Description
Foreliggende oppfinnelse vedrører ricinolsyrederivater med formel som angitt i krav 1 og med agonistisk aktivitet mot vanilloidreseptor type-1.
BAKGRUNN FOR OPPFINNELSEN
Vanilloidreseptor type 1 (VRI eller TRPV1) tilhører den store familien av transiente reseptor potensielle kationkanaler (TRPV) med seks frans-membrandomener. TRPV1 er den eneste TRPV-kanalen som for tiden er kjent for å kunne aktiveres med naturlige produkter, capsaicin og resiniferatoksin er det mest kjente og studer-te (Sterner and Szallasi, Trends Pharmacol. Sei. 1999, 20, 459-465). Det er til i dag kjent at, mens andre TRPV-kanaler, slik som TRPV2, TRPV3 og TRPV4 (også kjent som "VRl-lignende (VRL) reseptorer") alene er ansvarlig for mekaniske, osmotiske eller termiske stimulanser og distribueres i prinsippet likt i de ulike vevene hos pat-tedyr, virker TRPV1 spesielt som en molekylær integrator av indusert smertestimu-lering, f.eks. ved varme, protoner og vegetabilske toksiner og uttrykkes hovedsak-lig i perifer sensortype C og A5 fibre (Gunthorpe et al., Trends Pharmacol. Sei. 2002, 23, 183-191).
VRl-Knockout-studier på transgene mus viste entydig rollen til TRPV1 i i delvis per-sepsjon og transmisjon av "termisk" eller "inflammatorisk" smerte (Caterina et al., Science 2000, 288, 306-313; Davis et al., Nature 2000, 405, 183-187). Andre studier foreslo at TRPV1 også er involvert i intestinale inflammatoriske forstyrrelser (Yiangou et al., Lancet 2001, 357, 1338-1339), nevorpatisk smerte (Walker et al., J. Pharmacol. Exp. Ther. 2003, 304, 56-62), fekal inkontinens og patologisk hoste (Chung and Chang, Pulm. Pharmacol. Ther. 2002, 15, 335-338). TRPV1 spiller ty-delig en fundamental rolle også i kontrollen av urinblærefunksjon (Birder et al., Nat. Neurosci. 2002, 5, 856-860) og i kontrollen av nevronal plastisitet, kropps-temperatur, matinntak, energiforbruk og bevegelse (Di Marzo et al., Eur. J. Pharmacol. 2001, 420, 123-131).
Nevroner som uttrykker TPRVl-reseptorer kan desensibiliseres rett etter aktivering av enkelte agonister, slik som capsaicin. Ut fra et praktisk synspunkt, overvinnes startbrenningsfølelsen grunnet agonistvirkningen ved en paradokseffekt. VR1-takyfylakse kan også forklare årsaken til andre medisinske effekter beskrevet for capsaicin og chilipepper, slik som de velkjente antiemetiske og antiinflammatoriske effektene og den nevrobeskyttende effekten mot glutamatekscitotoksisitet. Capsaicin og dens analoge resiniferatoksin anvendes også i behandlingen av urininkontinens (hvori nerveenderkarakterisert vednærværet av VR deltar i overførselen av den blæretømmende refleksen), mens capsaicin syntetiske derivater, det mest kjente er olvanil, har blitt patentert som orale analgetika. Den farmasøytiske indu-strien er imidlertid fremdeles spesielt interessert i utviklingen av mer potente TRPVl-agonister.

