Description
Oppfinnelsen vedrører anvendelse av rekombinant-DNA-teknikker for å lage hybrid-proteinmolekyler. Slike molekyler kan anvendes ved behandling av medisinske forstyrrelser.
Mer bestemt vedrører oppfinnelsen et sammensmeltet gen som omfatter et fragment av det gen som koder for difteri-toksin sammensmeltet til et fragment av et gen som koder for en cellespesifikk polypeptidligand. Den henvises forøvrig til krav 1.
Oppfinnelsen vedrører videre en kloningsvektor, som beskrevet i krav 6, samt anvendelse av kloningsvektoren, som beskrevet i krav 7.
Litteraturen inneholder mange eksempler på sammensmeltede gener som koder for hybridproteiner. Eksempelvis beskriver Villa-Komaroff et al. (1978) P.N.A.S. U.S.A. 75, 3727-3731 et sammensmeltet gen bygd opp av et eukaryotisk strukturelt gen sammensmeltet til et ikke-cytoplasmisk bakterielt gen. Det sammensmeltede gen koder for et hybridprotein som transporteres ut av cytoplasmaet.
Hybridproteiner er også laget ved metoder som f.eks. anvender kobling av to forskjellige proteinmolekyler, som ikke innebærer rekombinant-DNA-teknikker. Eksempelvis er det foreslått å danne, ved kobling, terapeutiske hybridproteiner som består av et toksin koblet til en ligand med evne til å binde seg spesifikt til en utvalgt klasse av celler. Ett forsøk på å lage et slikt hybridprotein, rapportert av Chang et al. (1977) J. Biol. Chem. 252, 1515-1522, resulterte i et hybrid som besto av difteri-toksin A-kjede koblet til human-placentalt laktogen-hormon ved tverrbinding via en disulfidbinding. Hybridproteinet inhiberte ikke proteinsyntesen i disse celler, selv om det bandt seg til celler som inneholder laktogen-reseptorer.
Et hybridprotein bestående av ricin A-toksin koblet til jø-kjeden i human-chorionisk gonadotropin-hormon ved likeledes å tverrbinde via en disulfidbinding er også rapportert. Selv om den sies å ha spesifisitet, er dens bindingsevne ikke rapportert, og ekstremt høye konsentrasjoner var nødvendige for signifikant å inhibere proteinsyntese i Leydig-tumorceller hos rotte, hvilket gjør det vanskelig å skjelne mellom "ikke-spesifikk" adgang forårsaket av endocytose, og "spesifikk" adgang forårsaket av transport av den toksiske del av hybridet over den cytoplasmiske membran i målcellene, se Oeltman et al. (1979) J. Biol. Chem., 254. 1028-1032. Den samme mangel ble funnet hos et hybrid om besto av difteri A koblet til insulin under anvendelse av cystamin som tverrbindingsmiddel, se Miskimins et al. (1979) Biochem. Biophys. Res. Commun., 91, 143-151. Et hybrid som besto av ricin A koblet til epidermal vekstfaktor (EGF) ved hjelp av en heterobi-funksjonell tverrbinding er også laget, men bindekarakteristikkene tilveiebragt av EGF er ikke begrenset til spesifikke celler, men omfatter et stort utvalg av celletyper, se Cawley et al., (1980) Cell, 22, 563-570.
Det sammensmeltede gen i henhold til oppfinnelsen kan anvendes til å lage et overlegent difteri-toksin/hormon-hybridprotein hvor proteinet er syntetisert som en enkelt enhet, dvs.
at fragmenter knyttes sammen, ikke ved tverrbinding, men ved peptidbindinger.
Oppfinnelsen vil bli best forstått ved betraktning av tegningene, hvor: Fig. 1 er en skjematisk presentasjon av det nevnte difteri-
toksinmolekyl; Fig. 2 er en skjematisk presentasjon av et hybridprotein
molekyl; Fig. 3 er et restriksjonskart som viser lokaliseringen og orienteringen av difteri- tox-operonet på 3,9 kb BamH-I-restriksjonsfragmentet til corynefag /?<tox> (inklusive et punkt, NRU I, som ikke er funnet på tox-allel av villtype, men bare på mutant-tox<228->allelen, hvilket vil bli forklart mer detaljert nedenunder), og Fig. 4 er en skjematisk presentasjon av et sammensmeltet gen i henhold til oppfinnelsen, som koder for et hybridprotein (genfragmentene er merket på grunnlag av de utkodede proteinfragmenter).
