Oppfinnelsen vedrører forbindelser med formel I

tuert med R<2>og/eller R<4>,
R<2>fR<4>,
R<5> og R<6> er i hvert tilfelle uavhengig av hverandre Hal, A, OA,
OH, CN eller Ci-Ceacyl,
R<3> er et fenylradikal som er usubstituert eller substi-
tuert med R<5> og/eller R<6>, eller er Het<2>,
Het<2> er furyl eller tienyl,
A er alkyl med 1-6 C-atomer, og
Hal er F, Cl, Br eller I,
og deres fysiologisk akseptable salter og solvater.
Oppfinnelsen var basert på det formål å finne nye forbindelser som har verdifulle egenskaper, særlig de som kan anvendes til fremstillingen av medikamenter.
Det er blitt funnet at forbindelsene med formel I og
deres fysiologisk akseptable salter og solvater har verdifulle farmakologiske egenskaper sammen med god tolererbarhet, ettersom de har virkninger på sentralnervesystemet. Forbindelsene har en sterk affinitet for 5-HT2A-reseptorer; de utviser dessuten 5-HT2a~ reseptorantagonistiske egenskaper.
For in vitro-påvisningen av affiniteten for 5-HT2A-reseptorer er det mulig f.eks. å anvende den følgende test (eksempel Al). 5-HT2A-reseptorene eksponeres for både [<3>H}~
ketanserin (et stoff som er kjent for sin affinitet for resep-
toren) og testforbindelsen. Reduksjonen i affiniteten til [<3>H]-ketanserin for reseptoren er et tegn på affiniteten til test-
stoffet for 5-HT2A_reseptoren. Påvisning utføres analogt med beskrivelsen til J.E. Leysen et al., Molecular Pharmacology,
1982, 22:301-314, eller som også beskrevet i EP 0320983.
Virkningsfullheten av forbindelsene ifølge oppfinnelsen som 5-HT2A-reseptorantagonister kan måles in vit ro analogt med W. Feniuk et al., Mechanisms of 5-hydroxytryptamine-induced vasoconstriction, i: The Peripheral Actions of 5-hydroxytryptamine, red. Fozard, J.R., Oxford University Press, New York, 1989, s. 110. Kontraktiliteten i rottehalearterien forårsaket av 5-hydroksytryptamin formidles således ved hjelp av 5-HT2R-reseptorer. For testsystemet underkastes karringer fremstilt fra ventral rottehalearterie perfusjon med en oksygenmettet oppløs-ning i et organbad. Ved innføring av økende konsentrasjoner av 5-hydroksytryptamin i oppløsningen fås en respons på den kumula-tive konsentrasjon av 5-HT. Testforbindelsen tilsettes så til organbadet i egnede konsentrasjoner, og en andre konsentrasjonskurve måles for 5-HT. Styrken til testforbindelsen på skiftingen i den 5-HT-induserte konsentrasjonskurve til høyere 5-HT-konsentrasjoner er et mål for den 5-HT2A-reseptorantagonistiske egenskap in vitro.
Den 5-HT2A-antagonistiske egenskap kan bestemmes in vivo analogt med M.D. Serdar et al., Psychopharmacology, 1996, 125:198-205.
Andre forbindelser som likeledes utviser 5-HT2-antago-nistiske virkninger, er f.eks. beskrevet i EP 0320983. Lignende piperazinderivater som har antiarytmiske egenskaper, er f.eks. beskrevet i EP 0431945. Andre indolkarbonylderivater som har analgetiske egenskaper, er beskrevet i EP 0599240. EP 0624584 beskriver piperazinderivater som kalmodolininhibitorer. Forbindelsene ifølge oppfinnelsen skal anses som en utvelgelsesoppfin-nelse i forhold til den sist nevnte publikasjon. WO 99/11641 beskriver fenylindolderivater som har 5-HT2A-antagonistiske egenskaper.
