Abstract

La presente invención se refiere a un proceso para la preparación de 5- (2-aminopirimidin-4-il) -2-aril- 1H-pirrol-3-carboxamidas y a los compuestos intermediarios útiles de ese proceso. 5-(2-Aminopirimidin-4-il)-2-aril- 1H-pirrol-3-carboxamidas se describen y reivindican en la patente WC 2007/110344, que también describe procesos para su preparación. Estos compuestos pueden prepararse convenientemente mediante un proceso que permite obtener los productos deseados con altos rendimientos y pureza. La síntesis comienza con el acoplamiento de un acetal acetilado con una sal de beta-cetoéster; el compuesto resultante es acetilado y después se hace reaccionar con un dialquil acetal de N,N-dimetilformamida, para dar un intermediario que es ciclado para dar éster 5-(2- aminopirimidin-4-il) -2-aril-1H-pirrol-3-carb oxílico; enseguida el éster carboxílico es hidrolizado y el ácido carboxílico resultante finalmente es condensado con una forma apropiada de amoníaco para dar la carboxamida deseada. Los compuestos preparados de conformidad con el proceso de la presente invención están dotados de actividad inhibidora de proteína cinasas y, más particularmente, actividad inhibidora de Cdc7 o Cdc7/Cdks. Por lo tanto, los compuestos son útiles en el tratamiento de una variedad de cánceres, trastornos de la proliferación celular y enfermedades asociadas con proteína cinasas.
