Abstract
Se proporcionan aquí composiciones y métodos para mejorar la producción de acetil-CoA y compuestos derivados de acetil-CoA en una célula hospedera. En algunas modalidades, la célula hospedera se modifica genéticamente para comprender un ácido nucleico heterólogo que codifica una fosfoquetolasa (PK); y una disrupción funcional de una enzima endógena que convierte acetil fosfato a acetato. En algunas modalidades, la célula hospedera comprende además una secuencia de nucleótido heteróloga que codifica una fosfotransacetilasa (PTA). En algunas modalidades, la enzima que convierte el acetil fosfato a acetato es una glicerol-1-fosfatasa. En algunas modalidades, la glicerol-1-fosfatasa es GPP1IRHR2. En algunas modalidades, la glicerol-1-fosfatasa es GPP2/HOR2. Las composiciones y métodos descritos aquí proporcionan una ruta eficiente para la producción heteróloga de compuestos derivados de acetil-CoA, incluyendo, pero no limitado a, isoprenoides, policétidos, y ácidos grasos.