Chem., 2002, 45 (17), s. 3739-3745 er det kjent forbindelser med aktiveringspotensiale overfor vanilloidreseptor.
BESKRIVELSE AV OPPFINNELSEN
Forbindelsene ifølge oppfinnelsen har følgende generelle formel (I)
hvori
X representerer to hydrogenatomer, en n-binding, oksygen eller metylen;
R2 er et C6-C12-aryl- eller a ry la I kyl rest;
R3 er hydrogen, 2-hydroksyetyl eller 2-aminoetyl.
R2 er fortrinnsvis fenyl, benzyl eller fenetyl og R3 er fortrinnsvis hydrogen.
Disse forbindelsene er potente TPVRl-agonister og er derfor nyttige for behandlingen av smerte eller urininkontinens eller inflammatoriske tarmforstyrrelser.
Foreliggende oppfinnelse vedrører også farmasøytiske sammensetninger omfattende forbindelsene med formel (I) som aktive prinsipper i effektiv mengde. Forbindelsene ifølge oppfinnelsen kan være i racemisk eller enantiomerisk ren form, mer foretrukket 12R. Konfigurasjonen av dobbeltbindingen kan være E eller Z, mer foretrukket Z. De aktuelle forbindelsene kan også foreligge som farmakologisk akseptable salter av forbindelse med formel (I).
Forbindelsene ifølge oppfinnelsen kan syntetiseres f.eks. gjennom fremgangsmåtene rapportert under; andre reagenser og utgangsmaterialer kan velges for å oppnå andre forbindelser med formel (I).
Ifølge den enkleste syntetiske tilnærmelsen, fremstilles forbindelsene ifølge oppfinnelsen fra ricinolsyrevanillammid, ifølge skjema 1.
Ricinolsyrevanillamid, fremstilt i henhold til litteraturen, underkastes eventuelt cyk-lopropanasjon eller epoksidasjon og det resulterende intermediatet forestres med et passende acylerende middel. Aktiverte karboksylsyrederivater passende for fore-stringen er syrehalider, spesielt klorider, samt blandede anhydrider eller addukter med karbodiimid, ifølge fremgangsmåtene kjent i faget. Esteren av para-hydrok-sylet av vanillaminresiduet hydrolyseres deretter selektivt for å gi det tilsvarende hydroksyderivatet, som er et mål med foreliggende oppfinnelse. Om ønsket kan fenolhydroksylet deretter foretres med et aminoetyl eller et hydroksyetylresidu. Alternativt kan intermediatene nyttige for syntesen av forbindelsene ifølge foreliggende oppfinnelse oppnås også startende fra trikloretanolricinoleat ifølge skjema 2.
Trikloretanolricinoleat, fremstilt ifølge litteraturen, acyleres på 12-stillingen med en aktivert karboksylsyre, lignende det som er angitt over. Trikloretylesteren hydrolyseres deretter selektivt for å gi 12-acylricinolsyre som deretter omdannes til amid
ved kondensering med vanillamin.
De samme fremgangsmåtene kan med hell anvendes for trans- og 12-S-isomerer, eller for de mettede analogene av naturlig ricinolsyre.
Den biologiske aktiviteten av produktene ifølge oppfinnelsen ble målt både in vitro og in vivo.
In vitro-effekten av forbindelsene med formel (I) på TRPV1 ble studert ved å måle konsentrasjonen av intracellulært kalsium i HEK-293-celler (som overuttrykker hu-mant TRPV1); kalsiumkonsentrasjon i nærværet av 4 uM ionmycin ble tatt som den maksimale referanseverdien (Hayes et al., Pain 2000,88,205-215). Resultatene er rapportert i etterfølgende tabell 1.
Aktiviteten til forbindelsen ifølge eksempel 2 ble også evaluert i rotter gjennom in wVo-testen av indusert urininkontinens.
Sprague Dawley-rotter (250 ± 10 g) ble anestisert med kloralhydrat. Urinblæren ble åpnet gjennom et snitt langs abdomenmedianlinjen, og fettvevet som omgir urinrøret og urinlederne ble fjernet. Det proksimale urinrøret ble deretter festet med ikke-absorberende kirurgitråd for å danne delvis urinrørsobstruksjon. Etter 8 uker ble forandring av blærefunksjonalitet observert og det cystometriske mønste-ret ble forandret, antallet vannlatinger per time økte spesielt.
På behandlingsdagen ble forbindelsen ifølge eksempel 2, forbindelsen fremstilt iføl-ge Appendino et al. og resiniferatoksin som referansestandarden oppløst i etanol og dryppet i blæren ved konsentrasjon på 50 nM. Dryppingen varte 30 min: i løpet av denne perioden ble infusjonspumpen for saline skrudd av. Etter inkubasjon ble blæ ren tømt for enhver medikamentløsning, deretter ble infusjonspumpen skrudd på igjen.
