendelser hvor ytterligere reduksjon i antallet av levedyktige patanogene mikroorganismer eller mikroorganismer som forårsaker ubehageligheter, eller fjerning eller deaktivering av metabolske biprodukter, er ønsket.
Fremgangsmåte for fremstilling av polymer;
En liter avmineralisert vann ble tilsatt til en to-liters aluminiumsbeholder med vannkappe. Vannet ble dekket med nitrogengass og omrørt med mellom 50 og 150 omdreininger pr. minutt. Reaksjonstemperaturen ble innstilt på mellom 50 og 60°C med oppvarmet vann gjennom kappen på reaksjonsbeholderen. De enkelte monomerer ble utveid i ønskede mengder slik at den samlede vekt ble 400 g. Dersom den ble brukt, ble aminmonomeren tilsatt til reaksjonsbeholderen først og deretter ble pH regulert med 10% svovelsyre til 3 , 0 + 0 / !5 • De gjenværende monomerer ble tilsatt forsiktig til reaksjonsbeholderen. pH ble kontrollert på nytt og om nødvendig regulert. Temperaturen ble kontrollert og fikk komme tilbake innenfor grensen før tilsetning av initierings-midlene. I mellomtiden ble en av de følgende initiator-oppløsninger for polymerisering fremstilt: (la) 8 g 77 ppm jern(II)-ammoniumsulfat-oppløsning 20 g 11,5% hydrogenperoksydoppløsning
30 g 7% 1-thioglyserol-oppløsning
(lb) 8 g 77 ppm jern(II)-ammoniumsulfat-oppløsning
20 g 11,5% hydrogenperoksyd-oppløsning
30 g 7% thiourea-oppløsning
(2) 8 g 77 ppm jern(II)-ammoniumsulfat-oppløsning
30 g 20% natrium-bisulfitt-oppløsning
20 g 4,5% kaliumpersulfat-oppløsning
(3) 5 g 2,2<1->azobis-(2-amidinopropan)-hydroklorid
45 g avmineralisert vann
Deretter ble initiatoroppløsningen tilsatt til re-aks jonsbeholderen og den oppstartende reaksjon ble nøye overvåket. Rørerhastigheten ble sakte økt etter hvert som oppslemningen begynte å tykne. Avmineralisert vann ved værelsestemperatur ble sakte tilsatt etter hvert som temperaturen begynte å øke. Reaksjonen ble kjørt i 60 minutter fra tidspunktet for initiering. Dersom oppslemningen ble for tykk eller reaksjonstemperaturen ble for høy, ble en liten mengde oppslemning tatt ut og mer avmineralisert vann ved værelsestemperatur ble sakte tilsatt.
Ved slutten av reaksjonstiden ble en av de følgende oppløsninger for polymerherding tilsatt til reaksjonsbeholderen :
(1) 10 g 8% etylendiamintetraeddiksyre-oppløsning
15 g 20% natriumbisulfitt-oppløsning
(2) 10 g 8% etylendiamintetraeddiksyre-oppløsning
(3) kaldt vann for å nedsette reaksjonstemperaturen til under 45°C.
Etter at den passende herdeoppløsning var blitt tilsatt, ble polymeroppslemningen omrørt i 50 minutter. Oppslemningen ble sugt ut av reaksjonsbeholderen ved hjelp av vakuumutsugning og filtrert på en Buckner-trakt. Deretter ble polymeren fjernet fra filtertrakten, oppslemmet på nytt i en liter avmineralisert vann ved 80°C, omrørt i 10 minutter og filtrert. Filtreringstrinnet etter ny oppslemning ble gjentatt to ganger. Polymeren ble deretter oppslemmet i aceton av reagensgrad, omrørt 10 minutter og filtrert på en Buckner-trakt. Den vaskede polymer ble så pakket inn i oste-klede, plassert på et brett av rustfritt stål og tørket 16-24 timer i en ovn med luftgjennomstrømning ved 65°C.
