Forelaggende oppfinnelse vedrører en ny fremgangsmåte ved ;frems.tilling av 3.,,;3'-azo-bi-s-i(6-hydroksybenzosyre) med -formel og .salter derav. 'Den :nye :fremgangsmåte ifølge oppfinnelsen gjør :det mulig -å 'fremstille -forbindelsen med -formel I og .dens salter., cspesielt farmasøytisk aksepterbare ;salter, med meget høy -renhet., f..eks. 9B .% .éller ?mer-, -og i gode utbytter.
Forbindelsen med formel .1 -er :kj.erit 1 -seg :selv og -har funnet kommersiell anvendelse som .fargestoff.. En kjent :fremgang.s-måte for fremstilling av forbindelsen I .er .at 3,3'-azo-6-klor-6'-hydroksy-dibenzosyre hydrolyseres alkalisk ved høy temperatur, eventuelt i nærvær av kopper., se :f .eks., -tysk patent 27:86.13.. 'Ved .en .alternativ kjent f-remstilling.smetode fremstilles foreliggende forbindelse v.ed en tilsvarende alkalisk hydrolyse av 3 , 3" -azo-iiis- (6-klor.benzo.syre)., se f .eks. britisk patent 408.6 7;6.. Ved en tredje kjent fremstillingsmetode diazoteres :5-aminosalicylsyre og kobles med sal icyl syre i nærvær .av 3-verdig krom., hvoretter det dannede kromkomplekse.t spaltes .for frigjøring av forbindelse I., se f.eks. tysk -patent 590321.
Den førstnevnte fremstillingsmetode er den som anvendes
kommersielt. Det har vist seg at de produkter som erholdes ved denne hydrolyse av 3,3<1->azo-6-klor-6'-hydroksy-dibenzo-syre ikke er rene, men inneholder forholdsvis store mengder utgangsmateriale (f.eks. 20 - 30 %). Videre er det ønskede sluttprodukt I ikke stabilt ved de hårde hydrolysebetingelsene, men spaltes, bl.a. til 5-aminosalicylsyre.
Det ville i og for seg være teoretisk mulig å adskille sluttproduktet I fra utgangsmaterialet, f.eks. ved anvendelse av væskekromatografisk analyseteknikk. Slike separasjonsmetod-er er imidlertid økonomisk umulige å tillempe for praktisk preparativt bruk. På grunn.av de meget likeartede egenskap-ene hos utgangs- og sluttforbindelsene har det heller ikke vært mulig å adskille disse ved krystallisering.
Den alternative syntesevei via kromkomplekset av forbindelse I gir heller ikke noe rent produkt, ettersom forurensninger av krom alltid forekommer. På grunn av slike forurensninger kan forbindelse I som fremstilles på denne måte f.eks. ikke anvendes som legemiddel.
Hensikten med forliggende oppfinnelse er å tilveiebringe en teknisk enkel og økonomisk lønnsom ny fremgangsmåte for fremstilling av 3,3'-azo-(bis-6-hydroksy-benzosyre) med formel I, hvorved man får det ønskede produkt i betydelig høyere renhet enn ved kjente fremstillingsmetoder. Gjennom oppfinnelsen blir det fremfor alt mulig å fremstille forbindelse I med en renhet på minst 98 %, hvorigjennom det blir mulig å anvende foreliggende forbindelse som legemiddel, f.eks. for behandling av inflammatoriske tarmsykdommer, f.eks. ulcerøs kolitt, hvorunder forbindelsen ved oral administrering trans-porteres intakt til tykktarmen, hvor den reduseres til lok-alt virkende 5-aminosalicylsyre.
For dette formål foreslås ifølge oppfinnelsen en flertrinns-metode for fremstilling av forbindelsen med formel I og salter derav. Ved fremgangsmåten ifølge oppfinnelsen går man ut fra forbindelser med formel II
hvor betyr en lavere alkylgruppe
, og R 9 er en lavere alkylgruppe
, eller fenyl eller substituert fenyl. Foretrukne lavere alkylgrupper er sådanne med 1-3 karbonatom-er som metyl. En fenylgruppe R_ kan være mono- eller poly-substituert med grupper som er inerte under reaksjonsbeting-elsene så som alkyl, alkoksy, halogen, etc. Som utgangsmateriale kan man også anvende syreaddisjonssalter av forbindelse II, spesielt salter med en mineralsyre eller en sterk organisk syre.
Utgangsforbindelsen med formel II diazoteres på i og for seg kjent måte til tilsvarende diazoniumsalt. Diazoteringen skjer fortrinnsvis på vanlig måte ved behandling med sal-petersyrling i sur vannløsning.