Forbindelsene med formel I er derfor egnet både innen
veterinær- og humanmedisinen for behandling av funksjonelle forstyrrelser i sentralnervesystemet, og også av betennelse. De kan anvendes til profylaksen og til kontrollen av følgesykdommene av hjerneinfarkter {cerebral apopleksi) , slik som slag og cerebrale iskemier, og til behandlingen av ekstrapyramidale motorbivirk-ninger av neuroleptika, og også av Parkinsons sykdom, til den akutte og symptomatiske terapi av Alzheimers sykdom og behandlingen av amyotrof lateralsklerose. De er likeledes egnet som
terapeutika for behandlingen av hjerne- og ryggmargstraumer. Særlig er de imidlertid egnet som farmasøytisk aktive forbindelser for anxiolytika, antidepressive midler, antipsykotika, neuroleptika, antihypertensjonsmidler og/eller for positiv påvirkning av tvangsnevrotisk oppførsel (obsessiv-kompulsiv forstyrrelse, OCD; f.eks. WO 9524194), angsttilstander og også fysiologiske endringer som ledsager angsttilstander, slik som f.eks. takykardi, tremorer eller svetting (f.eks. EP 319962), panikkanfall, psykoser, schizofreni, anoreksi, vrangforestil-linger, agorafobi, migrene, Alzheimers sykdom, søvnforstyrrelser og også søvnapné, tardive dyskinesier, læreforstyrrelser, aldersavhengige hukommelsesforstyrrelser, spiseforstyrrelser, slik som bulimi, rusmiddelmisbruk, slik som f.eks. av alkohol, opiater, nikotin, slike psykostimuleringsmidler som f.eks. kokain eller amfetaminer {f.eks. US 6004980), seksuelle funksjonsforstyrrelser, smertetilstander av alle typer og fibro-myalgi (f.eks. WO 9946245).
Forbindelsene med formel I er egnet for behandlingen av ekstrapyramidale bivirkninger (EPS) med neuroleptisk legemiddel-terapi. EPS er kjennetegnet ved Parkinson-lignende syndromer, akatisi og dystoniske reaksjoner (f.eks. EP 337136). De er videre egnet til behandlingen av anorexia nervosa, angina, Raynauds fenomen, koronare vasospasmer, ved profylaksen av migrene (f.eks. EP 208235), smerte og neuralgi (f.eks.
EP 320983), til behandlingen av Rett-syndromet med autistiske trekk, av Asperger-syndromet eller autisme og autistiske forstyrrelser, ved konsentrasjonssvikttilstander, utviklingsmessige sykdommer, hyperaktivitetstilstander med mental underutvikling og stereotypisk oppførselsmessige tilstander {f.eks.
WO 9524194).
I tillegg er de egnet til behandlingen av endokrine forstyrrelser, slik som hyperprolaktinemi, dessuten ved vasospasmer, trombotiske forstyrrelser (f.eks. WO 9946245), hyper-tensjon og gastrointestinale forstyrrelser.
De er dessuten egnet for behandlingen av kardiovaskulære forstyrrelser og også ekstrapyramidale symptomer, som beskrevet i WO 99/11641, på side 2, linjene 24-30.
Forbindelsene ifølge oppfinnelsen er videre egnet for å redusere intraokulart trykk og for behandlingen av glaukom. De er også egnet hos dyr for profylaksen og behandlingen av symptomer på intoksikasjon etter administreringen av ergovalin.
Forbindelsene er dessuten egnet for behandlingen av forstyrrelser i det kardiovaskulære system (WO 99/11641, side 3, linjene 14-15).
Forbindelsene ifølge oppfinnelsen kan også anvendes sammen med andre aktive forbindelser ved behandlingen av schizofreni. Mulige andre aktive forbindelser er forbindelsene som er
nevnt i WO 99/11641, på side 13, linjene 20-26.
De kan dessuten anvendes som mellomprodukter for fremstillingen av ytterligere farmasøytisk aktive forbindelser.
Oppfinnelsen vedrører anvendelsen av N-{indolkarbonyl)-piperazinderivater med formel I og deres fysiologisk akseptable syreaddisjonssalter. Oppfinnelsen vedrører også solvatene, f.eks. hydratene eller alkoholatene, av disse forbindelsene.
