Description
Oppfinnelsen vedrører fotostabile kosmetiske lysskjermings komposisjoner, som angitt i kravene 1-9, for beskyttelse av den menneskelige epidermis mot ultrafiolette stråler med bølgelengder mellom 280 og 320 nm (UV-B), samt en anvendelse som angitt i krav 10.
Særlig vedrører oppfinnelsen kosmetiske lysskjermings-komposisjoner som inneholder et lipofilt polysiloxan type UV-B skjermingsmiddel og et hydrofilt benzimidazol type UV-B skj ermingsmiddel.
I den europeiske patentpublikasjon EP-0813857 beskrives en solbeskyttelseskomposisjon som har en synergistisk UV-be-skyttelsesaktivitet som omfatter en vannfase; en første oljefase; en andre oljefase; et bløtgjørende middel for emulsjonstypen olje i vann og et annet bløtgjørende middel for emulsjonstypen vann i olje; et hydrofilt UV-A filter som f.eks. benzofenonderivater eller et hydrofilt UV-B filter som f.eks. 4-tert. butyl-4'-metoxydibenzoylmetan (PARSOL 1789®) eller et lipofilt UV-B filter som f.eks. octocrylene (UVINUL N-539®) eller et organosiloxanfilter som beskrevet i WO 93/04665 eller et organosiloxanfilter av benzotriazol typen som beskrevet i WO94/06404, hvor ved nevnte solbeskyttelseskomposisjon er karakterisert ved at de første og andre oljefaser er inkompatible.
Den ovenfor nevnte europeiske publikasjon nevner kun generelt at de kombinerte filtersubstanser har en synergistisk ultrafiolett beskyttende aktivitet uten å gi noen høring~~~ for hvordan velge en spesifikk filterkombinasjon for å oppnå en spesifikk synergistisk effekt. Videre referer ikke organosiloxanene som beskrevet i WO 93/04665 eller W094/- 06404 spesielt til organosiloxaner som inneholder en kromofor rest av benzmalonate typen.
Det er nå funnet at en solskjermingskomposisjon som omfatter et polysiloxan som har en chromofore rest av benzmalo-nattypen som lipofil UV-B skjermingsmiddel og et hydrofilt benzomidazol type UV-skjermingsmiddel frembringer synergistisk økede beskyttelsesregistreringer.
Ifølge krav 1 vedrører oppfinnelsen en kosmetisk lysskjermingskomposisjon som omfatter i en vandig fase 0,5 til 10 vekt% av 2-fenylbenzimidazol-sulfonisyre eller et salt derav, og minst en fettfase, som omfatter 1 til 30 wt% av en lineær eller syklisk polysiloksanforbindelse med den generelle formel Ia eller Ib,

Uttrykket "Ci-6-alkyl" henviser til grupper så som metyl, etyl, propyl, isopropyl, butyl, sec.butyl, isobutyl, pentyl og neopentyl. Utrrykket "Ci_6-alkoksy" henviser til de korresponderende alkoksygrupper.
Restene R er fortrinnsvis metyl.
Restene R<1> og R<2> er fortrinnsvis hydrogen, metoksy eller etoksy, mer foretrukket hydrogen eller en av R<1> og R<2> er hydrogen og den andre er metyl, metoksy eller etoksy.
Restene R<3> er fortrinnsvis metyl eller etyl, mer foretrukket etyl.
Fortrinnsvis er R<4> hydrogen eller metyl, R<5> og R6 er hydrogen og n er 1.
Egnede salter av 2-fenylbenzimidazol-sulfonsyre er alkali metallsalter, så som natrium- eller kaliumsalter, ammonium-salter, morfolinsalter, salter av primære, sekundære og tertiære aminer som monoetanolaminsalter, dietanolaminsalter og liknende.
Polysiloksånforbindelser som har en gruppe A med generell"~ formel Ila og Ilb og deres fremstilling er beskrevet i det europeiske patent EP 0538431 Bl. Disse polysiloksan-forbindelser er foretrukne.
De polysiloksånforbindelser som har en gruppe A med generell formel lic og deres fremstilling er beskrevet i euro-peisk patent EP 0358584 Bl.
