Oppfinnelsens område
Foreliggende oppfinnelse angår en bionedbrytbar matriks, og nærmere bestemt en multifunksjonell collagen-basert matriks og/eller bærersystem derav.
Bakgrunn for oppfinnelsen
Utlevering av legemidler, vekstfaktorer, hormoner, peptider og glycopeptider til ytre sår har klassisk funnet sted ved direkte topisk administrering og påføring til indre sår ved injeksjon i blodet eller ved absorpsjon i blodet gjennom fordøyelsessystemet. Regulert frigivelse av disse midler er blitt oppnådd ved innkapsling enten i større om-fang eller til et mikroskopisk nivå under anvendelse av syntetiske polymerer slik som silikon, og naturlige polymerer slik som gelatin og cellulose. Frigivelseshastigheten kan reguleres i perioder opp til et år ved riktig valg av det polymere system som anvendes for å regulere diffusjons-hastigheten (Langer, R.S. og Peppas, N.A., Biomaterials 2, 201, 1981) . Naturlige polymerer slik som gelatin og cellulose, oppløses langsomt i løpet av minutter til timer, mens silikon forblir intakt i perioder på måneder til år. Bionedbrytbare polymerer medfører en fordel når det gjelder regulert frigivelse til indre sår da bare en enkel kirurgisk prosedyre er nødvendig.
Collagen er en bionedbrytbar polymer som finnes i dyr og mennesker. Det har vært anvendt som et plasmafordrøy-ningsmiddel, bærer for legemiddelutlevering, glasslegeme-erstatning, hemostatisk middel, sømmateriale, hornhinne-erstatning, hemodialysemembran, sårforbindingsmateriale og kunstig hud, brokklapp, karprotese, vaginal-antikonsepsjons-middel og injiserbart middel for vevforøkelse (Chvapil et al., Int. Review of Connective Tissue Research 6, 1, 1973;
Chvapil, in Biology of Collagen, redigert av A. Viidik og
J. Vuust, Academic Press, kapittel 22, 1980). I de fleste
av disse anvendelsesområder rekonstitueres collagen og tverrbindes i en uløselig form.
Yannas et al. beskriver i US patentskrift 4 060 081 anvendelse av collagen og mucopolysaccharider som syntetisk hud. Et slikt materiale tverrbindes under anvendelse av glutaraldehyd, et bifunskjonelt tverrbindingsmiddel, som reagerer med frie aminer. En hovedulempe med anvendelse av tverrbundet collagen har vært den uheldige biologiske effekt av fritt glutaraldehyd, et vanlig middel som anvendes for å tverrbinde og uløseliggjøre collagen i mange anvendelsesområder. Utlutning av glutaraldehyd fra tverrbundet collagen har vist seg å være cytotoksisk overfor celler, spesielt fibroblaster (Speer et al., J. Biomedical Materials Research 14, 753, 1980; Cooke et al., British J. Exp. Path. 64, 172, 1983). Nylige undersøkelser antyder at glutaraldehydpoly-merer og ikke monomert glutaraldehyd danner tverrbindinger mellom collagenmolekylene, og disse tverrbindinger kan deretter omordnes for å frigi fritt glutaraldehyd og glutar-aldehydpolymerer (Cheung, D.T. og Nimni, M.C., Connective Tissue Research 10, 187-217, 1982).

De collagen-baserte bærersystemer ifølge oppfinnelsen
er basert på anvendelse som utgangsmateriale av et løselig eller uløselig collagen valgt fra gruppen bestående av collagener av type I, Il og III og blandinger derav.
Løselige collagener av type I, II og III fremstilles
ved begrenset enzymatisk oppslutning av vev anriket i slike collagentyper og omdannes til en collagen-basert løsning (dvs. et løselig collagen oppløst i et egnet løsningsmiddel slik som fortynnet saltsyre, fortynnet eddiksyre eller lignende) .
Uløselige collagener er avledet fra følgende typiske kilder: type I collagen; kveg, kylling og fiskeskinn, kveg og kyllingsener og kveg og kyllingben innbefattende foster-vev; type II collagen: kvegleddbrusk, neseskillevegg, ribbensbrusk; og type III collagen: kveg- og human-aorta og hud.