Under henvisning til fig. 1 består difteri-toksinmolekylet av flere funksjonelle "domener" som kan karakteriseres, idet man starter ved aminoterminalenden til molekylet, som hydrofob leder-signalsekvens s, enzymatisk aktivt fragment A, den fjorten amino-syre-eksponerte proteasesensitive disulfid-loop (DSL) £lf som inneholder et spaltningsdomene, og fragment B, som inkluderer hydrofob-domene h, DSL £2'°9 karboksyterminalende a "make-up" a.
Den prosess ved hvilken difteri-toksin intoksikerer sensitive eukaryotiske celler innebærer minst følgende trinn: (i) difteri-toksin binder seg til spesifikke reseptorer på overflaten av en sensitiv celle: (ii) mens det er bundet til sin reseptor, internaliseres toksinmolekylet i en endocytisk vesikkel; (iii) enten før internalisering, eller i den endocytiske vesikkel, spaltes toksinmolekylet (eller det forarbeides) ved et punkt i området på 47 000 dalton fra den N-terminale ende; (iv) etter hvert som pH-verdien til den endocytiske vesikkel avtar til under 6, innsettes den strukturelle inter-mediære form av toksin, mens den fremdeles er bundet til sin reseptor, i membranen; (v) så snart den er innleiret i membranen, danner det hydrofobe domene h en pore; (vi) en proteolytisk spaltning i mellom fragmentene A og B inntreffer; (vii) deretter frigis fragment A, eller et polypeptid som inneholder fragment A, inn i cytosolen; (viii) den katalytiske aktivitet hos fragment A, dvs. niktoin-amid-adenin-dinukleotid - adenosin-difosfat-ribosylering av forlengelsesfaktor 2, forårsaker død hos den intoksi-kerte celle. Det er innlysende at et enkelt molekyl i fragment A som er innført i cytosolen, er tilstrekkelig til å drepe cellen.
Hybridproteinene inkluderer, i sekvensiell orden, idet man begynner ved amino-terminalenden til hybridproteinet, følgende peptidfragmenter, knyttet sammen ved peptidbindinger: a) det enzymatiske aktive fragment A hos difteri-toksin (uten lederen fragment s, som er kuttet under sekresjon av
proteinet),
b) et fragment inklusive spaltningsdomenet som er nærstående nevnte fragment A i difteri-toksin, c) et fragment som omfatter minst den del av fragment B i difteri-toksin som kodes av den del av fragment B som koder genfragment til tox-operonet mellom t-^ og posisjonen ca. 90 basepar oppstrøms for posisjonen på tox-operonet til NRU I-punktet til tox<228->alleien, og d) et fragment som omfatter en del av en cellespesifikk polypeptidligand, idet delen inkluderer minst en del av bindingsdomenet til polypeptidliganden, idet den del av bindingsdomenet er effektiv med hensyn til å få hybridproteinet til å binde seg selektivt til en forutbestemt klasse av celler som skal angripes av enzymatisk aktivt fragment A fra difeteri-toksin. Den nødvendige del av toksinmolekylet som er inkludert, er avtegnet som delen av molekylet mellom linjene y og x i fig. 1.
Fortrinnsvis inkluderer også hybridproteinet protease-sensitivt DSL £2 fra toksinmolekylet, dvs. at delen av molekylet mellom linjene x og z også er inkludert. Linje z er fortrinnsvis ved punktet 47 aminosyrer fra karboksylterminalnden til B<1>, dvs. ved enden av £2, ikke nærmere, for å sikre at det generaliserte eukaryotiske bindingspunkt i fragment B er utelukket, slik at avbindingen vil bli styrt av avbindingsdomenet til den celle-spesifikke ligand. Det er vist at litt mer enn halvparten av fragment B må være tilveiebragt for molekylet slik at det kan virke som et effektivt toksin.
Under henvisning til fig. 2 så viser denne en skjematisk presentasjon av et hybridproteinmolekyl som her beskrevet, hvor y-z-delen av difteri-toksinmolekylet via en peptidbinding er tilknyttet fragment r i en cellespesifikk polypeptidligand; dvs. et polypeptid som selektivt binder seg til en forutbestemt klasse av celler som skal angripes av enzymatisk aktivt fragment A i difteri-toksinmolekylet. Fragment r kan bestå av hele liganden, eller en del av liganden som inkluderer hele avbindingsdomenet i liganden, eller en effektiv del av avbindingsdomenet.