Det dannede diazoniumsaltet av forbindelse II koples deretter i alkalisk løsning, fortrinnsvis i nærvær av et alkalimetallhydroksyd så som kaliumhydroksyd, med en forbindelse med formel III
hvor betyr hydrogen eller en lavere alkylgruppe Det herved dannede produkt med formel IV
hvor R-^, R2 og R har ^e ovenfor angitte betydninger,
isoleres så på i og for seg kjent måte, f.eks. ved surgjør-ing av reaksjonsblandingen, slik at produktet med formel IV faller ut. Det har vist seg at forbindelsene med formel IV har utmerkede krystallisasjonsegenskaper, slik at
disse forbindelser renses ved omkrystallisering. For omkrystallisering kan man anvende et stort antall inerte organiske løsningsmidler, f.eks. lavere alifatiske ketoner så som aceton, 2-butanon, etc, lavere alkanoler, så som metanoletanol, etc, aromatiske hydro-karboner så som toluen, etc, organiske syrer og estere så som eddiksyre, etylacetat, etc, eller blandinger derav.
Forbindelsen med formel IV som er renset ved omkrystallisering, hydrolyseres deretter i et vannholdig reaksjons-medium til det ønskede produktet med formel I under avspalt-ning av gruppene -R-j.' -R3 og -SO2R2' hvoretter sluttproduktet med formel I - eller salter derav - isoleres fra reaksjonsblandingen. Hydrolysen utføres fortrinnsvis i alkalisk miljø, spesielt i nærvær av et alkalimetallhydroksyd og eventuelt et inert vannblandbart løsningsmiddel, f.eks. alkanoner så som aceton, alkanoler så som metanol, etanol, etc. For en fagmann er det klart at hydrolysebetingelsene kan velges på slik måte at mengdene partielt hydrolyserte produkter blir så små som mulig, samtidig som ingen nevne-verdige mengder sekundære biprodukter dannes. Som eksempel kan nevnes at hydrolysen med fordel kan utføres under koking i vann mellom 0,5 og 2 timer ved anvendelse av natriumhydroksyd som base. I motsetning til mellomproduktet IV er det som ovenfor nevnt, forholdsvis vanskelig å rense sluttproduktet I, og det er derfor viktig at hydrolysen utføres på angitt måte, særskilt når sluttproduktet I skal anvendes som legemiddel.
Etter å ha utført hydrolysen isoleres sluttproduktet fra reaksjonsblandingen på i og for seg kjent måte, fortrinnsvis ved nøytralisering eller surgjøring av reaksjonsblandingen, f.eks. med en mineralsyre så som saltsyre eller svovelsyre, eller en sterk organisk syre så som eddiksyre.
Forbindelsen med formel I kan isoleres som sådan eller i form av et tilsvarende salt, fortrinnsvis et farmasøytisk akseptabelt salt så som et alkalimetallsalt. En fortrukket måte å utvinne slike salter på er å partielt nøytralisere reaksjonsblandingen med en syre til passende pH for utfell-ing av det ønskede salt. F.eks. kan man fremstille dinatrium- og dikaliumsaltene ved å surgjøre reaksjonsblandingen til pH 5-8, fortrinnsvis 6-7, med en mineralsyre eller en organisk syre så som eddiksyre. Alternativt kan man eventuelt også fremstille ønskede salter ved på i og for seg kjent måte og omsette den isolerte syre med en passende saltdannende base.
De utgangsforbindelser som anvendes ved fremgangsmåten iføl-ge oppfinnelsen med formel II kan fremstilles ved i og for seg kjente metoder eller analogt med i og for seg kjente metoder. F.eks. kan forbindelsene fremstilles ved reduk-sjon av en tilsvarende forbindelse med formel V hvor R^ og R^ har de ovenfor angitte betydninger og X er en gruppe som kan overføres til en aminogruppe, f.eks. en nitro- eller en arylazogruppe.
Selv om fremgangsmåten ifølge oppfinnelsen fremfor alt er egnet for fremstilling av 3,3 '-azo-bis(6-hydroksy-benzosyre) med farmasøytisk akseptabel renhet, er det også klart at de kan utnyttes for å fremstille denne forbindelsen for andre formål hvor renhetskravene ikke er like høye.
Oppfinnelsen skal nå illustreres nærmere i tilknytning til noen spesielle utførelseséksempler som ikke begrenser oppfinnelsen.
EKSEMPEL 1
(Fremstilling av utgangsmaterialet)
a) Metyl- 2- metansulfonyloksy- 5- nitro- benzoat.
98,5 g metyl-2-hydroksy-5-nitrobenzoat ble oppløst i 250 ml
pyridin, hvoretter 68,5 g metansulfonylklorid ble tilsatt. Etter oppvarming til 50°C i 10 min. ble løsningen helt på 5N iskald saltsyre. Den dannede olje ble ekstrahert med kloroform. Kloroformfasen ble vasket med vann, tørket og inndampet. 50 ml toluen ble tilsatt og inndampet. Dette ble gjentatt en gang, hvorunder oljen krystalliserte. Utbytte: 132 g av tittelsubstansen.
b) Metyl- 3- amino- 6- metansulfonyloksy- benzoat.
