remgangsmåte til fremstilling av forbindelse med den generelle formel. hvori X betyr gruppen i CO eller ^CH(OH), Rbetyr hydrogen eller en C-Cg-alkylgruppe og R3 betyr hydrogen eller en hydroksygruppe og Rbetyr adamantylresten eller en mettet eller en enkelt umettet C-C-cyklo-lkylrest, idet denne C-C-cykloalkylrest også kan være substituert med en C-C^-alkylgruppe eller et halogenatom, samt deres syreaddisjonssalter ved omsetning av tilsvarende N,N'-metylen-bis-oksazolidiner med ketoner med formel R^-CO-C(R2)H2 i surt medium og eventuelt etterfølgende reduksjon.
Description
Fra DE-OS 29 19 49 5 er det kjent forskjellige fremgangsmåter til fremstilling av forbindelser med den generelle formel
hvori X betyr gruppen CO eller CH(OH), R2 betyr hydrogen eller en C^-Cg-alkylgruppe og R-j betyr hydrogen eller en hydroksygruppe og R.^ betyr adamantylresten eller en mettet eller enkelt umettet C^-C^g-cykloalkylrest, idet denne C3-C1g-cykloalkylrest også kan være substituert med en C1~C4-alkylgruppe eller et halogenatom, samt deres salter. Den viktigste av disse fremgangsmåter består i at et amin med den generelle formel hvori R^ har den angitte betydning, omsettes med med en forbindelse med den generelle formel
idet X, R-, og R2 har den angitte betydning og E betyr en metylengruppe eller et hydrogenatom og gruppen -CHj-RRgRtø og R& og R^ er lavmolekylære alkylrester, som også kan være sluttet til en ring eller idet E også kan bety to hydrogenatomer når X betyr CO-gruppen og arbeides i nærvær av formaldehyd eller et formaldehydleverende stoff og eventuelt reduseres i de dannede forbindelser ved en iso-lert dobbeltbinding og/eller en CO-gruppe. De andre i DE-OS 29 19 49 5 angitte fremgangsmåter er mer omstendelige og kommer praktisk talt ikke i betraktning for den tekniske
fremstilling.
Fremgangsmåten ifølge oppfinnelsen går en ny vei for fremstilling av forbindelser med formel I, den gir eksempelvis bedre utbytter og er enklere å gjennomføre.
I formel I betyr R2 fortrinnsvis en C-^-C^-alkylgruppe, spesielt metyl eller etyl. Den mettede eller umettede C^-C^g-cykloalkylrest består fortrinnsvis av 3 til 12 C-atomer, spesielt 3 til 8 C-atomer. Hvis denne cykloalkylrest er substituert, da dreier det seg fortrinnsvis om en eller to like, respektiv forskjellige substituenter som metyl, etyl, klor, brom og/ eller fluor.
Fremgangsmåten ifølge oppfinnelsen foregår eksempelvis under omsetning av 1 mol av en forbindelse med formel II med 2 - 3 mol, spesielt 2,3 - 2,5 mol av et keton med formel R^-CO-CR2H2
i nærvær av en syre i et oppløsningsmiddel ved en temperatur mellom 20 til 150°C.
