hvori R 1 betyr hydrogen eller en hydroksygruppe, R2 betyr en hydroksygruppe, en hydroksymetylgruppe eller en C-j-C^-alkylgruppe, R^ betyr hydrogen eller en -C^-alkylgruppe og Alk betyr en rettlinjet eller forgrenet C2-Ck~alkylen-
gruppe.
Disse forbindelser har en bronkolytisk virkning
samt en coronarutvidende og positiv inotrop virkning.
Oppfinnelsen vedrører analogifremgangsmåte til-fremstilling av terapeutisk aktive forbindelser med den generelle formel
hvor Alk betyr en C2-C^-alkylengruppe,
Ar betyr fenyl som er substituert med en Cj-C^-alkyl-, <C1>"<C>^-alkoksy-, C2_C^-alkenoksy- eller -C^-alkynoksy-
gruppe, eller et halogenatom, eller Ar betyr en naftyl-
eller en eventuelt med en -C^-alkylgruppe substituert indolylgruppe, samt deres salter, idet fremgangsmåten er karakterisert ved at et amin med formel
Ar-0-CH2-CH(0H)-CH2-NH-Alk-NH2 (il)
hvori Ar og Alk har den angitte betydning, omsettes med en forbindelse med formel
hvori E betyr en C<->,-Cb-alkylmerkaptogruppe eller C.,-^-alkoksygruppe, og hvis ønsket overføres forbindelsene til
deres salter.
Forbindelsene fremstilt ifølge oppfinnelsen
er farmakodynamisk virksomme. Hovedvirkningen består i en blokkering av 3-reseptorene av det adrenergiske nerve-
system (isoprenalinsk kretsløpseffekt hemmes). Dessuten virker de blodtrykksenkende.
Forbindelsene ifølge oppfinnelsen har en god terapeutisk bredde og utmerker seg ved små bivirkninger.
Ved forbindelsene fremstilt ifølge oppfinnelsen med formel I består alkylengruppen Alk fortrinnsvis av 2,3 eller 4 karbonatomer (etylengruppen, propylengruppen, butylengruppen).
Hvis gruppen Alk er forgrenet inneholder den spesielt 3,4- eller 5 C-atomer (eksempelvis: -CHg-CHg-CH (CH^ ) -,
-CH2-CH(CH3)-, -CH2-CH(CH3)-CH2-, -CH2~CH (CH^ )-CH (CH3 ),\
Substituentene av resten Ar har spesielt følgende betydning: Alkyl med 1-4- C-atomer, eksempelvis metyl, etyl, propyl, isopropyl, tert.-butyl, butyl.
alkoksy med 1-6 C-atomer, eksempelvis metoksy, etoksy, propoksy, isopropyloksy, butoksy, tert.-butoksy.
alkenyloksy og alkinyloksy med 2-5 C-atomer, eksempelvis allyloksy, metallyloksy, propargyl-0 k s y .
halogen: fortrinnsvis fluor, brom eller klor;
Substituentene i Ar kan være like eller for-skjellige idet restene Ar kan inneholde flere av de nevnte substituenter, fortrinnsvis en, to eller også tre av de nevnte substituenter. Fsnylresten er spesielt substituert 1 orto- og/eller para-stilling.
Innen rammen av oppfinnelsen forstås med forbindelser med den generelle formel I også de mulige stereo-isomere og optisk aktive forbindel ser og blandinger herav, spesielt racematene. Blandinger av diastereoisomere kan på kjent måte adskille, eksempelvis ved fraksjonert krystallisasjon. Optisk aktive forbindelser kan fåes etter vanlige metoder, eksempelvis ved omkrystallisering av salter av de racemiske baser med formel I med optisk aktive syrer eller eventuelt ved anvendelse av optisk aktive utgangspro-dukter ved syntesen.