En vesentlig økning i antallet vannlatinger per time ble observert i ikke-behandlede opererte dyr. Derimot, i dyr behandlet med forbindelsen ifølge oppfinnelsen og med resiniferatoksin forble antallet vannlatinger likt det for ikke-opererte dyr. Svært lav aktivitet ble også observert også for produktet til Appendino et al.
Resultatene av eksperimentet er rapportert i den etterfølgende tabell 2.
Som potente agonister av TPRVl-reseptoren kan forbindelser (I) anvendes i behandlingen av urininkontinens, i lindringen av nevropatisk smerte og i terapien av inflammatoriske tarmforstyrrelser.
Oppfinnelsen vedrører også farmasøytiske sammensetninger omfattende forbindelser (I) i kombinasjon med passende bærere eller fortynningsmidler. De farmasøy-tiske sammensetningene ifølge oppfinnelsen kan administreres gjennom ulike ad-ministrasjonsruter, f.eks. gjennom den orale, rektale, intravenøse, intramuskulære, subkutane, intrathekale, epidurale eller intracerebroventrikulære ruten. Passende bærere for injiserbare formuleringer omfatter oljer, propylen- eller etylenglykol, fysiologisk løsning, etanol, vegetabilske oljer og isopropylmyristat eller andre løse-midler vanligvis anvendt for fremstillingen av injiserbare løsninger.
For å fremstille injiserbare formuleringer kan forbindelsene ifølge oppfinnelsen lø-ses, suspenderes eller emulgeres i vandige løsemidler, slik som fysiologisk løsning, 5 % dekstrose eller ikke-vandige løsningsmidler slik som vegetabilske oljer, mettede syntetiske glyserider, estere av langkjede alifatiske syrer eller propylenglykol. Formuleringene kan også inkludere passende eksipienser, slik som løsningshjelpe- midler, isotoniske midler, suspenderingsmidler, emulgatorer, stabilisatorer og kon-serveringsmidler.
For topisk anvendelse kan forbindelsene ifølge oppfinnelsen formuleres som kremer eller salver.
Den farmasøytiske sammensetningen av oppfinnelsen kan anvendes:
for å lindre smerte forårsaket av post-herpeslignende nevralgi, diabetisk nevropati, post-mastektomisyndrom, sympatisk refleksdystrofi, tvillingnervenevral-gi, oral nevropatisk smerte, osteoartritt, revmatoid artritt, fibromyalgi, Guillain-Barré-syndrom;
for å lindre smerte som ikke kan behandles forårsaket av bilateral perifer nevropat;
for å lindre kløe forårsaket av psoriasis, hemodialyse, aquagenisk kløe, vul-va-vestibulitt, notalgia paresthetica, underarmskløe;
for å behandle gruppehodepine, vasomotorisk rhinitt eller allergisk rhinitt (som intranasale dråper);
for å behandle blærehypersensibilitet eller ryggmargsdetrusorhyperrefleksi (som intrablæreløsning).
Forbindelsene ifølge foreliggende oppfinnelse har potent analgesisk effekt og mulig antiinflammatorisk aktivitet, og de farmasøytiske formuleringene inneholdende dem kan anvendes for å lindre eller behandle akutt eller kronisk inflammatorisk smerte, inflammasjon og tranginkontinens.
Forbindelsene ifølge oppfinnelsen kan anvendes i form av farmakologisk akseptable salter, alene eller i passende kombinasjon, eventuelt i tilsetning med andre aktive ingredienser.
Dosen av forbindelsene ifølge oppfinnelsen varierer avhengig av tilstandene og pa-sientens vekt, sykdommens alvorlighet, farmasøytisk form, administrasjonsrute og varighet, og kan etableres av ekspertklinikeren. I prinsipp vil dosen strekke seg fra 0,1 ug til 100 mg/kg, fortrinnsvis fra 1 ug til 100 mg/kg/die. Preparatet kan admi nistreres som enkel eller gjentatt dose. Forbindelsesprosentdelene i sammensetningene kan strekke seg fra 0,0001 til 10 vekt%, fortrinnsvis fra 0,0001 til 1 % basert på sammensetningens vekt.
Følgende eksempler illustrerer oppfinnelsen mer detaljert.
Eksempel I - 12, 4'- difenylacetylrinvanil
En løsning av 1,56 g rinvanil (3,6 mmol) i toluen (20 ml) tilsettes med 2 ekv. mol fenyleddiksyre (1,0 g, 7,2 mmol), 2 ekv. mol dicykloheksylkarbodiimid (1,45 g, 7,2 mmol) og 1 ekv. mol DMAP (440 mg, 3,6 mmol). Reaksjonen settes under omrø-ring ved romtemperatur og overvåkes med TLC (6:4 petroleumseter/etylacetat Rfp= 0,31; Rfa= 0,60). Etter 3 timer filtreres blandingen og løsemidlet inndampes. Det resulterende råstoffet kan enten anvendes som sådan for det etterfølgende trinnet eller gjenvinnes med kolonnekromatografi.