Fremgangsmåten gjennomføres generelt i et inert oppløsnings-respektiv suspensjonsmiddel ved temperaturer mellom 5 og 250°C, fortrinnsvis 20 og 150°C, spesielt mellom 40°C og 90°C. Som oppløsningsmiddel kommer det eksempelvis i betraktning: lavere alifatiske alkoholer (etanol, metanol, isopropanol, propanol), mettede alicykliske og cykliske etere (dioksan, tetrahydrofuran, dietyleter), lavere alifatiske ketoner (aceton), lavere alifatiske hydrokarboner eller halogenhydrokarboner (kloroform, 1,2-dikloretan), aromatiske hydrokarboner (benzen, xylen, toluen), iseddik, vann respektiv blandinger av disse). Reak-sjonsoppløshingen må i et hvert tilfelle innstilles surt, fortrinnsvis ved hjelp av mineralsyrer som saltsyre, bromhydrogen-syre, svovelsyre, fosforsyre. Imidlertid kan det hertil også anvendes organiske syrer og sure ioneutvekslere. Som organiske syrer kommer det eksempelvis på tale: mettede og enkelte umettede C^-C^-alifatiske mono- og dikarboksylsyrer som maursyre, eddiksyre, propionsyre, oksalsyre, maleinsyre, fumarsyre, aromatiske kaeboksylsyrer som benzosyre, C-^-C^-alkyl-benzosyrer eller alifatiske respektive aromatiske sulfonsyrer som metansulfonsyre, etansulfonsyre, benzensulfon-syre, toluensulfonsyre ( eksempelvis p-toluensulfonsyre).
Det kan også anvendes blandinger av disse syrer. Som sure ioneutvekslere kommer det eksempelvis i betraktning slike som er nevnt i boken av K. Dorfner, Ionenaustauscher, Verlag De gruyter, 3. opplag, 19 70, bind 1 av verket Ionenaustauscher av E. Helfferich, Verlag Chemie, 19 59 eller i Rompps Chemie Lexikon bind 3, side 1616-1618 (1976). I detalj dreier det seg eksempelvis om organiske ioneutvekslere hvis aktive, dvs. ionedannende gruppe er karboksylgruppen eller en organiske sur gruppe som eksempelvis sulfonsyregruppen (SO^H) eller fosforsyregruppen, mens den høymolekylære struktur (matrix) består av kunstharpiks (kcrylharpiks, polystyrenharpiks, syren-divinylbenzen-kopolymerisat, styren-acrylsyre-kopolymerisat, divinylbenzen-polymerisat, vinylbenzen-polymerisat, kondensasjonsprodukter av fenol og formaldehyd). Videre kommer det i betraktning: sure cellulose-, stivelse- og dekstran-ioneutvekslere, karbon-ioneutvekslere (av sulfonert høymolekylær humuskull), samt sure uorganiske ioneutvekslere som zeolitter og aliminiumsilikater.
Reaksjonsoppløsningens, respektiv reaksjonsblandingens pH-verdi skal ligge mellom 1 og 6, fortrinnsvis 2 og 3.
Utgangsstoffer med formel II kan eksempelvis fåes ved at
et fenyletylamin med den generelle formel
omsettes med formaldehyd som også kan anvendes i form av et vanlig formaldehydleverende stoff. Denne omsetning foregår
i et oppløsnings- eller suspensjonsmiddel ved temperaturer mellom 20 og 180°C, spesielt mellom 30°C og 150°C. Som opp-løsnings- respektiv suspensjonsmiddel kommer det i betraktning: vann, lavere mettet alifatiske C-^-C^-alkoholer, lavere alifatiske mettede etere med akrylrester av 1 - 5 C-atomer, aromatiske hydrokarboner som benzen, metylbenzener (toluen), dimetylbenzener (xylen). Vanligvis arbeider man ved det anvendte oppløsningsmiddels koketemperatur. Det ved reaksjonen dannede vann kan eksempelvis fjernes ved tilsetning av tørke-midler som natriumsulfat, kaliumkarbonat, kaliumklorid, molekylarsikter (eksempelvis type 4A), eller ved azeotrop destillering (eksempelvis ved anvendelse av aromatiske hydrokarboner som oppløsningsmiddel). Fremgangsmåten må gjennom-føres under utelukkelse av syrer, da fremgangsmåteproduktene er syrelabile.
