Description
Denne oppfinnelse angår en analogifremgangsmåte for fremstilling av oligomerer som kan finne anvendelse som legemidler. Disse oligomerene er anioniske forbindelser som
har særlig verdifulle virkninger mot humant immunsvikt virus,
og oligomerene er derfor egnet ved behandling av AIDS
(Acquired Immune Deficiency Syndrome).
Det foregår for tiden en betydelig forskning for å
utvikle kurative behandlingsmåter for virale infeksjoner i mennesker og dyr. Særlig er forekomsten av AIDS og det AIDS-relaterte kompleks (ARC) hos mennesker, under utvikling med en alarmerende hastighet. Overlevelsesraten over fem år for personer med AIDS er nedslående, og AIDS-pasienter som har fått sitt immunsystem alvorlig svekket av infeksjonen, lider av mange opportunistiske infeksjoner, innbefattet Kaposi 's sarkom og Pneumocystis carninii pneumonia. Det er ikke kjent noen helbredelsesmåte for AIDS, og de behandlinger som for tiden benyttes, mangler i stor utstrekning rimelig bevis på effekt, og har adskillige uønskede bivirkninger. Frykten for sykdommen har ført til at personer som har eller mistenkes for å ha sykdommen, fryses ut eller diskrimineres.
Retrovira utgjør en klasse av ribonukleinsyre (RNA)-virus som ved sin replikasjon benytter revers transkriptase for å danne en tråd av komplementært DNA (cDNA) hvorfra det dannes et dobbelt-trådet proviralt DNA. Dette provirale DNA inkorporeres deretter vilkårlig i vertscellens kromosomale DNA, hvorved viral replikasjon gjennom senere translasjon av den virale informasjon fra det integrerte virale genom, muliggjøres.
Mange av de kjente retrovira er onkogene eller tumor-forårsakende. De to først oppdagede humane retrovirus,
betegnet human T-celle leukemivirus I og II eller HTLV-I og II, viste seg faktisk å forårsake sjeldne humane leukemier etter infeksjon av T-lymfocytter. Det tredje av slike human-virus som ble oppdaget, HTLV-III som nå betegnes HIV, viste seg å forårsake celledød etter infeksjon av T-lymfocytter og er identifisert som det agens som forårsaker AIDS og ARC.
Envelope-proteinet til HIV er et 160 kDa glykoprotein.
Proteinet spaltes av en protease for å gi et 12 0 kDa eksternt protein, gp 120, og et transmembran-glykoprotein, gp 41.
gp 120-proteinet inneholder arainosyresekvensen som gjenkjenner CD4-antigenet på humane hjelper-T-celler (T4).
En mulighet som utforskes er å forebygge bindingen av HIV til dens mål, de humane T4-celler. Disse T4-cellene har et spesifikt område, et CD4-antigen, som påvirker gp 120. Dersom denne interaksjon kan brytes, kan vertscelle-infeksjonen hemmes.
Interferens med dannelsen av det virale envelope-glykoprotein skulle kunne forhindre den initiale virus-vertscelle-interaksjon eller den senere fusjon, eller forhindre viral duplikasjon ved å forhindre oppbygningen av det bestemte glykoprotein som fordres for å fullføre virusmembranen. Det er publisert [se H.A. Blough et al., Biochem. Biophys. Res. Comm. 141(1), 33-38 (1986)] at de ikke-spesifikke glykosylerings-hemmerne 2-deoksy-D-glukose og S-hydroksy-norvalin, hemmer HIV glykoproteiners ekspresjon og blokkerer dannelse av syncytier. Viral multiplikasjon av HIV-infiserte celler behandlet med disse midler stanses, antagelig fordi det nødvendige glykoprotein for den virale membrandannelse ikke er tilgjengelig. I en annen publikasjon [W. McDowell et al., Biochemistry 24(27), 8145-52 (1985)], ble glykosylerings-hemmeren 2-deoksy-2-fluor-D-mannose funnet å hemme antiviral aktivitet overfor influenza-infiserte celler ved å forhindre glykosyleringen av viralt membranprotein. Ifølge rapporten ble også den antivirale aktivitet av 2-deoksy-glukose og 2-deoksy-2-fluorglukose studert, og det ble fastslått at begge hemmet viral protein-glykosylering gjennom ulike mekanismer. Andre kjente glykosylerings-nemmere har imidlertid vist seg å ikke ha noen antiviral aktivitet. Den antivirale aktivitet av glykosylerings-hemmere vis a vis virus i sin alminnelighet og den virale aktivitet i særdeleshet, er ganske uforutsigbar.
Det har vært beskrevet i sørafrikansk patent 90/0094 av 31. oktober 1990 at en renset form av heparin, et sulfatert polysakkarid, bindes gjennom interaksjoner til et viralt protein som er ansvarlig for gjenkjenningen av celler og dermed gir en begrenset hemming av vertscelle-infeksjon. Heparin har imidlertid noen bivirkninger, spesielt hemoragi og øket koagulasjonstid, så vel som trombocytopeni. Bruk av heparin er kontraindisert i pasienter som har blødninger, eller har hemofili, purpura, trombocytopeni, intrakraniell hemoragi, bakteriell endokarditt, aktiv tuberkulose, forøket kapillær-permeabilitet, ulcerative lesjoner i gastrointestinaltractus, alvorlig hypertensjon, truende abort eller vicerale karcinomer. Kontraindikasjonen for anvendelse til blødere er særlig betenkelig siden mange slike personer nå er HIV-positive.
Det har lenge vært kjent at visse syntetiske, vann-oppløselige polymerer oppviser et bredt spekter av biologisk aktivitet [R.M. Ottenbrite i "Biological Activities of Polymers", Amer. Chem. Soc. Symp. Ser. No. 182, s. 205-220, red. CE. Carraher & C.G. Gebelein (1982)]. En kopolymer av divinyleter og maleinsyre-anhydrid er vist å være aktiv overfor en rekke virus, og bruken av denne innen cancer-kjemoterapien har vært gjenstand for studier i årevis [Breslow, D.S. Pure and Applied Chem., 46, 103 (1976)]. Polyakryl-, polymetakryl- og en rekke andre vann-oppløselige polymerer med alifatisk skjelett har også vist seg å ha et bredt spektrum av biologiske virkninger [W. Regelson et al., Nature, 186, 778. (1960)] . Den ekstreme toksisitet av disse polymerene har uheldigvis hindret deres kliniske anvendelse. Polymerene har dessuten en høy molekylvekt og kan ikke passere gjennom de renale membranene.