Alt etter fremgangsmåtebetingelser og utgangsstoffer får man sluttstoffene med formel I i fri form eller i form av deres salter. Saltene av slutt sto ffene kan på i og for seg kjent måte, eksempelvis med alkali eller ione-utvekslere igjen overføres i basene. Av sistnevnte lar det seg ved omsetning med organiske eller uorganiske syrer spesielt slike som er egnet til dannelse av terapeutisk anvendbare salter, danne salter. Som slike syrer skal eksempelvis nevnes: halogenhydrogensyrer, svovelsyrer, fos-forsyre, salpetersyre, perklorsyre, organiske mono-, di-eller trikarboksylsyrer av den alifatiske, alicykliske, aromatiske eller heterocykliske rekke, samt sulfonsyrer. Eksempler herpå er: maursyre, eddiksyre, propionsyre, rav-syre, glykolsyre, melkesyre, eplesyre, vinsyre, sitronsyre, ascorbinsyre, maleinsyre, fumarsyre, hydroksymaleinsyre-ell er pyrodruesyre; fenyleddiksyre, benzosyre, p-amino-benzo -
syre, antranilsyre, p-hydroksy-benzosyre, salicylsyre eller p-amino-salicylsyre. Embonsyre, metansulfonsyre, etansul-fonsyre, hydroksyetansulfonsyre, etylensulfonsyre, halogen-benzensulfonsyre, toluensulfonsyre, naftalinsulfonsyre eller sulfanilsyre eller også 8-klor-teofyllin.
Ved de videre nedenfor angitte fremgangsmåter til fremstilling av forbindelsen ifølge oppfinnelsen kan de i reaksjon tredende aminogrupper inneholde kjente og vanlige beskyttelsesgrupper, Det dreier seg derved om rester som er lett avspaltbare ved hydrolyse eller hydrogenolyse og ofte allerede avspaltes under reaksjonen. Hvis slike beskyttelsesgrupper ikke avspaltes ved fremgangsmåtereak-sjonen, foregår en avspaltning etter reaksjonen. Ofte inneholder utgangsforbindel sene allerede slike beskyttelsesgrupper på grunn av deres fremstilling.
Ved disse beskyttelsesgrupper dreier det seg eksempelvis om lett solvolytisk avspaltbare acylgrupper eller hydrerende avspaltbare grupper. De solvolytisk avspaltbare beskyttelsesgrupper avspaltes eksempelvis ved forsåpning med fortynnede syrer eller ved hjelp av basiske stoffer (pottaske, soda, vandige alkalioppløsninger, alkoholiske alkalioppløsninger, NH^) ved temperaturer mellom 10 og 150°C, spesielt 20-100°C. Hydrerende avspaltbare grupper som a-arylalkylrester (benzylrest) eller hydroksy-karbonylrest er (karbobenzoksyrest) avspaltes hensiktsmessig med katalytisk hydrogenering i nærvær av vanlige hydro-generingskatalysatorer, spesielt palladium-katalysatorer, platinoksyd eller også Raney-nikkel, i et oppløsnings- eller suspensjonsmiddel, eventuelt under forhøyet trykk ved temperaturer mellom 20 og 100°C, spesielt 40-80°C. Som oppløsnings- resp. suspensjonsmiddel kommer det eksempelvis i betraktning: vann, lavere alifatiske alkoholer, cykliske etere som dioksan, eller tetrahydrofuran, alifatiske etere, dimetylformamid og så videre samt blandinger av disse midler.
Som beskyttelsesgrupper som er avspaltbare ved hydrogenolyse kommer det eksempelvis på tale: benzylrest, a-fenyletylrest, i benzenkjernen substituert benzylrester (p-brom- eller p-nitrobenzylrest), karbobenzoksyrest, kar-bobenztiorest, tert.-butylhydroksykarbonylrest. Eksempler for hydrolytisk avspaltbare rester er: trifluoracetylrest, ftalylrest, tritylrest, p-toluensulfonylrest og lignende, samt lavere alkanoylrester som acetylrest, formylrest, tert.-butylkarboksyrest og lignende.