I de lineære polysiloksånforbindelser i hht. til formel Ia kan den kromoforbærende rest A være forbundet til ende-grupppene til polysiloksan (X=A) eller kan være statisk fordelt.
Lineære polysiloksånforbindelser hvori den kromoforbærende rest A er statistisk fordelt er foretrukne. Nevnte foretrukne polysiloksånforbindelser har minst én enhet som bærer den kromofore rest (s=l), fortrinnsvis har s en verdi på fra 2 til 10, mer foretrukket et statistisk gjennomsnitt på 4. Antallet av r av de andre silikonenhetene som foreligger i polysiloksånforbindelsene er fortrinnsvis 5 til 150, mer foretrukket et statistisk gjennomsnitt på 60.
Polysiloksånforbindelser hvori 20% eller mindre, fortrinnsvis mindre enn 10% av de totale siloksanenheter er enheter som bærer en kromofor rest, er foretrukne med hensyn til de kosmetiske egenskaper.
Forholdet mellom polysiloksanenheter som har en kromofor rest A med formel Ila, og de som har en kromofor rest A med formelen Ilb er ikke kritisk. Nevnte forhold kan være 1:1 til 19:1, fortrinnsvis 2:1 til 9:1, mer foretrukket 4:1.
Konsentrasjonen av polysiloksånforbindelsen i den kosmetiske lysskjemingskomposisjonen er fortrinnsvis 2-20 vekt%, mer foretrukket 5 vekt%.
Konsentrasjonen av 2-fenylbenzimidazol sulfonsyre eller ét~ salt derav er fortrinnsvis 0,5 til 5 vekt%, mer foretrukket 1-2 vekt%.
Forholdet av 2-fenylbenzimidazol sulfonsyre eller et salt derav til polysiloksånforbindelsen som definert ovenfor, er ikke kritisk. For eksempel er forholdet 1:1 til 1:20, fortrinnsvis 1:5.
Følgelig inneholder en foretrukket kosmetisk lysskjermingskomposisjon i en vandig fase 0,5-5 vekt% av 2-fenylbenzimidazol-sulfonsyre eller et salt derav, og minst en fettfase, som inneholder 2-20 vekt% av en lineær polysiloksånforbindelse med generell formel Ia, hvor X betyr metyl;

En mer foretrukket kosmetisk lysskjermingskomposisjon inneholder og i en vandig fase 1-2 vekt% av 2-fenyl-benzimidazol-sulfonsyre eller et salt derav og minst en fettfase, som inneholder 5 vekt% av en lineær polysiloksånforbindelse med den generelle formel Ia, hvor X betyr metyl;

Polysiloksånforbindelsene Ia eller Ib hvori A er en rest med formel Ila eller Ilb kan fremstilles som beskrevet i EP 0538431 Bl ved silylering av de korresponderende benzal-malonater i henhold til det følgende reaksjonsskjema:
hvori R<1>, R2 og R<3> er som definert ovenfor.
Silyleringen av 4-(2-propynyloks)fenyl-metylen-dietylester kan utføres ved å ta i bruk kjente prosedyrer for addisjon av silisiumbundne hydrdogenatomer til grupper som inneholder alifatisk umettethet. Slike reaksjoner katalyseres generelt av et platingruppemetall eller et kompleks av et slikt metall. Eksempler på katalysatorer som kan anvendes er platin på kull, klorplatinsyre, platin acetyl acetonat, komplekser av platinforbindelser med umettede forbindelser, f.eks. olefiner og devinyl disiloksaner, komplekser av rhodium og palladium forbindelser og komplekser av platinforbindelser båret på uorganiske substrater. Addisjons-reaksjonen kan utføres ved redusert, atmosfærisk eller øket trykk. Et løsningsmiddel kan anvendes, f.eks. toluen eller xylen i reaksjonsblandingen selv om nærvær av løsningsmid-del ikke er avgjørende. Det er også foretrukket å utføre reaksjonen ved øket reaksjonstemperatur, f.eks. fra 50°C opp til 150°C.
