Abstract

본 발명은 아미로펙틴에서 α-1.6형의 글루코시드 결합을 가수분해하는 성질을 갖고 산성배지에서와 약 60℃의 온도에서 효소활성을 갖는 열안정성 플루라나아제에 관한 것이다. 또한, 본 발명은 이 플루라나아제를 생성시키는 미생물의 균주와 이 플루라나아제의 제조방법에 관한 것이다. 또한 본 발명은 DNA분자에 관한 것이고, 본 발명은 이 DNA분자를 함유하는 합성벡터와 이 DNA분자를 함유 하는 염색체 집성벡터에 관한 것이다.

플루라아나제, 이를 제조하는 미생물, 이 플루라나아제의 제조방법과 이의 용도
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 플라스미드 pUBDEBRA1의 제한효소지도를 나타내고,
제2도는 플라스미드 pLD1을 제한효소지도를 나타내며,
제3도는 플라스미드 pUBCDEBR11DNSI의 제한효소지도를 나타낸다.

Claims (20)

  1. 균주 바실러스 데라미피칸스에 의하여 또는 이 균주의 유도체나 돌연변이체에 의하여 제조됨을 특징으로 하는 플루라나아제.
  2. N-말단 배열(SEQ ID NO:1)이 다음과 같이 아미노 카르복실 감응에서와 좌에서 우로 위치함을 특징으로 하는 플루라나아제:
  3. 제4도 (SEQ ID NO :11)에 예시된 바와같은 1~928아미노산의 아미노산 배열 또는 이로부터 유도된 수정배열로 이루어짐을 특징으로 하는 단리되고 정제된 플루라나아제.
  4. 299아미노산의 부가적배열(SEQ ID NO:12)을 함유하는 전구물질의 형태로 합성함을 특징으로 하는 플루라나아제.
  5. 야생상태의 알칼리성 플로테아제를 부호화하는 유전자를 함유하는 바실러스속의 미생물로 이종방법에 따라 바실러속의 미생물에서 삭제된 알칼리성 플로테아제를 부호화 하는 유전자를 제조함을 특징으로 하는 플루라나아제.
  6. 호기성 조건하에서 탄소 및 질소급원과 무기염을 함유하는 적당한 배양기에서 플루라나아제를 제조하고 얻은 플루라나아제를 수확할 수 있는 균주 바실러스 데라미피칸스 또는 이 균주의 유도체의 배양으로 이루어짐을 특징으로 하는 제1항 내지 제4항중 어느 한 항에 따른 플루라나아제의 제조방법.
  7. 플루라나아제를 부호화하는 DNA단편의 단리, 이 DNA단편을 적당한 벡터에 삽입, 이 벡터를 적당한 숙주에 주입 또는 이 DNA단편을 적당한 숙주의 염색체에 주입, 이 숙주의 배양, 플루라나아제의 합성과 플루라나아제의 수확으로 이루어짐을 특징으로 하는 제1항 내지 제4항중 어느 한 항에 따른 플루라나아제의 제조방법.
  8. 전분의 당화에 사용하는 제1항 내지 제5항중 어느 한 항에 따른 플루라나아제의 용도.
  9. 바실러스 데라미피칸스의 플루라나아제를 부호화하는 누클레오티드 배열(SEQ ID NO:10)또는 이러부터 유도된 수정배열로 이루어지는 DNA분자.
  10. 제9항에 있어서, 바실러스 데라미피칸스 T89.117D의 플루라나아제의 전체 유전자 (SEQ ID NO:8)로 이루어짐을 특징으로 하는 DNA 분자.
  11. 제9항에 따른 DNA분자를 함유하는 합성벡터 또는 염색체 집성백터.
  12. 합성벡터 pUBDEBRA1
  13. DUATORCP 집성벡터 pUBDEBRA11DNSI
  14. 제9항에 따른 DNA분자를 함유하는 바실러스 리케니포르미스의 형질전환된 균주.
  15. 제11항에 따른 합성벡터 또는 염색체 집성벡터를 함유하는 바실러스 리케니포르미스의 형질전환된 균주.
  16. 합성벡터 pUBDEBRA1 또는 염색체 집성벡터 pUBDEBRA11DNSI를 함유하는 바실러스 리케니포르미스의 형질전환된 균주.
  17. 제14항, 제15항, 또는 제16항에 따른 바실러스 리케니포르미스의 형질 전환된 균주에 의하여 플루라나아제.
  18. 바실러스 데라미피칸스의 단리되고 정제된 배양물과 이러부터 유도되거나 돌연변이된 배양물.
  19. 균주 바실러스 데라미피칸스 T 89.117D(LMG P-13056) 또는 이 균주의 유도체나 돌연변이체에 의하여 제조됨을 특징으로 하는 플라라나아제.
  20. 바실러스 데라미피칸스 T 89 .117D의 단리되고 정제된 배양물과 이로부터 유도되거나 돌연변이된 배양물.