Når den ligand som anvendes er stor, er det ønskelig at så lite av den ikke-avbindende del som mulig av liganden blir inkludert, slik at bindingsdomenet til molekylet er beliggende nær det hydrofobe domene h i fragment B. Det er også ønskelig å inkludere alt eller det meste av avbindingsdomenet til ligandmolekylet. Når det gjelder a-melanocytt-stimulerende hormon (MSH), som er et lite peptid med 13 aminosyrer, eller /3MSH, som inneholder 17 aminosyrer, kan den del av molekylet som består av 9 aminosyrer ved karboksyterminalenden til molekylet, anvendes, eller hele molekylet kan anvendes.
De områder innen celle-spesifikke ligander hvor avbindingsdomenet er beliggende, er nå kjent for et antall slike ligander. Videre kan nylige fremskritt i fastfase-polypeptid-syntese gjøre det mulig for fagmannen på dette område å bestemme avbindingsdomenet til praktisk talt enhver slik ligand, ved å syntetisere forskjellige fragmenter i liganden og teste dem med hensyn på evne til å binde seg til klassen av celler som skal angripes.
Hybridproteinmolekylene som beskrives her, er praktisk talt ikke-toksiske for alle pattedyrceller med unntagelse av cellene fra den spesifikke klasse som de ligandbindende domene binder seg til. Således er hybridproteinene meget mer spesifikke enn mange andre terapeutiske midler, f.eks. generelle cytotoksiske anti-cancerdroger.
Hybridproteinene i hvilke fragmenter er knyttet via peptidbindinger, er også tverrbundne hybrider overlegne fordi de her beskrevne proteiner kan tilveiebringes i en homogen prøve, i hvilken alle de identiske molekyler er effektive og selektive for en spesiell klasse av celler.
Den spesifikke klasse av celler som bindes og angripes av disse hybridproteiner bestemmes av den spesifikke polypeptidligand som tilfører avbindingsdomenet til hybridmolekylet. Enhver cellespesifikk polypeptidligand kan anvendes som har et avbindings-domene som er spesifikt for en spesiell klasse av celler som skal angripes. Polypeptidhormoner er nyttige slike ligander. Hybrider laget under anvendelse av en del av avbindingsdomenet av a- eller Ø-MSH (MSH = melanocytt-stimulerende hormon), f.eks., binder seg selektivt til melanocytter, idet de gjør hybridene nyttige ved behandling av melanomer. Andre ligander tilveiebringer forskjellige spesifisiteter, f.eks. gjenkjenner avbindingsdomenet til substans P reseptorer på overflatene av neuroner som er involvert i transmisjon av smerte, slik at hybrider laget under anvendelse av substans P kan anvendes for å ødelegge slike neuroner for å mildne kronisk smerte. Disse hybrider kan også anvendes på kart-arealer i nervesystemet som inneholder substans P-reseptorer. Andre spesifikt-bindende ligander som kan anvendes inkluderer somotostatin, interleukin I, interleukin II og interleukin III. Interleukin II er av spesiell betydning på grunn av sin rolle i allergiske reaksjoner og autoimmune sykdommer, f.eks. lupus, som involverer aktiverte T-celler. I tillegg, siden alle inter-leukinene er spesifikke for T-celler, kunne hybrider som er laget med dem bli anvendt for å behandle cancer som involverer immun-systemet, og inhibere avstøtning av transplanterte organer.
Andre anvendelige polypeptidligander som har cellespesifikke avbindingsdomener er follikkelstimulerende hormon (spesifikt for ovarieceller); luteiniserende hormon (spesifikt for ovarieceller); thyroid-stimulerende hormon (spesifikt for thyroid-celler); vasopressin (spesifikt for uterus-celler så vel som blære- og innvoll-celler); prolaktin (spesifikt for brystceller); og veksthormon (spesifikt for visse benceller).
Hybridproteinene som her er beskrevet fremtilles fortrinnsvis under anvendelse av rekombinant-DNA-teknikker som involverer å danne det ønskede sammensmeltede gen som koder for hybridproteinet, og deretter utkoder det smeltede gen, under anvendelse av konvensjonelle prosesser. Under henvisning til fig. 3 tillater lokaliseringen og orienteringen av diferi- tox-operonet på 3.9 kb BamH-I restriksj onsfragmentet til corynefag /3<tox+> tox-operonet å bli splittet ved en ønsket lokalisering, og den ønskede del av det operon som skal sammensmeltes med den ønskede del av genet for en utvalgt polypeptidligand. En mer detaljert beskrivelse av tox-operonet, og en beskrivelse av kloning av fragment A, inneholdes i Leong et al. (1983) Science 220, 515, herved tatt med som referanse. Fragment A, klonet som beskrevet deri for å lage plasmid pDT201, ble deponert i American Type Culture Collection, Rockville, MD i mai, 1983, og har fått ATCC Accession nr. 39359.
