Abstract
La presente invención se refiere a la detección de una secuencia de ácidos nucleicos objetivo por un ensayo de PCE-NR (Escisión de PTO y No Hibridación Dependiente de la Extensión). La presente invención adopta la ocurrencia de la inhibición de la hibridación entre el HO con el CTO por la formación del dúplex extendido dependiente del objetivo. Por lo tanto, la presente invención puede detectar secuencias objetivo incluso cuando el HO no se escinde. A este respecto, el diseño de la porción de marcado 5´ de secuencias de PTO, CTO y HO se puede realizar fácilmente y las condiciones para las reacciones también se pueden establecer fácilmente. Además, la detección del híbrido entre el CTO y el HO se puede realizar en un recipiente diferente de aquel para la extensión del CTO.
