Description
Oppfinnelsens bakgrunn
Foreliggende oppfinnelse er rettet mot nye taxaner som har eksepsjonell anvendbarhet som antitumormidler.
Taxanfamilien av terpener, hvorav baccatin III og taxol er medlemmer, har vært gjenstand for betydelig interesse både innen biologien og kjemien. Taxol selv anvendes som et kjemo-terapeutisk middel mot kreft og har et bredt spekter av tumor-inhiberende aktivitet. Taxol har en 2'R,3'S-konfigurasjon og den følgende strukturformel:

Colin et al. rapporterte i US patentskrift nr.
4 814 470 at visse taxolanaloger har en aktivitet som er signi-fikant større enn aktiviteten til taxol. Én av disse analogene, vanligvis henvist til som docetaxel, har den følgende strukturformel:
Selv om taxol og docetaxel er anvendbare kjemoterapeutiske midler, er det begrensninger på deres effektivitet, inkludert begrenset virkningsfullhet mot visse typer av kreft, og toksisitet for individer når de administreres ved forskjellige doser. Følgelig består det et behov for ytterligere kjemoterapeutiske midler med forbedret virkningsfullhet og mindre toksisitet.
Oppsummering av oppfinnelsen
Blant gjenstandene for foreliggende oppfinnelse er derfor tilveiebringelsen av taxaner som er like gode som taxol og docetaxel når det gjelder virkningsfullhet som antitumormidler, og med hensyn til toksisitet. Generelt har disse taxanene en annen estersubstituent enn formiat, acetat og heterosubstituert acetat i C-7, en hydroksysubstituent i C-10 og en rekke C-3'-substituenter.
Foreliggende oppfinnelse er derfor i korte trekk rettet mot taxanforbindelsene som sådanne, mot farmasøytiske preparater som omfatter taxanene og en farmasøytisk akseptabel bærer, og mot anvendelse av taxanene til fremstilling av orale farmasøytisk preparater for inhibering av tumorvekst hos pattedyr.
Nærmere beskrivelse av foretrukne utførelsesformer
Ved én utførelsesform av foreliggende oppfinnelse til-svarer taxanene ifølge foreliggende oppfinnelse formel (1):

Taxaner som har den generelle formel 1, kan fås ved behandling av et p-laktam med et alkoksid som har den tetra-sykliske taxankjerne og en C-13-metalloksidsubstituent, slik at det dannes forbindelser som har en p-amidoestersubstituent i C(13) (som beskrevet mer fullstendig av Holton, US patentskrift nr. 5 466 834), etterfulgt av fjerning av hydroksybeskyttelses-gruppene. p-laktamet har den følgende strukturformel (3): hvor P2 er en hydroksybeskyttelsesgruppe, og X3 og X5 er som tidligere definert, og alkoksidet har strukturformel (4):
hvor M er et metall eller ammonium, Pi0 er en hydroksybeskyttelsesgruppe, og R2, Rg, R7 og R14 er som tidligere definert.
Alkoksid 4 kan fremstilles fra 10-deacetylbaccatin III (eller et derivat derav) ved selektiv beskyttelse av C-10-hydroksylgruppen, og så forestring av C-7-hydroksylgruppen, etterfulgt av behandling med et metallamid. Ved én utførelses-form av foreliggende oppfinnelse beskyttes C(10)-hydroksylgruppen i 10-deacetylbaccatin III selektivt med en silylgruppe f.eks. under anvendelse av et silyamid eller bis-silylamid som silyleringsmiddel. Foretrukne silyleringsmidler omfatter tri-(hydrokarbyl)silyltrifluormetylacetamider og bis-tri(hydro-karbyl)silyltrifluormetylacetamider (hvor hydrokarbylresten er substituert eller usubstituert alkyl eller aryl) , slik som N,0-bis-(trimetylsilyl)trifluoracetamid, N,0-bis-(trietylsilyl)trifluoracetamid, N-metyl-N-trietylsilyltrifluoracetamid og N,0-bis-(t-butyldimetylsilyl)trifluoracetamid. Silyleringsmidlene kan anvendes enten alene eller i kombinasjon med en katalytisk mengde av en slik base som en alkalimetallbase. Alkalimetall-amider, slik som litiumamidkatalysatorer generelt, og litium-heksametyldisilazid spesielt, er foretrukket. Oppløsningsmidlet for den selektive silyleringsreaksjon er fortrinnsvis et eterisk oppløsningsmiddel, slik som tetrahydrofuran. Alternativt kan imidlertid andre oppløsningsmidler, slik som eter eller di-metoksyetan, anvendes. Temperaturen hvor den C(10)-selektive silylering utføres, er ikke begrenset til et snevert område. Generelt utføres den imidlertid ved 0 °C eller høyere.