I en utførelsesform av oppfinnelsen underkastes en collagen-basert løsning eller et uløselig collagen dispergert og svellet i et egnet væskemedium (f.eks. fortynnet saltsyre, fortynnet eddiksyre eller lignende) en temperatur på mellom 0 og -100°C for derved å bringe det collagen-baserte materiale i fast form. Eksempler på bifunksjonelle aktive succinimidylestere innbefatter bifunksjonelt N-hydroxysuccinimid, 3,3^-dithio-(sulfosuccinimidyl)-propionat og bis-(sulfosuccinimidyl)-suberat. Ved anvendelse av et carbodimid-tverrbindingsmiddel nedsenkes den collagen-baserte svamp eller ark i en carbodiimidløsning i en konsentrasjon på fra 0,1 til 10% (w/v) opprettholdt ved en temperatur på fra 2 til 40°C, og ved en pH på mellom 2 og 11 i et tidsrom på fra 2 til 9 6 timer. En slik bærerforbindelse er valgt fra gruppen bestående av collagen type IV og V, fibronectin, laminin, hyaluronat, proteoglycaner, epidermisk vekstfaktor, blodplate-avledet vekstfaktor, angiogenesefaktor, antibiotikum, antifungalt middel, spermacidalt middel, hormon, enzym og enzyminhibitor.
Generelt kan bærerforbindelsen innføres på et hvilket
som helst tidspunkt under bearbeidelsen av det collagen-baserte materiale til den collagen-baserte matriks i svamp-eller arkform. Fortrinnsvis innarbeides bærerforbindelsen før det løselige eller uløselige collagen bringes i fast form ved dannelse av den collagen-baserte mellomproduktmatriks, eller før den collagen-baserte mellomproduktmatriks bringes i fast form før den strenge dehydratisering. Bærer-materialene tilsettes i en mengde på fra 1 til 30 (w/w) basert på vekten av løselig eller uløselig collagen. Generelt innarbeides bindevevfaktorer slik som fibronectin, collagen

Collagener av type IV og V finnes assosiert med basalmembraner og glattmuskelceller i vev. Typiske kilder for proteoglycaner innbefatter kveg- og human brusk og synovialvæske, neseskillevegg og ribbensbrusk, og hud. Typiske kilder for glycoproteiner innbefatter EHS tumor, kveg- og humane nyrer, brusk, ben og placenta såvel som blod fra kveg og mennesker. Typiske kilder for hyaluronat innbefatter haneskjell og glasslegeme fra kveg.
Foretrukne bærerforbindelser for den collagen-baserte matriks innbefatter fibronectin, laminin, type IV collagen og komplekser av hyaluronat og proteoglycaner. En verdi for svellingsforholdet på mellom 2,5 til 5 er nødvendig for en collagen-basert matriks som kommer i kontakt med åpne sår, mens et svellingsforhold på mellom 2,5 til 10 er anvendbart for en collagen-basert matriks innbefattende bærerforbindelser for subkutan implantasjon. Svellingsforholdet defineres som volumet av absorbert vann pr. volumenhet av collagen-basert matriks.
Når det gjelder innvendige sår og korttidsfrigivelse
av legemiddel, anbringes den collagen-baserte matriks innbefattende en bærerforbindelse i form av et ark eller en slange i direkte kontakt med vev.
