Abstract
Den foreliggende oppfinnelse beskriver indeno[2,lalinden- og isoindolo[2,la]indolderivater av formel (I), dens salter og stereoisomerer, hvori X, R,, R2, R3, R4, R6, R7, R8, Rg, R,,, R13 · og R14 er som beskrevet i spesifiseringen. Oppfinnelsen tilveiebringer også en fremgangsmåte for å forberede forbindelsen med formel (I), en farmasøytisk forbindelse inneholdende slike forbindelser og metode for å fremstille et medikament. Foreliggende oppfinnelse angår en forbindelse med den generelle formel (I) som vist nedenfor,
Forbindelser med formel (I) er brukbare ved behandling av forskjellige forstyrrelser som involverer 5-HT reseptorer (serotonin) (WO 03/065046 A2), fortrinnsvis de som har en diskriminatorisk profil og sterk affinitet mot den spesielle reseptor. Mediering av 5-ht6reseptorer er foreslått for behandling av forskjellige CNS forstyrrelser, hematologiske forstyrrelser, spiseforstyrrelser, sykdommer forbundet med smerte, respiratoriske sykdommer, genito-urologiske forstyrrelser, kardiovaskulære sykdommer og cancer.
Disse forbindelser kan formuleres til forskjellige doseringsformer hvorved en effektiv mengde kan avleveres til pasienten som trenger det, enten for å oppnå en terapeutisk eller diagnostisk fordel.
WO 2004/000205, WO 2004/000845, WO 2004/000849, WO 2004/055026 Al, WO 2004/048331 Al, WO 2004/048330 Al og WO 2004/048328 A2 (alle i navnet Suven Life Sciences Limited) beskriver noe av den kjente teknikk.
WO 03/066056 Al rapporterer at antagonisme av 5-HT6-reseptor kan fremme neu-ronal vekst i sentralnervesystemet hos et pattedyr. Videre beskriver WO 03/065046 A2 nye varianter av 5-HT6-reseptoren og foreslår at human 5-HT6-reseptor assosieres med hematologiske forstyrrelser, smertesykdommer, respiratoriske sykdommer, genito-urologiske forstyrrelser, kardiovaskulære sykdommer og cancer.
Det er et av hovedmålene for foreliggende oppfinnelse å tilveiebringe en forbindelse med formel (I),

En annen gjenstand for oppfinnelsen er å tilveiebringe en forbindelse med formel (I) som kan være agonist, partialagonist eller antagonist av 5-HT reseptorsubtypene.
I henhold til dette tilveiebringer oppfinnelsen en forbindelse med den generelle formel

Foreliggende oppfinnelse angår også en fremgangsmåte for fremstilling av forbindelse med formel (I) som inkluderer:

Oppfinnelsen angår også anvendelsen av en forbindelse som definert ovenfor fremstilling av et medikament for behandling av forstyrrelser i sentralnervesystemet.
Oppfinnelsen angår også medikamenter og deres fremstilling i forskjellige doseringsformer inneholdende minst en forbindelse med formel (I).
Egnede grupper som representeres ved substituenter som Ri, R2, R3, R4, R5, R6, R7, Rs, R9, Rio, Ru, R12, R13og R14kan, der det er mulig, være valgt blant: halogen som fluor, klor, brom eller jod; substituerte eller usubstituerte (Ci-C3)alkylgrupper, slik som metyl, etyl, 2-klorprop-l-yl, isopropyl og liknende; (Ci-C3)alkoksy slik som metoksy, etoksy, propyloksy; syklo(C3-C5)alkoksygrupper slik som syklopropyloksy, syklobuty-loksy, syklopentyloksy, der sykloalkoksygruppen kan være substituert;.