Spesielt kommer det på tale de ved peptid-syntesen vanlige beskyttelsesgrupper og de der vanlige av-spaltningsfremgangsmåter. Blant annet henvises her også
til boken av Jesse P..Greenstein og Milton Winitz, Chemis-
try og Amino Acids", N.Y. 1961, John Waley and Sons, Inc. Volum 2, eksempelvis side 883 ff. Også- karbalkoksygruppen (eksempelvis lavmolekylære) kommer på tale.
Hvis det i utgangsstoffene dessuten foreligger alkoholiske hydroksygrupper og/eller primære aminogrupper, kan disse eventuelt være beskyttet ved ovennevnte beskyttelsesgrupper, idet avspaltningen foregår på samme måte.
Fremgangsmåten ifølge oppfinnelsen kan gjennom-føres i oppløsning eller i smelte ved temperaturer mellom 20 og 150UC, eventuelt under forhøyet trykk. Som oppløsnings-middel egner det seg eksempelvis vann eller organiske oppløsningsmidler som alkoholer, toluen, xylen, dioksan, alkohol-vannblandinger. Forbindelsene med formel III kan anvendes som frie baser eller også i form av vanlige syre-addisjonssalter (eksempelvis sulfater eller hydroklorider). Hvis i formel III E og gruppen "NHg sammen danner gruppen =NH, dreier det seg om karbonsyrediimid eller cyanamid.
Utgangsstoffet med formel II fåes eksempelvis
ved omsetning av forbindelsene med formel Ar-0-CH2_CH(0H-CHg-Hal (Hal= klor, , brom ell er jod) eller halogenhydriner
med formel
med et diamin H2N-Alk-NH2 på kjent måte, Fortrinnsvis anvendes diamin i overskudd. Ved usymmetrisk forgrenede di-aminer er det hensiktsmessig før omsetningen å beskytte aminogruppen som ikke skal reagere med de andre reaksjons-komponenter. Hertil har det eksempelvis vist seg egnet benzylgruppen, da den etter omsetningen kan fjernes på kjent måte under milde betingelser.
Forbindelsene fremstilt ifølge oppfinnelsen er egnet til fremstilling av farmasøytiske sammensetninger og tilberedninger. De farmasøytiske sammensetninger resp. legemidler inneholder som virksomt stoff en eller flere av forbindelsene fremstilt ifølge oppfinnelsen og eventuelt i blanding med andre farmakologiske resp. farmasøytisk virk-
somme stoffer. Fremstilling av legemidlene foregår på
kjent måte, idet de kjente og vanlige farmasøytiske hjelpestoffer kan anvendes, samt andre vanlige bære- og for-tynn ing smi dl er .
Som slike bære- og hjelpestoffer kommer det eksempelvis på tale slike stoffer som er anbefalt resp. angitt i følgende litteratursteder som hjelpestoffer for farmasi, kosmetikk og tilgrensende områder: Ullmanns Encyklopfldie der technischen Chemie, bind 4 (1 953), side 1 -39, Journal og Pharmaceutical Sciences, bind 52 (1963), side 918 ff, H.v. Czetsch-Lindenwald, Hilfsstoffe fflr Pharmazie und angrenzende Gebiete, Phar, Ind., hefte 2,
1961, dise 72 ff, Dr. H.P. Fiedler, Lexikon der Hilfsstoffe filr Pharmazie, Kosmetik und angrenzende Gebiete Cantor KG. Aulendorf i Wflrttemberg 1971.
Den farmakologiske og galeniske håndtering av forbindelsene fremsitlt ifølge oppfinnelsen foregår etter de vanlige standardmetoder. Eksempelvis sammenblandes godt virksomme stoffer eller virksomt stoff og hjelpe-resp. bærestoffer ved omrøring eller homogenisering (eksempelvis ved hjelp av kolloidmøller, kulemøller) idet vanligvis arbeides det i temperaturer mellom 20 og 80°C, fortrinnsvis 20-50°C.