Eksempel II - 12- Fenvlacetvlrinvanil
Det rå 12,4'-difenylacetylrinvanil (3,6 mmol, beregnet) av eksempel I løses i diklormetan (20 ml) behandles med 5 ekv. mol pyrrolidin (1,52 ml, 1,30 g, 18,0 mmol). Reaksjonen omrøres magnetisk ved romtemperatur og overvåkes med TLC (6:4 petroleumseter/etylacetat Rfp=0,8; Rfa=0,5). Etter 3 timer opparbeides reaksjonen ved vasking med 2N H2S04og saltlake. Den organiske fasen tørkes (Na2S04), filtreres og inndampes, og residuet renses med kolonnekromatografi
(37 g silikagel, pakket med 7:3 petroleumseter/etylacetat eludert med 6:4, 4:6; fraksjoner på omkring 20 ml samles opp). 1,6 g (80 %) fenylacetylrinvanil oppnås. Forbindelsen er en olje ved romtemperatur, men i fryseren stivner den til et hvitt pulver.

Eksempel III - 2', 2', 2'- trikloretvlricinoleat
3 g ricinolsyre (M.W. =298,47; 10,07 mmol) løses i 30 ml toluen, tilsettes med 2 ekv. mol trikloretanol (M.W. = 149,40; 20,14 mmol; 3,0 g; d = l,55; 1,9 ml), 1 ekv.
mol dicykloheksylkarbodiimid (M.W.= 202; 10,07 mmol; 2,0 g) og 1 ekv. mol DMAP (M.W.= 122; 10,07 mmol; 1,23 g). Den resulterende blandingen omrøres magnes-tiskved romtemperatur og reaksjonen overvåkes med TLC (8:2 heksan/hetylacetat Rfp= 0,14; Rfa= 0,53). Etter 18 timer filtreres blandingen og løsemidlet inndampes. Råstoffet renses med filtrering på silikagel, ved anvendelse av 9:1 petroleumseter/etylacetat som eluent. 4,3 g produkt (kvantitativt utbytte) oppnås.

Eksempel IV - 2', 2', 2'- Trikloretvl 12- fenvlacetvlricinoleat
4,3 g 2',2',2'-trikloretylricinoleat (10,7 mmol) løses i 30 ml toluen og tilsettes med 2,5 ekv. mol fenyleddiksyre (3,4 g, 25,2 mmol), 2,5 ekv. mol dicykloheksylkarbo-
diimid (5,0 g, 25,2 mmol) og 1,5 ekv. mol DMAP (1,8 g, 15,0 mmol). Blandingen omrøres ved romtemperatur og reaksjonen overvåkes med TLC (alumina, petroleumseter/etylacetat 8:2, Rfp=0,50; Rfa=0,76). Etter 30 minutter filtreres dicykloheksylurea fra og løsemidlet inndampes for å oppnå et råstoff, som deretter renses med kolonnekromatografi (35 g aluminagel, 95:5 petroleumseter-etylacetat, fraksjoner: omkring 20 ml). 4,6 g produkt oppnås (84 %).

Eksempel V - 12- fenvlacetvlricinolsvre
4,6 g (8,4 mmol) 2',2',2'-trikloretyl 12-fenylacetylricinoleat løses i 40 ml av en 1:1 eddiksyre/MeOH-løsning, tilsettes deretter med 4,6 g aktivert sinkpulver under sterk omrøring, og reaksjonen overvåkes med TLC (petroleumseter/etylacetat 8:2; Rfp=0,69; Rfa=0,36). Etter 18 timer filtreres blandingen gjennom kiselgur, vasking med etylacetat. Filtratet konsentreres, vaskes med vann og med en natriumbikar-bonatmettet løsning, tørkes deretter over natriumsulfat, filtreres og inndampes. Råstoffet renses med kolonnekromatografi (60 g silikagel, 95:5 petroleumseter-etylacetat, fraksjoner: omkring 20 ml). 1,85 mg produkt (53 %) oppnås.