Fiberfremstilling
Fibrer ble fremstilt ved hjelp av standardteknikker for tørrspinning av akryl. Polymeren ble oppløst i dimetyl-formamid opptil et nivå på 23-33% faste stoffer, varmet opp til mellom 90 og 110°C, filtrert, deretter varmet opp til 110-150°C og ekstrudert gjennom en spinnedyse med 1400 hull ved en spinningshastighet på 270 ypm. Nitrogen som avsug-ningsgass fraktet oppløsningsmidlet bort etter hvert som de enkelte filamentene ble dannet. Fibrene samlet opp ved ut-løpet av spinnecellen hadde en denier pr. filament som spunnet på 9,0. Restoppløsningsmiddel ble ekstrahert fra den spundne fiber i 90-100°C vann etter hvert som fiberen ble strukket enten 4,5x eller 6, 0x hvorved man fikk denier pr. filamenter som strukket på henholdsvis 2,0 og 1,5. Standard etterbehandlingsmiddel for tekstilfiber ble påført ved å bruke en rullepåfører for å redusere friksjonen mellom fibrene og gi glatthet under etterfølgende tekstilbearbeid-ing. Det påførte etterbehandlingsmiddel inneholdt et kationisk antistatikum fremfor et anionisk antistatikum, etter-som det anioniske antistatikumet ville reagere med det protoniserte amin. Deretter ble den strukne fiber krympet mekanisk og samlet opp ved utløpsenden av en strekke-uttrekkingsmaskin. Det mekanisk krympede garn ble deretter tørket i en tørker mellom 90 og 120°C hvorved man fikk endelig denier pr. filament på henholdsvis 3,0 og 2,2.
Fremgangsmåte for måling av antimikrobiell aktivitet
Den bestandige antimikrobielle aktivitet hos polymerer, fibrer og stoffer ble evaluert ved hjelp av testfremgangsmåten med rystekolbe og metoden med parallell strek.
A. Rystekolbetest. Ved testfremgangsmåten med rystekolbe ble 70 ml steril kaliumhydrogenfosfatbuffer-oppløsning ved pH 7,2 tilført sterile 250 ml Erlenmeyer-kolber. Hver kolbe ble så inokulert med 5 ml av et bakterie-inokulum hvorved man fikk 10 4 kolonidannende enheter/ml oppløsninger. Kolbene ble rystet godt og tellinger ved 0-tidspunktet ble gjort ved å fjerne 1 ml alikvoter fra hver kolbe og lage 1:10 og 1:100 fortynninger i fosfatbuffer. Duplikat-plater med fortynningen 1:100 i tryptonglukoseekstraktagar ble fremstilt, inkubert ved 37°C i 18-24 timer og telt. Et gram testpolymer eller -fiber ble tilsatt aseptisk til hver kolbe. En ekstra kolbe ble fremstilt som inneholdt bakterieinokulumet, men ingen testoppløsning, for å forsikre seg om at bakteriekulturen var levedyktig i test-oppløsningen. Kolbene ble plassert på en veivtapp-drevet ryster og rystet i 1-2 timer. Tellingene etter 1-2 timers kontakttid ble gjort ved å fjerne 1 ml alikvoter fra hver kolbe og lage 1:10 og 1:100 fortynninger i fosfatbuffer. Duplikat-plater av begge fortynningene ble fremstilt i tryptonglukoseekstraktagar, inkubert ved 37°C i 18-24
timer og fortynningene som inneholdt et antall på 30-300,
ble telt. Antallene ved 0-tid og antallene etter 1 times kontakttid ble så korrigert med hensyn på sine respektive fortynningsfaktorer og 100 ganger differansen mellom de opprinnelige antall og sluttantallene dividert med de opprinnelige antall var den prosentvise reduksjon for test-gjenstanden. Rystekolbefremgangsmåten gav resultater for de relative bioaktiviteter hos forskjellige polymerer, fibrer og stoffer mot en lang rekke mikroorganismer. Resultatene er gjengitt i tabellene II-VI og VIII. Testfremgangsmåten med rystekolbe er beskrevet i Malek, J.R. og Speier, J.L., The Journal of Coated Fabrics, Vol. 12, juli, 1982, ss. 38-45.