Applikasjonen av de virksomme stoffer resp. legemidler kan foregå på huden eller slimhuden eller i det indre av kroppen, eksempelvis oralt, enteralt, pulmonalt, rektalt, nasalt, vaginalt, lingialt, intravenøst, intra-arterielt, intrakardialt, intramuskulært, intraperitonealt, intracutant, subcutant.
Spesielt er også tilsetningen av andre virksomme legemidler mulig resp. gunstig.
Forbindelsene fremstilt ifølge oppfinnelsen viser på våkne kaniner (metode etter Bartsch et al, 1974 "Arzneimittelforschung (Drug Res.) 24, 303 (1974)), ved en ved isoprenalin utløst tachycardi en god 3-adrenolytisk virkning. Således ble det eksempelvis på kaniner ved intra-venøs applikasjon funnet en ED^q på 0,007 mg/kg. På narkotisert hund senkes det arterielle trykk tydelig, idet det samtidig minskes på grunn av en senkning av hjertefrekvensen, hjertetidsvolumet, mens karmostanden, eksempel-
vis bare endres lite eller eventuelt senkes. Fastlegging av blodtrykk og hjertefrekvens foregår eksempelvis på narkotisert hund ved hjelp av et elektronisk manometer i forbindelse med en direkte skriver. Hjertefrekvensen fast-slås derved fra registreringskurven ved valg av enkelt-impulser pr. minutt. Hjertetidsvolumet bestemmes ved hjelp av kuldefortynningsmetoden ifølge H, Slama og J. Piiper, "Kreislaufforschung 53, 322, (1 964.) . Den perifere karmot-stand kan da beregnes av hjertevolum og midlere artielt blodtrykk på kjent måte. Eksempelvis ble det etter 1 mg/kg (intravenøst) hos hund iakttatt en senkning av den midlere areterielle blodtrykk rundt 35 % en nedsettelse av hjertefrekvensen rundt 13 % og en nedsettelse av hjertetidsvolumet rundt 10 % og en nedsettelse av den samlede mot-stand av de perifere kar rundt 27 %. Disse virkninger er sammenlignbare med disse av det kjente legemiddel Acebu-tolol.
Den laveste allerede tydelig blodtrykkssenkende dosis i det ovennevnte nye forsøk er eksempelvis: 0,03 mg/ kg pr. os, 0,003 mg/kg intravenøst.
Som generelt dosisområde for den blodtrykkssenkende 3-adrenolytiske virkning (dyreforsøk som ovenfor) kommer det eksempelvis på tale: 0,3 til 30 mg/kg pr. os, spesielt 1-10 mg/kg, 0,03 til 3 mg/kg intravenøst, spesielt 0,1-1 mg/kg.
Indikasjoner for forbindelsene fremstilt ifølge oppfinnelsen kommer på tale: essensielle og renale hyper-tonier, angina pectoris, kronisk coronarinsuffensiens, for-styrrelse i energiseringsdannelsen og energiseringsutbredning av hjertet.
De farmasøytiske tilberedninger inneholder vanligvis mellom 3 til 300 mg av de ifølge oppfinnelsen aktive komponenter.
Administreringen kan eksempelvis foregå i form av tabletter, kapsler, piller, drageer eller i flytende form; Som flytende anvendelsesformer kommer det eksempelvis på tale: oljeaktige eller alkoholiske resp. vandige opp-løsninger samt suspensjoner og emulsjoner. Foretrukne anvendelsesformer er tabletter som inneholder mellom 10 og 200 mg eller oppløsninger som inneholder mellom 0,5 til 5% aktivt stoff.
Enkeltdosene av de ifølge oppfinnelsen aktive komponenter kan eksempelvis ligge a) ved orale1 legemiddelformer mellom 10 og 4-00 mg, b) , ved parenterale legemiddelformer (eksempelvis intravenøst, intramuskulært) mellom 10 og 50 mg.
(Dosene er hver gang referert til den frie ba s e ) .