Det er gjort forsøk på å omgå toksisitets- og utskillingsproblemene ved syntese av lavmolekylær (1000 til 10.000) alifatiske polymerer [R.M. Ottenbrite i "Biological Activities of Polymers", Amer. Chem. Soc. Symp. Ser. No. 182, s. 205-220, red. CE. Carraher & C.G. Gebelein (1982)]. Det har vist seg at slike polymerer er mindre toksiske, men de har betydelig redusert antiviral aktivitet. Disse lavmolekylære alifatiske polymerene kan klassifiseres som "random coil"-polymerer. Slike polymerer har en uforutsigbar konfigurasjon på grunn av fleksibiliteten i molekyl-skjelettets tilkoblingsgrupper. Konfigurasjonen av "random coil"-polymerer i oppløsning kan generelt beskrives som globulær.Virkningsmekanismen av slike vann-oppløselige polymerer er ukjent, men et postulat er at polymeret binder seg til den virale membran, f.eks. encefalomyokarditt, gjennom en ionisk tiltrekning som setter viruset ut av stand til å infisere vertscellene.
Et ytterligere forsøk på anvendelse av en syntetisk polymer består i å anbringe ioniske grupper som har en mer definert geometri, på polymerets skjelett. Det finnes adskillige eksempler på ikke-ioniske, syntetiske polymerer som har en mer lineær geometri i ikke-vandig oppløsning enn de alifatiske polymerene beskrevet ovenfor [J. Macromolecular Sci-Reviews in Macromol. Chem. Phys. C26(4), 551 (1986)]. Involverte faktorer som forårsaker denne "non-random coil"-struktur er komplekse og mangelfullt klarlagte. Generelt har slike polymerer enten et meget begrenset antall dreibare bindinger som ikke er parallelle med polymeraksen, eller forekommer hydrogen-bindings- eller dipolare interaksjoner som favoriserer lineære strukturer. Disse polymerene angis å ha et "stivt skjelett". Et polyamid avledet fra tereftalsyre og p-diaminobenzen (kommersielt kjent som Kevlar™ fra DuPont) er et velkjent eksempel på slike polymerer.
Syntetiske, vann-oppløselige, stive polymerer er adskillig mindre vanlig, men noen få høymolekylære eksempler er kjent (se f.eks. US-patent 4.824.916 og 4.895.660). Denne polymertype "non-random coil"-struktur resulterer i høye oppløsnings-viskositeter for en gitt molekylvekt og konsentrasj on.
Det ville helt klart vært ønskelig å finne en behandling og AIDS- og ARC-helbredeIse som oppviste minimalt eller ingen bivirkninger og utgjorde en klar forbedring i forhold til de polymerer som tidligere er anvendt som farmasøytika.
Det er nå oppdaget at anioniske oligomerer hemmer viral replikasjon uten de bivirkninger som heparin og kjente polymerer oppviser. Oligomerene har en ordnet anion-avstand, et stivt skjelett og er vann-oppløselige.
De nye oligomerene fremstillet i henhold til foreliggende oppfinnelse, er anioniske karbonyl-holdige polyurea-forbindelser, som har en antalls-midlere molekylvekt, Mn, på
<10.000 og er vann-oppløselige, har et stivt skjelett og en regelmessig anion-avstand. Oligomerene innbefatter de av deres salter som er farmasøytisk akseptable når de benyttes som farmasøytiske midler. Andre anvendelser for disse anioniske oligomerer er som virksomme fortykningsmidler i vandige oppløsninger eller som milde ioniske overflateaktive midler. Generelt har vann-oppløselige polymerer, innbefattet oligomerene fremstillet i henhold til foreliggende oppfinnelse, et bredt bruksområde som fortykningsmidler, dispergeringsmidler og flokkulanter. De foreliggende oligomerene kan benyttes til bestemte formål innen olje-området, grubedrift, papirfremstilling, tekstilproduksjon, fremstilling av kosmetiske ingredienser og
næringsmiddelproduksjon. Disse lavmolekylære polymerene, dvs. oligomerene, kan dessuten benyttes som utgangsmaterialer for fremstilling av høymolekylære polymerer og kopolymerer.
Oppfinnelsen angår således en fremgangsmåte for fremstilling av en vann-oppløselig oligomer med stivt skjelett, som har en molekylvekt på mindre enn (<)10.000 som omfatter repeterende enheter koblet gjennom karbonyl-ledd, der oligomerene har anioniske grupper og i det vesentlige lineær geometri, slik at det i et vandig miljø foreligger en regelmessig avstand mellom anioniske grupper. Fortrinnsvis har hver repeterende enhet minst to anioniske grupper og oligomerene antar fortrinnsvis en lineær geometri.
De nye oligomerene som fremstilles i henhold til foreliggende oppfinnelse er polyurea-forbindelser som har en antalls-midlere molekylvekt, Mn, på < 10.000 som er vann-oppløselig, har et stivt skjelett, en regelmessig anion-avstand og en tilnærmet lineær geometri i et vandig miljø. Oligomerene har fortrinnsvis lineært skjelett og kan også foreligge i saltform, hvorav de farmasøytisk akseptable salter er særlig foretrukket.
Oligomerene fremstillet i henhold til oppfinnelsen, er representert gjennom formel I:

Uttrykket "farmasøytisk akseptabelt kation" er å forstå som et kation som er akseptabelt for farmasøytiske formål. De kationer som ikke er av vesentlig giftighet i den mengde som administreres for å oppnå den ønskede effekt og som ikke har en egen klar farmakologisk aktivitet, er inkludert i uttrykket "farmasøytisk akseptabelt kation". Disse saltene innbefatter for eksempel salter av alkalimetaller, så som natrium og kalium; jordalkalimetaller, så som kalsium og magnesium; ammonium; lettmetaller fra gruppe HIA innbefattet aluminium; og organiske primære, sekundære og tertiære aminer, så som trialkylaminer, innbefattet trietylamin, prokain , dibenzylamin, N,N '-dibenzyletylendiamin, dihydroabietylamin, N- (C-l-Qi) alkylpiperidin og eventuelt andre passende aminer. Natrium- og kaliumsalter er å foretrekke. Uttrykket "farmasøytisk akseptabel" betyr egnet for administrasjon til varmblodige dyr, spesielt mennesker og innebærer at de- er ugiftige, f.eks. egnet for farmasøytisk bruk og ikke giftig for det varmblodige dyr. De farmasøytisk akseptable kationer av oligomerer av de nye forbindelsene fremstilles ved konvensjonlle ionebytter-prosesser eller ved å behandle R<1->syren med en passende base.