Egnede, farmasøytisk akseptable syreaddisjonssalter av forbindelser med formel (I) kan fremstilles. De ikke-toksiske syreaddisjonssalter inkluderer de som har farmakologisk akseptable anioner som kloridet, bromidet, jodidet, nitratet, sulfatet, bisulfatet, fosfatet, det sure fosfat, acetatet, laktatet, ciratet, det sure ciratet, tartratet, bitartra-tet, suksinatet, maleatet, fumaratet, glukonatet, sakkaratet, benzoatet, metansul-fonatet, etansulfonatet, benzensulfonatet, p-toluensulfonatet, palmoatet og oksalatet. Farmasøytisk akseptable salter som utgjør en del av oppfinnelsen er ment å være definert av men ikke begrenset til listen ovenfor.
I tillegg kan farmasøytisk akseptable salter av forbindelsen med den generelle formel (I) oppnås ved å konvertere derivater som har tertiære aminogrupper til de tilsvarende, kvaternære ammoniumsalter i metodene som velkjent i litteraturen ved bruk av kvaterniserende midler. Mulige, kvaterniserende midler er for eksempel alkylhalider som metyljodid, etylbromid og n-propylklorid inkludert arylalkylhalider som benzylklo-rid og 2-fenyletylbromid.
De farmasøytisk akseptable salter som utgjør en del av oppfinnelsen kan fremstilles ved behandling av forbindelsen med formel (I) med 1-6 ekvivalenter av en syre som nevnt ovenfor i oppløsningsmidler som vann, alkoholer, etere, etylacetat, dioksan, DMF og et lavere alkylketon som aceton, eller blandinger derav.
Videre kan de eksepsjonelle salter dannes som mellomprodukter under rensing, fremstilling av andre salter, eller identifisering og karakterisering av forbindelser med formel (I) eller intermediater som er involvert i fremstilling av forbindelser med formel
(I).
De farmasøytisk akseptable salter av forbindelser med formel (I) kan foreligge som solvater, for eksempel med vann, metanol, etanol, dimetylformamid, etylacetat og liknende. Kilden for et slikt solvat kan være fra oppløsningsmidlet for krystallisering, inherent ved oppløsningsmiddelfremstilling eller -krystallisering, eller i forbindelse med et slikt oppløsningsmiddel.
Det kan være nødvendig å fremstille radiomerkede forbindelser relatert til den generelle struktur (I). Egnede isotoper som kan fremstilles ved innarbeiding av isotoper av hydrogen, karbon, nitrogen, oksygen, fosfor, fluor, klor, jod, brom og mTeknitium, eksemplifiseres ved<2>H,3H,nC,<13>C,<14>C, 1<3>N, 15N,<15>0,<18>F,<99m>Tc,<31>P, S,<123>I og 125I. Disse forbindelser som inneholder de ovenfor nevnte isotoper og/eller andre isotoper av andre atomer ligger innenfor oppfinnelsens ramme. Isotopisk merkede forbindelser ifølge oppfinnelsen er populære i medikament- og/eller substratvevfordeling og mål-okkuperingsanalyser. For eksempel er isotopisk merkede forbindelser spesielt nyttige ved SPECT (enkelt fotoemisjonsberegnet tomografi) og ved PET (positronemisjons-tomografi).
En effektiv mengde av en forbindelse med den generelle formel (I) eller deres derivater som definert ovenfor, kan benyttes for å fremstille et medikament, sammen med konvensjonelle, farmasøytiske hjelpestoffer, bærere eller additiver. 'Terapeutisk effektiv mengde" er definert som en mengde av en forbindelse ifølge oppfinnelsen som (i) behandler eller forhindrer den spesielle sykdom, tilstand eller forstyrrelser, (ii) svekker, lindrer eller eliminerer ett eller flere symptomer av den spesielle sykdom, tilstand eller forstyrrelse, eller (iii) forhindrer eller forsinker starten av ett eller flere symptomer av den spesielle sykdom, tilstand eller forstyrrelse som definert heri.
Slik terapi inkluderer flere valg: for eksempel administrering av to kompatible forbindelser samtidig i en enkelt doseform eller administrering av hver forbindelse individu-elt i en separat dosering; eller, hvis nødvendig, ved samme tidsintervall eller separat for å maksimalisere den fordelaktige effekt eller å minimalisere potensielle bivirk-ninger av medikamentene, i henhold til kjente, farmakologiske prinsipper.