Under henvisning til fig. 3 og 4 er den del av difteri- tox-operonet (fig. 3) som anvendes for å lage det sammensmeltede gen (fig. 4) som koder de nevnte hybridproteiner, fortrinnsvis den del som er antydet av de stiplede linjer som avgrenser fragment D; dvs. delen av tox-genet fra første Sau 3AI-punkt til SPH I-punktet. Fragment D inkluderer således det 831-basepar (bp) lange gen-fragment som koder fragment A (inklusive 177 bp-sekvensen foran det strukturelle gen som inkluderer promotoren og delen som koder signalfrekevensen, S i fig. 1); den del av genfragmentet som koder det hydrofobe domene i fragment B; og genfragmentet som koder DSL £2.
Som vist i fig. 3, inkluderer de første 177 bp til det gen-fragment som koder fragment A, foran tox-promotoren, noe DNA som ikke er del av tox-operonet. Denne DNA er irrelevant og er ikke transkribert; den er inkludert bare fordi Sau 3AI-punktet ved begynnelsen av fragment A som koder genfragment er det mest bekvemme restriksjonspunkt nær tox-promotoren (som er avsatt som linjen like til venstre for Hind III).
Det skulle være mulig å oppnå fragment D ved ganske enkelt å eksitere det fra tox-operonet via spaltning ved Sau 3AI og SPH I. Alternativt kan genfragmenter sammensmeltes som følger. Først oppnås det genfragment som koder fragment A. Deretter oppnås det genfragment som koder det meste av fragment B, fra Sau 3AI til Sau 3AI (B<1> i fig. 3). Det genfragment som koder fragment B' er blitt klonet i plasmid pUC8, til å lage plasmid pDT301, og ble deponert i American Type Culture Collection, Rockville, Md i mai 1983, og har fått ATCC Accession nr. 39360. Fragment B' kuttes ned, under anvendelse av enzym Bal31, ca. 2 00 bp, til posisjonen NRU I-punktet på tox<228> (villtypen tox-allel har ikke et NRU I-punkt), slik at man får B<11> (fig. 3). Fragmentene A og B" er sammensmeltet, et fragment som koder £2 er sammensmeltet til B<*1>, og et fragment som koder den ønskede del av polypeptidliganden er sammensmeltet til £2. ;I ovennevnte skjema kuttes fortrinnsvis det fragment B' som kutter genfragment ned til den presise lokalisering av NRU I, men kan også kuttes ned til enhver lokalisering mellom lokaliseringen 90 bp foran NRU I, og slutten av de £2-kodende område (dvs. i 208 bp-området mellom lokaliseringen 90 bp foran NRU I, og begynnelsen av det €2-kodende området). Eller det B<1->kodende fragment kunne kuttes ned til karboksyterminal-enden til det £2-kodende område (129 bp nedstrøms for NRU I), i hvilket tilfelle sammensmeltning av et syntetisk £2-kodende område er unødvendig. (Det skulle være innlysende at, når det B'-kodende område er kuttet ned og et syntetisk £2-kodende fragment er sammensmeltet med det, så vil det generelt være et lite antall basepar som normalt finnes mellom NRU I og SPH I som ikke vil være til stede i det sammensmeltede gen, slik at strengt tatt ikke alt av D i fig. 3, eller alt av y-z i fig. 1 og 2, nødvendigvis vil være inkludert). ;En alternativ metode, mindre foretrukket enn ovenstående skjema, er å sammensmelte det ligand-kodende genfragment direkte til B* 1 uten å anvende det £2-kodende fragment.
Det sammensmeltede gen, enten inklusive eller under ute-latelse av det £2-kodende fragment, kan alternativt lages ved anvendelse av det B<11->kodende genfragment fra den mutante tox<228->allel, snarere enn villtype-allelen. Den tox<228->allel, som inneholder NRU I-punktet, forarbeides lett slik at det gir det B<1>'-kodende fragment. Denne tox<228->allel er beskrevet i Uchida et al.
(1973) Jour. Biolog. Chem. 248, 3838, som herved tas med som referanse.
Mer detaljert kan sammensmeltede gener som koder hybridproteiner som her beskrives, lages på følgende måte.
