인체 염기성 FGF 뮤테인의 제조방법
[도면의 간단한 설명]
제1도는 실시예 1에 사용된 rhbFGF 뮤테인 CS23의 DNA 뉴클레오티드 서열 및 뉴클레오티드 서열을 코딩하는 단백질의 아미노산 서열을 나타낸다,
제2도는 실시예 1에서 수득된 플라스미드 pTB960의 제작과정을 나타낸다,
제3도는 실시예 5에서 수득된 정제 시료의 SDS-PAGE 패턴 및 마커를 나타낸다.

Claims (13)
- 성숙 인체 염기성 섬유아세포 성장 인자(hbFGF)의 적어도 하나의 구성 아미노산이 다른 아미노산으로 대체된 뮤테인의 코딩 뉴클레오티드 서열 및 그 상부의 T7프로모터를 함유하는 벡터.
- 제1항에 있어서, 적어도 하나의 구성 아미노산이 적어도 하나의 시스테인 잔기이고, 다른 아미노산이 세린 잔기인 벡터.
- 제1항에 있어서, 뮤테인이 성숙 hbFGF 구성 아미노산의 69- 및 87-위치의 시스테인 잔기가 세린 잔기로 대체된 뮤테인인 벡터.
- 제1항의 벡터에 의해 형질전환된 형질전환체.
- 제4항에 있어서, 숙주가 1ac 프로모터의 하부의 T7 RNA 폴리머라제 유전자를 갖는 에스케리키아 콜리(Escherichia coli)인 형질전환체.
- 제5항에 있어서, 이. 콜리(E. coli)MM 294(DE 3)/pCM 901의 특징을 갖는 형질전환체.
- 제5항에 있어서, 이 콜리(E. coli)MM 294(DE 3)/PCM 901의 특징을 갖는 형질전환체.
- 제4항의 형질전환체를 배지에서 배양함을 특징으로 하는, 성숙 hbFGF의 적어도 하나의 구성 아미노산이 다른 아미노산으로 대체된 뮤테인의 제조방법.
- 제항에 있어서, 제5하의 형질전환체의 대수성장기에 약 3-500μM의 이소프로필티오갈락토피라노시드(IPTG)를 배지에 가한 후 배양하는 방법.
- 제9항에 있어서, 3∼300μM의 IPTG를 사용하는 방법.
- 제10항에 있어서, 6∼200μM의 IPTG를 사용하는 방법.
- 제11항에 있어서, 6∼80μM의 IPTG를 사용하는 방법.
- 제8항에 있어서, 생성 뮤테인-함유 용액을 담체로 가교 다당류 황산염, 교환기로 술폰산기를 갖는 합성 중합체 및/또는 겔 여과용 합성 중합체를 사용하는 크로마토그래피에 의해 정제하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
