Claims (2)
- (ⅰ) 다음 일반식(Ⅱ)의 화합물을 알카리금속 수소화물 존재하에 고리화반응시키는 단계와, (ⅱ) 다음 일반식(Ⅲ)의 얻어진 생성물을 다음 일반식(Ⅳ)의 아세톤페논과 반응시켜 다음 일반식(Ⅴ)의 화합물을 생성케 하는 단계와, (ⅲ) 그후 제조하고저 하는 물질에 따라서 (a) 아세트산 암모늄과 아세트산의 존재하에 화합물(Ⅴ)을 고리화 반응시키고, 다음 일반식(Ⅵ)의 얻어진 화합물을 처음에 가성소다로 처리하고, 다음에 염산으로 처리하여 X가 -NH-이고 R2가 벤질인 일반식(Ⅰ)의 피롤로피리딘을 생성케하거나. 또는 (b) 강산의 존재하에 화합물(Ⅴ)를 고리화반응시켜서 X가-O-이고, R2가 벤질인 일반식(Ⅰ)의 화합물을 알킬화시켜 R2가 메틸이나 에틸인 화합물을 생성케 하는 단계와, (ⅳ) 요구에 따라서는, 이와 같이 얻어진 화합물(Ⅰ)을 탈벤질화시키고, 동시에 R1이 니트로페닐을 나타내는 경우 아미노페닐이 되게 환원시켜서 R2가 수소인 일반식(Ⅰ)의 화합물을 생성케하는 단계와, (ⅴ) 또한 요구에 따라서는, R2가 수소인 일반식(Ⅰ)의 화합물을 알킬화시켜 R2가 메틸이나 에틸인 화합물을 생성케 하는 단계와, (ⅵ) 요구에 따라서, (ⅲ),(ⅳ)또는 (ⅴ)단계로부터 얻어진 일반식(Ⅰ)의 피리딘 유도체를 공지된 방법으로, 약학적으로 사용가능한 산부가염으로 저환시키는 단계로 구성되는 것을 특징으로 하는 다음 일반식(Ⅰ)의 피리딘 유도체를 제조하는 방법.(Ⅰ)(Ⅱ)(Ⅲ)(Ⅳ)(Ⅴ)(Ⅵ)상기식에서 X는 -0- 또는 -NH- 이고 R1은 하나 또는 두개의 할로겐원자로 임의치환된 페닐기이거나, 또는 메틸, 에틸, 트리플루오로메틸,메톡시 또는 페닐기이거나, 또는 R2가 벤질기가 아닌 경우에는 아미노기이고, 또는 나프틸이나 벤조디옥사닐기이며, R2는 수소 또는 벤질기이거나, 또는 X가 -0-인 경우 R2는 메틸이나 에틸기일 수 있고, (Ⅳ)에서의 R1은 상기와 같거나 또는 니트로페닐이다.
- 제1항에 있어서, X는 -0-이고 R2는 메틸이나 에틸이며, 그의 염이 제조되는 경우에는 염산염인 것을 특징으로 하는 제조방법.※ 참고사항 : 최초출원내용에 의하여 공개하는 것임.