Eksempel VI - 12- fenvlacetvlrinvanil
1,85 g 12-fenylacetylricinolsyre løses (4,4 mmol) i 15 ml tørr diklormetan og tilsettes med 2 ekv. mol vanillaminhydroklorid (835 mg, 4,4 mmol), 4 ekv. mol TEA (2,45 ml, 1,78 g, 17,6 mmol) og 1,2 ekv. mol polyfosforsyre (50 % EtOH-løsning, 3,4 ml, 1,68 g, 5,28 mmol). Reaksjonen omrøres ved romtemperatur og overvåkes med TLC (6:4: Rfp= 0,67; Rfa=0,37). Etter 3 timer inndampes løsemidlet og råstoffet renses med kolonnekromatografi (50 g silikagel, eluert med 7:3 petroleumseter/etylacetat, fraksjoner: omkring 20 ml). Produktet renses ytterligere ved filtre-
ring gjennom alumina (6:4 til 4:6 petroleumseter/etylacetat). 512 mg fenylacetylrinvanil oppnås (23 %).
Eksempel VII - 2', 2', 2'- trikloretvl 12- benzovlricinoleat
200 mg 2',2',2'-trikloretylricinoleat (M.W.=429,85; 0,46 mmol) løses i 2 ml toluen og tilsettes med 1 ekv. mol benzosyre (M.W.= 122,12; 0,46 mmol, 56 mg), 1 ekv. mol dicykloheksylkarbodiimid (M.W.=206,33; 0,46 mmol, 95 mg) og 1 ekv. mol DMAP (M.W. = 122,17; 0,46 mmol, 56 mg). Blandingen omrøres ved romtemperatur og reaksjonen overvåkes med TLC (95:5 heksan/etylacetat, Rfp= 0,05; Rfa=0,32). Etter 30 minutter tilsettes én eller flere ekvivalenter av benzosyre, dicykloheksylkarbodiimid og DMAP. Selv om reaksjonen ikke er fullført, opparbeides den etter ytterligere 18 timer omrøring: dicykloheksylurea filtreres og filtratet inndampes. Råstoffet renses med kolonnekromatografi (5 g silikagel, 95:5 heksan/etylacetat, fraksjoner på omkring 5 ml ble samlet opp). 195 mg produkt oppnås (79 %).

Eksempel VIII - 12- benzovlricinolsvre
185 mg 2',2',2'-trikloretyl 12-benzoylricinoleat (M.W. = 533,95; 0,35 mmol) løses i 2 ml av en 1:1 eddiksyre/MeOH-løsning, tilsettes deretter 200 mg aktivert sinkpulver under sterk omrøring og reaksjonen overvåkes med TLC (8:2 heksan/etylacetat; Rfp=0,42; Rfa=0,17). Etter 2 timer inndampes løsemidlet og råstoffet renses med kolonnekromatografi (2,5 g silikagel, 8:2 petroleumseter/etylacetat, fraksjoner: omkring 5 ml). 31 mg benzoylrinvanil (38 %) oppnås.

Eksempel X - 9, 10- metylrinvanil
I en tohalset rundbunnet kolbe og under nitrogenatmosfære, løses 300 mg rinvanil (M.W.=433,62; 0,69 mmol) i 29 ml vannfritt toluen (29 ml) og behandles med 15 ekv. mol dietylsink (1,0 M i heksan; 10,35 mmol; 10,35 ml) og 15 ekv. mol bis-iodometan (M.W.=268,84; 10,35 mmol; 2,78 g; d=3,325g/ml; 837 pl). Løsningen omrøres ved 65 °C og blir rose når et hvitt faststoff begynner å presipitere. Reaksjonen overvåkes med TLC på sølvsilika (6:4 petroleumseter/etylacetat; Rf p=0; Rfa=0,l).
Etter 7 timer avkjøles blandingen til 0 °C, tilsettes med 2N H2S04og ekstraheres med etylacetat, deretter vaskes det organiske sjiktet med NaHC03og saltlake. Et ter tørking (Na2S04) og inndamping renses råstoffet med kolonnekromatografi (15 ml silika, pakkes med petroleumseter/etylacetat 7:3 og elueres med en 6:4 blan-ding av de samme løsemidlene; fraksjoner: omkring 8 ml). 119 mg produkt (40 %) oppnås.