Fremstillingen av de nye lysskjermingskomposisjoner omfatter innlemmelsen av en polysiloksånforbindelse som definert ovenfor, og 2-fenyl-benzimidazol-sulfonsyre eller et salt derav, ev. i kombinasjon med andre kjente UV-A og/eller UV-B filtere i en kosmetisk base som er vanlig for lys-skj ermingsmidler.
Egnede UV B filtere, dvs. substanser som har absorpsjons-maksima mellom 290 og 320 nm er f.eks. de følgende organiske forbindelser hvilket tilhører de videste klasser av substanser: p-Aminobenzoisyrderivater så som etyl-, propyl-, butyl-og isobutyl p-aminobenzoat og liknende;
Akrylater så som 2-etylhexyl 2-cyano-3,3 difenylakrylat (octokrylen), etyl 2-cyano-3,3 difenylakrylat og liknende;
Anilinderivater så som metyl anilium methosulfat og liknende;
Anthranilisyre derivater så som mentylantramilat og liknende;
Benzpfenonderivater så som benzofenon-3, benzofenon-4 og liknende;
Kamferderivater så som metyl-benzyliden kamfer (PARSOL 5000), 3-benzyliden kamfer, kamfer benzalkonium metosulfat, polyacrylamidometyl benzyliden, kamfer, sulfa benzyliden kamfer, sulfometyl benzyliden kamfer, tereftaliden-kamfer sulfonsyre og liknende;
Cinnamatderivater så som octyl methoxycynnamate (PARSON MCX) eller ethoxyetyl methoxycinnamat og liknende samt kanelsyrederivater bundet til siloksaner.
Gallussyre så som digalloyl trioleat o.l.;
Salisylatderivater så som isopropylbenzyl salisylat, butylsalisylat, octylsalisylat (Neo Heliopan OS), isooctyl salisylat eller homomentylsalisylat (homosalat, Heliopan) og liknende;
Triazolderivater så som hydroksyfenylbenztriazol, 2-2'metylene-bis-(6-(2H-benzotriazol-2-yl)-4-(1,1, 3, 3, -te-trametylbutyl)-fenol (TINOSORB M) og liknende;
Triazolderivater så som oktyl triazon (Uvinul T-150), dioktyl butamido triazon (Uvasorb HEB) og liknende.
Pigmenter så som mikropartikulært Ti02, ZnO og liknende.
Utrykket "mikropartikulært" henviser til en partikkel-størrelse fra 5 nm - 200nm, særlig fra 15 - 100 nm. Ti02 partiklene kan også være belagte med metalloksider, så som f.eks. aluminium eller zirkoniumoksider eller med organiske belegg så som f.eks. polyoler, methicon, aluminiumsstearat, alkylsilan. Slike belegg er velkjente i faget.
Formuleringen kan videre inneholde UV-A filtere, så som
Dibenzoylmetanderivater så som 4-tert. butyl-4'me-toksydibenzoyl-metan og liknende;
Triazineforbindelser som beskrevet i den europeiske patentpublikasjonen EP 0693483 Al, EP 0704437 A2, EP 0704444 Al og EP 0780382 Al, f.eks. 2,2 ' -[6-(4-metok-sydfenyl)-1,3,5-triazine-2,4-diyl] bis [5-[2-etylhexyl) oxy]-; som er tilgjengelige under handelsnavnet TINOSORB S fra Ciba Speciality Chemicals Holding Switzerland.
Som kosmetiske baser som er vanlige for lysskjermingskomposisjoner innenfor omfanget til foreliggende oppfin-nelse kan det anvendes enhver konvensjonell formulering som korresponderer til de kosmetiske krav, f.eks. kremer, lotion, emulsjoner, salver, gel, oppløsninger, sprayer, , sticks og melker; se også [Sunscreens, Development, Eva-luation and Regulatory Aspects, ed. N.Y. Lowe, N.A. Shaath, Marcel Dekker, Inc. New York and Basel, 1990].
De følgende eksempler forklarer oppfinnelsen mer i detalj.
Eksempel 1
Fremstilling av { (4-[2-propynyloxy) fenyl] metylen} dietylester som beskrevet i EP 0538431 Bl.