Vektorer
De foretrukne vektorer er plasmidene pUC7, pUC8, pUC9 og pBR322. pUC-plasmidene, beskrevet av Viera et al. (1982) Gene 19, 259 (herved tatt med som referanse) er spesielt godt egnet for dobbelt-fordøyingskloning (fordøying med to forskjellige restrik-sjonsenzymer), en prosess som tillater de sammensmeltede gener å lages på utvetydig måte.
Sammensmeltet gen
Nedenunder er det vist et flytskjema for konstruksjon av difteri-toksin-MSH-sammensmeltede gener som inneholder den protease-sensitive loop £2 mellom tox-sekvensene og ligand-
(i dette tilfelle, MSH) sekvensene.

Under henvisning til ovenstående flytskjema, i trinn (i), oppnås først det fragment A-kodende genfragment og renses, som beskrevet i Science, JEd. I trinn (ii) oppnås det fragment B'<1->kodende fragment ved nedkutting av et større fragment B<1->kodende område under anvendelse av enzym Bal31.
Som vist i fig. 3, er det genfragment som koder fragment B' 1023 bp-området mellom to Sau3Al-punkter, det første av hvilke er den DNA-sekvens som koder det tredje arginin i Z^, og det annet av hvilke er 4 9 bp før enden av det strukturelle tox-gen. Det gen-fragment som koder B<1> er, som nevnt ovenfor, blitt klonet i pUC8. Det plasmid som bærer dette fragment i samme orientering som lac Z-genet er betegnet pDT3 01; ( lac Z- og B'-genene er utenfor rammen på pDT301).
Ved igjen å henvise til det ovenfor angitte flytskjema åpnes pDT3 01 via HindIII-fordøying, og deretter kuttes det B'-kodende fragment ned ved eksponering for exonukleasen Bal31 i varierende tidsrom. Endene av de resulterende avkortede genfragmenter (av varierende lengder) blir deretter butt-ende-ligert med EcoRl og Pstl bindeledd, og fragmentene fordøyes deretter med SauRl. Dette resulterer i en heterogen (hva størrelse angår) populasjon av genfragmenter som koder en del av B<1>.
Disse fragmenter reklones deretter i enten BamHl- Pstl- eller BamHl- EcoRl-punktene i pUC8. Kloning i disse punkter tillater utvelgelse bare av kloner som har BamHl (Sau3AL) på én ende og Pstl eller EcoRl på den annen.
Kloning i dobbelt-fordøyet pUC8 muliggjør også bare én fragment-orientering, og utvelgelse av blå kolonier (de som utkoder lac Z) på X-G-plater verifiserer en forbindelse innen rammen mellom det avkortede B'-kodende område og lac Z. De foretrukne kloner er de hvor det B'-kodende område er 830 bp langt; dvs. at genet er blitt kuttet ned 2 00 bp, til posisjonen til NRU I på tox<228>.
Det neste trinn (iii) er in vitro-syntesen av det genfragment som koder den protease-sensitive loop £2- Dette utføres ved hjelp av konvensjonell fastfase-oligonukleotid-syntese. Sekvensen til dette fragment, inklusive bindeleddene Pstl og EcoRl som er festet etter syntese, er vist nedenunder:
Neste trinn (iv) er å klone det B<1>'-kodende fragment fra trinn (ii) og det i2-kodende fragment fra trinn (iii) til BamHl-EcoRl-punkter på pUC8.
Deretter tilveiebringes det ønskede fragment som koder en del av den celle-spesifikke ligand, enten fra en naturlig kilde eller ved oligonukleotid-syntese. For eksempel kan genfragmenter som koder spesifikke avbindingsdeler av a- og /3-MSH syntetiseres via konvensjonell fastfase-oligonukleotid-syntese.
DNA-sekvensene til disse genfragmenter, sammen med de passende bindeledd, er vist nedenunder:
De unike Pstl- og EcoRl-punkter i pUC8 tillater subkloning av enhver av de ovennevnte syntetiske MSH-frekvenser, nedstrøms for det £2"kodende fragment.

Et sammensmeltet gen som inneholder substans P-genet, snarere enn a- eller /3-MSH-genet, gjøres basisk, som beskrevet ovenfor. Substans P-genet syntetiseres ved anvendelse av konvensjonell fastfase-oligonukleotidsyntese. Substans P-gen-sekvens er:
Som det klart fremgår av ovenstående, må delen av den genetiske sekvens for polypeptidliganden være tilstrekkelig til å muliggjøre at den tilsvarende aminosyresekvens får hybridproteinet til å binde seg til den forutbestemte klasse av celler. Fortrinnsvis vil genet for polypeptidhormonet inkludere alt eller meste-parten av den genetiske sekvens for avbindingsdomenet av liganden.
