Description
Foreliggende oppfinnelse vedrører analogifremgangsmåte ved fremstilling av nye fenoksypropanolaminer.
Fenoksypropanolaminene som fremstilles ifølge oppfinnelsen er forbindelser med formel

og fysiologisk forenlige salter derav.
Det her anvendte uttrykk "lavere" betegner rester med 1-6 karbonatomer, idet rester med 1-4 karbonatomer er foretruk-ket. Alkyl- og alkoksygrupper kan være rettkjedet eller for-grenet. Eksempler er metyl, etyl, propyl, isopropyl, n-butyl og isobutyl hhv. metoksy, etoksy, propoksy, isopropoksy, but-oksy og isobutoksy.
Forbindelsene med formel I danner med syrer salter. Eksemp-
ler på slike salter er salter med fysiologisk forenlige mineral-syrer såsom saltsyre, hydrogenbromid, svovelsyre, fosforsyre; eller med organiske syrer såsom metansulfonsyre, eddiksyre, propionsyre, sitronsyre, oksalsyre, ravsyre, eplesyre, fumar-
syre, fenyleddigksyre eller salisylsyre. Karboksylsyrer med formel I kan foreligge som salter. Eksempler for slike salter er alkalimetall-, jordalkalimetall-, ammonium- og alkylammo-niumsalter såsom Na-, K-, Ca-, trimetylammonium- og etanol-ammoniumsalter.
Forbindelsene med formel I inneholder i det minste to asymme-triske karbonatomer og kan således foreligge som optisk aktive enantiomerer, som diastereomerer eller som racemater.
Forbindelsene med formel I kan ifølge oppfinnelsen oppnås ved at man
a) omsetter et epoksyd med formel
eller et P -ketohaiogenid med formel med et primært amin med formel eller et sekundært amin med formel
idet n, Y og Z i formel II-V har ovennevnte betydning,

med formel IV,
og reduserer en i en erholdt forbindelse foreliggende Xi<->C(0)- eller X<2>C(0)-gruppe til en X<1->CHOH- hhv. X -CHOH-gruppe, og eventuelt
funksjonelt modifiserer om ønsket en reaksjonsdyktig substituent i en gruppe Z av en forbindelse med formel I,
ved å forsåpe en estergruppe -C(0)R , hydrogenere en dobbeltbinding, foretre en hydroksygruppe eller redusere en ester- eller amidgruppe - C(0}R 2 til en tilsvarende hydroksymetyl- henholdsvis aminometylgruppe, eller eventuelt
omdanner en forbindelse med formel I, hvor R er hydrogen til en forbindelse med formel I, hvor R betyr lavere alkanoyl eller fenyllavere alkanoyl, og eventuelt
overfører om ønsket en forbindelse med formel I i et salt.
Omsettingen av en forbindelse med formel II med en forbindelse med formel IV eller V kan foretas på en for omsettingen av epoksyder med aminer til aminoalkholer i og for seg kjent måte. Hensiktsmessig sammenblandes reaksjonspartnerne i et egnet oppløsningsmiddel og oppvarmes. Som oppløsnings-midler er inerte organiske oppløsningsmidler, f.eks. dimetyl-sulfoksyd (DMSO), acetonitril eller eter såsom tetrahydrofuran (THF) eller dioksan;' eller alkoholer såsom etanol, aktuelle. Reaksjonstemperaturen er ikke kritisk, hensiktsmessig arbeider man ved temperaturer mellom 60°C og reaksjons-blandingens kokepunkt.
Omsettingen av en forbindelse med formel III med en forbindelse med formel IV eller V kan likeledes gjennomføres på i og for seg kjent måte, hensiktsmessig i nærvær av et apro-tisk oppløsningsmiddel såsom et halogenert hydrogenkarbon, f.eks. kloroform, ved en temperatur inntil 2 00°C.