B. Metoden med parallellstrek. For å bekrefte bestandigheten til den antimikrobielle bestanddel, ble metoden med parallellstrek anvendt. Sterilisert agarvekstmedium ble fremstilt og 15 ml helt over i hver flatbunnet petri-skål med diameter 100 mm. Agaren fikk så stivne til en gel. En 1 ml prøve av dyrkningsvæske fra den ønskede testorganis-me ble overført til 9 ml sterilt avmineralisert vann og blandet ved omrøring med en 4 mm inokuleringssløyfe. En sløyfefull av det fortynnede inokolum ble overført til en steril agarplate ved å lage fem 7,5 cm lange parallelle streker 1 cm fra hverandre i midten av platen uten å fylle sløyfen på nytt, idet det ble passet på at agaroverflaten ble brutt. Et avlangt prøvestykke ble forsiktig presset på tvers av de fem inokuleringsstreker for å sikre nøye kontakt med agaroverflaten. Prøven ble så inkubert i 18-24 timer ved 37°C. De inkuberte plater ble undersøkt med hensyn på for-styrrelse som skyldes vekst, langs strekene av inokulum under prøven, og med hensyn på en klar sone utenfor prøve- kanten. Den gjennomsnittlige bredde til inhiberingssonen rundt prøvegjenstanden ble beregnet med en linjal inndelt i millimeter. Resultatene er gjengitt i tabell VII. Eksempel 1
Det ble fremstilt polymerprøver etter fremgangsmåten beskrevet ovenfor. Monomerer av akrylnitril (AN) og metylakrylat (MA) ble kopolymerisert med forskjellige protoniserte aminholdige monomerer. Disse polymerene ble så smittet med Klebsiella pneumoniae i rystekolbetesten. Resultatene fra disse forsøkene er gjengitt i tabell II. Dette eksemplet viser at kopolymeriseringen av protoniserte aminholdige monomerer i akrylpolymerer overfører antimikrobiell aktivitet.
Eksempel 2
For å bestemme virkningene av protoniserte aminholdige endegrupper på polymerbioaktivitet, ble en rekke polymeri-seringsinitiatorer testet. Polymerprøver ble fremstilt i henhold til den beskrevne fremgangsmåte og ble smittet med Klebsiella pneumoniae i rystekolbetesten. Eksempel 3

Eksemplene 4, 5, 6 og 7 omfatter gjenstandene 18 og 19. Som det vil ses av tabell V, omfatter gjenstand 18 ikke noe protonisert amin, mens gjenstand 19 inneholder et protonisert amin.
Eksempel 4
Det ble fremstilt polymerer i en kontinuerlig utflyt-reaksjon og disse ble deretter spunnet til fibrer i henhold til fremgangsmåten beskrevet ovenfor. Bioaktiviteten til polymeren og fiberen ble bestemt i rystekolbetesten mot Klebsiella pneumoniae. Resultatene i disse forsøkene er gjengitt i tabell V. Disse resultatene viser at bioaktive polymeregenskaper beholdes når polymeren omdannes til fiber.
Eksempel 5
Fiberprøver ble fremstilt i henhold til fremgangsmåten beskrevet ovenfor og de ble smittet med et bredt spekter av mikroorganismer i rystekolbetesten. Resultatene fra disse forsøkene er gjengitt i tabell VI. Disse resultatene viser at fibrer laget av akrylpolymerer som inneholder protonisert amin, har et høyt bioaktivitetsnivå mot et bredt spekter av mikroorganismer.
Eksempel 6
Fiberprøver ble fremstilt i henhold til fremgangsmåten beskrevet ovenfor og de ble smittet med Klebsiella pneumoniae i parallellstrek-metoden for å bekrefte bestandigheten av aminholdige akrylfibrer. Resultatene fra disse forsøkene er gjengitt i tabell VII.