Bionedbrytningshastigheten og frigivelseshastigheten kan reguleres ved variasjon av tverrbindingsdensiteten. For anvendelsesområder hvori langvarig frigivelse ønskes, påføres et ikke-porøst diffusjonslag av en bionedbrytbar polymer
slik som poly-3-(hydroxybutyrat) på hver side av den collagen-baserte matriks. For materialer som skal anvendes på kjøtt-sår, anbringes et diffusjonskontrollag slik som beskrevet i
det tidligere angitte US patent tilhørende Yannas, for å forhindre diffusjon av vann eller andre små flyktige molekyler fra den collagen-baserte matriks, idet diffusjonskontrollaget må adhere tett til det collagen-baserte matrikslag. Kombinasjonen av collagen-basert matrikslag og diffusjonskontrollag må være sterk nok til å kunne syes og må ha en strekkstyrke på minst 7 kg/cm og må være i stand til å deformeres til minst 10% før sammenbrudd. Syntetiske ikke-bionedbrytbare polymerer slik som silikonpolymerer (Silastic Medical Grade Adhesive) eller bionedbrytbare polymerer slik som poly-melkesyre, polyglycolsyre, poly-3-(hydroxybutyrat)
og copolymerer av disse materialer, kan anvendes som diffusjonskontrollaget.
Silikonpolymerer er foretrukne materialer for å kon-trollere diffusjonsgraden av små molekyler. Herding av klebemidlet i diffusjonskontrollaget kan finne sted uten fukting av bærerforbindelsen i den collagen-baserte matriks ved tørking ved romtemperatur. En film med tykkelse på 0,5 til 1,5 mm påføres matrikslaget og tillates å herde ved romtemperatur i minst 2 timer under anvendelse av et vakuum på 35,5 cm Hg.
Tykkelsen av den collagen-baserte matriks kan varieres fra 1 til flere hundre mm. For kjøttsår er en tykkelse på 2 til 3 mm ønskelig og muliggjør nær kontakt mellom bærerforbindelsen i den collagen-baserte matriks og sårskiktet.
Når den implanteres subkutant eller direkte på dermale kjøttsår, vil en hvilken som helst collagen-basert matriks
ifølge oppfinnelsen ikke stimulere den inflammatoriske prosess. I tillegg vil kronisk implantasjon resultere i fibroblast- og endothelial-cellemigrering inn i den collagen-baserte matriks. Nedbrytnings- og resorpsjonsgraden av
collagen-baserte matriser er som bestemt in vitro ved bestemm-else av den tid som 1 mg av materialet motstår oppløselig-gjøring av 100 enheter av bakteriell collagenase. En enhet av collagenase frigir aminosyrer i collagen ekvivalent med 1,0 mikromol L-leucin i løpet av 5 timer ved 37°C. En kom-binasjon av dannelse av succinimidyl-aktiv ester og streng dehydratisering eller carbodiimidbehandling og streng
dehydratisering øker collagenase-resistenstiden signifikant i forhold til den som observeres ved en hvilken som helst av de tidligere anvendte prosedyrer. Eksempel 1
Fremstilling av løselige og uløselige collagener
Det anvendte collagen ble fremstilt fra kalvehud etter at narvelaget var fraskilt fra lærhuden eller dermis. Lærhuden ble kuttet i biter, vasket, svellet, frysetørket og lagret ved -20°C. Løselig collagen ble erholdt ved å an-bringe 250 g av det frysetørkede, uløselige collagen i 1,5 1 HCl ved en pH på 2,5 inneholdende 1,5 g krystallinsk pepsin (Sigma Chemical Company). Blandingen ble omrørt over natten ved 4°C og ble deretter filtrert gjennom et osteklede, Whatman filterpapir nr. 4 etterfulgt av Whatman filterpapir nr. 42, Gelman 5 ,um filter og l,2^um filter og til slutt gjennom Millipore® 0,65^um og0,45^um filtere. Den løselige fraksjon erholdt etter sekvensfiltrering, inneholdt collagener av type I, III og V. Den løselige collagenfraksjon ble dialysert mot en 1,0 M NaCl-løsning inneholdende 50 mM tris justert til pH 7,5, og nok fast NaCl ble tilsatt for å heve molariteten på løsningen til 1,7. Bunnfallet ved 1,7 M NaCl er collagen av type III, og dette ble oppsamlet ved sentrlfugering ved 10.000 x g i 60 minutter og ble deretter dialysert mot 0,005 M eddiksyre, frysetørket og lagret ved

Uløselige collagenløsninger ble erholdt ved disperger-ing av oppmalt, frysetørket lærhud under anvendelse av en Wiley-mølle i HC1 ved pH 2,5. Det svellede collagen ble deretter dialysert mot 0,005 M eddiksyre og ble frysetørket.
