제1도는 체혈자로부터 표준 혈액 단위를 채혈하고 이를 냉동백의 HES 및 적혈구세포와 혼합하는 것을 도식으로 나타낸 것이며, 제2도는 본 발명에 따라 사용하기 위한 냉동 프레임중에 사용되는 플레이트의 평면도이고, 제3도는 제1도에 상응하는 평면도이며 플레이트상에서 냉동 백의 위치를 나타낸다.
Claims (22)
- 적혈구세포로부터 혈청 및 혈소판을 분리시키기 위해 단위 혈액을 원심분리시키고, 적혈구세포를 HES와혼합시키며, 이 혼합물을 냉동시키는 단계를 포함하고, 혼합전에 적혈구 세포가 90% 이상의 포장된 세포 용적(Packed Cell Volume)을 가지며 혼합물중의 HES 가 7% w/v HES/적혈구세포 미만으로 존재함을 특징으로 하여, HES를 저온보호제로 사용하는 적혈구 세포를 냉동시키는 방법.
- 제1항에 있어서 HES가 6% w/v HES/적혈구세포의 비로 존지함을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, HES가 40% w/v HES의 용액 형태임을 특징으로 하는 방법.
- 제1항 내지 제3항중 어느 한 항에 있어서, HES가 150,000 내지 200,000의 평균분자량을 가짐을 특징으로 하는 방법.
- 제1항 내지 제4항중 어느 한 항에 있어서, 혈액 단위가 표준 헌혈 단위 450ml에 기준함을 특징으로 하는 방법.
- 제1항 내지 제5항중 어느 한 항에 있어서, 적혈구 세포를 HES와 혼합하기전에 항웅고제로 처리하는 방법.
- 제1항 내지 제6항중 어느 한 항에 있어서, 냉동을 액체질소중에서 실시함을 특징으로 하는 방법.
- 제1항 내지 제7항중 어느 한 항에 있어서, 혼합물이 냉동백중에 있음을 특징으로 하는 방법.
- 제8항에 있어서, 냉동백이 냉동동안에 두개의 평행한 구멍을 갖는 판으로된 프레이중에 고정됨을 특징으로 하는 방법.

- 혈청 및 혈소판을 제거하고 실질적으로 200ml의 용적을 차지하는 90%이상의 적혈구세포 포장 세포 용적을 성취하기 위해, 450ml표준 혈액 채혈 단위를 원심분리시키며, 실질적으로 35ml의 40% w/v HES용액(이 HES는 150,000 내지 200,000의 평균 분자량을 갖는다) 을 함유하는 냉동백증에 도입시키고, 이 냉동백을 평행인 상태를 유지하면서, 서로에 대해 약간 정상적으로 이동할 수 있게 한 두개의 평행한 곡면판을 가진 냉동 프레임중에 위치시키며, 이 냉동 프레임을 액체 질소중에 위치시켜, HES 및 적혈구세포가 냉동될때까지 진탕시킴없이 그 내부에 방치시키는 단계를 포함함을 특징으로 하는 적혈구 세포 냉동방법.
- 제1항 내지 제18항중의 어느 한 항 또는 제21항에 따른 방법으로 냉동시킨 적혈구세포를, HES와 적혈구세포의 혼합물을 진탕없이 약 10분 동안 약 43.5℃에서 따뜻한 물중에 함침시킴을 특징으로 하여, 수혈용 적혈구세포를 제조하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개되는 것임.