Selektiv forestring av C(7)-hydroksylgruppen i et C(10)-beskyttet taxan kan oppnås ved å anvende hvilket som helst av mange forskjellige vanlige acyleringsmidler, inkludert, men er ikke begrenset til, substituerte og usubstituerte karboksyl-syrederivater, f.eks. karboksylsyrehalogenider, -anhydrider, -dikarbonater, -isocyanater og -halogenformiater. C(7)-hydroksylgruppen i det C(10)-beskyttede 10-deacetylbaccatin III kan f.eks. acyleres selektivt med dibenzyldikarbonat, diallyl-dikarbonat, 2,2,2-trikloretylklorformiat, benzylklorformiat eller et annet vanlig acyleringsmiddel. Generelt er acylering av C(7)-hydroksygruppen i et C(10)-beskyttet taxan mer effektiv og mer selektiv enn det C(7)-acyleringer av et slikt 7,10-di-hydroksytaxan som 10-DAB er; sagt på en annen måte er det en betydelig forskjell i reaktiviteten til de gjenværende C(7)-, C(13)- og C(1)-hydroksylgrupper så snart C(10)-hydroksylgruppen er blitt beskyttet. Disse acyleringsreaksjonene kan eventuelt utføres i nærvær eller fravær av en aminbase.
Derivater av 10-deacetylbaccatin III som har alterna-tive substituenter i C(2), C(9) og C(14), og fremgangsmåter for deres fremstilling er kjent innenfor teknikken. Taxanderivater som har acyloksysubstituenter som er forskjellige fra benzoyl-oksy i C(2), kan fremstilles f.eks. som beskrevet av Holton et al., US patentskrift nr. 5 728 725, eller Kingston et al.,
US patentskrift nr. 6 002 023. Taxaner som har acyloksy- eller hydroksysubstituenter i C(9) i stedet for keto, kan fremstilles som f.eks. beskrevet av Holton et al., US patentskrift nr. 6 011 056, eller Gunawardana et al., US patentskrift nr. 5 352 806. Taxaner som har en beta-hydroksysubstituent i C(14), kan fremstilles fra naturlig forekommende 14-hydroksy-10-deacetylbaccatin III.
Fremgangsmåter for fremstilling og oppløsning av p-laktamutgangsmaterialet er generelt vel kjent. For eksempel kan P-laktamet fremstilles som beskrevet av Holton, US patentskrift nr. 5 430 160, og de resulterende enantiomerblandinger av p-laktamer kan oppløses ved hjelp av en stereoselektiv hydrolyse under anvendelse av en lipase eller et enzym, som beskrevet f.eks. av Patel, US patentskrift nr. 5 879 929, eller Patel,
US patentskrift nr. 5 567 614, eller et leverhomogenat, som beskrevet f.eks. i PCT-patentsøknad nr. 00/41204. Ved en foretrukket utførelsesform hvor p-laktamet er furylsubstituert i C(4)-stillingen, kan p-laktamet fremstilles som illustrert i det følgende reaksjonsskjema:
hvor Ac er acetyl, NEt3 er trietylamin, CAN er cerisk ammonium-nitrat, og p-TsOH er p-toluensulfonsyre. Okseleveroppløsningen
kan utføres f.eks. ved å kombinere den enantiomere p-laktam-blanding med en okseleversuspensjon (fremstilt f.eks. ved å til-sette 20 g nedfryst okselever til et blandeapparat og så til-sette en pH 8-buffer til et totalvolum på 1 1).
Forbindelser med formel 1 ifølge foreliggende oppfinnelse kan anvendes til inhibering av tumorvekst hos pattedyr, inkludert mennesker, og administreres fortrinnsvis i form av et farmasøytisk preparat som omfatter en effektiv antitumormengde av en forbindelse ifølge foreliggende oppfinnelse i kombinasjon med minst én farmasøytisk eller farmakologisk akseptabel bærer. Bæreren, også kjent innenfor teknikken som en eksipiens, et vehikkel, et hjelpestoff, et adjuvans eller et fortynnings-middel, er hvilket som helst stoff som er farmasøytisk inert, gir en egnet konsistens eller form til preparatet, og som ikke reduserer den terapeutiske virkningsfullhet av antitumorforbindelsene. Bæreren er "farmasøytisk eller farmakologisk akseptabel" dersom den ikke gir en skadelig, allergisk eller annen uønsket reaksjon når den administreres på passende måte til et pattedyr eller menneske.