Hvis hud fra voksent kveg er kilden for lærhuden, er det frem-stilte, uløselige collagen typisk type I.
Løselig collagen av type IV ekstraheres fra EHS muse-sarcomatumor etter vasking av tumoren i kaldt, destillert vann 3 ganger ved sentrifugeringsfiltrering ved 10.000 x g i 30 minutter. Pelletten (0,500 g) ble homogenisert i 0,5 1 0,5 M eddiksyre justert til pH 2,5 med HC1, og 0,5 g pepsin ble tilsatt. Homogenatet fikk blandes ved 4°C i 24 til 48 timer, ble filtrert gjennom et osteklede, sentrifugert ved 10.000 x g i 30 minutter. Pelletten ble resuspendert i
0,5 M NaCl inneholdende 50 mM tris, NaCl ble tilsatt inntil en sluttkonsentrasjon på 4,5 M ble erholdt, hvorpå blandingen ble sentrifugert ved 10.000 x g i 30 minutter, hvoretter
pelletten ble dialysert mot 0,1 N eddiksyre 3 ganger og ble frysetørket.
Eksempel 2 Fremstilling av kvegserum- fibronectin
80 ml friskt uttrukket kvegblod ble oppsamlet i et polypropylenrør inneholdende 20 ml 5% trinatriumcitrat og
0,1 mM fenylmethylsulfonylfluorid. Blodet ble sentrifugert ved 300 x g i 10 minutter, og plasmaet ble deretter separert
fra cellelaget og ble ytterligere sentrifugert ved 30.000 x g ved en temperatur på 15°C i 30 minutter. Til plasmaet ble deretter tilsatt ytterligere 1 mM fenylmethylsulfonylfluorid og ble deretter helt gjennom en gelatin-sefarosekolonne. Kolonnen ble vasket med en fosfatbufferløsning (0,182 M fosfat og 0,145 M NaCl), 2 kolonnevolumer 1 M NaCl i en fosfatbuffer, 2 kolonnevolumer fosfatbufferløsning, 2 kolonnevolumer 4 M urea i en fosfatbuffer. Prøven ble deretter dialysert mot 0,05 M tris-HCl inneholdende 4,5 M urea (pH 7,2) og ble anbragt på en DEAE (diethyl-aminoethylcellulose) ionebytterkolonne ved romtemperatur. Kolonnen ble eluert med en 0 til 0,3 M lineær NaCl-gradient
i en 0,05 M tris-HCl bufferløsning (pH 7,2) inneholdende 4,5 M urea. Det eluerte fibronectin ble dialysert mot 0,180 M fosfat inneholdende 0,145 M NaCl og 1,0 M urea og ble fryst ved -20°C.
Eksempel 3
Fremstilling av laminin
Laminin ble fremstilt fra lathrytisk EHS-tumorer dyrket i C57/6J mus. Tumorene ble dissekert og homogenisert i 3,4 M NaCl inneholdende 50 mM tris-HCl justert til pH 7,4 og proteaseinhibitorer.Homogenatet ble sentrifugert ved 16.000 x g i 60 minutter, og pelletten ble oppsamlet og resuspendert i 3,4 M NaCl. Etter sentrifugering ved 16.000 x g i 60 minutter en ytterligere gang ble homogenatet resuspendert i 0,5 M NaCl inneholdende 50 mM tris-HCl, pH 7,4, og proteaseinhibitorer, og ble omrørt i 18-24 timer ved 4°C, etterfulgt av sentrifugering ved 16.000 x g i 60 minutter. SephacrylV C-300 ekvilibrert med 1,0 M CaCl2, 50 mM tris-HCl, pH 7,5, ved 22°C, og hulvolumet ble oppsamlet og dialysert mot 0,4 M NaCl, 50 mM tris-HCl, pH 7,4, og ble lagret ved 4°C. Laminin ble oppløst på nytt i 0,1 M ammoniumhydroxyd, pH 11,0, hvorpå pH ble justert til 7,2.