Når de foreliggende oligomerer er beregnet for annet enn farmasøytisk bruk, kan salter som ellers ikke ville være så akseptable, benyttes. Eksempler på slike ytterligere salter innbefatter barium, sink og titan.
Oligomerene fremstillet i henhold til foreliggende oppfinnelse, er lavmolekylære, vann-oppløselige polymerer med stivt skjelett. Oligomerene har dessuten regelmessig anion-avstand. Med "regelmessig anion-avstand" eller "regelmessig avstand mellom anioniske grupper" menes at de anioniske gruppene (R<1>) forekommer i polymerskjelettet med intervaller som bestemmes av det anvendte utgangsmateriale, og at forekomsten av de anioniske gruppene kontrolleres på en forutsigbar måte. Uten å ta stilling til noen bestemt teori, antas de anioniske grupper i oligomerene å være den del som bindes til HIV og/eller cellemembranen og derved forstyrrer replikasjonsevnen til viruset.
Uttrykket "tilnærmet lineær geometri" i et vandig miljø refererer til oppløsnings-konfigurasjonen av oligomeren. En metode som er velkjent på området for karakterisering av oppløsnings-konfigurasjonen av polymere molekyler er basert på den følgende formel kjent som Mark-Houwink-ligningen ["Introduction to Physical Polymer Science" red. L.H. Sperling, pub. John Wiley & Sons (1985), s. 81-83],
hvor ir er egenvi skos it eten; M er den vekt-midlere molekylvekt; K er en konstant som har relasjon til kjedebindings-dimensjonen og a er en konstant bestemt av polymer-konfigurasjonen. Egenviskositeten [ ir] for en "random coil"-polymer er 0,5<a<0,9; og for en lineær polymer 0,9<=o;<l,8. Denne formel relaterer oppløsningsviskotitet " ir" til molekylvekten "M". Når det gjelder foreliggende oppfinnelse er lineære polymerer definert som polymerer som har "a"-verdier større eller lik 0,9. For en "rigid rod"-polymer er den teoretiske øvre grense 1,8. For en gitt molekylvekt, vil en høyere oppløsningsviskositet oppnås fra polymerer med lineær konfigurasjon sammenlignet med polymerer som forekommer som en "random coil". En ytterligere faktor er at "ot"-verdien er en funksjon av det anvendte oppløsningsmiddel. For en gitt vann-oppløselig polymer kan "o;" være forskjellig ved ulike salt-konsentrasjoner. I forbindelse med denne oppfinnelse settes saltkonsentrasjonen ved de nivåer som forekommer i serum (ca.
80 g/L NaCl, 4 g/L KC1).
I denne sammenheng benyttes betegnelsen "oligomer" for alle mulige verdier for n, dvs. 3 opp til15. Oligomerene er fortrinnsvis lineære. Selvsagt vil n-verdien være direkte relatert til molekylvekten av den resulterende oligomer. Det er vesentlig at disse oligomerene har tilstrekkelig lav molekylvekt slik at de kan passere nyrenes utskillingsmembran, men være i stand til å hemme HIV-viruset. Den gjennomsnittlige molekylvekt styres av reagensenes støkiometriske forhold. Den antalls-midlere molekylvekt (Mn) er <10.000, fortrinnsvis fra ca. 500 til ca. 10.000 og helst fra ca. 1000 til ca. 6000.
I sammenheng med foreliggende oppfinnelse anses de her beskrevne oligomerer og fysiologisk akseptable salter derav, som likeverdige. Fysiologisk akseptable salter refererer seg til salter av de baser som vil danne et salt med minst én sur gruppe i Rx-gruppen og som ikke vil forårsake vesentlige uheldige fysiologiske effekter når de administreres som her beskrevet. Egnede baser innbefatter for eksempel alkalimetall-og jordalkalimetall-hydroksyder, -karbonater og -bikarbonater, så som natriumhydroksyd, kaliumhydroksyd, kalsiumhydroksyd, kaliumkarbonat, natriumkarbonat, magnesiumkarbonat og lignende, ammoniakk, primære, sekundære og tertiære aminer og lignende.
Særlig foretrukne baser er alkalimetallhydroksyder, -karbonater og -bikarbonater. Fysiologisk akseptable salter kan fremstilles ved konvensjonelle ionebytter-prosesser eller ved å behandle R^syren med en passende base. Eksempler på ytterligere salter er tidligere beskrevet.
Preparatene hvor de nye forbindelsene inngår er i fast eller flytende form. Disse formuleringene kan foreligge i form av et sett slik at de to komponentene blandes et passende tidsrom før bruk. Enten de er blandet på forhånd eller foreligger som et sett, fordrer formuleringene vanligvis et farmasøytisk akseptabelt bæremiddel eller hjelpestoff.
Oligomerene fremstillet i henhold til foreliggende oppfinnelse, er oppløselige i vann og i saltoppløsninger, spesielt ved fysiologisk pH og i saltvann. De lar seg således lett formulere til passende vandige farmasøytiske doseringsformer. Også etter at denne oligomer-formulering er administrert, forblir oligomeren oppløselig in vivo.
En foretrukket betydning for X i den tidligere beskrevne formel I er
Anti-HIV anioniske oligomerer kan benyttes for å hindre syncytium-danneIse i celler infisert med HIV-I-virus eller andre beslektede vira som har gpl20 overflate-protein. Anti-HVI-anioniske oligomerer kan benyttes for å behandle AIDS og ARC og andre sykdommer forårsaket av retrovirus-HIV-I eller andre beslektede vira som har gpl2G-overflate-protein. De anioniske oligomerene fremstillet i henhold til oppfinnelsen, kan benyttes som en ren forbindelse eller som blandinger, så som de med n-betydninger av en bestemt formel I, eller som blandinger med andre kjente midler for de her aktuelle antivirale anvendelser. For alle fremstillede oligomerer representerer imidlertid n den antalls-midlere repetisjonslengde av fordelingen.