De farmasøytiske preparatene som inneholder antitumorforbindelsene ifølge foreliggende oppfinnelse, kan formuleres på hvilken som helst vanlig måte. Korrekt formulering er avhengig av administreringsveien som velges. Preparatene ifølge oppfinnelsen kan formuleres for hvilken som helst administreringsvei så lenge som målvevet er tilgjengelig via den veien. Egnede administreringsveier omfatter, men er ikke begrenset til, oral, parenteral (f.eks. intravenøs, intraarteriell, subkutan, rektal, intramuskulær, intraorbital, intrakapsulær, intraspinal, intraperitoneal eller intrasternal), topisk (nasal, transdermal, intraokular), intravesikulær, intratekal, enteral, pulmonal, intralymfatisk, intrakavital, vaginal, transuretral, intra-dermal, aural, intramammal, bukkal, ortotopisk, intratrakeal, intralesjonal, perkutan, endoskopisk, transmukosal, sublingual og intestinal administrering.
Farmasøytisk akseptable bærere for anvendelse i preparatene ifølge foreliggende oppfinnelse er vel kjent for fagfolk innen teknikken og velges på grunnlag av en rekke faktorer: den bestemte antitumorforbindelse som anvendes og dens konsentra-sjon, stabilitet og påtenkte biologiske tilgjengelighet; syk-dommen, forstyrrelsen eller tilstanden som behandles med preparatet; individet, dets alder, størrelse og generelle tilstand; og administreringsveien. Egnede bærere bestemmes lett av fagfolk innen teknikken (se f.eks. J.G. Nairn i Remington's Pharmaceutical Science (A. Gennaro, red.), Mack Publishing Co., Easton, Pa.
(1985), s. 1492-1517).
Preparatene formuleres fortrinnsvis som tabletter, dispergerbare pulvere, piller, kapsler, gelkapsler, tabletter med hard overflate, geler, liposomer, granulater, oppløsninger, suspensjoner, emulsjoner, siruper, eliksirer, runde, flate pastiller, drasjeer, sugetabletter eller hvilken som helst annen doseringsform som kan administreres oralt. Teknikker og preparater for å lage orale doseringsformer som kan anvendes ved foreliggende oppfinnelse, er beskrevet i de følgende litteratur-henvisninger: Modern Pharmaceutics, kapitlene 9 og 10 (Banker & Rhodes, red., 1979); Lieberman et al., Pharmaceutical Dosage Forms: Tablets (1981); og Ansel, Introduction to Pharmaceutical Dosage Forms, 2. utgave (1976).
Preparatene ifølge oppfinnelsen for oral administrering omfatter en effektiv antitumormengde av en forbindelse ifølge oppfinnelsen i en farmasøytisk akseptabel bærer. Egnede bærere for faste doseringsformer omfatter sukkere, stivelser og andre vanlige stoffer, inkludert laktose, talkum, sukrose, gelatin, karboksymetylcellulose, agar, mannitol, sorbitol, kalsiumfosfat, kalsiumkarbonat, natriumkarbonat, kaolin, alginsyre, akasie, maisstivelse, potetstivelse, natriumsakkarin, magnesiumkarbonat, tragant, mikrokrystallinsk cellulose, kolloidalt silisium-dioksid, croskarmellosenatrium, talkum, magnesiumstearat og stearinsyre. Slike faste doseringsformer kan videre være ubelagte eller kan belegges ved hjelp av kjente teknikker; f.eks. for å forsinke desintegrasjon og absorpsjon.
Antitumorforbindelsene ifølge foreliggende oppfinnelse formuleres også fortrinnsvis for parenteral administrering, f.eks. formulert for injeksjon via intravenøs, intraarteriell, subkutan, rektal, intramuskulær, intraorbital, intrakapsulær, intraspinal, intraperitoneal eller intrasternal vei. Preparatene ifølge oppfinnelsen for parenteral administrering omfatter en effektiv antitumormengde av antitumorforbindelsen i en farma-søytisk akseptabel bærer. Doseringsformer som er egnet for parenteral administrering, omfatter oppløsninger, suspensjoner, dispersjoner, emulsjoner eller hvilken som helst annen doseringsform som kan administreres parenteralt. Teknikker og preparater for å lage parenterale doseringsformer er kjent innenfor fagområdet.