Eksempel 4
Fremstilling av collagen- baserte svamper og ark
1,2 g løselig eller uløselig collagen ble tilsatt til 120 ml av en fortynnet HCl-løsning med pH 3,0, og blandingen ble malt i en Waring blander ved lav hastighet i 1 minutt og deretter ved høy hastighet i 1 minutt. Løsningen eller dispersjonen ble deretter helt over i en vakuumkolbe og ble avluftet ved et vakuum på 100 millitoll i 10 minutter. Collagendispersjonen eller løsningen som skal omdannes til svamper, ble avkjølt til 0°C og fryst ved -100°C før fryse-tørking ved -65°C under et vakuum på mindre enn 10 millitorr. Eksempel 5 Fremstilling av succinylert, succinimidylester-tverrbundet, intermediær collagen- basert matriks
9 g ravsyreanhydrid ble oppløst i 80 ml destillert
vann og ble blandet i 30 minutter ved 37°C. Etter at ravsyreanhydrid forelå i løsning, ble pH justert til 7,2, og volumet ble bragt til 100 ml. Denne løsning ble anbragt i en Waring blander, og 1,0 g collagen ble tilsatt, blandingen ble malt
i 2 minutter og fikk stå ved 22°C i 1 time og ble deretter
anbragt på et Whatman filterpapir nr. 4 for å fjerne uomsatt ravsyreanhydrid, og ble deretter vasket med 100 ml destillert vann.

Eksempel 6
Fremstilling av collagen-basert matriks inneholdende fibronectin
Til 1,2 g løselig eller uløselig collagen i 120 ml HCl, pH 3,0, ble tilsatt 0,12 g fibronectin (2,5 mg fibronectin/ml) i 0,1 M urea inneholdende 0,182 M fosfat og 0,145 M NaCl, og blandingen ble dispergert i en Waring blander i 2 minutter. Ark og svamper ble fremstilt i henhold til fremgangsmåten beskrevet i eksempel 4.
Eksempel 7
Svamper og ark dannet med belegg av collagen type I med laminin, fibronectin og collagen av type IV

Eksempel 9
Fremstilling av collagen-basert matriks inneholdende collagen av type IV
Til 1,2 g løselig eller uløselig collagen av type I iHCl, pH 2,0, ble tilsatt 0,012 g collagen av type IV i 0,1 M ammoniumacetat, pH 7,2, til et sluttvolum på 120 ml. Blandingen ble dispergert i en Waring blander i 2 minutter og ble formet til ark eller svamper i henhold til fremgangsmåten beskrevet i eksempel 4.
Eksempel 10 Cyanamid- tverrbinding av collagen- basert matriks
Produktet ifølge eksempel 4 og 6 til 9 ble tverrbundet ved nedsenkning i en vandig løsning inneholdende 1 vekt% cyanamid ved pH 5,5 i 24 timer ved 22°C. Etter fjerning fra badet ble svampene og arkene grundig vasket i flere por-sjoner vann i løpet av 24 timer, ble fyst og frysetørket ved -65°C i et vakuum på mindre enn 50 millitorr.
Eksempel 11 Tverrbinding av collagen-basert matriks ved streng dehydratisering
Produktene ifølge eksempel 4 og 6 til 10 ble anbragt 1 en vakuumovn ved romtemperatur og utsatt for et vakuum på
mindre enn 50 millitorr. Etter 1 time ble prøvene oppvarmet til 110°C og holdt ved denne temperatur i 7 2 timer på hvilket tidspunkt temperaturen ble nedsatt til 40°C. Prøvene ble
deretter fjernet fra vakuumovnen og lagret ved -20°C.
Eksempel 12 Tverrbinding av collagen-basert matriks under anvendelse av succinlmidyl- aktiv ester
2 g collagen tverrbundet med succinimidyl-aktiv ester fremstilt i henhold til eksempel 5, ble anbragt i en Waring
blander inneholdende 400 ml HCl, pH 2,0, og dispergert i 2 minutter. Denne blanding ble avluftet ved at den ble anbragt i et vakuum på 300 millitorr og deretter anbragt ved
romtemperatur i et miljø med 100% relativ fuktighet i 24 timer. Materialet ble deretter avkjølt til 0°C, fryst og frysetørket ved -6 5°C under anvendelse av et vakuum på mindre enn 50 millitorr, eller ble lufttørket for dannelse av hhv. svamper eller ark.
