Description
Foreliggende oppfinnelse angår fremstilling av (E)-2-(p-fluorfenethyl)-3-fluorallylamin med farmakologisk aktivi-tet ved å inhibere monoaminoxydase-B, og som er anvendbar ved behandling av Parkinsons sykdom.
Nærmere bestemt angår oppfinnelsen en analogifremgangsmåte for fremstilling av terapeutisk aktiv (E)-2-(p-fluor-fenylethyl)-3-fluorallylamin
en kraftig og selektiv enzym-aktivert, irreversibel inhibitor av MAO-B, som er anvendbar ved behandling av pasienter som lider av Parkinsons sykdom.
Egnede farmasøytisk akseptable og addisjonssalter
er slike salter avledet fra organiske og uorganiske syrer slik som saltsyre, hydrobromsyre, sulfonsyre, svovelsyre, fosforsyre, salpetersyre, maleinsyre, fumarsyre, benzosyre, ascorbinsyre, pamoinsyre, ravsyre, methansulfonsyre, eddiksyre, propionsyre, vinsyre, sitronsyre, melkesyre, eplesyre, mandelsyre, kanelsyre, palmitinsyre, itaconsyre og benzensulfonsyre.
Analogifremgangsmåten ifølge oppfinnelsen er kjenneteg-net ved at
(a) (E)-2-(p-fluorfenethyl)-3-fluoracrylsyreester
reduseres under dannelse av (E)-2-(p-fluorfenethyl)-3-fluorallylalkohol, og (b) den allyliske hydroxygruppe av (E)-2-(p-fluor-fenethyl)-3-fluorallylalkohol erstattes med en primær aminogruppe under dannelse av (E)-2-(p-fluorfenethyl)-3-fluorallylamin.
Generelt kan fremstilling av forbindelsen utføres ved prosedyrer velkjent innen faget (eksempelvis U.S.
Patent 4.454.158) hvori en diester av p-fluorfenyl-ethylsmørsyre halomethyleres på kjent måte, først ved behandling av diesteren med en sterk syre for å danne det tilsvarende carbanion, hvorpå carbanionet bringes i kontakt med et egnet halomethyleringsmiddel. Den sterke base må være ikke-nucleofi 1 og må ha tilstrekkelig styrke til å fjerne et proton fra methindelen tilstøtende carboxygruppen av utgangsesteren. Slike egnete baser er kjent innen faget. Eksempler er: (a) et alkyllithium (eksempelvis n-butyllithium), (b) et aryllithium (f.eks. fenyllithium), (c) et lithiumdialkylamid (f.eks. lithiumdiisopropylamid), (d) natrium eller lithiumamid, (e) et metallhydrid (f.eks. natrium eller kaliumhydrid), (f) et metallalkoholat (f.eks. natrium eller kalium-tert.-butoxyd), eller (g) lithium eller dilithiumacetylid. Reaksjonen mellom diesteren og basen kan utføres i et aprotisk, organisk løsningsmiddel (slik som tetrahydrofuran (THF), diethylether, dimethylformamid (DMF), dimethylsulfoxyd (DMSO), dimethoxyethan eller dioxan, eller blandinger derav, under anvendelse av et temperaturområde på fra 0 til 70°C, fortrinnsvis 45°C, og en reaksjonstid på 5 minutter til 2 timer. Foretrukne baser for dannelse av carbanionet er natriumhydrid i dimethoxyethan, kalium-tert.-butoxyd/n-butyllithium i THF, eller natrium-tert.-butoxyd i THF.
Etter halomethyleringen foretrekkes det selektivt å fjerne en av estergruppene ved syrehydrolyse. For å bevirke selektiv splitting foretrekkes det å ha en blandet diester hvori en estergruppe lett splittes (eksempelvis barer en estergruppe t-butyl, benzyl, difenylmethyl eller trifenylmethyl) mens den andre bærer en rettkjedet alkylgruppe (eksempelvis methyl, ethyl, propyl eller n-buty1).