Egnede bærere som brukes ved formulering av flytende doseringsformer for oral eller parenteral administrering, omfatter ikke-vandige, farmasøytisk akseptable, polare oppløs-ningsmidler, slik som oljer, alkoholer, amider, estere, etere, ketoner, hydrokarboner og blandinger derav, samt vann, salt-oppløsninger, dekstroseoppløsninger (f.eks. DW5), elektrolytt-oppløsninger eller hvilken som helst annen vandig, farmasøytisk akseptabel væske.
Egnede ikke-vandige, farmasøytisk akseptable, polare oppløsningsmidler omfatter, men er ikke begrenset til, alkoholer (f.eks. a-glyserolformal, p-glyserolformal, 1,3-butylenglykol, alifatiske eller aromatiske alkoholer med 2-30 karbonatomer, slik som metanol, etanol, propanol, isopropanol, butanol, t-butanol, heksanol, oktanol, amylenhydrat, benzylalkohol, glyserol, glykol, heksylenglykol, tetrahydrofurfurylalkohol, lauryl-alkohol, cetylalkohol eller stearylalkohol, fettsyreestere av fettalkoholer, slik som polyalkylenglykoler (f.eks. polypropy-lenglykol, polyetylenglykol), sorbitan, sukrose og kolesterol); amider (f.eks. dimetylacetamid (DMA), benzylbenzoat-DMA, di-metylformamid, N-(P-hydroksyetyl)laktamid, N,N-dimetylacetamid-amider, 2-pyrrolidinon, l-metyl-2-pyrrolidinon eller polyvinylpyrrolidon); estere (f.eks. l-metyl-2-pyrrolidinon, 2-pyrrolidinon, acetatestere, slik som monoacetin, diacetin og triacetin, alifatiske eller aromatiske estere, slik som etylkaprylat eller

Mindre viktige tilleggsbestanddeler kan være inkludert i preparatene ifølge oppfinnelsen for mange forskjellige formål som er vel kjent innenfor den farmasøytiske industri. Disse bestanddelene vil for det meste gi egenskaper som øker retensjon av antitumorforbindelsen på administreringsstedet, beskytter stabiliteten til preparatet, regulerer pH-verdien, letter be-arbeidelse av antitumorforbindelsen til farmasøytiske preparater og lignende. Hver av disse bestanddelene er fortrinnsvis indi-viduelt til stede med mindre enn ca. 15 vekt% av totalprepa-ratet, mer foretrukket mindre enn ca. 5 vekt%, og mest foretrukket mindre enn ca. 0,5 vekt% av hele preparatet. Noen bestand-deler, slik som fyllstoffer eller fortynningsmidler, kan utgjøre opptil 90 vekt% av hele preparatet, noe som er vel kjent innenfor formuleringsteknikken. Slike additiver omfatter kuldebeskyt-telsesmidler for forhindring av ny utfelling av taxanet, over-flateaktive, fukte- eller emulgeringsmidler (f.eks. lecitin, "Polysorbat" 80, "Tween" 80, "Pluronic" 60, polyoksyetylen-stearat), konserveringsmidler (f.eks. etyl-p-hydroksybenzoat), mikrobielle konserveringsmidler (f.eks. benzylalkohol, fenol, m-kresol, klorbutanol, sorbinsyre, timerosal og paraben), midler for regulering av pH eller buffermidler (f.eks. syrer, baser, natriumacetat, sorbitanmonolaurat) , midler for regulering av osmolalitet (f.eks. glyserol), fortykningsmidler (f.eks. aluminiummonostearat, stearinsyre, cetylalkohol, stearylalkohol, guargummi, metylcellulose, hydroksypropylcellulose, tristearin, cetylvoksestere, polyetylenglykol), fargestoffer, farger, strømningshjelpemidler, ikke-flyktige silikoner (f.eks. syklo-metikon), leirestoffer (f.eks. bentonitter), festemidler, volum-økende midler, smaksmidler, søtningsmidler, adsorberingsmidler, fyllstoffer (f.eks. slike sukkere som laktose, sukrose, mannitol eller sorbitol, cellulose eller kalsiumfosfat), fortynningsmidler (f.eks. vann, saltoppløsning, elektrolyttoppløsninger) , bindemidler (f.eks. slike stivelser som maisstivelse, hvetestivelse, risstivelse eller potetstivelse, gelatin, tragant-gummi, metylcellulose, hydroksypropylmetylcellulose, natrium-karboksymetylcellulose, polyvinylpyrrolidon, sukkere, polymerer, akasie), desintegrasjonsmidler (f.eks. slike stivelser som maisstivelse, hvetestivelse, risstivelse, potetstivelse eller karboksymetylstivelse, kryssbundet polyvinylpyrrolidon, agar, alginsyre eller et salt derav, slik som natriumalginat, croskarmellosenatrium eller crospovidon), smøremidler (f.eks. silika, talkum, stearinsyre eller salter derav, slik som magnesiumstearat, eller polyetylenglykol), beleggingsmidler (f.eks. konsentrerte sukkeroppløsninger, inkludert gummi arabicum, talkum, polyvinylpyrrolidon, karbopolgel, polyetylenglykol eller titandioksid) og antioksidanter (f.eks. natrium-metabisulfitt, natriumbisulfitt, natriumsulfitt, dekstrose, fenoler og tiofenoler).