I det således erholdte reaksjonsprodukt med formel
I kan en reaksjonsdyktig substituent, spesielt en gruppe -C(0)R<2> eller -C(R3)=CH-(CH2)m-C(0)R2 funksjonelt modifiseres. Forestringen av en karboksylgruppe kan foretas på i og for seg kjent måte, f.eks. ved hjelp av alkylhalogenider såsom metyljodid, og en base. Forsåpningen av en estergruppe foretas hensiktsmessig under alkaliske betingelser, f.eks. ved hjlep av vandig-alkoholisk alkalihydroksyd, f.eks. vandig-metanolisk kaliumhydroksyd. En i en sidekjede R"<*>" eller R"<1>"<1 >foreliggende dobbeltbinding kan f.eks. hydreres i nærvær av en katalysator såsom palladium på kull, i et oppløsningsmid-del såsom en lavere alkanol f.eks. etanol, for enkeltbinding. En hydroksyrest R"<*>""<*>" kan foretres på i og for seg kjent måte, f.eks. ved omsetting med et mesylat eller halogenid som til-svarer eterresten og i nærvær av en base såsom kaliumhydroksyd i et oppløsningsmiddel såsom en lavere alkanol, f.eks. n-propanol, eller i nærvær av kalium-t-butylat i et oppløs-ningsmiddel såsom DMSO.
En eventuelt mono- eller di-laverealkylert karbamoylgruppe
R<1> eller R<11> kan ved hjelp av reduksjon f.eks. med komplek-se metallhydrider såsom LiAlH^, reduseres til den tilsvarende aminometylgruppe. På analog måte kan en lavere-alkoksy-karbonylgruppe reduseres til hydroksymetylgruppen.
Alkanoyleringen eller fenalkanoyleringen av hydroksygruppene som foreligger i Y-stilling til N-atomet av en diol med for-
mel I eller en alkohol med formel I, kan gjennomføres på
i og for seg kjent måte, f.eks. ved hjelp av den tilsvarende karboksylsyre eller et tilsvarende syrehalogenid i nærvær av en sterk syre såsom saltsyre.
Foretrukne forbindelser med formel I er disse, hvor den i en fenylgruppe Z foreliggende substituent R<11> er hydroksy, sulfamoyl eller en gruppe R<1>, -0-(CH_) -0H eller -0-(CH9)

Spesielt foretrukkede forbindelser med formel I er
dessuten slike, hvor C-atomet som er bundet til en metyl— rest Y, har R-konfigurasjonen, hvor C-atomet som er bundet

til en fenoksymetylrest X eller X har S-konfigurasjonen og C-atomet som er bundet til en fenylrest X 2 har R-konfigurasjonen.

Aminene med formel I samt de fysiologisk forlikelige
salter derav kan anvendes som aktivstoffer i farmasøytiske preparater ved behandling av fettsyke og/eller diabetes mellitus, spesielt av overvektige voksne diabetikere. Ved ekspe-rimenter med dyr kunne ved administrasjon av ovennevnte forbindelser iakttas en øket katabolisme, frem for alt av fette-, ne. Dessuten ble det funnet at forbindelsene stimulerer dannelsen av brunt fettvev ved rotter og overvektige hyperglykemiske mus. Som kjent tilskrives defekter i det brune fettvev en vesentlig rolle når det oppstår fettsyke. På overvektige hyperglykemiske mus har forbindelsene en utpre-get antidiabetisk effekt, idet de virker hypoglykemisk og
forminsker glykosurien. Disse forbindelser viser kun en liten virkning på hjerteaktivitet og kretsløp. Doseringen kan avhengig av virkningsstyrken av de enkelte forbin-
delser og pasientens individuelle behov utgjøre 0,5-100 mg fortrinnsvis 2-200mg pr. dag for voksne, idet dosen kan administreres som enkeltdose eller i flere doser fordelt over en dag.
Dessuten kan det med ovennevnte forbindelser ved eksperi-menter med dyr påvises en økning av proteininholdet og en minskning av fettinnholdet i kroppen. Disse forbindelser fører således til en økning av den magre kropsmasse på bekostning av fett. Derfor kan de først og fremst anvendes i humanmedisinen ved behandling av tilstander som er for-bundet med øket proteinnedbrytning, f.eks. ved rekonvale-
sens efter operasjoner. Herved ligger administrasjonsdosene i samme ramme som ved behandling av fettsyke og/eller diabetes mellitus.
Ovennevnte forbindelser kan også anvendes i ernæringen av kjøttproduserende dyr såsom storfe, griser, sauer og fjær-kre. Herved kan administrasjonsdosene og administrasjonsfor-mene være de samme som for vitaminer. Disse forbindelser kan også anvendes som fortilsetninger i doser på 0,01-100 mg/kg efter substans, dyreart og alter.