Uttrykket "farmasøytisk akseptabel" indikerer at substansen eller preparatet må være kompatibelt kjemisk og/eller toksikologisk, med de andre bestanddeler omfattende en formulering og/eller pattedyret som behandles med denne.
Uttrykket "behandling", eller "å behandle" eller tilsvarende uttrykk, omfatter alle be-tydninger som preventiv, profylaktisk og palliativ.
De farmasøytiske preparater ifølge oppfinnelsen kan formuleres på konvensjonell måte ved bruk av en eller flere farmasøytisk akseptable bærere. Således kan de aktive forbindelser ifølge oppfinnelsen formuleres for oral, bukkal, intranasal, parental (for eksempel intravenøs, intramuskulær eller subkutan) eller rektal administrering eller en form som er egnet for administrering ved inhalering eller insufflering.
De farmasøytiske preparater så vel som de formulerte medikamenter som fremstilles ifølge oppfinnelsen kan i tillegg til minst en forbindelse med formel (I) omfatte ytterligere, konvensjonelle hjelpestoffer som er velkjente for fagfolk på området som bærere, fyllstoffer, solventer, diluenter, fargestoffer, beleggsmidler, matriksmidler og/eller bindemidler. Slik det er velkjent for fagfolk på området er valget av hjelpestoffer og mengdene av disse som skal benyttes avhengig av den tilsiktede administreringsvei, for eksempel den orale, rektale, intravenøse, intraperitoneale, intramuskulære, intra-nasale, bukkale eller topiske vei.
Dosen av den aktive forbindelse kan variere avhengig av faktorer som administreringsvei, pasientens alder og vekt, arten og alvoret av sykdommen som skal behandles og liknende faktorer. Derfor henviser enhver referanse her til en farmakologisk effektiv mengde av forbindelsene med den generelle formel (I) til de ovenfor nevnte faktorer. En foreslått dose av de aktive forbindelser ifølge oppfinnelsen for enten oral, parenteral, nasal eller bukkal administrering, til et gjennomsnittlig voksent menneske, for behandling av tilstandene som nevnt ovenfor, er 0,1 til 200 mg av den aktive bestanddel per enhetsdose som kan administreres, for eksempel 1 til 4 ganger per dag.
Medikamenter som er egnet for parenteral, topisk eller inhalatorisk administrering kan fortrinnsvis velges fra gruppen bestående av oppløsninger, suspensjoner, lett rekonsti-tuerbare, tørre preparater og også spray.
Egnede medikamenter, for eksempel medikamenter for oral eller perkutan bruk kan frigi sulfonamidforbindelsene med den generelle formel (I) på forsinket måte hvorved fremstillingen av disse medikamenter for forsinket frigivning generelt er kjente for fagfolk på området.
Medikamenter som er egnet for oral administrering er for eksempel tabletter, sukker-belagte piller, kapsler eller multi parti kler som granuler eller pellets, eventuelt presset til tabletter, fylt i kapsler eller suspendert i en egnet væske, oppløsning eller suspensjon. Slike farmasøytiske preparater kan ha eksipienter som bindemidler (for eksempel pregelatinisert maisstivelse, polyvinylpyrrolidon eller hydroksypropylmetylcellulo-se); fyllstoffer som laktose, mikrokrystaMinsk cellulose eller kalsiumfosfat); smøre- midler som magnesiumstearat, talkum eller silika; disintegranter som potetstivelse eller natriumstivelsesglykolat; eller fuktemidler som natriumlaurylsulfat. Tablettene kan være belagt ved velkjente metoder på området. Flytende preparater for oral administrering kan ha form av for eksempel oppløsninger, siruper eller suspensjoner eller de kan presenteres som et tørrprodukt for konstituering med vann eller en annen egnet vehikkel før bruk. Slike flytende preparater kan fremstilles ved konvensjonelle midler med farmasøytisk akseptable additiver som suspensjonsmidler (for eksempel sorbitolsirup, metylcellulose eller hydrogenen, spiselig fett); emulgeringsmidler som lecitin eller akasia; ikke-vandige vehikler som mandelolje, oljeestere av etylalkohol; og preserveringsmidler som metyl- eller propyl p-hydroksybenzoater eller sorbinsyre.