Ved en foretrukket utførelsesform omfatter et farma-søytisk preparatet ifølge oppfinnelsen minst ett ikke-vandig, farmasøytisk akseptabelt oppløsningsmiddel og en antitumorforbindelse med en oppløselighet i etanol som er minst ca. 100, 200, 300, 400, 500, 600, 700 eller 800 mg/ml. Selv om man ikke er bundet til noen bestemt teori,- antas det at etanoloppløselig-heten til antitumorforbindelsen kan være direkte relatert til dens virkningsfullhet. Antitumorforbindelsen kan også være i stand til å krystallisere fra en oppløsning. Med andre ord kan en krystallinsk antitumorforbindelse, slik som forbindelse 1393, oppløses i et oppløsningsmiddel slik at det dannes en oppløs-ning, og så rekrystalliseres etter avdamping av oppløsnings-midlet uten dannelse av noen amorf antitumorforbindelse. Det er også foretrukket at antitumorforbindelsen har en IDso-verdi (det vil si den legemiddelkonsentrasjonen som gir 50 % inhibering av kolonidannelse) som er minst 4, 5, 6, 7, 8, 9 eller 10 ganger mindre enn den til paklitaxel når man måler i henhold til frem-gangsmåten angitt i arbeidseksemplene.
Doseringsformadministrering via disse veier kan være kontinuerlig eller oppdelt, f.eks. avhengig av pasientens fysiologiske tilstand, hvorvidt formålet ved administreringen er terapeutisk eller profylaktisk, og andre faktorer som er kjent og som lar seg fastslå av en erfaren lege.
Dosering og regimer for administreringen av de farma-søytiske preparatene ifølge oppfinnelsen kan lett bestemmes av fagfolk innen behandling av kreft. Det skal forstås at doseringen av antitumorforbindelsene vil være avhengig av alderen, kjønnet, helsen og vekten til mottakeren, typen av ledsagende behandling dersom det er noen slik, hyppigheten av behandling og typen av den ønskede effekt. For enhver administreringsmåte vil den aktuelle mengde av antitumorforbindelse som avleveres, samt doseringsplanen som er nødvendig for å oppnå de fordelaktige effekter som er beskrevet her, også delvis avhenge av slike faktorer som den biologiske tilgjengelighet av antitumorforbindelsen, forstyrrelsen som behandles, den ønskede terapeutiske dose og andre faktorer som vil være åpenbare for fagfolk innen teknikken. Dosen som administreres til et dyr, særlig et menneske, i sammenheng med den foreliggende oppfinnelse, bør være tilstrekkelig til å bevirke den ønskede terapeutiske respons hos dyret over et rimelig tidsrom. En effektiv mengde av antitumorforbindelsen, enten den administreres oralt eller via en annen vei, er fortrinnsvis enhver mengde som vil resultere i en ønsket terapeutisk respons når den administreres via den veien. Fortrinnsvis fremstilles preparatene for oral administrering på en slik måte at en enkeltdose i ett eller flere orale preparater inneholder minst 20 mg av antitumorforbindelsen pr. m<2> pasientkroppsoverflateareal, eller minst 50, 100, 150, 200, 300, 400 eller 500 mg av antitumorforbindelsen pr. m<2 >pasientkroppsoverflateareal, hvor det gjennomsnittlige kropps-overflateareal for et menneske er 1,8 m<2>. En enkeltdose av et preparat for oral administrering inneholder fortrinnsvis fra ca.