De farmasøytiske preparater inneholder aktivstoffet sammen med et aksepterbart farmasøytisk, organisk eller uorganisk bære-materiale såsom f.eks. vann, gelatine, gummi arabicum, laktose, stivelse, magnesiumstearat, talkum, planteolje, polyalkylengly-koler, vaseline o.l.. De farmasøytiske preparater administreres fortrinnsvis oralt, f.eks. i form av tabletter, kapsler, piller, pulver, granulater, oppløsninger, sirupper, suspensjo-ner eller eliksierer. Administrasjonen kan imidlertid også skje parenteral, f.eks. i form av sterile oppløsninger, suspen-sjoner eller emulsjoner. De farmasøytiske preparater kan være steriliserte og/eller inneholder bestanddeler såsom konserve-ringsmidler, stabilisatorer, fuktemidler, emulgatorer, salter for å variere det osomotiske trykk, og buffersubstanser.
Ovennevnte forbindelsers aktivitet vil fremgå tydelig fra de efterfølgende forsøksresultater:
1) Virkning på oksygenforbruket
Hvite hanrotter med en vekt på 160-180 g ble satt i stoffskiftebur efter 24 timers fasting. Burene ble luftet med konstant 6 liter romluft/minutt, som ble ekvilibrert ved et duggpunkt på 11°C. Av den utpustede luft ble det efter ny ekvilibrering i perioder på 14 minutter samlet prøver og oksygen- og C02_innholdet ble analysert. Efter en tilpasnings-tid på 4 timer fikk dyrene som var delt i grupper på 6, enten placebo (5 % gummi arabicum) eller testforbindelsen (suspendert i 5% gummi arabicum) pr. os. Derefter ble be-stemmelsen gjennomført i løpet av 12 timer. I tabell I er det angitt prosentsatsen av oksygenforbruket efter medikasjon i løpet av de første 3 timer og av den totale forsøkstid (12 timer) oksygenforbruket av tilpasningsperioden, idet tilsvarende korrekturer for forandringer i placebo-gruppen ble tatt hensyn til.
2) Katabolsk virkning på lipidene
Grupper på 4 hvite hanrotter med en vekt på 320-360g ble holdt uten tilgang til for i stoffskiftebur. Oksygenforbruket og (X^-produksjonen ble målt i 12 timer. Efter
4 timer fikk dyrene placebo (5 % gummi arabicum) eller testforbindelsen (suspendert i gummi arabicum) pr. os. I tabell II er det angitt avtagelsen av den respiratoriske.kvotient (CO2/O2) i 8 timer efter administrasjon av testforbindelsen i sammenligning med de siste 3 timer før administrasjon av testforbindelsen.De iplacebo-gruppen inntrufne forandringer ble tatt hensyn til ved beregningen.
3) Virkning på urin- og blodglykose og dannelsen av brunt fettvev
Hyperglykemiske fettmus av hunnkjønn ble tilpasset en formengde som var begrenset på 3 g/dag/dyr. Testforbindel-sene (suspendert, i 5% gummi arabicum) eller placebo (5 % gummi arabicum), ble administrert oralt i 15 dager to ganger daglig. Urin ble samlet i 6 dager pr. uke og uringlukusen ble bestemt. Blodglukosen og vekten av det interskapulære brune fettvev ble bestemt på slutten av forsøket.
Forsøksresultatene er angitt i tabell III som prosentsatser av kontrollverdiene.
De i de efterfølgende eksempler anvendte utgangsmaterialer, spesielt aminene med formel V, hvor X er lavere-alkyl eller eventuelt ved fluor, klor, trifluormetyl eller lavere-alkoksy monosubstituert fenyl, og aminene med formel IV, er kjent hhv. kan fremstilles på i og for seg kjent måte f.eks. som i det etterfølgende eller i de europeiske patentansøkninger 6735, 21636 og 94595 beskrevet.