Den lett splittbare estergruppe kan selektivt hydrolyseres ved behandling med en organisk eller uorganisk syre, enten med eller uten tilsatt løsningsmiddel, under anvendelse av et temperaturområde på fra 0° til 25°C og en reaksjonstid på fra 1 til 10 timer. Omgivende temperatur foretrekkes. Valget av syre for hydrolysen er ikke kritisk, bortsett fra at syren skal velges slik at den kan lett fjernes etter hydrolysetrinnet. Trifluoreddiksyre er foretrukket da dens lave kokepunkt muliggjør at den lett kan fjernes fra hydrolyseproduktet. Når en estergruppe bærer benzyl, difenylmethyl eller trifenylmethyl, og den andre er en rettkjedet C^-C^-alkylgruppe, kan den lett splittede estergruppe også selektivt splittes ved å underkaste den blandede diester katalytisk hydrogenolyse under anvendelse av konvensjonelle prosedyrer, eksempelvis ved behandling under en hydrogenatmosfære i nærvær av en katalysator (f.eks. Pd/C) ved omgivende temperatur i 1 til 48 timer. Som det fremgår for fagmannen, kan estergruppene velges slik at begge grupper kan splittes samtidig ved syrehydrolyse eller katalytisk hydrogenolyse.
Etter selektiv hydrolyse omdannes den halomethyl-erte monoester til dens acrylatester ved behandling med en base. Reaksjonen kan utføres under anvendelse av et vandig eller ikke-vandig løsningsmiddel med sterke baser slik som natriumhydroxyd og lignende, eller svake baser slik som triethylamin eller natriumbicarbonat kan anvendes. Med sterke baser må forsiktighet utvises for å unngå anvendelse av et overskudd av base for å unngå interaksjon med dobbeltbindingen. Valget av basen, reaksjonsløsningsmidlet og reaksjonsbetingelsene vil være innlysende for fagmannen. En foretrukken prosedyre er å anvende vandig natriumhydroxyd i THF ved omgivende temperatur. Generelt kan et temperaturområde på fra 0° til 25°C og en reaksjonstid på 15 minutter til 2 timer anvendes.
Acrylatesteren reduseres under dannelse av allylalkoholen. Det anvendte reduksjonsmiddel for denne omdannelse kan være et hvilket som helst reagens som er kjent innen faget for å være i stand til selektiv reduksjon av en esterfunksjon eller carboxylsyrefunksjon til den tilsvarende carbinol i nærvær av en dobbeltbinding. Et foretrukket reduksjonsmiddel er diisobutylaluminiumhydrid (DIBAL-H) i hexan, THF, diethylether eller diklormethan, eller blandinger derav. Ved en foretrukket prosedyre avkjøles en løsning av acrylat-methylesteren i THF til 0° til -78°C (fortrinnsvis -60° til -70°C), DIBAL-H oppløst i hexan tilsettes, og temperaturen på blandingen tillates å stige til omgivende temperatur. Reaksjonstiden kan være 2 til 24 timer.
Allylalkoholen kan omdannes til det ønskede allyl-primære amin under anvendelse av prosedyrer som er kjent innen faget for å være anvendbare for å erstatte en allylisk hydroxylgruppe med en allylisk primær aminogruppe. En foretrukket laboratoriemetode innbefatter den direkte dannelse av et imidoderivat, fortrinnsvis fthalimidet, og etterfølgende splitting av imidogruppen for å danne den primære aminogruppen. Imidoderivatet kan hensiktsmessig fremstilles ved behandling av allylalkoholen med det egnete imid (f.eks. fthalimid, succinimid eller maleimid) i nærvær av et triarylfosfin (f.eks. trifenylfosfin) eller et trialkylfosfin og diethylazodicarboxylat i et aprotisk, organisk løsningsmiddel (f.eks. THF eller dioxan). Reaksjonen kan utføres under anvendelse av et temperaturområde på fra 0 til 70°C og en reaksjonstid på fra 1 til 24 timer. Omgivende temperatur foretrekkes. Imidoderivatet kan splittes, fortrinnsvis ved omsetning med hydrazin i et organisk løsningsmiddel slik som en alkanol (f.eks. ethanol) ved tilbakeløpstemperatur (50 til 100°C) og en reaksjonstid på fra 30 minutter til 10 timer. Det foretrekkes å tilsette en syre (f.eks. saltsyre) etter hydrazinbehandlingen for å omdanne produktet til syreaddisjonssaltet. Andre reagenser kan anvendes for å spalte imidofunksjonen. Eksempelvis kan imidet oppvarmes med en sterk uorganisk syre (f.eks. saltsyre eller svovelsyre) eller en blanding av saltsyre og eddiksyre. Syrer slik som hydrobromsyre, som er reaktive overfor olefiner, kan vanligvis ikke anvendes. Sluttproduktene renses hensiktsmessig og isoleres som syreaddisjonssaltet under anvendelse av konvensjonelle rensemetoder.