Til en rørt suspensjon av 4-hydrokxybenzaldehyd (425,8 g) og K2CO3 (807,6 g)i aceton (2,96 ml) ved reflukstemperatur 60°C og under en nitrogenatmosfære, ble det dråpevis tilsatt 3-bromo-propyn (502,4 g) over et tidsrom på to timer. Reaksjonen ble varmet til refluks i mer enn 33 timer. Etter avkjøling til romtemperatur ble reaksjonsblandingen filtrert, overskudd av K2CO3 ble fjernet, og det ble vasket flere ganger med aceton. Filtratet ble vasket med mettede, vandige oppløsninger av NaHC03 og NaCl. Den vandige fase ble ekstrahert med dietyleter. De samlede organiske ekstrakter ble tørket over Na2S04, filtrert og konsentrert til et volum på 1 1. Oppløsningen ble holdt i kjøleskap over natten. Krystallene ble filtrert fra og vasket med kald dietyleter. Filtratet ble holdt i kjøle-skapet, og noen flere krystaller ble utfelt og fjernet. Denne prosedyre ble gjentatt tre ganger hvilket resulterte i 1.385 g av 4-(2-propynyloxy) benzaldehyd i 83% utbytte. Materialet ble analysert ved gaskromatografi og viste seg å være 99,9% rent. Den resulterende forbindelse(449,2 g) ble tilsatt i små mengder til en rørt oppløsning av dietyl-malonat (448,5 g), piperidin (23,84 g), toluen (1.400 ml) og eddiksyre (59 g) ved 50°C. Eddiksyren var blitt tilsatt i tre like porsjoner etter hhv. 1, 1,5 og 2 timer. Reaksjonsblandingen ble oppvarmet til refluks. Etter 4 timer fikk blandingen bli avkjølt til romtemperatur og vasket med mettet, vandig oppløsning av NaHCC>3 og NaCl, tørket med Na2S04, filtrert og konsentrert, hvilket ga 853,4 g av et mørkebrunt oljeaktig produkt. Dietyleter (458 mlL) og n-hexan (358 ml)ble tilsatt, og oppløsningen ble holdt i kjøleskapet over natt. Oppløsningen ble filtert, hvilket ga 564,8 g med lysebrune krystaller (67% utbytte) som hadde et smeltepunkt på 45,5 til 48°C. Omkrystal-lisering av etanol og n-hexan, 543 g av tittelforbindelse som lysebrune krystaller. Materialet ble analysert ved gaskromatografi, og viste seg å være 99,9% rene.
Eksempel 2
Fremstilling av en organosiloksanforbindelse med generell formel Ia hvori r betyr metyl, s er 0, r er 20, X er A og A betyr en bénzalmalonatrest med formel Ila og Ilb hvori R<1*>"" og R2 er hydrogen, R<3> er etyl og R<4>, R<5> og R<6> er hydrogen, n er 1 som beskrevet i EP 053 431 Bl.
5 g av {[4-(2 propynyloxy) fenyl] metylen}-dietyl ester ble oppløst i 20 g toluen og oppvarmet under nitrogen til omtrent 80°C. 13,2 g av et hydrosiloksan som hadde en poly-meriseringsgrad på 20 og 10 mpc SiH grupper (3,62% SiH) ble deretter tilsatt dråpevis etter at et platin-divinyl-tetra-metyl-disiloksan kompleks også var tilsatt, hvilket ga IO"<4>
mol med Pt per mol SiH av hydrosiloksane. Blandingen ble oppvarmet til refluks og opprettholdt inntil all SiH var forsvunnet fra den infrarøde spektroskopiske anlyse. Den fikk deretter avkjøles til romtemperatur. Toluen ble fordampet for å gi etter vasking 16,5 g av en noe brun polymer som hadde en gjennomsnittlig struktur A-[ (CH3) 2SiO]2o-A, hvori A er en rest med formel Hai og Ilbi.
