제1도는 본 발명에 따라 수득된 메탄올 중에서의 사프라마이신 Y3의 자외선(uv) 흡수스펙트럼이다.
제2도는 본 발명에 따라 수득된, 클로로포름 중에서의 사프라마이신 Y3의 적외선(IR) 흡수 스펙트럼이다.
제3도는 본 발명에 따라 수득된, 메탄올 중에서의 사프라마이신 Yd-1의 자외선 흡수 스펙트럼이다.
Claims (4)
- 사프라마이신 A를 생성시킬 수 있는 스트렙토마이세스 속(genus)의 균주로, 탄소와 질소원 및 필수미량원소를 함유하는 배지를 접종시키고 호기조건하에서 약 25 내지 35℃의 온도 및 약 4.0 내지 7.5의 pH수준으로 배양함으로써 예비 배양물(preculture)을 제조하고, 수득된 바이오매스(biomass)를 단리시킨 후, 이 바이오매스의 수성 현탁액을 R이 수소 또는 C1-C4-알킬인 일반식 HOOC-CH(NH2)-CH2-R의 아미노산과 반응시키고 글리신, L-티로신 및 L-메티오닌과 반응시키거나, HOOC-CH(NH2)-CH2-R, 글리신, L-티로신, 및 L-메티오닌 중에서 선택된 아미노산에 의해 형성된 펩티드와 반응시킨 다음, 시안화물염을 배양여과액에 첨가한 후, 수득된 일반식(Ⅰ)의 사프라마이신 A유도체를 단리 및 정제시키고, 임의로, 산부가염으로 전환시키거나; 또는 -R이 수소인 일반식(Ⅰ)의 사프라마이신 A유도체(사프라마이신 Y3)를 제조하기 위하여-사프라마이신 A를 생성시킬 수 있는 스트렙토마세이스속의 균주로, 탄소와 질소원 및 필수미량 원소를 함유하는 배지를 접종시키고 호기존건하에서 약 25°내지 35℃의 온도 및 약 4.0 내지 7.5의 pH수준으로 배양함으로써 배양물을 제조한 다음, 수득된 사프라마이신 Y3를 단리 및 정제시키고, 임의로 산부가염으로 전환시킴을 특징으로 하여, 하기 일반식(Ⅰ)의 화합물 및 이의 산부가염을 제조하는 방법.상기식에서, R은 수소 또는 C1-C4-알킬이다.
- 제1항에 있어서, 수득된 바이오매스의 수성 현탁액을 R이 수소인 일반식 HOOC-CH(NH2)-CH2-R의 아미노산과 반응시키고 글리신, L-티로신 및 L-메티오닌과 반응시키거나, HOOC-CH(NH2)-CH2-R (R=H), 글리신, L-티로신 및 L-메티오닌중의 아미노산에 의해 형성된 펩티드와 반응시킴을 특징으로 하여, R이 수소인 일반식(Ⅰ)의 화합물을 제조하는 방법.
- 제1항에 있어서, 수득된 바이오매스의 수성 현탁액을 R이 메틸인 일반식 HOOC-CH(NH2)-CH2-R의 아미노산과 반응시키고 글리신, L-티로신 또는 L-메티오닌과 반응시키거나, HOOC-CH(NH2)-CH2-R (R=CH3), 글리신, L-티로신 또는 L-메티오닌중의 아미노산에 의해 형성된 펩티드와 반응시킴을 특징으로 하여, R이 메틸인 일반식(Ⅰ)의 화합물을 제조하는 방법.
- 제1항에 있어서, 배지를 균주 스트렙토 마이세스 라벤듈리 No. 314(FERM-P 3218)로 접종시킴을 특징으로 하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