Oppfinnelsen vedrører følgelig forbindelsene med formel I og en fremgangsmåte for fremstillingen av forbindelser med formel I ifølge krav 1.
Fremgangsmåten for fremstillingen av forbindelser med formel I ifølge krav 1 er kjennetegnet ved at

Oppfinnelsen vedrører også forbindelsene med formel I ifølge krav 1 og deres fysiologisk akseptable salter og solvater for bruk som medikamenter.
For alle radikaler som opptrer et antall ganger, slik som f.eks. A eller Hal, gjelder det at deres betydninger er uavhengige av hverandre.
Radikalet A er alkyl og har 1-6, fortrinnsvis 1, 2, 3 eller 4, særlig 1 eller 2, C-atomer. Oppfinnelsen vedrører følgelig særlig de forbindelsene med formel I hvor minst ett av de nevnte radikaler har en av de foretrukne betydninger angitt ovenfor. Forbindelsene med formel I og også utgangsforbindelsene for deres fremstilling fremstilles ellers ved hjelp av i og for seg kjente fremgangsmåter, slik som de er blitt beskrevet i litteraturen (f.eks. i standardverkene, slik som Houben-Weyl, Methoden der organischen Chemie [Methods of Organic Chemistry], Georg-Thieme-Verlag, Stuttgart; Organic Reactions, John Wiley & Sons, Inc., New York), nemlig under slike reaksjonsbetingelser som er kjent og egnet for de nevnte reaksjoner. I dette tilfellet kan det også gjøres bruk av i og for seg kjente varianter som ikke er nærmere nevnt her.
Om ønsket kan utgangsforbindelsene for den krevde fremgangsmåte også dannes in situ på en slik måte de ikke isoleres fra reaksjonsblandingen, men umiddelbart omsettes videre, hvorved man får forbindelsene med formel I. På den annen side er det mulig å utføre reaksjonen trinnvis.
I forbindelsene med formlene II og V er radikalet L fortrinnsvis Cl eller Br; det kan imidlertid også være I, OH eller ellers foretrukket en reaktivt, funksjonelt modifisert OH-gruppe, særlig alkylsulfonyloksy med 1-6 {f.eks. metansulfonyl-oksy) eller arylsulfonyloksy med 6-10 C-atomer (f.eks. benzen-sulfonyloksy, p-toluensulfonyloksy, 1- eller 2-naftalensulfonyl-oksy) eller ellers triklormetoksy, alkoksy, slik som f.eks. metoksy, etoksy, propoksy eller butoksy, dessuten også fenoksy.
Forbindelsene med formel I kan fortrinnsvis fås ved å omsette forbindelser med formel II med forbindelser med formel
III.
Som regel er utgangsforbindelsene med formlene II og III kjent; de ukjente forbindelsene med formlene II og III kan lett fremstilles analogt med de kjente forbindelsene.
Omsetningen av forbindelsene II og III forløper i hen-hold til slike metoder som er kjent fra litteraturen for alky-leringen eller acyleringen av aminer. Det er imidlertid også mulig å omsette forbindelsene i nærvær av et inert oppløsnings-middel. Egnede oppløsningsmidler er f.eks. slike hydrokarboner som benzen, toluen eller xylen; slike ketoner som aceton eller butanon; slike alkoholer som metanol, etanol, isopropanol eller n-butanol; slike etere som tetrahydrofuran (THF) eller dioksan; slike amider som dimetylformamid (DMF) eller N-metylpyrrolidon; slike nitriler som acetonitril, og om det er passende også blandinger av disse oppløsningsmidlene med hverandre eller blandinger med vann. Tilsetningen av et syrebindende middel, f.eks. et alkalimetall- eller jordalkalimetallhydroksid, -karbonat eller -bikarbonat, eller et annet salt av en svak syre med alkali- eller jordalkalimetallene, fortrinnsvis kalium, natrium eller kalsium, eller tilsetningen av en organisk base, slik som trietylamin, dimetylanilin, pyridin eller kinolin, eller av et overskudd av piperazinderivat med formel II kan være gunstig. Avhengig av de anvendte betingelser er reaksjonstiden mellom noen få minutter og 14 dager; reaksjonstemperaturen er mellom ca. 0 og 150 °C, normalt mellom 20 og 130 °C.