Eksempel 3
Fremstilling av en organosiloksanforbindelse som har den generelle formelen Ia hvori R betyr metyl, r er 59, s er 4, X er metyl og A betyr an benzalmalonatrest med formel Ila og Ilb hvori R<1> og R2 er hydrogen, R3 er etyl, R<4>, R5 og R<6 >ar hydrogen som beskrevet i den europeiske publikasjon EP 0709080 Al.
13,28 g av {[4-(2-propynyloxy) fenyl] metylen}-dietyl ester ble oppløst i 75 g toluen og oppvarmet under nitrogen til omtrent 70°C. 4 4 g av en hydrosiloksan som hadde en poly-meriseringggrad på 65 og 6 mpc SiH grupper (2,36% SiH) ble^.
deretter tilsatt dråpevis etter at et platin-divinyl-tetra-metyl-disiloksan kompleks også var tilsatt, hvilket ga IO"<4>
mol Pt per mol av SiH av hydrosiloksan. Blandingen ble oppvarmet til refluks og opprettholdt inntil all SiH var forsvunnet ved infrarød spektroskopisk analyse. Den fikk
deretter avkjøles til romtemperatur. Toluen ble fordampet for å etterlate etter vasking 52 g av en brun, viskos polymer som hadde den gjennomsnittlige struktur (CH3)3SiO-
[ (CH3) 2SiO]59-[ (CH3) ASiO]4-Si (CH3) 3 hvori A har formelen Hai og Ilbi.
Forholdet av forbindelser som har en rest Hai til forbindelser som har en rest Ilbi er 4:1.
Eksempel 4
Egenede kosmetiske lysskjermingskomposisjoner.
En solskjermings O/V lotion som inneholder 5 vekt% av en polysiloksan med formel Ia i hht. til eksempel 3 og 1 vekt% av 2-fenylbenzimidazol-sulfonsyre, ble fremstilt med de følgende ingredienser:
CTFA: Cosmetic, Toiletry and Fragrance Association.
Del A: Tilsatt og smeltet i reaktoren.
Del B: Blandet, nøytralisert og oppvarmet på koleplate
til 85°C.
Del C: Tilsatt ved 50°C.
Eksempel 5
Bestemmelse av solbeskyttelsesfaktoren.
Bestemmelsen av solbeskyttelsesfaktoren ble utført i henhold til COLIPA protokollen (European Cosmetic, Toiletry and Perfumery Association, sobeskyttelsesfaktor testmetode, oktober 1994) .
En solarsimulator SU 2000 har en lampeintensitet i området fra 5.0 til 5,5 mW/cm<2> ble brukt. Strålingsområdet 6x1 cm<2>; doseprogresjon 25%.
De følgende filterkombinasjoner ble formulert under anvendelse av den samme kosmetiske base som beskrevet i eksempel 4 for å gi en stabil solskjermingslotion med identiske for-de lings kar a kt ert rekk.
En applikasjonsdose på 2 mg/cm<2> ble brukt i henhold til COLIPA protokollen for in vivo SPF målinger i mennesker, på et applikasjonsområde på 50 cm<2>.
Som sammenlikning ble de følgende solskjermings-komposisjoner testet: En solskjermingskomposisjon som inneholdt en polysiloksanforbindelse i henhold til eksempel 3, og et annet kjent lipofilt UV-B filter, nemlig 2-etylhexyl-p-metoksycinnamat (OMC) (tilgjengelig under handelsnavnet PARSOL MCX).
En solbeskyttelseskomposisjon som inneholdt en polysil-oksånf orbindelse i henhold til eksempel 3, og et annet kjent hydrofilt UV-B filter, nemlig dietanolaminsalter til p-methoxykanelsyre (MC-DEA) (tilgjengelig under handelsnavnet PARSOL Hydro).
Resultatene er oppført i den følgende tabell:
P3: Polysiloksan i henhold til eksempel 3.
PBSA: 2-fenylbenzimidazol-sulfonsyre.
OMC: 2-etylhexyl-p-metoxycinnamat.
MC-DEA: Dietanolaminsalt av p-metoxykanalsyre.
In vivo SBF: Gjennomsnittsverdi for bestemmelse på 5
frivillige.