Generelt, som i de ovenstående eksempler, utføres de mani-pulerende operasjoner ved anvendelse av kloningsvektorer; f.eks. fager eller plasmider. Det genetiske materiale som koder for avbindingsdomenet i polypeptidliganden kan enten være klonet DNA eller en syntetisk oligonukleotidsekvens, alt etter hva som måtte være mest passende for de spesielle ligand-gen som anvendes. Generelt vil det sammensmeltede gen sitte på en kloningsvektor, f.eks. et plasmid eller en fag, som anvendes for transformering av dyrkede mikroorganismer. Hybridproteinet innhøstes så fra dyrkningsmediene for cellene ved anvendelse av konvensjonelle teknikker.
Hybridproteinene som beskrives her administreres til et pattedyr, f.eks. et menneske, som lider av en medisinsk forstyr-relse, f.eks. cancer, karakterisert ved nærvær av en klasse av uønskede celler som en polypeptidligand selektivt kan binde seg til. Mengden av protein som administreres vil variere med typen sykdom, hvor langt sykdommen er kommet og størrelsen og hvilket pattedyr det er som lider av sykdommen. Generelt vil mengdene være i samme område som for andre cytotoksiske midler som anvendes ved behandling av cancer, selv om lavere mengder i visse tilfeller vil være nødvendig på grunn av hybridproteinenes spesifisitet.
Hybridproteinene kan administreres under anvendelse av hvilken som helst konvensjonell metode, f.eks. ved injeksjon, eller ved hjelp av et implantat som frigis med tiden. Når det gjelder MSH-hybrider, kan topiske kremer anvendes for å drepe primær-cancer-celler, og injeksjoner eller implantater kan anvendes for å drepe metastatiske celler. Hybridproteinene kan kombineres med hvilken som helst ikke-toksisk, famasøytisk akseptabel bærersubstans.
Claims (7)
1. Sammensmeltet gen,
karakterisert ved at det omfatter et fragment av det gen som koder for difteri-toksin sammensmeltet til et fragment av et gen som koder for en cellespesifikk polypeptidligand,
idet nevnte sammensmeltede gen koder for et hybridprotein,
idet nevnte fragment av nevnte difteri-toksin-gen inkluderer de deler som koder for (a) det enzymatisk aktive fragment A i difteri-toksin, (b) et fragment som inkluderer spaltningsdomenet £1 som er nærliggende nevnte fragment A i difteri-toksin, (c) et fragment som omfatter minst en del av det hydrofobe domene av fragment B i difteri-toksin og som ikke inkluderer det generaliserte eukaryotiske bindingspunkt i nevnte fragment B, og idet fragmentet av et gen som koder for nevnte polypeptidligand inkluderer minst en del av bindingsdomenet til nevnte polypeptidligand, idet nevnte del av bindingsdomenet er effektiv til å for-årsake at nevnte hybridprotein binder seg selektivt til en forutbestemt klasse av celler som skal angripes av nevnte enzymatisk aktive fragment A.
2. Sammensmeltet gen som angitt i krav 1, karakterisert ved at den del som koder for fragment c) er delen mellom og posisjonen på Nrul-punktet.
3. Sammensmeltet gen som angitt i krav 1 eller 2, karakterisert ved at nevnte fragment i difteri-toksin-genet inkluderer delen £2 mellom den del som koder for fragment c) og det genfragment som koder for liganden.
4. Sammensmeltet gen som angitt i hvilket som helst av kravene 1 til 3, karakterisert ved at nevnte ligand er et hormon.
5. Sammensmeltet gen som angitt i hvilket som helst av kravene 1 til 3, karakterisert ved at det genfragment som koder for nevnte ligand inkluderer en del som koder for en del av a- eller /3-melanocytt-stimulerende hormon som er effektivt til å binde seg til maligne melanocytt-celler.
6. Kloningsvektor, karakterisert ved at den inneholder det sammensmeltede gen som angitt i hvilket som helst av kravene 1 til 5, og kan uttrykke genet.
7. Anvendelse av kloningsvektor som angitt i krav 6, for ekspresjon av en aminosyresekvens ved anvendelse av konvensjonelle fremgangsmåter.