20 til ca. 600 mg av antitumorforbindelsen pr. m<2> pasientkropps-overf lateareal , mer foretrukket fra ca. 25 til ca. 400 mg/m<2>, enda mer foretrukket fra ca. 40 til ca. 300 mg/m<2>, og enda mer foretrukket fra ca. 50 til ca. 200 mg/m2. Fortrinnsvis fremstilles preparatene for parenteral administrering på en slik måte at en enkeltdose inneholder minst 20 mg av antitumorforbin-deisen pr. m pasientkroppsoverflateareal, eller minst 40, 50, 100, 150, 200, 300, 400 eller 500 mg av antitumorforbindelsen pr. m<2> pasientkroppsoverflateareal. En enkeltdose i ett eller flere parenterale preparater inneholder fortrinnsvis fra ca. 20 til ca. 500 mg av antitumorforbindelse pr. m<2> pasientkropps-overf lateareal, mer foretrukket fra ca. 40 til ca. 400 mg/m<2>, og enda mer foretrukket fra ca. 60 til ca. 350 mg/m<2>. Doseringen kan imidlertid variere, avhengig av doseringsplanen som kan regu-leres etter behov for å oppnå den ønskede terapeutiske effekt. Det bør legges merke til at områdene for effektive doser som er gitt her, ikke er ment å begrense oppfinnelsen og utgjør foretrukne doseområder. Den mest foretrukne dosering vil være skreddersydd for det enkelte individ, slik det vil forstås og lar seg bestemme av fagfolk innen teknikken uten for mye eksperimentering.
Konsentrasjonen av antitumorforbindelsen i et flytende farmasøytisk preparatet er fortrinnsvis mellom ca. 0,01 mg og ca. 10 mg pr. ml av preparatet, mer foretrukket mellom ca.
0,1 mg og ca. 7 mg pr. ml, enda mer foretrukket mellom ca.
0,5 mg og ca. 5 mg pr. ml, og mest foretrukket mellom ca. 1,5 mg og ca. 4 mg pr. ml. Forholdsvis lave konsentrasjoner er generelt foretrukket fordi antitumorforbindelsen er mest oppløselig i oppløsningen ved lave konsentrasjoner. Konsentrasjonen av anti-tumorf orbindelsen i et fast farmasøytisk preparatet for oral administrering er fortrinnsvis mellom ca. 5 vekt% og ca. 50 vekt%, basert på den totale vekt av preparatet, mer foretrukket mellom ca. 8 vekt% og ca. 40 vekt%, og mest foretrukket mellom ca. 10 vekt% og ca. 30 vekt%.
Ved én utførelsesform fremstilles oppløsninger for oral administrering ved å oppløse en antitumorlorbindelse i hvilket som helst farmasøytisk akseptabelt oppløsningsmiddel som er i stand til å oppløse forbindelsen (f.eks. etanol eller metylenklorid) for å danne en oppløsning. Et passende volum av en bærer som er en oppløsning, slik som "Cremophor" EL-oppløsning, tilsettes til oppløsningen mens det omrøres for å danne en farma-søytisk akseptabel oppløsning for oral administrering til en pasient. Om ønsket kan slike oppløsninger formuleres til å inneholde en minimal mengde av, eller til å være fri for, etanol, som er kjent innenfor teknikken for å forårsake skadelige fysiologiske effekter når de administreres ved visse konsentrasjoner i orale preparater.
Ved en annen utførelsesform fremstilles pulvere eller
tabletter for oral administrering ved å oppløse en antitumorforbindelse i hvilket som helst farmasøytisk akseptabelt opp-løsningsmiddel som er i stand til å oppløse forbindelsen (f.eks. etanol eller metylenklorid), hvorved det dannes en oppløsning. Oppløsningsmidlet kan eventuelt være i stand til å fordampe når oppløsningen tørkes under vakuum. En ytterligere bærer kan tilsettes til oppløsningen før tørking, slik som "Cremophor" EL-oppløsning. Den resulterende oppløsning tørkes under vakuum, hvorved det dannes et glass. Glasset blandes så med et binde-middel for å danne et pulver. Pulveret kan blandes med fyllstoffer eller andre vanlige tabletteringsmidler og bearbeides for å danne en tablett for oral administrering til en pasient. Pulveret kan også tilsettes til hvilken som helst flytende bærer, som beskrevet ovenfor, for å danne en oppløsning, emulsjon, suspensjon eller lignende for oral administrering.