Mengden av anti-HIV-anioniske oligomerer som trengs for å hindre syncytium-danneIse i HIV-infiserte celler, kan være en hvilken som helst effektiv mengde. Eksperimentelt er det fastslått at anti-HIV-anioniske oligomerer benyttet i en konsentrasjon på 10 mg/ml vandig formulering, resulterte i fullstendig hemming av syncytium-danneIse og reduserte dessuten forekomsten av p24-antigen, en indikator på viral replikasjon, til mindre enn 300 pg/ml. Mengden av anti-HIV-anioniske oligomerer som må gis for å behandle AIDS eller ARC eller andre sykdommer forårsaket av HIV-infeksjon, kan variere betydelig med den spesielle enhetsdose som benyttes, behandlingsperioden, pasientens alder og kjønn, natur og grad av forstyrrelsen som behandles, og andre faktorer som vil være velkjent innen dette medisinske fagfelt. Anti-HIV-anioniske oligomerer kan dessuten benyttes sammen med andre kjente midler egnet for behandling av retrovirale sykdommer, og midler som er kjent for å være egnet for behandling av symptomer på, og komplikasjoner forbundet med, sykdommer og tilstander forårsaket av retrovira.
Den anti-HIV-effektive mengde av anti-HIV-anioniske oligomerer vil i alminnelighet variere fra ca. 0,1 mg/kg til 50 0 mg/kg legemsvekt og kan administreres én eller flere ganger per dag. Anti-HIV-anioniske oligomerer kan gis peroralt eller parenteralt sammen med et farmasøytisk bæremiddel ved
bruk av konvensjonelle enhetsdoseformer.
For oral administrasjon kan anti-HIV-anioniske oligomerer formuleres som faste eller flytende preparater, så som kapsler, piller, tabletter, pastiller, smelter, pulvere, oppløsninger, suspensjoner eller emulsjoner. De faste enhetsdoseformene kan være en kapsel som kan være av ordinær hård- eller myk-gelatintype og som for eksempel inneholder overflateaktive midler, glattemidler og inerte fyllstoffer, så som laktose, sukrose, sorbitol, kalsiumfosfat og maisstivelse. De anioniske oligomerene kan dessuten tabletteres med konvensjonelle tablettbasismaterialer, så som laktose, sukrose og maisstivelse kombinert med bindemidler, så som akasiegummi, maisstivelse eller gelatin, sprengmidler som bidrar til å bryte opp og løse opp tabletten etter administrasjon, så som potetstivelse, alginsyre, maisstivelse og guargummi, slippmidler beregnet på å forbedre tablettgranuleringsstrømmen og forhindre adhesjon av tablettmaterialet til overflater på tablett-stanseverktøy, for eksempel talk, stearinsyre eller magnesium-, kalsium- eller sink-stearat, farvestoffer, og smaksforbedrende midler beregnet for å forbedre tablettenes estetiske kvaliteter og gjøre dem mer akseptable for pasienten. Egnede hjelpestoffer for bruk ved flytende, perorale doseringsformer innbefatter fortynningsmidler, så som vann og alkoholer, for eksempel etanol, benzylalkohol og polyetylenglykoler, enten med eller uten tilsetning av et farmasøytisk akseptabelt overflateaktivt middel, suspenderingsmiddel eller emulgeringsmiddel.
De anti-HIV-anioniske oligomerene fremstillet i henhold til oppfinnelsen, kan også administreres parenteralt, hvilket vil si subkutant, intravenøst, intramuskulært eller intra-peritonealt, som injiserbare doser av de anioniske oligomerer i et fysiologisk akspetabelt fortynningsmiddel sammen med et farmasøytisk bæremiddel som kan være en steril væske eller blanding av væsker, så som vann, saltvann, vandig dekstrose og lignende sukkeroppløsninger, i en alkohol så som etanol, isopropanol eller heksadecylalkohol, glykoler, så som propylenglykol eller polyetylenglykol, glycerolketaler så som 2,2-dimetyl-l, 3-dioksolan-4-metanol, etere så som poly(etylenglykol) 400, en olje, en fettsyre, en fettsyreester eller -glycerid eller et acetylert fettsyreglycerid med eller uten tilsetning av et farmasøytisk akseptabelt overflateaktivt middel, så som en såpe eller en detergent, suspenderingsmiddel så som pektin, "karbomerer", metylcellulose, hydroksypropylmetylcellulose eller karboksymetylcellulose, eller emulgeringsmidler og andre farmasøytiske tilsetningsstoffer.
Eksempler på oljer som kan benyttes i parenterale formuleringer av forbindelser fremstillet i henhold til oppfinnelsen, er oljer av petroleums-, animalsk, vegetabilsk eller syntetisk opprinnelse, for eksempel jordnøttolje, soyaolje, sesamolje, bomullsfrø-olje, maisolje, olivenolje, vaselin og mineralolje. Egnede fettsyrer innbefatter oljesyre, stearinsyre og isostearinsyre. Egnede fettsyreestere er for eksempel etyloleat og isopropylmyristat. Egnede såper innbefatter fettalkalimetaller, ammonium- og trietanolamin-salter, og passende detergenter innbefatter kationiske detergenter, for eksempel dimetyldialkylammoniumhalogenider, alkylpyridiniumhalogenider og alkylaminacetater; anioniske detergenter, for eksempel alkyl-, aryl- og olefinsulfonater, alkyl-, olefin- eter- og monoglyceridsulfater, og
-sulfosuccinater; ikke-ioniske detergenter, for eksempel fettamin-oksyder, fettsyrealkanolamider og polyoksy-etylenpolypropylen-kopolymerer; samt amfotære detergenter, for eksempel alkyl-S-aminopropionater og 2-alkylimidazolin-kvartære ammoniumsalter så vel som blandinger. De parenterale preparatene vil typisk inneholde fra ca. 0,5 til ca. 25 vektprosent anti-HIV-anionisk oligomer i oppløsning.Konserveringsmidler og buffere kan også med fordel benyttes. For å minske eller eliminere irritasjon på injeksjonsstedet kan slike preparater inneholde et ikke-ioniske overflateaktivt middel som har en HLB (hydrophile-lipophile balance) på ca.12til ca. 17. Mengden av overflateaktivt middel i en slik formulering varierer fra ca. 5 til ca. 15 vektprosent. Det overflateaktive midlet kan være en enkelt komponent som har
den ovenfor angitte HLB eller kan være en blanding av to eller flere komponenter som har den ønskede HLB. Eksempler på overflateaktive midler benyttet i parenterale formuleringer er polyetylensorbitanfettsyreestere, for eksempel sorbitanmonooleat.