De foregående prosedyrer kan illustreres ved etterfølgende eksempler.
Eksempel 1
2-( p- fluorfenethyl)- 3- fluorallylamin HC1
Trinn A. Ethyl- 2-( tert.- butoxycarbonyl)- p- fluorfenyl-butyrat
En løsning av 25 g p-fluorfenylsmørsyre i 349 ml tert.-butylacetat ble behandlet med 1,77 ml perklorsyre og ble deretter omrørt ved omgivende temperatur i 1,5 time. Denne ble deretter helt over i 350 ml vann inneholdende 48 g NaOH, og den tertiære butylester ble isolert ved etherekstraksjon som en lys gul olje. En løsning av lithiumdiisopropylamid ble fremstilt fra 22,74 g diisopropylamin og 143,7 ml 1,6M n-butyllithium i 200 ml THF. Denne ble avkjølt til -78°C og en løsning av 26,76 g tert.-butyl-p-fluorfenyl-butyrat i 100 ml THF ble langsomt tilsatt. Etter 1 time ble en løsning av 12,19 g ethylklorformiat i 100 ml THF tilsatt, og omrøringen ble fortsatt ved omgivende temperatur i 24 timer. Blandingen ble deretter helt over i vann, ble nøytralisert med fortynnet vandig HC1, og produktet ble isolert ved etherekstraksjon. På denne måte ble 32,27 g urent malonat erholdt som en orange olje.
Trinn B. Ethyl- 2- ( tert.- butoxycarbonyl) - 2- ( dif luormethyl) -
p- fluorfenylbutyrat
19,81 g fast natrium-tert.-butoxyd ble tilsatt til en løsning av 32,14 g urent ethyl-2-(tert.-butoxycarbonyl)-p-fluorfenylbutyrat i 400 ml THF. Blandingen ble omrørt i 1 time og ble deretter oppvarmet til 45°C ved hvilket tidspunkt ClCHF2~gass ble hurtig tilsatt i løpet av 15 minutter. Omrøringen ble fortsatt i 1 time under en atmosfære av ClCHF2f under hvilket tidsrom temperaturen falt til omgivende temperatur. Reaksjonsblandingen ble helt over i vann/saltvann, og det urene produkt ble isolert som en orange olje (34,55 g) ved etherekstraksjon.
Trinn C. ( E)- ethyl- 2-( p- fluorfenethyl)- 3- fluoracrylat
En løsning av 30,28 g ethyl-2-(tert.-butoxycarbonyl)-2-(difluormethyl)-p-fluorfenyl-butyrat i 168 ml trifluoreddiksyre ble omrørt i 1 time hvorpå overskuddet av trifluoreddiksyre ble fjernet ved fordampning. 25,82 g av restoljen ble oppløst i 230 ml THF og ble langsomt behandlet med 80 ml 2M NaOH slik at pH ikke steg over 7,02. Etter endt tilsetning ble løsningen omrørt i ytterligere 15 minutter, og produktet ble ekstrahert i ether. Etheren ble fordampet, og residuet ble filtrert gjennom en kort kolonne av silica under anvendelse av 5% EtoAc i lett petroleum som løsningsmiddel. Fordampning av løsningsmiddelet ga 15,75 g av hovedsakelig rent produkt som en lys orange olje.