For bukkal administrering kan preparatet ha form av tabletter eller lozenger formulert på i og for seg kjent måte.
De aktive forbindelser ifølge oppfinnelsen kan formuleres for parenteral administrering ved injeksjon, inkludert anvendelse av konvensjonelle kateteriseringsteknikker eller infusjon. Formuleringer for injeksjon kan presenteres i enhetsdoseformer, for eksempel i ampuller eller i multidosebeholdere med et tilsatt preserveringsmiddel. Prepara-tene kan ha former som suspensjoner, oppløsninger eller emulsjoner i olje- eller vandige vehikler, og kan inneholde formuleringsmidler som suspenderings-, stabiliserings-og/eller dispergeringsmidler. Alternativt kan den aktive bestanddel foreligge i pulver-form for rekonstituering med en egnet vehikkel, for eksempel sterilt, pyrogenfritt vann, før bruk.
De aktive forbindelser ifølge oppfinnelsen kan også formuleres i rektal preparater som suppositorier eller retensjonsenemaer, for eksempel inneholdende konvensjonelle suppositoriebaser som kakaosmør og andre glyserider.
For intranasal administrering eller administrering ved inhalering blir de aktive forbindelser ifølge oppfinnelsen hensiktsmessig avgitt i form av en aerosolspray fra en trykksatt beholder eller en forstøver, eller fra en kapsel ved bruk av en inhalator eller insufflator. Når det gjelder en trykksatt aerosol, kan et egnet drivmiddel, for eksempel diklordifluormetan, triklorfluormetan, diklortetrafluoretan, karbondioksid eller en annen gass, og doseringsenheten, bestemmes ved å tilveiebringe en ventil for å avgi en tildosert mengde. Medikamentet for trykksatt beholder eller forstøver kan inneholde en oppløsning eller suspensjon av den aktive forbindelse, mens det for en kapsel er foretrukket at innholdet foreligger i form av et pulver. Kapsler og patroner (for eksempel laget av gelatin) for bruk i en inhalator eller insufflator kan formuleres inne holdende en pulverblanding av en forbindelse ifølge oppfinnelsen og en egnet pulver-base som laktose eller stivelse.
Aerosolformuleringer for behandling av betingelsene som angitt ovenfor (for eksempel migrene) hos et midlere voksent menneske er fortrinnsvis anordnet slik at hver tildo-serte dose eller "puff" av aerosol inneholder 20 ug til 1 000 ug av forbindelsen ifølge oppfinnelsen. Den totale, daglige dose med en aerosol vil ligge innen området 100 ug til 10 ug. Administreringen kan foretas flere ganger daglig, for eksempel 2, 3, 4 eller 8 ganger daglig, gitt for eksempel 1, 2 eller 3 doser hver gang.

Kommersielle reagenser ble benyttet uten ytterligere rensing. Romtemperatur er 25-30 °C. Smeltepunktene er ikke ikke-korrigert. IR-spektra ble tatt opp ved bruk av KBr og i fast tilstand. Hvis ikke annet er sagt, ble alle massespektra gjennomført ved bruk av ESI-betingelser.lH NMR-spektra ble tatt opp ved 400 MHz på et brukerinstrument. Deuterert kloroform (99,8 % D) ble benyttet som løsemiddel. TMS ble benyttet som indre referansestandard. De kjemiske skiftverdier uttrykkes som deler-per-million(8)-verdier. En omrørt oppløsning av l-benzensulfonyl-3-(l-metylpyrrolidin-3-yl)-lH-indol (0,286 mol) ble tatt opp i en 100 ml trehalset rundkolbe sammen med 40ml N,N-dimetylacetamid, kaliumacetat (0,286 mol, 0,281 g) og diklorbis(tri-o-tolylfosfin)palladium (0,0143 mol, 0,0126 g). Reaksjonsblandingen ble holdt under nitrogenatmosfære og så varmet opp til 160 °C under omrøring i 16 timer. Etter ferdig reaksjon ble oversky-tende dimetylacetamid destillert av under redusert trykk og resten renset ved silika-gelkolonnekromatografi ved bruk av 20 % metanol i etylacetat som elueringsmiddel. Den endelige, ønskede forbindelse med den generelle formel (I) kunne renses videre ved fremstilling av deres syreaddisjonssalt som hadde smeltepunkt i området 167,8-173,1 °C.