Vanligvis anvender man pr. 1 mol fenyletylamin med formel
IV 1,5 - 5 mol formaldehyd. Fortrinnsvis anvender man 3 mol formaldehyd respektiv ekvivalten av et formaldehydleverende stoff pr. 2 mol fenyletylamin med formel IV. Som formaldehydleverende stoffer kommer det eksempelvis på take: polyformal-dehyd, paraformaldehyd. Ved anvendelse av formaldehydleverende stoffer, hvor formaldehydet frigjøres i surt medium, bør formaldehydfrigjøringen forankoples den egentlige reaksjon og overskytende syre forsiktig nøytraliseres for at de ønskede dimere oksazolidiner ikke utsettes for surt miljø.
Eksempelvis kan N,N<1->metylen-bis(4-metyl-5-fenylroksazolidin) fåes som følger: 400 g (2,6 mol)£-norefedrin-base oppløses i 800 ml vann, varmt, og tilsettes dråpevist 330 g 35 %ig vandig formalinoppløsning. Reaksjonsblandingen oppvarmes 4 timer i kokende vannbad. Deretter ekstraheres flere ganger med metylenklorid, tørkes med I^CO^ og oppløsningsmiddelet fjernes i vann. Det faste residue omkrystalliseres fra diisopropyleter.
Utbytte 423 g (94,5%), smeltepunkt 98 - 99°C.
Fremstilling av denne forbindelse kan også foregå på følgende modifiserte måte: 10 g (0,066 mol)£-norefedrin-base, 4 g (0,132 mol) paraformaldehyd, 6,6 g vannfri natriumsulfat suspenderes i 100 ml tørr xylen og oppvarmes under omrøring i 5 timer under tilbakeløp. De uorganiske bestanddeler frafUtreres,oppløsningsmiddelet inndampes i vakuum og det faste råprodukt omkrystalliseres fra et isopropyleter.
Utbytte 6,1 g = 55%, smeltepunkt 98 - 99°C.
10 g (0,066 mol)<£->norefedrin-base, 4 g (0,132^01) paraformaldehyd suspenderes i 150 ml xylen og oppvarmes i vann-utskiller i 5 timer under tilbakeløp. Oppløsningsmiddelet inndampes i vakuum og det faste residue omkrystalliseres fra diisopropyleter.
Utbytte 7,6 g = 68%, smeltepunkt 100°C.
Utgangsstoffet med formel II, hvori R, betyr en hydroksygruppe, kan inneholde en vanlig beskyttelsesgruppe som avspaltes etter omsetningen. Det dreier seg herved spesielt om rester som er lett avspaltbare ved hydrolyse i ikke surt medium og eventuelt allerede avspaltes under reaksjonen.
Hvis slike beskyttelsesgrupper ikke avspaltes ved fremgangs-måtereaksjonen, foregår en avspalting etter reaksjonen. Ofte inneholder utgangsforbindelsene pga. deres fremstilling allerede slike beskyttelsesgrupper. Ved disse beskyttelsesgrupper dreier det seg eksempelvis om lett solvolyttisk avspaltbare acylgrupper. De solvolyttiske avspaltbare beskyttelsesgrupper avspaltes eksempelvis ved forsåpning ved hjelp av basiske stoffer (pottaske, soda, vandig alkalioppløsninger, alkoholi-ske alkalioppløsninger, vandig NH^) ved temperaturer mellom 10 og 150°C, spesielt 20 - 100°C. Som oppløsning- respektiv suspensjonsmiddel hertil kommer det eksempelvis i betraktning: vann, lavere alifatiske alkoholer, cykliske etere som dioksan eller tetrahydrofuran, alifatiske etere, dimetylformamid og så videre, samt blandinger av disse mideler. Eksempler for hydrolyttiske avspaltbare rester er: trifluoracetylrest, ptalylrest, tritylrest, p-toluen-sulfonylrest og lignende samt lavere alkanoylrester som acetylrest, formylrest, tert.-butyloksykarbonylrest og lignende. Også karbalkoksygruppen (eksempelvis lavmolekylær) kommer på tale.