100 mg 9,10-metyl rinvanil (M.W.=447,65; 0,22 mmol) løses i 2 ml toluen og tilsettes med 2 ekv. mol fenyleddiksyre (M.W. = 136; 0,44 mmol; 60 mg), 2 ekv. mol dicykloheksylkarbodiimid (M.W.= 202; 0,44 mmol; 89 mg) og 1 ekv. mol DMAP (M.W.= 122; 0,22 mmol; 27 mg). Blandingen omrøres ved romtemperatur og reaksjonen overvåkes med TLC (6:4 petroleumseter/etylacetat Rfp=0,23; Rfa= 0,42). Etter 3 timer fjernes dicykloheksylurea ved filtrereing og løsemidlet inndampes. Det resulterende råstoffet kan anvendes som sådan for den etterfølgende reaksjonen eller renses med kromatografi (5 g silikagel, 8:2 petroleumseter/etylacetat som eluent).

Eksempel XII - 9, 10- metylen- 12- fenvlacetvlrinvanil I
Råstoffet 9,10-metylen-12,4'-difenylacetylrinvanil fra eksempel XI (0,22 mmol, teoretisk) løses i diklormetan (20 ml) og tilsettes med 5 ekv. mol pyrrolidin (M.W.=71,12; 1,1 mmol; 78 mg; d=0,86 g/ml; 90 pl). Etter 3 timer er reaksjonen fullført (TLC over alumina, eluent: 6:4 petroleumseter/etylacetat; Rfp=0,8; Rfa=0,5; Rfb=0,45; TLC på silika med den samme eluenten: Rfp=0,42; Rfa=0,39; kun en flekk vises). Det organiske sjiktet vaskes med 2N H2S04og saltlake og tør-kes over natriumsulfat. Etter inndamping av løsemidlet, renses råstoffet ved kolonnekromatografi (10 ml silica, 7:3 til 4:6 petroleumseter/etylacetat). 75 mg produkt (60 %) oppnås.

Eksempel XIII - TPRVl- bindinqsassay
Affinitet med human TRPV1 ble målt ved utskifting av [3H]RTX (48 Ci/mmol, NEN-Dupont) fra HEK cellemembraner (50 pg/rør) ifølge fremgangsmåten beskrevet av Ross (Ross et al., Br. J. Pharmacol. 2001, 132, 631-640). Under disse betingelsene var Kd og Bmax for [<3>H]RTX 0,5 nM og 1,39 pmol/mg protein. Ki av utskiftingen av 1 nM [3H]RTX ble beregnet fra IC50-verdien (oppnådd med GraphPad programvar) ved anvendelse av Cheng-Prusoffs ligning. Spesifikk binding ble beregnet med 1 pM RTX (Alexis Biochemicals) og var 48,1+5,6 %. Verdiene for forbindelsene av oppfinnelsen er rapportert i tabell 1.
Eksempel XIV - 4,-( 2- aminoetvl)- 12- fenylacetvlrinvanil ( hvdroklorid)
I en tohalset rundbunnet kolbe og under nitrogenatmosfære, tilsettes en løsning av 382 mg fenylacetylrinvanil (M.W.=552; 0,69 mmol) løst i THF (5 ml) til en suspen-sjon av NaH (60 %, 57 mg, 1,4 mmol, 2 molarekvivalenter) i THF (10 ml). Etter omrøring ved romtemperatur i 10 min, tilsettes et overskudd av 1,2-dibrometan (0,7 ml). Løsningen omrøres ved romtemperatur i 16 timer, fortynnes deretter med mettet NH4CI og ekstraheres med eter. Inndamping av løsemidlet gir en olje, som filtreres gjennom et tynt silikasjikt (15 g) ved anvendelse av petroleumseter (100 ml) for å fjerne overskuddet av 1,2-dibromoetan, deretter 1:1 petroleumseter/etylacetat (100 ml) for å eludere produktet. Etter inndamping av løsemidlet, oppnås et klebrig residu, som løses direkte i dimetylformamid (10 ml) og behandles med et overskudd av natriumazid (NaN3) (300 mg). Etter omrøring i 16 timer, fortynnes reaksjonen med vann (omkring 50 ml) og ekstraheres med 3:1 petroleumseter/eter (2 x 30 ml). Etter vasking med mettet NaCI og tørking (Na2S04), inndampes det organiske sjiktet og residuet løses i THF (20 ml), tilsettes deretter med trifenylfosfin (917 mg, 3,5 mmol) og vann (0,62 ml, 3,5 mmol). Etter omrøring i 5 timer ved romtemperatur, fortynnes reaksjonen med vann og ekstraheres med etylacetat. Etter tørking (Na2S04) og inndamping, renses residuet ved kromatografi på en si I i— kagelkolonne (10 g), ved anvendelse av etylacetat som eluent. 130 mg produkt oppnås (samlet utbytte: 40 %).