I tillegg kan forbindelser med formel I fremstilles ved å omsette aminer med formel IV med en bestanddel med formel V som omfatter radikalet R<1>. Som regel er de respektive kompo-nentene kjent eller kan fremstilles ved hjelp av kjente fremgangsmåter, som allerede beskrevet.
En erholdt base med formel I kan omdannes til det led-sagende syreaddisjonssalt ved å anvende en syre. For denne omsetningen er egnede syrer de som gir fysiologisk akseptable salter. Uorganiske syrer kan således anvendes, f.eks. svovel-syre, hydrogenhalogensyrer, slik som saltsyre eller hydrobrom-syre, fosforsyrer, slik som ortofosforsyre, salpetersyre, sulf-aminsyre, dessuten organiske syrer, nærmere bestemt alifatiske, alisykliske, aralifatiske, aromatiske eller heterosykliske en-eller flerbasiske karboksyl-, sulfon- eller svovelsyrer, slik som maursyre, eddiksyre, propionsyre, pivalinsyre, dietyleddik-syre, malonsyre, ravsyre, pimelinsyre, fumarsyre, maleinsyre, melkesyre, vinsyre, eplesyre, benzosyre, salisylsyre, 2-fenyl-propionsyre, sitronsyre, glukonsyre, askorbinsyre, nikotinsyre, isonikotinsyre, metan- eller etansulfonsyre, etandisulfonsyre, 2-hydroksyetansulfonsyre, benzensulfonsyre, p-toluensulfonsyre, naftalenmono- og -disulfonsyrer og laurylsvovelsyre.
De frie basene med formel I kan om ønsket frigjøres fra sine salter ved behandling med sterke baser, slik som natrium-eller kaliumhydroksid, eller natrium- eller kaliumkarbonat, dersom det ikke er til stede noen ytterligere sure grupper i molekylet. I de tilfellene hvor forbindelsene med formel I har frie syregrupper, kan saltdannelse likeledes oppnås ved behandling med baser. Egnede baser er alkalimetallhydroksider, jord-alkalimetallhydroksider eller organiske baser i form av primære, sekundære eller tertiære aminer.
Oppfinnelsen vedrører også et farmasøytisk preparat som omfatter minst én forbindelse med formel I ifølge oppfinnelsen, og deres fysiologisk akseptable salter og solvater, og også, dersom det er passende, vehikler og/eller eksipienser og, dersom det er passende, andre aktive forbindelser. I dette tilfellet kan medikamentene bringes i en egnet doseform sammen med minst ett fast, flytende og/eller halvflytende vehikkel eller en eksi-piens, og dersom det er passende, i kombinasjon med én eller flere ytterligere aktive forbindelser.
Oppfinnelsen vedrører også anvendelsen av forbindelsene ifølge oppfinnelsen og/eller av deres fysiologisk akseptable salter og solvater til fremstillingen et medikament for behandlingen av psykoser, schizofreni, depresjon, neurologiske forstyrrelser, hukommelsesforstyrrelser, Parkinsons sykdom, amyotrof lateralsklerose, Alzheimers sykdom, Huntingtons sykdom, slike spiseforstyrrelser som bulimi, anorexia nervosa, premenstruelt syndrom og/eller til positiv påvirkning av tvangsnevrotisk oppførsel (obsessiv-kompulsiv forstyrrelse, OCD).