Den som eventualitetsreaksjon tilknyttede reduksjon av ketogruppen av forbindelsen hvori X betyr gruppen ) CO til forbindelse hvori X er gruppen ^CHOH, samt reduksjon av en dobbelbinding av resten R, gjennomføres vanligvis ved hjelp av katallytisk hydrogenering. Som katalysatorer kommer det eksempelvis på tale de vanlige finfordelte metallkatalysator-er som edelmetallkatalysatorer, eksempelvis Raney-nikkel, platina eller spesielt palladium. Fremgangsmåten kan gjennom-føres ved normal temperaturer eller forhøyede temperaturer. Hensiktsmessig arbeider man i et temperaturområde på ca. 40 til 200°C, eventuelt under forhøyet trykk (1 - 100, spesielt 1-50 bar). Inneholder den fenoliske hydroksyl-gruppe benzylbeskyttelsesgruppen, så avspaltes denne sam-tidig ved den katallytiske hydrogenering når eksempelvis det anvendes en palladiumkatalysator.
Reduksjonen av ketogruppen er imidlertid likeledes mulig på annen måte, eksempelvis ved hjelp av komplekse metallhydrider (eksempelvis litiumaluminiumhydrid, natriumborhydrid, karbo-borhydrid, litium-tri-tert.-butoksy-aluminiumhydrid) eller ved hjelp av aluminiumalkoholater ifølge Meerwein og Ponndorf (eksempelvis ved hjelp av aluminiumisopropylat) ved temperaturer mellom 0 og 150°C, spesielt 20 og 100°C. Som opp-løsnings- respektiv suspensjonsmiddel for denne reaksjon kommer det eksempelvis i betraktning: lavere alifatiske alkoholer, dioksan, tetrahydrofuran, vann eller aromatiske hydrokarboner som benzen, toluen samt blandinger av disse.
En selektiv reduksjon av en dobbeltbinding av resen R^ er eksempelvis mulig under skånende betingelser ved hydrogen-eringen i nærvær av edelmetallkatalysatorer (Pd, Pt) eller Raney-nikkel.
Det anvendte utgangsamin med formel IV kan tilhøre den diastereomere rekke av efedrin (erytro-rekken) eller pseudoefe-drin. Det kan såvel anvende de rene enantiomere som også de tilsvarende racemater. Følgelig fåes bis-oksazolidinet med formel II som enten tilhører erytro-rekken eller treo-rekken. Bis-oksazolidiner av de to rekker adskiller seg i stillingen av CH^-gruppen og fenylringen. Stillingen av de to substituenter fremgår av de to nedenforstående stereo-formler:
Alt etter om man går ut fra tilsvarende rene enantiomere eller racemater av utgangsstoffene IV får man bis-oksazolidinene med formel II likeledes i form av rene enantiomere eller racematene. Ved spaltningen av bis-oksazolidinene under sure betingelser, berøres ikke av symmetrisentrene således at dette foregår under bibehold av den eventuelt foreliggende konfigurasjon.
De fremgangsmåteprodukter med formel I såvel også utgangs-bis-oksazolidinene med formel II som fremkommer som racemater, kan spaltes på i og for seg kjente måte, eksempelvis ved hjelp av en optisk aktiv syre under skånende betingelser ved lave temperaturer i de optisk aktive isomerer. Det er selvsagt også mulig fra begynnelsen å anvende optisk aktive respektiv også diastereomere utgangsstoffer, idet da som sluttprodukt I fåes en tilsvarende ren optisk aktiv form, respektiv diastereomer konfigurasjon. Eksempelvis dreier det seg om forbindelser av norefedrin-konfigurasjonen (erytro-rekken) og pseudonorefedrin-konfigurasjonen (treo-rekken).
Det kan også opptre diastereomere racemater, da i de fremstile forbindelser to eller flere assymetriske karbonatomer er til-stede. Adskillelsen er mulig på vanlig måte, eksempelvis som krystallisering.
