인체 r-인터페론활성을 감는 폴리펩타이드의 제조방법

제1도는 유전자단편 IFN-1을 pBR 322로 삽입시킨 하이브리드 플라스미드를 나타내고, 제2도는 유전자 단편 IFN-2를 pUC 8으로 삽입시킨 하이브리드 플라스미드를 나타내며, 제3도는 유전자단편 IFN-Ⅲ를 pUC 8으로 삽입시킨 하이브리드 플라스미드를 나타내고, 제4도는 DNA-서열 I 을 pUC 8으로 삽입시킨 하이브리드 플라스미드를 나타내며, 제5도는 DNA-서열 IB를 pUC 8으로 삽입시킨 하이브리드 플라스미드를 나타내고, 제6도는 DNA-서열 IA를 pUC 8으로 삽입시킨 하이브리드 플라스미드를 나타내며, 제7도는 DNA-서열 IC를 pMX2로 삽입시킨 하이브리드 플라스미드를 나타내며, 제8도는-인터페론 유전자단편 IA*를 pMX2로 삽입시킨 하이브리드 플라스미드를 나타내고, 제9도는-인터페론 유전자단편 IB를 pMX2M로 삽입시킨 하이브리드 플라스미드를 나타내며, 제10도는-인터페론 유전자단편 IC*를 pMX2로 삽입시킨 하이브리드 플라스미드를 나타낸다.

Claims (3)

  1. 일반식(1)의 DNA를 갖는 합성유전자를 숙주미생물에서 발현시킴을 특징으로 하여, 아미노산 서열 5내지 127, 1내지 127 및 5내지 146으로 이루어진 부분서열의 인체감마 인터페론(IFN-)또는 이들부분 서열 및 감마 인터페론활성을 갖는 인체감마 인터페론의 동족체(이때, 동족체중의 천연의 아미노산서열은 아미노산의 교환, 삽입 또는 제거에 의해서 변형되었다)를 유전공학적으로 제조하는 방법.
    (Pr)X-XY-R-YZ-Z (1)
    상기식에서, x,y 및 z는, 각기 독립적으로, 0 또는 1을 나타내나 X가 Met-Cys-Tyr-Cys-Gln인 경우에는 y 및 z가 모두 다 1은 아니며, Pr은 숙주미생물내에서 또는 시험관내에서 제거되는 프리펩타이드를 코드화하는 뉴클레오티드 서열이고, X는 Met, Met-Cys-Tyr-Cys-Gln 또는 H-Cys-Tyr-Cys-Gln을 코드화하는 뉴클레오티드 서열이며, R은 아미노산 서열 5 내지 127 또는 이 아미노산 서열의 변형체를 코드화하는 뉴클레오티드 서열이고, Y는 아미노산 서열 128내지 146또는 이 아미노산 서열의 변형체를 코드화하는 뉴클레오티드 서열이며, Z는 하나 또는 2개의 정지코돈을 나타낸다.