De farmasøytiske preparatene kan anvendes som medikamenter innen human- og veterinærmedisin. Egnede bærerstoffer er organiske eller uorganiske stoffer som er egnet for enteral (f.eks. oral) eller parenteral administrering eller topisk applikasjon, og som ikke reagerer med de nye forbindelsene, f.eks. vann, vegetabilske oljer, benzylalkoholer, polyetylen-glykoler, gelatin, slike karbohydrater som laktose eller stivelse, magnesiumstearat, talkum og vaselin. Særlig anvendes tabletter, belagte tabletter, kapsler, siruper, suspensjoner, dråper eller suppositorier for enteral administrering, oppløs-ninger, fortrinnsvis olje- eller vannoppløsninger, dessuten suspensjoner, emulsjoner eller implantater for parenteral administrering, og salver, kremer eller pulver brukes for topisk applikasjon. De nye forbindelsene kan også lyofiliseres, og de erholdte lyofilisater kan f.eks. anvendes til fremstillingen av inj eksj onspreparater.
De angitte preparater steriliseres og/eller kan inneholde slike eksipienser som smøremidler, konserveringsmidler, stabiliseringsmidler og/eller fuktemidler, emulgeringsmidler, salter for påvirkning av det osmotiske trykk, bufferstoffer, fargestoffer, smaksstoffer og/eller aromastoffer. De kan om ønsket også inneholde én eller flere ytterligere aktive forbindelser, f.eks. ett eller fleire vitaminer.
I dette tilfellet administreres stoffene ifølge oppfinnelsen som regel analogt med kjente preparater, fortrinnsvis i doser mellom ca. 0,1 og 500 mg, særlig mellom 5 og 300 mg, pr. doseenhet. Den daglige dose er fortrinnsvis mellom ca. 0,01 og 250 mg/kg, særlig mellom 0,02 og 100 mg/kg kroppsvekt.
I dette tilfellet administreres stoffene ifølge oppfinnelsen som regel fortrinnsvis i doser mellom ca. 1 og 500 mg, særlig mellom 5 og 100 mg, pr. doseenhet. Den daglige dose er fortrinnsvis mellom ca. 0,02 og 10 mg/kg kroppsvekt. Den bestemte dose for hver påtenkte pasient avhenger imidlertid av alle slags faktorer, f.eks. av virkningsfullheten til den bestemte forbindelse som anvendes, av alderen, kroppsvekten, den generelle helsetilstand, kjønnet, av kosten, av administrerings-tiden og -veien, av utskillingshastigheten, den farmasøytiske kombinasjonen og alvorligheten av den bestemte forstyrrelse som behandlingen gjelder for. Oral administrering er foretrukket.
Ovenfor og nedenunder er alle temperaturer angitt i °C. I eksemplene nedenunder betyr "vanlig opparbeidelse": Om nød-vendig fjernes oppløsningsmidlet, om nødvendig tilsettes vann, om nødvendig reguleres blandingen, avhengig av sluttproduktets konstitusjon, til en pH-verdi mellom 2 og 10, og ekstraheres med etylacetat eller diklormetan, den organiske fase fraskilles, tørkes over natriumsulfat, filtreres og konsentreres, og resten renses ved hjelp av kromatografi på silikagel og/eller ved krystallisasjon.

Fremre del av hjernebarken fra rotte homogeniseres i iskald buffer. Homogenatet sentrifugeres i 10 minutter ved 4 °C og 50 000 x g. Pelleten oppslemmes på nytt i 2,5 ml iskald tris-buffer, røres sammen med ytterligere 10 ml buffer, og det sentrifugeres som beskrevet. Pelleten oppslemmes så på nytt i buffer og fortynnes, hvorved man får et homogenat som inneholder 60 mg materiale/ml. 0,1 ml av suspensjonen, 100 ul av en 5 nM oppløsning av [<3>H]ketanserin, 100 ul av en oppløsning av testforbindelsen {konsentrasjon i området fra IO"<5> til 10~<10> mol pr. liter) tilsettes til inkubasjonsrørene og røres sammen til 1 ml med buffer. Rørene inkuberes i 15 minutter ved 37 °C. Etter av-slutning av inkubasjonen ved å senke rørene ned i et isbad filtreres den avkjølte suspensjon gjennom et glassfilter under vakuum. Filtrene vaskes 3 x med 5 ml kald buffer og overføres så til scintillasjonsrør. Filtrene analyseres ved hjelp av væske-scintillasjonsspektrometri i 8 ml Triton X scintillatorvæske.