Hydrokloridsaltet oppnås ved å løse produktet i minimumsmengden av THF og til-sette 1 ekvivalent av en 1,0 M hydrokloridsyreløsning i dietyleter ved 0 °C. Etter inndamping av løsemidlet samles presipitatet og tørkes under vakuum.
Eksempel XV - 9, 10- epoksv- 12- fenvlacetvlrinvanil
En løsning av 12,4'-difenylacetylrinvanil fremstilt som i eksempel 1 (300 mg, 0,45 mmol) i vannfri CH2CI2(6 ml) tilsettes med 2,5 molare ekvivalenter av meta-klor-perbenzosyre (MCPBA, 242 mg av 80 % syre, 1,12 mmol), og løsningen omrøres magnetisk i 3 timer, vaskes deretter med Na2S203, tørkes (Na2S04), filtreres og inndampes. Residuet løses direkte i CH2CI2(5 ml) og tilsettes med pyrrolidin (155 mg, 0,180 ml, 5 molar ekv.). Etter omrøring ved romtemperatur i 16 timer, vaskes blandingen med 2N H2S04og mettet NaCI, tørkes deretter (Na2S04). Etter inndamping av løsemidlet renses residuet ved kolonnekromatografi på nøytral alumina (3 g, 6:4 petroleumseter/etylacetat som eluent), for å oppnå 120 mg produkt (41 %).

4'-(2-aminoetyl)-12-fenylacetylrinvanil*HCI injiserbar løsning består av 4'-(2-aminoetyl)-12-fenylacetylrinvanil*HCI (1,0 mg), natriumklorid (9,0 mg), benzylalkohol (15,0 mg) og vann injiserbare preparater (opp til 1 ml).
I standardprosedyren løses natriumklorid og benzylalkohol i vann for injiserbare preparater, deretter tilsettes 4'-(2-aminoetyl)-12-fenylacetylrinvanilhydroklorid.
Eksempel XVII - 12- fenylacetylrinvanil iniiserbar løsning
12-fenylacetylrinvanil injiserbar løsning består av 12-fenylacetylrinvanil (1,0 mg), propylgallat (0,5 mg), olivenolje for injiserbare preparater (opp til 1 ml).
Eksempel XVIII - Emulsjon for topisk anvendelse
Emulsjoner for topisk anvendelse, f.eks. 12-fenylacetylrinvanilemulsjoner, består av 12-fenylacetylrinvanil (1,0 g), flytende parafin (25,0 g), stearylalkohol (12,0 g), cetylalkohol (5,0 g), metyl-p-hydroksybenzoat (0,028 g), propyl-p-hydroksybenzoat (0,012 g), PEG-40-stearat (1,0 g), glyserin (12,0 g), renset vann (opp til 100 g).
I standardprosedyren smeltes flytende parafin, stearylalkohol og cetylalkohol ved 70-75 °C under omrøring, deretter løses 12-fenylacetylrinvanil i det resulterende sjiktet, løsningen holdes ved 70-75 °C. De gjenværende komponentene, tidligere løst i renset vann ved 70-75 °C, tilsettes deretter under sterk omrøring ved 70-75 °C. Det resulterende produktet avkjøles langsomt under omrøring.

4. Anvendelse av forbindelsene ifølge kravene 1-3 for fremstilling av medika-menter for behandling av urininkontinens, for lindring av nevropatisk smerte og for terapi av inflammatoriske forstyrrelser i tarmen.
5. Sammensetninger inneholdende én eller flere av forbindelsene ifølge et hvil-ket som helst av kravene 1-3, i tilsetning med passende bærere.