Oligomerene fremstillet i henhold til oppfinnelsen, kan også benyttes profylaktisk, dvs. for å forhindre overføring av virus fra et infisert individ til en ikke infisert mottager. Virus spres proporsjonalt via utveksling av blod, men kan også overføres via utveksling av andre kroppsvæsker. Oligomerene fremstillet i henhold til oppfinnelsen, kan således formuleres med vanlige detergentprodukter for bruk ved rengjøring, spesielt i forskningslaboratorier og kliniske laboratorier, samt i sykehus hvor blodprodukter fra infiserte personer behandles. Oligomerene ble fremstillet ved å modifisere fremgangsmåten til Kershner (US-patent 4.895.660, som forøvrig er ytterligere beskrevet nedenfor) ved å erstatte en del av én av de difunksjonelle monomerene med et monofunksjonelt "end-capping"-middel og foreta omsetningen uten overflateaktivt middel. Oligomerene fremstilles i henhold til oppfinnelsen ved omsetning av et aromatisk diamin med en difunksjonell elektrofil i nærvær av en syreakseptor, med vann som oppløsningsmiddel eller i vann med opp til 1 mol med-oppløsningsmiddel som ikke er blandbart med vann, ved en temperatur fra 0 til 100°C og ved en pH mellom 7 og 9, særlig under anvendelse av et monofunksjonelt "end-capping"-middel.
Polyurea- forbindelser ( med formel I ovenfor)
Den foretrukne fremgangsmåte for fremstilling av polyurea-forbindelser med formel I ovenfor, er tidligere beskrevet (Kershner US-patent 4.824.916) og er ytterligere forklart nedenfor. De ulike reaktanter og betingelser er også beskrevet.
Diaminer: Et bredt utvalg alifatiske og aromatiske diaminer er inkludert. Hydrokarbylen-diradikalene som diaminene er sammensatt av, kan innbefatte metylen, etylen, butylen, isopropyliden, fenylen, bifenylen og andre diradikaler. Utvalget av mulige substituenter er tilsvarende bredt og innbefatter hydroksyl, alkenyl, lavere alkyldeler, karboksylat, sulfonat og halogener. Substituentene er ikke nødvendigvis anioniske i vann ved nøytral pH-verdi.
Difunksjonelle elektrofile forbindelser: Fosgen (karbonyldiklorid) , karbonyldibromid, Cl3COCOCl, Cl3COC02CCl3, disyrehalogenider av alifatiske og aromatiske to-basiske syrer, så som oksal-, malon-, rav-, glutar-, adipin-, sebacin-, ftal-, isoftal-, 2,6-naftalsyrer.
Syreakseptorer: flere baser har vært benyttet, så som
natriumkarbonat, natriumhydroksyd og tributylamin.
Diverse tilsetningsstoffer: forskjellige overflateaktive midler kan tilsettes. Egnede overflateaktive midler kan være ikke-ioniske, så som sorbitan-monolaurat, sorbitan-monostearat, etylenglykoldistearat, polyetylenoksy/polypropylenoksypolymer. Slike overflateaktive midler kan være vanskelige å fjerne fra produktet og bruk av overflateaktivt middel er derfor ikke å foretrekke.
Oppløsningsmidler: ved fremgangsmåter hvor ett oppløsningsmiddel benyttes anvendes polare aprotiske oppløsningsmidler, så som N,N-dimetylacetamid og N,N-dimetylformamid. En kombinasjon av vann og et annet oppløsningsmiddel, så som toluen, karbontetraklorid, benzen, aceton, etylendiklorid og lignende kan også benyttes. Forholdet mellom organiske og vandige oppløsningsmidler er typisk ca. 0,5 til ca. 2.
For tidligere beskrevne prosesser tilsettes disyre-halogenidet til en omrørt oppløsning eller suspensjon av de øvrige utgangsmaterialene. I enkelte tilfeller tilsettes basen under karbonyldihalogenid-tilsetningen. Temperaturen holdes mellom 0 og 50°C, fortrinnsvis 20 til 30°C. Et reaktantforhold (molart forhold mellom diamin og disyrehalogenid) fra ca. 0,9 til 1,2 kan benyttes, mens tilnærmet ekvimolare mengder foretrekkes.
Reaksjonsblandingen omrøres med tilstrekkelig hastighet til å oppnå blanding av reaktantene. Reaksjonshastigheten avhenger delvis av berøringsflaten mellom fasene og kraftig omrøring er derfor å foretrekke. En kommersiell mikser kan benyttes for dette formål.
Fremgangsmåten benyttet i henhold til oppfinnelsen, for fremstilling av polyurea-forbindelsene er en modifikasjon av prosessen beskrevet ovenfor.
Diaminer: diaminene fremstillet i henhold til foreliggende oppfinnelse, er primært aromatiske og har de formler som er beskrevet i tidligere avsnitt. Slike diaminer er substituert med minst én gruppe som har en ladning ved nøytral pH, fortrinnsvis sulfonat. Monovalente alifatiske substituenter kan tillates. Et lite utvalg alifatiske sammenbindende grupper som binder aromatiske radikaler sammen, så som trans-substituert etylen og acetylen, kan benyttes. Foretrukne diaminer er slike hvor karbon-nitrogen-bindingene er tvunget til å være parallelle, så som PDS, BPDS, StDS og 2, 5-diaminobenzensulfonsyre.
Difunksjonelle elektrofiler: for fremstillingen av polyurea-fosgen (karbonyldiklorid) og karbonyldibromid, og andre urea-forløpere, så som karbonyldiimidazol, heksakloraceton, Cl3COC02CCl3, CC13C0C1 og C130C0C1 kan benyttes. For fremstillingen av polyamider benyttes aromatiske disyrer, så som isoftal- og tereftalsyre (TPC), 2,6-naftalendisyre. Disse disyrene har nøytrale eller ladede substituenter, så som monovalente alkylradikaler (metyl, etyl, butyl) og/eller ladede grupper, så som sulfonater, fosfater og lignende. Et eksempel på en slik ladet difunksjonell elektrofil forbindelse er natrium 2,5-bis(klorkarbonyl)benzensulfonat (TPCS).