Ved fremstilling av utgangsaminet i eksempel 1 og 10e
ble (RS)-4-(2-tienyl)-2-butanol, acetylklorid og aluminiumklorid i metylenklorid omsatt til (RS)-3-(5-acetyl-2-tienyl)-1- metylpropylacetat. Med NaOH i metanol ble dette forsåpet til (RS)-5-(3-hydroksybutyl)-2-tienylmetylketon. Dette rea-gerte i etanol i nærvær av natriumetanolat med trietylfosfono-acetat til (E)-5-/TRS)-3-hydroksybutyl7- -metyl -2-tiofena-crylsyreetylester. Omsettingen med p-toluensulfoklorid og derefter behandlingen med natriumazid ga (E)-5-/~(RS)-3-azidobutyl/-P-metyl -2-tiofenakrylsyreetylester. Derfra ble ved reduksjon med trifenylfosfin og derefter hydrolyse oppnådd (E)-5-/TRS)-3-aminobutyl7-Z<5->metyl -2-tiof enakryl-syreetylester, £ ^20 = 17465.
Ved fremstilling av utgangsaminet i eksempel 5 ble 2- (2-tienyl)etyl-p-toluensulfonat, acetylklorid og aluminiumklorid i metylenklorid omsatt til 2t-,/75-acetyl-2-rtienyl) etyl7~ p-toluensulfonat (smp. 111-112°C, fra etanol). Dette ble mednatriumazid i DMSO overført til 5-(2-azidoetyl)-2-tienyl-metylketonet. Oksydasjon med natriumhypobromit leverte 5-(2-azidoetyl)-2-tiofenkarboksylsyre med smp. 53-55°C, som med tionylklorid ga det tilsvarende syreklorid som ble omsatt med ammoniakk til 5-(2-azidoetyl)-2-tiofenkarboksylsyreamid (smp. 105-105°C, fra etanol). Reaksjon med trifenylfosfin og hydrolyse ga 5-(2-aminoetyl)-2-tiofenkarboksylsyrearnidet, smp. 134-136°C (fra acetonitril).
Ved fremstilling av utgangsaminet i eksempel 6 og 10g) ble 5-/TRS)-2-hydroksypropyl7_2-tienylmetylketon og trietyl-fosfonoacetat i etanol i nærvær av natriumetylat omsatt til (E)-5-/7RS)-2-hydroksypropyl/- /3-metyl -2-tiofenakrylsyre-etylester. Med p-toluensulfoklorid ble derav oppnådd (E)-(i-metyl-5-^TRS) 2-/7p-toluensulfonyl)oksy/propyl/ -2-tiofen-akrylsyreetylesteren (smp. 121°C, fra metylenklorid-alkohol). Reaksjonen med natriumazid i DMSO ga (E)-5-/7RS)-2-azidopro-pyl/-P-metyl -2-tiofenakrylsyreetylesteren. Reduksjon med trifenylfosfin og hydrolyse førte til (E)-5-/TRS)-2-amino-propyl/- ( i -metyl -2-tiofenakrylsyreetylesteren, f 320 = 17970.
Ved fremstilling av utgangsaminet i eksempel 7, ble 4-(5-acetyl-2-tienyl)-2-butanon med etylenglykol, o-maursyretri-etylester og p-toluensulfonsyre i metylenklorid omsatt selek-tivt til metyl-5-</2- (2-metyl-l,3-dioksolan 2-yl)etyl/-2-tienyl-keton. Oksydasjon med natriumhypobromit og etterfølgende hydrolyse ga 5-(3-oksobutyl)-2-tiofenkarboksylsyre. Med NaBH^ ble derav oppnådd 5-(3-hydroksybutyl)-2-tiofenkarboksylsyren som i dimetylacetamid med metyljodid og natriumbikarbonat ble overført i metylesteren. Behandlingen med p-toluensulfoklorid i pyridin og reaksjonen med natriumazid i DMSO ga 5-(3-azidobutyl)-2-tiofenkarboksylsyremetylestern fra hvilken man ved forsåpning oppnådde den tilsvarende syre. Med tionylklorid ble det fremstilt dennes syreklorid fra hvilket med konsentrert ammoniakk i dietyleter man oppnådde 5-(2-azidobutyl)-2-tiofenkarboksylsyreamidet. Reduksjon av azido-gruppen med trifenylfosfin og etterfølgende hydrolyse ga (R,S)-5-(3-aminobutyl)-2-tiofenkarboksylsyreamidet, smp. 65-75°C, £256 = 7780, 6275 = 9900.