Eksempel 13
Fremstilling av collagen-basert matriks inneholdende proteaseinhibitorer
Svamper og ark fremstilt i henhold til eksempel 4 til 9, ble anbragt i 20 ml HCl ved pH 2,0, inneholdende 25% cystein eller 0,1% (w/v) a-2-macroglobulin. Blandingen ble fryst og frysetørket ved -65°C ved et vakuum på mindre enn 50 millitorr, eller ble lufttørket ved romtemperatur.
Eksempel 14
Enzymatisk nedbrytning in vitro
1 cm 2 av hvert av produktene i de foregående eksempler ble anbragt i 2,0 ml 10 mM tris-HCl, pH 7,4, inneholdende 25 mM calciumklorid og 100 enheter av collagenase av type IV fra Clostridium histolyticum (Sigma Chemical Co.) ble tilsatt pr. mg prøve. Prøvene ble anbragt i omgivelser ved 37°C, og intaktheten av hver prøve ble kontrollert visuelt hvert 10. minutt. Tiden ble nedtegnet når hver prøve var visuelt blitt nedbrutt til biter som var mindre enn 0,5^,um. Resultatene er oppført i tabell I.
Eksempel 15 In vitrobestemmelse av svelleforhold
Svelleforholdet av denaturert collagen er omvendt
beslektet med tverrbindingsgraden. Produktene ifølge de ovenfor angitte eksempler ble kokt i 2 minutter i destillert vann og ble anbragt mellom to servietter. En vekt på 1 kg ble anbragt på toppen av serviettene inneholdende materi-alene i 20 sekunder. Våtprøvevekten ble nedtegnet, og prøven ble deretter tørket ved 110°C i 3 timer. Eksempel 16 Mekaniske egenskaper av collagen- baserte matriser
Svamper og ark fremstilt i henhold til de ovenfor angitte eksempler, ble kuttet i rektangulære (4,0 cm x 1,0 cm) remser og nedsenket i fosfatbufferløsning ved pH 7,5 i 20 minutter før mekanisk testing. Remsene ble testet i uni-aksialt strekk ved 22°C i en belastningsgrad på 10%/minutt under anvendelse av en Instron® testanordning modell 1122. Endene av remsene ble anbragt i pneumatiske gripeanordninger som ble lukket under et trykk på 2,8 kg/cm 2 med en målelengde på 20 mm. Spenning-belastningskurver ble erholdt, fra hvilke Youngs modul ved høye (HSM) og lave belastninger ble beregnet. Belastningen ved hvilken området med lav modul sluttet, ble angitt som eT, og belastningen ved svikt ble angitt som e_. Belastningsresultåtene er angitt i tabell I.
Eksempel 17
Subkutan bioforenelighet av collagen- basert matriks
Svamper og ark fremstilt i henhold til de ovenfor angitte eksempler, ble testet med hensyn til subkutan bioforenelighet etter sterilisering ved eksponering av 2,5 M rad gammabestråling. Implantering ble foretatt under sterile betingelser under anvendelse av 3 50 g hvite hunnmarsvin som testdyr.
Et 1 cm kutansnitt ble foretatt på en side av ryggen, og huden ble skilt fra muskelhinnelaget, og en 1 cm x 1 cm bit av implantat ble anbragt i dette rom. Kantene av huden ble festet sammen over implantatet under anvendelse av sår-klips.
Dyrene ble avlivet på den 6., 9. og 12. dag etter implantering, og vevet inneholdende bæreren, ble anbragt i Carsons fiksativ og bearbeidet for histologiske studier. Resultatene er angitt i etterfølgende tabell II.