Syreakseptorer: et utvalg uorganiske baser, så som alkalimetall- eller divalente metallhydroksyder, -karbonater, -bikarbonater, -fosfater, kan benyttes. Syreakseptorer med en viss bufferkapasitet foretrekkes når basen i sin helhet tilsettes før tilsetningen av den difunksjonelle elektrofile forbindelse. Organiske baser, så som trialkylaminer, kan benyttes, men er ikke å foretrekke.
Monofunksjonelle "end capping"-midler: et utvalg av slike midler som begrenser molekylvekten, kan benyttes. Slike midler kan være alifatiske eller aromatiske forbindelser som reagerer med diaminene eller de difunksjonelle elektrofiler. Eksempler på egnede monofunksjonelle midler er aminer, så som anilin, metylanilin, metylamin, etylamin, butylamin, dietylamin, ammonium-N-metylanilin, fenol og kresol. Eksempler på monofunksjonelle reaktive amin-agens er benzoylklorid, metylbenzoylklorid, acetylklorid og fenylklorformiat. Disse"end capping"-midlene kan også inneholde ladede substituenter, for eksempel kalium 2-sulfofenol eller kalium 4-sulfoanilin. Diverse tilsetningsstoffer: tilsetningen av overflateaktive midler er ikke nødvendig eller foretrukket, og kan komplisere isoleringsprosesen.
Oppløsningsmidler: et enkelt oppløsningsmiddel, vann, er å foretrekke når den difunksjonelle elektrofile forbindelse er flytende ved reaksjonstemperaturen. Et eksempel på en slik difunksjonell elektrofil forbindelse er fosgen. Når faste, reaktanter som ikke er oppløselige i vann benyttes, kan små mengder av et med-oppløsningsmiddel som ikke er blandbart med vann, være ønskelig. For eksempel tilsettes, når tereftaloyl-klorid benyttes, en minimal mengde metylenklorid for å forbedre kontakten mellom reaktantene. Eksempler på slike med-oppløsningsmidler som ikke er blandbare med vann, er kloroform, karbontetraklorid, toluen og metylenklorid. Typiske forhold mellom organiske og vandige oppløsningsmidler er 0 til 1, fortrinnsvis 0 til 0,1.
Prosessen foretas ved temperaturer som får reaksjonen til å gå, typisk fra ca. 0 til 100°C. Foretrukne temperaturer er 0 til 25°C. Når det benyttes lavtkokende utgangsmaterialer, for eksempel fosgen (kp. 6°C) er det fordelaktig å foreta omsetningen ved temperaturer som ligger ved eller under kokepuktet. Trykket er ikke vesentlig og normaltrykk kan benyttes. Reaksjonens pH må overholdes omhyggelig for å optimalisere prosessen. Ved lav pH (<6) er reaksjonen meget langsom, mens den difunksjonelle elektrofile forbindelse ved høy pH (>10) er ustabil overfor angrep av hydroksyd eller annen base. Nedbrytning av polyurea-forbindelsen kan også skje ved høy pH. Fortrinnsvis holdes pH mellom 7 og 9.
Når det ikke benyttes "end capping"-middel, kan molekylvekt-kontroll oppnås ved forsiktig regulering av reaktantenes støkiometriske forhold. Diaminet eller den difunksjonelle elektrofile forbindelse kan benyttes i overskudd, for eksempel fra 1 til 100% molart overskudd. Dette støkiometriske forhold må erstatte eventuell difunksjonell elektrofil forbindelse som ødelegges ved hydrolyse før reaksjonen med diaminet. Når fosgen for eksempel benyttes ved høy pH, fordres et stort overskudd for å kompensere for den hurtige ødeleggende reaksjon med hydroksyd. Siden graden av denne bireaksjon er vanskelig å kontrollere, anvendes fortrinnsvis et monofunksjonelt "end capping"-middel for å kontrollere molekylvekten. Selv om de nevnte teknikker kan benyttes for å kontrollere den antalls-midlere molekylvekt, er produktene blandinger av polymerer med flere molekylvekter som erkarakterisert veden fordeling.
Tilsetningsrekkefølgen av reaktantene er ikke av avgjørende betydning. Den foretrukne rekkefølge er imidlertid å tilsette den difunksjonelle elektrofile forbindelse først. Når det benyttes syreakseptorer som ikke er buffere, så som hydroksyd, er det mest fordelaktig å tilsette en porsjon i begynnelsen for å oppnå den ønskede pH og deretter tilsette det gjenværende samtidig med den difunksjonelle elektrofile forbindelse.
Endelig er det ønskelig å foreta disse polymeriseringene ved høye konsentrasjoner. Dette reduserer mengden av oppløsningsmiddel som må fjernes for å isolere produktet. I enkelte tilfeller utfelles produktet dessuten fra reaksjonsoppløsningen mot slutten av omsetningen, og kan da isoleres ved enkel avdekantering av oppløsningsmidlet. Det meste av det uorganiske salt som er et resultat av reaksjonen med syreakseptoren, fjernes i denne prosess. Konsentrasjonen er ikke av avgjørende betydning og kan være fra 0,5 til 50 vektprosent, uttrykt som vekten av diamin i forhold til vekten av oppløsningsmiddel. Et foretrukket område er 5 til 20 vektprosent.
Produktet kan isoleres ved utfelling av reaksjonsopp-løsningen i et oppløsningsmiddel som er blandbart med vann, men utgjør et dårlig oppløsningsmiddel for produktet. Eksempler på slike oppløsningsmidler er aceton, metanol, etanol, isopropanol.

Eksempel A
Fremstilling av HBPDS med formelen
Til en 2 L kolbe forsynt med dråpetrakt og magnetrører ble det tilsatt 49,99 g (0,145 mol) 4,4'-diamino(1,1'-bifenyl)-2,2'-disulfonsyre og 600 mL vann.Diaminet ble solubilisert ved tilsetning av 30 mL (0,15 mol) 5M NaOH. Den resulterende oppløsning ble tilsatt 20,56 g (0,298 mol) natriumnitritt. Reaksjonsblandingen ble deretter avkjølt til 0°C og i løpet av 30 minutter tilsatt 60 mL konsentrert H2S04oppløst i 360 mL vann. Det ble dannet et gult faststoff.Blandingen ble deretter tilsatt 300 mL vann og holdt ved 0°C i 1 time. Reaksjonsblandingen ble deretter filtrert. Det gule faststoffet ble etter oppløsning i 800 mL vann, anbragt i en 1 L kolbe og oppvarmet til det var igjen ca. 50 mL vann. Nitrogen-gass oppsto under oppvarming. Den konsentrerte oppløsningen ble tilsatt 20,14 g (0,146 mol) K2C03, hvorpå oppløsningen ble kokt. Absolutt etanol (1,5 L) ble deretter tilsatt og det utfeltes et brunt faststoff. Faststoffet ble frafiltrert og tørket over natten i en 50°C ovn. Produktet,HBPDS, ble oppnådd i et utbytte på 32,33 g (53%) og ble viderekarakterisert ved<i>H NMR 5 6,70 (dd, 1H), 7,05 (d, 1H), 7,14 (d, 1H).