132 g av nitroforbindelsen fra trinn a) ble hydrogenert i
500 ml eddiksyre og 3 g 10 % palladium-på-karbon ved 1-3 atmosfærer. Etter at den teoretiske mengde hydrogen var forbrukt, ble blandingen filtrert og inndampet. Den ble oppløst ill isopropanol ved 65 oC, hvoretter HC1 i etanol ble tilsatt. Hydrokloridet krystalliserte ved avkjøling. Etter filtrering ble substansen rystet med natriumkarbonat
i vann og kloroform. Kloroformfasen ble inndampet. Aminet ble omkrystallisert fra metanol/vann. Utbytte: 93 g av den rene tittelsubstansen.
EKSEMPEL 2
a) Metyl- 2- hydroksy- 5-[( 4- metansulfonyloksy- 3- metoksykarbon-ylfenyl)- azo]- benzoat.
12,2 g metyl-3-amino-6-metansulfonyloksy-benzoat (fremstilt ifølge eksempel 1), 75 ml 2N saltsyre og 200 g is ble diazo-tert ved 0°C med 3,5 g natriumnitritt i 20 ml vann. Diazon-iumsaltløsningen ble blandet raskt med en nylaget løsning av 15,2 g metylsalicylat, 12 g 85 % kaliumhydroksyd og 1 1 is-vann under kraftig omrøring. Koplingsløsningen ble surgjort etter ca. 15 sek. med saltsyre. Det utfelte oljeaktige produkt ble ekstrahert med kloroform. Etter tørking, inndamp-ing og omkrystallisering fra 2-butanon fikk man tittelsubstansen i et utbytte på 12,8 g (63 %). Renheten ble bestemt ved væskekromatografisk analyse (HPLC) på en "reversed
phase"-kolonne (u-Bondapak C^g) med metanol/vann (75:25)
som elueringsmiddel. Total mengde forurensninger var bare ca. 0,6 flateprosent.
b.) 3, 3' - azo- bis ( 6- hydroksy- benzosyre) .
41 g metyl-2-hydroksy-5-[(4-metansulfonyloksy-3-metoksykar-bonylfenyl)-azo]benzoat fremstilt ifølge trinn a), ble hydro-lysert ved koking i 800 ml IN natriumhydroksyd i 4 timer. Det hydrolyserte produktet ble utfelt ved tilsetning av kons-entrert saltsyre til pH ca. 2. Fellingen ble filtrert og vasket med vann. Utbytte: 29,7 g (98 %) av tittelsubstansen.
EKSEMPEL 3
Dinatrium- 3, 3'- azo- bis( 6- hydroksy- benzoat).
624 g metyl-2-hydroksy-5-[(4-metansulfonyloksy-3-metoksykar-bonylfenyl)-azo]benzoat, fremstilt ifølge trinn a) i eksempel 2, ble porsjonsvis i løpet av 15 min. satt til 3 1 vann . og 428 g natriumhydroksyd under koking. Etter koking i 30 min. tilsattes 367 ml eddiksyre i løpet av 15 min. til pH 6. Suspensjonen ble omrørt i 1,5 timer uten kjøling, hvorunder temperaturen sank til 70°C. Etter kjøling til 30°C ble suspensjonen filtrert og vasket med 700 ml metanol. Etter tørking fikk man tittelsubstansen i et utbytte på 520 g (98,5 %). Renheten var>99,0 % ifølge ionepar-væskekromatografisk analyse på "reversed phase"-kolonne (u-Bondapak C^g) og med metanol/vann (57:43) som elueringsmiddel og tetra-butylammonium som motion.
Claims (3)
1. Fremgangsmåte ved fremstilling av 3 , 3'-azo-bis-(6-hydroksybenzosyre) med formel I
og salter derav,
karakterisert ved at man på i og for seg kjent måte diazoterer en forbindelse med formel II eller et salt derav:
hvor er lavere alkyl og R2 er lavere alkyl eller eventuelt substituert fenyl,
kopler det erholdte diazoniumsalt i alkalisk vannløsning til en forbindelse med formel III
hvor R3 er hydrogen eller lavere alkyl,
under dannelse av en forbindelse med formel IV
_ hvor R^, R2 og R^ har de ovenfor angitte betydninger, og at man isolerer og renser forbindelsen med formel IV ved omkrystallisering fra et organisk løsningsmiddel, og deretter alkalisk hydrolyserer den, og på i og for seg kjent måte isolerer derav den erholdte forbindelsen med formel I eller et salt derav.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at koplingen av diazoniumsaltet av forbindelsen med formel II til forbindelsen med formel III foretas i nærvær av kaliumhydroksyd.
3. Fremgangsmåte ifølge kravene 1-2 for fremstilling av dinatriumsaltet av forbindelsen med formel I, karak-terisert ved at man hydrolyserer forbindelsen med formelen IV med natriumhydroksyd og feller dinatriumsaltet ved nøytralisering til pH 5-8.
