Abstract
본 발명은 프락토실트랜스피라제 활성 및 β-갈락토시다제의 활성을 갖는 오레오바시디움 플루란스(Aureobasidium pullulans) MW-4218(KFCC-10754)를 알지네이트를 담체로 하여 함께 고정화한 후 설탕과 유당을 기질로 하여 동시에 각각 프락토올리고당과 갈락토올리고당으로 전환시켜 두 올리고당이 혼합된 복합 올리고당을 제조하는 방법에 관한 것으로, 본 발명에 따르면 동일한 미생물에서 유래된 두 효소를 함께 고정화하여 사용하기 때문에 효소의 반응조건 등이 비슷하여 간단하게 반응을 수행할 수 있으며, 두가지 올리고당을 동시에 제조하므로 경제적이다.

Claims (6)
- 탄소원의 종류에 따라 프라토실트랜스피라제 또는 β-갈락토시다제를 생산할 수 있는 오레오바시디움 플루란스(Aureobasidium pullulans) MW-4218(KFCC-10754)를 각각 설탕 및 유당을 탄소원으로 하는 배지에서 배양하여 얻은 균체를 혼합하여 알지네이트에 고정화시켜 얻은 프락토실 트랜스피라제 활설 및 β-갈락토시다제 활성을 갖는 고정화 세포를 유당과 설탕을 함유하는 기질 용액과 반응시켜 갈락토올리고당과 프락토올리고당을 동시에 제조하는 것을 특징으로 하는 복합 올리고당의 제조방법.
- 제1항에 있어서, 고정화 균체는 혼합 균체(습균체)와 알지네이트의 비를 1:5~1:10의 범위로 하여 고정화시켜 얻은 것임을 특징으로 하는 복합 올리고당의 제조방법.
- 제1항에 있어서, 기질 용액은 50~90%의 설탕 및 10~60%의 유당을 함유함을 특징으로 하는 복합 올리고당의 제조방법.
- 제1항에 있어서, 반응 pH가 5~7인 것을 특징으로 하는 복합 올리고당의 제조방법.
- 제1항에 있어서, 반응 온도가 40~60℃인 것을 특징으로 하는 복합 올리고당의 제조방법.
- 제1항에 있어서, 반응은 회분식 혹은 연속식으로 수행하는 것을 특징으로 하는 복합 올리고당의 제조방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