Emulsjoner for parenteral administrering kan fremstilles ved å oppløse en antitumorforbindelse i hvilket som helst farmasøytisk akseptabelt oppløsningsmiddel som er i stand til å oppløse forbindelsen (f.eks. etanol eller metylenklorid), hvorved det dannes en oppløsning. Et passende volum av en bærer som er en emulsjon, slik som "Liposyn" II- eller "Liposyn" III-emulsjon, tilsettes til oppløsningen mens det omrøres for å danne en farmasøytisk akseptabel emulsjon for parenteral administrering til en pasient. Om ønsket kan slike emulsjoner formuleres til å inneholde en minimal mengde av, eller til å være fri for, etanol eller "Cremophor"-oppløsning, som er kjent innenfor teknikken for å forårsake skadelige fysiologiske effekter når de administreres ved visse konsentrasjoner i parenterale preparater.

Om ønsket kan emulsjonene eller oppløsningene som er beskrevet ovenfor for oral eller parenteral administrering, emballeres i I.V.-poser, ampuller eller andre vanlige beholdere i konsentrert form og fortynnes med hvilken som helst farma-søytisk akseptabel væske, slik som saltoppløsning, for å danne en akseptabel taxankonsentrasjon før bruk, slik det er kjent innenfor teknikken.

Eksempel 1
10- trietylsilyl- 10- deacetyl- 7- propionyl- baccatin III
Til en oppløsning av 1,0 g (l,84mol) 10-deacetylbaccatin III i 50 ml THF ved -10 °C under nitrogenatmosfære ble det tilsatt 0,857 ml (2,76 mmol, 1,5 molekv.) N,0-(bis)-TES-trifluoracetamid i løpet av et tidsrom på 3 minutter. Dette ble etterfulgt av tilsetning av 0,062 ml av en 0,89 M THF-oppløsning av litiumbis(trimetylsilyl)amid (0,055 mmol, 0,03 molekv.). Etter 10 minutter ble 0,038 ml (0,92 mmol, 0,5 molekv.) metanol tilsatt, og etter ytterligere 5 minutter ble 4 ml (0,055 mmol, 0,03 molekv.) eddiksyre tilsatt. Oppløsningen ble fortynnet med 300 ml etylacetat og vasket to ganger med 100 ml mettet, vandig natriumbikarbonatoppløsning. Til en oppløsning av 1,0 g (1,517 mmol) 10-trietylsilyl-10-deacetylbaccatin III og 37,0 mg (0,303 mmol) DMAP i 20 ml diklormetan ved romtemperatur under nitrogenatmosfære ble det tilsatt 0,920 ml (11,381 mmol) pyridin og 0,329 ml (3,794 mmol, 2,5 molekv.) propionylklorid i den rekkefølgen. Blandingen ble omrørt ved romtemperatur i 6 timer, fortynnet med 350 ml etylacetat og ekstrahert med 50 ml 10 % vandig kobber-sulfatoppløsning. Det organiske lag ble vasket med 50 ml mettet, vandig natriumbikarbonatoppløsning, 50 ml saltoppløsning, tørket over natriumsulfat og konsentrert under redusert trykk. Råproduktet ble oppløst i 75 ml etylacetat, 100 mg Norit A ble tilsatt, blandingen ble filtrert gjennom celitt og konsentrert under redusert trykk, hvorved man fikk 1,13 g materiale. Rekrystallisering fra etylacetat/heksaner (oppløst i 6,5 ml reflukserende etylacetat, så ble 24 ml heksaner tilsatt, fikk avkjøles til romtemperatur og stå i 17 timer) ga 787 mg (72,5 %) av et hvitt, krystallinsk, fast stoff. En andre rekrystallisering (ca. 340 mg materiale oppløst i 2 ml reflukserende etylacetat, så tilsatt 10 ml heksaner, fikk avkjøles til romtemperatur og stå i 17 timer) ga 181 mg (16,7 % av et hvitt, krystallinsk, fast stoff. Det kombinerte utbyttet etter rekrystallisering var 89,2 %.