Ved fremstilling av utgangsaminet i eksempel 8 blir (RS)-5-(3-hydroksybutyl)-2-tienylmetylketon omsatt med p-toiuensulfo-klorid til (RS)-3-(5-acetyl-2-tienyl)-1-metylpropyl-p-toluensulfonat, smp. 61-63°C. Med natriumazid oppnår man derav metyl-(RS)-5-(3-azidobutyl)-2-tienylketonet, som hydreres katalytisk til 2-acetyl-5-i/TRS)-3-aminobutyl7tiofen.
Ved fremstilling av utgangsaminet i eksempel 9 ble 2-(p-tolu-ensulfonyloksy)-propyltiofen omsatt med acetylklorid og alumi-niumtriklorid i metylenklorid til 5-acetyl-2-(p-toluensulfo-14
nyloksy)-propyltiofen. Med natriumazid i DMSO ble derav oppnådd 5-(3-azidopropyl)-2-tienylmetylketonet. Oksydasjon med natriumhypobromit ga 5-(3-azidopropyl)-2-tiofenkarboksylsyre, smp. 71-72°C. Omsettingen av denne syre med tionylklorid og etterfølgende behandling med konsentrert ammoniakk førte til 5-(3-azidopropyl)-2-tiofenkarboksylsyreamid, smp. 85-87°C. Derav oppnår man efter behandling mea trifenylfosfin og hydrolyse 5-(3-aminopropyl)-2-tiofenkarboksylsyreamidet, smp. 143,5-155°C (fra vann).
Ved fremstilling av utgangsmaterialet i eksempel 10a ble p-(2-brometyl)-acetofenon omsatt med natriumazid i DMSO til p-(2-azidoetyl)acetofenon. Oksydasjon med natriumhypobromit ga p-(2-azidoetyl)benzosyre (smp. 130-131°C, fra aceton-heksan), som med tionylklorid gikk over til det tilsvarende syreklorid og derefter med ammoniakk til p-(2-azidoetyl)benzamid. Behandling med trifenylfosfin og hydrolyse ga p-(2-aminoetyl)-benzamidet, smp. 132-133°C (fra etanol).
Ved fremstilling av utgangsaminet i eksempel 10b, 5-(3-aminopropyl)-2-tiofenkarboksylsyre-metylesteren, forestres 5-(3-azidopropyl)-2-tiofenkarboksylsyren med metyljodid og hydreres katalytisk den herved oppnådde 5-(3-azidopropyl)-2-tiofenkarboksylsyremetylester.
Ved fremstilling av utgangsaminet i eksempel 10f ble c*.-metyl-2-tiofenetanol, acetylklorid og aluminiumklorid i metylenklorid omsatt til (RS)-2-(5-acetyl-2-tienyl)-1-metyletylacetat. Med natronlut i metanol ble dette forsåpet til 5-/7RS)-2-hydroksypropyl/-2-tienylmetylketon og derefter omsatt med p-toluensulfoklorid til (RS)-2-(5-acetyl-2-tienyl)-1-metyl-etyl-p-toluensulfonat, smp. 101-103°C. Med natriumazid i DMSO fikk man derfra 5-/TRS)-2-azidopropyl/-2-tienylmetylketonet som ble oksydert med brom i natronlut til 5-/TRS)-2-azidopro-pyl/-2-tiofenkarboksylsyre. Med SOCI2 ble derfra fremstilt det tilsvarende syreklorid og fra dette med ammoniakk 5-/TRS)-2-azidopropyl7_2-tiofenkarboksylsyreamidet (smp. 79-80°C, fra dietyleter). Behandling med trifenylfosfin og hydrolyse ga 5-/(RS)-2-aminopropyl/-2-tiofenkarboksylsyre-amidet, smp. 91-92°C fra acetonitril.
Ved fremstilling av utgangsaminet i eksempel 24 ble p-amino-sulfonylbenzaldehyd omsatt med dietyl-cyanometylfosfonat/NaH 1 tetrahydrofuran til l-cyano-2-(4-aminosulfonylfenyl)-etan som i metanol ble hydrert med Raney-kobalt som katalysator til 3- (4-aminosulfonylfenyl)-propylamin.