Eksempel 15: Næringsinntaksmåling
Wistar hannrotter (100-270 g), tilveiebrakt fra National Institute of Nutrition, Hydera-bad, India, ble benyttet. Dyrene akklimatiseres i dyrefasiliteten i minst 7 dager før de underkastes enhver behandling. I løpet av dette tidsrom huses dyrene (i grupper på tre) i translucide bur og gis næring og vann ad libitum. Minst 24 timer før behandling-en starter tilpasses dyrene enkelthusbetingelser.
Den kroniske effekt av forbindelser med den generelle formel (I) på næringsinntaket i velforede rotter bestemmes så som følger. Rottene ble huset i deres enkelte hjembur i 28 dager. Etter denne periode ble rottene oralt dosert med et preparat omfattende en forbindelse med formel (I) eller et tilsvarende preparat (vehikkel) uten forbindelsen en gang per dag. Rotten gis næring og vann ad libitum. Den 7., 14., 21. og 28. dag set-tes rotten hen med på forhånd veid næring. Næringsinntaket og vektøkningen måles.
Eksempel 16: Tablett inneholdende en forbindelse med formel (I):
Bestanddelene kombineres og granuleres ved bruk av et egnet oppløsningsmiddel som etanol. Formuleringen blir deretter tørket og formet til tabletter (inneholdende rundt 20 mg aktiv forbindelse) med en egnet tabletteringsmaskin.
Eksempel 17: Preparat for oral administrering
Bestanddelene blandes og dispergeres i kapsler inneholdende rundt 100 mg hver idet en kapsel vil approksimere en total, daglig dose.
Eksempel 18: Flytende oral formulering
Bestanddelene blandes for å danne en suspensjon for oral administrering.
Eksempel 19: Parenteral formulering
Den aktive bestanddel oppløses i en andel av vann for injeksjon. En tilstrekkelig mengde natriumklorid tilsettes så under omrøring for å gjøre oppløsningen isotonisk. Oppløsningen tildannes til vektmålet med resten av vannet for injeksjon, filtreres gjennom et 0,2 mikronmembranfilter og pakkes under sterile betingelser.
Eksempel 20: Suppositorieformulering
Bestanddelene smeltes sammen og blandes i et dampbad og helles i former inneholdende til sammen 2,5 g.
Eksempel 21: Topisk formulering
Alle bestanddeler bortsett fra vann kombineres og varmes opp til 60 °C under omrø-ring. En tilstrekkelig mengde vann ved rundt 60 °C tilsettes så under heftig omrøring for å emulgere bestanddelene, og vann tilsettes q.s. rundt 100 g.
Eksempel 22: Bindingsanalyse for human 5HT6reseptor
Materialer og metoder:

Inkuberinqsbetinqelser:
Reaksjonene gjennomføres i 50 mM TRIS-HCI (pH 7,4) inneholdende 10 mM MgCI2,0,5 mM EDTA i 60 min ved 37 °C. Reaksjonen avsluttes ved hurtig vakuumfiltrering på glassfiberfiltre. Radioaktiviteten som fanges på filtrene bestemmes og sammenliknes med kontrollverdier for å sikre alle interaksjoner av en eller flere testforbindelser med den klonede serotonin - 5HT6bindingssete.
