Abstract

La presente solicitud se refiere entre otras cosas a una célula aislada de vertebrado adecuada para la expresión recombinante de un polipéptido de interés, en el que la célula de vertebrado se altera para disminuir la función de la proteasa matriptasa endógena y en el que dicha célula comprende al menos un polinucleótido heterólogo que codifica un polipéptido de interés y en el que el polipéptido de interés es secretado por la célula. Se encontró que el uso de las células respectivas de vertebrados para producir un polipéptido recombinante de interés reduce significativamente la escisión del polipéptido de interés que se secreta en el medio de cultivo celular. También se proporcionan métodos mejorados de producción y de detección.