Eksempelvis kan det anbefales 3 ganger daglig 1-3 tabletter med et innhold fra 10-200 mg virksomt stoff, eller eksempelvis ved intravenøse injeksjoner 1-3 ganger daglig en ampulle fra 2-10 ml innhold med 10-50 mg stoff. Ved oral administrering er den minimale daglige dose eksempelvis 150 mg, den maksimale daglige dose ved oral administrering skal ikke ligger over 1 g.
For behandling av hunder og katter ligger den orale enkeltdose vanligvis mellom omtrent 1 og 3 mg/kg legemsvekt, den parenterale dose omtrent mellom 0,1 og 0,5 mg/ kg legemsvekt.
For behandling av hester og storfe ligger den orale enkeltdose vanligvis mellom omtrent 1 og 3 mg/kg, den parenterale dose omtrent mellom 0,3 og 1,0 mg/kg legemsvekt.
Den akutte toksisitet av forbindelsene ifølge oppfinnelsen på mus (uttrykt ved LD 50 mg/kg, metode ifølge Miller og Tainter: Pro c . So c . Ezper. Biol. a. Med. 57 (1 94-4-) 261) ligger eksempelvis ved oral applikasjon mellom 100 og 500 mg/kg.
Legemidlene kan anvendes i humanmedisinen, vete-rinærmedisinen samt i landbruk alene eller i blanding med andre farmakologisk aktive stoffer.
Eksempel 1
3-((3-/ naftyl- (1 ) - ok syl7-2-hydroksy-propylamino ))-propylguanidin
5,0 g 3-{(3-/"naftyl-(1 )-oksyl7-2-hydroksypropylamino ))-pro-pylamin oppløses i 50 ml etanol og 15 ml vann og denne opp-løsning kokes sammen med 5,75 g S-metyl-isotiourinstoffsulfat i 10 timer under omrøring og tilbakeløp. Man innstiller med 10 %- ig svovelsyre en pH-verdi på 4--5, frasuger det utfelte sulfat og vasker det med vann og aceton.
Utbytte: 6,5 g
Sulfatets sm.p. 290-293°C (under spaltning).
Til overføring i det bedre oppløselige oksalat utrøres med natronlut og kloroform, utrystes ennå tre ganger med kloroform de forenede kloroformekstrakter, tørkes og natriumsulfat og inndampes, og den alkoholiske oppløsning av residuet surgjøres med en alkoholisk oksalsyreoppløsning (pH 4--5). Det utkrystalliserte oksalat frasuges og omkrystalliseres fra vann.
Utbytte: 3, 7 g
Oksalatets sm.p. 220°C (under spaltning).
Utgangsproduktets fremstilling:
I 118 ml 1,3-diaminopropan dryppes under om-røring 11,8 g 3-/ naftyl-(1)-oksyl7"2-hydroksy-propylklorid. Man omrører i en time ved 60°C og avdestillerer overskytende
amin i vakuum. Residuet opptas i kloroform, utrystes tre ganger med vann, kloroformfasen tørkes over kaliumkarbonat, inndampes og den gjenblivende base oppløses i eter. Man surgjør med alkoholisk saltsyre, frasuger det dannede di-hydroklorid og omkrystalliserer fra isopropanol (8 g, sm.p. 180-193°C).
For fremstilling av basen oppløses dihydro-klorid i vann, gjøres alkalisk med natronlut og utrystes med butanol. Det etter tørkning og inndamping dannede residu anvendes direkte for neste trinn.
Analogt eks. 2 fremstilles de i tabell 1a opp-førte forbindelser med formel:
av det tilsvarende amin Ar-0-CH2-CH(OH)-CH^-NH- (CH2)3-NH"2 og S-metyl-isotiourinstoff eller O-metyl-isourinstoff.
De tilsvarende mengder av utgangskomponentene angitt i tabell 1b. Noen mindre avvikninger i forhold til eks. 1 er nevnt i fjerde spalte av tabell 1b.
De anvendte utgangsstoffer ifølge ppalte 2
av tabell 1b kan fremstilles analogt det i eks, 1 anvendte utgangsstoff.