Til en oppløsning av 493 mg (0,690 mmol) 10-trietylsilyl-10-deacetyl-7-propionyl-baccatin III i 4 ml vannfritt THF under nitrogenatmosfære ved -45 °C ble det tilsatt 0,72 ml
(0,72 mmol) av en 1 M oppløsning av LiHMDS i THF. Etter 0,5 time ble en oppløsning av 263 mg (0,814 mmol) av b-laktamet (for-tørket som beskrevet ovenfor) i 2 ml vannfritt THF tilsatt. Blandingen ble varmet opp til 0 °C, og etter 2 timer ble 0,5 ml mettet, vandig natriumbikarbonatoppløsning tilsatt. Blandingen ble fortynnet med 50 ml etylacetat og vasket to ganger med 5 ml saltoppløsning. Den organiske fase ble tørket over natriumsulfat og konsentrert under redusert trykk, hvorved man fikk 742 mg (104 %) av et svakt gult, fast stoff. Til en oppløsning av 206 mg (0,199 mmol) 2'-0-MOP-3I<->desfenyl-3'-(2-furyl)-10-trietylsilyl-7-propionyltaxotere i 1,7 ml pyridin og 5,4 ml acetonitril ved 0 °C ble det tilsatt 0,80 ml (2,0 mmol) av en vandig oppløsning inneholdende 49 % HF. Blandingen ble varmet opp til romtemperatur i 14 timer og ble så fortynnet med 20 ml etylacetat og vasket tre ganger med 2 ml mettet, vandig natriumbikarbonat og så med 8 ml saltoppløsning. Den organiske fase ble tørket over natriumsulfat og konsentrert under redusert trykk, hvorved man fikk 170 mg (100 %) av et hvitt, fast stoff. Råproduktet ble krystallisert med 2 ml opp-løsningsmiddel (CH2CI2:heksan = 1:1,7), hvorved man fikk 155 mg (90,5 %) hvite krystaller. In vitro- cytotoksisitet målt ved hjelp av cellekolonidannelses-analysen
400 celler (HCT116) ble platet ut i 60 mm petriskåler inneholdende 2,7 ml medium (modifisert McCoys 5a-medium inneholdende 10 % bovint fosterserum og 100 enheter/ml penicillin og 100 g/ml streptomycin). Cellene ble inkubert i en C02-inkubator ved 37 °C i 5 timer for festing til bunnen av petriskåler. For-bindelsene identifisert i eksempel 2 ble nyutblandet i medium ved 10 ganger sluttkonsentrasjonen, og så ble 0,3 ml av denne standardoppløsningen tilsatt til de 2,7 ml medium i skålen. Cellene ble så inkubert med legemidler i 72 timer ved 37 °C. Antitumorforbindelse 1393 ble oppløst i 100 % etanol, og så ble det blandet med et like stort volum "Cremophor" EL-oppløsning, slik at det ble dannet en oppløsning inneholdende 70 mg av forbindelse 1393 pr. ml. Denne oppløsningen ble fortynnet ved å anvende 9 vektdeler D % W (en vandig oppløsning inneholdende 5 % (vekt/volum) dekstrose) eller 0,9 % salt-oppløsning, slik at det ble dannet en farmasøytisk akseptabel oppløsning for administrering til en pasient.
Eksempel 5
Fremstilling av en suspensjon inneholdende forbindelse 1393 for oral administrering
Et oralt preparat av antitumorforbindelse 1393 ble fremstilt ved å oppslemme 25 mg av forbindelse 1393 som et fint pulver i 1 ml bærer inneholdende 1 % karboksymetylcellulose (CMC) i avionisert vann.

Fremstilling av en tablett inneholdende forbindelse 1393 for oral administrering
Antitumorforbindelse 1393 (100 mg) ble oppløst i metylenklorid (2 ml), og "Cremophor" EL-oppløsning (100 mg) ble tilsatt. Metylenkloridet ble avdampet under vakuum slik at det ble dannet et glass. Mikrokrystallinsk cellulose (600 mg) ble tilsatt til glasset, og det ble blandet slik at det ble dannet et pulver som kan bearbeides slik at det dannes en tablett.

Oppløsningen ble så fortynnet med 19 vektdeler "Liposyn" III (2 %) med omrøring slik at det ble dannet en emulsjon inneholdende 2 mg av forbindelse 1393 pr. ml for parenteral administrering.
Emulsjon 3
Antitumorforbindelse 1393 ble oppløst i 100 % etanol for å danne en oppløsning inneholdende ? mg av forbindelse 1393 pr. ml av oppløsningen. Oppløsningen ble så fortynnet med 9 vektdeler "Liposyn" III (2 %) med omrøring slik at det ble dannet en emulsjon inneholdende 4 mg av forbindelse 1393 pr. ml for parenteral administrering.