Forventet SBF: Beregnet verdi ved addisjon av in vivo SBF-verdi for 10 vekt% P3 (4,7) til in vivo SPF---verdien for 2 vekt% for den korresponderende verdien for de andre filtere, dividert med 2.
Synergi: % avvik av in vivo SPF fra forventet (beregnet)SPF.
Kombinasjonen av PBSA og polysiloksan i henhold til oppfinnelsen utviser en uproporsjonal økning av SPF. Vanligvis er SPF i filterkombinasjoner meget nær til beregnede verdi fra aktiviteten til de enkelte filtere, derfor vises en synergistisk effekt for denne kombinasjon.
Claims (10)
1. Kosmetisk lysbeskyttelseskomposisjon omfatter i en vandig fase 0,5-10 vekt% av 2-fenylbenzimidazol-sulfonsyre eller et salt derav, og minst en fettfase, som omfatter 1-30 vekt% av en lineær eller syklisk polysiloksånforbindelse med den generelle formel Ia eller Ib,
hvor
X betyr R eller A;
A betyr en gruppe med formel Ila, Ilb eller Ile;
R betyr hydrogen, Ci-6-alkyl eller fenyl;
R<1> og R<2> betyr hver uavhengig hydrogen, hydroksy, Ci_6-
alkyl eller Ci-6-alkoksy;
R<3> betyr Ci-6-alkyl;
R<4> betyr hydrogen eller Ci_6-alkyl;
R5 og R<6> betyr hver uavhengig hydrogen eller Ci-6-alkyl;
r har en verdi fra 0-250;
s har en verdi på fra 0-20;
r + s har en verdi på minst 3;
t har en verdi på fra 0-10;
v har en verdi på fra 0-10; og
v + t har en verdi på minst 3; n har en verdi på fra 1-6;
med det forbehold at i det tilfellet s er 0 er minst en X lik A.
2. Kosmetisk lysbeskyttelseskomposisjon i henhold til krav 1, hvori A er en rest med formel Ila eller Ilb.
3. Kosmetisk lysbeskyttelseskomposisjon i henhold til krav 1 eller 2, hvori forholdet av organosiloksanforbindelser som har en kromofor rest med formel Ila til organosiloksanforbindelser som har en kromofor rest med formelen Ilb er 1:1 til 19:1, fortrinnsvis 2:1 til 9:1, mer foretrukket 4:1.
4. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1 til 3, hvori R er metyl.
5. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1 til 4, hvori R<1> og R2 er hydrogen, metoksy eller etoksy, eller en av R<1> og R2 er hydrogen og den andre er metyl, metoksy eller etoksy.
6. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1 til 5, hvori R<3> er metyl eller etyl.
7. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1 til 6, hvorav R<4> er hydrogen eller metyl; R<5 >og R<6> er hydrogen og n er 1.
8. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1-7 som inneholder i en vandig fase 0,5-5, fortrinnsvis 1-2 vekt% av 2-fenylbenzimidazol-sulfonsyre eller et salt derav, og minst en fettfase, som omfatter 2-20, fortrinnsvis 5 vekt% av en lineær polysil-oksånf orbindelse med den generelle formel Ia, hvor X betyr metyl;
A betyr en gruppe med formel Ila eller Ilb;
R betyr metyl
R<1> og R<2> betyr hydrogen, metoksy eller etoksy, eller en av
R<1> og R2 er hydrogen og den andre er metyl, metoksy eller etoksy, fortrinnsvis hydrogen;
R<3> betyr metyl eller etyl, fortrinnsvis etyl;
R<4> betyr hydrogen eller metyl, fortrinnsvis
hydrogen;.
R<5> og R<6> betyr hydrogen;
r er 5-150, fortrinnsvis 4;
s er 2-10, fortrinnsvis 60;
n har en verdi på 1.
9. Kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1-8, hvori nevnte komposisjon inneholder i til-legg vanlig UV-A filter og/eller UV-B filtere.
10. Anvendelsen av en kosmetisk lysbeskyttelseskomposisjon i henhold til et av kravene 1-9 som et absorberende middel for det ultrafiolette lys.