Eksempel B
Fremstilling av TPCS med formelen
En 500 mL kolbe forsynt med mekanisk rører, termometer og tilbakeløpskjøler ble tilsatt 40,49 g (0,143 mol) av mononatrium-saltet av 2-sulfotereftalsyre, 160 mL klorbenzen, 2,4 mL
(0,031 mol) dimetylformamid og 23 mL (0,315 mol) tionylklorid. Oppløsningen ble oppvarmet til 105°C og omrørt i 2 timer under nitrogen. I løpet av denne tiden ble det observert gass-utvikling. Denne oppløsningen ble avkjølt til romtemperatur, hvorpå et faststoff utfeltes.Faststoffet ble frafiltrert og tørket over natten i en vakuumovn ved romtemperatur. Produktet ble oppnådd som gult faststoff i et utbytte på 20,56 g (47%).
For å bekrefte strukturen av produktet ble noe av produktet omdannet til metylesteren.
Til en 25 mL kolbe forsynt med en magnetrører og et nitrogen-innledningsrør ble det tilsatt 0,9509 g (3,12 mmol) av produktet ovenfor, 0,6874 g (6,47 mmol) Na2C03og 10 mL metanol. Fremstilling av BPDS/P/T med formelen
Oligomer A ( n=6)
Til en 1 L kolbe forsynt med en kanyleåpning, termometer-brønn, pH-elektrode, tørriskjøler, fosgen-gass-innløp og et mekanisk røreverk, ble det tilsatt 10,00 g (28,19 mmol) BPDS, 1,35 g (9,40 mmol) toluidin-hydroklorid og 400 ml vann. Reaksjonsblandingen ble omrørt og avkjølt til 12°C. Den omrørte suspensjon ble deretter omsatt med 13 mL 5M NaOH inntil alt faststoffet var oppløst. Reaksjonsblandingen ble deretter tilsatt 10,1 g (102 mmol) fosgen i løpet av 27 minutter. Under fosgentilsetningen ble 5M NaOH tilsatt etter behov ved hjelp av en sprøyte for å holde pH mellom 7 og 8 (ekstreme pH-verdier fra 6 til 9 forekom). Totalt 31 mL NaOH ble tilsatt. Omrøring av reaksjosnblandingen ble fortsatt i 30 minutter til, hvorpå pH ble justert til 9,5 og reaksjonsblandingen omrørt i ytterligere 30 minutter. Den ble overført til en 2 L kolbe og råproduktet utfelt ved tilsetning av 1000 mL aceton. Råproduktet ble filtrert og lufttørket for å gi 18,6 g hvitaktig pulver med Mn= 2500. Egenviskositeten var 0,39 dL/g i H20, 0,15 dL/g i HBSS. Produktet ble viderekarakterisert ved

Ca. halvparten av suspensjonen oppnådd etter tilsetning av aceton ble filtrert på grunn av tilstopningsproblemer av fritten. Produktet ble oppnådd som et gult faststoff i et utbytte på 3,5 g og Mn= 3800. Egenviskositeten var 0,18 dL/g i H20.

Fremstilling av PDS/P/T med formelen
Oligomer A ( n=9)
Når fremgangsmåten i Eksempel IA ble gjentatt ved bruk av følgende reagensmengder:
ble produktet oppnådd som et brunt pulver i et utbytte på 2,95 g og Mn= 2900. Egenviskositeten var 0,12 dL/g i H20 og 0,07 dL/g i HBSS.
Oligomer B ( n=15)
Når fremgangsmåten i Eksempel IA ble gjentatt ved bruk av følgende reagensmengder:
ble produktet oppnådd som et brunt pulver i et utbytte på 3,83 g og Mn= 4650. Egenviskositeten var 0,12 dL/g i H20 og 0,14 dL/g i HBSS.

Eksempel 1
EN ANTI-HIV-OLIGOMERS EVNE TIL Å FORHINDRE SYNCYTIE-DANNELSE OG EKSPRESJON AV P24 VIRALT KJERNE-ANTIGEN VED BRUK AV JM-CELLER OG GB8 VIRUS-STAMME.
For å vise at en oligomer fremstillet i henhold til oppfinnelsen blokkerer HIV-infeksjon, ble CD4<+>T-celler (JM) eksponert for et klinisk isolat av HIV-I, GB8. Viruset ble først inkubert med en oligomer i 15 minutter og deretter ble cellene tilsatt. Etter 2 timers adsorpsjon ble virus-inokulatet fjernet og cellene vasket tre ganger for å fjerne spor av det benyttede virus. Anti-viral aktivitet ble bestemt etter tre dagers inkubering ved å avsette det gjennomsnittlige antall av dannede syncytier i fire-parallellers kulturer mot log10-konsentrasjonen av anionisk polymer eller av andre testforbindelser. Styrken av en oligomer ble også målt ved å bestemme viralt kjerne-antigen (P24 test-Abbott) i den overstående væske. Heparin, dekstran-sulfat, rs CD4, ATZ og/eller ddC-data er, når de er inkludert i noen av de etterfølgende tabeller, angitt som positive kontroller.
Eksempel II
Virusinfeksjon av JM-celler ble foretatt i nærvær av forskjellige konsentrasjoner av testforbindelser. JM-celler
(1 x IO<5>) og 50-100 syncytie-dannende enheter av virus (GB8) ble tilsatt til dobbelte sett brønner i en vevskultur-plate inneholdende 1 mL volumer vekstmedium med eller uten medikament. Platen ble inkubert i 2 dager ved 3 7°C og deretter vurdert med henblikk på forekomst av syncytier. Samtidig ble cellene vasket og vekstmediet skiftet ut. Etter ytterligere 2 dagers inkubering ble de celle-frie overstående væsker høstet og undersøkt på nivåer av P24 viralt kjerne-antigen ved å benytte Coulter™ HIV-antigen-testsett. Resultatene er angitt i
tabellene II-III. I tabellene betyr N.D. = ikke påvist og N.A. = ikke målt.