Trinn .0. ( E) - 2- ( p- f luorf enethyl) - 3- f luorallylalkohol
En løsning av 15,70 g av acrylatet i 350 ml hexan avkjølt i -10°, ble langsomt behandlet med en løsning av diisobutylaluminiumhydrid i hexan (1M løsning, 196 ml). Løsningen ble omrørt ved omgivende temperatur i 90 minutter, ble deretter avkjølt til 10°C og ble behandlet i rekkefølge med 196 ml CH30H og 245 ml 6M vandig HC1. Vann ble tilsatt, og produktet ble isolert ved etherekstraksjon etterfulgt av destillasjon av løsningsmidlene under dannelse av 11,36 g praktisk talt ren alkohol.
Trinn E. ( E)- 1- fluor- 2-( p- fluorfenethyl)- 3- fthalimido-
propen
En løsning av 11,36 g av den urene alkohol, 8,43 g fthalimid og 5,03 g trifenylfosfin i 400 ml THF ble avkjølt til 0°C og ble langsomt behandlet med en løsning av 9,99 g diethylazodicarboxylat i 50 ml THF. Omrøringen ble fortsatt ved omgivende temperatur over natten, hvorpå løsningen ble fordampet under dannelse av 30 g av en orangefarget pasta. Kromatografi på silica (20% EtoAc i petroleumether som elueringsmiddel) muliggjør separering av 13,90 rent produkt som et lysegult, fast materiale.
Trinn F. En blanding av 0,26 g av fthalimidet og 80 mg hydrazinhydrat i 5 ml ethanol ble kokt under tilbake-løpskjøling i 2,5 time. 1,2 ml 6N vandig HC1 ble tilsatt, og blandingen ble fordampet til tørrhet. Residuet ble oppløst i 10 ml NaOH, og det urene amin ble isolert ved ethereks-traks jon og ble deretter oppløst i 10 ml TKF og ble behandlet med 194 mg di-tert.-butyldicarbonat. Løsningen ble kokt under tilbakeløpskjøling i 2 timer hvorpå det urene N-BOC-derivat ble isolert ved etherekstraksjon. Rensing ble oppnådd ved silicakromatografi (25% EtoAc i petroleumether) hvorpå 180 mg rent materiale ble erholdt som en praktisk talt fargeløs olje. Denne ble oppløst i 12 ml hydrogen-klorid-mettet ether, fikk stå over natten og ble deretter filtrert under dannelse av 30 mg av hydrokloridsaltet av (E)-2-(p-fluorfenethyl)-3-fluorallylamin som fargeløse plater; smeltepunkt 131°C.
Monoaminoxydase-(MAO) inhibitorer har vært anvendt klinisk i over 20 år. I 1970-årene ble det fastslått at MAO var to enzymer med forskjellige fordelinger, substrat- og inhibitorsensibiliteter. I mennesket metaboliserer type A-enzymet primært norepinefrin og serotonin, mens type B-enzymet er viktig for metabolismen av dopamin. MAO type A inhiberes selektivt av klorgylin. L-deprenyl inhiberer selektivt MAO type B. L-deprenyl har i et stort antall kliniske forsøk vist seg å være gunstig for Parkinsonpasienter. Inhibitoren anvendes i kombinasjon med L-dopa og en decarboxylaseinhibitor og har særlig verdi ved regulering av "på-av"-effekten og dyskinese etter "endt dose". Enn videre antyder analyse av data fra et stort antall Parkinsonpasienter behandlet med L-deprenyl pluss "Madopar"
(L-dopa + benserazid) sammenlignet med "Madopar" alene at L-deprenylbehandling forlenger signifikant antatt levealder. Relevant for denne observasjonen er det blitt vist i mus og aper at L-deprenylbehandling blokkerer den neurotoksiske effekt av MPTP (N-methyl-4-fenyl-1,2,3,6-tetrahydropyridin) som fremkaller Parkinsonsyndromet i
mennesket. Det er blitt foreslått at et MPTP-lignende neurotoksin kan være årsaken til idiopatisk Parkinsons sykdom.