EKSEMPEL 1
2 g (E)-5-/TRS)-3-//TR)-2-hydroksyfenetyl7amino7-butyl/-P metyl-2-tiofenakrylsyreetylester (smp. 72°) og 850 mg fenylglycideter ble oppvarmet i 20 ml DMSO til 90°C. Efter 20 timer ble ytterligere 850 mg fenylglycideter tilsatt og i ytterligere 20 timer oppvarmet til 90°C. Efter avkjøling ble reaksjonsblandingen helt på vann og ekstrahert med metylenklorid. Kromatografi av råproduktet ga 1,8 g (E) -5-/~(RS) -3-^2~(R) -2-hydroksyf ene tyl/-^"(RS )-2-hydroksy-3-fenoksypropyl7aming7butyl7-

EKSEMPEL 55
2,1 g p-/7lR)-2-/7RS)-2-hydroksy-3-fenoksypropyl7amino7~ butyl? -N-butylbenzamid ble i 92 ml THF blandet porsjonsvis med 920 mg LiAlH4 og kokt i 4 timer ved tilbakeløp. Reaksjonsblandingen ble blandet med 25 ml 2N NaOH, fortynnet med vann og ekstrahert 2 x med metylenklorid. Metylenkloridopp-løsningen ble vasket 2 x med vann, tørket og inndampet i vakuum. Det ble oppnådd 2.06 g (RS) -1-//7R) -3-/oc - (butylamino) - p-tolyl/-l-metylpropyl7amino7-3-fenoksy-2-propanol, /Oc_7 265
= +6°C (0,1 % i MeOH).

Analogt med eksempel 55 ble det av 5-/3-/bis-/"(RS)-2-hydroksy-3-fenoksypropyl7aming7propyl7 -N-butyl-2-tiofenkarboksylsyre-amid oppnådd 1,1'-,/23-Z5-/Tbutylamino)metyl7-2-tienyl7propyl7-imino7bis/7RS)-3-fenoksy-2-propano<l>?, ^ 220 = 2°120' ^241 = 9090, i 2?0 = 3350, £277 = 2800.
EKSEMPEL 57
Til en suspensjon av 760 mg LiAlH4 i 60 ml THF ble det dryppet i 20 minutter en oppløsning av 1,79 g 5-/3-//7RS)-2-hydroksy-2-fenoksypropyl/amino/propyl/ -N,N-dibutyl-2-tiofenkarboksyl-syreamid i 28 ml THF ved 25°C og rørt i enda 2,5 timer ved 25°C. Opparbeidelsen skjedde som angitt i eksempel 55 og ga 1,73 g (RS)-l-/^3-/5-/7dibutylamino)metyl7 -2-tienyl7propyl7~ amino7~3-fenoksy-2-propanol, ^ 220 = 13050, <^ 240 = 9350, { 2?0 = 1950, <f2?7 <=><1>560.

1. Analogifremgangsmåte ved fremstilling av terapeutisk aktive fenoksypropanolaminer med formel
hvor Q betyr hydrogen eller en gruppe
n er tallet 1 eller 2, - og
R er hydrogen, lavere-alkanoyl eller fenyl-lavere-alkanoyl,
X er eventuelt i orto-stilling mono-fluorert eller -klorert
fenoksymetyl,
X 2 er eventuelt i orto-stilling moho-fluorert
eller -klorert fenoksymetyl eller eventuelt ved fluor, klor, trifluormetyl eller lavere-alkoksy monosubstituert fenyl,
Y er hydrogen eller metyl,
Z er en gruppe med formel
R er eventuelt N-mono- eller N-di-lavere-akylert amino-
metyl eller en rest -C(0)R 2,<-->CC((HR3,R)=3C)H--(C(HCH22)<ar>)<m>+<-1C-C>(<0>(<0>)<R>)<R22,,>-C(H,R3)-(CH2)p-0H eller -C (R3) =CH-C (CH-,) =CH-COOCH- ,

eventuelt omdanner en forbindelse med formel I, hvor R er hydrogen til en forbindelse med formel I, hvor R betyr lavere alkanoyl eller fenyl-lavere alkanoyl, og eventuelt
overfører om ønsket en forbindelse med formel I i et salt. 2?