Eksempel 7
Fiberprøver fremstilt ved hjelp av fremgangsmåten beskrevet ovenfor ble brukt til å produsere ikke-vevde produkter og de ble smittet med Klebsiella i rystekolbetesten. Resultater fra disse forsøk er gjengitt i tabell VIII. Disse resultatene viser at det høye nivå varig bioaktivitet funnet i akrylfibrer som inneholder protonisert amin, opp-rettholdes i ikke-vevde stoffer.
Claims (14)
1. Fremgangsmåte for overføring av bestandig antimikrobiell aktivitet som er effektiv når det gjelder å nedsette antallet levedyktige mikroorganismer, og som gir en prosent-vis nedsettelse av Klebsiella i rystekolbetesten på 70-100%, til en produsert artikkel, karakterisert ved at den produserte artikkel lages av en akrylnitrilblanding av minst 80 vekt% akrylnitril, opp til 13 vekt% av en nøytral etylen-umettet monomer og fra 0,1 til 10 vekt% av en forbindelse som inneholder protonisert amin, idet den antimikrobielle aktivitet er til stede i akrylnitrilblandingen.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at forbindelsen som inneholder protonisert amin, er en komonomer som inneholder protonisert amin.
3. Fremgangsmåte ifølge krav 1, karakteri sert ved at forbindelsen som inneholder protonisert amin, er en polymerendegruppe som inneholder protonisert amin.
4. Fremgangsmåte ifølge krav 2, karakteri sert ved at komonomeren som inneholder protonisert amin, er en akrylmonomer valgt fra gruppen bestående av:
dimetylaminoetylmetakrylat (DMAM),
dietylaminoetylmetakrylat (DEAM),
tertiært-butyl-aminoetylmetakrylat (TBAM),
dimetylaminoneofenylakrylat (DMANPA),
2-metyl-5-vinyl-pyridin (MVP) og
dimetylaminopropylmetakryloamid (DMAPMA).
5. Fremgangsmåte ifølge krav 3, karakteri sert ved at polymerendegruppen som inneholder protonisert amin, er en amidinogruppe bundet til polymerkjeden og valgt fra gruppen bestående avt hiourea og 2-amidino-propanhydroklorid.
6. Fremgangsmåte ifølge krav 2 eller 3, karakterisert ved at den produserte artikkel er et filter som vil kunne komme i kontakt med mikroorga-nismene .
7. Fremgangsmåte ifølge krav 2 eller 3, karakterisert ved at den produserte artikkel er en medisinsk anordning som vil kunne komme i kontakt med biologiske fluider.
8. Fremgangsmåte ifølge krav 2 eller 3, karakterisert ved at den produserte artikkel er en beholder for emballering av matvarer.
9. Fremgangsmåte ifølge krav 2 eller 3, karakterisert ved at den prosentvise reduksjon av Klebsiella ved rystekolbetesten er 99,9-100%.
10. Fremgangsmåte ifølge krav 2 eller 3, karakterisert ved at akrylnitrilblandingen er en fiber som er påført et etterbehandlingsmiddel som inneholder kationisk antistatikum.
11. Fremgangsmåte for å begrense antallet levedyktige mikroorganismer forbundet med et pattedyr, karakterisert ved å bringe huden eller andre deler av pattedyret i kontakt med en produsert artikkel dannet av en akrylnitrilblanding av minst 85 vekt% akrylnitril, opp til 13 vekt% av en nøytral etylen-umettet monomer og fra 0,1 til 10 vekt% av en forbindelse som inneholder protonisert amin.
12. Fremgangsmåte ifølge krav 11, karakterisert ved at den produserte artikkel er et rør.
13. Fremgangsmåte ifølge krav 11, karakterisert ved at den produserte artikkel er et vevd stoff.
14. Fremgangsmåte ifølge krav 11, karakterisert ved at den produserte artikkel er en sutur.