Eksempel III
FORSKJELLIGE ANTI-HIV-OLIGOMERERS EVNE TIL Å HINDRE VIRUS-INDUSERT CELLEDØD VED BRUK AV MT4-CELLER OG STAMME RF.
Forskjellige oligomerer ble oppløst i RPMI og deretter undersøkt på anti-HIV-aktivitet ved å foreta
dobbeltfortynninger av oppløsningene over en 96-brønns
mikrotiter-plate. Hver brønn ble deretter tilsatt 5 xlO<4>celler og 100 TCID50 virus og platene inkubert ved 37°C i 7 dager. MTT ble tilsatt til hver brønn og platene inkubert i ytterligere 2 timer. De blå formazan-krystallene ble oppløst ved bruk av sur isopropanol og absorbansen målt ved 54 0 nm. Resultatene er angitt i Tabell IV.
Eksempel IV
EVNE TIL FORBEHANDLING AV CELLER MED FORSKJELLIGE OLIGOMERER OG BLOKKERE HIV-I-INFEKSJON VED Å BENYTTE JM-CELLER OG GB8-STAMME AV HIV-I
JM-celler ble forbehandlet over natten ved 37°C med
20 fxg/ mh av forskjellige forbindelser eller de forble ubehandlet. Cellene ble vasket tre ganger i RPMI-medium og
deretter infisert med HIV-I (GB8) i 2 timer ved romtemperatur. Cellene ble igjen vasket tre ganger i RPMI-medium og resuspendert i friskt medium før de ble fordelt på dobbelte sett av brønner og inkubert ved 37°C. Etter 2 dager ble antallet syncytier bestemt og den celle-frie overstående væske høstec og undersøkt med henblikk på P24 viralt kjerne-antigen ved bruk av Coulter™ HIV-antigen-testsettet. Resultatene er angitt i Tabell IV.
Eksempel V
EN ANTI-HIV-I-OLIGOMERS EVNE TIL Å FORHINDRE SYNCYTIE-DANNELSE OG P24 VIRALT KJERNE-ANTIGEN EKSPRESJON AV ULIKE VIRALE
STAMMER (GB8 OG RF) OG CELLER (JM OG C8166)
Celler ble infisert med stamme RF eller GB8 i 24 timer ved 37°C med en veksthastighetskonstant (multiplisity of infection) på 0,001. Cellene ble vasket tre ganger for å fjerne gjenværende virus og deretter sådd ut igjen på friskt vekstmedium. Cellene ble deretter behandlet i 24 og 48 timer etter infeksjon (p.i.) med de angitte konsentrasjoner testforbindelser. Syncytier og P24 antigen-nivåer ble bestemt på de angitte dager p.i. etter fremgangsmåter beskrevet tidligere. Resultatene er vist i Tabell V-VII.
Resultatene i Tabell V ovenfor indikerer at omstendigheter assosiert med viralt induserte cytopatologiske forandringer så som syncytie-dannelse, kan hemmes selv når forbindelser administreres til tidligere infiserte celler. Disse resultatene indikerer også at de anioniske oligomerene virker gjennom en mekanisme i tillegg til å blokkere viral tilknytning til overflateproteinet av CD4-celler.
Resultatene i Tabell VI ovenfor indikerer at oligomerene fremstillet i henhold til oppfinnelsen, er effektive overfor forskjellige virale stammer og forskjellige celle-typer selv når de tilsettes 24 timer etter virusinfeksjon.
Resultatene av disse undersøkelser viser at oligomerene fra Eksempel IA befridde kulutrer for syncytier, stabiliserte infeksjonen og reduserte virus-antigen-nivåene i celler som hadde etablerte,infeksj oner.
Claims (4)
1. Analogfremgangsmåte for fremstilling av en terapeutisk aktiv oligomer som er en polyurea-forbindelse med formel:
hvor: R er en 4-metylfenylgruppe; n er 3 til 15; X utgjør og R<2>er et hydrogenatom eller et farmasøytisk akseptabelt kation,
karakterisert vedomsetning av et aromatisk diamin med en difunksjonell elektrofil i nærvær av en syreakseptor, med vann som oppløsningsmiddel eller i vann med opp til 1 mol med-oppløsningsmiddel som ikke er blandbart med vann, ved en temperatur fra 0 til 10 0°C og ved en pH mellom 7 og 9,
særlig under anvendelse av et monofunksjonelt "end-capping" middel.
2. Fremgangsmåte ifølge krav 1 for fremstilling av en oligomer som er StDS/P/T og betegnet poly[imino(3-sulfo-l,4-fenylen)-1,2-etendiyl-(2-sulfo-l,4-fenylen)iminokarbonyl], alfa-{[(4-metyl-fenyl)aminokarbonyl}-omega-[(4-metylfenyl)amino- og er representert ved formel I i krav 1 idet R er 4-metylfenyl, R2 er hydrogen, X er
og n er definert som for formel I i krav 1, fortrinnsvis 6 eller 9,
karakterisert vedbruk av tilsvarende utgangsmaterialer.
3. Fremgangsmåte ifølge krav 1 for fremstilling av en oligomer som er PDS/P/T betegnet poly[imino(2, 5-disulfo-l, 4-fenylen)imino-karbonyl], alfa-{[(4-metylfenyl)amino]karbonyl}-omega-[(4-metyl-fenyl)amino]- og er representert ved formel I i krav 1 idet R er 4-metylfenyl, R<2>er hydrogen, X er
og n er definert som for formel I i krav 1, fortrinnsvis 9 eller 15,karakterisert vedbruk av tilsvarende utgangsmaterialer.
4. Fremgangsmåte ifølge krav 1 for fremstilling av en oligomer som er PDS/P/T betegnet poly{imino[2,2 '-disulfo(1,1'-bifenyl)-4,4'-diyl]iminokarbonyl}, alfa-{ [ (4-metylfenyl)amino]karbonyl}-omega-[(4-metylfenyl)amino]- og er representert ved formel I i krav 1 idet R er 4-metylfenyl, R<2>er hydrogen, X er