I mennesker og dyr omdannes L-deprenyl hurtig til L-amfetamin og methamfetamin, og disse produkter kan bidra til enkelte av de bivirkninger som observeres med L-deprenyl. Enn videre har L-deprenyl bare begrenset selektivitet overfor MAO-B-enzymet. En forbindelse med forøket selektivitet overfor MAO-B-enzymet og som er fri for amfetamin-lignende effekter, vil således være av betydelig verdi ved Parkinsons sykdom. Forbindelsen ifølge oppfinnelsen har en biokjemisk og farmakologisk profil som oppfyller disse kriterier. (E)-2-(p-fluorfenethyl)-3-fluorallylamin er en kraftig og selektiv enzym-aktivert, irreversibel inhibitor av MAO-B. In vitro er IC50-verdiene for inhibering av rottehjerne mitokondrisk MAO-A og -B 6,8 x 10~^M og 3,6 x 10~^M, og et selektivitetsforhold på 190 i favør av MAO-B-inhibering. In vivo er forbindelsen aktiv oralt, er kraftig, og med et selektivitetsforhold på 44 forblir den selektiv overfor MAO-B.
Etter gjentatt daglig administrering i 14 dager forble (E)-2-(p-fluorfenethyl)-3-fluorallylamin selektiv overfor MAO-B-enzymet. Fra den irreversible inhibering, virkningsmåte og lange halveringstid av enzymet er en lavere daglig dose nødvendig for å inhibere enzymet. I rotter er de nødvendige orale doser for å inhibere med 50% MAO-A- og MAO-B-enzymet i forskjellige vev, vist i tabell 1.
MPTP gitt parenteralt til mus, fortrenger striatalt dopamin. Denne effekt kan forhindres av MAO-B-, men ikke MAO-A-inhibitor. (E)-2-(p-fluorfenethyl)-3-fluorallylamin beskytter mus mot den neurotoksiske effekt av MPTP slik det fremgår fra tabell 2.
Ikke-selektive og MAO-A-selektive inhibitorer er kjent for å fremkalle alvorlige cardiovaskulære interaksjoner med tyraminholdige næringsmidler, den såkalte "ostereaksjon". L-deprenyl er fri for denne effekt. Gitt akutt i 10 og 50 ganger ED50-dosen for inhibering av MAO-B, er (E)-2-(p-fluorfenethyl)-3-fluorallylamin er også fritt for interaksjoner med tyramin slik det fremgår fra tabell 3.
<*>Tyraminpotensieringen ble beregnet fra en dose-responskurve av tachycardiaresponsen på intraduodenalt (i.d.) tyramin (0,3-50 mg/kg) og intravenøst (i.v.) tyramin (1,25-80 /ug/kg) ved sammenligning av ED50-dosen for tyramin i kontrolldyr mot den i behandlete dyr.
For dens sluttelige anvendelse for behandling av Parkinsons sykdom kan (E)-2-(p-fluorfenethyl)-3-fluorallylamin anvendes i doseområder på fra 0,01 til 1 mg pr. kg kroppsvekt pr. dag.
Mår den anvendes for å behandle Parkinsons sykdom, vil selvsagt den effektive dose av forbindelsen variere i henhold til arten og strengheten av sykdommen og den bestemte pasient som behandles. Generelt kan effektive resultater oppnås ved administrering av forbindelsen ved et doseringsnivå på fra 0,1 til 1 mg pr. dag pr. kg kroppsvekt. I praksis bør behandlingen startes systemisk ved lavere doser, hvoretter dosen administeres oralt i faste doseringsformer, f.eks. kapsler, tabletter eller pulvere, eller i vaskeformer, f.eks. løsninger eller suspensjoner. Forbindelsene kan også injiseres parenteralt i form av sterile løsninger eller suspensjoner. Faste, orale former kan inneholde konvensjonelle eksipienter slik som f.eks. lactose, sucrose, magnesiumstearat, harpikser og lignende materialer. Væskeformige, orale former kan inneholde forskjellige smaksgivende midler, fargestoffer, konserveringsmidler, stabiliseringsmidler, oppløseliggjørende midler eller suspenderingsmidler. Om ønsket, kan additiver slik som saltvann eller glucose, tilsettes for å gjøre løsningene isotoniske. Når forbindelsen anvendes ved samtidig behandling med L-dopa, er lavere mengder av L-dopa nødvendig, og enhver bivirkning forbundet med store mengder av L-dopa, vil således bli redusert eller unngått.