Eksempel 12 3-((3-/ -2-metyl-indolyl- (4-) -oks_y7-2-hydroksy-propylamino ))-butylguaniden
Den som angitt nedenfor dannede hydrogenerings-
oppløsning som inneholder 3~ i{ 3-/_ 2-metyl-indolyl-(4-)-oksyl7~ 2-hydroksy-propylamino))-butylamin omsettes etter filtrering rnea S-metyl-isotiourinstoffsulfat (3,1 g i 13 ml vann) i løpet av 10 timers . koking. Man innstiller med fortynnet svovelsyre en pH på 5 og lar det stå i 10 timer. Nu suges det fra og omkrystalliseres fra vann. Utbytte: 3,9 g.
Sulfatets sm.p. 242-244°C (inneholder 1 mol vann).
Utgangsaminets fremstilling:
34., 4- g 1-/ 2-metyl-indolyl-(4)-oks_v7-2, 3-epoksy-propan omsettes sammen med 31 g 1rbenzylamino-3-amino-butan (fremstilt av 1,3-diaminobutan og benzylklorid i nærvær av kaliumkarbonat og etterfølgende fraksjonert vakuumdesti 11ering ,kp. 0,1 mm 95-96°C) i 250 ml dioksan 4 timer ved- koketempera-
tur. Man avdestillerer i vakuum og oppløser residuet i eddik-ester. Eddikesteroppløsningen utrystes flere ganger med van-dig vinsyreoppløsning, de forenede vandige faser gjøres alkalisk med 20 %- ig natronlut og utrystes deretter fem ganger med diklormetan. Etter tørking med natriumsulfat filtreres og inndampes på rotasjonsfordamper.
Residuet oppløser man nu 1 75 ml aceton og blander med en acetonisk maleinsyreoppløsning (11,5 g i 50 ml aceton). Neste dag frasuges, vaskes med etanol og tørkes.
13,5 g av det således dannede maleat (sm.p. 162-164°C) opptas i vann, oppløsningen gjøres alkalisk med 10$-ig natronlut og utrystes med eter under tilsetning av en liten etanolmengde. Etter tørking med natriumsul fat avdestillere r man og oppløser den gjenblivende base i etanol.
Etter tilsetning av 0,7 g av en 10 %- lg palla-diumkull hydrogeneres ved 6o°C. Det således dannede 3-C(3-_/ 2-metyl-indolyl- (4-) -oks_^7-2-hydroksy-propylamino ) ) - butylamin omsettes direkte videre uten isolering.
Eksempel 13
Anvendelse av et utgangsstoff med formel III hvori E betyr'en -C^-alkoksygruppe.
3-((3-/ naftyl - (1) - oksjjr7-2-hydroksy-propylamino ))-
propylguanidin
5,0 g 3((3-/ naftyl-(1 )-oks_y7"-2-hydroksypropyl-amino))-propylamin oppløses i 50 ml etanol og 15 ml vann,
og denne oppløsning kokes under omrøring og tilbakeløp sammen med 5, 5 g ,0-metyl-isourinstof f sulfat i 15 timer. Man innstiller med 10 %- ig svovelsyre en pH-verdi på 4--5, frasuger det utfelte sulfat, og vasker med vann og aceton.
Utbytte: ' 5 g.
.Sulfatets sm.p. 291-293°C (under' spaltning).

Ar betyr fenyl som er substituert med en C-j-C^-alkyl-, C|-<C>^-alkoksy-, C^-C^-alkenoksy- eller C^-C^-alkynoksy-gruppe eller et halogenatom, eller Ar er en naftyl- eller en eventuelt med en -C^-alkylgruppe substituert indolylgruppe, samt deres salter,
karakterisert ved at
et amin med formel
hvori Ar og Alk har den angitte betydning, omsettes med en forbindelse med formel
hvori E betyr en Cj-C^-alkylmerkaptogruppe eller -C^-alkoksygruppe, og hvis ønsket overføres forbindelsene til deres salter.