Eksempel 1
En oppløsning av 4-karboksy-3-(4-klorbenzoyl)indol og 2-klor-l-metylpyridiniumjodid i N-metylpyrrolidon (NMP) behand-les med en oppløsning av 4-fenetylpiperazin og etyldiisopropyl-amin (EDIPA) i NMP og omrøres deretter ved romtemperatur i 3 timer. Blandingen opparbeides på vanlig måte, og råproduktet fås. Dette oppløses i aceton, og hydrokloridet utfelles ved å anvende vandig saltsyre. Etter tørking fås [3-(4-klorbenzoyl)-lH-indol-4-yl]-4-fenetylpiperazin-l-ylmetanonhydroklorid-hydrat, smp. 187,4 °C.
Forbindelsene nedenunder fås analogt: [3-(4-fluorbenzoyl)-lH-indol-4-yl]-(4-fenetylpiperazin-l-yl)metanon-hydroklorid, smp. 139-141 °C,

[3-(4-fluorbenzoyl)-lH-indol-5-yl]-{4-fenetylpiperazin-l-yl)metanon, hydroklorid, smp. 139-141 °C, [3-(4-klorbenzoyl)-lH-indol-5-yl]-{4-fenetylpiperazin-l-yl)metanon, smp. 168,5 °C,
[3-(4-fluorbenzoyl)-lH-indol-5-yl]-[4-(4-fluorfenetyl)-piperazin-l-yl]metanon-hydroklorid, smp. 198-200 °C,
[3-(furan-2-yl-karbonyl)-lH-indol-5-yl]-(4-fenetylpiperazin-l-yl)metanon-hydroklorid, smp. 268-269 °C,
[3-(tiofen-2-yl-karbonyl)-lH-indol-5-yl]-(4-fenetylpiperazin-l-yl)metanon-hydroklorid, smp. 191-192 °C,
Eksempel 2
Ved hjelp av katalytisk hydrogenering med hydrogen og Pd/C i metanol fås det fra [3-(4-klorbenzoyl)-lH-indol-4-yl]-[4-(4-fluorfenetyl)piperazin-l-yl]metanon forbindelsen: [3-benzoyl-lH-indol-4-yl]-[4-{4-fluorfenetyl)piperazin-l-yl] metanon-hydroklorid, smp. 257 °C.
De følgende eksempler vedrører farmasøytiske preparater.
Eksempel A
Injeksjonsflasker
En oppløsning av 100 g av en aktiv forbindelse med formel I og 5 g dinatriumhydrogenfosfat i 3 1 dobbeltdestillert vann reguleres til pH 6,5 ved å anvende 2 N saltsyre, det sterilfiltreres, fylles i injeksjonsflasker, lyofiliseres og lukkes aseptisk. Hver injeksjonsflaske inneholder 5 mg aktiv forbindelse.
Eksempel B
Suppositorier
En blanding av 20 g av en aktiv forbindelse med formel I smeltes sammen med 100 g soyalecitin og 1400 g kakaosmør, helles i støpeformer og får avkjøles. Hvert suppositorium inneholder 20 mg aktiv forbindelse.
Eksempel C
Oppløsning
En oppløsning fremstilles fra 1 g av en aktiv forbindelse med formel I, 9,38 g NaH2P04 x 2 H20, 28,48 g Na2HP04 x 12 H20 og 0,1 g benzalkoniumklorid i 940 ml dobbeltdestillert vann. Den reguleres til pH 6,8, røres ut til 1 1 og steriliseres ved bestråling.. Denne oppløsningen kan brukes i form av øyedråper.
Eksempel D
Salve
500 mg av en aktiv forbindelse med formel I blandes med 99,5 g vaselin under aseptiske betingelser.
Eksempel E
Tabletter
En blanding av 1 kg aktiv forbindelse med formel I,
4 kg laktose, 1,2 kg potetstivelse, 0,2 kg talkum og 0,1 kg magnesiumstearat presses på vanlig måte til tabletter, slik at hver tablett inneholder 10 mg aktiv forbindelse.