Eksempel 18
Fremstilling av diffusjonskontrollag på et matrikslag
Under anvendelse av svamper og ark fremstilt i henhold til eksempel 4 til 10, ble et 1 mm lag av Silasti<_®>Medical Grade klebemiddel påført på overflaten av matrikslaget under anvendelse av en spatel. Diffusjonskontrollaget ble herdet ved påføring av et vakuum på 100 millitorr i et tidsrom på 2 timer ved 22°C. Det resulterende kompleks av diffusjonskontroll- og matrikslag ble anbragt i kontakt med dermale kjøttsår.
Eksempel 19
Bioforenelighet av diffusjonskontroll- og matrikslag på dermale kjøttsår
Svamper og ark fremstilt i henhold til eksempel 18,
ble testet etter bestrålingssterilisering som forbindings-materialer på åpne dermale sår på hvite Hartley albino hunnmarsvin med en vekt på 350 g. Hvert dyr ble separat f6ret og veiet i 4 dager før testing. En dag før testing ble dyrene barbert under anvendelse av en elektrisk klipper etterfulgt av hårfjerningsbehandling med "Nair" og vasket. Dyret ble deretter bedøvet ved eksponering for ether, og ryggen ble vasket med povidon-jod og alkoholløsninger. En 2 cm x 2 cm bit av materiale sammensatt av matriks og diffusjonskontrollag oppbløtet i en fosfatbufferløsning (0,182 M fosfat og 0,154 M NaCl) ble anbragt på et dermalt kjøttsår med samme størrelse med matrikslaget mot panniculus carnosus. Matriks- og diffusjonskontrollagene ble sydd til sårskiktet ved kantene av forbindingen under anvendelse av kromgarvet tarmstreng. Dyret ble bandasjert ved at en steril bomulls-forbindingssvamp ble anbragt oversårforbindingen og deretter omviklet med en "Elasticon" elastisk tape (Johnson and Johnson Products, Inc.) festet rundt nakken. 3 dyr ble anbragt i et bur under forsøket.
Dyrene ble overvåket daglig, og deres bandasjer ble undersøkt med hensyn til flenger. Dyrene ble avlivet på den 6., 9. og 12. dag etter implantasjon, og implantatet og sårskiktene ble skåret ut, fiksert og bearbeidet for histo-logisk undersøkelse. Disse resultater er angitt i tabell III.
Utallige modifikasjoner og variasjoner av foreliggende oppfinnelse er mulig i lys av den ovenfor angitte lære, og oppfinnelsen kan således utøves på andre måter enn hva som spesielt er beskrevet, men innen rammen for de etterfølgende krav.
Claims (50)
- I 1. Fremgangsmåte for fremstilling av en ny collagen-basert matriks, karakterisert ved at a) et collagen valgt fra gruppen bestående av collagen av type I, II og III, blandes med et væskeformig medium under dannelse av et collagen-basert substrat;b) et tverrbindingsmiddel valgt fra gruppen bestående av et carbodiimid og en bifunksjonell succinimidyl-aktiv ester innføres i produktet fra trinn a); ogc) produktet fra trinn b) underkastes forhøyet temperatur under et vakuum, under dannelse av angitte collagen-baserte matriks.
- 2. Fremgangsmåte ifølge krav 1,karakterisert ved at trinn c) utføres før trinn b).
- 3. Fremgangsmåte ifølge krav 1 eller 2, karakterisert , ved at angitte væskeformige medium er et løsningsmiddel for angitte collagen.
- 4. Fremgangsmåte ifølge krav 1 eller 2, karakterisert ved at angitte væskeformige medium er et dispergeringsmiddel for angitte collagen.
- 5. Fremgangsmåte ifølge krav 1 eller 2,i karakterisert ved at tverrbindingsmidleter angitte carbodiimid og foreligger i løsning i en konsentrasjon på fra 0,1 til 10% (w/v).
- 6. Fremgangsmåte ifølge krav 5,karakterisert ved at carbodiimidløsningen og det collagen-baserte substrat opprettholdes ved en temperatur på fra 2 til 40°C i et tidsrom på fra 2 til 96 timer før ytterligere bearbeidelse.
