제1도는 실시예 1내지 3에 기술된 융합단백질의 개략적 표현을 나타내는데, S는 단백질 A암호서열을 나타내며, IGB는 IgB결합 영역을 나타내고,
제4도는 실시예에 기술된 바와 같이 pZZ-IGF-I의 구조에 대한 클로닝 전략을 나타낸다. AMP는 베타-락타마제를 코드하는 유전자이고, S는 신호서열이며, A-E는 단백질 A의 IgG결합 영역이고, ori는 복제의 기원이며, Z는 합성단편이고, IGF-I는 IGF-I에 대한 유전자이고, F1은 파아지(phage)f1으로부터의 복제의 기원이며, 1acZ는 베타-갈락토시다제에 대한 유전자이다.
제5도는 실시예 2와 3에 기술된 클로닝 전략을 나타낸다. Amp는 베타-락타마제를 코드하는 유전자이고, S는 암호서열이며, Z는 합성단편이고, IGF-I는 IGF-I에 대한 유전자이다.
Claims (11)
- 아미노산 서열을 코드하는 클로운 되거나 합성된 DNA절편을 발현벡터로 도입하는 단계, 상기 DNA의 절편은 상기 면역원 운반체 단백질을 코드하는 유전자에 융합되며, 이러한 발현벡터를 이용하여 숙주세포를 형질 전환시키는 단계, 법 상기 숙주세포로 하여금 융합단백질을 발현하게하는 것을 특징으로하는 면역원 운반체 단백질에 융합된 아미노산 서열로 구성된 융합 단백질의 제조방법.
- 제1항에 있어서, DNA절편은 플라스미드로 도입되는 것을 특징으로 하는 제조방법.
- 제2항에 있어서, 플라스미드는 천연 혹은 인공의 출신인것을 특징으로 하는 제조방법.
- 제1항 내지 제3항중 어느 하나에 있어서, DNA 절편은 적절한 제한효소로 분해하여 도입되는것을 특징으로 하는 제조방법.
- 제1항 내지 제4항중 어느 하나에 있어서, 천연단백질의 항원결정인자를 코드하는 합성 DNA 절편은 발현 벡터로 도입되고 거기에서 상기의 운반체 단백질을 코드하는 유전자에 융합되는 것을 특징으로하는 제조방법.
- 제1항 내지 제5항중 어느 하나에 있어서, 운반체단백질은 면역글로불린결합단백질(IGB)인것을 특징으로 하는 제조방법.
- 제6항에 있어서, 운반체 단백질은 황색포도상구균 또는 그것과 관련된 단백질인 것을 특징으로 하는 제조방법.
- 제1항 내지 제7항중 어느 하나에 있어서, 아미노기 말단끝과 카르복실기 맡단끝 모두는 운반체 단백질에 융합되는 것을 특징으로 하는 제조방법.
- 제9항의 융합단백질을 이용하여 동물을 면역하여 동물을 출혈시켜 동물혈청으로부터 생산된 항체를 회수하는 것으로 구성되는 것을 특징으로 하는 다클로운성 항체의 제조방법.
- 제9항에 있어서, 면역된 동물은 포유동물인 것을 특징으로 하는 제조방법.
- 제10항에 있어서, 포유동물은 토끼와 쥐로부터 선택되는 것을 특징으로 하는 제조방법.※ 참고사항: 최초출원 내용에 의하여 공개하는 것임.