Eksempel F
Belagte tabletter
Analogt med eksempel E presses tabletter og belegges så på vanlig måte med et belegg av sukrose, potetstivelse, talkum, tragant og fargestoff.
Eksempel G
Kapsler
2 kg aktiv forbindelse med formel I fylles i harde gelatinkapsler på vanlig måte, slik at hver kapsel inneholder
20 mg av den aktive forbindelse.
Eksempel H
Ampuller
En oppløsning av 1 kg aktiv forbindelse med formel I i 60 1 dobbeltdestillert vann fylles i ampuller, lyofiliseres under aseptiske betingelser og lukkes aseptisk. Hver ampulle inneholder 10 mg aktiv forbindelse.
Farmakologisk forsøksrapport
Testdataene for noen representative forbindelser viser deres IC5o-vedier i M x liter'<1>, deres smeltepunkt i °C og eksemplet, siden og linjen ovenfor hvor forbindelsene er nevnt.
Affiniteten til 5-HT2A-reseptoren ble testet ved hjelp av metoden til J.E. Leysen et al., Molecular Pharmacology, 1982, 21: 301-314.
Claims (5)
1. Forbindelser,
karakterisert ved at de har formel I
hvor: R<1> er et fenylradikal som er usubstituert eller substi-
tuert med R<2>og/eller R<4>,R<2>, R<4>, R<5> og R<6> er i hvert tilfelle uavhengig av hverandre Hal, A, OA,
OH, CN eller Ci-Csacyl, R<3> er et fenylradikal som er usubstituert eller substi-
tuert med R<5> og/eller R<6>, eller er Het<2>,Het<2> er furyl eller tienyl, A er alkyl med 1-6 C-atomer, og Hal er F, Cl, Br eller I, og deres fysiologisk akseptable salter og solvater.
2. Fremgangsmåte for fremstilling av forbindelser med formel I ifølge krav 1,
karakterisert ved at a) en forbindelse med formel II
hvor L er Cl, Br, I eller en fri eller reaktivt, funksjonelt modifisert OH-gruppe, og R<3> har betydningen angitt i krav 1, omsettes med en forbindelse med formel III
hvor R<1> har betydningen angitt i krav 1, eller b) en forbindelse med formel IV
hvor R<3> har betydningen angitt i krav 1, omsettes med en forbindelse med formel V

hvor L er Cl, Br, I eller en fri eller reaktivt, funksjonelt modifisert OH-gruppe, og R<1> har betydningen angitt i krav 1, eller c) dersom det er hensiktsmessig, omdannes ett av radikalene R<1> og/eller R<3> til et annet radikal R<1>og/eller R<3> ved hjelp av spalting av f.eks. en OA-gruppe med dannelse av en OH-gruppe og/eller omdannelse av.en CHO-gruppe til en CN-gruppe, og/eller en erholdt base med formel I omdannes til et av dens salter ved behandling med en syre.
3. Forbindelser med formel I ifølge krav 1, og deres fysiologisk akseptable salter og solvater, for bruk som medikamenter.
4. Farmasøytisk preparat,
karakterisert ved at det omfatter minst én forbindelse med formel I ifølge krav 1, og deres fysiologisk akseptable salter og solvater, og eventuelt vehikler og/eller eksipienser, og eventuelt andre aktive forbindelser.
5. Anvendelse av forbindelser ifølge krav 1 og/eller av deres fysiologisk akseptable salter og solvater til fremstilling av et medikament for behandling av psykoser, schizofreni, depresjon, neurologiske forstyrrelser, hukommelsesforstyrrelser, Parkinsons sykdom, amyotrof lateralsklerose, Alzheimers sykdom, Huntingtons sykdom, slike spiseforstyrrelser som bulimi, anorexia nervosa, premenstruelt syndrom og/eller til positiv påvirkning av tvangsnevrotisk oppførsel (obsessiv-kompulsiv forstyrrelse, OCD) .