Alt etter fremgangsmåtebetingelser- og utgangsstoffer får
man sluttstoffene med formel I i fri form eller i form av deres salter. Sluttstoffenes salter kan på i og for seg kjent måte, eksempelvis med alkali eller ioneutvekslere igjen overføres i basene. Av sistnevnte lar det seg ved omsetning med organiske eller uorganiske syrer, spesielt slike som er regnet til dannelse av terapeutiske anvend-
bare salter, fremstilles salter. Som slike syrer skal eksempelvis nevnes: halogenhydrogensyrer, svovelsyre, fosforsyrer, salpetersyre, perklorsyre, organiske mono-, di- eller ti-karboksylsyrer fra den alifatiske, alicykliske, aromatiske eller hetrocykliske rekke samt sulfonsyrer. Eksempelet her-på er maursyre, eddiksyre, propiosyre, ravsyre, glykolsyre, melkesyre, eplesyre, vinsyre, sitronsyre, ascorbinsyre, maleinsyre, fumarsyre, hydroksymaleinsyre eller druesyre, fenyl-eddiksyre, benzosyre, p-amino-benzosyre, antraninsyre, p-hydroksy-benzosyre, salisylsyre eller p-amino-salisylsyre, embonsyre, metansulfonsyre, etansulfonsyre, hydroksy-etansulfonsyre, etylensulfonsyre, halogenbenzensulfonsyre, toluensulfonsyre, naftalinsulfonsyre eller sulfanilsyre eller også 8-klor-teofyllin.
Eksempel 1
Reaksjonsblandingen av 16,92 g (0,05 mol)£- N,N<1->metylen-bis-(4-metyl-5-fenyl-oksazolidin) (fremstilt fra£-norefedrin), 15,14 g (0,12 mol) acetylcykloheksan, 35 ml isopropanol og 19 ml 5,ln isopropanolisk saltsyre oppvarmes 6 timer under omrøring og tilbakeløp. Reaksjonsblandingen hensettes natten over ved værelsetemperatur. Det utkrystalliserte produkt frasuges og vaskes med 5 ml isopropanol og 20 ml aceton. Man får eksempelvis 32,58 g (67% av det teoretiske) £-[3-hydroksy-3-fenyl-propyl-(2)] [3-cykloheksyl-3-okso-propyl]-amin-hydroklorid med smaltepunkt 219 - 221°C
Claims (2)
1. Fremgangsmåte til fremstilling av forbindelser med den generelle formel
hvori X betyr gruppen CO eller CH (OH) , R,, betyr hydrogen eller en C-^-Cg-alkylgruppe og R^ betyr hydrogen eller en hydroksygruppe og R^ betyr adamantylresten eller en mettet eller enkelt umettet C^-C^g-cykloalkylrest, idet denne C^-C^g-cykloalkylrest også kan være subsituert med C^-C^-alkyl-gruppe eller et halogenatom, samt deres syreaddisjonssalter, karakterisert ved at et N,N'-metylen-bis-oksazolidin med formel II
hvori R^ har den angitte betydning, omsettes med et keton med formel III
hvori R^ og R^ har overnevnte betydning i nærvær av en syre eller sur ioneutveksler og eventuelt reduserer de dannede forbindelser med formel I, hvori R, inneholder en dobbeltbinding og/eller X betyr CO-gruppen, eventuelt denne dobbeltbinding og/eller denne CO-gruppe.
2. Fremgangsmåte ifølge krav 1 til fremstilling av forbindelse med den generelle formel I, hvori X har den i krav 1 angitte betydning, og R2 og R^ betyr hydrogen, og R^ betyr en mettet eller enkelt umettet Cg-Cg-cykloalkylrest, karakterisert ved at det anvendes et N,N'-metylen-bis-oksazolidin med formel II og et keton med formel III, hvor R^ R2 og R^ har den ovenfor angitte betydning .
